The isolated retina of the goldfish has proven a valuable resource for studying the variability of firing of retinal ganglion cells. Three major areas of study are considered here: the variability of maintained discharges, the correlated firing of neighboring ganglion cells, and the variability of responses to light. The sources of variability, its relationship to retinal processing, and its possible functional role in perception are examined through these three aspects of variability. The results are related to similar studies in mammals (mainly cats). This retrospective is biased toward my studies over 30 years.

Glaucoma is an eye disease characterized by a progressive vision loss usually starting in peripheral vision. However, a deficit for scene categorization is observed even in the preserved central vision of patients with glaucoma. We assessed the processing and integration of spatial frequencies in the central vision of patients with glaucoma during scene categorization, considering the severity of the disease, in comparison to age-matched controls. In the first session, participants had to categorize scenes filtered in low-spatial frequencies (LSFs) and high-spatial frequencies (HSFs) as a natural or an artificial scene. Results showed that the processing of spatial frequencies was impaired only for patients with severe glaucoma, in particular for HFS scenes. In the light of proactive models of visual perception, we investigated how LSF could guide the processing of HSF in a second session. We presented hybrid scenes (combining LSF and HSF from two scenes belonging to the same or different semantic category). Participants had to categorize the scene filtered in HSF while ignoring the scene filtered in LSF. Surprisingly, results showed that the semantic influence of LSF on HSF was greater for patients with early glaucoma than controls, and then disappeared for the severe cases. This study shows that a progressive destruction of retinal ganglion cells affects the spatial frequency processing in central vision. This deficit may, however, be compensated by increased reliance on predictive mechanisms at early stages of the disease which would however decline in more severe cases.

Motion-sensitive neurons in the visual systems of many species, including humans, exhibit a depression of motion responses immediately after being exposed to rapidly moving images. This motion adaptation has been extensively studied in flies, but a neuronal mechanism that explains the most prominent component of adaptation, which occurs regardless of the direction of motion of the visual stimulus, has yet to be proposed. We identify a neuronal mechanism, namely frequency-dependent synaptic depression, which explains a number of the features of adaptation in mammalian motion-sensitive neurons and use it to model fly motion adaptation. While synaptic depression has been studied mainly in spiking cells, we use the same principles to develop a simple model for depression in a graded synapse. By incorporating this synaptic model into a neuronally based model for elementary motion detection, along with the implementation of a center-surround spatial band-pass filtering stage that mimics the interactions among a subset of visual neurons, we show that we can predict with remarkable success most of the qualitative features of adaptation observed in electrophysiological experiments. Our results support the idea that diverse species share common computational principles for processing visual motion and suggest that such principles could be neuronally implemented in very similar ways.

Receptive fields of retinal ganglion cells in turtle have excitatory and inhibitory components that are balanced along the dimensions of wavelength, functional ON and OFF responses, and spatial assignments of center and surround. These components were analyzed by spectral light adaptations and by the glutamate agonist, 2-amino-4-phosphonobutyric acid (APB). Extracellular recordings to stationary and moving spots of light were used to map changes in receptive fields. ON spike counts minus OFF spike counts, derived from flashed stationary light spots, quantified functional shifts by calculating normalized mean response modulations. The data show that receptive fields are not static, but rather are dynamic arrangements which depend on linked, antagonistic balances among the three dimensions of wavelength, ON and OFF response functions, and center/surround areas.

