We compared the reliability and bias of genomic evaluation of Holstein bulls for milk, fat, and protein yield with two methods of genomic best linear unbiased prediction (GBLUP) and single-step GBLUP (ssGBLUP). Four response variables of estimated breeding value (EBV), daughter yield deviation (DYD), de-regressed proofs based on Garrick (DRPGR) and VanRaden (DRPVR) were used as dependent variables. The effects of three weighting methods for diagonal elements of the incidence matrix associated with residuals were also explored. The reliability and the absolute deviation from 1 of the regression coefficient of the response variable on genomic prediction (Dev) using GBLUP and ssGBLUP methods were estimated in the validation population. In the ssGBLUP method, the genomic prediction reliability and Dev from un-weighted DRPGR method for milk yield were 0.44 and 0.002, respectively. In the GBLUP method, the corresponding measurements from un-weighted EBV for fat were 0.52 and 0.008, respectively. Moreover, the un-weighted DRPGR performed well in ssGBLUP with fat yield values for reliability and Dev of 0.49 and 0.001, respectively, compared to equivalent protein yield values of 0.38 and 0.056, respectively. In general, the results from ssGBLUP of the un-weighted DRPGR for milk and fat yield and weighted DRPGR for protein yield outperformed other models. The average reliability of genomic predictions for three traits from ssGBLUP was 0.39 which was 0.98% higher than the average reliability from GBLUP. Likewise, the Dev of genomic predictions was lower in ssGBLUP than GBLUP. The average Dev of predictions for three traits from ssGBLUP and GBLUP were 0.110 and 0.144, respectively. In conclusion, genomic prediction using ssGBLUP outperformed GBLUP both in terms of reliability and bias.

Preservation of colostrum for neonatal dairy calves has seldom been seldom in recent years, much of the peer reviewed literature having been published in the 1970s and 1980s. First milking colostrum is high in bioactive immune enhancers such as immunoglobulins, lactoferrins, lysozymes and cytokines and is vital to confer passive immunity to newborn dairy calves to promote their health, welfare and future productivity. Bovine colostrum is advisedly restricted from the bulk milk supply for the first 8 milkings post calving due to high somatic cell counts and the risk of antimicrobial residues. As such, many producers refer to ‘colostrum’ as not only the first milking post calving, but also the aformentioned ‘transition’ milk. Colostrum is preserved in order to protect supply for feeding when production may be poor or where there is a glut of colostrum such as in seasonal calving systems. There are multiple reasons for newborn calves not to have access to their dam's colostrum, including multiple births, acute mastitis or maladapted maternal behaviour, especially in first lactation heifers. Shortages in colostrum may also be precipitated by purposeful discarding of colostrum from cows infected with Mycobacterium avium subsp paratuberculosis and Mycoplasma bovis. Broadly, colostrum may be preserved using low temperature (refrigeration or freezing) or chemical preservatives. The aim of this scoping review article was to identify options for preservation and gaps in research and to propose best practice for colostrum preservation.

The aims of this study were to: (1) estimate genetic correlation for milk production traits (milk, fat and protein yields and fat and protein contents) and fatty acids (FA: C16:0, C18:1 cis-9, LCFA, SFA, and UFA) over days in milk, (2) investigate the performance of genomic predictions using single-step GBLUP (ssGBLUP) based on random regression models (RRM), and (3) identify the optimal scaling and weighting factors to be used in the construction of the H matrix. A total of 302 684 test-day records of 63.875 first lactation Walloon Holstein cows were used. Positive genetic correlations were found between milk yield and fat and protein yield (rg from 0.46 to 0.85) and between fat yield and milk FA (rg from 0.17 to 0.47). On the other hand, negative correlations were estimated between fat and protein contents (rg from −0.22 to −0.59), between milk yield and milk FA (rg from −0.22 to −0.62), and between protein yield and milk FA (rg from −0.11 to −0.19). The selection for high fat content increases milk FA throughout lactation (rg from 0.61 to 0.98). The test-day ssGBLUP approach showed considerably higher prediction reliability than the parent average for all milk production and FA traits, even when no scaling and weighting factors were used in the H matrix. The highest validation reliabilities (r2 from 0.09 to 0.38) and less biased predictions (b1 from 0.76 to 0.92) were obtained using the optimal parameters (i.e., ω = 0.7 and α = 0.6) for the genomic evaluation of milk production traits. For milk FA, the optimal parameters were ω = 0.6 and α = 0.6. However, biased predictions were still observed (b1 from 0.32 to 0.81). The findings suggest that using ssGBLUP based on RRM is feasible for the genomic prediction of daily milk production and FA traits in Walloon Holstein dairy cattle.

