Laboratory investigations were designed to study the influence of temperature, pH and oxygen tension on the growth of Arthrobotrys oligospora, a nematode-trapping microfungus. Experiments were performed to evaluate the potential role of A. oligospora in destroying third-stage larvae of Cooperia spp. on agar plates and in cattle faeces. The fungus had a growth rate optimum at 23°ree;C and pII 6. Anaerobic cultivation for 23 hours at 23°ree;C and 39°ree;C inhibited fungal growth, but it did not destroy the fungus, which regained growth upon a subsequent shift to aerobic conditions at 23°ree;C. Under experimental conditions in petri-dishes containing agar, the nematode-trapping efficiency of the fungus was striking in that 100% of a population of third-stage larvae of Cooperia spp. was captured within three days of the experiment. The trapping efficiency in faeces was shown to depend upon the inoculation level. At a concentration of approximately 2500 conidia per g faeces, 99% of the larvae were destroyed. The possibilities of using nematode-trapping fungi in controlling animal-parasitic nematodes arc discussed.

Suspensions of embryonated (L2) Toxocara canis and T. cati eggs were maintained under freezing conditions in order to study the impact on their long term viability The eggs had been removed from the uteri of adult Toxocara spp. worms and the suspended in a 0.4% formalin solution before being frozen in 20 ml plastic tubes in the freezer compartment of a domestic refrigerator. Assessment of embryonated egg viability over a period of 34 days was conducted by microscopic examination under a high light intensity Embryonated T. cati eggs were found to exhibit a greater resistance to freezing than those of T. canis. A difference in the size, design of the egg shell or the metabolism of the larval form may have accounted for the observed differences in the percentage viabilities over time between the two species of Toxocara.

Protease production from Duddingtonia flagrans (isolate AC001) was optimized and the larvicidal activity of the enzymatic extract was evaluated on infective horse cyathostomin larvae (L3). Duddingtonia flagrans was grown in liquid medium with eight different variables: glucose, casein, bibasic potassium phosphate (K2HPO4), magnesium sulphate (MgSO4), zinc sulphate (ZnSO4), ferrous sulphate (FeSO4), copper sulphate (CuSO4) and temperature. The Plackett–Burman analysis showed a significant influence of MgSO4, CuSO4 and casein (P < 0.05) on protease production by D. flagrans in liquid medium. Central composite design indicated that the highest proteolytic activity was 39.56 U/ml as a function of the concentrations of casein (18.409 g/l), MgSO4 (0.10 g/l) and CuSO4 (0.50 mg/l). A significant difference (P < 0.01) was found for the larval number between the treated and control groups at the end of the experiment. A reduction of 95.46% in the number of free-living larvae was found in the treated group compared with the control. The results of this study suggest that protease production by D. flagrans (AC001) in liquid medium was optimized by MgSO4, CuSO4 and casein, showing that the optimized enzymatic extract exerted larvicidal activity on cyathostomins and therefore may contribute to large-scale industrial production.

A study has been carried out to determine the morphological changes to the adult liver fluke, Fasciola hepatica after treatment in vivo with artemether. Rats were infected with the triclabendazole-resistant Sligo isolate of F. hepatica, dosed orally with artemether at a concentration of 200 mg/kg and flukes recovered at 24, 48 and 72 h post-treatment (p.t.). Surface changes were monitored by scanning electron microscopy and fine structural changes to the tegument and gut by transmission electron microscopy. Twenty-four hours p.t., the external surface showed minor disruption, in the form of mild swelling of the tegument. The tegumental syncytium and sub-tegumental tissues appeared relatively normal. Forty-eight and seventy-two hours p.t., disruption to the tegumental system increased, with isolated patches of surface blebbing and reduced production of secretory bodies by the tegumental cells being the main changes seen. The gastrodermal cells showed a relatively normal morphology 24 h p.t. By 48 h, large numbers of autophagic vacuoles and lipid droplets were present. Autophagy increased in magnitude by 72 h p.t. and substantial disruption to the granular endoplasmic reticulum was observed. Results from this study show that flukes treated in vivo with artemether display progressive and time-dependent alterations to the tegument and gut. Disruption to the gut was consistently and substantially more severe than that to the tegument, suggesting that an oral route of uptake for this compound predominates. This is the first study providing ultrastructural information on the effect of an artemisinin compound against liver fluke.

