1. Adult rats were trained to consume the entire day's ration in 2 h. They showed marked difference in food intake when a diet lacking in protein was given. Two broad stages were observed, the first phase did not show any drop in calorie intake whereas the second showed distinct severe calorie deficiency.

2. Definite changes were noted in the metabolic profiles of the protein-deficient group even though no evidence for a fall in calorie intake (stage 1) was provided by the measurements of body and organ weights, urinary end-products, liver constituents and liver enzymes.

3. The levels of urinary end-products and liver DNA and RNA decreased in the proteindeficient and pair-fed control groups compared with the control group.

4. The liver glycogen content remained unchanged in the protein-deficient groups whereas the lipid content and the ratio of vitamin A alcohol to ester increased significantly.

5. The activity of xanthine oxidase in the liver was reduced by 75% in the pair-fed control group and by 95% in the protein-deficient group when compared with the control group. However, the activity of succinic dehydrogenase varied, depending on the unit of activity used to express it.

The sympathetic nervous system is involved in the control of energy metabolism and expenditure. Diet-induced thermogenesis is mediated partly by the ß-adrenergic component of this system. The aim of the present study was to investigate the role of genetic variation in the ß2-adrenoceptor in diet-induced thermogenesis. Data from twenty-four subjects (fourteen men and ten women; BMI 26·7(sem 0·8) kg/m2; age 45·2(sem1·4) years) with different polymorphisms of the ß2-adrenoceptor at codon 16 (Gly16Gly, Gly16Arg or Arg16Arg) were recruited for this study. Subjects were given a high-carbohydrate liquid meal, and the energy expenditure, respiratory exchange ratio, and plasma concentrations of NEFA, glycerol, glucose, insulin and catecholamines were measured before and over 4 h after the meal. The AUC of energy expenditure (diet-induced thermogenesis) was not significantly different between polymorphism groups, nor was the response of any of the other measured variables to the meal. In a multiple regression model, the only variable that explained a significant proportion (32 %) of the variation in diet-induced thermogenesis was the increase in plasma adrenaline in response to the meal (P<0·05). The ß2-adrenoceptor codon16 polymorphisms did not contribute significantly. In conclusion, an independent contribution of the codon 16 polymorphism of the ß2-adrenoceptor gene to the variation in thermogenic response to a high-carbohydrate meal could not be demonstrated. The interindividual variation in thermogenic response to the meal was correlated with variations in the plasma adrenaline response to the meal.

1. The vitamin A content of human liver tissue was determined in 363 autopsy samples. The sample comprised a total of 181 subjects dying after accidents and 182 dying from coronary heart disease among Singapore ethnic groups of both sexes.

2. The medium vitamin A reserve was 146 mg/kg in accident victims and 141 mg/kg in those who had died of coronary heart disease. Of all the samples 16% contained less than 40 mg/kg, 45% had 100–300 mg/kg, while 9% contained more than 500 mg/kg liver.

3. Among the accident victims, Indians had the lowest median liver vitamin A reserve (118 mg/kg) compared with that in other ethnic groups (137 mg/kg in Chinese, 191 mg/kg in Malays, 155 mg/kg in Caucasians).

4. The ethnic distribution of vitamin A reserve in coronary deaths was similar to that in accident victims.

5. There was no significant difference between the sexes in hepatic vitamin A reserve.

6. The distribution of vitamin A reserve in all the groups was skewed to the right.

1. To elucidate the causal relation between leucine and the lowering of hepatic NAD content of rats fed on a leucine-excessive diet (Yamada et al. 1979), the effect of leucine on intestinal absorption of tryptophan was investigated.

2. Co-administration of [3H]tryptophan and leucine, with leucine at ten times the level of tryptophan, delayed absorption of L-[side chain 2,3-3H]tryptophan from the digestive tract and incorporation of [3H]tryptophan into portal blood, the liver and a protein fraction of the liver. After 120 min, more than 95% of tryptophan was absorbed whether [3H]tryptophan was administered with or without leucine.

3. Co-administration of a mixture of ten essential amino acids, in proportions simulating casein, with [3H]tryptophan markedly delayed absorption of tryptophan from the digestive tract. The addition of supplementary leucine to the amino acid mixture, however, caused no further delay.

