An interspecific hybrid Jatropha curcas (Jc) × Jatropha integerrima (Ji) was developed between jatropha (Jc) from Mexico and Ji from Thailand. Jc is a large canopy plant with soft wood, while Ji is a tall plant with semi-hard wood. The F1 hybrid and their parents were grown under field conditions at a spacing of 1 m × 1.5 m. One-year-old plants were harvested and determined for biomass yield, calorific value and chemical composition of wood, as well as for heterosis (hybrid vigour) of these characters. The F1 plants gave an average fresh wood weight of 18.07 kg/plant, a moisture content of 46.56%, a dry wood weight of 9.56 kg/plant and a wood density of 0.62 g/cm3. The F1 hybrid had less ash (2.60%) than Jc (6.93%), but a higher heat value of wood (18.73 MJ/kg) than Jc (17.77 MJ/kg). Heterosis over mid-parent was very high and positive in dry wood weight, fresh wood weight and number of secondary branches at 542, 310 and 450%, respectively, while negative heterosis was found in moisture content ( − 24.86%). The desirable traits found in the F1 hybrid can be fixed by cutting propagation of the selected plants.

Amylose-free (waxy) maize has been a vegetable (cooked ears) and staple food in Southeast Asia for centuries, resulting in hundreds of landraces (LRs) across the region. The recessive waxy allele induces soft grains with preferred cooking and flavour properties. We hypothesized that eating preferences resulted in the additional selection for different starch properties, reflected in altered starch granule morphology or amylopectin structure. A total of 41 LRs were available as starting material that had been used by different ethnic groups in Vietnam and Thailand. Unluckily, some LR were not pure waxy, but we successfully regained the original pure waxy status for most. Twenty LR were chosen for analysis of starch traits according to their purity. Four different waxy mutations were identified, including two unknown alleles. This is a strong proof for parallel independent selection of waxy maize in the region. Starch granule morphology and size were similar among all LRs. Gelatinization properties differed only between waxy and wild-type LR, and all waxy LR were comparable to a commercial waxy hybrid. The fine structure of waxy amylopectin had fewer short chains compared with that in wild-type. So far, the differences observed in starch properties are likely associated exclusively with the waxy trait. Despite the strong selection for amylose-free starch, there was no evidence for additional region wide selection for other special starch properties in our collection. In conclusion, all analyses did not encourage further targeted research on allelic variation of other starch metabolism genes for future use in the food and feed industry.

Rhubarb includes approximately 60 species in the genus Rheum. It has been utilized for thousands of years for medicinal purposes, but only recently identified for its culinary use. In the mid 1700s, edible petioles were discovered on seedlings from rhubarb species. Hundreds of cultivars have since been identified for a wide range of uses from tarts to wine. Unfortunately, propagation by seed and irregular naming has resulted in a plethora of similarly named cultivars and multitude of phenotypes. Fifteen morphological characters were evaluated to differentiate rhubarb cultivars in the USDA, ARS Rheum collection in Palmer, Alaska. Two years of morphological data, focusing on horticultural characteristics indicated variation between the years. To improve cultivar resolution, the results suggest using 1 year's data instead of combining data from different years. The mean °Brix observed was 3.8, with a range from 2.2 to 6.1. Flesh colour and basal skin colour were poorly correlated (R2 = 0.462); overall skin colour was more red at the base than in the middle of the petiole. Rhubarb character categories, in particular petiole number and petiole base thickness, need to be modified to better anticipate the range of expected values, and thereby contribute improved reproducibility and reliability to separate cultivars based on morphological characters.

