In 1895, by means of the Golgi method, Santiago Ramón y Cajal discovered a cell having a unique morphology in the avian retina. This cell had its cell body in the amacrine cell level of the inner nuclear layer, only a few rudimentary dendrites at the outermost level of the inner plexiform layer (IPL), and a long axon coursing horizontally and terminating in the IPL. Despite having defined amacrine cells as cells without axons, Cajal named this cell type “association amacrine cell” (AAC). This discovery was not confirmed by other investigators for nearly a century. Very recently, however, isthmo-optic target cells (IOTCs), which receive the terminals of centrifugal fibers emanating from the isthmo-optic nucleus, have been identified as one type of AAC. As summarized and discussed in this review, the morphology of the AACs as described by Cajal has been completely confirmed. However, since these cells appear to be classical polarized, monoaxonal neurons and lack the dendritic interactions that are typical of amacrine cells, they should be regarded as a distinct type of retinal interneuron and not as amacrine cells.

The purpose of this study was to specify isoluminance at different retinal eccentricities and to characterize macular pigment optical density (MPOD) and distribution using the steady-state visual evoked potential (VEP). Red–green (R/G) and blue–green (B/G) gratings were generated within two circular stimulus fields (radius = 0.55 or 1.1 deg) and within four annular fields (maximum mean radius = 6.0 deg) on a color monitor. Temporal frequency was 15 Hz. Isoluminance was determined for each stimulus using minimum flicker photometry. Steady-state onset–offset VEPs were recorded to the same annular stimuli as the luminance ratio between adjacent chromatic components was changed from 0.25 to 0.85 in 11 automated steps (0.5 representing photometric isoluminance). Fourier analysis showed that the power of the first harmonic was minimized at the isoluminant ratio specific to each subject. Relative OD was computed by comparing the isoluminant ratio at any location with that for the most eccentric annulus. To compensate for the broadband characteristics of the monitor, OD values were corrected according to minimum flicker measurements made through known concentrations of carotenoid solution. MPOD was additionally measured using minimum motion photometry.

There was high correlation between the isoluminant ratios determined by minimum flicker and VEPs for both R/G and B/G stimulation (r = 0.91, P < 0.005, slope = 1). Calibrated OD values computed from VEP estimates of B/G isoluminance correlated with those derived from minimum flicker (r = 0.96, P < 0.0005, slope = 0.85) and motion photometry (r = 0.94, P < 0.0005, slope = 0.88). OD values derived from B/G VEPs increased toward the fovea and corresponded closely with minimum flicker and minimum motion assessment of MP distribution profiles. The steady-state VEP can be used to determine isoluminance at different retinal eccentricities. MPOD and distribution can be measured by steady-state VEPs to B/G stimuli.

Brain-derived neurotrophic factor (BDNF) is a preferred ligand for a member of the tropomyosin-related receptor family, trkB. Activation of trkB is implicated in various activity-independent as well as activity-dependent growth processes in many developing and mature neural systems. In the subcortical visual system, where electrical activity has been implicated in normal development, both differential survival, as well as remodeling of axonal arbors, have been suggested to contribute to eye-specific segregation of retinal ganglion cell inputs. Here, we tested whether BDNF is required for eye-specific segregation of visual inputs to the lateral geniculate nucleus and the superior colliculus, and two other major subcortical target fields in mice. We report that eye-specific patterning is normal in two mutants that lack BDNF expression during the segregation period: a germ-line knockout for BDNF, and a conditional mutant in which BDNF expression is absent or greatly reduced in the central nervous system. We conclude that the availability of BDNF is not necessary for eye-specific segregation in subcortical visual nuclei.

