From the study of the worm egg output of nearly fifteen hundred sheep on many hill farms in Scotland it has been shown that:-

(i) Worm egg output in ewes and gimmers is lowest in early winter and highest in spring and early summer; this is followed by a marked decrease towards the winter months.

(ii) Generally in ewe hoggs the worm egg output is the same as that in older sheep, but shows greater variability; on occasion the spring peak may be absent.

(iii) The level of worm egg output of hoggs is generally higher than that of older sheep, and in this respect gimmers tend to fall between hoggs and ewes.

(iv) Weather conditions may have an influence on the height and duration of the spring peak of worm egg production.

Seasonal fluctuations in the prevalence and intensity of infection with Heligmosomoides polygyrus (Nematoda) were studied in Apodemus sylvaticus (wood mouse, n = 399), sampled from three contrasting habitats in southern England, to test the hypothesis that both intrinsic (host sex, age) and extrinsic (season, site) factors influence parasite burdens. Maximum likelihood techniques based on log linear analysis of contingency tables were employed to generate a minimum sufficient model for prevalence of infection and 3-way ANOVA with negative binomial errors was used to evaluate the relative contribution of the principal factors and their interactions in explaining variation in worm burdens. Host age could not be entered into either statistical model because of some incomplete subsets of data. However, it was evident that in general juvenile mice carried lower worm burdens compared with adults, although these increased towards the winter season. Host sex was not a significant factor, other than in making a contribution to a weak significant interaction between sex and site, arising from male bias in one site and female bias in the remaining two. The principal determinants of variation in worm burden were the extrinsic factors, site and season, with an approximately equal weighting, and their interaction. These effects arose because worm burdens were lower at Dungeness and showed quite different seasonal patterns to the Egham and Isle of Wight sites. We propose that the unique character of the Dungeness habitat was not conducive to optimal transmission of H. polygyrus throughout most of the year (excepting spring) and we suggest possible explanations for these observations.

The objective of this paper is to critically evaluate the significance of parasite-altered host behaviour in the Toxocara mouse model particularly in the light of the Manipulation Hypothesis. Murine behaviours were examined in both outbred and inbred strains of mice infected with different doses of Toxocara canis ova. Behaviours investigated included activity, exploration, response to novelty, anxiety, learning, memory and social behaviour. Subsequent modifications to the behaviour of infected mice were investigated with respect to dose administered and larval accumulation in the brain. There was substantial variation in the number of larvae recovered from brains of individual mice, which received similar doses of Toxocara ova. Furthermore, the numbers of larvae recovered at different doses differed significantly between an outbred and inbred strain of mouse. Alterations in infected host behaviour occurred and were related to the number of larvae recovered from the brain. For social behaviour in outbred mice, a high infection in the brain reduced levels of aggressive behaviour and increased levels of flight and defensive behaviours. In contrast, outbred mice with a low infection in the brain displayed a greater level of risk behaviour in respect of predator odour and the light/dark box compared to control or high infection mice. Post-infection, outbred mice were more immobile whereas inbred mice showed reduced immobility and increased digging and climbing. Impaired learning ability was observed in outbred mice with moderate and high levels of infection in the brain compared to control and low infection mice. Toxocara infection has an impact upon a diverse range of murine behaviours with little evidence for a specific and hence an adaptive alteration. Many of the effects on murine host behaviour by Toxocara are likely to be pathological side effects of infection rather than as a consequence of adaptive host-manipulation. Observed changes in murine behaviour may be relevant to human toxocariasis.

Cases of canine onchocerciasis caused by Onchocerca lupi are increasingly reported from Europe and the western United States of America. The zoonotic role of this parasite had already been suspected in Europe as the clinical signs and histopathology seen in two ocular cases from Albania and the Crimean region were very similar to those of canine ocular onchocerciasis. In the most recent reports of human onchocerciasis, O. lupi has been morphologically and molecularly identified as the causative agent of ocular infestation in two patients from Turkey, and one patient from Tunisia. Here, we report an additional case of nodular lesions involving two, and possibly more, immature worms in a patient from Iran. The parasite was found to belong to the genus Onchocerca based on morphological features and the species was confirmed as O. lupi from a partial sequence analysis of 12S ribosomal DNA.