Pepper (Capsicum sp.) is important in human diets in many parts of the world and a major source of several antioxidants, including carotenoids, ascorbic acid, tocopherols and phenolics. More information on genetic diversity within Capsicum for antioxidant (AO) content and antioxidant activity (AOA) could contribute to improved human health. We evaluated 46 Capsicum accessions from AVRDC—the World Vegetable Center Capsicum core collection for content of nine AO (five carotenoids, ascorbic acid, tocopherols α and γ, and total phenolics) and two AOA assays for 2 years in south Taiwan. Ample genetic diversity exists within C. annuum to increase AO content. Based on dry weight values, non-pungent C. annuum entries as a group were significantly greater than pungent entries for contents of β-cryptoxanthin (36%), ascorbic acid (65%), total phenolics (36%) and α-tocopherol (11%). Group means of the brown-fruited entries exceeded the means of red-fruited entries for capsanthin (34%), zeaxanthin (37%), lutein (36%), β-cryptoxanthin (71%), β-carotene (82%), ascorbic acid (19%) and α-tocopherol (40%). Red-fruited C. annuum entries ‘Verdano Poblano’ and ‘Guajillo Ancho’ from Mexico ranked among the entries highest for all carotenoids, ascorbic acid and α-tocopherol. The inhibition of lipid peroxidation (ILP) assay could be adopted for AOA characterization or selection because of high variation among entries and consistent entry performance over years. ILP was positively correlated with phenolics (r=0.72**) and ascorbic acid (r=0.58**) contents. Significant positive correlations were detected between most carotenoids as well as a significant positive correlation between ascorbic acid and total phenolics contents (r=0.78**).

Ninety-eight high-yielding recombinant inbred lines (RILs) of WAB638-1/PRIMAVERA rice were evaluated with the parents under reproductive stage drought stress. The study aimed at characterizing flowering time response under drought stress and consequently classifying the genetic resources for efficient use in breeding programmes. Two field trials were conducted during the 2016 and 2017 dry seasons using the randomized complete block design with two replications. In 2016, 12 RILs were evaluated with the parents for the rooting attributes under drought stress and well-watered treatments. Analysis of variance revealed a significant (P < 0.05) variation among the genotypes for the traits evaluated. Drought stress reduced most of the characters in this study, including grain yield. However, the inbred lines exhibited considerable tolerance to drought stress as indicated by yield-related stress indexes, including stress susceptibility and drought tolerance indexes. Delayed flowering (FD) was recorded in 75 genotypes (including WAB638-1), while 25 genotypes (including PRIMAVERA) had no delay under drought stress compared to the control. The genotypes were grouped into flowering delay (D) genotypes (FD > 1 d) and no delay (N) genotypes (FD < 1 d), which significantly differ (P ⩽ 0.05) for numbers of days to 50% flowering. The flowering delay genotypes may be suitable for intermittent drought, while genotypes with little or no delay may be selected for terminal drought conditions. There was a significant correlation (r > 0.5) between the root length and number of days to 50% flowering. The study indicated that root parameters, such as the root length, may contribute to the drought adaptation mechanisms of the RILs.

Germplasm molecular and phenotypic characterization is instrumental to its utilization and to genetic variability incorporation into rice breeding programmes. The diversity within 192 Japanese rice accessions was analysed for 22 agro-morphological traits and 24 single sequence repeat markers. A total of 181 alleles were detected, 38 of which were exclusive. The number of alleles/marker ranged from 2 to 16, with an average of 7.54 alleles/locus and the He value ranged from 0.01 to 0.82, with an average of 0.46. The accessions showed diversity at molecular and phenotypic level and few showed also good agronomic performance. Tocher's method applied on a total-dissimilarity matrix was used to determine cluster formation of 13 diversity groups. Most of the accessions (81%) were clustered within a group, whereas eight accessions (Kyuushuu, Eika Ine, Ishiwari Mochi, Col/Fukui/1965, Ookuma Nishiki, Suzume Shirazu, Iwate Ryoon and Toga) did not cluster with other accessions.

