Polyacrylamide gel electrophoresis of cell proteins was investigated as a possible typing method for Corynebacterium diphtheriae. A method was developed using stock strains which were representatives of the five gravis serotypes described by Robinson & Peeney (1936). This technique was then applied to recent isolates sent to our laboratory for identification.

Cattle and sheep were housed with infected pigs for 11 days. Small amounts of virus were recovered intermittently from the pharynx, milk and rectal swabs of the cattle, but no evidence of subclinical infection was found. Some indication of virus growth in the sheep was obtained in that large amounts of virus were recovered from the pharyngeal region 4 to 7 days after exposure and six of the eight sheep developed significant titres of neutralizing antibody which were maintained in four animals for at least 6 weeks.

A prospective longitudinal study was conducted on 96 smallholder duck farms in Indonesia over a period of 14 months in 2007 and 2008 to monitor bird- and flock-level incidence rates of H5 highly pathogenic avian influenza (HPAI) infection in duck flocks, and to identify risk factors associated with these flocks becoming H5 seropositive. Flocks that scavenged around neighbouring houses within the village were at increased risk of developing H5 antibodies, as were flocks from which carcases of birds that died during the 2 months between visits were consumed by the family. Duck flock confinement overnight on the farm and sudden deaths of birds between visits were associated with lower risk of the flock developing H5 antibodies. Scavenging around neighbouring houses and non-confinement overnight are likely to be causal risk factors for infection. With this study we have provided insights into farm-level risk factors of HPAI virus introduction into duck flocks. Preventive messages based on these risk factors should be included in HPAI awareness programmes.

Non-steroidal anti-inflammatory drugs (NSAIDs) may inhibit antibody production by peripheral blood mononuclear cells; one consequence of this could be decreased effectiveness of vaccines in NSAID users. Because many older adults use low-dose aspirin for primary or secondary prevention of coronary events, any inhibitory effect of aspirin on vaccine immune response could reduce the benefits of vaccination programmes in older adults. We tested whether immune response to vaccination differed between users vs. non-users of low-dose aspirin, using data from four randomized trials of monovalent 2009 pandemic influenza A(H1N1) vaccine. Geometric mean haemagglutination inhibition antibody titres were not significantly lower in low-dose aspirin users compared to non-users. Our results provide reassurance that influenza vaccination effectiveness is probably not reduced in older adults taking chronic low-dose aspirin.

Pasteurisation at 62.8° C. (145° F.) for 30 minutes leads to a marked reduction in the total bacterial content of milk, varying for different specimens from 94 to 99·8 per cent., when the process is carried out under the most exact conditions. At lower temperatures, e.g. 60° C., the percentage reduction may vary widely—from 51·6 to 97·4 per cent. The percentage reduction has been found to be slightly less when the same specimens of milk are pasteurised in a large scale apparatus than when treated in small quantities under laboratory conditions, the average reduction being 91·2 per cent, in the former case and 98·4 per cent, in the latter. The reasons for this are discussed.

An effectively pasteurised milk should not contain lactose-fermenting bacilli in 1 c.c, and the B. coli content is a valuable index of the efficiency of a pasteurising process.

Milk containing B. tuberculosis, when pasteurised at 62·8° C. for 30 minutes (1) in tubes in a water bath in the laboratory, and (2) on a large scale by a commercial plant, is rendered non-infective to guinea-pigs. By laboratory pasteurisation, even at 60° C. for 30 minutes, milk containing tubercle bacilli is rendered non-infective.

Five pigs were treated orally with norfloxacin for 5 consecutive days in two well-separated periods. This was done to determine the lowest dose required to free the pigs of Enterobacteriaceae. In the first period of the study, the animals were treated with 400 mg per day while in the second treatment 800 mg norfloxacin was given. Daily faecal culturing indicated that the faeces became free of Enterobacteriaceae in 3–5 days when treated with 400 mg/day, while all animals were found negative on culturing after 3 days of treatment with 800 mg/day. Investigation of the concentration of norfloxacin in the faeces revealed that a substantial fraction of the dose was either absorbed or inactivated by faecal substances. An in vitro study in faeces confirmed that a substantial part, some 75% of the dose, may have been inactivated by intestinal contents. This finding helps to explain the much lower concentration of 120 mg norfloxacin per kg of faeces in the pig in comparison with the almost tenfold higher concentration reported to develop in man during treatment with an identical dose.