Intrinsic interneurons within the dorsal lateral geniculate nucleus (dLGN) provide a feed-forward inhibitory pathway for afferent visual information originating from the retina. These interneurons are unique because in addition to traditional axodendritic output onto thalamocortical neurons, these interneurons have presynaptic dendrites that form dendrodendritic synapses onto thalamocortical neurons as well. These presynaptic dendrites, termed F2 terminals, are tightly coupled to the retinogeniculate afferents that synapse onto thalamocortical relay neurons. Retinogeniculate stimulation of F2 terminals can occur through the activation of ionotropic and/or metabotropic glutamate receptors. The stimulation of ionotropic glutamate receptors can occur with single stimuli and produces a short-lasting inhibition of the thalamocortical neuron. By contrast, activation of metabotropic glutamate receptors requires tetanic activation and results in longer-lasting inhibition in the thalamocortical neuron. The F2 terminals are predominantly localized to the distal dendrites of interneurons, and the excitation and output of F2 terminals can occur independent of somatic activity within the interneuron thereby allowing these F2 terminals to serve as independent processors, giving rise to focal inhibition. By contrast, strong transient depolarizations at the soma can initiate a backpropagating calcium-mediated potential that invades the dendritic arbor activating F2 terminals and leading to a global form of inhibition. These distinct types of output, focal versus global, could play an important role in the temporal and spatial roles of inhibition that in turn impacts thalamocortical information processing.

The specific role of ON and OFF retinal information channels in the generation of the horizontal optokinetic nystagmus (OKN) of the frog was studied. Coil recordings of monocular eye and head OKN were obtained before and after intravitreal injection of two drugs that block either ON or OFF channels. The intravitreal injection of 2-amino-4-phosphonobutyrate (APB), a glutamate analog that selectively blocks the ON retinal channel, strongly reduced or even cancelled the monocular OKN of the head and of the eye. The intravitreal injection of another glutamate analog, the cis-2, 3-piperidine dicarboxylic acid (PDA) that especially blocks the OFF retinal channel, did not affect the gain velocity of the slow phase of both the horizontal monocular head and eye OKN, for low stimulus velocities. Our results suggest that the retinal ON information channel, but not the OFF channel, is involved in the generation of the slow phase of the OKN of the frog, at least at low drum velocities.

In birds, the nucleus of the basal optic root (nBOR) of the accessory optic system (AOS) and the pretectal nucleus lentiformis mesencephali (LM) are involved in the analysis of optic flow and the generation of the optokinetic response. In several species, it has been shown that the AOS and pretectum receive input from visual areas of the telencephalon. Previous studies in pigeons using anterograde tracers have shown that both nBOR and LM receive input from the visual Wulst, the putative homolog of mammalian primary visual cortex. In the present study, we used retrograde and anterograde tracing techniques to further characterize these projections in pigeons. After injections of the retrograde tracer cholera toxin subunit B (CTB) into either LM or nBOR, retrograde labeling in the telencephalon was restricted to the hyperpallium apicale (HA) of the Wulst. From the LM injections, retrograde labeling appeared as a discrete band of cells restricted to the lateral edge of HA. From the nBOR injections, the retrograde labeling was more distributed in HA, generally dorsal and dorso-medial to the LM-projecting neurons. In the anterograde experiments, biotinylated dextran amine (BDA) was injected into HA and individual axons were reconstructed to terminal fields in the LM and nBOR. Those fibers projecting to the nBOR also innervated the adjacent ventral tegmental area. However, tracing of BDA-labeled axons revealed no evidence that individual neurons project to both LM and nBOR. In summary, our results suggest that the nBOR and LM receive input from different areas of the Wulst. We discuss how these projections may transmit visual and/or somatosensory information to the nBOR and LM.

