Martin placed the lateral embryo, which occurs only in grasses, adjacent to the broad embryo at the base of his family tree of seed phylogeny. Since Poales and Poaceae are derived monocots, we questioned the evolutionary relationship between the lateral embryo and other kinds of monocot embryos. Information was compiled on embryo and seed characteristics for the various families of monocots, kind of embryogenesis for families in Poales and germination morphology of families with lateral (only Poaceae) and broad embryos. The kinds of monocot embryos are broad, capitate, lateral, linear fully developed, linear underdeveloped and undifferentiated, but only broad and lateral embryos are restricted to Poales. Asterad embryogenesis occurs in Poaceae with a lateral embryo and in Eriocaulaceae, Rapataceae and Xyridaceae with a broad embryo. In developing grass seeds, the growing scutellum (cotyledon) pushes the coleoptile, mesocotyl and coleorhiza to the side. In the organless broad embryo, the cotyledonary sector is larger than the epicotyledonary sector. During germination of grass seeds, the coleorhiza and then the coleoptile emerge, while in a seed with a broad embryo the elongating cotyledon pushes the epicotyledonary sector outside the seed, after which a root–shoot axis is differentiated at a right angle to the cotyledon inside the seed. Broad and lateral embryos are closely related; however, the lateral embryo is more advanced in seed/embryo traits and germination morphology than the other kinds of monocot embryos, suggesting that its position on the family tree of seed phylogeny should be higher than of the other monocot embryos.

Our study aimed to establish the response of Salminus franciscanus to hypophysation and describe the main morphological events of its embryonic process. Wild fish were captured in São Francisco River and selected broodstock (females: 66.4 ± 11.1 cm and 4.04 ± 2.32 kg; males: 58.3 ± 10.2 cm and 3.62 ± 1.12 kg) were kept at 26.1 ± 0.6°C for induction of final maturation/gamete release via the hypophysation technique. In females, two doses (0.8 and 5.6 mg/kg body weight) of crude pituitary extract of common carp (Cyprinus carpio) were administered with a 14 h interval. For males, a single dose (2.7 mg/kg body weight) of crude pituitary extract was applied at the same time as the females’ second dose. Oocytes and sperm were manually stripped 8 h after a females’ second hormonal dose. Fertilization was carried out using the dry method. Eggs were kept in funnel-type 60 L incubators at 24.3 ± 0.3°C and were analyzed and photographed every 10 min. After hormonal induction, 60% of females and 100% of males reacted positively and no broodstock mortality was recorded. The females released an average of 385.2 ± 78.4 g of oocytes and the fertilization rate observed was 50.4 ± 12.3%. The blastopore closure occurred at 7.5 h, somite formation at 12 h and hatching at 20 h post-fertilization. In general, the results of this study improve the understanding of the reproductive biology of dourado and confirm its potential for fish farming in the neotropical region.

This study describes the embryonic development of Moenkhausia oligolepis in laboratory conditions. After fertilization, the embryos were collected every 10 min up to 2 h, then every 20 min up to 4 h, and afterwards every 30 min until hatching. The fertilized eggs of M. oligolepis measured approximately 0.85 ± 0.5 mm and had an adhesive surface. Embryonic development lasted 14 h at 25ºC through the zygote, cleavage, blastula, gastrula, neurula, and segmentation phases. Hatching occurred in embryos around the 30-somites stage. The present results contribute only the second description of embryonic development to a species from the Moenkhausia genus, being also the first for this species. Such data are of paramount importance considering the current conflicting state of this genus phylogenetic classification and may help taxonomic studies. Understanding the biology of a species that is easily managed in laboratory conditions and has an ornamental appeal may assist studies in its reproduction to both supply the aquarium market and help the species conservation in nature. Moreover, these data enable the use of M. oligolepis as a model species in biotechnological applications, such as the germ cell transplantation approach.