In the present study rubella HI antibodies were determined in cases of congenital malformation and bad obstetric history to determine the role of rubella in such cases in India, as it has been reported to be rare in Japan in contrast to Western countries. The incidence of antibodies was statistically significant in cases of congenital eye, c.n.s., visceral and miscellaneous malformations and cases of spontaneous abortion and still-birth compared with controls of matching age groups. This is further supported by the demonstration of IgM antibodies in seven cases of congenital malformations and ten cases of spontaneous abortion. Our findings show that congenital rubella is not rare in India.

Studies of several cultures of Salmonella typhi-murium var. binns revealed the fact that all contained specific components characteristic of S. typhi-murium. These cultures lacked the somatic factor V. It is proposed, therefore, to emend the antigenic formula of S. typhi-murium var. binns from IV, V: —: 1, 2, 3, to IV: i: 1, 2, 3. Under the emended variety would also be included the organisms described by Kauffmann as S. typhi-murium var. copenhagen and by Edwards as S. aertrycke var. storrs.

The analysis of the serological results of blood specimens submitted during the course of the past 2 years for the Widal reaction has been made.

The sera from cases of typhoid and paratyphoid fever were examined in particular for their agglutinin content for the “H” and “O” antigens of B. typhosus and B. paratyphosus B. The results obtained indicate that it is essential to use the “O” antigen of B. typhosus, but no particular advantage has been derived, from the point of view of early diagnosis, from the use of B. paratyphosus B “O” antigen.

A comparison of the value of suitable living and killed antigens has been made. Either type of antigen appears to be equally effective, but for convenience and stability the suspensions issued by the Standards Laboratory, Oxford, have proved entirely satisfactory.

Cases showing the serological difficulties which arise when a mixed specific and non-specific form of B. paratyphosus B “H” antigen is used are described.

Group-A streptococci belonging to opacity-factor (OF)-positive M types were poorly haemolytic on horse-blood agar, but members of OF-negative M types, and M-negative variants of OF-positive strains gave good haemolysis. Horse-serum extracts of strains of OF-negative serotypes 6 and 12, and M-negative variant cultures of OF-positive serotypes 4 and 49, had higher titres of streptolysin S than did similar extracts of OF-positive, M-positive cultures of types 4 and 49. However, much larger amounts of streptolysin S could be extracted with ribonuclease (RNAase)-digested yeast ribonucleic acid (RNA) and M-positive OF-positive cultures treated in this way gave extracts at least as strong as did their M-negative variants or the OF-negative strains. Extraction of streptolysin S from OF-negative strains by serum could be inhibited by previous incubation of the serum with extracellular OF, suggesting that the production of diffusable OF by M-positive variants of OF-positive serotypes interferes with the extraction of streptolysin S by serum and leads to poor haemolysis on blood agar.

The haemolysis of all strains on blood agar was greatly improved by the incorporation of 0·1% (w/v) RNAase-digested yeast RNA into the medium, but the improvement was most marked in OF-positive serotypes.

In 2016, we reviewed preventive control measures for secondary transmission of Shiga-toxin producing Escherichia coli (STEC) in humans in European Union (EU)/European Free Trade Association (EEA) countries to inform the revision of the respective Norwegian guidelines which at that time did not accommodate for the varying pathogenic potential of STEC. We interviewed public health experts from EU/EEA institutes, using a semi-structured questionnaire. We revised the Norwegian guidelines using a risk-based approach informed by the new scientific evidence on risk factors for HUS and the survey results. All 13 (42%) participating countries tested STEC for Shiga toxin (stx) 1, stx2 and eae (encoding intimin). Five countries differentiated their control measures based on clinical and/or microbiological case characteristics, but only Denmark based their measures on routinely conducted stx subtyping. In all countries, but Norway, clearance was obtained with ⩽3 negative STEC specimens. After this review, Norway revised the STEC guidelines and recommended only follow-up of cases infected with high-virulent STEC (determined by microbiological and clinical information); clearance is obtained with three negative specimens. Implementation of the revised Norwegian guidelines will lead to a decrease of STEC cases needing follow-up and clearance, and will reduce the burden of unnecessary public health measures and the socioeconomic impact on cases. This review of guidelines could assist other countries in adapting their STEC control measures.