While much attention has been given to the correlation between cytochrome-oxidase (CO) compartments and patterns of cortico-cortical projections originating from supragranular layers in the striate cortex, little is known in this regard about patterns of cortico-subcortical projections originating from infragranular cortex. We studied the tangential distribution of the striate cortex neurons projecting to the superior colliculus and used two approaches to analyze the relationship of this distribution to the arrangement of CO “blobs.” First, chi-square analysis indicated that significantly fewer labeled neurons were found within the CO blob compartment than the number expected for a random distribution. Second, spatial cross-correlation analysis – which circumvents the inherent subjectivity of delineating blob boundaries – revealed an area around blob centers in which there was a decreased probability of encountering labeled cells. The size of this area compared well with that of our outlines of CO blobs. We conclude that corticotectal projection neurons in the striate cortex are distributed preferentially within the interblob compartment of the infragranular striate cortex. These results demonstrate that the spatial distribution of cortico-subcortical projection neurons within infragranular cortex can correlate with the CO architecture of the primary visual cortex.

Tritiated neurotransmitter candidates were unilaterally injected in visual cortical regions with abundant corpus callosum connections. D-aspartate (Asp) or gamma-aminobutyric acid (GABA) was injected along the area 17/18 border in cat, and the area 17/18a and 17/18b borders in rat. Retrograde Asp label was found contralaterally in supragranular and infragranular laminae in areas 17, 18, 19, PMLS, and PLLS in cat, and in areas 29, 18b, 17, and 18a in rat. No contralateral GABA label was found in cat or rat. Thus, the cat and rat corpus callosum may use Asp or a related substance as a neurotransmitter.

The thalamic reticular nucleus (TRN) supplies an important inhibitory input to the dorsal thalamus. Previous studies in non-primate mammals have suggested that the visual sector of the TRN has a lateral division, which has connections with first-order (primary) sensory thalamic and cortical areas, and a medial division, which has connections with higher-order (association) thalamic and cortical areas. However, the question whether the primate TRN is segregated in the same manner is controversial. Here, we investigated the connections of the TRN in a New World primate, the marmoset (Callithrix jacchus). The topography of labeled cells and terminals was analyzed following iontophoretic injections of tracers into the primary visual cortex (V1) or the dorsal lateral geniculate nucleus (LGNd). The results show that rostroventral TRN, adjacent to the LGNd, is primarily connected with primary visual areas, while the most caudal parts of the TRN are associated with higher order visual thalamic areas. A small region of the TRN near the caudal pole of the LGNd (foveal representation) contains connections where first (lateral TRN) and higher order visual areas (medial TRN) overlap. Reciprocal connections between LGNd and TRN are topographically organized, so that a series of rostrocaudal injections within the LGNd labeled cells and terminals in the TRN in a pattern shaped like rostrocaudal overlapping “fish scales.” We propose that the dorsal areas of the TRN, adjacent to the top of the LGNd, represent the lower visual field (connected with medial LGNd), and the more ventral parts of the TRN contain a map representing the upper visual field (connected with lateral LGNd).

Arrestins participate in the termination of phototransduction in both vertebrates and invertebrates. However, the visual arrestins of invertebrates and vertebrates differ significantly from one another in that the invertebrate visual arrestins become phosphorylated rapidly in response to light while those in the photoreceptors of vertebrates do not. In an effort to understand the functional relevance of arrestin phosphorylation, we examined this process in the photoreceptors of the horseshoe crab Limulus polyphemus. We report that Limulus visual arrestin can be phosphorylated at three sites near its C-terminus and show that arrestin molecules phosphorylated on one, two, and three sites are normally present in both light- and dark-adapted photoreceptors. Light adaptation increases the amount of arrestin phosphorylated at three sites.

