Investigations of acquired immunity to Fasciola hepatica in calves, sheep and rabbits are described. With injections of extracts of imma- ture parasites retarded growth of the challenge infection was observed between the 6th and 7th week. Following a previous experience of a natural infection or viable fluke implants, retardation and reduced take of the challenge infection were observed. Considerable individual variation was present and the level of significance between treatment and control groups was low.

During the examination of an abnormal specimen of the Scottish Red Deer (Cervus elaphus) in the winter of 1930, my colleague, Mr. W. C. Miller, M.R.C.V.S., discovered several specimens of a bursate nematode lying on the connective tissue between the latissimus dorsi and the external intercostal muscles. Struck by the unusual habitat of such a worm, search was subsequently made in similar situations in other deer, and in two animals the same parasite was found.

Bithynids snails are a widespread group of molluscs in European freshwater systems. However, not much information is available on trematode communities from molluscs of this family. Here, we investigate the trematode diversity of Bithynia tentaculata, based on molecular and morphological data. A total of 682 snails from the rivers Lippe and Rhine in North Rhine-Westphalia, Germany, and 121 B. tentaculata from Curonian Lagoon, Lithuania were screened for infections with digeneans. In total, B. tentaculata showed a trematode prevalence of 12.9% and 14%, respectively. The phylogenetic analyses based on 55 novel sequences for 36 isolates demonstrated a high diversity of digeneans. Analyses of the molecular and morphological data revealed a species-rich trematode fauna, comprising 20 species, belonging to ten families. Interestingly, the larval trematode community of B. tentaculata shows little overlap with the well-studied trematode fauna of lymnaeids and planorbids, and some of the detected species (Echinochasmus beleocephalus and E. coaxatus) constitute first records for B. tentaculata in Central Europe. Our study revealed an abundant, diverse and distinct trematode fauna in B. tentaculata, which highlights the need for further research on this so far understudied host–parasite system. Therefore, we might currently be underestimating the ecological roles of several parasite communities of non-pulmonate snail host families in European fresh waters.

An unidentified monogenean diplozoid species was collected from the gills of moggel in the Vaal River and Vaal Dam, South Africa. Specimens were removed from gills of the hosts and observed using light and electron microscopy to compare these diplozoids with known species. The second internal transcribed spacer (ITS2) of the ribosomal gene was amplified, sequenced and compared to that of other diplozoid taxa. Morphological species delimitation was used to determine the identity of these diplozoids, but they did not match the description of any diplozoid taxa. This species is recognized by the specific size of the hooks, number of plicae in posterior and trapezoid anterior projection of the median sclerite connecting to the clamp jaws via a single sclerite, occasionally with two small additional sclerites. Genetic characteristics based on sequence data from the ITS2 region also distinguish this taxon from all other diplozoid taxa. This South African diplozoid grouped in the same clade as Paradiplozoon ichthyoxanthon Avenant-Oldewage, 2013. Data clearly indicate that diplozoids collected from moggel represent a new, distinct taxon of Paradiplozoon Akhmerov, 1974 and are described here as Paradiplozoon vaalense n. sp.

Morphological and molecular sequence data were combined with cross-hybridization studies and used to identify a new Steinernema sp. from Free State, South Africa. Molecular and morphological data indicate that the new species belongs to the ‘glaseri-group’ of Steinernema spp. Key morphological diagnostic characters for S. innovationi n. sp. include the morphometric features of the third-stage infective juveniles: total body length = 1054 (1000–1103) μm, tail length = 108 (97–117) μm, location of the excretory pore = 88 (82–91) μm, and D% = 58 (54–63), E% = 115 (104–137) and H% = 43 (37–46). Additionally, the morphology of the spicules and gubernaculum of the first-generation males are considered key diagnostic traits. Steinernema innovationi n. sp. was also characterized by analysis of both rDNA and mitochondrial gene sequence data, which further indicate the uniqueness of this Steinernema species.

The histopathology of the livers and lungs of pigs after a primary infection with eggs of A. suum was compared with the lesions and cellular response found in resistant pigs after reinfection.

The author's previously published work on experimental infection of guinea pigs, rabbits and pigs is also discussed and summarized.

