The citrus mealybug, Planococcus citri, is a highly destructive pest of citrus, occurring only in the aerial parts of plants. Humidity will be one of the key factors to consider when using entomopathogenic nematodes (EPN) as biological control agents. Different adjuvants can be added to suspensions of EPNs, to improve control as a foliar application. An aqueous suspension containing Heterorhabditis zealandica and 0.3% Zeba® significantly increased P.citri mortality by 22% at 80% relative humidity (RH) with a temperature cycle starting at 22°C for 14 h and 11°C for 11 h. The same polymer formulation was tested for Steinernema yirgalemense and mortality of P. citri increased by 21% at 60% RH and by 27% at 80% RH. The addition of Nu-Film-P® and Zeba® to H. zealandica suspensions did not significantly retard application run-off on citrus leaves. The combination of Nu-Film-P® and Zeba®, however, was able to significantly retard sedimentation, increasing the average number of nematodes deposited on 2-cm2 leaf discs by 10 nematodes. In an aqueous suspension, nematodes settle rapidly to the bottom, resulting in an uneven distribution of nematodes. Xanthan gum, at a concentration of 0.2%, was highly effective at retarding sedimentation, with 72% of the initial nematode number still in suspension after 1 h. Zeba®, at a concentration of 0.3%, despite not being as effective as Xanthan gum, nevertheless still retarded sedimentation significantly. This is the first report of the potential of Nu-Film-P® and Zeba® to improve EPN performance against P. citri when used above ground in citrus orchards.

Confusion exists over the species status and host-specificity of the tapeworm Rodentolepis (= Hymenolepis) nana. It has been described as one species, R. nana, found in both humans and rodents. Others have identified a subspecies; R. nana var. fraterna, describing it as morphologically identical to the human form but only found in rodents. The species present in Australian communities has never been identified with certainty. Fifty one human isolates of Rodentolepis (= Hymenolepis) nana were orally inoculated into Swiss Q, BALB/c, A/J, CBA/CAH and nude (hypothymic) BALB/c mice, Fischer 344 and Wistar rats and specific pathogen free (SPF) hamsters. Twenty four human isolates of R. nana were cross-tested in flour beetles, Tribolium confusum. No adult worms were obtained from mice, rats or hamsters, even when immunosuppressed with cortisone acetate. Only one of the 24 samples developed to the cysticercoid stage in T. confusum; however, when inoculated into laboratory mice the cysticercoids failed to develop into adult worms. The large sample size used in this study, and the range of techniques employed for extraction and preparation of eggs provide a comprehensive test of the hypothesis that the human strain of R. nana is essentially non-infective to rodents.

Entomopathogenic nematodes were isolated from soil samples collected during three separate surveys, two in the United Kingdom and one in the Netherlands. The nematodes were identified by their restriction fragment length polymorphisms on hybridization of Southern blots with a ribosomal DNA repeat unit clone. A total of eight steinernematid species/RFLP types and two heterorhabditids were isolated. The distributions of the majority of these species/RFLP types were significantly different between the three surveys and some of the species displayed a close association with certain habitat types.

Sera of 908 blood donors, average age 35 years, living in towns and villages of the Slovak Republic were tested for Toxocara canis antibodies using an ELISA-IgG test. The seroprevalence in the healthy population was estimated at 13·65%. Antibodies were predominantly demonstrated in the sera from persons living in rural communities (17·09%) compared to the sera investigated from urban communities (11·82%). Women had a significantly higher seropositivity than men. The serological response in 2703 sera from patients suspected of toxocarosis was compared with their clinical manifestation, sex and age. Toxocara antibodies were found in 27·41% of suspected patients. Although the seroprevalence in adults was significantly higher among women than men, in children, boys had a higher seroprevalence than girls. The main clinical manifestations were leucocytosis and eosinophilia (46%), ocular disorders (36%), lung, liver and neurological disorders (26%). The most severe forms of this disease were observed in 2 to 5-year-old children with geophagy.

Out of a total of 282 soil samples obtained from public areas, 58 samples contained Toxocara spp. eggs. This gave an overall prevalence of 20.6%. Highway rest areas were contaminated and childrens sand boxes had the highest percentage of recovery of all.

