To identify appropriate genotypes for breeding varieties suitable for semi-arid areas, yield components, as well as morphophonological traits of 127 safflower genotypes, were assessed at the Seed and Plant Improvement Institute, Iran. Cluster analysis revealed four distinct groups. The largest group (third cluster) includes 48 genotypes with characteristics such as mostly yellow-coloured flowers, mostly thorny with few thistle genotypes, relatively early-flowering, relatively high number of heads and grains per plant, highest 1000-grain weight with the highest grain yields. In contrast, the smallest group (second cluster) contains 13 genotypes with characteristics such as yellow florets, semi-dwarf, thorny, moderately late-flowering, low number of heads per plant and number of grains per head, low 1000-grain weight, and low grain yield. Principal component analysis results showed that the three components accounted for 29.5, 15.9 and 11.1% of the total variation, respectively. Graphic representation of the biplots based on the first and second principal components showed that the experimental genotypes were classified into four groups. The results also indicated that stem diameter, the number of branches, heads per plant and grains per head had the highest correlation with the first component, and grain yield, flower colour, flowering initiation and plant height had the highest correlation with the second component. These traits can be considered as an appropriate index for the selection of elite safflower genotypes for breeding varieties suited to cultivation in semi-arid conditions.

Cotton fibre yield and quality are markedly influenced by drought and high-temperature stress. We examined the traits of the leaf stomata in 39 cotton genotypes subjected to exogenous phytohormone abscisic acid (ABA) signalling, electrolyte leakage under 40°C thermal stress, and relative GhHsfA, GhbZIP and GhHSP70 expression levels under two treatments. Stomatal density and area ranged from 66 to 182/mm2 and 663 to 1305 μm2, respectively. Under exogenous ABA signalling, the changes in stomatal aperture (ΔSAp) were in the range of 2.5–31.2%; ΔSAp and relative GhHsfA, GhbZIP and GhHSP70 expression levels were significantly correlated, respectively. Electrolyte leakage increased unequally among cotton genotypes after heat stress. The changes in electrolyte leakage (ΔEL) and relative GhHsfA, GhbZIP and GhHSP70 expression levels were very strongly correlated, respectively. Their relative expression levels could be used as references for the rapid identification of stress-tolerant cotton strains. Cluster analysis of the 39 cotton genotypes indicated that Xinluzao36, Shiyang1, shinong98-7 and Zhongmiansuo293 are heat- and drought-resistant. We integrated both analysis of physiological parameters and molecular methods to identify cotton varieties with the drought and heat tolerance, in order to provide a reference for the selection of materials and methods for the research and production of cotton.

Drought stress negatively affects the cotton production all over the world. The negative impact of drought varies for different species due to some morphological and root attributes that help some species to better stand under drought. But the extent of disturbance varies for different cotton species. To find out such variation, two cotton species (Gossypium hirsutum and Gossypium arboreum) were studied under normal and drought conditions for 2 years. Two genotypes for each species were included, i.e. PC-1 and COMILLA (G. arboreum) and IUB-13 and IUB-65 (G. hirsutum). The experiment was laid out under a completely randomized design following factorial arrangement. Genotype × treatment × year interaction of cotton genotypes was studied for different root, morphological, physiological and fibre-related traits. Traits such as above ground dry biomass, above ground fresh biomass, chlorophyll contents, leaf area, seed cotton yield, sympodial branches/plant, fibre strength and ginning out-turn were higher in G. hirsutum genotypes as compared to G. arboreum genotypes. However less reduction under drought in all above mentioned traits was recorded for G. arboreum, than G. hirsutum. Furthermore, root traits; primary root length, lateral root numbers, root fresh weight and root dry weight were enriched under drought condition in G. arboreum genotypes than in G. hirsutum genotypes, which is a clear manifestation of higher drought tolerance ability in G. arboreum genotypes transferrable to G. hirsutum genotypes through interspecific crossing or other means.

