We assessed inheritance of resistance to sugarcane brown rust (Puccinia melanocephala) in selfing F1 populations of wild sugarcane germplasm Erianthus rockii ‘Yundian 95-19’ and E. rockii ‘Yundian 95-20’. We tested parent and selfing F1 individuals for the brown rust resistance gene, Bru1, that has been shown to confer resistance to brown rust in sugarcane. The Bru1 gene was not detected in E. rockii ‘Yundian 95-19’, E. rockii ‘Yundian 95-20’ or their selfing F1 individuals, and we found there was segregation of resistance in the two selfing F1 populations (segregation ratio: 3:1). The results confirmed resistance in E. rockii ‘Yundian 95-19’ and E. rockii ‘Yundian 95-20’ to sugarcane brown rust is controlled by a novel, single dominant gene.

As a member of the tribe Brassiceae, Eruca sativa, although a minor crop worldwide, is considered a valuable genetic resource for cabbage, rapeseed and other Brassica crops. Self-incompatibility (SI) in Brassica has been extensively studied, but information on SI in E. sativa is limited. Of six chemicals used to treat the stigmas to overcome SI in five E. sativa lines, gibberellin was the most effective. As gibberellin is well known for its ability to break dormancy and to promote cell elongation, its effectiveness may help to understand the mechanisms of SI. Urea and ammonium sulphate were also effective. These two chemicals are known to affect protein stability, which may help explain their effects on SI. Although table salt has been reported as being effective in overcoming SI in B. rapa, B. oleracea and B. napu, it was not effective in E. sativa. Sucrose and alcohol also had negligible effect. There was significant variation among the genotypes in SI intensity and response to chemicals, but the genotype–chemical interaction was not significant. The data presented in this paper add to our understanding of SI in E. sativa and may lead to a better use of this genetic resource.

The genetics of the root system is still not dissected for wheat and lack of knowledge prohibits the use of marker-assisted selection in breeding. To understand the genetic mechanism of root development, Synthetic W7984 × Opata M85 doubled-haploid (SynOpDH) mapping population was evaluated for root and shoot characteristics in PVC tubes until maturity. Two major quantitative trait loci (QTLs) for total root biomass were detected on homoeologous chromosomes 2A and 2D with logarithm of the odds scores between 6.25–10.9 and 11.8–20.86, and total phenotypic effects between 12.7–17.7 and 26.6–40.04% in 2013 and 2014, respectively. There was a strong correlation between days to anthesis and root and shoot biomass accumulation (0.50–0.81). The QTL for biomass traits on chromosome 2D co-locates with QTL for days to anthesis. The effect of extended vegetative growth, caused by photoperiod sensitivity (Ppd) genes, on biomass accumulation was always hypothesized, this is the first study to genetically support this theory.

Cotton is one of the most important crops among natural fibres. Fibre quality determines the spinning ability, which is negatively correlated with yield and yield-contributing traits. Limited efforts have been made to improve fibre quality and yield in diploid cotton. Therefore, screening a large panel of germplasm lines can help identify genotypes with better fibre quality and yield. We evaluated 712 desi cotton genotypes for fibre quality traits. The genotypes showed a significant difference for all the traits, suggesting considerable variability for fibre quality improvement. Fibre length and strength showed high phenotypic and genotypic coefficients of variation. Heritability was high for fibre strength, length, and elongation. Fibre length and strength were positively correlated; however, micronaire was negatively correlated with these two traits. Superior accessions were identified for fibre length (11), strength (20), uniformity (7), and elongation (25) among genotypes. Most of the desi lines (71%) had medium micronaire values. Twenty accessions identified were ideal for spinning, showing the fibre strength-to-length ratio of one. Cluster analysis based on Euclidean distance grouped all 712 accessions into four major clusters. Principal component analysis biplot revealed that accessions AC3418, 360-SP1, AC3522B, Kanpur A, Gao16CB-9, and AC3370 were genetically diverse. The superior accessions for fibre quality identified in this study are potential lines for the diploid cotton improvement programme.

