Ca absorption has been shown to be unaffected by high luminal concentrations of two commonly consumed soyabean phyto-oestrogens (PO) (genistein and daidzein) in Caco-2 cells grown under oestrogen-depleted conditions. However, these compounds exhibit dose-dependent biphasic effects in some tissues, such as reproductive tissue and bone. Thus, in light of this biphasic activity, the effect of lower concentrations of genistein and daidzein on Ca absorption requires further investigation. Therefore, the aim of the present study was to investigate the effect of a range of concentrations of genistein and daidzein on Ca absorption in the human Caco-2 intestinal-like cell model. Caco-2 cells were seeded onto permeable filter supports and allowed to differentiate into monolayers. On day 21, the Caco-2 monolayers (n 12 per treatment), grown in oestrogen-deplete media, were then exposed to 10 nm-1,25-dihydroxycholecalciferol (1,25 (OH)2D3), or 1, 10 and 50 μm-genistein or -daidzein for 24 h. After exposure, transepithelial and transcellular transport of 45Ca and fluorescein transport were measured. As expected, 1,25 (OH)2D3 stimulated Ca absorption in Caco-2 cells, by up regulating transcellular transport. Ca absorption was unaffected by either PO at luminal concentrations of 1, 10 or 50 μm, typical of intakes by Western and Asian populations as well as supplemental levels, respectively. The results of this model suggest that the proposed beneficial effects of supplemental levels of these PO compounds on bone mass in postmenopausal women more probably arise from direct effects on bone cells, and not by an indirect effect of these compounds on Ca absorption.

We have observed a stiffening of the gastric wall in mice following oral administration of viscous methyl cellulose (MC) solution as a vehicle for food chemicals, an effect which has not previously been reported. To further investigate this effect, young male ICR mice (n 48) were divided into four groups (one control and three experimental groups) of twelve mice each. Experimental groups I, II and III were administered 0·2 ml of 0·5 % (w/v) MC viscous solution/d via a stomach tube for 2, 3 and 4 weeks, respectively. Stomachs were collected following 12 h fasting. The weight of the stomach and mucosa of seven mice per group was measured, and the mucosa was used for the measurement of 8-oxo-2′-deoxyguanosine (8-oxodG). Stomach tissue slices from the remaining five mice per group were stained with haematoxylin and eosin to measure mucosal thickness, and were immunostained with an HIK1083 antibody to determine the number of gastric gland mucous cells. The mucosal thickness of the groups administered the MC solution for 3 and 4 weeks, and the size of the HIK1083-immunostained area of the groups administered the MC solution for 2, 3 and 4 weeks were significantly increased (P< 0·01) compared with those of the control group. The amount of 8-oxodG was not significantly affected. These results indicate that oral administration of viscous MC solution leads to the thickening of the gastric mucosa, which may be related to an increase in the number of gland mucous cells.

Evidence on the association between children’s food insecurity (FI) and dietary patterns (DPs) is scarce. This study assessed the association between children’s FI and a priori and a posteriori-defined DPs in a Portuguese population-based sample of children. A cross-sectional study including 2800 children from the 10-year-old follow-up of the Generation XXI birth cohort was performed. Data on food security status, assessed by the Self-administered Food Security Survey Module for children (SAFSSMC), dietary intake and socio-demographics were collected. A previously developed Healthy Eating Index (HEI) was adapted for this study. Using the HEI score and its food groups, linear and logistic regression models were performed. Using latent class analysis, five a posteriori-defined DPs were identified. The DPs names considered an overall picture of the DP. Food security status as a categorical (food security/FI) and continuous variable (SAFSSMC raw score: higher scores representing higher FI) was used. Multinomial logistic regression models were used to assess the association between food security status and DPs. Children’s FI (9·4 %) was inversely associated with the HEI score (β=–0·695;95%CI:–1·154,–0·235), representing worse diet quality. A higher SAFSSMC raw score was associated with low fruit and vegetables (OR=1·089;95%CI:1·023,1·159) and seafood and eggs consumption (OR=1·073;95%CI:1·009,1·142) and high consumption of meat and meat products (OR=1·091;95%CI:1·026,1·160), salty snacks (OR=1·067;95%CI:1·003,1·136) and soft drinks (OR=1·097;95%CI:1·031,1·168). The SAFSSMC raw score was positively associated with ‘Low consumption’ (OR=1·119;95%CI:1·016,1·232), ‘Energy-dense foods’ (OR=1·155;95%CI:1·028,1·298) and ‘Snacking’ (OR=1·119;95%CI:1·006,1·245) DPs. FI was associated with worse dietary choices. Intervention strategies targeting food insecure children should be developed to promote healthy dietary habits.