The current study was undertaken to identify the sources of tolerance to bruchid in cowpea, by screening a set of germplasm accessions as a source for natural resistance. A total of 103 diverse accessions of cowpea were evaluated for resistance to Callosobruchus maculatus Fab. under no-choice artificial infestation conditions. Significant differences among the cowpea accessions were observed for oviposition, adult emergence, exit holes and per cent seed weight loss (PSWL) caused by the bruchid infestation. The accessions showed variation in physical seed parameters viz., colour, shape, testa texture, length, width and seed hardness. Among the seed biochemical parameters studied, per cent sugar content ranged from 0.322 (IC330950) to 1.493 (IC249137), and per cent phenol content ranged from 0.0326 (EC390261) to 1.081 (EC528423). Correlation studies indicated that PSWL had significant positive correlation (r = 0.335) with exit holes, oviposition (r = 0.219), adult emergence (r = 0.534) and seed roundness (r = 0.219). Adult emergence had a significant negative correlation with seed hardness (r = −0.332). Correlation with biochemical parameters indicated that PSWL had a significant positive correlation (r = 0.231) with sugar content and a significant negative correlation with phenol content (r = −0.219). None of the accessions were found to be immune to bruchid infestation. However, out of studied accessions, EC528425 and EC528387 were identified as resistant based on PSWL and moderately resistant based on adult emergence. These resistance sources of cowpea germplasm can be used as potential donors for development of bruchid tolerant/resistant cultivars.

The genetic structure of marram grass populations at coastal and inland locations,200 m apart, was investigated at three sites by means of amplified fragmentlength polymorphism (AFLP) DNA markers. We expected a genetic differentiation betweencoastal and inland populations and more genetic variation in the coastal areas as a resultof different events of colonization by different plant materials. An assignment testshowed that the sampled Ammophila arenaria could be assigned to twogroups based on AFLP data. The spatial distribution of the two AFLP types of A.arenaria varied with sampling location. In two of the three locations, mainlyone type (1) was found in the newly formed dunes. This type did also occur furtherlandward, but the second type (2) was preferentially found in inland populations. Geneticdiversity was very low and of similar value in both coastal and inland populations. Foreach site, outlier loci with respect to FST value wereidentified, which may be indicative of different selection pressures in coastal comparedwith inland clusters. However, no identical outlier loci were found at all three sites.Possible explanations for the observed difference in distribution of type 1 and 2populations between coastal and inland sites are discussed.

Seeds of wild and cultivated species of Luffa were studied to determine the variability in morphological (seed size, colour, seed-coat surface and 100-seed weight) and biochemical (oil and protein) characteristics. A total of 80 accessions of three cultivated species (71 accessions) and three wild species (9 accessions) of Luffa under the present investigation from diverse regions (12 states of five regions) of India showed variations in seed size, colour, seed-coat surface, 100-seed weight, and seed protein and oil contents both within the accessions of the same species and between different species. Significant variability in seed morphological traits was observed. Both seed oil and soluble seed protein contents were highest in some accessions of the cultivated species (25–27% oil and 8–10% protein, respectively, on a fresh seed weight basis). Using the 2D scatter plot diagram derived from the principal components analysis, the morphological and biochemical traits of the 80 Luffa accessions classified the wild species into one cluster (cluster I) and the cultivated species into a second major cluster (cluster II). The present investigation on the correlation between seed morphology and biochemical traits in the cultivated and wild species of Luffa can help in identifying the genotypes of Luffa species with valuable traits for further exploring the potential of this valuable crop as a source of edible oil, food and fodder in edible seed types or as a source of industrial oil/biodiesel in non-edible seed types. The protein-rich seed could be further explored to be utilized in the fortification of food products for value addition.