Housekeeping genes (HKG) are paramount for accurate gene expression analysis during preimplantation development. Markedly, quantitative reverse transcription polymerase chain reaction (RT-qPCR) in ovine embryos currently lacks HKGs. Therefore, we tested 11 HKGs for RT-qPCR normalization during ovine parthenogenetic preimplantation development. Seven HKGs reached the qPCR efficiency threshold (97.20–105.96%), with correlation coefficients ranging from −0.922 to −0.998 and slopes from −3.22 to −3.59. GeNorm ranked glyceraldehyde 3-phosphate dehydrogenase (GAPDH) and TATA-binding protein (TBP) as the best HKG pair, while H3 histone, family 3A (H3F3A) was the third HKG. Relative gene expression was measured for zinc finger protein X-linked (ZFX) and developmental pluripotency-associated 3 (DPPA3) transcripts during ovine parthenogenetic preimplantation development. ZFX did not show any transcript abundance fluctuation among oocytes, cleavage-stage embryos, and morulae. DPPA3 transcript abundance was also similar among all developmental stages, therefore suggesting that it may not display a maternal gene expression profile. In silico analysis of ovine DPPA3 mRNA and protein showed high conservation to bovine orthologues. However, DPPA3 orthologues differed in regulatory motifs. In conclusion, GAPDH, TBP and H3F3A are stable HKGs in ovine parthenogenetic embryos and allow accurate RT-qPCR-based gene expression analysis.

This study investigated the breeding parameters and embryogenic development of diploid and heat shock-induced triploid eggs of Anabas testudineus (Bloch, 1792). To this effect, broodstocks of A. testudineus were induced to spawn using the Ovaprim® hormone. After fertilization, the eggs were divided into two groups and one portion heat shocked at 41°C (for 3 min), at approximately 4 min after fertilization. Results of fertilization, hatchability, as well as the sequence and timing of embryogenic development were collated from three breeding trials. Fertilization percentages were similar in both treatments (≈90%) while hatchability was higher in the diploid eggs (79.56%) than the triploid induced eggs (50.04%). Both treatments had the same sequence of embryogenetic stages; however, the timing of development was significantly delayed in the triploids (i.e. beyond the 2-cell stages) as compared with the observations in the control group (diploid eggs). Consequently, hatching time was 5 h faster in the diploid eggs [i.e. 18 hours post fertilization (hpf)] compared with the triploid induced eggs (23 hpf). The most critical stage of embryonic development in which mass mortality occurred in the different treatments was the somite stage. The status of triploid hatchlings was affirmed using erythrocyte morphology in 2-month-old fingerlings.

It has been established that embryogenesis is accompanied by the intense production of nitric oxide (NO). Based on available data, the rate of NO production is roughly equal in all embryos of the same poultry species. However, the rate of NO oxidation to nitrate in embryos of meat breeds is higher than in embryos of egg breeds. In broiler embryos about 90% of all produced NO is oxidised to nitrate. In embryos from egg breeds only several per cent of the NO oxidised to nitrate and the other NO is stored in the embryonic tissues included in NO donors. The intensity of NO oxidation in bird embryo is not depended on sex, age of the layer and feeding regime. Intensity of oxidation varies by no more than 10% within a breed strain or cross. Breeding to increase meat productivity is always associated with an increase in the intensity of NO oxidation in the embryo. There is no direct relationship between the increase in NO oxidation and the live weight gain. It can vary from hundreds of percent to several percent depending on the breed. Moreover, morphological differences between breeds with high and low intensity of embryonic NO oxidation are manifested only after hatching, as synthesis of NO is much lower than in the embryo. It has been presumed that NO oxidation is associated with the synthesis or activation of the factor(s) involved with enhanced growth of meat tissue. This is predetermined genetically and can be partly induced by exogenous stimulants, such as green light. The rate of embryonic NO oxidation can therefore be a useful criterion in practical selection of poultry and in the evaluation of growth enhancers acting at the embryonic level.

Pluripotency-associated transcription factors (PATFs) modulate gene expression during early mammalian embryogenesis. Despite a strong understanding of PATFs during mouse embryogenesis, limited progress has been made in ruminants. This work aimed to describe the temporal expression of eight PATFs during both sheep and cattle preimplantation development. Transcript availability of PATFs was evaluated by reverse transcriptase quantitative polymerase chain reaction (RT-qPCR) in eggs, cleavage-stage embryos, morulae, and blastocysts. Transcripts of five genes were detected in all developmental stages of both species (KLF5, OCT4, RONIN, ZFP281, and ZFX). Furthermore, CMYC was detected in all cattle samples but was found from cleavage-stage onwards in sheep. In contrast, NR0B1 was detected in all sheep samples but was not detected in cattle morulae. GLIS1 displayed the most significant variation in temporal expression between species, as this PATF was only detected in cattle eggs and sheep cleavage-stage embryos and blastocysts. In silico analysis suggested that cattle and sheep PATFs share similar size, isometric point and molecular weight. A phenetic analysis showed two patterns of PATF clustering between cattle and sheep, among several mammalian species. In conclusion, the temporal expression of pluripotency-associated transcription factors differs between sheep and cattle, suggesting species-specific regulation during preimplantation development.