A long-term N balance study was carried out to determine the adequacy of an estimated protein requirement level recommended for young healthy Nigerian women and the effect of the menstrual cycle on the requirement. Eleven healthy young women, 25 (SD 2·6) years, were fed on a diet providing 0·6 g protein (N × 6.25)/kg per d and an average energy intake of 0·17 (SD 0·012) MJ/kg per d. Urine, faeces, sweat and menstrual fluids were collected for estimation of N balance. Menstrual N loss varied among individuals ranging from 46 to 124 mg N/d with an average of 89 (SD 21.8) mg N/d. Individual N balance was found to vary according to the day of the menstrual cycle. Positive N balances were recorded at about ovulation while negative balances were observed just before the onset of menstruation. The average N balance ranged from + 8·49 (SD 5·64) to − 430 (SD 7·84) mg N/kg per d. Nevertheless, an overall cumulative positive N balance of + 5·7 (SD 6·98) mg N/kg per d which did not change significantly with time was observed for the last 5 d of two consecutive 20 d diet periods, although three subjects were in negative N balance. Blood biochemical measurements were stable except for one subject who had elevated serum aspartate aminotransferase (EC 2.6.1.1) levels. These findings suggest that our estimate of protein requirements was sufficient to achieve N balance equilibrium in a majority (70 %) of young women. However, to satisfy 97·5 % of the population, slight adjustments might be necessary in the energy intake since subjects who were in cumulative negative N balance also lost weight.

Maternal nutritional programming by a high-fat (HF) diet is related to hepatic lipid accumulation and steatosis in offspring. Conjugated linoleic acid (CLA) might ameliorate impaired hepatic lipid homoeostasis; therefore, the aim was to investigate the potential preventive effect of maternal CLA consumption on TAG metabolism alterations induced by HF diets in adult male rat offspring receiving or not receiving CLA. Female Wistar rats were fed a control (C) diet, HF diet or HF diet supplemented with CLA (HF+CLA) for 4 weeks before mating and throughout pregnancy and lactation. After weaning, for 9 weeks, male offspring of C or HF rats continued with the same diets as their mothers (C/C or HF/HF groups, respectively) and male offspring of HF+CLA rats were fed HF or HF+CLA diets (HF+CLA/HF or HF+CLA/HF+CLA groups, respectively). Nutritional parameters, serum and liver TAG levels, the TAG secretion rate (TAG-SR) and the activities as well as gene expression of key hepatic enzymes involved in TAG regulation were assessed. The most interesting results were that maternal CLA decreased epididymal white adipose tissue weight and prevented serum and liver TAG accumulation induced by a HF diet in adult male offspring receiving or not receiving CLA. The prevention of liver steatosis in HF+CLA/HF+CLA and HF+CLA/HF offspring was associated with an increased hepatic TAG-SR. Overall, this study provides evidence that maternal CLA consumption programmes TAG regulation and in this way contributes to lowering lipid levels in tissues and preventing liver steatosis in particular.

Asthma-obesity is a multifactorial disease with specific asthma phenotypes that aggravate due to overweight and an unbalanced diet. Furthermore, obese asthmatic patients are corticotherapy-resistant. Therefore, the aims of the present study were to evaluate the effects of an interdisciplinary intervention on food consumption, body composition, lung function and adipokines in asthmatic and non-asthmatic obese adolescents and to investigate the influence of nutrients on lung function. Obese non-asthmatic (n 42) and obese asthmatic (n 21) adolescents of both sexes were enrolled in the present study. Food intake, adipokine levels, body composition, asthma symptoms and lung function were assessed across the study. After the intervention of 1 year, there was a reduction (P ≤ 0·01) in BMI, body fat percentage, visceral and subcutaneous fat and an increase (P ≤ 0·01) in lean mass and all lung function variables in both groups, except the relation between forced expiratory volume in 1 s and forced vital capacity (FEV1:FVC) in non-asthmatic patients. Moreover, both groups decreased lipid and cholesterol consumption (P ≤ 0·01). The highest energy consumption (β = −0·021) was associated with lower values of FVC. Similarly, carbohydrate consumption (β = −0·06) and cholesterol were negative predictors (β = −0·05) in FEV1:FVC. However, the consumption of Ca (β = 0·01), fibres (β = 1·34) and vitamin A (β = 0·01) were positive predictors of FEV1:FVC. Asthma-obesity interdisciplinary treatment promoted an improvement on food consumption and lung function in adolescents and demonstrated that the consumption of nutrients influenced an increase in lung function.