Eosinophil counts, interleukin 5 (IL-5) concentrations in peripheral blood and larval recovery after infection with Toxocara canis were compared between IL-5 transgenic (Tg) and nontransgenic counterparts, C3H/HeN mice. In Tg mice characterized by a constant high level of peripheral eosinophils, eosinophils in peripheral blood fell to the lowest level on day 4 of a T. canis infection and then returned to the preinfection level on day 14. Serum IL-5 level fell to the lowest level on day 4 of infection, then recovered rapidly and peaked on day 14 postinfection. In contrast, the eosinophil and IL-5 levels in the peripheral blood peaked on days 11 and 7 of infection, respectively in C3H/HeN mice. The degree of eosinophil infiltration into the lung 4 days after infection was far more pronounced in Tg than C3H/HeN mice. The highest number of larvae recovered from the lungs of infected Tg and C3H/HeN mice occurred on day 4 postinfection. Strains of Tg and C3H/HeN mice vaccinated with excretory and secretory (ES) antigens of T. canis larvae were infected with T. canis and the recovery of larvae on day 21 analysed. There were no significant differences in the mean number of larvae recovered from nonvaccinated Tg and C3H/HeN mice or vaccinated Tg and C3H/HeN mice. However, significant differences were demonstrated in the mean total number of larvae recovered from vaccinated and nonvaccinated Tg or C3H/HeN mice. These results suggest that immunocompetent cells other than eosinophils may play a significant role in the expulsion and killing of T. canis larvae in infected mice.

In the Chambezi-Luapula area of the Northern Province of Northern Rhodesia, 2,575 Africans were examined for intestinal helminths and 2,017 for urinary helminths. The examinations were carried out in situ in the native villages in widespread localities representing different topographical parts of the area.

The average infection rates of the helminthic infections found were as follows: hookworm, 52.2%; Strongyloides, 13.3%; Ascaris lumbricoides, 3.7%; Enterobius vermicularis, 0.85%; Trichuris trichiura, 0.3%; Trichostrongylus sp., 0.1%; Schistosoma haematobium, 14.7%; S. mansoni, 6.99%; Hymenolepis nana, 0.09%; H. diminuta, 0.04%; Taenia sp., 0.04%.

The survey revealed regions or foci of relatively high and relatively low incidence in the case of each of the major species. The relationship of this to topographical and other factors is discussed.

Examination of 459 Africans for Mf. bancrofti revealed three positives, but none of these had definitely contracted the infection in N. Rhodesia. Mf. Perstans was found in 5.4% of those examined; this infection is endemic in the country.

Praziquantel (PZQ) is now widely used for the treatment of human schistosomiasis. However, in recent years, there has been a growing concern about the resistance of Schistosoma to PZQ. The mechanisms of PZQ action against Schistosoma and resistance of Schistosoma to PZQ are poorly understood. Here, we report differential susceptibilities to PZQ between male and female cercariae in the PZQ-susceptible and PZQ-resistant isolates of Schistosoma mansoni, using tail loss as a measurement of PZQ action. The miracidia were collected by hatching eggs collected from faeces of infected mice. Single-sex cercaria lines were made by infecting a single Biomphalaria glabrata snail with a single miracidium. The sex of each single-sex cercaria line was identified by a direct W1-specific polymerase chain reaction (PCR) technique. Single-sex cercariae of two isolates were exposed to four different concentrations of PZQ, respectively. The tail shedding of cercariae was observed under a dissecting microscope for five time points up to 100 min after adding PZQ. The results showed that male cercariae have higher tail-shedding rates than that of female cercariae when PZQ-susceptible isolates of S. mansoni are exposed to the same concentration of PZQ. But this phenomenon was not observed in the PZQ-resistant isolates. This sexual differential resistance phenomenon of S. mansoni suggests that resistance to PZQ is induced by decreasing the PZQ susceptibility of male worms. The experiment described here may also be useful for developing tests to detect PZQ resistance in the field.

A systematic review was conducted focusing on the distribution of Fasciola species and their snail intermediate hosts (IHs) in East and Southern Africa. The reviewed literature showed that both Fasciola hepatica and F. gigantica are present in East and Southern Africa, and infect a wide range of domestic and wild ruminants. Fasciola gigantica was reported in six East African and five Southern African countries, where Radix natalensis (found in low altitudes) was reported to be the main IH. Fasciola hepatica was reported in Tanzania and Ethiopia (East Africa), and in South Africa and Zimbabwe (Southern Africa), where Galba truncatula (found in high altitudes) was documented as the IH in all countries except in Zimbabwe. Both Fasciola species were documented in Tanzania, Ethiopia, Zimbabwe and South Africa. An overlap of the two was observed in areas with an intermediate altitude in Ethiopia and South Africa, where Pseudosuccinea columella was widespread and assumed to transmit both species. Pseudosuccinea columella has been reported in South Africa and Namibia, and proven to transmit F. gigantica in South Africa; its role in Namibia in the transmission of Fasciola species has not been reported. Other lymnaeid species such as R. rubiginosa were reported in South Africa, and R. auricularia in South Africa and Botswana; their role in the transmission of Fasciola species has not been proven. Future studies should aim to determine the role of P. columella in the geographical spread of the two species in East and Southern African countries.