In this study, a Müller cell culture preparation from young rats was used to investigate the regulation of GLAST transport activity in native cells. Immunohistochemical analysis confirmed GLAST to be the predominant glutamate transporter expressed by the cells through five passages. [3H]-glutamate uptake assays showed the typical Na+-dependent glutamate transport which was blocked by L-(-)-threo-3-hydroxyaspartate (L-THA), a competitive inhibitor. Glutamate transport was decreased significantly in Müller cells exposed to phorbol-12-myristate-13-acetate (PMA), a protein kinase C (PKC) activator. A similar effect on [3H]-D-aspartate (nonmetabolizable glutamate analog) uptake ruled out the possibility that the decrease was a consequence of altered metabolism. However, PMA did not affect Na+-dependent [3H]-glycine transport, indicating the absence of a nonspecific change in the electrochemical gradients. The PMA effect on glutamate uptake was evidenced by partial blocking with a specific PKC inhibitor, bisindolymaleimide II (Bis II). Activation of PKC did not change the Km, but the Vmax was significantly reduced. Image analysis of Müller cells with biotinylated cell membranes immunolabeled with GLAST shows a reduction of GLAST in the plasma membrane. In conclusion, these data show that rat Müller cells in primary cultures express GLAST and that PKC activation affects GLAST transport activity by decreasing cell surface expression.

Intracellular recording techniques were used to evaluate the effects of norepinephrine (NE) on the membrane properties of superficial layer (stratum griseum superficiale and stratum opticum) superior colliculus (SC) cells. Of the 207 cells tested, 44.4% (N = 92) were hyperpolarized by ≥3 mV and 8.7% (N = 18) were depolarized by ≥3 mV by application of NE. Hyperpolarization induced by NE was dose dependent (EC50 = 8.1 μM) and was associated with decreased input resistance and outward current which had a reversal potential of −94.0 mV. Depolarization was associated with a very slight rise in input resistance and had a reversal potential of −93.1 mV for the single cell tested. Pharmacologic experiments demonstrated that isoproterenol, dobutamine, and p-aminoclonidine all hyperpolarized SC cells. These results are consistent with the conclusion that NE-induced hyperpolarization of SC cells is mediated by both α2 and β1 adrenoceptors. The α1 adrenoceptor agonists, methoxamine and phenylephrine, depolarized 35% (6 of 17) of the SC cells tested by ≥3 mV. Most of the SC cells tested exhibited responses indicative of expression of more than one adrenoceptor. Application of p-aminoclonidine or dobutamine inhibited transsynaptic responses in SC cells evoked by electrical stimulation of optic tract axons. Inhibition of evoked responses by these agents was usually, but not invariably, associated with a hyperpolarization of the cell membrane and a reduction in depolarizing potentials evoked by application of glutamate. The present in vitro results are consistent with those of the companion in vivo study which suggested that NE-induced response suppression in superficial layer SC neurons was primarily postsynaptic and chiefly mediated by both α2 and β1 adrenoceptors.

After being severed, optic axons in goldfish regenerate and eventually restore the retinotectal map; refinement of the map depends upon impulse activity generated by the ganglion cells. Because little is known about the changes in activity and receptive-field properties of ganglion cells during regeneration, we made extracellular recordings from them in the intact eye up to 95 days after sectioning their axons in the optic tract. Their receptive fields were classified as OFF-, ON–OFF-, or ON-centers, and their axonal conduction velocities measured by antidromic activation. The rate of encountering single units dropped drastically at 4–8 days postsection when only a few OFF-center units could be recorded, recovering to normal between 42 and 63 days. Receptive-field centers were normal in size, except for the few OFF-centers at 4–8 days which were abnormally large. Maintained discharge rates of all types were depressed up to 42 days, but ON–OFF-center units were more spontaneously active than normal around 42 days. Light-evoked responses in OFF-center units were subnormal at 4–8 days, becoming supernormal at 16 days and normal thereafter. ON–OFF- and ON-center units started to regain responsiveness at 16 days, and became supernormal at 42 days, before returning to normal. Conduction velocities of all fiber groups dropped to a minimum at 8 days, the fastest being affected most. There was a gradual recovery to normal conduction velocity by 63 days. The conduction latencies of OFF- and ON–OFF-center units recovered to normal by 42 days, and ON-center units by 63 days. Recovery of ganglion cell responsiveness correlates with functional recovery in the retinotectal system: OFF-center units recover light-evoked responses at about the time OFF activity first reappears in the tectum. ON- and ON–OFF-center units recover later, exhibiting supernormal spiking activity around the time that ON responses reappear in the tectum.