Thymus is a widely distributed genus in the Mediterranean region with several species endemic to the Iberian Peninsula. The genetic relationships among the 12 major Thymus taxa, T. albicans, T. caespititius, T. camphoratus, T. capitellatus, T. carnosus, T.lotocephalus, T. mastichina L. ssp. mastichina, T. pulegioides, T. villosus ssp. lusitanicus, T. villosus ssp. villosus, T. zygis ssp. sylvestris and T. zygis ssp. zygis, which occur in Portugal were assessed by AFLP (Amplified Fragment Length Polymorphisms) markers. A general agreement was found between the genetic relationships estimated by the AFLP markers and the accepted Thymus taxonomy based on morphological traits and essential oil content. The AFLP markers also supported suggestions for refinement of the taxonomy of this genus.

Picrorhiza kurroa (family Scrophulariaceae) is a high value medicinal herb used in herbal drug formulations like picroliv, picrolax, etc. It has been collected recklessly from its natural habitat posing endangered status for its existence, thereby necessitating characterization of genetic diversity for its sustainable utilization and conservation. In this study, picrosides content and genetic profiles of 26 accessions of P. kurroa from different locations in north-western Himalayas have been analysed. Picroside-I (P-I) content ranged from 0.37 to 2.7% in fresh shoots whereas total picrosides content (P-I+P-II) ranged from 3.7 to 10.9% in dry rhizomes. High picrosides content accession PKS-1 was identified, both for shoots and rhizomes. To study genetic diversity and correlate picrosides content with their genetic factors, genetic profiling was done using simple sequence repeats (SSRs) identified from P. kurroa transcriptomes. Out of 361 SSR primers tested on 26 accessions, 35 primers yielded polymorphic profiles. Overall low genetic diversity was observed in P. kurroa accessions. The highest polymorphism information content (PIC) of 0.55 was given by SSR marker PKSTS-P9 (TGGTG)4. Mean allele number was 2.97. Mean observed and expected heterozygosities were 0.597 and 0.452, respectively. Among all accessions Nei's genetic diversity (H) was 0.39 and Shannon's information index (I) was 0.58. Cluster analysis of STRUCTURE was comparable to DARWIN. Microsatellite markers would be helpful in the development of DNA diagnostics for the authentication of quality plant material as well as planning a genetic improvement strategy.

Vavilov (1935) described China as the earliest and largest independent centre of origin of cultivated plants. Gansu Province occupies the geographic centre of China. With an area of 454,000 km2, latitudes ranging from 32°3′N to 42°57′N and elevations varying from 500 to 6200 m, the province has an extremely diverse climate and topography. Stretching 1653 km along the ancient Silk Road, Gansu also has a long history of agriculture (6000 years) and for centuries was exposed to dynamic cultural exchanges between the East and the West. The unique combination of these conditions allowed rapeseed to be introduced from Europe and the Middle East, to evolve over a long time of cultivation and to diversify to adapt to many types of ecological conditions and cropping systems in Gansu. Currently, rich genetic resources exist in Gansu for Brassica rapa, B. juncea, B. chinensis and Eruca sativa. Eight landraces are described to demonstrate the diversity of rapeseed genetic resources in Gansu. It is generally recognized that B. chinensis originated in China. Information provided in this paper suggests that Gansu and its adjacent regions could also be a centre of diversity for B. rapa and B. juncea.

Plant height is important for crop yield improvement. In this study, a dwarf mutant, Gmdwarf1, was screened from a γ-ray-treated soybean population. Compared with the wild type, the mutant exhibited later germination, smaller and darker green leaves, and less-elongated shoots. Genome-wide transcriptome detection through RNA-seq analysis revealed that not only gibberellin-related genes but many other genes involved in hormone biosynthetic pathways were also significantly influenced in the mutant. We presumed that Gmdwarf1 might play essential roles in the plant hormone pathways. Future functional analysis of this dwarf mutant would help us to understand the underlying mechanisms and be beneficial for improving soybean yield.

Okra (Abelmoschus esculentus L. Moench) is considered as a treasure house of nutrients and it is one of the major vegetables widely spread all over tropical, subtropical and warm temperate regions of the world. Yellow vein mosaic virus (YVMV) and enation leaf curl virus are the most destructive diseases of okra as they affect both crop growth and yield. Due to the frequent breakdown of resistance and lack of a stable source of resistance in the cultivated species, interspecific hybridization is considered as a reliable approach for durable resistance. Cultivated species from The United States Department of Agriculture and wild accessions from The National Bureau of Plant Genetic Resources were screened at YVMV hotspot (Guntur, Andhra Pradesh) to identify the potential donors for disease resistance. Accessions IC141032 and IC141012 were found to be free from both viruses and categorized as resistant lines. Interspecific hybridization between A. tetraphyllus and A. esculentus revealed a high crossability index of around 80% when A. esculentus was utilized as a female parent. The bottleneck of hybrid sterility was partially overcome by the colchicine treatment of interspecific F1 hybrids. Good seed set was observed when raw colchiploids were backcrossed to the recurrent parent.

