In normal adult cats, a monoclonal antibody directed toward the NR-1 subunit of the N-methyl-d-aspartate (NMDA) receptor (Pharmingen, clone 54.1) produced dense cellular and neuropil labeling throughout all layers of the lateral geniculate nucleus (LGN) and adjacent thalamic nuclei, including the thalamic reticular, perigeniculate, medial intralaminar, and ventral lateral geniculate nuclei. Cellular staining revealed well-defined somata, and in some cases proximal dendrites. NMDAR-1 cell labeling was also evident in the LGN of early postnatal kittens, suggesting that developing LGN cells possess this receptor subunit at or before eye opening. Within the A-layers of the adult LGN, staining encompassed a wide range of soma sizes. Soma size comparisons of NMDAR-1 stained cells with those stained with an antibody directed toward a nonphosphorylated neurofilament protein (SMI-32), which selectively stains Y-relay cells (Bickford et al., 1998), or an antibody to glutamic acid decarboxylase (GAD), which stains for GABAergic interneurons, suggested that NMDA receptors are utilized by relay cells and interneurons. NMDAR-1 staining was also observed in the LGN of cats with early monocular lid suture. Although labeling was apparent in both deprived and nondeprived A-layers of LGN, the distribution of soma sizes was significantly different. In the deprived A-layers of LGN, staining was limited to small- and medium-sized cells. Cells with relatively large soma were lacking. However, cell density measurements as well as soma size comparisons with cells stained for Nissl substance suggested these differences were due to deprivation-induced cell shrinkage and not to a loss of NMDAR-1 staining in Y-cells. Taken together, these results suggest that NMDA receptors are utilized by both relay cells and interneurons in LGN and that alterations in early visual experience do not necessarily affect the expression of NMDA receptors in the LGN.

It is often assumed that the ultimate goal of a motion-detection system is to faithfully represent the time-dependent velocity of a moving stimulus. This assumption, however, may be an arbitrary standard since the requirements for a motion-detection system depend on the task that is to be solved. In the context of optomotor course stabilization, the performance of a motion-sensitive neuron in the fly's optomotor pathway and of a hypothetical velocity sensor are compared for stimuli as are characteristic of a normal behavioral situation in which the actions and reactions of the animal directly affect its visual input. On average, tethered flies flying in a flight simulator are able to compensate to a large extent the retinal image displacements as are induced by an external disturbance of their flight course. The retinal image motion experienced by the fly under these behavioral closed-loop conditions was replayed in subsequent electrophysiological experiments to the animal while the activity of an identified neuron in the motion pathway was recorded. The velocity fluctuations as well as the corresponding neuronal signals were analyzed with a statistical approach taken from signal-detection theory. An observer scrutinizing either signal performs almost equally well in detecting the external disturbance.

The vertebrate eye allows to capture an enormous amount of detail about the surrounding world which can only be exploited with sophisticated central information processing. Furthermore, vision is an active process due to head and eye movements that enables the animal to change the gaze and actively select objects to investigate in detail. The entire system requires a coordinated coevolution of its parts to work properly. Ray-finned fishes offer a unique opportunity to study the evolution of the visual system due to the high diversity in all of its parts. Here, we are bringing together information on retinal specializations (fovea), central visual centers (brain morphology studies), and eye movements in a large number of ray-finned fishes in a cladistic framework. The nucleus glomerulosus-inferior lobe system is well developed only in Acanthopterygii. A fovea, independent eye movements, and an enlargement of the nucleus glomerulosus-inferior lobe system coevolved at least five times independently within Acanthopterygii. This suggests that the nucleus glomerulosus-inferior lobe system is involved in advanced object recognition which is especially well developed in association with a fovea and independent eye movements. None of the non-Acanthopterygii have a fovea (except for some deep sea fish) or independent eye movements and they also lack important parts of the glomerulosus-inferior lobe system. This suggests that structures for advanced visual object recognition evolved within ray-finned fishes independent of the ones in tetrapods and non-ray-finned fishes as a result of a coevolution of retinal, central, and oculomotor structures.