1. The effect of supplementing barley diets with urea (U), extracted decorticated groundnut meal (GNM) or Peruvian fish meal (PFM) on plasma free amino acid concentrations in sheep have been examined and the first limiting amino acid has been indicated by measuring the changes in the concentration of the plasma essential amino acids (PEAA) during a rumen infusion of a volatile fatty acid (VFA) mixture.

2. Three wethers fitted with rumen and re-entrant duodenal cannulas were given isonitrogenous, isoenergetic diets containing (g/kg dry matter (DM)) U 20, GNM 106 or PFM 78, the crude protein (nitrogen × 6.25) contents being 139, 145 and 148 respectively. The sheep were fed hourly, the mean daily dm intake being 0.634 kg.

3. Plasma concentrations of valine, threonine, lysine, isoleucine and leucine were linearly related to their concentrations in duodenal digesta.

4. A VFA mixture was infused into the rumen for 6 h to supply (mmol/min) acetate 1.47, propionate 0.22 and n-butyrate 0.27. Blood samples were taken 6 h before, during and 12 h after the end of the infusion.

5. The concentration of all PEAA decreased relative to the pre-infusion and post-infusion controls but there were no significant differences between diets.

6. The mean decreases in concentration averaged over all three diets showed that the decrease in concentration of methionine (41.5%) was far greater than for any other essential amino acid suggesting that under these conditions methionine was the first limiting amino acid.

Early life nutrition and feeding practices are important modifiable determinants of subsequent obesity, yet little is known about the circadian feeding pattern of 12-month-old infants. We aimed to describe the 24-h feeding patterns of 12-month-old infants and examine their associations with maternal and infant characteristics. Mothers from a prospective birth cohort study (n 431) reported dietary intakes of their 12-month-old infants and respective feeding times using 24-h dietary recall. Based on their feeding times, infants were classified into post-midnight (00.00–05.59 hours) and pre-midnight (06.00–23.59 hours) feeders. Mean daily energy intake was 3234 (sd 950) kJ (773 (sd 227) kcal), comprising 51·8 (sd 7·8) % carbohydrate, 33·9 (sd 7·2) % fat and 14·4 (sd 3·2) % protein. Mean hourly energy intake and proportion of infants fed were lower during post-midnight than pre-midnight hours. There were 251 (58·2 %) pre-midnight and 180 (41·8 %) post-midnight feeders. Post-midnight feeders consumed higher daily energy, carbohydrate, fat and protein intakes than pre-midnight feeders (all P<0·001). The difference in energy intake originated from energy content consumed during the post-midnight period. Majority (n 173) of post-midnight feeders consumed formula milk during the post-midnight period. Using multivariate logistic regression with confounder adjustment, exclusively breast-feeding during the first 6 months of life was negatively associated with post-midnight feeding at 12 months (adjusted OR 0·31; 95 % CI 0·11, 0·82). This study provides new insights into the circadian pattern of energy intake during infancy. Our findings indicated that the timing of feeding at 12 months was associated with daily energy and macronutrient intakes, and feeding mode during early infancy.

Evidence suggests that differences in meal timing between weekends and weekdays can disrupt the body’s circadian rhythm, leading to a higher BMI. We aimed to investigate the associations between mealtime variation from weekdays to weekends (eating midpoint jetlag), dietary intake and anthropometric parameters, based on individuals’ chronotype. The study utilised data from National Health and Nutrition Examination Survey 2017–2018. Food consumption was estimated by weighted average of participants’ food intake on weekdays and weekends. Eating midpoint jetlag, defined as the difference between the midpoint of the first and last mealtimes on weekends and weekdays, was calculated. Chronotype was assessed by participants’ mid-sleep time on weekends, adjusted for sleep debt. Linear regression analysis was conducted to investigate the associations between variables. The sample was categorised into chronotype tertiles. Among individuals in the third chronotype tertile, there was a positive association between eating midpoint jetlag and BMI (β = 1·2; 95 % CI (1·13, 1·27)). Individuals in the first tertile showed a positive association between eating midpoint jetlag and energy (β = 96·9; 95 % CI (92·9, 101·7)), carbohydrate (β = 11·96; 95 % CI (11·2, 12·6)), fat (β = 3·69; 95 % CI (3·4, 3·8)), cholesterol (β = 32·75; 95 % CI (30·9, 34·6)) and sugar (β = 8·84; 95 % CI (8·3, 9·3)) intake on weekends. Among individuals with an evening tendency, delaying meals on weekends appears to be linked to a higher BMI. Conversely, among individuals with a morning tendency, eating meals later on weekends is associated with higher energetic intake on weekends.