Soil salinity is a major limitation to legume production in many areas of the world. Identification of the genetic source of salt tolerance is critical in soybean breeding for improving soybean production in salt-affected regions. Vietnam has unique sources of soybean germplasm and varieties are grown in the area where exposure to salinity is frequent. However, there is little research on the identification of salt tolerant sources in the Vietnamese gene pool. The present study compared 18 Vietnamese soybean cultivars for their differences in salt tolerance. Under a range of NaCl stress from 0 to 200 mM NaCl, there was a large variation in salt tolerance among the 18 soybean lines evaluated. The soybean accession PI 675847 A (Vietnamese variety DT2008), was identified as a useful source of salt tolerance. During vegetative growth, PI 675847 A had lower leaf scorch scores, higher cell membrane stability, better photosynthesis and biomass accumulation under NaCl stress than the other 17 strains evaluated. In addition, PI 675847 A maintained better growth and seed yield in salt-affected soils compared with the sensitive lines. Analyses of ion contents in plant leaves under saline conditions showed that PI 675847 A was able to limit uptake and transport of Na+ and Cl−. Because of its higher productivity under saline conditions, PI 675847 A will be a useful germplasm source in soybean improvement programs for salt tolerance.

Using microsatellite loci, we assessed the mating system and genetic diversity of the dioecious tropical tree Genipa americana in a natural population (NP) and a progeny test (PT). For NP, we also estimated the paternity correlation within and among fruits and mean pollen dispersal distance. As expected for dioecious species, all offspring originated from outcrossing (t = 1). Mating among relatives (1 − ts) and paternity correlation (rp) were variable among progenies (1 − ts = 0.03–0.19; rp = 0.04–0.40), but greater in NP than in PT. Fixation index (F) was generally significant and lower in adults than in offspring, indicating selection against inbred individuals. Paternity correlation was higher within (0.40) than among (0.26) fruits, indicating a lower effective number of pollen donors (Nep) within (2.5) than among (3.8) fruits. Due to the higher rp in NP, the effective size within progenies (Ne) was lower (2.69) than PT (3.27). The pollen dispersal pattern was strongly leptokurtic, suggesting long-distance pollen dispersal (mean of 179 m). The results show that both populations can be used for seed collection in environmental reforestation programmes; however, considering that PT is structured in maternal progenies, NP is more suitable for seed collection due to the lower probability of mating among related trees.

Cultivated lentil suffers yield loss from stemphylium blight, caused by Stemphylium botryosum Wallr. Identification of sources of stemphylium blight resistance and knowledge of the mode of inheritance of resistance are important for developing resistant cultivars. The interspecific recombinant inbred line (RIL) population developed from a cross between the moderately resistant parent Lens culinaris cv. ‘Eston’ and the resistant parent L. ervoides (Brign.) Grande accession IG 72815 was evaluated for stemphylium blight resistance under controlled conditions at the University of Saskatchewan, Saskatoon, Canada, and under field conditions at the Pulses Research Centre (PRC), Ishurdi, Bangladesh. We hypothesized that resistance from both parents will lead to transgressive segregation indicative of pyramiding of resistance genes from the same. However, no resistant transgressive segregants were observed in the RIL population. A large proportion (50%) of the RILs had disease severity levels similar to the resistant parent IG 72815 in experiments conducted under natural disease pressure in Bangladesh. Under controlled conditions in Saskatoon, 38% of RILs had resistance levels similar to IG 72815. Across all environments, 14 RILs consistently had resistance levels similar to IG 72815. The distribution of disease severity scores for all RILs indicated polygenic inheritance of stemphylium blight resistance in the population. RILs with consistent resistant reactions should prove useful for lentil improvement programmes. This will contribute to increasing the productivity of lentil crops in North America and the Indo-Gangetic region, which account for more than 68% of world lentil production.