Although Taenia hydatigena is one of the most prevalent taeniid species of livestock, very little molecular genetic information exists for this parasite. Up to 100 sheep isolates of T. hydatigena were collected from 19 abattoirs located in the provinces of Tehran, Alborz and Kerman. A calibrated microscope was used to measure the larval rostellar hook lengths. Following DNA extraction, fragments of cytochrome c oxidase 1 (CO1) and 12S rRNA genes were amplified by the polymerase chain reaction method and the amplicons were subjected to sequencing. The mean total length of large and small hooks was 203.4 μm and 135.9 μm, respectively. Forty CO1 and 39 12S rRNA sequence haplotypes were obtained in the study. The levels of pairwise nucleotide variation between individual haplotypes of CO1 and 12S rRNA genes were determined to be between 0.3–3.4% and 0.2–2.1%, respectively. The overall nucleotide variation among all the CO1 haplotypes was 9.7%, and for all the 12S rRNA haplotypes it was 10.1%. A significant difference was observed between rostellar hook morphometry and both CO1 and 12S rRNA sequence variability. A significantly high level of genetic variation was observed in the present study. The results showed that the 12S rRNA gene is more variable than CO1.

Over 26000 specimens representing five euphausiid species collected, mostly in 1969, in the North-East Atlantic and northern North Sea were examined for parasites. Hysterothylacium sp. larvae were rare, occurring in only two euphausiid samples from inshore localities; the 9 larvae found (3.8 to 12.3 mm long) are described and illustrated. Infection with larval Anisakis simplex was virtually restricted to Thysanoessa spp. and appeared to be discontinuous. Infection was absent to the west of Scotland, and rare to the north of Scotland and at the Faroes. Larval A. simplex was not uncommon in offshore regions of the northern North Sea at depths between 100 and 200 m; the prevalence rate there for individual euphausiid species in individual samples was mostly 0 to 40% but an exceptionally high rate of 78.0% was recorded. Larvae in euphausiids do not become encapsulated but appear to remain active. Experimental observations on the sedimentation rate of A. simplex eggs and hatched larvae in sea water at 10°C are discussed in relation to embryonation and hatching at different temperatures, and depth of the sea. New host records are claimed for larval A. simplex in Nyctiplutnes couchii, and for larval Hysterothylacium sp. in Thysanoessa inermis. No individual euphausiid harboured more than one parasite, either larval nematode or ellobiopsid (see also SMITH, 1981). Morphological and morphometric observations on larval A. simplex from euphausiids are reported elsewhere (SMITH, 1983).

Dogs, naturally infected with Dirofilaria immitis, were treated with the residues of the alcoholic extracts of the rhizomes of Zingiber officinale (ginger). Twelve subcutaneous injections of the extract given at 100 mg/kg reduced microfilarial concentration in blood by a maximum of 98%. Fifty five days after the last injection there was 83% reduction in microfilarial concentration suggesting partial destruction of adult worms. Half of the treated dogs showed some lethargy at the beginning of treatment possibly due to the mass annihilation of microfilariae in blood.

1.In Kenya Dipetalonema reconditum is a common parasite of dogs, jackals and hyaenas. Usually microfilarial densities in the blood are very low. The adult worms are small; they can be detected at autopsy by searching the subcutaneous fascial spaces with a dissecting microscope.

2.The morphology of the adult worms is described and illustrated. The adults and microfilariae are readily distinguished from other species found in dogs in East Africa.

3.The main intermediate host of D. reconditum in dogs in Kenya is the flea Ctenocephalides felis. If fleas are kept on dogs for more than a week they can be used for xenodiagnosis to detect very low density infections.

4.The infective larvae have a characteristic caudal morphology with three terminal projections. They are very similar to the infective larvae of D. arbuta, D. vite and D. manson-bahri.

While carrying out an intensive survey of the helminths parasitic in domestic animals at Aberystwyth, an opportunity was Afforded the writer to examine the dogs and cats taken to the local police station for destruction by the lethal chamber process. These animals comprised the unclaimed “strays,” the incurably wounded or maimed, the vicious and the aged. Healthy but unwanted animals, also, were occasionally destroyed.

A single oral dose of an aqueous suspension containing 11,350,000 chlamydospores of a Mexican isolate of Duddingtonia flagrans (FTHO-8) given to sheep, resulted in a maximum reduction of 88% (range 86.7–90.4%) of the population of Haemonchus contortus infective larvae in the faeces. The effect of this treatment continued for 4–5 days after administration of the suspension. The possible use of this treatment as a method of control of ovine haemonchosis is discussed.