Seedling emergence is the preliminary factor defining wheat adaptability and stability under salt stress. This study was led to assess the salinity tolerance amongst 226 synthetic hexaploid wheats (SHWs) evaluated against two check cultivars, the tolerant ‘S-24’ and the susceptible ‘PBW-343’ at three sodium chloride treatments (0, 100 and 200 mM). Highly significant and positive correlation was observed between germination % and germination index (r = 0.85), and between seedling height and weight (r = 0.85). All four traits across three treatments were transformed into the salt tolerance trait index and salt tolerance index (STI). STI had significant positive correlation with all four parameters indicating reliability of this index for ranking the tolerance levels. STI-based 20 best performing genotypes were known as being promising candidates for wheat breeding. Local tolerant check was amongst the top three tolerant accessions. Two SHWs, AUS30288 {Croc_1/Aegilops squarrosa (466)} and AUS34444 {Ceta/Ae. squarrosa (872)} outperformed S-24 with STI of 61.8 and 55.7, respectively. SHW with same durum parents were included in tolerant and susceptible categories indicating that tolerance is contributed by the Ae. squarrosa syn. tauschii parent of SHWs. In conclusion, this baseline study revealed that continuous variation in the seedling emergence traits under salt stress is a conduit towards implementing genome-wide association studies. Likewise, new diversity has implications in development of salt tolerance germplasm after genetic dissection permitting unique Ae. squarrosa accessional diversity validation to target SHW donors for breeding.

Genetic variation in wild soybean (Glycine soja Sieb. and Zucc.) is a valuable resource for crop improvement efforts. Soybean is believed to have originated from China, Korea, and Japan, but little is known about the diversity or evolution of Korean wild soybean. Therefore, in this study, we evaluated the genetic diversity and population structure of 733 G. soja accessions collected in Korea using 21 simple sequence repeat (SSR) markers. The SSR loci produced 539 alleles (25.7 per locus) with a mean genetic diversity of 0.882 in these accessions. Rare alleles, those with a frequency of less than 5%, represented 75% of the total number. This collection was divided into two populations based on the principal coordinate analysis. Accessions from population 1 were distributed throughout the country, whereas most of the accessions from population 2 were distributed on the western side of the Taebaek and Sobaek mountains. The Korean G. soja collection evaluated in this study should provide useful background information for allele mining approach and breeding programmes to introgress alleles into the cultivated soybean (G. max (L). Merr.) from wild soybean.

To identify appropriate genotypes for breeding varieties suitable for semi-arid areas, yield components, as well as morphophonological traits of 127 safflower genotypes, were assessed at the Seed and Plant Improvement Institute, Iran. Cluster analysis revealed four distinct groups. The largest group (third cluster) includes 48 genotypes with characteristics such as mostly yellow-coloured flowers, mostly thorny with few thistle genotypes, relatively early-flowering, relatively high number of heads and grains per plant, highest 1000-grain weight with the highest grain yields. In contrast, the smallest group (second cluster) contains 13 genotypes with characteristics such as yellow florets, semi-dwarf, thorny, moderately late-flowering, low number of heads per plant and number of grains per head, low 1000-grain weight, and low grain yield. Principal component analysis results showed that the three components accounted for 29.5, 15.9 and 11.1% of the total variation, respectively. Graphic representation of the biplots based on the first and second principal components showed that the experimental genotypes were classified into four groups. The results also indicated that stem diameter, the number of branches, heads per plant and grains per head had the highest correlation with the first component, and grain yield, flower colour, flowering initiation and plant height had the highest correlation with the second component. These traits can be considered as an appropriate index for the selection of elite safflower genotypes for breeding varieties suited to cultivation in semi-arid conditions.