The objective of the present work was to determine the interaction of cone inputs in the response of horizontal cells using heterochromatic flicker photometry (HFP). Intracellular electrophysiological recordings were made in horizontal cells of isolated retinae of carp maintained in physiological solution, with the receptor side up. Sharp glass microelectrodes filled with 3 M KCl solution with resistances between 100 and 120 MΩ were used. Stimuli comprised six cycles of two 6-Hz sinusoidal light waves in counterphase adjusted for the same number of quanta: a green light (550 nm) from a monochromator with a Xenon lamp and an LED red light (628 nm). The stimulation program consisted of 10 steps with the 550-nm wave at constant amplitude, while the 628-nm wave varied in increments of 10% up to 100%, followed by another 10 steps with the 628-nm wave at constant amplitude while the 550-nm wave varied in increments of 10% up to 100%. We recorded responses from four different horizontal cell classes: H1 (monophasic, broadband, n = 37), H2 (biphasic, red-green color-opponent, n = 13), and H3 (biphasic, blue-yellow color-opponent, n = 2) cone horizontal cells; and RH (monophasic, broadband, n = 3) rod horizontal cells. H1 and RH horizontal cells showed a similar cancellation point at a heterochromatic mixture consistent with mixed inputs from 630- and 550-nm cones. No cancellation point was found for the H2 cell class. Fish H1 cells add cone inputs and signal “luminance” in light levels appropriate for cone stimulation. The same occurs with RH cells, which also signal “luminance,” but in light levels appropriate for rod work. For both cell classes there is an HFP cancellation point occurring at a combination of 628-nm and 550-nm lights in opposing phase that leads to the cancellation of the cell's response. No cancellation was found for H2 and H3 cells, which are the chromatically opponent horizontal cells in lower vertebrates.

A projection by the superior colliculus to the supraoculomotor area (SOA) located dorsal to the oculomotor complex was first described in 1978. This projection’s targets have yet to be identified, although the initial study suggested that vertical gaze motoneuron dendrites might receive this input. Defining the tectal targets is complicated by the fact the SOA contains a number of different cell populations. In the present study, we used anterograde tracers to characterize collicular axonal arbors and retrograde tracers to label prospective SOA target populations in macaque monkeys. Close associations were not found with either superior or medial rectus motoneurons whose axons supply singly innervated muscle fibers. S-group motoneurons, which supply superior rectus multiply innervated muscle fibers, appeared to receive a very minor input, but C-group motoneurons, which supply medial rectus multiply innervated muscle fibers, received no input. A number of labeled boutons were observed in close association with SOA neurons projecting to the spinal cord, or the reticular formation in the pons and medulla. These descending output neurons are presumed to be peptidergic cells within the centrally projecting Edinger–Westphal population. It is possible the collicular input provides a signaling function for neurons in this population that serve roles in either stress responses, or in eating and drinking behavior. Finally, a number of close associations were observed between tectal terminals and levator palpebrae superioris motoneurons, suggesting the possibility that the superior colliculus provides a modest direct input for raising the eyelids during upward saccades.

This study describes a phenomenon of transient expression of corticotropin releasing factor-like immunoreactivity (CRF-LI) in immature horizontal cells of the developing rat retina. These cells could be distinguished from those destined to become CRF-LI amacrine cells in the adult by their location within the outer portion of the neuroblastic layer (NBL) and by their ontogenetic pattern. Upon initial detection on postnatal day 3 (PD-3), faint CRF-LI cellular profiles were found in the outer portion of the NBL, limited to the central region of the retina. Subsequently, on PD-5, these profiles began to appear in the periphery, forming a single horizontal row along the outermost aspect of the developing inner nuclear layer (INL), concomitant with the establishment of the outer plexiform layer (OPL). The results of our birth-dating study combining immunohistochemistry and [3H]-thymidine autoradiography indicated that these cells were generated between embryonic day 14 and 18. These findings are consistent with them being horizontal cells. Between PD-7 and PD-9, CRF-LI in horizontal cells began to diminish progressively following a center-to-periphery gradient such that only sporadic, faintly immunoreactive patches of cells could be seen by the time of eye opening (PD-15). Around PD-19, it declined to levels below immunohistochemical detection. However, when rats were reared in complete darkness beginning at birth until PD-21, the period of CRF-LI expression in horizontal cells was prolonged and persisted throughout the first three postnatal weeks of development.