A polymerase chain reaction (PCR) technique has been used for the differentiation of T. canis and T. cati eggs isolated from soil and previously identified from microscopical observations. The method, using specific primers for the identification of the two Toxocara species, was assessed in both the field and laboratory. Successful results were obtained when only a single or large numbers of eggs were recovered from 40 g soil samples. The method is sensitive, allows analysis of material independent of the stage of egg development and can be adapted for the recovery of other species of parasites from soil.

Herring Clupea harengus L. viscera were examined for endoparasitic infections as part of a multidisciplinary stock identification project (WESTHER, EU Contract no. Q5RS-2002-01 056) which applied a range of stock discrimination techniques to the same individual fishes to obtain comparable results for multivariate analysis. Spawning and non-spawning adults, and juvenile herring were caught, over 3 years, by commercial and research vessels from numerous locations to the west of the UK and Ireland, along with control samples of spawning fish from the eastern Baltic Sea, and juveniles from sites in the eastern and western North Sea, and the north of Norway. The metacercariae of two renicolid digeneans (Cercaria pythionike and Cercaria doricha), one larval nematode (Anisakis simplex s.s.) and one larval cestode (Lacistorhynchus tenuis) were selected as tag species. Results were compared with those from herring collected between 1973 and 1982, which suggested remarkable stability in the parasite fauna of herring in the study area. These species were used to compare the parasite infracommunities of spawning herring. A significant variation in infracommunity structure was observed between different spawning grounds. These results suggest that the parasite fauna of herring are spatially variable but remain temporally stable in both the short and long term. Significant differences in prevalence and abundance of infections and comparisons of parasite infracommunity enabled the separation of putative herring stocks west of the British Isles. Distinctive patterns of parasite infection in two different spawning groups off the north coast of Scotland suggest that this area is occupied by two spawning populations, one recruiting from the west of Scotland, the other from outside this area, and most likely from the eastern North Sea. The distribution patterns of L. tenuis, C. doricha and C. pythionike suggest the potential for fish that spawn in three distinct International Council for the Exploration of the Seas (ICES) management units to be present in mixed aggregations found over the Malin Shelf, with significant implications for management in this area.

Human taeniosis due to Taenia saginata is cosmopolitan where beef is consumed; however, there is little or no information on the symptomatology over the early time-course of human infection. Copro-antigen detection is very useful in community screening for human taeniosis, particularly for T. solium, but there are no data on copro-antigen detection in pre-patent infection. In order to provide insight into this, a voluntary self-infection with T. saginata was undertaken and monitored over a 6-month period using a copro-antigen enzyme-linked immunosorbent assay (ELISA) that we developed using anti-T. saginata antibody based reagents. Tapeworm patency, defined as first proglottid appearance, occurred on day 86 post-infection (pi) and was followed by almost daily release of proglottids (range 1–8) until termination using praziquantel on day 180 pi. The first 10 weeks post-infection (wpi) were essentially asymptomatic, followed by main symptoms of involuntary proglottid discharge throughout the infection period, and abdominal discomfort peaking around 15–19 wpi. Copro-antigens could not be reliably detected until 2 weeks before proglottid patency but then remained highly elevated over the next 15 weeks until treatment. Copro-antigen levels reverted to negative 4 days post-treatment. This time-course study suggests that although copro-antigen ELISA is an excellent diagnostic tool for established patent infections of T. saginata, it may not be reliable for faecal antigen detection in the early infection phase prior to proglottid release for T. saginata and other human taenioses.