Variability in the electrophysiological properties of homotypic photoreceptors is widespread and is thought to facilitate functioning under disparate illumination conditions. Compound eyes of insects have three sources of variability: inter-individual, intra-individual, and intra-ommatidial, the latter two overlapping. Here, I explored the causes of variability in Periplaneta americana, a nocturnal insect characterized by highly variable photoreceptor responses. By recording from photoreceptors in dissociated ommatidia, including consecutive recordings from photoreceptors in the same ommatidium (SO), I studied the variability of six properties: whole-cell membrane capacitance (Cm), phototransduction latency, maximal conductance (Gmax) and the slope factor of the sustained Kv current, absolute sensitivity in dim light, and sustained light-induced current (LIC) amplitude in bright light. Coefficient of variation (CV) metrics were used to compare variances in four experimental groups: SO, same animal (SA), all data combined “full sample” (FS), and full sample of all SO recordings (FSSO). For the normally distributed parameters Cm, Gmax, slope factor, and latency, the highest CV values were found in FS and FSSO, intermediate in SA, and the lowest in SO. On average, SO variance accounted for 47% of the full-sample variance in these four parameters. Absolute sensitivity and LIC values were not normally distributed, and the differences in variability between SO and FS/FSSO groups were smaller than for the other four parameters. These results indicate two main sources of variability, intra-ommatidial and inter-individual. Inter-individual variability was investigated by exposing adult cockroaches to constant light or dark for several months. In both groups, the majority of CV measures for the six parameters decreased compared to control, indicating substantial contribution of phenotypic plasticity to inter-individual differences. Analysis of variability of resting potential and elementary voltage responses revealed that resting potential is mainly determined by the sustained Kv conductance, whereas voltage bump amplitude is mainly determined by current bump amplitude and Cm.

In the reverse Stroop task, observers are instructed to ignore the ink color in which a color word is printed (the distractor color) and to respond to the meaning of the color word (the target). Reaction times (RTs) are faster with congruent combinations when the ink color matches the word than with incongruent combinations when the ink color does not match the word. We manipulated the distracting ink color from congruent to incongruent and measured the transition from facilitation to interference. In Experiment 1, we confirmed that this transition could be assessed independently from the contextual influence of particular sets of stimuli and responses, implying that the color space in which interference and facilitation occurs is generalizable. In Experiment 2, we obtained reverse Stroop data for transitions between red and yellow, yellow and green, green and blue, and blue and red, and compared them with independent estimates of color appearance obtained by hue scaling for the same chromaticity samples. We find that the magnitude of the reverse Stroop effect can provide a reliable index of the similarity of color appearance between the distracting chromaticity and the color category represented by the target color word. Moreover, it will allow us to quantify the mapping between the chromaticity space defined at the cone photoreceptors and a cognitive color space defined at an advanced level of neural processing.

An automated, computerized color-vision test was designed to diagnose congenital red–green color-vision defects. The observer viewed a yellow appearing CRT screen. The principle was to measure increment thresholds for three different chromaticities, the background yellow, a red, and a green chromaticity. Spatial and temporal parameters were chosen to favor parvocellular pathway mediation of thresholds. Thresholds for the three test stimuli were estimated by four-alternative forced-choice (4AFC), randomly interleaved staircases. Four 1.5-deg, 4.2 cd/m2 square pedestals were arranged as a 2 × 2 matrix around the center of the display with 15-minute separations. A trial incremented all four squares by 1.0 cd/m2 for 133 ms. One randomly chosen square included an extra increment of a test chromaticity. The observer identified the different appearing square using the cursor. Administration time was ∼5 minutes. Normal trichromats showed clear Sloan notch as defined by log (ΔY/ΔR), whereas red–green color defectives generally showed little or no Sloan notch, indicating that their thresholds were mediated by their luminance system, not by the chromatic system. Data from 107 normal trichromats showed a mean Sloan notch of 0.654 (SD = 0.123). Among 16 color-vision defectives tested (2 protanopes, 1 protanomal, 6 deuteranopes, & 7 deuteranomals), the Sloan notch was between −0.062 and 0.353 for deutans and was <−0.10 for protans. A sufficient number of color-defective observers have not yet been tested to determine whether the test can reliably discriminate between protans and deutans. Nevertheless, the current data show that the test can work as a quick diagnostic procedure (functional trichromatism or dichromatism) of red–green color-vision defect.

We report the results of psychophysical experiments with the so-called barber pole stimulus providing new insights on the neuronal processes underlying the analysis of moving features such as terminators or line-endings. In experiment 1, we show that the perceived direction of a barber pole stimulus, induced by line-ending motion, is highly dependent on the spatial frequency and contrast of the grating stimulus: perceived direction is shifted away from the barber pole illusion at high spatial frequency in a contrast dependent way, suggesting that line-ends are not processed at high spatial scales. In subsequent experiments, we use a contrast adaptation paradigm and a masking paradigm in an attempt to assess the spatial structure and location of the receptive fields that process line-endings. We show that the adapting stimulus that weakens most the barber pole illusion is localized within the barber pole stimulus and not at line-endings' locations. Current models of line-endings' motion processing are discussed in the light of these psychophysical results.