In fish, many factors can affect reproduction during in vitro fertilization, therefore determination of the factors that affect affecting gamete quality is needed. However, few studies have focused on gamete quality and the ploidy status. This study was conducted to elucidate whether oocyte storage can affect ploidy status, survival, and embryo viability in the characid species Astyanax altiparanae. Oocytes were stored in Dulbecco's phosphate-buffered saline (PBS) at 26°C, then aliquots were fertilized immediately after extrusion (control) and also after 60, 120, 180, and 240 min of storage. Fertilization and hatching rates were measured, and the developmental stages were analyzed at each stage before describing the main abnormalities. Ploidy status was analyzed by flow cytometry and blood smear. In the control group, 100% of the samples were diploid. After treatment for 60 min, 95.56 ± 4.44% samples were diploid and 4.44 ± 4.44% were triploid. After 120 min, 94.44 ± 9.62% of the samples was diploid and 5.56 ± 5.56% were triploid; 100% of the samples were diploid after 180 min and, after 240 min, there was no survival. In other treatments, the highest percentage of hatching was after 60 min (88.93 ± 5.15%; P = 0.015), and treatment with 180 min storage resulted in the highest percentage of abnormal larvae (95.76 ± 12.67%; P = 0.012). These results show that oocyte storage can affect ploidy status and may be an interesting parameter for analysis in studies on chromosome set manipulation and micromanipulation.

The culture of ovules excised from velvetleaf (Abutilon theophrasti Medic., ♯4 ABUTH) capsules 5 days after anthesis was used to measure the effects of abscisic acid (ABA) and sucrose on embryo development and prevention of precocious germination. ABA at 1 × 10-7 M combined with 6% sucrose in the medium for the first 14 days of culture increased embryo development but prevented precocious germination. Higher concentrations of ABA inhibited embryo development. Without ABA, precocious germination increased directly with the concentration of sucrose in the medium, and embryos died. In vivo, ABA reached its highest concentration in ovules 5 days after anthesis but was undetectable after 16 days. Parental control of embryo development may involve ABA and an increasing concentration of osmoticum as seeds dehydrate during maturation.

Croftonweed has established itself as a pernicious weed in China. Experiments were conducted to test for anemophily, self-pollination, hand crossing, and apomixis. Additionally, pollen viability, seed set, and seed viability were investigated. Pollen cultured in vitro with the use of 14 culture media did not germinate, nor did self- or cross-pollen germinate on the stigma, and no pollen tubes were seen in the style. Young embryos developed into globular- or heart-shaped embryos before the opening of the captimulum, although neither meiosis to form an embryo sac nor fertilization occurred. Furthermore, seeds developed normally after closed flowers were emasculated and bagged. The plump seed ratio (plump seeds/total seeds) from apomixis (emasculation and bagged treatment) was high (41 ± 17.0%), and plump seed germination ratio exceeded 50%. These results provided additional evidence for the previous conclusion that croftonweed reproduces by apomixis. Apomixis, high seed production, and high seed germination may explain why this weed spreads so rapidly.