Dairy cattle breeding has historically focused on relatively small numbers of elite bulls as sires of sons. In recent years, even if generation intervals were reduced and more diverse sires of sons could have been selected, genomic selection has not fundamentally changed the fact that a large number of individuals are being analyzed. However, a relatively small number of elite bulls are still siring those animals. Therefore inbreeding-derived negative consequences in the gene pool have brought concern. The detrimental effects of non-additive genetic changes such as inbreeding depression and dominance have been widely disseminated while seriously affecting bioeconomically important parameters because of an antagonistic relationship between dairy production and reproductive traits. Therefore, the estimation of benefits and limitations of inbreeding and variance of the selection response deserves to be evaluated and discussed to preserve genetic variability, a significant concern in the selection of individuals for reproduction and production. Short-term strategies for genetic merit improvement through modern breeding programs have severely lowered high-producing dairy cattle fertility potential. Since the current selection programs potentially increase long-term costs, genetic diversity has decreased globally as a consequence. Therefore, a greater understanding of the potential that selection programs have for supporting long-term genetic sustainability and genetic diversity among dairy cattle populations should be prioritized in managing farm profitability. The present review provides a broad approach to current inbreeding-derived problems, identifying critical points to be solved and possible alternative strategies to control selection against homozygous haplotypes while maintaining sustained selection pressure. Moreover, this manuscript explores future perspectives, emphasizing theoretical applications and critical points, and strategies to avoid the adverse effects of inbreeding in dairy cattle. Finally, this review provides an overview of challenges that will soon require multidisciplinary approaches to managing dairy cattle populations, intending to combine increases in productive trait phenotypes with improvements in reproductive, health, welfare, linear conformation, and adaptability traits into the foreseeable future.

We compared the potential of dietary lipid supplements of different fatty acid compositions to affect milk performance when early lactation dairy goats were fed a high-concentrate diet. Thirty Alpine goats at 23 ± 5 d in milk were allocated to 1 of 10 blocks according to parity and milk fat concentration. Within each block, goats were randomly assigned to receive, during a period of 41 d, either CONT) a basal diet with a forage to concentrate ratio of 45:55, used as control, or PALM) the basal diet + 2% of a palmitic acid-enriched fat supplement, or FLAX) the basal diet + 7% of extruded flaxseed. Body weight, dry matter intake and milk yield were not different between treatments. As compared with CONT, goats fed PALM and FLAX had a greater milk fat concentration. Moreover, milk fat yield was numerically (but non-significantly) greater with PALM than with CONT. Milk fat from goats receiving PALM had a greater concentration of 16:0 as compared with CONT and FLAX, whereas a greater concentration of cis-9, cis-12, cis-15 18:3 was observed when goats were fed FLAX as compared with CONT and PALM. Under the conditions of the current experiment, dietary fat supplementation had only minor impacts on the yield of major milk constituents, with the exception of a modest increase in fat yield when goats were fed PALM. The impact of a greater concentration of 16:0 in milk fat of goats receiving this feed ingredient on the nutritive value of dairy products remains to be determined.

Recently, a selection index called Valor Económico Lechero (VEL) was developed for Chilean dairy cattle under pasture. However, a specific selection scheme has not yet been implemented. This study aimed to estimate genetic progress from selection on the VEL selection index based on selection schemes using progeny testing (PT) and genomic selection (GS). Under a PT-scheme, estimated genetic progress was 41.50, 3.44, and 2.33 kg/year for milk, fat, and protein yield, respectively. The realised genetic gain takes eight-year after the PT-scheme implementation, which may be a disincentive for implementing a PT-scheme, suggesting that importing frozen semen of proven bulls could be a preferred alternative. In this case, an option may be to conduct the genetic evaluation of those bulls using their progeny in Chile for the traits included in VEL selection index. In the case of implementing a specific selection scheme, compared to PT, a more profitable alternative might be the implementation of a GS-scheme, that would result in a faster genetic gain in the aggregate breeding value or merit for all the traits included in the selection objective (0.323–0.371 vs. 0.194 σg/year).