This study was conducted to determine the opinions of the farmers producing dry bean landraces (DBL) at the regional level of Turkey (eight provinces). In total, 140 questionnaire forms were filled by DBL producers. The population and then the sample size were determined according to 2016-year-preliminary-study. The capital structures of the farms according to the production types were evaluated using the Five-Capital-Model approach, and differences were tested by parametric and non-parametric statistical methods. The study showed that 37.86% of the DBL producers produce as the home-garden-type while 62.14% of them as the field-type, which is larger than 0.1 ha. While home-garden-type producers majorly produce for their own family needs, commercial ideas and concerns are at the forefront for field-type producers. While field-type producers have higher values in terms of five capitals, home-garden-type producers move within a more traditional way of production. More than half of the producers indicate that they will continue to produce DBL; this ratio is higher in home-garden-type producers. This indication of approximately 25% of both types of producers wherein their children will continue to produce DBL, there is a need for more human labour, efficiency and profitability concerns, production mostly by the elderly population increasing the risk of future depletion in DBL-genetic resources. This calls for more efforts to increase the awareness among the young population on the importance and protection of genetic resources and to make special policies for the protection of genetic resources by policy makers and develop models based on genetic resources.

A total of 54 horsegram landraces collected from three altitudinal zones (lower hills, mid hills and higher hills) of Himalayan region were characterized and evaluated for nine quantitative characters. Accessions collected from lower and higher hills were more diverse and differed significantly from mid hills accessions for characters viz., days to flowering, days to physiological maturity, plant height and grain yield per plant. The plotting of first and second principal component axes scores revealed that there was dispersion among the accessions collected from different altitudinal groups. It reveals a less variability among the landraces collected from same altitudinal group but high variability among the landraces collected from different altitudes. This suggests that the horsegram landraces are adapted to specific zone due to different selection pressure practiced by the farmers at different altitudes. Farmers in lower and higher hills select the landraces having high grain yield coupled with less reproductive phase duration and taller in nature.

Roselle (Hibiscus sabdariffa L.) is an economically important plant in Niger. Little is known about its genetic diversity and population structure. In this study, we performed the first diversity assessment of a collection of 124 roselle accessions and 16 accessions of its close relatives Hibiscuscannabinus and Abelmoschusesculentus. Our study was based on ten agromorphological traits and 267 amplified fragment length polymorphism markers. We identified two major distinct groups in H. sabdariffa using a Bayesian method. These two genetic groups were associated with statistical differences for three phenological characteristics: number of days to flowering, 100-seed weight and calyx size. However, the calyx colour, i.e. a morphological characteristic often used to classify different local ecotypes, did not superimpose this separation. Our findings suggested that roselle diversity is genetically structured; the two different groups were clearly associated with morphological differences but were not commonly used by farmers for their classification. The impact of the perceived ecotype structure and its consequence on farmer management is discussed.

In South Europe, rice is grown as an irrigated crop in river deltas where it plays an important role in soil desalinization. Specific varieties are needed for these tough conditions. We analyzed the genetic structure of a set of 305 varieties coming from the European Rice Germplasm Collection (ERGC) with 90 single nucleotide polymorphisms and compared it with a reference set representative of the diversity of Oryza sativa (mini-Germplasm Bank (GB)). These accessions had been characterized for their grain type and growth cycle duration. The polymorphism information contents of the ERGC were lower than those of the mini-GB, indicating a narrower genetic basis. Indeed, almost all ERGC accessions belong to the japonica group. Within the japonica group, both a dendrogram and a Bayesian clustering identified two major clusters. The first cluster encompassed tropical japonicas and American varieties from USA and Argentina characterized by long and narrow grains and medium to long duration. On a finer level, tropical japonicas appear separated from the other accessions. The second cluster is composed of European varieties mostly early or medium in duration and Asian temperate accessions, with a subgrouping based on grain format. A set of 200 accessions was composed for association mapping studies on traits such as salt tolerance.