4. In rats prefed a leucine-excessive diet for 1 week [3H]tryptophan was absorbed at the same rate as in rats fed on a control diet.

5. The results indicate that competition between tryptophan and leucine for intestinal absorption did not cause lowering of hepatic NAD.

1. Three experiments were conducted to examine the effect of monensin on growth performance, feed utilization and volatile fatty acids (VFA) in the forestomach and caecum of hamsters.

2. In Expt 1, monensin was fed at levels of 0, 5, 10 and 20 mg/kg to the growing male and female hamsters given a commercial diet (major component: lucerne (Medicago sativa) meal). In Expt 2, monensin was fed at levels of 0, 5, 15, 45 and 135 mg/kg to the growing male hamsters given a semi-purified diet containing 10 g urea/kg (main components: maize starch, sucrose, casein and cellulose). In Expt 3, monensin was fed at levels of 0 and 40 mg/kg to the growing male hamsters given the commercial diet containing lucerne meal or a semi-purified diet.

3. In Expt 1, monensin improved feed conversion efficiency and growth performances in the young growing hamsters, but monensin did not affect the hamsters at a later growing stage.

4. In response to monensin the proportion of acetic acid increased and that of propionic acid decreased in the forestomach, whereas the proportion of acetic acid decreased and that of propionic acid increased in the caecum in Expt 2. The hamsters given 135 mg monensin/kg ate less, developed diarrhoea and died.

5. The apparent digestibility of crude protein (nitrogen x 6.25) was improved by monensin but those of dry matter and neutral-detergent fibre (NDF) were decreased in hamsters given the semi-purified diet in Expt 3. Monensin did not appear to have a significant effect on the apparent digestibility of the diet containing lucerne meal.

6. The responses to monensin in hamsters are compared with those in ruminants.

Intake of marine-based n-3 fatty acids (EPA, docosapentaenoic acid and DHA) is recommended to prevent CHD. Stearidonic acid (SDA), a plant-based n-3 fatty acid, is a precursor of EPA and may be more readily converted to EPA than α-linolenic acid (ALA). While transgenic soyabeans might supply SDA at low cost, it is unclear whether SDA is associated with CHD risk. Furthermore, associations of other n-3 fatty acids with CHD risk remain inconsistent. The present ancillary study examined the association of erythrocyte SDA as well as other n-3 fatty acids with the risk of CHD. In a prospective nested case–control study of the Physicians' Health Study, we randomly selected 1000 pairs of incident CHD with matching controls. Erythrocyte fatty acids were measured using GC. We used conditional logistic regression to estimate relative risks. Mean age was 68·7 (sd 8·7) years. In a multivariable model controlling for matching factors and established CHD risk factors, OR for CHD for each standard deviation increase of log-SDA was 1·03 (95 % CI 0·90, 1·18). Corresponding values for log-ALA and log-marine n-3 fatty acids were 1·04 (95 % CI 0·94, 1·16) and 0·97 (95 % CI 0·88, 1·07), respectively. In conclusion, the present data did not show an association among erythrocyte SDA, ALA or marine n-3 fatty acids and the risk of CHD in male physicians.

The objective of the present study was to assess the effects of Enterococcus sp. strain TN-3 isolated from deep seawater on inhibition of eosinophil accumulation, IgE production and active cutaneous anaphylaxis (ACA). We investigated the effects of viable and non-viable TN-3 on allergen-induced peritoneal eosinophil accumulation in mice. Viable (5·4 × 1010 colony-forming units per 60 mg) or non-viable TN-3 (60 mg) was orally administered to BALB/c mice that had been sensitised with the cedar pollen (Cryptomeria japonica) allergen. Oral administration of non-viable TN-3 was effective in suppressing eosinophil accumulation while viable TN-3 was ineffective. We also examined the dose–response relationship for non-viable TN-3 in regard to eosinophil accumulation, IgE production and ACA in allergen-primed mice. Non-viable TN-3 was orally administered at doses of 15 mg (low dose), 30 mg (medium dose) and 60 mg (high dose) to BALB/c mice that had been sensitised with cedar pollen allergen. The anti-allergic effects expressed as inhibition of eosinophil accumulation, IgE production and ACA were found at the low and high doses, but not at the medium dose. These results suggest that non-viable TN-3 exhibited anti-allergic effects at doses of 15 and 60 mg.