Significant genetic diversity for sucrose and fibre percentages exists in the species that served as the foundation of present day sugarcane cultivars. However, information is lacking worldwide on the recent contributions of sugarcane crop wild relatives (mainly Saccharum, Erianthus and Miscanthus wild species) in developing new varieties. There is renewed interest in using those relatives for creating new varieties to use as a dedicated bioenergy crop with higher fibre. This study focuses on past data analysis of sugarcane breeding in Mauritius with the objective to assess the efficiency in exploiting sugarcane wild relatives since 1970s to date. Pedigree analyses helped retrace the parentages of elite inter-specific hybrids reaching the final stages of selection. The studies confirmed the high prevalence of a few ‘wonder canes’ (successful hybrids with wild canes produced in the beginning of last century) among the ancestors of Mauritian varieties. Among the wild relatives, eight Saccharum spontaneum, two S. robustum, and one Erianthus clones were involved in generating elite genotypes worth evaluating at the advanced variety trial stages. A few early generation hybrids were released in the past for industrial exploitation, the latest one being M 1002/02 in 2016, with sugar as the primary output. Recent studies on the biomass potential and fibre yield of inter-specific hybrids are giving promising results, which expands the horizon in the use of sugarcane wild relatives for the generation of novel type of sugarcane varieties for multiple end-uses.

In this study, agro-morphological and yield-related traits associated with drought tolerance in 80 barley genotypes belonging to 15 wild species together with the cultivated one, and their potential to improve adaptation to different levels of drought stress conditions (moisture environments) were studied. There was significant genetic variation among the genotypes and species for all of the measured traits, as well as differential responses of genotypes across environments. The results indicated high variation for grain yield (GY) under drought stress among the genotypes, and that some of the wild genotypes had consistently superior specific adaptation to the water stress conditions. The genotypes belong to wild barley species, especially Hordeum murinum and Hordeum marinum had lower GY but relatively higher yield stability under different environments. Traits such as number of seed per plant and hundred kernel weight were positively correlated with GY in all of the environments. High negative correlation between GY and days to ripening was observed only under intense drought environment, showing drought escape as a strategy of wild plants under highly stressed conditions. Grouping of the genotypes by principal component analysis completely separated cultivated barley and its progenitor (Hordeum vulgare ssp. spontaneum) from other wild genotypes; however, the other wild species were slightly separated from each other. In addition, the Iranian and foreign genotypes did not completely separate from each other. The identified wild barley genotypes with favourable characters and high drought tolerance could be used in genetic studies and barley improvement programmes especially for drought stress.

For more than four decades after the introduction of cv. Italia (Vitis vinifera L.) in Brazil, several somatic mutations in the genome of cv. Italia and its somatic mutants gave rise to phenotypes which generated at least five new cultivars of fine table grapes. Since no molecular marker proved to be effective in discriminating cv. Italia (V. vinifera L.) and its coloured mutants (Rubi, Benitaka, Brasil, Black Star), primers for the long terminal repeat (LTR) sequences were developed to analyse Inter Retrotransposon Amplified Polymorphism (IRAP) and Retrotransposon-Microsatellite Amplified Polymorphism (REMAP), and investigate how the coloured cultivars derived from clonal propagations of somatic mutations are genetically structured. Primers for LTR sequences of IRAP and REMAP markers were edited from grape sequence databases available at a GenBank. Twenty-four primers, denominated DKS001–DKS024, were edited. Three hundred and forty-nine DNA segments were amplified by individual DKS primers and DKS/ISSR (Inter Simple Sequence Repeats) primer combinations, at an average of 13.96 amplicons per primer pair. High genetic divergence between the five cultivars was inferred from polymorphism in retrotransposons IRAP and REMAP. The analysis of polymorphism of IRAP and REMAP retrotransposons was crucial to show that clonal propagation of somatic mutations may lead towards the formation of genetically divergent cultivars by the formation of genetically structured vineyards and show the mixture of genomes within each cultivar.