Cultivation of the geoduck Panopea zelandica (Quoy & Gaimard, 1835) requires knowledge on embryonic development to produce spat in hatcheries. This study investigated the development of P. zelandica embryos at 15°C and 35 ppt and the optimal sperm:egg ratios for fertilization under hatchery conditions. Panopea zelandica broodstock were induced to spawn by serotonin injection. Sperm and eggs were collected and fertilization was conducted at sperm:egg ratios of: 50:1, 100:1, 500:1, 1000:1 and 10,000:1 over 40 min. The optimal sperm:egg ratio was <500:1 and the normal embryo yield at 3 and 18 h post-fertilization (hpf) ranged from 83–96%. Panopea zelandica eggs (~80 μm diameter) developed the first and second polar bodies within 15–20 and 50–55 min post-fertilization, respectively. The blastula appeared at ~8 hpf, including the XR and XL cells and the presumptive shell field depression. Gastrulation occurred at 12–18 hpf with organic material apparent at the shell field depression. The mid-stage trochophore, which appeared at around 35 hpf had an apical plate with an apical tuft. The shell field spread to form the periostracum, which expanded and folded into right and left segments covering the late trochophore. The early D-stage veliger appeared at 45 hpf with the soft body being enclosed by two valves and the appearance of the velum. These observations will serve as the basis for future analyses of P. zelandica embryogenesis and for optimization of commercial production of D-veliger larvae.

The cattle tick Rhipicephalus (Boophilus) microplus is a hematophagous ectoparasite of major importance for the livestock industry. It shows a remarkable ability to survive over long periods without feeding. However, the mechanisms used to endure long-term starvation are poorly understood. It is believed that autophagy, a process of intracellular protein degradation, may play a significant role to confront adverse environmental conditions. To advance our understanding of autophagy in R. microplus, in the present study we report the molecular characterization of three autophagy-related (ATG) genes, namely, RmATG3, RmATG4 and RmATG6, as well as their expression profiles in different developmental stages and organs of the parasite. The deduced amino acid sequences derived from the characterized gene sequences were subjected to Basic Local Alignment Search Tool analysis. The testing produced significant alignments with respective ATG proteins from Haemaphysalis longicornis and Ixodes scapularis ticks. Real-time polymerase chain reaction assays revealed that RmATG4 and RmATG6 transcripts were elevated in egg and ovary tissue, when compared with larva and midgut samples, while RmATG3 expression in midgut was 2-fold higher than in egg, larva and ovary samples.

Sponges of the Mycale genus are an important source of several bioactive metabolites. The raw materials for studies on this genus mainly come from the wild populations. However, the scarce biomass of sponges in the wild has become an obstacle to the better investigation of the active substances. To establish aquaculture methods based on the sexual reproduction of the demosponge Mycale phyllophila and solve the sponge shortage problems, the sexual reproductive cycle of this species was studied. Our investigation is the first to report the sexual reproduction of the Mycale species in the coastal waters of China. We collected the sponge specimens from Dongshan Bay (Fujian, China) monthly from June 2012 to January 2014. Small samples of approximately 8 cm3 in volume were cut from 15 sponges (10 tagged individuals and five random individuals) with surgical scissors for histological analysis. The results indicated that M. phyllophila is a hermaphroditic and viviparous species. The primary oocytes measured 30–50 μm in diameter, while mature oocytes reached 300–400 µm by engulfing nearby nurse cells. Spermatic cysts containing asynchronous cells were found throughout the mesohyl, especially adjacent to the oscula. Macromeres in the middle and micromeres in the periphery were observed as the cleavage progressed. The reproductive activity of M. phyllophila lasted for 5–6 months and peaked when the average water temperature was above 25°C. This suggests that water temperature is an important environmental factor influencing the sexual reproduction cycle.

This study shows for the first time the presence of a jelly coat on oocytes of neotropical Characiformes fish. This structure could be responsible for the adhesiveness of Astyanax bimaculatus oocytes, a species widely distributed in South America including in the São Francisco River basin in Brazil. Adult specimens of A. bimaculatus were submitted to artificial reproduction in order to analyse the egg morphology and embryonic development. The eggs were fertilised and kept in incubators with a water temperature of 24°C so that embryogenesis could be monitored. Ovulated and unfertilised oocytes were also collected and submitted to routine histological techniques. Astyanax bimaculatus oocytes were found to be spherical, yellowish, and covered by a thin jelly coat with a slightly adhesive surface. The mean oocyte diameter was 1.03 ± 0.03 mm, the perivitelline space was 0.21 ± 0.02 mm and the jelly coat's thickness was 0.04 ± 0.01 mm. Positive periodic acid–Schiff (PAS) stain and Alcian blue stain pH 2.5 indicated the presence of neutral glycoproteins, and carboxylated acid glycoconjugates on the jelly coat that formed mucosubstances that may be associated with egg adhesiveness. At a water temperature of 24°C, blastopore closure and hatching occurred at 5 h and 17 h after fertilisation, respectively. The results of this study provide essential information for phylogenetic studies and for a better understanding of the reproductive strategy of A. bimaculatus, currently included in the incertae sedis group of the Characidae family due to the lack of monophyly among the families of the group.

