Genetically accessible variation to some of the abiotic and biotic stresses are limited in the cultivated lentil (Lens culinaris Medik.) germplasm. Introgression of novel alleles from its wild relative species will be useful for enhancing the genetic improvement of the crop. L. ervoides, one of the wild relatives of lentil, is a proven source of disease resistance for the crop. Here we introduce a lentil advanced backcross (LABC-01) population developed in cultivar ‘CDC Redberry’ background, based on L. ervoides alleles derived from an interspecific recombinant inbred population, LR-59-81. Two-hundred and seventeen individuals of the LABC-01 population at BC2F3:4 generation were screened for the race 0 of anthracnose (Colletotrichum lentis) and stemphylium blight (Stemphylium botryosum) under controlled conditions. The population showed significant variations for both diseases and the transfer of resistance alleles into the elite cultivar was evident. It also segregated for other traits such as days to flowering, seed coat colour, seed coat pattern and flower colour. Overall, we showed that LABC-01 population can be used in breeding programmes worldwide to improve disease resistance and will be available as a valuable genetic resource for future genetic analysis of desired loci introgressed from L. ervoides.

The present research was conducted to determine the reactions of 200 pure lines selected from bread wheat landraces collected from 18 provinces and seven regions of Turkey against bunt disease (Tilletia foetida) under field conditions for 3 years. Bunt disease reactions of pure lines were assessed based on the infected spike/total spike ratio. For visually assessed materials, the GGE-biplot method, where G = genotype effect and GE = genotype-by-environment effect, was used to group the reactions against bunt disease. Fifty-nine pure lines showed high resistance (with infection rates ranging from 0.1 to 10%); 24 in the moderate resistance (with infection rates ranging from 10.1 to 25%); 75 in the moderate susceptibility (with infection rates ranging from 25.1 to 45%); 38 in the susceptibility (with infection rates ranging from 45.1 to 70%) and finally four in the highly susceptibility (with infection rates of >70.1%). PC1 and PC2 of the GGE-biplot graph created over the years explained 76.49% of the total variation. The GGE-biplot graph provided efficient identification of resistant genotypes. The lowest PC1 values and PC2 values close to 0.0 explained the resistance of pure line to bunt disease best. The resistance of pure lines to bunt disease over the biplot decreased from the first section through the last section. Based on the results of present study, 19 pure lines (located within the first circle of the biplot graph) were selected for resistance breeding programmes against the diseases.

Germplasm accessions of wild Lactuca species are maintained worldwide in ex situ collections as gene reservoirs for quality and disease resistance traits for cultivated lettuce. Accessions of 12 Lactuca species from 6 genebanks were compared via morphological characterization and AFLP (Amplified Fragment Length Polymorphism)-based profiling to estimate the extent of duplication. A method of assessing redundancy within very similar, but not identical accessions, is proposed, based on 352 polymorphic AFLP products. Seven duplication groups showed a high level of AFLP similarity, and one pair of Lactuca saligna accessions displayed identical AFLP profiles. In several cases, the morphological assessment indicated that a taxonomic reclassification of accessions was necessary. Candidate duplicates were identified using population parameters and inter- and intra-accession variability. The implications of these findings on the conservation of wild species are discussed.

Sustainable utilization of plant genetic resources for food and agriculture has been increasingly discussed at both national and international forums. Besides exploitation, conservation of plant genetic resources has become an integral part of these discussions. Conservation aims at maintaining the diversity of living organisms, their habitat and the interrelationship between organisms and their environment. For achieving such goals, appropriate conservation strategies have to be adopted. Determining the genetic makeup of a particular plant species is of critical importance when planning a suitable conservation strategy. In this study, we sequenced the chloroplast trnK intron, matK and rbcL gene aimed at understanding the rarity of Nymphaea tetragona, a critically rare and endangered plant of India found at only one location. We extended our investigation to other Nymphaea species such as N. nouchali, N. pubescens and N. rubra that are commonly available throughout India. Interestingly, matK gene of N. tetragona revealed high number of non-synonymous substitutions. Molecular evolutionary analysis indicated that three of these sites may be under mild selective pressures. Such adaptive changes at the DNA and protein sequence level of matK gene may have been associated with the colonization of N. tetragona, suggesting that it could have migrated from China.