Hydatidosis is an important public health problem in several parts of Iran. The aim of this molecular study is to investigate Echinococcus granulosus genotypes as the causative agents of hydatidosis in the south-west of Iran (Khuzestan province). In this study, isolates of 334 hydatid cysts were collected from the liver and lungs of 141 sheep, 104 cattle, 84 goats and 5 human cases. DNA was extracted and examined by nested polymerase chain reaction (PCR) of ribosomal DNA (rDNA) internal transcribed spacer 1 (ITS1) and restriction fragment length polymorphism (RFLP)-PCR. In addition, fragments of genes coding for ITS1 were sequenced. The results of RFLP-PCR analysis revealed the presence of the G1 genotype in all human, cattle, goat and sheep isolates. Furthermore, no camel strain (G6) was detected among all samples in the regions studied. The molecular findings indicate that the predominant genotype involved in E. granulosus transmission in south-west Iran is the common sheep strain (G1 genotype), which occurs in human, cattle, sheep and goat populations. In conclusion, these results may have important implications for hydatid disease control in the areas studied.

1. With the aid of field growth curves, age-prevalence curves can be derived from field collections of snails, using cercarial shedding as proof of infection by schistosomes.

2. Such curves were obtained from eight collections of field snails: one sample each of Bulinus (Physopsis) nasutus productus and Biomphalaria pfeifferi, intermediate hosts respectively of Schistosoma haematobium and S. mansoni in Tanzania, and six samples of Biomphalaria glabrata, intermediate host of S. mansoni on St. Lucia, West Indies.

3. Catalytic curves were fitted to the data and in each case the two-stage curve gave the most satisfactory fit.

4. This curve is the resultant of two opposing forces: one force being the rate of infection and the other the rate of loss of infection in a snail population.

5. The rate of infection may be defined as the number of successful snail-miracidium contacts per 1,000 snails per week, while the rate of loss of infection is taken to represent the number of deaths per 1,000 infected snails per week.

6. The underlying assumptions of this form of analysis are discussed and, in view of the independent verification of some of the calculated rates, the values are considered realistic.

7. Besides providing quantitative data for inclusion in mathematical models of schistosome transmission, the technique offers an additional means of assessing the efficacy of certain methods of controlling transmission.

The entomopathogenic nematode Steinernema yirgalemense is considered a promising agent in the biocontrol of insects. However, little is known about the bacteria living in symbiosis with the nematode. In this study, we have identified the only available bacterial strain (157-C) isolated from S. yirgalemense, as a member of the species Xenorhabdus indica. Identification was based on 16S rDNA, recA, dnaN, gltX, gyrB and infB gene sequence analyses. The relatedness of strain 157-C to the type strain of X. indica (DSM 17 382) was confirmed with DNA–DNA hybridization. The phenotypic characteristics of strain 157-C are similar to those described for the type strain of X. indica. This is the first report associating X. indica with S. yirgalemense.

Eighty-nine samples of grey wolf (Canis lupus L.) faeces were collected between 2002 and 2004 from two areas in the western Beskidy Mts (south Poland). Helminth eggs were observed in 56.2% of faeces examined. These included: Alaria alata (2.2%), taeniid eggs (11.2%), Toxocara canis (5.6%), Toxascaris leonina (1.1%), Eucoleus aerophilus (14.6%), Ancylostoma caninum (12.3%), Uncinaria stenocephala (37%) and unidentified roundworm eggs of the family Strongyloididae (1.1%). Eucoleus aerophilus is recorded for the first time from Poland. The results are compared with the helminth fauna of other wolf populations in Europe.