Sesame (Sesamum indicum L.) is one of the oldest and most nutritional oilseed crops, of which domestication history has been poorly understood. This study suggested that sesame has undergone domestication bottleneck during its use for a long time. In this investigation, the molecular analysis included 4.4 Mbp of the genomic DNA of sesame comprising stearoyl acyl desaturase (sad), fatty acid desaturase 2 (fad2) and omega 3 fatty acid desaturase (o3fad) genes in 99 accessions of four populations of sesame germplasm namely: wild species, landraces, improved cultivars and introgressed lines. Results indicated that the improved cultivars and landraces lost 46.6 and 36.7% of nucleotide diversity, respectively, which indicate that the genetic diversity of the crop had been eroded due to selection after domestication. However, there was no significant reduction in genetic diversity of improved cultivars compared with landraces, indicating that unique improved cultivars generated through crosses were of less frequency in this population. Moreover, introgressed lines retained only 17.77% (π) and 4.57% (θ) of landrace diversity. To evaluate the impact of selection across fatty acid biosynthetic pathway, individual nucleotide diversity at three major genes involved in the pathway was surveyed. The analysis between wild and improved cultivars supported positive selection in fad2 and o3fad loci. Though locus-to-locus sequence variation was observed, positive results with two most important loci supported selection after domestication. Reduced diversity in these critical quality governing genes in improved cultivars suggested that future sesame cultivation would benefit from the incorporation of alleles from sesame's wild relatives.

Chickpea is one of the most important nutritious grain legume crops in the world. There is limited information available on micro- and macro-nutrients in chickpea. Therefore, an effort was made to evaluate a set of 40 chickpea genotypes belonging to all the gene pools including cultivated (Cicer arietinum) as well as wild, Cicer reticulatum from the primary gene pool, Cicer echinospermum from the secondary gene pool and Cicer microphyllum from the tertiary gene pools. Concentration in the seed of the micro- (Zn, Fe, Cu and Mn) and macro-nutrients (Ca, Mg and K) was studied. Substantial variation was observed among different gene pools for the concentration of all the nutrients. The cultivated chickpea exhibited higher seed Cu, Mn, Mg and Ca than wilds indicating positive domestication effect, whereas wild crop relatives were found to have higher levels than cultivated chickpeas for seed Zn, Fe and K concentrations. While comparing desi-type chickpeas with Kabulis, it was revealed that desi types possessed more Zn, Cu, Ca and Mg than Kabulis but reverse was true for seed Fe, Mn and K. Among different desi types (desi brown, desi green and desi black), desi brown types were generally associated with higher mineral nutrient levels. The present study led to the identification of most promising genotypes for different seed micro- and macro-nutrients. These promising lines may serve as genetic resources useful in gene discovery programmes and for alleviating malnutrition or hidden hunger in the developing world.

Hayachine-usuyukiso (Leontopodium hayachinense) is an alpine plant native to Mt Hayachine. This unique chrysanthemum is listed as an endangered plant by the Department of Conservation, Iwate Prefecture, and as a threatened plant by the Ministry of the Environment, Japan. We successfully cryopreserved the shoot tips from in vitro-grown L. hayachinense shoots using a vitrification protocol. Cold-hardened shoot tips were excised and pre-cultured on a solidified Murashige–Skoog medium containing 0.3 M sucrose for 1 d at 5°C. The shoot tips were then treated with loading solution for 20 min at 25°C, dehydrated in plant vitrification solution 2 for 120 min at 25°C and immersed in liquid nitrogen. The survival rate of the vitrified shoot tips was 63.3% after 30 d of regrowth. This protocol appears to be a promising technique for the cryopreservation of in vitro-grown shoots of this endangered plant.

Phenotyping of genetic resources remains the bottleneck in the characterization of genetic resources, since the advent of modern next-generation sequencing technologies has made genotyping much more cost- and time-effective. This article reports on the phenotyping of agriculturally important traits in perennial ryegrass (Lolium perenne). In the present study, water-soluble carbohydrate (WSC), crude protein and dry matter contents were recorded for 1320 individuals, pooled into 132 samples from 33 perennial ryegrass ecotypes and cultivars at five different harvest time points across the 2004 growing season. While, in general, the cultivars had higher WSC contents than the ecotypes, individual ecotypes did show potential to be used in breeding programmes, as they had higher values than all other accessions at particular cutting time points. In correlation analyses, positive relationships were observed between dry matter and glucose contents both early and late in the growing season. Principal components analysis allowed the split either between cultivars and ecotypes or between tetraploid cultivars and the rest of the accessions at four of the five cutting time points. In the analysis of variance, cutting time was the most significant factor influencing the variation in traits.

