We have studied the influence of chromatic adaptation upon the perceived visual position of a test stimulus using a Vernier alignment task. Maximum and minimum offsets in spatial position are generated when the adapting and test stimuli lie on the same and orthogonal axes in MBDKL color space, respectively. When the test stimuli lie on intermediate color axes, the measured positional shifts decrease as a function of the angular separation in color space (φ) from the adapting stimulus. At low stimulus contrasts, these shifts follow a sinusoidal function of φ and exhibit broad chromatic tuning and can be accounted for by a model in which the centroid is extracted from the linear combination of after-image, formed by the adapting stimulus, and the test stimulus. Such linear, broadband behavior is consistent with the response properties of chromatic neurons in the precortical visual pathway. At high contrast, and when adaptation gets closer to the S/(L+M) axis, the tuning functions become narrower and require sinusoids raised to increasingly higher exponents in order to describe the data. This narrowing of chromatic tuning is consistent with the tuning properties of chromatic neurons in the striate cortex, and implies the operation of a nonlinear mechanism in the combination of cone outputs.

This study investigated chromatic induction from inhomogeneous background patterns. Previous work showed that a background pattern detected by only S cones induced strong color shifts in a nearby test area (Monnier & Shevell, 2003). In that work, the S-cone patterns were composed with constant L- and M-cone stimulation over the entire background; in terms of L and M cones, therefore, the background was uniform. S-cone stimulation was varied over space to produce S-cone-isolated background patterns. These S-cone patterns, however, established spatial structure (the pattern) at both the receptoral level (S-cone stimulation) and the postreceptoral level (S/(L+M)). Here, these two levels of pattern representation were unconfounded to determine whether color shifts induced by S-cone patterns were due to spatial structure within an S-cone-specific neural pathway versus a pathway that combines responses from S cones and other cone types (e.g. S/(L+M)). The results showed that the induced color shifts were mediated by signals within a pathway that combines responses from multiple cone types. These results are consistent with a +s/−s spatially antagonistic neural receptive field, which is found in some neurons in V1 and V2.

Studies of functional variability in the compound eyes of flies reveal superior temporal resolution of photoreceptors from the frontal areas that mediate binocular vision, and in males mate recognition and pursuit. However, the mechanisms underlying differences in performance are not known. Here, we investigated properties of hover fly Volucella pellucens photoreceptors from two regions of the retina, the frontal-dorsal “love spot” and the lateral one. Morphologically, the microvilli of the frontal-dorsal photoreceptors were relatively few in number per rhabdomere cross-section, short and narrow. In electrophysiological experiments involving stimulation with prolonged white-noise and natural time intensity series, frontal-dorsal photoreceptors demonstrated comparatively high corner frequencies and information rates. Investigation of possible mechanisms responsible for their superior performance revealed significant differences in the properties of quantum bumps, and, unexpectedly, relatively high absolute sensitivity of the frontal-dorsal photoreceptors. Analysis of light adaptation indicated that photoreceptors from two regions adapt similarly but because frontal-dorsal photoreceptors were depolarized much stronger by the same stimuli than the lateral photoreceptors, they reached a deeper state of adaptation associated with higher corner frequencies of light response. Recordings from the photoreceptor axons were characterized by spike-like events that can significantly expand the frequency response range. Seamless integration of spikes into the graded voltage responses was enabled by light adaptation mechanisms that accelerate kinetics and decrease duration of depolarizing light response transients.

At the first retinal synapse, horizontal cells (HCs) contact both photoreceptor terminals and bipolar cell dendrites, modulating information transfer between these two cell types to enhance spatial contrast and mediate color opponency. The synaptic mechanisms through which these modulations occur are still debated. The initial hypothesis of a GABAergic feedback from HCs to cones has been challenged by pharmacological inconsistencies. Surround antagonism has been demonstrated to occur via a modulation of cone calcium channels through ephaptic signaling and pH changes in the synaptic cleft. GABAergic transmission between HCs and cones has been reported in some lower vertebrates, like the turtle and tiger salamander. In these reports, it was revealed that GABA is released from HCs through reverse transport and target GABA receptors are located at the cone terminals. In mammalian retinas, there is growing evidence that HCs can release GABA through conventional vesicular transmission, acting both on autaptic GABA receptors and on receptors expressed at the dendritic tips of the bipolar cells. The presence of GABA receptors on mammalian cone terminals remains equivocal. Here, we looked specifically for functional GABA receptors in mouse photoreceptors by recording in the whole-cell or amphotericin/gramicidin-perforated patch clamp configurations. Cones could be differentiated from rods through morphological criteria. Local GABA applications evoked a Cl− current in cones but not in rods. It was blocked by the GABAA receptor antagonist bicuculline methiodide and unaffected by the GABAC receptor antagonist TPMPA [(1,2,5,6-tetrahydropyridin-4-yl)methylphosphinic acid]. The voltage dependency of the current amplitude was as expected from a direct action of GABA on cone pedicles but not from an indirect modulation of cone currents following the activation of the GABA receptors of HCs. This supports a direct role of GABA released from HCs in the control of cone activity in the mouse retina.