Observations on the course of Hymenolepis nana infection in immunosuppressed mice are presented. Treatment of the host with hydrocortisone acetate caused superinfection of the bowel with worms and the development of normal cysticercoids in the mesenteric lymph node and liver. Natural infection of mice deprived of their T-cells by pre-adult thymectomy and administration of antithymocyte serum resulted in superinfection and widespread metastasis of aberrant cysticercoids that were greatly enlarged and without scolices, causing death after about five months. The significance of these findings and their possible relevance to human infection with H. nana are discussed.

In field experiments, conducted on parasite-free grass plots in two consecutive summers, artificially prepared cow pats containing Cooperia oncophora eggs were inoculated with the nematode-trapping fungus Arthrobotrys oligospora. Numbers of infective C. oncophora larvae isolated from the pats as well as from the surrounding herbage were subject to an approximately teh-fold reduction as compared with numbers in fungus-free pats and herbage surrounding these. This reduction was undoubtedly a result of entrapment of the parasite larvae within the faecal pats.

Diplozoon homoion gracile from the southern barbel (Barbus meridionalis) displays an egg-laying and an egg hatching rhythm, the latter being more clearly defined. Egg laying continues both day and night but significantly more eggs are laid at night. The oncomiracidia hatch at dusk. If parasite eggs are eaten by the host they pass through the gut undamaged and hatch normally.

Sera of Sudanese patients with active infections of Schistosoma mansoni or S. haematobium were tested in immunoblots for their reactivity with Mr31/32 000 proteins of adult S. mansoni (Sm31/32). All sera from patients with intestinal (n=123) and all but one from those with urinary schistosomiasis (n=35) had antibodies against Sm31/32. These and additional data suggest that both specificity and sensitivity of Sm31/32 to detect schistosome infections are close to 100%. Antibodies against these proteins developed also in monkeys after experimental infection with S. haematobium. Sm31/32 antigens reacted in immunoblots as a doublet with most S. haematobium-patient sera and as a broad band with many S. mansoni-sera suggesting that at least two components are present in the molecular weight region of Mr31/32 000. The data demonstrate the potential use of Sm31/32 from adult worms to diagnose patients with intestinal or urinary schistosomiasis in endemic areas.

All three strains of Neotricula aperta (Gastropoda: Pomatiopsidae) sampled from populations in northeast Thailand were found to be compatible with a Schistosoma mekongi isolate from Kratié District, eastern Cambodia. The infection rates were: 3.0%, α-strain; 6.0%, β-strain; and 20.5%, γ-strain. The greater infectivity to the γ-strain, over both the α- and β-strains, was statistically significant. Comparisons of infectivity patterns for the Kratié isolate, with those described in earlier studies using S. mekongi isolates from southern Laos, revealed differences among the strains. The γ-strain of N. aperta is responsible for endemic transmission of human schistosomiasis in southern Laos and at Kratié. Consequently, the findings of this study are of use in the prediction of changes in the distribution of Mekong schistosomiasis, particularly its introduction to northeast Thailand from the neighbouring countries, Cambodia and Laos.

1. The filarial worm Brugia patei was brought to London from East Africa in the larval stage developing in Mansonia mosquitoes. Subsequent transmission to domestic cats was possible in London through a laboratory colony of Mansonia uniformis and the worm was very infective to this species of mosquito. The worm was much less infective to Aedes togoi and Anopheles gambiae.

2. Subsequent passage of the worm through Aedes togoi has increased its infectivity to this species of mosquito. The infection rate has risen from 43·6% in females of A. togoi fed on the cat original- ly infected by larvae from Mansonia mosquitoes, to 59·2% in females fed on a cat containing the first generation of worms to pass through A. togoi, to 79·1% in females fed on cats containing the second generation of worms to pass through A. togoi, to 86·7% in females fed on cats containing the third generation of worms to pass through A. togoi, to 89·8% in females fed on cats containing the fourth generation of worms to pass through A. togoi.

3. The distributions of infective stage larvae in female mosquitoes were negative binomial distributions. These were related to the distributions of microfilarial intake immediately after infection which were also negative binomial. From one generation of worms passed through the new mosquito host to the next the mean and the k value of the negative binomial of the infective stage larvae approached the same parameters for the distribution of microfilarial intake. By the fourth generation of worms passed through A. togoi the distributions of microfilarial intake and infective stage larvae developing in A. togoi were similar, suggesting close adaptation to the new mosquito host. A similar relation between these distributions was found originally in Mansonia. There were however considerable differences in these distributions between the two species of mosquito.