In this paper, we describe variations among autochthonous olive cultivars from five different areas in Basilicata (Southern Italy) classified according to 33 chemical oil components and morphological traits. While all examined descriptors show no significant differences among cultivars, means and coefficients of variations have been highlighted. Principal component analysis has then been used to reduce the number of descriptors. Cultivars have been classified by cluster analysis into three groups. Following a discussion of cultivar group similarities, results suggest that an ‘a priori’ classification of cultivars according to growing area does not strictly correspond to phenotypic grouping. From the spatial distribution of cultivars, however, it has been possible to identify ‘superior’ genotypes in terms of olive oil composition.

In drought-affected maize production zones with short growing periods, the development and use of early maturing drought-tolerant cultivars can stabilize maize production. We evaluated 10 improved and 25 farmers' early maturing maize varieties under moisture deficit and well-watered conditions for 2 years to identify suitable genetic materials for breeding drought-tolerant cultivars. The varieties exhibited significant differences in grain yield and other traits under both moisture deficit and well-watered conditions. Changes in the rank order of the varieties for grain yield was not significant across the different levels of moisture supply in this study. Grain yield was significantly correlated with days to anthesis, days to silking, plant height, ear height, ear number and anthesis–silking interval (ASI) under the two irrigation treatments and with leaf death scores under moisture deficit, suggesting that the common traits were beneficial in maximizing grain yield under both sufficient water supply and moisture deficit. Grain yield and the traits significantly correlated with it differentiated the early maturing maize varieties into two distinct groups under well-watered condition and moisture deficit. The improved varieties were superior to the farmers' varieties in grain yield and other traits under moisture deficit, possibly due to selection of their progenitors for improved performance in multiple locations. We found some farmers' and improved varieties with similar yield potential and flowering time under well-watered conditions but with marked differences in grain yield and other traits under moisture deficit. Use of such promising landraces that would also be invaluable sources of desirable farmers-preferred end-use quality traits in combination with promising improved varieties as breeding materials could enhance the genetic grain from selection for drought tolerance in early maize.

Monitoring and protecting germplasm in genebanks using in situ collections while preserving its original genetic integrity is a priority of germplasm curation. Many germplasm accessions need to be regenerated due to their demand and/or seed condition. The regeneration of wild Helianthus (sunflower) species poses several challenges due to the diversity of 53 wild species. Fatty acid composition of sunflower oil is an important quality factor for the crop. Since oil quality is environmentally influenced, and evaluation of this trait is usually performed on oil from achenes from the original accessions of wild sunflower species, we conducted a study on 72 accessions of eight annual and four perennial taxa of wild sunflower species to compare the oil quality of the original accessions and those regenerated for genebank maintenance. The results showed that the fatty acid composition profiles of achenes from the original and regenerated accessions are not the same. It seems that selection for specific fatty acids in several species will require the analysis of both populations to identify germplasm accessions for use in breeding programmes. It should be borne in mind that accessions of wild species are open-pollinated segregating populations, so one would expect some variability in each succeeding generation. While there may be differences between the original and regenerated accessions, the interrelationships of fatty acids are generally similar in wild and cultivated sunflower species, so there should be no detrimental effects on oil quality when using the wild species for other traits. As more regenerated accessions become available, a more precise relationship between the original and regenerated accessions should emerge.

Nineteen kenaf genotypes from Cuba, Taiwan, the USA, El Salvador, Guatemala, Russia, Spain and Indonesia, and three wild types collected in South Africa were analysed for genetic diversity using AFLP analysis. All could be uniquely distinguished from one another, but only a low level of genetic diversity was present. The most distinct accession, Guatemala 4, was 85% similar to all other accessions. The accessions clustered more or less according to known pedigree and/or origin. Two of the three wild types (Hibiscus cannabinus c and H. cannabinus a) clustered separately from the commercial and Russian accessions. One of the wild types, H. cannabinus b clustered with some of the commercial accessions. Commercial accessions in the first subgroup all originated from central and North America, and surrounding islands (Cuba and El Salvador). The Russian accessions are all grouped together. The second subgroup was the only group that contained accessions from different geographical origins.