The patches in primary visual cortex constitute hot spots of metabolic activity, manifested by enhanced levels of cytochrome oxidase (CO) activity. They are also labeled preferentially by immunostaining for glutamic acid decarboxylase (GAD), γ-aminobutyric acid (GABA), and parvalbumin. However, calbindin shows stronger immunoreactivity outside patches. In light of this discrepancy, the distribution of the vesicular GABA transporter (VGAT) was examined in striate cortex of two normal macaques. VGAT immunoreactivity was strongest in layers 4B, 4Cα, and 5. In tangential sections, the distribution of CO, GAD, and VGAT was compared in layer 2/3. There was a close match between all three labels. This finding indicates that GABA synthesis is enriched in patches, and that inhibitory synapses are more active in patches than interpatches.

Previous neuroimaging studies demonstrated that retinal detachment (RD) subjects were associated with abnormal spontaneous brain activities; however, whether the altered interhemispheric functional connectivity (FC) occurred in RD patients remains unknown. The current study tried to explore the alternations of interhemispheric FC of the whole brain in unilateral RD patients using the voxel-mirrored homotopic connectivity (VMHC) method and their connections to clinical features. Methods: We recruited 30 patients with RD (16 males and 14 females) and 30 healthy controls (HCs) (16 males and 14 females) whose age and sex were closely matched. All subjects underwent the rs-fMRI scans. The VMHC method was applied to directly assess the hemispheres’ functional interaction. The VMHC in these brain areas, which could be used as biomarkers to differentiate RD from HC, was identified by the receiver operating characteristic (ROC) curve analyses. The relations between these patients’ clinical features and their mean VMHC signal values in multiple brain regions were calculated by Pearson correlation analysis. Results: RD patients had significantly lower VMHC values than HCs in the bilateral occipital lobe (Brodmann areas, BA 18), bilateral superior temporal gyrus (BA 39), and bilateral cuneus (BA 19). Moreover, the mean VMHC signal values of the bilateral cuneus were in positive correlation with the duration of the RD (r = 0.446, P = 0.013). Conclusion: Our results provided an evidence of disturbed interhemispheric FC in the visual area occurred in RD patients, which might provide some useful information to understand the neural mechanism of RD patients with acute vision loss. Furthermore, the VMHC values might indicate the progress of the RD.

We tested the hypothesis that gamma-aminobutyric acid (GABA) is the transmitter released by barnacle photoreceptors onto postsynaptic cells (I-cells). GABA was applied to I-cells either by superfusion or by ejecting it with pressure from a pipette positioned close to the I-cell's soma. The I-cell's response to GABA was compared with its response to light (i.e. to the photoreceptors' transmitter) by recording intracellularly from its soma. Bath-applied (100 µm to 10 mM) and pressure-applied GABA (10 mM in pipette) hyperpolarizes I-cells by increasing their conductance, as does the photoreceptors' transmitter. The response to pressure-applied GABA consists of two components; both persist when Co2+ or Cd2+ are added to the saline to block synaptic transmission in the preparation, indicating that GABA affects the I-cell directly rather than affecting a presynaptic cell. GABA hyperpolarizes the I-cell when applied to the cell over the soma and ipsilateral arbor or over the contralateral arbor. The I-cells' responses to GABA and to light both depend on extracellular K+ and are affected by changes in intracellular and extracellular Cl−. However, picrotoxin and β-guanidinopropionic acid block the response to pressure-applied GABA but do not block the response to light even at an order of magnitude higher concentration. Thus, GABA is not likely to be the transmitter that causes the hyperpolarizing response of the I-cell. It may be a neuromodulator or the transmitter of an unknown input to the I-cell.