Lactation is a physiologically demanding period in mink production, during which kit and dam losses may occur. Ambient temperature and quantitative water supply are thought to affect animal performance and well-being, but conclusive data in the literature are sparse. Therefore, effects of ambient temperature (Ta; low, about 5° medium, about 15° high, average 20-25°) and water supply (ad libitum (N), or 10 % extra supplementation in the food (E)) were investigated regarding effects on quantitative water intake and excretion, urine osmolality and solute excretion, and urinary cortisol and catecholamines as stress indicators in an experiment with twelve lactating mink with litters of three to seven kits in three consecutive periods, lasting 3, 3 and 2 d respectively. Kit ages ranged from 15 to 20 d at the end of the experiment. Water requirement for milk production (factorial calculations) and water available for evaporation (balance component) were estimated. Period, and hence mainly Ta, had a significant influence on intake of metabolizable energy, quantitative water intake and excretion, but there was less effect of water supply. The total water intake and excretion were very high in relation to the weight of the animals as an effect of lactation. Water intake and excretion, and urinary Na excretion, seemed to be less accurately regulated compared with corresponding functions in non-lactating animals. Rectal temperature increased with increasing Ta, possibly as a means of decreasing evaporative water loss. Water output in milk was estimated to increase from 118 g/d at low Ta to 134 g/d at high Ta. The amounts of water available for evaporation were estimated to be 42, 58 and 69 g/kg0·75 at low, medium and high Ta. Cortisol data did not indicate that the animals experienced negative stress. It was concluded that prolonged periods of high Ta may be hazardous for lactating mink because of decreased intake of metabolizable energy resulting in energy deficit and excessive mobilization of body reserves simultaneously as the requirement for intake of water increases considerably.

Advice on replacing unhealthy foods with healthier alternatives within the same food category may be more acceptable and might ease the transition towards a healthy diet. Here, we studied the potential impact of substitutions within the pizza category on the risk of type 2 diabetes (T2D). The study sample consisted of 2510 adults from the INCA2 French national survey. Based on their nutritional characteristics, the 353 pizzas marketed in France were grouped into 100 clusters that were used to run various scenarios of pizza substitutions, which were either isoenergetic (IE) or non-isoenergetic (NIE). We then used a model structurally similar to the Preventable Risk Integrated ModEl to assess the expected rate of change in risk of T2D. Pizzas characterised by a low energy, high vegetable content and whole grain dough were associated with a greater reduction in the risk of T2D. The rates of change in risk of T2D were markedly stronger in men and for NIE substitutions. When the rates of change were estimated in the subsample of pizza consumers, replacing the observed pizzas with the best pizza resulted in a T2D risk reduction of −6·7 % (–8·4 %; −4·9 %, IE) and −8·9 % (–11·2 %; −6·3 %, NIE), assuming that this is their usual diets. The greatest risk reduction induced by an IE substitution of the observed pizza with a mixed dish was similar to that observed with the best pizzas. Overall, this modelling study suggests that healthy swaps within a category can effectively supplement broader dietary changes towards a healthier diet.

This study evaluated the association between inflammatory diets as measured by the Dietary Inflammatory index (DII), inflammation biomarkers and the development of preeclampsia among the Chinese population. We followed the reporting guidelines of the Strengthening the Reporting of Observational Studies in Epidemiology statement for observational studies. A total of 466 preeclampsia cases aged over 18 years were recruited between March 2016 and June 2019, and 466 healthy controls were 1:1 ratio matched by age (±3 years), week of gestation (±1 week) and gestational diabetes mellitus. The energy-adjusted DII (E-DII) was computed based on dietary intake assessed using a seventy-nine item semiquantitative FFQ. Inflammatory biomarkers were analysed by ELISA kits. The mean E-DII scores were −0·65 ± 1·58 for cases and −1·19 ± 1·47 for controls (P value < 0·001). E-DII scores positively correlated with interferon-γ (r s = 0·194, P value = 0·001) and IL-4 (r s = 0·135, P value = 0·021). After multivariable adjustment, E-DII scores were positively related to preeclampsia risk (P trend < 0·001). The highest tertile of E-DII was 2·18 times the lowest tertiles (95 % CI = 1·52, 3·13). The odds of preeclampsia increased by 30 % (95 % CI = 18 %, 43 %, P value < 0·001) for each E-DII score increase. The preeclampsia risk was positively associated with IL-2 (OR = 1·07, 95 % CI = 1·03, 1·11), IL-4 (OR = 1·26, 95 % CI = 1·03, 1·54) and transforming growth factor beta (TGF-β) (OR = 1·17, 95 % CI = 1·06, 1·29). Therefore, proinflammatory diets, corresponding to higher IL-2, IL-4 and TGF-β levels, were associated with increased preeclampsia risk.