The presence and concentrations of each of the three subtypes of opioid receptors (mu, kappa, and delta) has been studied in the accessory optic nuclei (dorsal, lateral, and medial terminal nuclei and the interstitial nucleus of the superior fasciculus, posterior fibers: DTN, LTN, MTN, and inSFp) in normal young rats with radioligands directed towards each opioid receptor subtype. The changes in mu opioid receptors have also been investigated in monocularly enucleated rats in which one eye was removed and the rats sacrificed at postoperative day (PO) 2, 3, 5, 7, 14, and 30. As the MTN is the only accessory optic nucleus of the rat large enough for semiquantitative evaluation, the mu receptor population of the MTN has been subjected to optical microdensitometric analysis.

All four of the accessory optic nuclei (AOS nuclei) are found to contain exceedingly high levels of mu opioid receptor binding with the selective radioligand [3H]-[D-Ala, MePhe4, Gly-ol5] (DAGO), low levels of kappa opioid receptor binding using the radioligand [3H]-[ethylketocyclazocine] (EKC) together with the competing agents [D-Pro4]-morphiceptin and [D-Ser2, Thr6]-Leu-enkephalin, and an absence of delta opioid receptor binding with the radioligand [3H]-[D-A1a2, D-Leu5]-enkephalin (DADLE) combined with the competing agent [D-Pro4]-morphiceptin. Monocular enucleation, as studied on the mu opioid receptor population with this experimental approach, results in virtually a complete loss of mu opioid receptors throughout all four of the contralaterally located AOS nuclei, including both dorsal and ventral subdivisions of the medial terminal nucleus (MTNd, v). Kappa and delta receptors are very few (kappa receptors) or are lacking (delta receptors) in the AOS nuclei, and for this reason, the effects of monocular enucleation on these two opioid receptor subtypes have not been investigated. Monocular enucleation also produces a significant lowering in mu receptor binding in other primary optic nuclei (the lateral geniculate nuclei, nucleus of the optic tract, and superficial layers of the superior colliculus) and in the pars principalis of the medial geniculate nucleus (description of changes in mu receptors in non-accessory optic primary optic nuclei will be considered elsewhere).

Microdensitometric study of the MTNd, v shows that the decreased binding of mu receptors in this nucleus is barely detectable (about 6%) at PO2 and rises to 6–15% at PO3. At PO5 receptor loss reaches approximately 62%, whereas at PO7 it is about 81% complete. At PO14 and PO30, the mu receptor loss is nearly complete at around 93%. Mu receptor loss involves all of the AOS nuclei contralateral, but none ipsilateral, to ocular enucleation, an observation entirely consistent with the overwhelmingly crossed (about 97%) nature of the retinofugal projection to the rat accessory optic nuclei. These opioid receptors represent a prominent feature in the AOS and other primary optic nuclei of the rat. Their role in visuomotor control remains uncertain but probably involves the fine-tuning of information concerned with compensatory eye movements.

In switching from studying native cyclic nucleotide-gated (CNG) ion channels in rod cells to studying the corresponding cloned channels expressed in Xenopus oocytes, we changed our perfusion system to a more efficient one. This change involved replacing culture flasks and a small plexiglass/glass chamber with plastic syringes, metal needles, and plastic petri dishes. We now report that these new perfusion system components release agents that distort or obscure measured functional properties of rod CNG channels. The magnitude and time course of appearance of the artifacts vary widely among individual components (e.g. from syringe to syringe). The effects most resemble voltage-dependent block of the channels, giving a decrease in current at positive potentials, and producing distortions of the kinetics and voltage dependence of channel activation.