Ranked amongst the top five milk-producing countries globally, the Pakistan dairy industry can help to overcome food shortage and hunger, alleviate poverty and positively impact economic growth. This influencing role could potentially be more significant while the COVID-19 pandemic severely affects humanity, challenges the economy and increases the risk of global food shortage. However, its large national population of dairy livestock contrasts with Pakistan's top-five ranking, indeed, four to five Pakistani cows produce milk equivalent to one dairy cow of countries with a well-developed dairy industry. Low milk yield per cow negatively impacts the national production and compromises the development of an efficient processing sector, such that consumers are very often forced to use adulterated milk sold by local ‘milkmen.’ As a consequence, while committed to alleviating global hunger, Pakistan imports in excess of half a million tons of milk and milk-based products annually. Many studies have identified unproductive, inefficient and imprecise management issues combined with poor genetics and imbalanced nutrition as the leading barriers to improvement in the Pakistani dairy livestock sector. At an individual level, lack of awareness, affordability issues, illiteracy and low ambition of a large percentile of dairy farmers creates additional significant barriers. To address low productivity and poor genetics, Pakistani corporate farms and wealthier individual farmers import genetically improved breeds to attain high milk yields. However, they are then faced with the challenge of managing such breeds to attain sustainable and persistent milk yields under Pakistani climatic conditions, often risking excessive culling even to the point of business liquidation. In developed dairy industries, automated sensor-based livestock management systems are now available to help monitor, compute, and optimize procedures in real-time and are proven to increase productivity and profitability. The term precision livestock farming (PLF) is used to describe systems that monitor individual animals or groups of animals to overcome management deficiencies and optimize productivity. My stance in this Opinion Paper is that adopting and utilizing such precision technologies may support Pakistan in raising its livestock resources toward greater productivity, thereby helping to overcome the global food shortage.

The study in this research paper was undertaken with a hypothesis that accelerometer data can be used to improve monitoring of energy balance in dairy cows. Animals of high (select, S) and average (control, C) genetic-merit lines were allocated to two feeding systems, by-product (BP) and homegrown (HG). This culminated in four production systems referred to as BPS, BPC, HGS and HGC. Cows between their first and fourth lactations were included and a total of 8602 records were used. The target crude protein (CP) and metabolisable energy (ME) content in the BP diet was 185 g/kg DM and 12.3 MJ/kg DM while it was 180 g/kg DM, and 11.5 MJ/kg DM for the HG diet, respectively. Milk yield, body energy content (BEC) and animal activity were monitored while the animals were all housed for winter. Results showed that cows on homegrown feeds were significantly (P < 0.05) more active than cows on by-product feeds as indicated by higher motion index and number of steps per day. Feeding duration was not significantly different (P > 0.05) between cows under by-product feeding system irrespective of the energy balance of the cows. However, there were significant differences for cows under homegrown feeding system. Cows in negative energy balance had a longer feeding duration per day than cows in positive energy balance. Milk yield was negatively correlated (P < 0.05) to motion index and number of steps per day but not to lying time and feeding duration. The results showed differences in cow activity were related to diet content and body energy status. This is useful in precision farming where feeds are provided according to specific animal behaviour and feed requirements.

Bakery by-products (BP), rich in fats and sugars, are unconventional feed sources for cows whose effects on milk fat composition have not yet been evaluated. This research paper aimed to assess the effects of dietary BP inclusion rate and feeding period on the milk fatty acid composition. Twenty-four Simmental cows were fed a diet without BP (CON) for 1 week. Then they either continued with the CON diet or switched to one of the BP diets (with 15% or 30% BP in diet dry matter) for 3 weeks. Milk samples were taken before diet change and three times during BP feeding and analysed for fatty acid composition. Data showed that increasing BP content in the diet increased total fatty acid intake, especially of 18 : 1 n9. In the milk fat, the percentages of total monounsaturated fatty acids especially of the 18 : 1 origin linearly increased with increasing dietary BP level. The percentage of fatty acids de novo synthesized in the mammary gland (the sum of 4 : 0–14 : 0) remained similar among diets (32-34% of total fatty acids). The 16 : 0 percentage dropped from 32.5 to 29.6% and from 33.6 to 28.3% for 15% and 30% BP, respectively. Only 30% BP elevated the percentage of conjugated linoleic acids (CLA: by 59%) compared with CON throughout the 3 weeks. Proportions of 18 : 2 n6 and 18 : 3 n3 and the n6:n3 ratio were unaffected by BP and feeding time. BP feeding improved all those estimated health indices of the milk fat that are suggested to be related to coronary health. In summary, the inclusion of BP in dairy rations beneficially shifted the milk fatty acid profile to more 18 : 1 fatty acids at the expense of 16 : 0. At a 30% inclusion rate, BP feeding showed an additional benefit of increased CLA content in milk fat.