4. The experiment extended over a period of 6 years. The change in infectivity did not appear to be related to the microfilarial density, age of infection, or sex, in the mammalian host. Neither could it be related to the genetics of susceptibility in the mosquito, nor to mosquito culture techniques, nor to mosquito mortality.

5. It is concluded that the observed change in infectivity is best explained by a change in adaptation of the worm to the new intermediate host by selection within the mosquito of those microfilariae adapted to it. No major change in infectivity to the original intermediate host, Mansonia, was found during the experiment.

6. These findings are examined with reference to changes in adaptation to mosquito hosts in the history of Bancroftian filariasis.

Infective larvae of Dirofilaria immitis commonly escaped from the tips of the Iabella and rarely from the mid-portion of the labium of Aedes aegypti as they engorged blood or just probed on mice. During the infective feed a small quantity of liquid, not more than 1 µI in volume, accumulated around the emerging larvae. Liquid was never seen when uninfected mosquitoes engorged on mice. This fluid is of insect origin and probably haemolymph that exuded from the lumina of the labella or labium during worm emergence. The presence of the fluid prevents dessication of larvae on the surface of the skin and maintains them in a liquid medium until they enter the puncture wound that remains after withdrawal of the biting fascicle. The possible effects of the fluid on the geographical distribution of mosquito borne filariae are discussed.

The influence of habitat structure on helminth communities of three sigomdontinae rodent species (Akodon cursor, A. montensis and Oligoryzomys nigripes) was investigated in forest fragments within an agricultural landscape in south-eastern Brazil. This is a pionner study correlating the occurrence of helminth species of rodent hosts with microhabitat characteristics. Rodents were collected from 12 fragments and in a continuous conserved area. Up to 13 nematode, three cestode and two trematode species were identified, and habitat fragmentation was found to have more influence on the helminth composition of O. nigripes compared to the other two rodent species. Fragmentation appeared to limit the development of some helminths’ life cycles, e.g. with some species such as Trichofreitasia lenti, Protospirura numidica, Cysticercus fasciolaris and Avellaria sp., occurring mostly in areas with less anthropic impact. However, fragmentation did not seem to affect the life cycles of other dominant helminths, such as the trematode Canaania obesa, the nematodes Stilestrongylus lanfrediae,S. eta and S. aculeata, and the cestode Rodentolepis akodontis. The helminth community structure followed a nested pattern of distribution in A. montensis and O. nigripes. Stilestrongylus lanfrediae seemed to be more associated with dense understorey, C. obesa with open canopy and dense understorey, and Guerrerostrongylus zetta with organic matter on the ground. Their presence in each area may be explained by aspects of their life cycles that take place in the external environment outside the host.

P-glycoproteins (P-gps) are proteins that function as efflux pumps, removing lipophilic xenobiotic compounds from cells. There is evidence that P-gps play a role in the resistance of parasitic nematodes to anthelmintic drugs such as benzimidazoles and macrocyclic lactones. As anthelmintic resistance becomes more common, it is important to identify candidate resistance genes with the aim of understanding the molecular basis of resistance, and of developing assays to detect these resistance-associated changes. We identified several sequences from the genome of the parasite Haemonchus contortus with convincing homology to the known P-gp coding genes of the model nematode Caenorhabditis elegans. Nine of these sequences were successfully amplified by polymerase chain reaction (PCR) and shown to be most similar to the C. elegans sequences for pgp-1, pgp-2, pgp-3, pgp-4, pgp-9, pgp-10, pgp-11, pgp-12 and pgp-14. These partial P-gp sequences from H. contortus were used to design and optimize a quantitative real-time PCR assay to investigate potential changes in the expression levels of P-gp transcripts associated with drug resistance. No significant changes in P-gp mRNA expression levels were found in a rapidly selected ivermectin-resistant parasite isolate compared to its drug-sensitive parent, but the assay has the potential to be used on other isolates in the future to further investigate resistance-associated changes in P-gp gene expression.