Earlier, we reported that nucleus rotundus (Rt) together with its inhibitory complex, nucleus subpretectalis/interstitio-pretecto-subpretectalis (SP/IPS), had significantly higher activity in pigeons performing figure–ground discrimination than in the control group that did not perform any visual discriminations. In contrast, color discrimination produced significantly higher activity than control in the Rt but not in the SP/IPS. Finally, shape discrimination produced significantly lower activity than control in both the Rt and the SP/IPS. In this study, we trained pigeons to simultaneously perform three visual discriminations (figure–ground, color, and shape) using the same stimulus displays. When birds learned to perform all three tasks concurrently at high levels of accuracy, we conducted bilateral chemical lesions of the SP/IPS. After a period of recovery, the birds were retrained on the same tasks to evaluate the effect of lesions on maintenance of these discriminations. We found that the lesions of the SP/IPS had no effect on color or shape discrimination and that they significantly impaired figure–ground discrimination. Together with our earlier data, these results suggest that the nucleus Rt and the SP/IPS are the key structures involved in figure–ground discrimination. These results also imply that thalamic processing is critical for figure–ground segregation in avian brain.

This study aims to create a prediction model for state-space estimation and to elucidate the required information processing for identifying an external space in prism adaptation. Subjects were 57 healthy students. The subjects were instructed to rapidly perform reaching movements to one of the randomly illuminating light-emitting diode lights. Their movements were measured while wearing prism glasses and after removing that. We provided the following four conditions and control. In target condition, reaching error distance was visually fed back to the subject. In trajectory condition, the trajectory of fingertip movement could be seen, and the final reaching error was not fed back. Two restricted visual feedback conditions were prepared based on a different presentation timing (on-time and late-time conditions). We set up a linear parametric model and an estimation model using Kalman filtering. The goodness of fit between the estimated and observed values in each model was examined using Akaike information criterion (AIC). AIC would be one way to evaluate two models with different number of parameters. In the control, the value of AIC was 179.0 and 154.0 for the linear model and Kalman filtering, respectively, while these values were 173.6 and 161.1 for the target condition, 202.8 and 159.7 for the trajectory condition, 192.7 and 180.8 for the on-time condition, and 206.9 and 174.0 for the late-time condition. Kalman gain in the control was 0.07–0.26. Kalman gain relies on the prior estimation distribution when its value is below 0.5. Kalman gain in the trajectory and late-time conditions was 0.03–0.60 and 0.08–0.95, respectively. The Kalman filter, a state estimation model based on Bayesian theory, expressed the dynamics of the internal model under uncertain feedback information better than the linear parametric model. The probabilistic estimation model can clearly simulate state estimation according to the reliability of the visual feedback.

The appearance of cGMP-gated cation channel protein in the postnatal rat retina has been studied by fluorescence immunocytochemistry of radial retinal sections and immunoblots of retinal membrane proteins. Channel immunoreactivity was first detectable with RCNGC1-7H2 monoclonal antibody at postnatal day 7 (PN7) by both methods. Immunocytochemical label in retinal sections was localized to the outer segments, and immunoreactivity increased with increasing age. We also compared the developmental appearance of the cGMP-gated cation channel to that of other phototransduction proteins and developmental markers. RET-P2, a monoclonal antibody recognizing the 39-kDa rds/peripherin disc protein, first labeled outer segments at PN7, coincident with cGMP-gated cation channel expression. Double labeling of the same section of PN7 rat retina with RET-P2 and R309 (a polyclonal antiserum against the rod cGMP-gated cation channel) revealed identical patterns of labelling. Similarly, double labeling with RCNGC1-7H2 and an antibody against the rod cGMP-phosphodiesterase gave coincident labeling, suggesting coordinate expression mechanisms of phototransduction proteins with each other and with outer segment structural proteins.