Cyclic GMP has been shown in recent years to directly activate ion channels in bipolar and ganglion cells, and to indirectly regulate coupling between horizontal cells, and between bipolar and amacrine cells. In all of these cases, the effects of cyclic GMP are mimicked by nitric oxide. An increase in calcium concentration stimulates the production of nitric oxide by neuronal and endothelial forms of nitric oxide synthase, which in turn activates soluble guanylate cyclases, enhancing the synthesis of cyclic GMP. Though some effects of nitric oxide do not involve cyclic GMP, the nitric oxide-cyclic GMP cascade is well recognized as a signaling mechanism in brain and other tissues. The widespread occurrence of nitric oxide/cyclic GMP-regulated ion channel activity in retinal neurons raises the possibility that nitric-oxide-sensitive soluble guanylate cyclases play an important role in cell–cell communication, and possibly, synaptic transmission. Immunohistochemical studies have indicated the presence of soluble guanylate cyclase in retinal synaptic layers, but such studies are not suitable for determination of the density or quantitative subcellular distribution of the enzyme. Microanalytical methods involving microdissection of frozen retina also showed the presence of cyclase activity in retinal plexiform layers but these methods did not permit distinction between nitric oxide-sensitive and insensitive cyclases. In this study, we fractionated retinal homogenate into the cytosolic and synaptosomal fractions and investigated the specific activity and distribution of soluble guanylate cyclase and nitric oxide synthase. The results show that both enzymes are present in the synaptosomal fractions derived from inner and outer plexiform layers. The synaptosomal fraction derived from inner retina was highly enriched in cyclase activity. Nitric oxide synthase activity was also higher in the inner than outer retinal synaptosomal fraction. The results suggest that the nitric oxide-cyclic GMP system is operational in both synaptic layers of retina and that it may play a more significant role in the inner retina.

Linking propositions have played an important role in refining our understanding of the relationship between neural activity and perception. Over the last 40 years, visual evoked potentials (VEPs) have been used in many different ways to address questions of the relationship between neural activity and perception. This review organizes and discusses this research within the linking proposition framework developed by Davida Teller, and her colleagues. A series of examples from the VEP literature illustrates each of the five classes of linking propositions originally proposed by Davida Teller. The related concept of the bridge locus—the site at which neural activity can be said to first be proscriptive of perception—is discussed and a suggestion is made that the concept be expanded to include an evolution over time and cortical area.

We have produced and characterized a monoclonal antibody, MH1, which selectively labels rod horizontal cells and Miiller cells in the goldfish retina. Biochemical and tissue distribution studies indicate that MH1 may recognize four out of five classes of intermediate filament proteins in goldfish: vimentin, desmin, glial fibrillary acidic protein (GFAP), and keratin, but not neurofilament. The intermediate filament which is labeled strongest in the retina is vimentin. In the goldfish retina, the only type of horizontal cells recognized by MH1 appear to be rod horizontal cells. This result suggests that the rod horizontal cell, an interneuron, and Miiller (glial) cells share a common antigen: vimentin, which is usually only expressed in mesenchymal origin cells.

The development of rod horizontal cells in the goldfish retina was also studied using MH1. The cells were not labeled by MH1 until 4–6 weeks posthatching, a stage in which the animals are already visually active. MH1 also did not label any horizontal cell in the region close to the ora terminalis in the goldfish retina. These results suggest that either the emergence and maturation of rod horizontal cells occur late during goldfish retinal development or the expression of vimentin itself occurs late in the development of rod horizontal cells.

Ganglion cell activity in response to drifting or alternating stimulus patterns was recorded in an eyecup preparation of the turtle. The stimuli were sinusoidal gratings with various spatial frequencies and spatial phases. Spike histograms of the activity were Fourier-analyzed. The results indicated that the retinal circuitry always functioned in a nonlinear manner. The average, first, and second harmonics of the activity were all a function of stimulus conditions, including spatial phase. Similar outcomes were obtained with full-field (retina-wide) and a small-field (1 mm diameter) stimulation. In the middle of the spatial-frequency range, the ratio of second to first harmonic activity was higher in response to static, contrast-reversing gratings than for drifting, constant-contrast ones. The findings suggest that, as distinct from the usual X, Y, and W segregation of ganglion cell properties in other retinas, there is a single class of cells whose responses seem to have attributes of one or another of the usual categories depending on stimulus characteristics. This may be the result of a degree of stimulus-dependent functional modification in the retinal input circuitry to the turtle ganglion cells.