Jatropha curcas L. is a potential bioenergy crop but has a lack of improved cultivars with high yields and oil content. Therefore, increasing our understanding of J. curcas germplasm is important for designing breeding strategies. This study was performed to investigate the genetic diversity and population structure of Indonesian J. curcas populations from six different islands. To construct a reference, we de novo assembled the scaffolds (N50 = 355.5 kb) using 182 Gb Illumina HiSeq sequencing data from Thai J. curcas variety Chai Nat. Genetic diversity analysis among 52 Indonesian J. curcas accessions was conducted based on yield traits and single nucleotide polymorphism (SNP) markers detected by mapping genotyping-by-sequencing reads from Indonesian population to Chai Nat scaffolds. Strong variation in yield traits was detected among accessions. Using J. integerrima as an outgroup, 13,916 SNPs were detected. Among J. curcas accessions, including accessions from other countries (Thailand, the Philippines and China), 856 SNPs were detected, but only 297 SNPs were detected among Indonesian J. curcas populations, representing low genetic diversity. Through phylogenetic and structural analysis, the populations were clustered into two major groups. Group one consists of populations from Bangka and Sulawesi in the northern part of Indonesia, which are located at a distance of 1572.59 km. Group two contains populations from islands in the southern part: Java, Lombok-Sumbawa, Flores and Timor. These results indicate that introduction of diverse J. curcas germplasms is necessary for the improvement of the genetic variation in the Indonesian collections.

Cotton fibre yield and quality are markedly influenced by drought and high-temperature stress. We examined the traits of the leaf stomata in 39 cotton genotypes subjected to exogenous phytohormone abscisic acid (ABA) signalling, electrolyte leakage under 40°C thermal stress, and relative GhHsfA, GhbZIP and GhHSP70 expression levels under two treatments. Stomatal density and area ranged from 66 to 182/mm2 and 663 to 1305 μm2, respectively. Under exogenous ABA signalling, the changes in stomatal aperture (ΔSAp) were in the range of 2.5–31.2%; ΔSAp and relative GhHsfA, GhbZIP and GhHSP70 expression levels were significantly correlated, respectively. Electrolyte leakage increased unequally among cotton genotypes after heat stress. The changes in electrolyte leakage (ΔEL) and relative GhHsfA, GhbZIP and GhHSP70 expression levels were very strongly correlated, respectively. Their relative expression levels could be used as references for the rapid identification of stress-tolerant cotton strains. Cluster analysis of the 39 cotton genotypes indicated that Xinluzao36, Shiyang1, shinong98-7 and Zhongmiansuo293 are heat- and drought-resistant. We integrated both analysis of physiological parameters and molecular methods to identify cotton varieties with the drought and heat tolerance, in order to provide a reference for the selection of materials and methods for the research and production of cotton.

A natural population of Vriesea friburgensis var. paludosa was genetically and demographically characterized. Four selected enzymatic systems were used. In total, 10 allozymic loci were analysed. The proportion of polymorphic loci was 70% and the number of alleles per locus was 2.3· The mean observed and expected heterozygosities were 0.234 and 0.226, respectively. An excess of heterozygotes was revealed by the value of the fixation index (—0.035). These indices showed the presence of high levels of genetic diversity in the population studied. A demographic survey revealed almost six times more vegetative than sexually reproductive plants. Reproductive plants were found only in clusters. The genetic diversity exhibited by this population, associated with vegetative as well as sexual propagation, resulting in clusters or colonies, would allow the employment of a sustainable management strategy for species conservation as well as its exploitation as an ornamental.

Asian palmyrah palm (Borassus flabellifer L.) is a multipurpose, economically important tree widely distributed in the Indian subcontinent and Southeast Asia. The multifaceted uses (~800 uses) of the tree as food, wood and medicine make it a viable industrial crop. However, the dioecious nature of the tree coupled with long juvenile phase (12–20 years to initiate flowering), long stature and considerable variations in the productivity of male and female trees necessitates the application of new biotechnological tools for crop improvement. This review provides comprehensive information on the extent of genetic diversity of the crop, problems associated with palmyrah cultivation along with its multifaceted application and research gaps to be addressed for the effective utilization and conservation of palmyrah palm genetic resources.