Nitric oxide (NO) is generated in human and animal tissues from L-arginine. Its known effects include mediation of the parasympathetic nervous system (effects of aсеtуlchоline), smooth muscle tonus, apoptosis, and tissue differentiation. The physiological effects of NO attract significant theoretical and practical interest in medicine and agriculture due to its possible regulation of physiological functions.

It is known that embryo development involves intensive NO production, but the exact role of NO in embryogenesis is not clear. Elucidation of the features of NO metabolism in the embryo and their correlation with its development is necessary for the clarification of this role. The avian embryo is a convenient object for such research, because of its relatively large size and its development outside the dam, which allows control of all stages of the development.

This article reviews the scientific information on the physiological effects of NO, current hypotheses about their mechanism, and, finally, some perspectives of practical application in poultry breeding. It also reviews of the basic results of our own research that is conducted according to the program of RAAS. It has been observed that NO metabolites contents in embryo amnion and allantois of both egg laying and meat poultry breeds are very different. The value of the difference is more than one order of magnitude, which is seen in more than 30 lines and crosses of hen, quail, turkey and ostrich. It is possible that elucidation of the mechanism of action will allow active regulation of embryo development, as well as the potential for using the indices of NO metabolites content in selection work.

Assessments of seed storage physiology among Arecaceae (palm) species are often inconclusive because seeds exhibit diverse responses to low temperature and moisture conditions. Interrelationships between dry matter accumulation, cell structure and water relations during seed development of the endangered Hawaiian endemic palm, Pritchardia remota, suggest that damage from drying results from mechanical strain. Endosperm and fruits accumulate dry mass through most of the 400 d gestation period, but embryos reached maximum dry mass about 250 d post-anthesis (DPA). Mostly sucrose and some triacylglycerols accumulated in the cytoplasm and vacuoles of embryo cells, and organelles in mature embryo cells de-differentiated. Water content and water potential decreased as embryos matured and embryos contained about 0.45 g H2O (g dry mass)− 1 ( − 26 MPa) at shedding. Mature embryos survived drying to 0.16 g g− 1 ( − 49 MPa), but further drying was lethal. A model of allowable cell shrinkage is consistent with the substantial, but incomplete, desiccation tolerance acquired in P. remota embryos, and provides a new framework to explain variation in critical water contents as embryos develop. We suggest that desiccation tolerance, which distinguishes recalcitrant and orthodox physiologies among seeds, can be quantified by mechanical strain when embryo cells shrink during drying.

The aim of this study was to evaluate the oocytes, post-fertilization events and embryonic development in Brycon insignis, under both scanning electron microscopy and stereomicroscopy. Oocytes and embryos were sampled from spawning up to hatching. Stripped oocytes were spherical, non-adhesive, greenish-brown, possessed a single micropyle, pore-canals and had a mean diameter of 1.46 mm. In 63% of oocytes the germinal vesicle was peripheric. The main post-fertilization events were the fertilization cone formation (20 s), micropyle closure (100–180 s) and agglutination of supernumerary spermatozoa (100–180 s). Embryonic development lasted 30 h at ~24 °C and was characterized by seven stages. Zygote, cleavage, blastula and gastrula stages were first observed at 0.25, 1, 3 and 6 h post-fertilization, respectively. Fertilization rate was determined at the moment of blastopore closure, 10–11 h post-fertilization. The segmentation stage began at 11 h post-fertilization and comprised the development of somites, notochord, optic, otic and Kupffer's vesicles, neural tube, primitive intestine, and development and release of the tail. The larval stage began 21 h post-fertilization and was characterized by the presence of somites, growth and elongation of the larvae. At the hatching stage, embryos presented vigorous contractions of the tail and body leading to chorion rupture (30 h). The morphological characteristics described for B. insignis were similar to that described for other teleost species, and such knowledge is important for a better understanding of reproductive features of a species and useful for ecological and conservational studies.