The prevalence of antibodies against Entamoeba histolytica was studied in the Mexican population using an immunoenzyme assay in solid phase (ELISA) and semiautomatic equipment. The antigen was a mixture of membrane proteins obtained by Triton X-100 extraction from an axenic culture of Entamoeba histolytica HM1-IMSS. The method was standardized by comparing serum samples from amoebic liver abscess patients with healthy volunteers. From the 60538 samples supplied by the National Seroepidemiology Survey, antibodies were found in 4·49 % (4·32–;4·65% at 95 % confidence limit). More significant titres occurred in the central region of the country. The ratio female to male was 1·25:1. The population living in metropolitan areas had probably been infected at a younger age than those living in the country.

Important differences were found in the seroprevalence obtained by ELISA compared with a study which used indirect haemagglutination (IHA) in the same sample frame.

Eight hundred and seventy-seven sera from 360 adults aged 18–50 who were under permanent observation from October 1980 to March 1981 have been studied by haemagglutination-inhibition (HI) and erythrocyte elution-inhibition (EI) tests – a simplified method of antineuraminidase antibody titration. It was demonstrated in some subjects infected with influenza A H1N1 and H3N2 viruses that the antibody rise was to one of the surface antigens only – haemagglutinin or neuraminidase. These subjects made up 5·2–25·8% of all examinees. The protective effect of antibodies to neuraminidase was similar to that of antihaemagglutinins. Interaction of both types of antibodies was observed in protection against the disease. Data have been obtained on the influence of antineuraminidase antibodies in decreasing the severity of natural infection with influenza A.

A study of heterologous immunologic responses to haemagglutinin and neuraminidase among persons immunized with live influenza A H1N1 and H3N2 vaccines and among children naturally infected with influenza A H3N2 demonstrated the presence of immunologic memory for antineuraminidase antibody synthesis. Thus, the suggestion of a common antigenic structure for neuraminidase Nl and N2 is made.

Serum cholesterol in rabbits was elevated, by intravenous inoculation of 20 % Tween 80, to amounts thought to have been sufficient to support growth of Mycoplasma gallisepticum when 20 % of such serum was added to rabbit infusion broth.

However, better growth of the organism was obtained with a supplement of 5 % rabbit serum and commercial cholesterol. Investigation showed that normal rabbit serum may be inhibitory, which would explain these findings. The commercial cholesterol was not antigenic.

Studies were made in a modified hospital ward containing 19 beds, 14 of them in the open ward, one in a window-ventilated side-room, two in rooms with partial-recirculation ventilators giving 7–10 air changes per hour, and two in self-contained isolation suites with plenum ventilation (20 air changes per hour), ultra-violet (UV) barriers at doorways and airlocks.

Preliminary tests with aerosols of tracer bacteria showed that few bacteria entered the plenum or recirculation-ventilated rooms. Bacteria released inside mechanically ventilated cubicles escaped into the corridor, but this transfer was reduced by the presence of an airlock. UV barriers at the entrance to the airlock and the cubicle reduced the transfer of bacteria from cubicle to corridor.

During a period of 4 years while the ward was in use for surgical and gynaecological patients, the incidence of post-operative sepsis and colonization of wounds by multiple-resistant Staphylococcus aureus was lower (though not significantly lower) in the plenum-ventilated rooms than in the open ward, the recirculator-ventilated cubicles and the window-ventilated cubicles. Nasal acquisition of multiple-resistant Staph. aureus was significantly less common in the plenum-ventilated than in the recirculator-ventilated cubicles and in the other areas. Mean counts of bacteria on settle-plates were significantly lower in the plenum-ventilated cubicles than in the other areas; mean settle-plate counts in the recirculator-ventilated cubicles were significantly lower than in the open ward and in the window-ventilated side-room; similar results were shown by slit-sampling of air. Mean settle-plate counts were significantly lower in all areas when the ward was occupied by female patients. Staph. aureus was rarely carried by air from plenum-ventilated or other cubicles to the open ward, or from the open ward to the cubicles; though staphylococci were transferred from one floor area to another, they did not appear to be redispersed into the air in sufficient numbers to infect the patients. Ultra-violet irradiation caused a significant reduction in the total and staphylococcal counts from the floors of airlocks, and a significant reduction of total counts in the air.