One type of interplexiform cell (IPC) in the dace retina was discriminated physiologically from other cell classes and identified morphologically with HRP staining. This type responded with slow hyperpolarizing potentials to white diffuse light, and in addition a slow hyperpolarization (after potential) was observed after the cessation of light with relatively high intensities. The latency of the ON phase of the response was always longer than that of the second-order neurons and the amacrine cells. Morphologically, this type of IPC was similar in appearance with the dopaminergic IPC. The conventional synaptic specialization between this type of IPC and horizontal cells was observed, and the IPC was presynaptic.

Human visual evoked potentials (VEPs) were recorded for abrupt 6.25-Hz reversals of 2 c/deg square-wave gratings combining red–green contrast with different levels of luminance contrast. Response characteristics— amplitudes and peak latencies as a function of luminance contrast—were compared for four different pairs of red–green colors and an isochromatic yellow grating. For each of the red–green color pairs, the plots of VEP amplitudes and latencies were nonsymmetrical with respect to isoluminance. The amplitude dropped to a minimum within a region of rapid phase change, at a different contrast for each color pair but always at a luminance contrast for which the greener color had the higher luminance. When the contrast-response curve for each of the four red–green pairs was modeled by a simple |CL − CM| opponency of L- and M-cone contrast using a fixed CL/CM weighting ratio of about two, there was a close correspondence between the contrast giving a null in the modeled response and that giving a minimum in the VEP amplitude. So for the stimulus parameters applied here, the reversal VEP appeared to be dominated by L/M-opponent response contributions for which the signed CL/CM-cone weighting ratio was close to a value of minus two rather than to a value of minus one, which is characteristic of the psychophysical red–green detection mechanism and representative of CL/CM weighting ratios for precortical cells in the parvocellular pathway.

Visual localization was studied by flashing small stimuli on a green background and requiring observers to press keys to indicate whether the stimulus appeared to the left or right of fixation. The results suggest that, for small (0.25 deg) briefly flashed (17 ms) stimuli at an eccentric location (10 deg), color contrast is not useable and localization presumably must rely on the magnocellular pathway. When stimulus size and duration were increased at 10-deg eccentricity, isochromatic stimuli could be localized at less than 10% luminance contrast (again suggesting use of the magnocellular high sensitivity luminance-contrast system), but isoluminant color-contrast stimuli could also be localized (suggesting use of the color-contrast sensitive parvocellular system). Thus, the results indicate that, dependent on stimulus conditions, both magnocellular and parvocellular pathways were utilized by normal observers in this localization task.

The aim of this project was to investigate the interaction between Y retinal ganglion cells and the cells of the dorsal lateral geniculate nucleus (LGNd) of the cat, with particular reference to center-surround antagonism and intrageniculate inhibition. Responses of cells in the LGNd were studied by stimulating the retina with spots of light of constant contrast but varying size. The peak discharges of nonlagged X (XN) cells were strongly suppressed with increase in spot size but the responses of lagged X (XL) cells and nonlagged Y (YN) cells were inhibited much less strongly. The effect of the Y system on these responses was examined by producing a selective block of conduction in Y fibers in one optic nerve by means of a pressure cuff (Y-blocking). These effects were assessed by measuring the peak discharge rates and by calculation of a “peak suppression index.” Y-blocking had no significant effect on the peak suppression index of XL, cells in either lamina or on YN cells in the normal (not Y-blocked) lamina but had significant effects on the responses of XN cells, causing a decrease in peak suppression index, both for cells in laminae receiving their principal excitatory input from the Y-blocked eye (both lamina A and lamina A1 ) as well as those in lamina A (but not lamina A1 ) receiving their excitatory input from the normal eye. These effects were obtained with relatively large spots of light. Thus Y optic fibers have both intralaminar (monocular) and interlaminar (binocular) inhibitory effects on XN cells. In addition to these suppressive effects, the experiments also show that ipsilaterally projecting Y fibers have facilitatory effects on XN cells in lamina A when small spots of light, about optimal size for the XN cell, are used. These results suggest that the Y system plays a powerful role in shaping the responses of XN cells, possibly enhancing visual acuity.