A network model of disparity estimation was developed based on disparity-selective neurons, such as those found in the early stages of processing in the visual cortex. The model accurately estimated multiple disparities in regions, which may be caused by transparency or occlusion. The selective integration of reliable local estimates enabled the network to generate accurate disparity estimates on normal and transparent random-dot stereograms. The model was consistent with human psychophysical results on the effects of spatial-frequency filtering on disparity sensitivity. The responses of neurons in macaque area V2 to random-dot stereograms are consistent with the prediction of the model that a subset of neurons responsible for disparity selection should be sensitive to disparity gradients.

We recorded optokinetic eye movements of the crab, Carcinus maenas, in split-drum experiments. The patterns were either oscillated in antiphase on both sides mimicking translational image flow or they were oscillated in phase producing rotational image flow. Eye movements elicited by the rotational stimulus were larger than those produced by the pseudotranslational pattern movements. The smaller response to the latter is mainly a consequence of binocular interaction, the strength of which depends on both the phase-shift and amplitude of pattern oscillation. We develop two hypotheses to explain our results: either (1) signals from each eye modify the gain of the linkage signals coming from the other eye, or (2) the signals coming from the other eye modify the gain of the control loop itself. Quantitative evaluation of the data favors the second of these two hypotheses, which comprises the models of Barnes and Horridge (1969) and Nalbach et al. (1985). In addition, we found that it is the signals from the two slow channels of the crab's movement-detecting system that are transferred from one eye to the other, while signals of the fastest channel act almost exclusively ipsilaterally. We discuss our results as an adaptation by which an animal with panoramic vision compensates exclusively the rotational component of image flow during locomotion. The fact that freely walking crabs distinguish the two components of image flow better than restrained crabs indicates that further visual and nonvisual signals help to disentangle image flow.

The PC pathway conveys both chromatic and achromatic information, with PC neurons being more responsive to chromatic (L−M) than to achromatic (L+M) stimuli. In considering the evolution of color vision, it has been suggested that the dynamic range of chromatic PC-pathway processing is tuned to the chromatic content of the natural environment. Anomalous trichromats, with reduced separation of their L- and M-cone spectral sensitivities, have diminished chromatic input to PC-pathway cells. Dichromats, with absent L or M cones, should have no chromatic input to PC-pathway cells. Therefore, the PC-pathway dynamic range of color defectives should be released from any constraint imposed by the chromatic environment. Here we ask whether this results in compensatory enhancement of achromatic PC-pathway processing in color defectives. This study employed a psychophysical method designed to isolate PC-pathway processing using achromatic stimuli. In a pulsed-pedestal condition, a four-square stimulus array appeared within a uniform surround. During a trial, one of the test squares differed from the other three, and the observer's task was to choose the square that was different. A four-alternative, forced-choice method was used to determine thresholds as a function of the contrast of the four-square array to the surround. Seven color defective and four normal observers participated. Results showed no systematic differences between normals and color defectives. There was no enhancement of achromatic processing as compensation for reduced chromatic processing in the PC-pathway system in color defectives. From physiological recordings, PC-pathway achromatic contrast gains of dichromatic and trichromatic New World primates and trichromatic Old World macaques have also been shown to be similar to each other. Our study and the animal studies imply that PC-pathway contrast gain parameters were regulated by factors other than the environmental chromaticity gamut, and may have arisen in a nontrichromatic common ancestor to both Old and New World primates.