The abundance of six species of trematodes: Aphanurus stossichi, Bacciger israelensis, Diphterostomum israelense, Plagioporus idoneus, Lepocreadium album and L. pegorchis, parasitic in the digestive tract of marine teleostei (Sparidae) collected near Jounieh (east Mediterranean), was analysed as a function of the host-size. In two parasite/host systems, infections were observed from the lowest size classes of the sample, with a clear tendency to an increase of abundance in older fish. In four others, parasites appear only above a rather high threshold class, young individuals never being infected. In the last three parasite/host systems, host invasion may occur early or late, but infection decreases above a well defined size class, old fishes rarely or never being infected. A given trematode species. when parasitizing several host species, shows similar abundance/host size relationships, e.g. P. idoneus in Diplodus vulgaris and Oblada melanura. When more than one species of trematode infects a single host species, curves can be markedly distinct; for instance, L. pegorchis was collected from Pagellus erythrinus below 15 cm. whereas D. israelense parasitized the same fish approximately above the same size. There is no evidence that such a replacement of one trematode by another in the course of host growth is a result of interspecific competition.

Schistosoma species have traditionally been arranged in groups based on egg morphology, geographical origins, and the genus or family of snail intermediate host. One of these groups is the ‘S. indicum group’ comprising species from Asia that use pulmonate snails as intermediate hosts. DNA sequences were obtained from the four members of this group (S. indicum, S. spindale, S. nasale and S. incognitum) to provide information concerning their phylogenetic relationships with other Asian and African species and species groups. The sequences came from the second internal transcribed spacer (ITS2) of the ribosomal gene repeat, part of the 28S ribosomal RNA gene (28S), and part of the mitochondrial cytochrome c oxidase subunit 1 (CO1) gene. Tree analyses using both distance and parsimony methods showed the S. indicum group not to be monophyletic. Schistosoma indicum, S. spindale and S. nasale were clustered among African schistosomes, while S. incognitum was placed as sister to the African species (using ITS2 and 28S nucleotide sequences and CO1 amino acid sequences), or as sister to all other species of Schistosoma (CO1 nucleotide sequences). Based on the present molecular data, a scenario for the evolution of the S. indicum group is discussed.

The relative importance of ecology and evolution as factors determining species richness and composition of the helminth communities of fish is a matter of current debate. Theoretical studies use host–parasite lists, but these do not include studies on a temporal or spatial scale. Local environmental conditions and host biological characteristics are shown to influence helminth species richness and composition in four fish species (Eugerres plumieri, Hexanematichthys assimilis, Oligoplites saurus, and Scomberomorus maculatus) in Chetumal Bay, Mexico. With the exception of H. assimilis, the helminth communities had not been previously studied and possible associations between environmental and host biological characteristics as factors determining helminth species richness and composition using redundancy analysis (RDA) are described. Thirty-four helminth species are identified, with the highest number of species (19 total (mean = 6.3 ± 2.1)) and the lowest (9 (4.0 ± 1.0)) occurring in H. assimilis and S. maculatus, respectively. The larval nematodes Contracaecum sp. and Pseudoterranova sp. were not only the helminth species shared by all four host species but also were the most prevalent and abundant. Statistical associations between helminth community parameters and local ecological variables such as host habitat use, feeding habits, mobility, and time of residence in coastal lagoons are identified. Phylogeny is important because it clearly separates all four host species by their specialist parasites, although specific habitat and feeding habits also significantly influence the differentiation between the four fish species.

The susceptibility of Bulinus truncatus, B. tropicus, B. natalensis, B. coulboisi, B. globosus, B. cernicus and B. wrighti to Schistosoma haematobium from Egypt were examined. Eighteen different populations of B. truncatus were investigated.

A new method of measuring the compatibility between schistosomes and the intermediate snail hosts, the total cercarial production per 100 exposed snail, is introduced.

All B. truncatus populations became infected and could be divided in four groups according to the total cercarial production per 100 exposed snails. In the first group the cercarial production varied from 250 000 to 500 000 and in the last group between 5000 and 20 000. The first group had a high infection rate of 62 to 100% and a low mortality of approximately 10% and the last group showed a very low infection rate and a high mortality.

B. globosus (Rhodesia), B. cernicus and B. wrighti became infected, while B. globosus (Zaire), B. coulboisi, B. natalensis and B. tropicus were refractory.

The shortest prepatent period was 26 days at a room temperature of 26°C. The maximum cercariae production for one snail in one day was 600 and the longest duration of infection was 200 days.