Recombinant inbred lines (RILs) generated from a cross between a cultivated species and its wild progenitor species serve as important germplasm for introgressing valuable genes from a wild species to a cultivated species. During this breeding process, it is equally important to prevent the loss of agronomically important genes in the cultivated species. In an effort to establish an efficient selection system for the single Rsv1 gene conferring durable resistance to soybean mosaic virus (SMV) in the soybean cultivar Hwangkeum (also known as Suweon 97), which is a parent of a RIL population from Hwangkeum (cultivated soybean) × IT182932 (wild soybean), in the present study, we unexpectedly identified an additional necrosis-conditioning gene unmasked by a recombination in the middle of the Rsv1-containing nucleotide-binding leucine-rich repeat gene cluster region and the Rsv3 gene in Hwangkeum. Thus, Hwangkeum contains at least three SMV resistance genes consisting of two tightly linked genes at the Rsv1 locus and the Rsv3 locus. The results of this study provide additional important information for use of the Hwangkeum genome in soybean breeding programmes.

The importance of plant genetic resources for the future of agricultural production and for achieving food security necessitates the study of the factors affecting their most efficient exploitation, particularly in breeding programmes. The established negative correlation between competitive and yielding ability is emerging as an important, yet overlooked cause of seed and variety degradation. Because of this negative correlation, the low yielding, strong competing plants within the variety or the germplasm under study acquire a survival advantage over the high yielding, weak competing plants when propagated under dense stands, leading to a gradual cultivar degeneration and identity loss. Moreover, this gradual degeneration prevents selecting for the positive and novel adaptive variation that is endlessly released by the genome in response to biotic (e.g. mutating pathogens) and abiotic stresses. The application of nonstop selection on individual plants grown in the absence of competition using a novel selection equation demonstrates an effective means to counteract the negative effects and accelerate progress through selection.

Soybean [Glycine max (L.) Merr.] is one of the most important legume crops in the world. However, soybean grain yield is extensively affected by environmental stresses such as soil salinity. In this study, we evaluated the germplasm of 51 Indonesian soybean accessions for salt tolerance to identify salt-tolerant germplasms for possible use in breeding for soybean salt tolerance. Based on experiments under hydroponic conditions, adding 100 mM of NaCl to a 1/2 concentration of Hoagland and Arnon solution, several Indonesian soybean germplasms, such as Java 7, Seputih Raman, Tambora, Ringgit (JP 30217), Sinyonya (early) and Sinyonya (late) were identified as salt-tolerant in terms of salt tolerance rate (STR) and leaf chlorophyll content (SPAD value) taken with the Konica Minolta SPAD-502 chlorophyll meter. The selected salt-tolerant germplasms were further evaluated under soil medium cultivation in pots irrigated with 100 mM NaCl for around 5 weeks. The six selected soybean germplasms again showed higher salt tolerance in terms of SPAD, STR and shoot dry weight. Expression analysis of the salt tolerance gene Ncl revealed a significant positive correlation between Ncl expression and salt tolerance, suggesting that Ncl is essential for salt tolerance in the Indonesian soybean germplasms we tested. The salt-tolerant Indonesian soybean germplasms identified in this study could be used in local soybean breeding practices for the improvement of salt tolerance.

The pattern of inheritance of powdery mildew resistance expressed by two bread wheat (cultivar ‘Alcedo’)/Aegilops markgrafii introgression lines was explored using F2 populations bred from crosses made with the powdery mildew-susceptible cultivar ‘Kanzler’. Disease reaction was tested at both seedling and adult plant stages. Two resistance loci, designated QPm.ipk-1A and QPm.ipk-7A, were identified as mapping to the distal ends of chromosome arms 1AS and 7AL, respectively. Whereas QPm.ipk-1A was expressed throughout the plant's life, QPm.ipk-7A was only effective at the seedling stage. The map location of both resistance loci indicated that resistances originated from A. markgrafii. The possible genetic relationship of these disease-resistant genes to known Pm genes is discussed in the context of synteny.

Genebanks play an important role in the conservation of global plant biodiversity. The European Search Catalogue for Plant Genetic Resources (EURISCO) was created as a central entry point to provide information on these collections. However, a major challenge lies in the heterogeneity of scientific plant names. This makes the selection of suitable plant material, e.g. for research or breeding purposes, significantly more difficult. For this reason, the taxonomic backbone of EURISCO has been completely revised. Search terms entered by users are now automatically checked against taxonomic reference repositories, allowing a variety of synonyms to be identified. In addition, a fuzzy search has been implemented, which makes the search function tolerant of erroneous data (e.g. caused by typing errors). Besides improvements of the search interface, more support will be given to EURISCO's data providers. The new developments provide a tool that makes it easier to identify problem cases within the data, such as accepted/non-accepted taxonomic names, and will successively improve the quality of taxonomic information in EURISCO.