We studied the visuomotor properties of 54 corticostriatal (CS) and 38 corticotectal (CT) neurons in a region of area 6 that largely corresponds to the cat's frontal eye fields in five cats trained to do simple oculomotor tasks. Overall, these cells were similar to the general population of area 6 neurons described in the previous paper (Weyand & Gafka, 1998), with very few showing pre-saccadic activity. Likewise, CS and CT cells were similar to each other, although only CS cells showed activity exclusively related to the delivery of the reward and CT cells were more likely to be active during saccades. Variability in visual response latencies and the observation that some cells showed initial visual suppression suggest CS and CT cells reflect the output of a variety of intracortical circuits. Despite similar response properties and overlapping laminar origin, CS and CT circuits appear largely independent. Among 32 cells that we could electrically activate (either synaptically or antidromically) from the superior colliculus, only two could also be activated from stimulating electrodes in the striatum. Similarly, 23 of 25 cells electrically activated from the striatum could not be activated from the superior colliculus. Although few of these efferent cells exhibited pre-motor activity, many exhibit properties that could contribute to gaze control.

We measured the photopic spectral sensitivity of multiunit activity in the torus longitudinalis and optic tectum of goldfish. Since negative contrast stimuli are most effective for exciting torus longitudinalis, spiking activity was evoked by the shadow of a disc moving through a monochromatic light beam projected upon a screen. The amount of activity evoked in torus longitudinalis generally increased with the monochromatic stimulus radiance at the same rate for all wavelengths, indicating a univariant response. Spiking activity in tectum, however, increased at different rates across the spectrum, indicating color-dependent responses. The action spectra for torus longitudinalis were all similar and relatively flat as expected of a homogeneous, broad-band luminance processing system, and about 1 log unit more sensitive than the tectal action spectra. The latter generally displayed sharp peaks and dips in sensitivity indicative of opponent processing.

During optic nerve regeneration in the frog, axons transiently grow along the opposite optic nerve forming a retino-retinal projection. In the present study, we crushed the left optic nerve in the frog Litoria (Hyla) moorei and later applied horseradish peroxidase (HRP) or diamidino yellow (DY) to the right optic nerve. In one series, retinae were examined 3 days after application of the tracer. The retino-retinal projection was found to be maximal at 5 weeks, fell significantly by 7 weeks, and returned to close-to-normal levels by 24 weeks. In a second series, we applied DY at 5 weeks as before but did not sacrifice the frogs until 7 weeks. Numbers of labeled ganglion cells were not significantly different from those frogs in the first series labeled and examined at 5 weeks. We conclude that ganglion cells giving rise to the retino-retinal projection had not died in appreciable numbers, presumably being sustained by collateral axons in the brain.

Studies into the mechanisms underlying the active emmetropization process by which neonatal refractive errors are corrected, have described rapid, compensatory changes in the thickness of the choroidal layer in response to imposed optical defocus. While high frequency A-scan ultrasonography, as traditionally used to characterize such changes, offers good resolution of central (on-axis) changes, evidence of local retinal control mechanisms make it imperative that more peripheral, off-axis changes also be tracked. In this study, we used in vivo high resolution spectral domain-optical coherence tomography (SD-OCT) imaging in combination with the Iowa Reference Algorithms for 3-dimensional segmentation, to more fully characterize these changes, both spatially and temporally, in young, 7-day old chicks (n = 15), which were fitted with monocular +15 D defocusing lenses to induce choroidal thickening. With these tools, we were also able to localize the retinal area centralis, which was used as a landmark along with the ocular pectin in standardizing the location of scans and aligning them for subsequent analyses of choroidal thickness (CT) changes across time and between eyes. Values were derived for each of four quadrants, centered on the area centralis, and global CT values were also derived for all eyes. Data were compared with on-axis changes measured using ultrasonography. There were significant on-axis choroidal thickening that was detected after just one day of lens wear (∼190 µm), and regional (quadrant-related) differences in choroidal responses were also found, as well as global thickness changes 1 day after treatment. The ratio of global to on-axis choroidal thicknesses, used as an index of regional variability in responses, was also found to change significantly, reflecting the significant central changes. In summary, we demonstrated in vivo high resolution SD-OCT imaging, used in combination with segmentation algorithms, to be a viable and informative approach for characterizing regional (spatial), time-sensitive changes in CT in small animals such as the chick.