Non-destructive high-throughput phenotyping based on phenomics is an emerging technology for assessing the genetic diversity of various traits and screening in breeding programmes. In this study, non-destructive measurements of leaf temperature and chlorophyll fluorescence were conducted to investigate the physiological responses of soybean (Glycine max) to salt stress so as to set up a non-destructive screening method. Two-week-old seedlings of soybean in the V2 stage were treated with 0, 12.5, 25, 50 and 100 mM NaCl to induce salt stress. Three parameters, photosynthesis rate, stomatal conductance and chlorophyll fluorescence, decreased significantly, while soybean leaf temperature increased by exhibiting a positive correlation with NaCl concentration (P< 0.001). Soybean leaf temperature increased significantly at 50 mM NaCl when compared with the untreated control, although no visual symptom was observed. We selected leaf temperature as a major physiological parameter of salt stress as its measurement is much easier, faster and cheaper than that of other physiological parameters. Therefore, leaf temperature can be used for evaluating the responses to salt stress in soybean as a non-destructive and phenomic parameter. The results of this study suggest that non-destructive parameters such as chlorophyll fluorescence and leaf temperature are useful tools for assessing the genetic diversity of soybean with regard to salt stress tolerance and to screen salt stress-tolerant soybean for breeding.

The development of crop varieties with enhanced economic value requires considerable resources, time and intellectual input. The individual/agencies/institutions other than the originator/developers can easily multiply and commercialize pure-line varieties which are the only cultivar option in predominantly self-pollinated crops like dolichos bean and derive economic benefits from them. Seeking legal rights on crop varieties is the best and the only way to protect them from unlawful claim of their origin/development and deriving economic benefits from them. Seeking legal protection from unauthorized commercialization of crop varieties under Indian PPV & FR Act requires establishment of their distinctness (D), uniformity (U) and stability (S). DUS of crop varieties are established predominantly using morphological descriptors. Five genotypes which include two released varieties (HA 3 and HA 4) (referred to as reference varieties) and one advanced breeding line, HA 11–3 and two elite germplasm accessions, Kadalavare and GL 66 (referred to as candidate varieties) were characterized for 25 morphological descriptors to establish their DUS. All the candidate varieties differed from reference varieties for at least one characterization descriptors, indicating their distinctness. The absence of off-types indicated the uniformity of reference and candidate varieties and hence was deemed stable. Thus, the study demonstrated the utility of morphological descriptors for establishing DUS dolichos bean genotypes.

Breeding for resistance to biotic stress and higher yield is a continuous process. Thus, the identification of desirable parents with good combining ability and nature of gene action for the target trait is of utmost importance. Hence, in this present investigation, 10 lines and three testers of Okra were crossed in line × tester mating design to generate 30 testcross progenies and their evaluation along with parents and check in a randomized complete block design with three replications. To depict the true picture of genetic variation among the parental genotypes, molecular diversity analysis was also carried out using genomic-simple sequence repeats before crossing to ascertain that sufficient variability is present among the parents. The molecular analysis grouped the parental genotypes into four clusters (I–IV). The analysis of variance revealed that all the treatments were significant for most of the traits. The combining ability analysis suggested Pusa A-4 as the best general combiner for earliness, Pusa Bhindi-5 for high yield, and DOV-92 for fruit length, plant height, yield per plant, and coefficient of infection for Yellow Vein Mosaic Virus Disease resistance. Similarly, the specific combining ability analysis suggested that the cross combinations DOV-92 × Pusa Bhindi-5 followed by DOV-92 × Pusa A-4 and DOV-92 × Pusa Sawani exhibit high economic heterosis for yield per plant as well as for disease resistance. Finally, estimation of the degree of dominance and predictability ratio was also worked out which indicated the prevalence of non-additive gene action for most of the traits pointing towards sufficient scope for heterosis breeding in Okra.
