The effect of artesunate (ART) on the survival time of adult worm pairs of Schistosoma mansoni and on their egg output during in vitro culture was assessed. ART significantly decreased the survival time of both paired male and female worms at concentrations of 5, 10, 20 and 40 mg l− 1 during in vitro cultivation. An inhibitory effect of ART on the daily egg output of paired female worms during in vitro cultivation was also observed.

The life cycle of Gastrodiscoidcs hominis has been described using Helicorbis coenosus as the experimental intermediate host and the pig as the definitive host.

The morphology of the miracidium, redia and metacercaria has been described. Data have been furnished on the infection and longevity, of and production of cercariae by the snail host, and the growth and development of the adult-fluke in the definitive host.

Vaccines against gastrointestinal nematodes are one potential option for the control of parasitic gastroenteritis in ruminants. Excretory/secretory (E/S) and hidden antigens are being studied as candidates for vaccines against Haemonchus spp., which is a major parasite in cattle and small ruminants that are raised in warm climates. Protection has been observed after vaccination with some E/S proteases, particularly cysteine proteases and with some glycans that are abundant on the surfaces and in the secretory products of helminths. However, the most promising results are being obtained with glycoprotein antigens extracted from the microvillar surfaces of the Haemonchus contortus intestinal cells. These antigens are called ‘hidden′ because they are not exposed to the host's immune system during infection. Thus far, recombinant forms of these antigens have not been usefully protective. However, because only 5 μg of antigen is required per dose, production of a native antigen vaccine from adult parasites has been found to be practical and commercially viable. Trials indicate that a vaccine made from one particular isolate will cross-protect against geographically distant isolates.

1. Observations were made on various methods of infecting S. ornatum with 0. gutturosa, the behaviour of the parasite in its vector, and the maintenance of the vector in the laboratory.

2. S. ornatum reared from pupae brought into the laboratory were successfully infected with 0. gutturosa after feeding on infected cattle. The use of in vitro methods of infection was unsuccessful as microfilariae were ingested but did not undergo larval development.

3. The migratory route of 0. gutturosa microfilariae ingested by S. ornatum was studied and it was found that a mean of only 25% penetrate out of the abdominal midgut. The dimensional changes of 0. gutturosa were noted from the time of ingestion by S. ornatum until the infective stage was reached.

4. 0. gutturosa took 13–15 days to reach the infective stage in S. ornatum when maintained at a constant temperature of 23°C.

5. The development and structure of the peritrophic membrane of S. ornatum are described.

6. Preliminary observations on the seasonal variations of some factors affecting the transmission of O. gutturosa by S. ornatum suggest that microfilariae were only available for ingestion during the period of vector activity, after which they migrated into the deeper regions of the skin of cattle.

Entomopathogenic nematodes (EPN) were evaluated for their potential use as biological control agents against Phlyctinus callosus, the banded fruit weevil (BFW). The susceptibility of larvae and adults to EPN was evaluated using 400 infective juveniles (IJ) per insect after 4 days in 24-well bioassay trays. The nematode isolates used were all able to infect BFW, although the larvae were found to be more susceptible than were the adults. The percentage mortality for BFW larvae ranged from 41 to 73% and for BFW adults from 13 to 45%. The most effective isolate, SF41 of Heterorhabditis zealandica, was used to investigate the effect of vertical movement of nematodes in sand and sandy loam soil, at specified concentration and temperature. A higher (82.2 ± 0.084%) percentage mortality rate was obtained with the sandy loam soil, than with the use of sand (67.5 ± 0.12%). The LD50 and LD90 values after 4 days of incubation were 96 and 278 IJ/50 μl, respectively. Nematodes were inactive below 15°C, with the highest mortality of 74 ± 0.081% for BFW larvae recorded at 25°C. Heterorhabditis zealandica was able to complete its life cycle successfully in sixth-instar BFW larvae after a period of 22 days. The study showed BFW larvae not to be as susceptible to nematode infection as they need a high concentration (400 IJ/larva) and 4 days to give effective control.