Experimenting with different doses of miracidia, 5 to 50 per snail, resulted in a one hundred per cent infection and a low mortality of 10%. There was only a slight variation in the cercarial production.

Rainbow trout were experimentally infected with the ectoparasitic monogenean Gyrodactylus derjavini at three temperatures, 5.5°C, 11.6°C and 18.7°C. Subsequently, the infection level was monitored during the following 6 weeks. The population growth was positively correlated with temperature in the initial part of the investigation. At 18.7°C growth of the parasite population was fast reaching a peak abundance within 3 weeks, whereafter the infection level decreased significantly. Peak abundance was recorded in week 5 at 11.6°C. A very slow but steady parasite reproduction was noted at 5.5°C, but no peak abundance was reached within 6 weeks. The decline in parasite population after reaching peak levels is likely to be caused by a host response. The host response is also indicated by the parasites' active selection of particular microhabitats. This anti-monogenean response developed fastest at 18.7°C, more slowly at 11.6°C and was not recorded within 6 weeks at 5.5°C.

Monogenean richness of marine fish is highly variable among host families and species. On the basis of 160 fish species selected because their parasite fauna was considered as ‘adequately’ investigated, an attempt was made to find associations between parasite richness and various characters of the hosts. No clear correlation was found with maximal size, although the smallest species (less than 10cm) seem to never harbour monogeneans. Correlations were found with nectonic, migrating and gregarious behaviours. However, taxonomy of fish appears to be extremely important as a determinant of monogenean richness. In the absence of a satisfactory phylogeny of hosts, no attempt was made to control the analysis for phylogeny. However, removing some families characterized by a high parasite richness (sparids, sciaenids, mugilids) shows that the association with gregariousness holds, whereas the others tend to disappear.

The plant-parasitic nematodes have this in common with their relatives parasitic in animals, that they are unusually resistant to chemical control. It is not merely that substances lethal to the nematode are harmful to the animal or plant, for in both cases there are usually in the nematode’s life cycle free-living stages, and these are also highly resistant. Apparently the nematode cuticle, egg shell, and vitelline membrane are effective barriers to the penetration of many substances which might be expected to be good contact poisons. Several of the best modern insecticides have given disappointing results against plant eelworms (Ellis and Clayton, 1948). On the other hand some of the simpler halogenated hydrocarbons have been fairly effective, against both animal nematodes (CCl4, C2C14) and plant eelworms in the soil (C2H4Br2, C3H4Cl2).

Eighty three carcasses of pigs were examined at three abattoirs in Moshi, Arusha and Mbulu in northern Tanzania. Taenia solium metacestodes were found in all the three abattoirs with an overall prevalence of 13.3%. During routine meat inspection in Kiboroloni, Moshi 6.2–6.9% of the pigs were found to harbour T. solium metacestodes. The mean number of hooks on the protoscolices was 27 and the length of the small hooks varied from 105 mm to 130 μm while that of the larger hooks varied from 168 mm to 174 mm confirming that the cysts were metacestodes of T. solium. Although cystlike lesions were recovered from livers of both pigs and cattle, no booklets of sizes in the range of 5–37mm were found indicating that no Taiwan Taenia metacestodes were recovered during carcass examination.

A comparison of the characteristics of two strains of S. haematobium in experimental infections in hamsters is made. A limited study of these strains is also made in Mastomys.

Growth changes in male and female adult worms are compared, as are changes in the uterine egg counts.

The quantitative distribution of eggs in the tissues of the hamster is recorded. The approximate mean daily egg output per female worm for each strain is calculated at specified time intervals, and for the entire period of observations.

The recorded characteristics of the two strains of parasite are compared with those described for other strains of S. haematobium.

It is considered that the hamster provides a useful model for such studies provided that standardised infection procedures are employed.

It is considered that differences in host characteristics and susceptibility of individual animals account for the apparent variability recorded with regard to growth of parasites and egg depositions.