This study assessed the impact of improved green fodder production activities on technical efficiency (TE) of dairy farmers in climate vulnerable landscapes of central India. We estimated stochastic production frontiers, considering potential self-selection bias stemming from both observable and unobservable factors in adoption of fodder interventions at farm level. The empirical results show that TE for treated group ranges from 0.55 to 0.59 and that for control ranges from 0.41 to 0.48, depending on how biases are controlled. Additionally, the efficiency levels of both adopters and non-adopters would be underestimated if the selectivity bias is not appropriately accounted. As the average TE is consistently higher for adopter farmers than the control group, promoting improved fodder cultivation would increase input use efficiency, especially in resource-deprived small holder dairy farmers in the semi-arid tropics.

This study evaluated the effect of replacing soybean meal by a blend of ground corn and urea-ammonium sulphate (GCU-S) in the diet of lactating buffaloes on milk production and composition, digestibility, N balance and blood metabolites. Twelve multiparous dairy Murrah buffaloes (Bubalus bubalis), at 100 ± 4 d in milk and yielding 10 ± 2.5 kg/d, were randomly distributed in a triple 4 × 4 Latin square, with four different inclusions of GCU-S at U-S levels: 0 (control), 8.0, 16.4 and 24.1 g/kg dry matter (DM) total. Replacing soybean meal with GC-US had a significant depressing effect on absolute DM intake, which was still numerically evident but no longer significant when expressed on a body weight-related basis. Intakes of crude protein (CP) and N as well as N-urinary excretion were also significantly depressed. Digestibility of dry matter was improved and, as a proportion of DMI, intake of total digestible nutrients increased significantly. Perhaps as a result, feed efficiency (kg DMI required per kg 6% fat-corrected milk) was significantly improved. Efficiency of protein use for milk production was significantly improved, but not when expressed as milk protein output. N transfer into milk, as a proportion of total N intake, also increased significantly. However, GC-US inclusion had no significant effect on milk production or milk composition, nor did it affect serum metabolites, digestibilities (apart from dry matter) or N measures of balance apart from those mentioned above. Inclusion of this blend of ground corn with urea and ammonium suplate can be recommended for dairy buffalo because, although it decreases DMI and N-excretion, it improves DM digestibility and feeding efficiency whilst maintaining milk production and composition.

This study aimed to conduct a meta-analysis based on a random-effects model to combine different published heritability estimates and genetic correlations for milk and serum minerals in dairy cows. In total, 59 heritability and 25 genetic correlation estimates from 12 articles published between 2009 and 2021 were used. The heritability estimates for milk macro-minerals were moderate to high and ranged from 0.311 (for Na) to 0.420 (for Ca). On the other hand, milk micro-minerals had lower heritabilities with a range from 0.013 (for Fe) to 0.373 (for Zn). The heritability estimates for serum macro-minerals were generally low and varied from 0.126 (for K) to 0.206 (for Mg). The estimates of genetic correlation between milk macro-minerals varied from −0.024 (between Na and K) to 0.625 (between Mg and P). The genetic correlations of milk Ca and P with milk yield were −0.171 and −0.211, respectively. The estimates of genetic parameters reported in this meta-analysis study are appropriate to utilize in breeding plans when valid estimates are not available for milk minerals in dairy cow populations.

The present study assesses the sensitivity of dairy animals to thermal stress, and projects the economic losses due to heat stress in the Trans and Upper Gangetic plains region of India with Representative Concentration Pathway (RCP) 4.5 climate scenario for the time slice 2010–2039 and two subperiods, 2020–2029 and 2030–2039. The projections were carried out for two different scenarios of population and productivity growth of dairy animals, Business-as-Usual (BAU) and Alternate, whereby land, feed and fodder constraints were applied. The potential annual loss in milk production due to heat stress in the region was estimated to be around 361 and 377 thousand tons for the time slice 2010–2039 under BAU and Alternate scenario, respectively. In economic terms these losses, at current prices, would be equivalent to INR 11.93 billion and INR 12.44 billion, respectively. This gives an indication of the level of financial investment that can be made in adaptation measures to arrest the loss due to climate change.