An examination was made of the relations between antigen, antibody and fixation of complement with foot-and-mouth disease virus (FMDV). It was found that complement fixation in this system follows the same principles as models developed in other antigen/antibody systems. The assumption that there is a relation of direct proportionality between the amount of complement fixed and the amount of antiserum reacting with constant antigen was found to be incorrect. An alternative method was proposed for the quantitative differentiation of FMDV strains by comparing the titres of an antiserum when reacting with optimum amounts of homologous or heterologous antigens.

Captive, healthy, adult badgers have blood containing haemoglobin at 13·3 g/dl, and 8·4×1012/l red cells with an MCV of 46·2 f1 and an MCH of 15·6 pg. They have 5·1×109 white cells/1 of which 3·29×109 are polymorphs, 1·49×109 are lymphocytes, 0·26×109 are monocytes, 0·07×109 are eosinophils and 0·01×109 are basophils. These values are somewhat less in adult animals just trapped from the wild, and are lower still in wild cubs.

Changes associated with tuberculosis are a rise, and then a fall in red blood count and white blood count, an increase in the proportion of polymorphs and momocytes and a fall in lymphocytes late in the disease. This picture is similar to that seen in widespread, disseminated, tuberculin negative, tuberculosis in humans, a type of disease similar to that occurring in many badgers.

BCG vaccination of badgers did not produce any measurable change in the blood picture.

The investigation of an outbreak of cryptosporidiosis in a hospital day-care centre is reported. Twenty-eight (27%) children and one member of the staff were infected. Most of the cases were symptomatic and the major presenting symptom was watery diarrhoea. Shedding of oocysts continued for a mean of 12 days after diarrhoea had subsided and most of the cases were still excreting oocysts when the first follow-up sample was examined. Intermittent shedding or a carrier state were not seen in any of the cases. Giardia intestinalis cysts were detected in 19 children, 7 of whom showing mixed infection with Cryptosporidium sp., but this association was not statistically significant. All cases recovered without specific therapy. No definite proof could be found to demonstrate the source of the outbreak or the route of transmission but some observations suggesting person-to-person transmission are discussed.

We investigated the presence of Mycobacterium spp. in livestock in northern China. Of the 163 clinical samples selected for this study, 20 were from throat swabs of dairy cows, and 143 were tissue samples (including lung tissue from one reindeer, hilar lymph node tissue from 55 cows, and liver tissue from 87 sheep). A total of 41 mycobacterial isolates were identified including two isolates of M. caprae and 39 non-tuberculous mycobacteria (NTM) isolates. Multi-locus variable-number tandem repeat analysis (MLVA) profiles of the two M. caprae isolates proved to be unique. This is the first report of M. caprae isolates from livestock in China. This study also confirms previous reports that NTM is common in livestock in northern China.

COVID-19 research has been produced at an unprecedented rate and managing what is currently known is in part being accomplished through synthesis research. Here we evaluated how the need to rapidly produce syntheses has impacted the quality of the synthesis research. Thus, we sought to identify, evaluate and map the synthesis research on COVID-19 published up to 10 July 2020. A COVID-19 literature database was created using pre-specified COVID-19 search algorithms carried out in eight databases. We identified 863 citations considered to be synthesis research for evaluation in this project. Four-hundred and thirty-nine reviews were fully assessed with A MeaSurement Tool to Assess systematic Reviews (AMSTAR-2) and rated as very low-quality (n = 145), low-quality (n = 80), medium-quality (n = 208) and high-quality (n = 151). The quality of these reviews fell short of what is expected for synthesis research with key domains being left out of the typical methodology. The increase in risk of bias due to non-adherence to systematic review methodology is unknown and prevents the reader from assessing the validity of the review. The responsibility to assure the quality is held by both producers and publishers of synthesis research and our findings indicate there is a need to equip readers with the expertise to evaluate the review conduct before using it for decision-making purposes.