Adult Strongyloides ratti were expelled from the small intestine of rats starting 14—18 days after a primary infection. In a secondary infection very few adult worms developed and most of these were expelled before day 14. At the time of expulsion the worms migrated posteriorly in the intestine and their size decreased.

The life-cycle of Apatemon (A.) gracilis was completed in the laboratory. The snail host is Lymnaeaperegra (Müller). The cercaria is redescribed from a wide range of material. Metacercariae were found in naturally infected rainbow trout (Salmo gairdneri Richardson), three-spined sticklebacks (Gasterosteus aculeatiis L.) and stone loach (Nemacheilus barbatulus (L.)) from Scotland and in three-spined sticklebacks from Iceland. In trout, most metacercariae were found in the pericardial cavity, in sticklebacks, the eye, and in loach, the body cavity. In infection experiments, cercariae from naturally infected Scottish snails developed in threespined sticklebacks, rainbow trout and brown trout (Salmo trutta L.). Under experimental conditions cercariae did not penetrate stone loach, although this species is naturally infected with A gracilis. The phenomenon of fish host specificity is briefly discussed. Development of the metacercaria is described. Excystation of metacercarial cysts with pepsin and trypsin solutions is unlike that reported for any other digenean; the contents of the cyst appear to be under pressure. In pepsin, layers of the cyst wall peel back from one end. When transferred to trypsin, one pole of the cyst ruptures and the worm is forcibly expelled.

A case of cystic echinococcosis (CE) in a domestic cat is described from Saint Petersburg, Russia. Ultrasonography showed numerous cysts with hyperechoic walls and anechoic contents within the cat's abdominal cavity. Molecular identification based on mitochondrial DNA genes indicated that the causative agent was Echinococcus granulosus sensu stricto (G1 strain). This is the first report of CE in a cat caused by E. granulosus sensu stricto with molecular confirmation.

The toxicity of cadmium to a population of Lymnaea peregra and L. stagnalis naturally infected with a range of digeneans and collected from a number of sites in the lower Thames Valley, UK was investigated. Lymnaeid snails were exposed to 100 μg l-1 cadmium and the effects on host survival and emergence of cercariae recorded. Overwintered L. peregra, but not L. stagnalis, showed significantly reduced survival compared to seasonally infected snails, i.e. snails which have acquired an infection during the spring or summer. A significant increase in survival with increasing snail size was demonstrated for L. stagnalis and for seasonally infected L. peregra only. Only L. stagnalis infected with Diplostomum spathaceum and L. peregra infected individually with D. spathaceum, Sanguinicola inermis, Echinoparyphium recurvatum and Notocotylus attenuatus demonstrated a significantly reduced survival compared to laboratory-bred controls. The exposure of L. stagnalis to cadmium resulted in a significant reduction in the emergence of D. spathaceum over a 5-day period but cadmium-exposed L. peregra showed no difference in the emergence of E. recurvatum cercariae over a 3-day exposure period. The mechanisms and importance of metal toxicity to snail–digenean interactions are discussed.

The previous observations of the author and others on the development ot Cysticercus fasciolaris and C. pisiformis have been confirmed and amplified by experimental studies, and the developmental processes of the two forms are compared. While definite information has been obtained on the age of the various developmental stages, there is considerable variation due to the subtleties of the host-parasite relationship.

The study of the development of the rostellar hooks of taeniids has been extended by the use of differential staining, histochemical tests and polarized light, and new facts, such as the chemical and optical differences existing between cuticle and hooklets and between blade and base of hooks, have been brought to light.

Forty-three specimens of Leptodactylus podicipinus (Anura: Leptodactylidae) were collected in the south-eastern Pantanal, municipality of Corumbá, State of Mato Grosso do Sul, Brazil in February and July 2007, and examined for endoparasites. Forty (93%) specimens were infected with at least one helminth species. The predominant parasites were nematodes (Aplectana sp., Cosmocerca podicipinus, Oswaldocruzia lopesi, Physalopteroides venancioi, Rhabdias sp.), but the trematode Catadiscus propinquus also showed high prevalence. The trematodes Infidum infidum and Travtrema stenocotyle were also found, but in only one specimen. Adult frogs showed higher parasite diversity than subadults. Leptodactylus podicipinus was preferentially infected by direct life-cycle parasites and was reported as a new host record for seven helminth species.