Rod and cone photoreceptors send their signals to ON- and OFF-retinal ganglion cells through different pathways in the primate retina. We hypothesized that increments and decrements of light may be processed differently by the rod-bipolar pathway because of the funneling of the rod signal through the rod bipolar cell. We tested this hypothesis using a psychophysical adaptation paradigm, which has provided evidence that photopic increments and decrements of light are processed by ON- and OFF-pathways in the human visual system. We had observers adapt to either a rapid-on or rapid-off sawtooth waveform, under both photopic and scotopic conditions. We then measured detection thresholds for one cycle of a rapid-on or rapid-off sawtooth stimulus. For photopic stimuli, sawtooth adaptation asymmetrically raised thresholds for test stimuli in a manner that depended on the polarity of the adaptation stimulus. For scotopic stimuli, thresholds were raised, but no significant selective adaptation effect was found. By repeating the photopic condition with sawtooth stimuli which had been filtered using an impulse response function derived for the rod system, we demonstrated that the lack of selective adaptation was not a consequence of the sluggish temporal response of the rod-bipolar pathway. We conclude instead that the reduced effectiveness of sawtooth adaptation is due to channeling of rod photoreceptor signals through the rod bipolar cell before reaching ON- and OFF-ganglion cells.

Many complex behaviors rely on guidance from sensations. To perform these behaviors, the motor system must decode information relevant to the task from the sensory system. However, identifying the neurons responsible for encoding the appropriate sensory information remains a difficult problem for neurophysiologists. A key step toward identifying candidate systems is finding neurons or groups of neurons capable of representing the stimuli adequately to support behavior. A traditional approach involves quantitatively measuring the performance of single neurons and comparing this to the performance of the animal. One of the strongest pieces of evidence in support of a neuronal population being involved in a behavioral task comes from the signals being sufficient to support behavior. Numerous experiments using perceptual decision tasks show that visual cortical neurons in many areas have this property. However, most visually guided behaviors are not categorical but continuous and dynamic. In this article, we review the concept of sufficiency and the tools used to measure neural and behavioral performance. We show how concepts from information theory can be used to measure the ongoing performance of both neurons and animal behavior. Finally, we apply these tools to dorsal medial superior temporal (MSTd) neurons and demonstrate that these neurons can represent stimuli important to navigation to a distant goal. We find that MSTd neurons represent ongoing steering error in a virtual-reality steering task. Although most individual neurons were insufficient to support the behavior, some very nearly matched the animal’s estimation performance. These results are consistent with many results from perceptual experiments and in line with the predictions of Mountcastle’s “lower envelope principle.”

Averaged grating-evoked cortical potentials were recorded from area 17 of awake cats. Peak latency of early components of the visual-evoked potential (VEP) response to stimulus onset increased as a function of spatial frequency, while amplitude tended to be largest at intermediate spatial frequencies. Latency increased and amplitude generally decreased to lower spatial-frequency stimuli (<0.25 cycle/deg) in the presence of a uniform flickering field (UFF). The UFF had a relatively small or opposite effect on peak latency and amplitude for higher spatial-frequency stimuli (>0.50 cycle/deg). The VEP response to stimulus offset was present only at low spatial frequencies and was virtually eliminated by the presence of the UFF. The effects were similar whether the target and UFF background were simultaneously presented or briefly separated; however, the UFF had no effect when the two were spatially separated. The effects of the UFF background on VEP onset response increased with increasing temporal frequency from 2–8 Hz; offset responses were affected similarly at all temporal frequencies. These effects are similar to those observed in humans and suggest that two spatio-temporally tuned mechanisms contribute to the early VEP response. In the cat, the mechanisms seem to correspond to X and Y cells in the dorsal lateral geniculate nucleus.

A photon-counting retinal densitometer is described that has been designed optically and electronically for improved sensitivity and reliability. The device allows measurement of visual pigments through the undilated natural pupils of subjects at relatively low levels of measuring lights, and serves also as an adaptometer for direct comparisons between pigment bleaching or regeneration and light or dark adaptation. Instrumental control and data collection are by computer to permit rapid and simple data analysis and comparisons between subjects. The methods by which the sensitivity and reliability have been enhanced are described in detail, and some examples of experimental results are presented.

In the Royal College of Surgeons, rat photoreceptor degeneration occurs over the first several months of life, causing deterioration of visual cortical responsiveness seen as greater numbers of cells being nonresponsive to visual stimulation, poor tuning of those cells that do respond, and an overall tendency for domination by the contralateral visual input. If the progress of degeneration in one eye is slowed by intraretinal cell transplantation, cortical responses to stimulation of the remaining, untreated, eye are much stronger, better tuned and histograms of ocular dominance resemble more those in normal rats. This suggests that the rescued eye is able to enhance performance in the untreated eye by some form of postsynaptic mechanism.