Weanling rats were offered food ad libitum, or fasted for 18 h, or fasted and refed for times ranging from 5 to 30 min. Five protein kinase C (PKC) isoforms (α, ε, ζ, and μ) were detected in the hindlimb muscles by Western immunoblotting. PKC forms ε and were abundant in plantaris, but not in soleus muscle, and no difference in localization was detected between fed rats and those fasted for 18 h. PKC forms α and μ were affected by fasting and refeeding. PKC-μ was found only in the cytosolic fraction of the plantaris muscle of the fasted animal, but in the fully-fed animals it was also associated with the membrane fraction. The pattern of localization observed in the fully-fed state was restored in the fasted rats by 20 min refeeding. In contrast, PKC-α was not detected in the cytosolic fraction of the plantaris in fasted animals but rapidly reappeared there on refeeding, being restored to 20 % and 80 % of the fed value within 5 and 30 min of refeeding respectively. The timing of these changes was correlated with the increase in serum insulin concentration, which was significantly elevated above the fasted value by 5 min and at subsequent times. These data suggest a possible role for PKC isoforms α and μ in the metabolic changes that occur in skeletal muscle on transition between the fasted and the fed state.

1. Chicks and quail (Coturnix japonica) were used in the measurement of net protein utilization and true digestibility of nitrogen of isolated soya-bean protein and a mixture of casein and gelatin in zinc-deficient diets.

2. The net protein utilization values for both protein sources were increased when the diets were supplemented with Zn or with the disodium salt of ethylenediaminetetra-acetic acid, as was the true digestibility of nitrogen of the isolated soya-bean protein.

The effect of oven-drying on the gross energy content of rat carcasses was investigated. No losses of carcass energy could be ascribed to oven-drying per se.

In work aimed at developing methodologies for validation of estimates of the dietary intake of free-living individuals, Bingham and colleagues have examined the use of urinary nitrogen (UN) excretion as an index of protein intake (Bingham & Cummings, 1985; Bingham, 1994; Bingham et al. 1995). The basis of this approach is that in subjects in N equilibrium, N intake is assumed equal to N excretion. Thus, if N excretion is measured it should indicate N and dietary protein intakes. In practice, since most N excretion occurs via the urine, which is relatively simple to collect, Bingham and colleagues examined the relationship between 24 h UN and N intake (DN). They argued that in subjects on typical UK diets UN should bear a fixed relationship to DN, and measured it carefully in a group of subjects. They showed that in a group of eight individuals in which intake and N excretion were measured meticulously, UN was 81% DN (SD 2, range 78–83%). They argued that measurement of this ratio could be used to assess the validity of food intake measurement and concluded ‘In healthy individuals eating normal western diets, 24 h urine N from an 8 day collection verified for its completeness by the PABA check method, should establish urine N to within 81(SD 5)% of the habitual dietary intake, range 70–90%. If the dietary assessment from 18 days of records or 24 h recalls or the diet history falls within these limits, it can be stated that there is no evidence of interference with normal dietary habits, or of reporting errors….’ (Bingham & Cummings, 1985).

1. Young rats were given, for 9 weeks, vitamin E-deficient diets containing either 20% or 10% casein, with and without a dietary supplement of 350 ppm D-α-tocopheryl acetate. For the next 5 weeks the casein content of the low-protein diets was decreased to 7%.

2. The low-protein diets induced severe growth depression.

3. The dialuric acid-induced haemolysis test showed that the rats given the 20% casein vitamin E-deficient diet were depleted of vitamin E, but that the rate of depletion on the lowcasein diet was slower.

4. Haemoglobin levels were slightly decreased by the 10% casein diets after 9 weeks, but this difference was not found after 14 weeks, comparing 20% and 7% casein. Dietary vitamin E had no effect on haemoglobin levels or erythrocyte counts.