The helminth fauna in 109 Eurasian otters (Lutra lutra L.) from France, Portugal and Spain was analysed, together with 56 faecal samples collected in Portugal and 23 fresh stools from otters included in a reintroduction programme. Seven helminth species were found in L. lutra in southwest Europe: Phagicola sp. (Trematoda), Aonchotheca putorii, Eucoleus schvalovoj, Strongyloides lutrae, Anisakis (third stage larvae) and Dirofilaria immitis (Nematoda), and Gigantorhynchus sp. (Acanthocephala). Eucoleus schvalovoj was the dominant species throughout southwest Europe. Strongyloides lutrae was significantly more prevalent in the Iberian Peninsula than in France. Apart from these two dominant nematodes and A. putorii, the other helminth species were incidental parasites of L. lutra in southwest Europe. The helminth fauna of L. lutra in southwest Europe is, in general, poorer than that reported in eastern Europe and in all other aquatic mustelids in southwest Europe. Phagicola specimens are reported for the first time in a non-marine wild carnivore in Europe. The prevalences of E. schvalovoj and S. lutrae obtained by necropsy were higher than those observed by coprological analysis using a formalin-ether concentration method (Ritchie). Nevertheless, the culture of fresh faeces appears to be the best method to study infection of L. lutra by Strongyloides.

A comparative study has been made of the Pseudophyllidean cestodes harboured by Pinnipeds, mainly from the Northern Hemisphere. Thirty species of this group of cestodes have been recorded in the literature, and as a result of the present study these may be divided into the following:

Valid species: Diphyllobothrium cordatum (Leuckart, 1868); D. elegans (Krabbe, 1865); D. glaciale (Cholodkovsky, 1915); D. hians (Diesing, 1850); D. lanceolatum (Krabbe, 1865); D. scoticum (Rennie & Reid, 1912); Diplogonoporus tetrapterus (v. Siebold, 1848); D. fasciatus (Krabbe, 1865); and Pyramicocephalus phocarum (Fabricius, 1780).

An established adult infection of Nematospiroides dubius was unaffected by the administration of immune lymphocytes and immune sera whereas an incoming larval infection was expelled. Past experiments have shown that the immune inoculum at least had the ability to recognize adult stages, leading to the hypothesis that adult stages secrete or excrete an immunomodulatory shield around themselves in the gastrointestinal tract. This hypothesis was given further credence by the demonstration that soluble antigens derived from adults abolished the generation of homologous immunity to this parasite. Modulation of immunity was reflected by increased fecundity, increased worm size, and increased survival time in the gut.

Histological examination of the small intestine of mice infected with Heligmosomoides polygyrus indicates that the final site of the larvae is in the circular muscle layer of the muscularis externa of the anterior small intestine. The larvae become embedded in the muscle between the first and third days after infection and are subsequently sequestered by a localized leucocytic response. There is no evidence of a true cyst. Histological evidence suggests that the larvae actively feed on the tissues of the host during their development.

The Angiostrongylus lungworms are of public health and veterinary concern in many countries. At the family level, the Angiostrongylus lungworms have been included in the family Angiostrongylidae or the family Metastrongylidae. The present study was undertaken to determine the usefulness and suitability of the nuclear 18S (small subunit, SSU) rDNA sequences for differentiating various taxa of the genus Angiostrongylus, as well as to determine the systematics and phylogenetic relationship of Angiostrongylus species and other metastrongyloid taxa. This study revealed six 18S (SSU) haplotypes in A. cantonensis, indicating considerable genetic diversity. The uncorrected pairwise ‘p’ distances among A. cantonensis ranged from 0 to 0.86%. The 18S (SSU) rDNA sequences unequivocally distinguished the five Angiostrongylus species, confirmed the close relationship of A. cantonensis and A. malaysiensis and that of A. costaricensis and A. dujardini, and were consistent with the family status of Angiostrongylidae and Metastrongylidae. In all cases, the congeneric metastrongyloid species clustered together. There was no supporting evidence to include the genus Skrjabingylus as a member of Metastrongylidae. The genera Aelurostrongylus and Didelphostrongylus were not recovered with Angiostrongylus, indicating polyphyly of the Angiostrongylidae. Of the currently recognized families of Metastrongyloidea, only Crenosomatidae appeared to be monophyletic. In view of the unsettled questions regarding the phylogenetic relationships of various taxa of the metastrongyloid worms, further analyses using more markers and more taxa are warranted.