Hayachine-usuyukiso (Leontopodium hayachinense) is an alpine plant native to Mt Hayachine. This unique chrysanthemum is listed as an endangered plant by the Department of Conservation, Iwate Prefecture, and as a threatened plant by the Ministry of the Environment, Japan. We successfully cryopreserved the shoot tips from in vitro-grown L. hayachinense shoots using a vitrification protocol. Cold-hardened shoot tips were excised and pre-cultured on a solidified Murashige–Skoog medium containing 0.3 M sucrose for 1 d at 5°C. The shoot tips were then treated with loading solution for 20 min at 25°C, dehydrated in plant vitrification solution 2 for 120 min at 25°C and immersed in liquid nitrogen. The survival rate of the vitrified shoot tips was 63.3% after 30 d of regrowth. This protocol appears to be a promising technique for the cryopreservation of in vitro-grown shoots of this endangered plant.

Grapevine (Vitis vinifera L.) is a major worldwide crop of high economic importance, tightly interwoven with the traditions and the culture of many civilizations. The Greek vineyard is one of the oldest in the world composed of an ample number of highly diverse indigenous landraces. However, over the last decades the local cultivated grapevine germplasm has undergone a drastic reduction of diversity, due to the established market preferences for international varieties. In the current work a combined approach involving both, ampelographic traits and microsatellite markers has been undertaken, to study the genetic diversity within and among 96 grapevine genotypes belonging to 36 V. vinifera subsp. vinifera cultivars, predominantly representing autochthonous Greek landraces. Results revealed high genetic diversity for the Greek cultivars yielding a mean number of alleles per locus 14.69 and mean polymorphic information content 0.848. Hierarchical cluster analysis, employing both, ampelographic and microsatellite data, showed a clear distinction based on the origin of the germplasm; Anatolian versus Mediterranean. Principal component analysis, based on the most informative ampelographic traits, coupled to the results from genetic structure analysis further corroborated the proposal of germplasm differentiation on the basis of geographic origin. This information can be further utilized for the reconstitution of the Greek vineyard and can significantly contribute towards a rational conservation and utilization strategy for breeding or for direct cultivation of the Greek indigenous grapevine germplasm.

The genetic structure of marram grass populations at coastal and inland locations,200 m apart, was investigated at three sites by means of amplified fragmentlength polymorphism (AFLP) DNA markers. We expected a genetic differentiation betweencoastal and inland populations and more genetic variation in the coastal areas as a resultof different events of colonization by different plant materials. An assignment testshowed that the sampled Ammophila arenaria could be assigned to twogroups based on AFLP data. The spatial distribution of the two AFLP types of A.arenaria varied with sampling location. In two of the three locations, mainlyone type (1) was found in the newly formed dunes. This type did also occur furtherlandward, but the second type (2) was preferentially found in inland populations. Geneticdiversity was very low and of similar value in both coastal and inland populations. Foreach site, outlier loci with respect to FST value wereidentified, which may be indicative of different selection pressures in coastal comparedwith inland clusters. However, no identical outlier loci were found at all three sites.Possible explanations for the observed difference in distribution of type 1 and 2populations between coastal and inland sites are discussed.

In this study, agro-morphological and yield-related traits associated with drought tolerance in 80 barley genotypes belonging to 15 wild species together with the cultivated one, and their potential to improve adaptation to different levels of drought stress conditions (moisture environments) were studied. There was significant genetic variation among the genotypes and species for all of the measured traits, as well as differential responses of genotypes across environments. The results indicated high variation for grain yield (GY) under drought stress among the genotypes, and that some of the wild genotypes had consistently superior specific adaptation to the water stress conditions. The genotypes belong to wild barley species, especially Hordeum murinum and Hordeum marinum had lower GY but relatively higher yield stability under different environments. Traits such as number of seed per plant and hundred kernel weight were positively correlated with GY in all of the environments. High negative correlation between GY and days to ripening was observed only under intense drought environment, showing drought escape as a strategy of wild plants under highly stressed conditions. Grouping of the genotypes by principal component analysis completely separated cultivated barley and its progenitor (Hordeum vulgare ssp. spontaneum) from other wild genotypes; however, the other wild species were slightly separated from each other. In addition, the Iranian and foreign genotypes did not completely separate from each other. The identified wild barley genotypes with favourable characters and high drought tolerance could be used in genetic studies and barley improvement programmes especially for drought stress.