This article links the concept of access and benefit-sharing as it pertains to crop genetic resources to climate change adaptation and argues that systems for access and benefit-sharing can, and should, be designed to contribute to climate change adaptation for agriculture. The access and benefit-sharing provisions of the two international agreements that together provide the international legal framework for access and benefit-sharing – the Convention on Biological Diversity (with its Nagoya Protocol) and the International Treaty on Plant Genetic Resources for Food and Agriculture – are presented and analysed. How these agreements are implemented is central to adaptation, as the effects of climate change threaten crop genetic resources and future adaptive capacity, and, if properly maintained and utilized, crop genetic resources will be essential to climate change adaptation across the globe. This article, therefore, argues that an important adaptation strategy linked to such implementation is to direct benefit-sharing for crop genetic resources towards adaptation efforts and to ensure facilitated and efficient access to crop genetic resources for adaptation purposes. Some options for how this can be pursued at both the international and national level are offered.

Genetic diversity and relationship of 92 bread wheat (Triticum aestivum L.) genotypes from India and exotic collections were examined using simple sequence repeat (SSR) markers and phenotypic traits to identify new sources of diversity that could accelerate the development of improved wheat varieties better suited to meet the challenges posed by heat stress in India. Genetic diversity assessed by using 82 SSR markers was compared with diversity evaluated using five physiological and six agronomic traits under the heat stress condition. A total of 248 alleles were detected, with a range of two to eight alleles per locus. The average polymorphic information content value was 0.37, with a range of 0.04 (cfd9) to 0.68 (wmc339). The heat susceptibility index was determined for grain yield per spike, and the genotypes were grouped into four categories. Two dendrograms that were constructed based on phenotypic and molecular analysis using UPGMA (unweighted pair group method with arithmetic mean) were found to be topologically different. Genotypes characterized as highly heat tolerant were distributed among all the SSR-based cluster groups. This implies that the genetic basis of heat stress tolerance in these genotypes is different, thereby enabling wheat breeders to combine these diverse sources of genetic variability to improve heat tolerance in their breeding programmes.

Annual wild soybean characterized by low 100-seed weight (100SW), high protein content (PRC) and low oil content (OIC) may have favourable exotic genes/alleles for broadening the genetic base of the cultivated soybean. To evaluate the wild alleles/segments, a chromosome segment substitution line population comprising 151 lines with N24852 (wild) as the donor and NN1138-2 (cultivated) as the recurrent parent was analysed using single-marker analysis, interval mapping, inclusive composite interval mapping and mixed linear composite interval mapping. On 14 segments of ten chromosomes, 17 quantitative trait loci (QTL) were identified, with two segments each containing two QTL for 100SW and OIC and one segment containing two QTL for PRC and OIC, respectively. All the seven wild alleles/segments for 100SW were associated with negative effects and three were associated with positive effects, but one was associated with a negative effect for PRC, and five were associated with negative effects, but one was associated with a positive effect for OIC. Except Satt216 and Sat_224 for 100SW, the identified QTL/segments have been reported from cultivated soybean mapping populations. The detected wild segments may provide materials for further characterization, cloning and pyramiding of the alleles conferring the seed-quality traits.

Taxonomy plays an essential role in genebank documentation. It is often the first level at which users search material, and it determines the protocols used in the management of collections. Especially, when plant genetic resources information is pooled in systems such as EURISCO, the European catalogue of ex situ plant genetic resources, problems regarding technical handling of taxonomic nomenclature, such as lack of standardization and low quality of data, become apparent. These problems were studied by analysing the content of EURISCO and mapping the taxon names in EURISCO on those used in the United States Department of Agriculture genebank system GRIN-Tax. Thus, the number of spelling errors and the level of standardization could be quantified and improved. An analysis of the content of EURISCO was made, showing a highly unbalanced distribution over crops: 50% of the accessions belong to ten genera only. Mapping EURISCO on the crops listed in Annex 1 of the International Treaty on Plant Genetic Resources for Food and Agriculture showed that 67% of the accessions in EURISCO belong to crops in that list.