The purpose of this study is to determine how the viewing distance affects the pass/fail results of the CN Lantern (CNLan). The CNLan is a color vision test designed for the railway industry. It presents 15 triplets of colored lights that could be any combination of red, green and yellow. The test was viewed from 4.6 m and 2.3 m. Sixty-seven color-defectives participated in the first part of the study. Sixty-six percent of the subjects repeated the experiment 10 days later. There was a significant (P < 0.05) decrease in the mean number of errors from 7.6 to 4.3 as the distance decreased. There was also a corresponding increase in the percentage of subjects who passed from 9.0% at 4.6 m to 20.9% at the 2.3 m viewing distance. None of the subjects who passed at the longer distance failed at the shorter distance. The replication results were statistically identical to the first session (P > 0.05). Decreasing the CNLan viewing distance by 50% does decrease the number of errors and increase the pass rate. This indicates that some color-defectives could work in the railway yards where the sighting distances for the signal lights are shorter than on the main track.

The watercolor illusion is a color spreading effect at long-range diffusing from a thin colored contour juxtaposed to a chromatic one of higher contrast and a object-hole effect across a large area (Pinna, 1987; Pinna et al., 2001, 2003; Pinna & Reeves, 2006). The watercolored figure appears evenly colored by an opaque light veil of chromatic tint (coloration effect), with a clear surface color property spreading from the lighter contour. At the same time, the watercolored figure manifests a strong figure-ground organization and a solid figural appearance comparable to a bas-relief illuminated from the top (object-hole effect). It appears similar to a rounded surface segregated in depth, which extends out from the flat surface. The complementary region appears as a hole or empty space. The phenomenal properties of coloration and object-hole effects raise some questions. Can the two effects be considered relatively independent? Under what conditions can a possible dissociation occur? How does the dissociation of one effect, say the coloration, influence the object-hole effect and vice versa? To answer these questions two new effects related to the watercolor illusion were psychophysically studied: (1) the “uneven watercolor,” based on a modified watercolor figure without volumetric and three-dimensional properties but with a strong coloration effect and (2) the “watercolor surface capture,” where oblique lines within a watercolor figure appear bulging, curved in depth and segregated from those that are perceived as placed in the background or perceived through holes. The results of two experiments suggest that the coloration effect can be dissociated from the object-hole one. These results are discussed in the light of a simple summation hypothesis of the underlying effects composing the whole figurality. This hypothesis can suggest further investigation both in the phenomenal and in the neurophysiological domain.

The visual response to a flash given in the dark is known to saturate according to the Michaelis-Menten relationship. Nevertheless, the incremental response from increasing levels of mean luminance tends to follow a Weber-Fechner relationship well into the saturation range determined from the Michaelis-Menten results. This sensitivity transformation from Michaelis-Menten to Weber-Fechner is an important characteristic of light adaptation in the vertebrate retina. Recent studies concerning the role of calcium in photoreceptor adaptation have shown that the relaxation from peak to plateau in the response of isolated photoreceptors was absent under conditions in which adaptation was blocked. Comparing the pronounced relaxation from peak to plateau in turtle horizontal cells with the absence of such relaxation in the catfish response, we noted also that turtle incremental sensitivity shows a Weber-Fechner relationship while catfish incremental sensitivity more closely follows the local slope of the Michaelis-Menten relation. Based on these observations, we have obtained an expression to relate the relaxation from peak to plateau with the sensitivity transformation. We assume that adaptation shifts the half-maximum point of the Michaelis-Menten curve so that the light response relaxes to a plateau value equal to a specified fraction φ of the peak response. We show that this manipulation alone results in a transformation from Michaelis-Menten kinetics to Weber-Fechner sensitivity.