1. Pigs were fed on semi-purified food. The magnesium content of the experimental diet was 1·0 and that of the control diet 1·9 g/kg.

2. In the low-Mg group serum triglycerides and blood lactate values were increased and base excess and standard bicarbonate values were decreased, indicating metabolic acidosis.

3. A significant positive dependence was found between blood pH and serum Mg:Ca value as well as between blood pH and serum Mg, and also between body temperature and blood lactate values in the low-Mg group. None of these dependences was significant in the control group with magnesium acetate supplementation in the food.

4. No specific histopathological changes were found in heart, liver or kidney of the experimental animals.

5. The results indicate that an increase in serum triglycerides does not need to depend on the amount or quality of food fat.

6. The present study shows that, in pigs, a low-Mg diet may cause metabolic disturbances in instances when the food Mg content is distinctly higher than the normal values recommended by the (UK) Agricultural Research Council (1966) and (US) National Research Council (1968).

1. The effect of cholesterol feeding during and after a period of protein malnutrition lasting 4 weeks was examined in the rat. Indices measured were plasma total cholesterol, triglycerides and protein levels, and aorta total cholesterol, triglycerides, hexosamine and hydroxyproline concentrations.

2. In both plasma and aorta, total cholesterol and triglycerides levels were higher in the low-protein diet group than in the standard-protein diet group, when cholesterol was supplied in both diets.

3. During the malnutrition period, cholesterol feeding led to a greater decrease in plasma protein than that promoted by the low-protein diet without cholesterol, while aorta hexosamine levels decreased to a lesser extent.

4. Cholesterol feeding with a standard-protein diet promoted a slight and temporary increase in aorta hydroxyproline levels, while a decrease in aorta hexosamine concentration was observed. Cholesterol feeding with the low-protein diet, on the other hand, also promoted a decrease in aorta hexosamine levels but to a lesser extent.

5. During the recovery period, cholesterol feeding impaired the return of plasma protein, aorta hexosamine and lipid levels to that of the control values.

6. These findings demonstrate that cholesterol feeding promotes different changes in aorta and plasma, depending on whether or not protein is supplied by the diet in adequate amounts. This point could be important in relation to the development of atherosclerosis during recovery from a period of malnutrition.

1. The effects of level of feeding and level of protein in the early postnatal period on the cellularity of subcutaneous adipose tissue and body fat content of pigs were investigated in two experiments.

2. In Expt 1, piglets were given a common liquid diet at energy intakes equivalent to 2·8 or 5·2 times energy for maintenance (M) between 1·8 and 6·5 kg live weight (LW), and a common dry diet at 2·8 or 4·5 M between 6·5 and 20 kg LW. Between 20 and 75 kg LW all pigs were given a second dry diet at 4·0 M.

3. In both experiments the effects of nutritional treatments on fat cell number at 20 and 75 kg LW (Expt 1) and at 45 kg LW (Expt 2) were assessed by measuring the DNA content of the subcutaneous adipose tissue contained in the left shoulder joint. Fat cell size was assessed in the same tissue by measuring the diameter of collagenase-released adipocytes.

4. In Expt 1, raising the level of feeding between 1·8 and 6·5 kg LW increased body fat content and average fat cell diameter at both 6·5 (P < 0·01) and 20 kg LW (P < 0·05) but had no effect on either measurement at 75 kg LW. Similarly, raising the level of feeding between 6·5 and 20 kg LW increased body fat content and fat cell size at both 20 (P < 0·01) and 75 kg LW (P < 0·05). There was an almost twofold increase in the DNA content of subcutaneous adipose tisuue between 20 and 75 kg LW. However, it was not significantly affected at either weight by level of feeding before or subsequent to 6·5 kg LW.

5. In Expt 2, reducing the level of dietary protein between 1·8 and 6·5 kg LW increased body fat content (P < 0·01) and fat cell size (P < 0·01) at the latter weight. Although level of dietary protein to 6·5 kg LW had no effect on body fat content or the weight of subcutaneous adipose tissue in the shoulder joint at 45 kg LW, pigs given the lowest-protein diet contained less DNA (P < 0·05) in the subcutaneous adipose tissue and had larger (P < 0·05) fat cells than those given the highest-protein diet to 6·5 kg LW. Reducing the protein content of the diet fed subsequent to 6·5 kg LW increased the body fat content (P < 0·01) and fat cell size (P < 0·01) at 45 kg LW.