Research on resistance to black pod caused by Phytophthora megakarya has been carried out in Cameroon for 40 years. It has yielded a significant database which is currently being exploited for the modelling of disease resistance levels of the major clones selected as candidate parents for the creation of new resistant and consequently more productive cocoa varieties. These outputs are expected to meet the increasing need of producers for improved planting material to extend cultivation into emerging cocoa-growing areas or to renew old cocoa production basins. These outputs have been made possible through an international collaborative effort and, more recently, within the scope of regional or international initiatives sponsored by CFC/ICCO/IPGRI and by CAOBISCO. New selection approaches and methodologies, such as rapid field assessment and early screening tests, have been promoted, along with the use of molecular tools for genome mapping and disease resistance quantitative trait locus (QTL) identification. Promising material has been introduced, evaluated in the laboratory and on-farm, and compared to the local cocoa germplasm. The basic knowledge developed from this local database has enabled the definition of an empirical disease resistance sorting and subsequently a prediction of the yield gains expected from the use of different cocoa clones and possible combinations (progenies) available on-station and in farmers' stands in different agro-ecological growing conditions; or which have to be created following strategies in the new breeding programme.

The study of unknown and therefore unexploited genetic material from landraces and wild relatives could be essential to help modern plant breeders to respond to ongoing requirements and new challenges in food production. The present study evaluates the most relevant physicochemical values and nutrient contents of a genetically unique array of traditional melon varieties, cultivated in Spain at least since the 19th century, and compares them with modern melon hybrids available on the market. This research is complemented with an assessment of variety, environment and repetition effects on each trait to determine their stability. Spanish melon landraces displayed extraordinary diversity with respect to juiciness (70.59–95.97 g/100 g water fresh weight), firmness (20.75–149.89 N), soluble solids content (9.57–16.53 °Brix), pH (5.04–6.38), total sugars (360.21–877.36 mg/g dry weight), carotenoids (0.01–2.05 μg/g fresh weight) and ascorbic acid values (7.55–44.33 mg/100 g fresh weight). A subset of these landraces, belonging to Piel de Sapo and Rochet market classes, revealed remarkably superior values of ascorbic acid in comparison with all commercial varieties, doubling ascorbic acid values with respect to their corresponding market class. Furthermore, most of these landraces exhibited high acidity and accumulated high levels of sugars, fulfilling those sensory and physicochemical characteristics that researchers and breeders have spent many years seeking. The possibilities of these landraces to be used in improvement projects are innumerable; they should be surely taken into account in the near future.

The aims of the current study were to investigate the effect of drought stress on phenological and morphological traits of sunflower genotypes and to determine the important traits for identifying drought-tolerant and drought-sensitive cultivars. For this purpose, a lattice square-design experiment was conducted with 64 sunflower genotypes in an 8 × 8 pattern with two replications under non-stress and moisture-stress conditions (irrigation holding at the flowering stage) during 2 years 2016 and 2017. Measured and recorded traits were included the some phenological and morphological traits seeds. Sil-96 genotype showed the highest yield under both non-stress and moisture-stress conditions. Among the morphological traits, the head and stem diameters were highly significant to determine the final yield. In comparing the genotypes, it was concluded that the number of seeds per head was the most influential component affecting the yield. Furthermore, 1000-seed weight was the most important factor affecting grain yield under moisture-stress conditions. The result of association analysis study shows that 45 of inter-simple sequence repeat markers in a general linear model are associated with yield and yield component traits and 23 of them were verified in a mixed linear model (MLM) association approach. Also, 32 markers were informative for morpho-physiological traits and 24 of them verified using the MLM. Finally, 19 informative markers were identified for phenological traits and 10 of them were verified by the MLM.

Understanding the genetic composition and population structure of plant species at a molecular level is essential for the development of adequate strategies aimed at enhancing the conservation and use of their genetic resources. In addition, such knowledge can help to plan ahead for a scenario under which wild and cultivated species come into contact with their genetically modified (GM) counterpart(s). Using ten simple sequence repeat markers, we genotyped 409 samples pertaining to the species in the Manihot genus known to occur in Colombia, i.e. cassava (Manihot esculenta) and its wild relatives Manihot brachyloba, Manihot carthaginensis and Manihot tristis. High genetic variation was observed in all the species (HE= 0.212–0.603), with cassava showing highest diversity. Most of the genetic variation was found within species populations. Our results suggest that outcrossing events among populations occur much more frequently in M. tristis and M. esculenta, and particularly so in the latter, where the exchange of varieties among local farmers plays a key role in maintaining and introducing new genetic diversity. The occurrence of gene flow within and among populations of Manihot species in Colombia becomes relevant in a biosafety context, where gene flow from GM cassava, if introduced to the country, might have detrimental effects on the structure and dynamics of populations of wild species. The baseline information on the genetic diversity and structure of the four Colombian species that we have presented here provides a first and indispensable step towards the development of targeted interventions necessary to preserve their genetic resources.