Fish-borne zoonotic nematodes may infect humans when fish or squid are ingested raw or inadequately cooked. Human infections may have serious consequences, including the unexpected deaths of infected people. This kind of disease is poorly known in general, and the characteristics of such infections in South American countries as a whole have never been assessed. In this paper the present status of fish-borne nematodiases in humans in South American countries is characterized. Potentially zoonotic nematode species are very common in both freshwater and marine fish in South America. Reports of human infections have only been found in some countries, and their incidence (especially with anisakids and Gnathostoma spp.) varies from country to country. Apparently they are more abundant in countries with strong traditions of eating raw fish, and are more frequent on the western coast of South America. So far fish-borne nematodes have been reported in Argentina, Brazil, Chile, Colombia, Ecuador and Peru. In recent years, cases of human infection have appeared in probably underestimated numbers. People need to be clearly informed about risky feeding habits, and physicians need to learn more about zoonotic diseases.

The infection dynamics of the gill monogeneans Cichlidogyrus tilapiae and C. sclerosus on Oreochromis niloticus with respect to habitat type (reservoir, stream, ponds and cages), host sex, size and seasons was determined between January and November 2008. During the study period, 45.2% of the 650 fish examined were infected with Cichlidogyrus spp. The infected hosts harboured an average of 8.6 ± 3.4 parasites/fish. Across habitat types, the proportion of infected fish was not statistically different. In contrast, the number of parasites recorded on infected fish from different habitat types differed significantly. The highest parasite number was recorded in reservoir-dwelling fish and lowest in stream-dwelling hosts. Concerning sex, more female O. niloticus were infected and harboured a high number of parasites than male and sexually undifferentiated fish. A weak negative relationship was found between rainfall and monthly parasite infections. However, a higher number of parasites and proportion of infected hosts were found during dry than in wet seasons, except in ponds. Results of this study show that differential exposure due to changes in fish behaviour associated with habitat modification and sex may account for the infection difference across the sampled sites. Meanwhile, rainfall and the associated hydrological events are important factors regulating monogenean infections in tropical aquatic environments. The continuous presence of Cichlidogyrus spp. in fish provides evidence of possible parasite outbreaks, indicating the application of biosecurity measures as crucial for the success of intensive fish farming.

A survey of intestinal parasites in dogs was carried out in a rural region around Cantanhede, in northern Portugal, where 301 dog faecal samples were collected from small-ruminant farms. Saturated salt flotation and formol–ether sedimentation techniques were used. An enquiry was conducted in 234 farms and a risk factor evaluation for zoonotic helminths was determined among the 195 farmers who owned dogs. The overall parasite prevalence in faecal samples of dogs was 58.8%, with specific prevalences for Ancylostomidae being 40.9% followed by species of Trichuris (29.9%), Toxocara (8%), Isospora (4%), Capillaria (0.7%) and Spirometra (0.3%). Taeniidae eggs were present in five samples (1.7%) which were analysed with the polymerase chain reaction (PCR) technique and revealed to be from Taenia sp., and not Echinococcusgranulosus. This rural region has a traditional small-farm system, in which farm products are mainly for in-house consumption and home slaughtering is a current practice (57%). Analysis showed home slaughtering to be a statistically significant risk factor for the presence of Ancylostomidae (P= 0.007) and Toxocara sp. (P= 0.049). Owning cattle was found to be a significant risk factor for Taenia sp. (P= 0.031).

The purpose of the present paper is to bring together brief descriptions of the various species of the genus Anguillulina, more generally known under the synonymous generic name of Tylenchus Bastian, 1865. In the case of the plant-parasitic species, in addition to particulars on dimensions and morphology, data are given on life-history, hosts and geographical distribution; such matters as symptoms, pathology and methods of control are not dealt with.