Seeds of wild and cultivated species of Luffa were studied to determine the variability in morphological (seed size, colour, seed-coat surface and 100-seed weight) and biochemical (oil and protein) characteristics. A total of 80 accessions of three cultivated species (71 accessions) and three wild species (9 accessions) of Luffa under the present investigation from diverse regions (12 states of five regions) of India showed variations in seed size, colour, seed-coat surface, 100-seed weight, and seed protein and oil contents both within the accessions of the same species and between different species. Significant variability in seed morphological traits was observed. Both seed oil and soluble seed protein contents were highest in some accessions of the cultivated species (25–27% oil and 8–10% protein, respectively, on a fresh seed weight basis). Using the 2D scatter plot diagram derived from the principal components analysis, the morphological and biochemical traits of the 80 Luffa accessions classified the wild species into one cluster (cluster I) and the cultivated species into a second major cluster (cluster II). The present investigation on the correlation between seed morphology and biochemical traits in the cultivated and wild species of Luffa can help in identifying the genotypes of Luffa species with valuable traits for further exploring the potential of this valuable crop as a source of edible oil, food and fodder in edible seed types or as a source of industrial oil/biodiesel in non-edible seed types. The protein-rich seed could be further explored to be utilized in the fortification of food products for value addition.

Chickpea is one of the most important nutritious grain legume crops in the world. There is limited information available on micro- and macro-nutrients in chickpea. Therefore, an effort was made to evaluate a set of 40 chickpea genotypes belonging to all the gene pools including cultivated (Cicer arietinum) as well as wild, Cicer reticulatum from the primary gene pool, Cicer echinospermum from the secondary gene pool and Cicer microphyllum from the tertiary gene pools. Concentration in the seed of the micro- (Zn, Fe, Cu and Mn) and macro-nutrients (Ca, Mg and K) was studied. Substantial variation was observed among different gene pools for the concentration of all the nutrients. The cultivated chickpea exhibited higher seed Cu, Mn, Mg and Ca than wilds indicating positive domestication effect, whereas wild crop relatives were found to have higher levels than cultivated chickpeas for seed Zn, Fe and K concentrations. While comparing desi-type chickpeas with Kabulis, it was revealed that desi types possessed more Zn, Cu, Ca and Mg than Kabulis but reverse was true for seed Fe, Mn and K. Among different desi types (desi brown, desi green and desi black), desi brown types were generally associated with higher mineral nutrient levels. The present study led to the identification of most promising genotypes for different seed micro- and macro-nutrients. These promising lines may serve as genetic resources useful in gene discovery programmes and for alleviating malnutrition or hidden hunger in the developing world.

Significant genetic diversity for sucrose and fibre percentages exists in the species that served as the foundation of present day sugarcane cultivars. However, information is lacking worldwide on the recent contributions of sugarcane crop wild relatives (mainly Saccharum, Erianthus and Miscanthus wild species) in developing new varieties. There is renewed interest in using those relatives for creating new varieties to use as a dedicated bioenergy crop with higher fibre. This study focuses on past data analysis of sugarcane breeding in Mauritius with the objective to assess the efficiency in exploiting sugarcane wild relatives since 1970s to date. Pedigree analyses helped retrace the parentages of elite inter-specific hybrids reaching the final stages of selection. The studies confirmed the high prevalence of a few ‘wonder canes’ (successful hybrids with wild canes produced in the beginning of last century) among the ancestors of Mauritian varieties. Among the wild relatives, eight Saccharum spontaneum, two S. robustum, and one Erianthus clones were involved in generating elite genotypes worth evaluating at the advanced variety trial stages. A few early generation hybrids were released in the past for industrial exploitation, the latest one being M 1002/02 in 2016, with sugar as the primary output. Recent studies on the biomass potential and fibre yield of inter-specific hybrids are giving promising results, which expands the horizon in the use of sugarcane wild relatives for the generation of novel type of sugarcane varieties for multiple end-uses.