The work has demonstrated that one of the more important areas of Scotland where Dicrocoelium dendriticum is endemic carried infected colonies of Helicella itala which live in association with the ant Formica fusca. Workers of the ant were shown to acquire typical metacercarial cysts after the ingestion of slime balls, and the cysts gave rise to mature lancet flukes in sheep and hamsters, eggs appearing in the faeces after 56 and 48 days respectively. Higher egg counts were obtained from hamsters to which more cysts were administered. This is the first occasion on which an ant intermediate host has been demonstrated for Dicrocoelium in Britain.

1. The method of maintaining schistosomes in vitro is described in detail.

2. Observations made on cultured S. mansoni are given.

3. Investigation of glycogen content shows that there is no significant difference between the reserve of glycogen in fresh and cultured worms. It is suggested that the worms do not rely on their reserve of glycogen but can take up carbohydrate from the medium.

4. The method could be adapted for the culture of other helminths besides schistosomes.

1. The intake of microfilariae of Onchocerca volvulus in two series of females of Simulium damnosum fed on an infected human volunteer in West Africa and dissected after staining in London was from 0–171 microfilariae per female in one series and from 0–152 microfilariae per female in the other series. The distribution of microfilariae per female became more normal when plotted on a geometric scale.

2. The migration of the microfilariae was followed in the two series of flies by comparing the geometric (Williams) mean numbers of microfilariae in the stomach, abdomen, thorax and head, from 2 minutes to 48 houis after engorgement on blood. Microfilariae left the blood meal rapidly after engorgement, 40–50% leaving the stomach within the first 60 minutes.

An investigation was conducted into the parasitic infection of an indigenous cichlid, Oreochromis mossambicus, collected seasonally from the Nwanedi-Luphephe dams of the Limpopo River System from July 2007 to April 2009. Of 157 host specimens examined, 115 (73.25%) were infected by at least one gill parasite. In all, 1565 monogenean specimens were collected, belonging to five different species and two genera: Cichlidogyrus (C. halli, C. sclerosus, C. tilapiae and C. dossoui) and Scutogyrus (S. longicornis). Infracommunities were poor, with only 27 (17.20%) hosts harbouring four of the five species observed. Cichlidogyrus halli was the dominant species, with a prevalence of 73.25%. Prevalence values for each of the other four species were less than 50%. The mean intensities for each of the five species were low ( < 8 parasites/host). The parasite abundance and intensity levels were not influenced by either the sex or the size of the host. The abundance of all parasite species except for S. longicornis exhibited seasonal fluctuations, reaching peaks in winter and summer. The spatial distribution of each parasite was studied on different regions of the gill, and positive associations among some species were revealed.

Morphological and molecular analyses of a stem nematode causing a severe disease on infected sowthistle (Sonchus bulbosus) plants, involving the formation of gall-like structures on infected leaves and stems, have led to the description of a new species named Ditylenchus oncogenus n. sp. Morphologically, the new species is characterized by a medium to large body size (all adults more than 1 mm in length); a delicate stylet (9.0–11.0 μm long) with minute, rounded knobs; a long post-vulval uterine sac (c. 65% of the vulva–anus distance); six incisures at the lateral fields and characteristic D. destructor-pattern of spicules (with pronounced ventral tumulus and anteriorly pointed, less sclerotized, cuticle parts present within the lamina). The results of molecular analysis of rRNA gene sequences, including the D2–D3 expansion regions of 28S rRNA, internal transcribed spacer (ITS) rRNA, partial 18S rRNA gene, the protein-coding mitochondrial gene, cytochrome oxidase c subunit I (COI), and the heat-shock protein 90 (hsp90) gene, support the new species status. The results of a host-suitability test indicated that the new species does not parasitize potato (Solanum tuberosum) tubers and broad bean (Vicia faba) seedlings. Histopathological observations on naturally infected sowthistle tissues revealed that D. oncogenus n. sp. causes floral stem neoplasia and midrib leaf gall formation on the type, and to date only known, host. The galls were characterized by extensive hyperplasia, where several necrotic cells in the neoplasic area were directly damaged by feeding of the nematode, whereas a number of adjacent cells showed typical cytological changes, such as granulated cytoplasm with hypertrophied nuclei and nucleoli.