The diplostomid genus Austrodiplostomum currently contains two species, i.e. A. mordax and A. ostrowskiae. Adults of these species inhabit the intestine of cormorants of the genus Nannopterum, whereas larval forms (metacercariae) are found in the eyes (vitreous humor) of freshwater fishes. Records of both species have been established across a wide geographic range in the Americas. Diplostomid adults and metacercariae were collected from a wide geographical range that spans from south-eastern Mexico, southwards to Central and South America. Even though the diplostomid has been largely reported in Mexico as Diplostomum (Austrodiplostomum) compactum, our specimens were identified morphologically as A. ostrowskiae. Sequences of the mitochondrial gene cytochrome c oxidase (cox 1) were obtained for 86 individuals, including 15 adults recovered from the intestine of Nannopterum brasilianus, in five localities, and 71 metacercariae from the eyes of ten fish species, in 13 localities. Sequences were used to evaluate the genetic diversity, and to test conspecificity of these specimens with the available sequence of A. ostrowskiae. Sequences were aligned with another 12 taxa representing five genera of Diplostomatidae, forming a dataset of 104 taxa with 478 nucleotides. The genetic divergence estimated among the 86 sequenced individuals, and that of A. ostrowskiae from the double-crested cormorant, Nannopterum auritus, in the USA, was very low, ranging from 0 to 0.8%. The maximum likelihood (ML) and Bayesian consensus trees showed that all sequences nested within a monophyletic lineage, with strong bootstrap and Bayesian posterior probability support values (100/1.0). In conclusion, a link between the metacercariae in fish and the adults in cormorants was established, indicating also that a single species is found in the distribution range comprising southern USA, southwards to Venezuela. Previous records of this species, particularly from Mexico, need to be corrected.

The effect of intestinal flora on the establishment, development and pathogenicity of Ascaris suum larvae in piglets (Large White breed) was investigated. The infected piglets with Ascaris and Escherichia coli showed signs of pneumonia, cough with respiratory difficulties initially even though these moderated with time. They lost appetite and showed signs of unthriftiness with loss of weight. The packed cell volume was normal but the differential leucocyte counts of the pigs infected with Ascaris larvae and bacteria had high neutrophils, unlike the very high lymphocyte count observed in piglets with ascarids only. The piglets had generalized serous atrophy of body fat. The pericardial and perirenal fats were gelatinous. There was a firm and nodular grey and red hepatization with abscess pockets in the intermediate and anterior one third of the diaphragmatic lobes of the lungs. The liver contained greyish-white and depressed focus immediately dorsal to the area of attachment to the gall bladder with multifocal areas. There was no significant gross lesion in the control animals. Cultural and microscopic examinations of some internal organs of the infected animals showed that bacteria were carried to the lungs by the migrating Ascaris larvae. The combined synergistic effect of Ascaris larvae and E. coli was also investigated and it was concluded that the two agents (A. suum larvae and E. coli) worked together synergistically.

During the examination of an abnormal specimen of the Scottish Red Deer (Cervus elaphus) in the winter of 1930, my colleague, Mr. W. C. Miller, M.R.C.V.S., discovered several specimens of a bursate nematode lying on the connective tissue between the latissimus dorsi and the external intercostal muscles. Struck by the unusual habitat of such a worm, search was subsequently made in similar situations in other deer, and in two animals the same parasite was found.

A case of intestinal anisakiasis caused by Anisakis sp. larva type I in a woman from Italy who consumed raw marinated anchovies, is reported. The diagnosis was based on the morphological features characteristic of anisakid larval stages, which were readily recognized in a large granuloma removed after emergency surgical treatment.

Morphological and molecular sequence data were combined with cross-hybridization studies and used to identify a new Steinernema sp. from Free State, South Africa. Molecular and morphological data indicate that the new species belongs to the ‘glaseri-group’ of Steinernema spp. Key morphological diagnostic characters for S. innovationi n. sp. include the morphometric features of the third-stage infective juveniles: total body length = 1054 (1000–1103) μm, tail length = 108 (97–117) μm, location of the excretory pore = 88 (82–91) μm, and D% = 58 (54–63), E% = 115 (104–137) and H% = 43 (37–46). Additionally, the morphology of the spicules and gubernaculum of the first-generation males are considered key diagnostic traits. Steinernema innovationi n. sp. was also characterized by analysis of both rDNA and mitochondrial gene sequence data, which further indicate the uniqueness of this Steinernema species.