This experiment aimed to investigate the effects of inulin supplementation on milk production and composition, feed intake, nutrient digestibility and rumen fermentation parameters in lactating ewes. The experimental treatments were (1) control group (basal diet), (2) basal diet plus 2% inulin (w/w) and (3) basal diet plus 4% inulin (w/w). The experiment was carried out for 21 d in a fully randomized design involving eighteen Ghezel ewes. Production and composition (percentages of fat, protein, lactose and fat-free solids and fatty acid profiles) of milk were measured. Faeces were collected in the last 3 days of the experiment to determine digestibility. On the last day of the experiment, rumen fluid samples were taken from the esophagus 3 h after feeding and fermentation parameters (pH, ammonia nitrogen (N-NH3), volatile fatty acids (VFA) and protozoal population) were examined. Daily milk production was not significantly affected by inulin supplementation, but the fat and protein content of the milk was increased whilst urea nitrogen (MUN) and unsaturated fatty acids were decreased (P < 0.05). The dry matter (DM) intake results showed that there was no significant difference between different diets. The highest digestibility of DM and NDF belonged to the inulin fed group (P < 0.05). Inulin consumption numerically increased the pH of the rumen fluid of the animals and significantly decreased the rumen N-NH3 value (P < 0.05). Inulin supplementation also significantly increased total VFA, acetate, and butyrate levels (P < 0.05). In general, it can be concluded that inulin supplementation can improve rumen fermentation, DM and NDF digestibility,as well as compositional aspects of the ewe's milk production.

The form of oral calcium (Ca) supplement and the Ca source influence Ca absorption dynamics resulting in different postpartum calcemia. The objective of this study was to investigate whether an oral Ca supplement (mainly CaCO3) offered for voluntary consumption would maintain or increase postpartum blood Ca to the same degree as a Ca bolus (mainly CaCl2) providing an equivalent dose of a Ca. A total of 72 Holstein cows were blocked by expected parturition date and parity. Within each block of 3 animals, cows were randomly assigned to one of three treatments, including an oral Ca supplement offered for voluntary consumption (Ca-drink, n = 23), an oral Ca bolus (Ca-bolus, n = 24), or an untreated group (CON, n = 25). Treatments were administered once within 15 min postpartum. The Ca-drink provided 45 g of Ca (CaCO3 source) and was mixed in 20 L of lukewarm water and offered to cows for 30 min. The Ca-bolus provided 43 g of Ca (71% from CaCl2 and 29% from CaSO4) and was administered once. Both Ca-bolus and CON cows received 20-l of lukewarm water at parturition to standardize the volume of fluids (Ca-drink or 20-l lukewarm water) offered at parturition. Dairy cows offered Ca-drink had a 28% higher fluid consumption than Ca-bolus and CON cows. Milk yield and milk composition expressed in percentage protein, fat, lactose, and urea did not differ, whilst there was a small but significant increase in DMI in cows receiving the Ca-drink compared to CON, while Ca-bolus did not differ from other groups. This was consistent with reduced BW losses between week 1 and 3 in cows receiving the Ca-drink suspension. Treatment by time interactions were present for blood Ca, glucose, and urea concentrations. Blood Ca was relatively stable in Ca-drink cows, while higher fluctuations were observed in Ca-bolus cows. In Ca-bolus cows, blood Ca increased from 15 min to 6 h, decreased from 6 to 24 h, and finally increased again from 24 to 48 h. At 24 h post administration, blood Ca was greater in cows receiving the Ca-drink than cows receiving the Ca-bolus. Blood glucose was greater in Ca-bolus cows at 15 min after treatment administration compared with Ca-bolus and CON, while blood urea was higher in CON than Ca-drink and Ca-bolus throughout the sampling period. These results indicate that voluntary oral Ca resulted in a relatively stable calcemia, whereas higher fluctuations were observed in cows receiving the Ca-bolus. Due to a lack of differences between Ca-drink and Ca-bolus compared with CON, it is not possible to conclude regarding the efficacy in maintaining postpartum blood Ca.

CXCL8 (also known as IL-8) is a member of the CXC subfamily of chemokines that binds two of the seven transmembrane G-protein-coupled receptors (GPCRs), CXCR1 and CXCR2, to mediate and regulate leucocyte accumulation and activation at sites of inflammation. They are known to play a critical role in both disease susceptibility and infection outcome. The aim of this study was to investigate the entire sequences of CXCL8 and CXCR2 genes in thirty-one Simmental sires to evaluate the effects of genomic variants on the indexes of the bulls for milk, fat and protein yields, and for somatic cell score (SCS). Five new single nucleotide polymorphisms (SNPs) were found in CXCR2 gene. The analysis of association indicated that one SNP in CXCL8 and two in CXCR2 influenced the considered traits. To evaluate the existence of functional haplotypic effects, combinations among the three genomic variants (SNP 1 in CXCL8, SNP 6 and SNP 7 in CXCR2) were investigated. Four different haplotypic alleles were identified in the experimental population, one of which at a high frequency (61%). Bulls with Hap 4 (G-C-G at SNP 1, SNP 6, and SNP 7 respectively) had more favourable indexes for SCS (P < 0.05). These results suggest that the SNPs in CXCL8 and CXCR2 may be potential genetic markers to improve udder health in the Simmental breed.