The strength of the flanking inhibitory regions of the receptive fields of fly lamina cells (LMC) decreases as the mean luminance is lowered. Simultaneously, the biphasic temporal flash (impulse) response of the lamina cells becomes monophasic on lowering luminance. For a moving-edge stimulus at high mean luminance, this implies that the spatial integration by the larnina cell yields a temporal waveform which is congruent to the waveform of the temporal impulse response of the lamina cell. In other words, the temporal waveform generated by the moving edge is matched to the temporal waveform most preferred by the lamina cell. The edge is the stimulus causing the largest amplitude response at high (above 1 cd/m2) levels of luminance. On lowering luminance, the now monophasic nature of the spatial and temporal impulse responses of the lamina gives a preference not for the edges but for the center of a uniform region. We describe this theory and its behavioral corroboration in walking flies (Lucilia cuprina).

Pupillary area was measured in urethane-anesthetized cats as a function of retinal illuminance. When appropriate corrections are made for differences in experimental procedures, it was found that the pupillary response of the urethane-anesthetized cat's eyes to light was basically unchanged from that of the alert behaving cat. This preparation may therefore be a very satisfactory one in which to study the pupillary response pathway in a higher mammal.

The spatial region involved in vernier acuity (integration region) was behaviorally determined in cats and compared with published human data and cortical scaling parameters for the two species. Integration region was estimated from vernier thresholds obtained with: (1) gaps of various sizes between the contiguous ends of the offset contours (gap effects); and (2) irrelevant contours placed at various distances from the vernier offset (interference effects). The gap studies showed that thresholds decreased as the gap grew from 0 to 4–7 min but then increased progressively with separations greater than 7 min. In the interference studies, thresholds were increased 2–6 times when the irrelevant contours were located within 40 min of the offset. Comparing these data with available human measures showed large species differences. Converting the interference distances for both species into cortical distances using published estimates of cortical magnification factor did not resolve the discrepancy. However, a comparison of the cat data with published human data, taken at an eccentricity at which the estimated human cortical magnification factor equals that of the cat, i.e. 7 deg, showed the effects of gap size and interference distance on vernier thresholds to be remarkably similar between the two species. This similarity supports the hypothesis that positional acuity depends upon mechanisms in striate cortex, and also suggests that such acuity may be mediated by a generic neural mechanism common to mammals.

Receptive fields of ganglion cells have been studied in cats possessing a chronic, arrested lesion of central retinal degeneration. Lesions were characterized by an ophthalmoscopically sharp border separating apparently normal retina from the region of the lesion. Under direct ophthalmoscopic guidance, a succession of recordings was obtained from ganglion cells having cell bodies at various positions relative to the lesion. Cells located more than 1 deg outside the ophthalmoscopic border had normal visual sensitivity as assessed by area-threshold experiments. Inside the lesion cells within 1 deg of the border had reduced sensitivity which often precluded functional classification by the usual visual tests. Ganglion cells located more than 1 deg inside the border of large lesions were blind and some had abnormal patterns of maintained discharge of action potentials. Nevertheless, the antidromic latencies of these blind cells fell into the familiar conduction groups (T1/T2/T3). Receptive-field maps of cells near the border of the lesion often appeared truncated, with the missing portion of the field covered by the lesion. These observations were consistent with the abnormal form of area-thresholdcurves. Altlhough the responsiveness of cells near the lesion was abnormally low for grating stimuli, cutoff spatial frequency and orientation bias of these cells were within normal limits.

A psychophysical procedure, classical conditioning of respiration, was used to measure contrast sensitivity to positive- and negative-contrast discs (8-deg diameter) in goldfish after crushing one optic nerve intraorbitally. In five out of six fish, sensitivity recovered to normal. Recovery times for negative contrasts were significantly shorter than for positive contrasts. The average times postcrush of initial responding to negative and positive contrasts were 23.8 and 30.6 days, and for threshold to come within 0.5 log unit of control values was 29.8 and 39.8 days, respectively. Thereafter, recovery to normal sensitivity was significantly faster for positive contrasts. These results parallel prior observations of neural activity in tectum after optic nerve crush: an early phase of OFF responding followed by a more sudden recovery of ON responding.

Although a number of global mechanisms have been proposed over the years that explain how crabs might separate the rotational and translational components of their optic flow field, there has been no evidence to date that local mechanisms such as motion parallax are used in this separation. We describe here a study that takes advantage of a recently developed suite of computer-generated visual stimuli that creates a three-dimensional world surrounding the crab in which we can simulate translational and rotational optic flow. We show that, while motion parallax is not the only mechanism used in flow-field separation, it does play a role in the recognition of translational optic flow fields in that, under conditions of low overall light intensity and low contrast ratio when crabs find the distinction between rotation and translation harder, smaller eye movements occur in response to translation when motion parallax cues are present than when they are absent. Thus, motion parallax is one of many cues that crabs use to separate rotational and translational optic flow by showing compensatory eye movements to only the former.