Assessing genetic diversity and identifying trait-specific germplasm within germplasm collections is necessary for a varietal development programme. Agronomic features were investigated in 318 diverse dry bean germplasm accessions, including check varieties. We observed a lot of genetic variability for the traits studied. A wide range of variations was noticed for days to 50% flowering, days to maturity, pod length, the number of seeds per pod and 100-seed weight (HSW). For eight of the agronomic features evaluated, the analysis of variance revealed substantial differences among the accessions. For all characters, phenotypic coefficient of variation estimations were more significant than genotypic coefficient of variation. Plant height, days to 50% flowering, seed yield (q/ha) and HSW had high heritability and genetic advance as a per cent of the mean. Association analysis revealed a significant positive relationship between HSW, plant height, pod length and seed yield (q/ha). According to a hierarchical clustering analysis based on agronomic features, the diversity of dry bean germplasm has no significant association with their geographical origin. The number of pods per plant, plant height, days to maturity, days to 50% flowering and seed yield had relatively long vectors based on principal components 1 and 2, indicating that genotypes differ significantly. Additionally, the trait-specific donors and bean common mosaic virus disease-resistant accessions, IC360831, ET4515, EC150250, IC340947, IC564797B, EC565693 and ET8409 could be of value for dry bean improvement.

Understanding of genetic diversity and its distribution is essential for promoting the use of genetic resources. The development of core collections using molecular tools has been proposed as a strategy for increasing the economical use and conservation of genetic resources. In this study, we investigated the genetic variation among different geographical origins and potential entries that constituted a core collection of oil palm, using 29 microsatellite markers and by evaluating 788 oil palm accessions. Our results revealed important genetic diversity (HT= 0.759) between oil palm accessions from Angola and Cameroon, which exhibited a low coefficient of genetic differentiation between populations (GST= 0.022). However, the inclusion of oil palm accessions from Indonesia in the analysis resulted in a high coefficient of genetic differentiation between populations (GST= 0.251). We found that the combination of stratified sampling based on a sorting method and a heuristic algorithm was the most effective method for the development of an oil palm core collection set. Using this method, two core collections were identified. The first core collection, comprising 289 entries, contained 271 retained alleles in a sample representing 37% of the entire collection. The second one is a mini core collection, comprising 91 entries, that contained 271 retained alleles with a total He value of 0.72 in a sample representing 11% of the entire collection. The information reported in this study will be of great interest to oil palm researchers because new strategies for breeding programmes can be developed based on these advances.

Southern Sudan has a low population density, abundant land and tremendous agricultural potential. A large number of domesticated crops are grown in the region in a range of cropping systems. There are also numerous useful wild plant genetic resources. Little collecting work has been done in the region, there are few accessions from southern Sudan stored ex situ and publications on the actual and potential plant genetic resources for agriculture are sparse. The region has been a centre of civil conflict with little respite since independence in 1956. The farmers’ fields and natural environments represent in situ genebanks, which following the cessation of hostilities will become extremely important for the rehabilitation of subsistence agriculture, the promotion of cash crop production and the revitalization of the regional economy. Several wild plant species are highly nutritious and merit the attention of plant scientists. This article describes some of the domesticated, semi-domesticated and wild plant genetic resources of the area, and suggests why these are important for agricultural rehabilitation following implementation of a peace accord.

Jackfruit (Artocarpus heterophyllus Lam.) is a nutritious crop from the Moraceae family. The current study was undertaken to evaluate the phenotypic diversity of fruit characteristics using a set of 27 standardized fruit descriptors to describe 28 jackfruit genotypes. These data were used to identify the superior jackfruit genotype that could be used for commercial cultivation. The data revealed a wide range of differences among the genotypes for all the traits studied. Cluster analysis classified the genotypes into four major groups that confirmed the wide diversity among them. Principal component analysis (PCA) also revealed that 80.22% of the variability among the jackfruit genotypes was explained by the first five principal components (PCs). Based on the overall results, the Indian Council of Agricultural Research, Research Complex for Eastern Region (ICAR-RCER) JS 6/3 and 10/3 genotypes were found to be the most promising for table purposes (medium fruit size, pulp percentage >50 and total soluble solid (TSS) >20°Brix), whereas the ICAR-RCER JS 7/7 genotype with large fruit size, pulp percentage >50 and TSS >20°Brix was found to be suitable for processing. The coefficient of variation was the least for traits such as TSS (12.56%) and average seed length (13.56%). Hence, priority may also be given to the TSS and seed size when exploring promising genotypes and operating a selection procedure for crop improvement in jackfruit. The information generated under the study forms a potential baseline for fruit breeders to use in selecting genotypes with superior fruit qualities for jackfruit crop improvement programmes in the future.