An unidentified monogenean diplozoid species was collected from the gills of moggel in the Vaal River and Vaal Dam, South Africa. Specimens were removed from gills of the hosts and observed using light and electron microscopy to compare these diplozoids with known species. The second internal transcribed spacer (ITS2) of the ribosomal gene was amplified, sequenced and compared to that of other diplozoid taxa. Morphological species delimitation was used to determine the identity of these diplozoids, but they did not match the description of any diplozoid taxa. This species is recognized by the specific size of the hooks, number of plicae in posterior and trapezoid anterior projection of the median sclerite connecting to the clamp jaws via a single sclerite, occasionally with two small additional sclerites. Genetic characteristics based on sequence data from the ITS2 region also distinguish this taxon from all other diplozoid taxa. This South African diplozoid grouped in the same clade as Paradiplozoon ichthyoxanthon Avenant-Oldewage, 2013. Data clearly indicate that diplozoids collected from moggel represent a new, distinct taxon of Paradiplozoon Akhmerov, 1974 and are described here as Paradiplozoon vaalense n. sp.

Investigations of acquired immunity to Fasciola hepatica in calves, sheep and rabbits are described. With injections of extracts of imma- ture parasites retarded growth of the challenge infection was observed between the 6th and 7th week. Following a previous experience of a natural infection or viable fluke implants, retardation and reduced take of the challenge infection were observed. Considerable individual variation was present and the level of significance between treatment and control groups was low.

Infective larvae of Dirofilaria immitis commonly escaped from the tips of the Iabella and rarely from the mid-portion of the labium of Aedes aegypti as they engorged blood or just probed on mice. During the infective feed a small quantity of liquid, not more than 1 µI in volume, accumulated around the emerging larvae. Liquid was never seen when uninfected mosquitoes engorged on mice. This fluid is of insect origin and probably haemolymph that exuded from the lumina of the labella or labium during worm emergence. The presence of the fluid prevents dessication of larvae on the surface of the skin and maintains them in a liquid medium until they enter the puncture wound that remains after withdrawal of the biting fascicle. The possible effects of the fluid on the geographical distribution of mosquito borne filariae are discussed.

All three strains of Neotricula aperta (Gastropoda: Pomatiopsidae) sampled from populations in northeast Thailand were found to be compatible with a Schistosoma mekongi isolate from Kratié District, eastern Cambodia. The infection rates were: 3.0%, α-strain; 6.0%, β-strain; and 20.5%, γ-strain. The greater infectivity to the γ-strain, over both the α- and β-strains, was statistically significant. Comparisons of infectivity patterns for the Kratié isolate, with those described in earlier studies using S. mekongi isolates from southern Laos, revealed differences among the strains. The γ-strain of N. aperta is responsible for endemic transmission of human schistosomiasis in southern Laos and at Kratié. Consequently, the findings of this study are of use in the prediction of changes in the distribution of Mekong schistosomiasis, particularly its introduction to northeast Thailand from the neighbouring countries, Cambodia and Laos.

The histopathology of the livers and lungs of pigs after a primary infection with eggs of A. suum was compared with the lesions and cellular response found in resistant pigs after reinfection.

The author's previously published work on experimental infection of guinea pigs, rabbits and pigs is also discussed and summarized.

Sera of Sudanese patients with active infections of Schistosoma mansoni or S. haematobium were tested in immunoblots for their reactivity with Mr31/32 000 proteins of adult S. mansoni (Sm31/32). All sera from patients with intestinal (n=123) and all but one from those with urinary schistosomiasis (n=35) had antibodies against Sm31/32. These and additional data suggest that both specificity and sensitivity of Sm31/32 to detect schistosome infections are close to 100%. Antibodies against these proteins developed also in monkeys after experimental infection with S. haematobium. Sm31/32 antigens reacted in immunoblots as a doublet with most S. haematobium-patient sera and as a broad band with many S. mansoni-sera suggesting that at least two components are present in the molecular weight region of Mr31/32 000. The data demonstrate the potential use of Sm31/32 from adult worms to diagnose patients with intestinal or urinary schistosomiasis in endemic areas.