The interdependence of the development of the eye and oculomotor system during embryogenesis is currently unclear. The occurrence of clinical anophthalmia, where the globe fails to develop, permits us to study the effects this has on the development of the complex neuromuscular system controlling eye movements. In this study, we use very high-resolution T2-weighted imaging in five anophthalmic subjects to visualize the extraocular muscles and the cranial nerves that innervate them. The subjects differed in the presence or absence of the optic nerve, the abducens nerve, and the extraocular muscles, reflecting differences in the underlying disruption to the eye’s morphogenetic pathway. The oculomotor nerve was present in all anophthalmic subjects and only slightly reduced in size compared to measurements in sighted controls. As might be expected, the presence of rudimentary eye-like structures in the socket appeared to correlate with development and persistence of the extraocular muscles in some cases. Our study supports in part the concept of an initial independence of muscle development, with its maintenance subject to the presence of these eye-like structures.

The resistances of the horizontal cell syncytium in the vertebrate retina are modulated in a time-dependent fashion during light stimulation. Therefore, the spatial properties of horizontal cells are expected to change with time after the illumination conditions are altered. This study was designed to investigate time- and intensity-dependent changes in the receptive-field properties of L1-type horizontal cells in the turtle Mauremys caspica. Photoresponses were elicited by monochromatic (650 nm) light stimuli of 2-s duration covering retinal spots of different radii. The length constants were derived from the relationships between amplitude and spot radius that were constructed for different time intervals after onset of the light stimulus. For a given stimulus intensity, the length constant transiently increased to a peak value and then slowly recovered to a plateau level. When the length constant was compared to the amplitude of the response to full-field illumination for the entire duration of the light stimulus, an ellipse-like curve was obtained indicating that for a given membrane potential, two different values of the length constant could be obtained. Dopamine considerably reduced the size of the receptive fields but did not affect the time-dependent changes in the length constant. These results indicate that changes in the membrane resistance underlie short-term modulation of the receptive-field properties of turtle L1-type horizontal cells after onset of a light stimulus.

In the 1990s, seminal work from Newsome and colleagues made it possible to study the neuronal mechanisms of simple perceptual decisions. The key strength of this work was the clear and direct link between neuronal activity and choice processes. Since then, a great deal of research has extended these initial discoveries to more complex forms of decision making, with the goal of bringing the same strength of linkage between neural and psychological processes. Here, we discuss the progress of two such research programs, namely our own, that are aimed at understanding memory-guided decisions and reward-guided decisions. These problems differ in the relevant brain areas, in the progress that has been achieved, and in the extent of broader understanding achieved so far. However, they are unified by the use of theoretical insights about how to link neuronal activity to decisions.

The isolated retina of the goldfish has proven a valuable resource for studying the variability of firing of retinal ganglion cells. Three major areas of study are considered here: the variability of maintained discharges, the correlated firing of neighboring ganglion cells, and the variability of responses to light. The sources of variability, its relationship to retinal processing, and its possible functional role in perception are examined through these three aspects of variability. The results are related to similar studies in mammals (mainly cats). This retrospective is biased toward my studies over 30 years.

The notion of a set of neurons that form a “bridge locus” serving as the immediate substrate of visual perception is examined in the light of evidence on the architecture of the visual pathway, of current thinking about perceptual representations, and of the basis of perceptual awareness. The bridge locus is likely to be part of a tangled web of representations, and this complexity raises the question of whether another scheme that relies less on geography might offer a better framework. The bridge locus bears a close relationship to the neural correlate of consciousness (NCC), and like the NCC may be a concept which is no longer precise enough to provide a useful basis for reasoning about the relationship between brain activity and perceptual experience.

Although there are many glutamate receptors in the retina, 2-amino-4-phosphonobutyrate (L-AP4) is an agonist that acts selectively at metabotropic glutamate receptors (mGluR6) of ON bipolar cells. We explored the properties of agonists that activate this receptor. The effects of various glutamate analogs on the b-wave of the electroretinogram (ERG) were used as a measure of their activity. Conformational comparisons among agonists suggest that ligands in an extended conformation preferentially bind to the ON bipolar synaptic receptor. But this property is insufficient to explain the selectivity of mGluR6 because some inactive glutamate analogs could also match this extended conformation. Comparative molecular field analysis (CoMFA) was used to compare the electrostatic and steric potentials of agonists with their action at the ON bipolar synapse. Steric potentials beneath a plane defined by the three putative binding sites plays a key role in determining agonist activity. The CoMFA model was used to predict the activity of glutamate analogs and correlations between predicted and measured activity support the model.