Genetic diversity is a valuable asset for crop improvement. In this study, a total of 50 rice genotypes were screened for salinity tolerance at the reproductive stage using gravel-based hydroponics, soil, controlled mini-field and field methods. Different morpho-agronomic, physiological markers and tolerance indices were used to classify tolerant and susceptible genotypes. Our results showed high genetic variability in response of rice genotypes to salinity using different screening methods. The significant effect (P < 0.01) of salinity include increased Na+ sequestration in the flag leaf, increased unfilled grains (except for the field method) and reduced pollen fertility, total yield, panicle length and the average number of filled grains per panicle. Plant height (except for the soil method) and K+ ion concentration in the flag leaf were not significantly affected by salinity (P > 0.05). Genetic diversity analysis indicated that the germplasm evaluated exhibits moderate diversity (PIC ${\overline{\bf X}}$ = 0.2085). Cluster analysis using single nucleotide polymorphisms showed that the genetic make-up of rice germplasm somehow did not necessarily indicate their over-all tolerance or susceptibility to salinity. This study proved that the controlled mini-field method is the most advantageous among the screening methods while geometric mean productivity, stress tolerance index and yield index are the tolerance indices that can be classified as better predictors of salinity tolerance considering the yield potentials of the genotypes. The genotypes Nona Bokra and Mushkan 41 can be used for breeding in the future through low Na+:K+ ratio while Damodar and Bhura Rata 4-10 for breeding salt-tolerant cultivars with higher yield potentials.

Soybean (Glycine max (L.) Merr.) is an important legume crop with high commercial value widely cultivated globally. Thus, the genetic characterization of the existing soybean germplasm will provide useful information for enhanced conservation, improvement and future utilization. This study aimed to assess the extent of genetic diversity of soybean elite breeding lines and varieties developed by the soybean breeding programme of the International Institute of Tropical Agriculture (IITA), Ibadan, Nigeria. The genetic diversity of 65 soybean genotypes was studied using single-nucleotide polymorphism (SNP) markers. The result revealed that 2446 alleles were detected, and the indicators for allelic richness and diversity had good differentiating power in assessing the diversity of the genotypes. The three complementary approaches used in the study grouped the germplasm into three major clusters based on genetic relatedness. The analysis of molecular variance revealed that 71% (P < 0.001) variation was due to among individual genotypes, while 11% (P < 0.001) was ascribed to differences among the three clusters, and the fixation index (FST) was 0.11 for the SNP loci, signifying moderate genetic differentiation among the genotypes. The identified private alleles indicate that the soybean germplasm contains diverse variability that is yet to be exploited. The SNP markers revealed high diversity in the studied germplasm and found to be efficient for assessing genetic diversity in the crop. These results provide valuable information that might be utilized for assessing the genetic variability of soybean and other legume crops germplasm by breeding programmes.

Seed composition, including the protein, lipid and sucrose contents of 334 accessions of wild soybean (Glycine soja) collected in Japan, was evaluated using near-infrared reflectance spectroscopy (NIRS) technology. The distribution of protein, lipid and sucrose contents and correlations among these three classes of seed components were determined. Protein, lipid and sucrose levels ranged in accessions from 48.6 to 57.0, 9.0 to 14.3 and 1.24 to 3.53%, respectively. Average levels of protein, lipid and sucrose in the accessions were 54, 11 and 2.5%, respectively. High negative correlations were observed between the protein and lipid contents, and the protein and sucrose contents. Mean levels of the three constituents were compared among collection sites classified by climatic conditions. The total protein content of accessions from regions with a high annual mean temperature was high. The protein content of accessions from the II-1 region was higher than those from the III-3 region, and the sucrose content from the II-1 region was lower than those from regions III-2 and IV-3. The lipid content of plants from the II-1 region was lower than those from other regions, and the accessions in region II had a higher protein content and lower sucrose and lipid contents than the other regions. These results provide diverse and wide-ranged protein, lipid and sucrose contents information of Japanese wild soybean resources according to climatic region; thus, providing a foundation for the future development and selection of new soybean varieties with desired traits in global environmental changes.