Naturally occurring cell death has been hypothesized to sculpt various features of the organization of the mature visual pathways, including the recent proposal that the selective elimination of ganglion cells in the temporal retina shapes the formation of decussation patterns. Through a class-specific interocular competition, ganglion cells in the two temporal hemiretinae are selectively lost to produce the decussation patterns characteristic of each individual cell class (Leventhal et al., 1988). The present study has tested this hypothesis by asking whether the removal of one retina in newborn ferrets, which should disrupt binocular interactions at the level of the terminals, alters the decussation pattern of the alpha cells, a cell class that is entirely decussating in the normal adult ferret. Enucleation on the day of birth was found to increase the uncrossed projection by ≈50%, but not a single uncrossed alpha cell was found in the temporal retina. Either alpha cells never project ipsilaterally during development, or if they do, they cannot be rescued by early enucleation. While naturally occurring cell death plays many roles during development, creating the decussation pattern of the ferreth's alpha cell class via a binocular competition at the level of the targets is unlikely to be one of them.

MacDRP is a Macintosh application that calculates Density Recovery Profiles.

Although the retinotectal projection of goldfish has long been known to have a high degree of retinotopic order, the structural basis for this in terms of the precise positioning of axonal arbors from neighboring retinal ganglion cells has not been determined. In studying this, a small number of neighboring retinal ganglion cells was selectively labeled by a microinjection of DiI into the retina. Following axoplasmic transport for several days, the tectum was removed and flat-mounted for fluorescence microscopy. The injection labeled a small number of axons and their terminal arbors which ranged in size from 108 × 134 μm to 394 × 331 μm with a mean of 233 × 219 μm. This mean size corresponds to about 1/15 of the length of one tectal axis. Although individual arbors labeled from one small retinal injection were always observed near the same retinotopic position, they were almost never coextensive. Overlap between pairs of arbors along the lines of projection perpendicular to the tectal surface averaged 57% of the area of a single arbor. These results indicate that neighboring retinal ganglion cells do not converge onto the same locus but instead project as a continuous retinotopic array of partially overlapping terminal fields.

This paper responds to Philipona and O'Regan (2006), which attempts to account for certain color phenomena by appeal to singularities in the space of “accessible information” in the light striking the retina. Three points are discussed. First, it is unclear what the empirical significance/import is of the mathematical analysis of the data regarding the accessible information in the light. Second, the singularity index employed in the study is both mathematically and empirically faulty. Third, the connection drawn between their findings and some data from the World Color Survey is lacking in quantitative analysis in places where it is needed. The difficulties raised prevent Philipona & O'Regan's conclusions from being accepted.

This study was performed in order to establish whether selective depletion of somatostatin (SS) in the rat primary visual cortex obtained by cysteamine (CSH) administration results in changes of visual evoked potentials (VEPs). VEPs in response to a contrast reversal (0.5 Hz) of an optimal sinusoidal grating (0.1 cycle/deg, contrast 90%, mean luminance 15 cd/m2) were recorded from different layers of the binocular portion of the primary visual cortex of anesthetized rats with saline injection as well as before and after CSH treatment (90 mg/kg, s.c). VEPs of CSH treated rats, as compared to those obtained either in saline-injected animals or before drug administration, are reduced in amplitude at intermediate cortical layers whereas they are increased at deeper layers. VEP changes depend on CSH treatment and not on the extended anesthesia since no alterations in the VEP profile can be observed in saline-injected animals maintained in the same experimental condition. Forty-eight hours following CSH treatment, the VEP profile is comparable to that of saline-injected animals. Immunocytochemical analysis of the visual cortex of rats recorded 7 h after CSH treatment shows a 20–30% reduction in the number of SS-containing cortical cells. The highest reduction can be observed in cortical layer 5 although a significant decrease is also found in layers 2–3. In contrast, the pattern of SS immunoreactivity of the visual cortex of rats recorded 48 h after CSH administration is similar to that obtained in control conditions. These results indicate that a selective toxin for somatostatinergic systems induces a transient decrease of SS-containing cell number in selected cortical layers. Accordingly, CSH can serve as a useful pharmacological tool for the study of somatostatinergic function in the rat visual cortex since changes in VEPs can be related to a reduction of somatostatinergic neurons associated to CSH treatment. In particular, the present results suggest that one of the possible actions of somatostatinergic neurons in the rat visual cortex is to modulate the excitatory-inhibitory balance.