Human cystic echinococcosis (CE) is known to be endemic in the Tibet Autonomous Region (TAR), China; however, there is relatively little data from hospital records or community prevalence studies, and the situation regarding occurrence of human alveolar echinococcosis (AE) is unclear. Here we review the available reports about human echinococcosis in the seven prefectures of TAR. In addition, two pilot studies by mass screening using ultrasound (with serology) were undertaken (2006/7) in Dangxiong County of Lhasa Prefecture (north central TAR) and Dingqing County of Changdu Prefecture (eastern TAR). In Dangxiong County a prevalence of 9.9% (55/557) for human CE was obtained but no human AE cases were detected. By contrast, in Dingqing County (N= 232 persons screened), 11 CE cases (4.7%) and 12 AE cases (5.2%) (including one mixed CE and AE case) were diagnosed by ultrasound. Hospital records and published reports indicated that CE cases were recorded in all of seven prefectures in Tibet Autonomous Region, and AE cases in four prefectures. Incidence rates of human CE were estimated to range from 1.9 to 155 per 100,000 across the seven prefectures of TAR, with a regional incidence of 45.1 per 100,000. Incidence of AE was estimated to be between 0.6 and 2.8 cases per 100,000. Overall for TAR, human AE prevalence appeared relatively low; however, the pilot mass screening in Dingqing in eastern TAR indicated that human AE disease is a potential public health problem, possibly similar to that already well described in Tibetan communities bordering TAR in north-west Sichuan and south-west Qinghai provinces.

The parasitic nematode Trichinella spiralis, an aetiological agent of the disease known as trichinellosis, infects wild and domestic animals through contaminated pig meat, which is the major source for Trichinella transmission. Prevention of this disease by interrupting parasite transmission includes vaccine development for livestock; however, major challenges to this strategy are the complexity of the T. spiralis life cycle, diversity of stage-specific antigens, immune-evasion strategies and the modulatory effect of host responses. Different approaches have been taken to induce protective immune responses by T. spiralis immunogens. These include the use of whole extracts or excretory–secretory antigens, as well as recombinant proteins or synthesized epitopes, using murine experimental models for trichinellosis. Here these schemes are reviewed and discussed, and new proposals envisioned to block the zoonotic transmission of this parasite.

A new entomopathogenic nematode species from Australia, Heterorhabditis gerrardi n. sp. (Nematoda: Heterorhabditidae) is described. Morphological and molecular studies together with cross-hybridization tests indicated that this nematode represents a new undescribed species, closely related to members in the ‘indica-group’. However, the new species can be distinguished from other species in this genus by a combination of several qualitative and quantitative morphological traits. Key diagnostic features include: body size and excretory pore position of the third-stage infective juveniles; male bursa with a reduction of bursal rays, usually affecting the terminal set of papillae, with symmetrical or asymmetrical loss of one or two pairs; vulva of hermaphrodites more anteriorly located than in other species in the indica-group (V% average: 43), with non-protruding or slightly protruding lips, and longer tail length (average: 106 μm). The new species can be further characterized by molecular traits of sequence data from the internal transcribed spacer (ITS) region of ribosomal DNA. Additionally, the bacterial symbiont of this new species, Photorhabdus asymbiotica Kingscliff strain, was phenotypically characterized and compared with other P. asymbiotica strains. The Kingscliff strain revealed many characters not present in other strains of this species. We hypothesize that the newly found traits may contribute to the maintenance of this mutualistic association of the bacterium with its nematode host.

Nine genera, listed on pages 122–128 are recognized in the subfamily Rhabditinae Micoletzky, 1922, in place of three genera recognized by Osche (1952) in his recent revision of the genus Rhabditis Dujardin [1844] (sensu lato). Two of the genera—Pelodera Schneider, 1866 and Rhabditis (sensti stricto)—are subdivided into four and five subgenera respectively. One genus—Brevibucca Goodey, 1985—is accepted only provisionally in the Rhabditinae. The genus Neorhabditis Schuurmans Stekhoven, 1954 (syn. Pararhabditis Schuurmans Stekhoven, 1951; non Baylis and Daubney, 1926) is regarded as not belonging in the Rhabditinae

A 10-year (1998–2008) retrospective study was carried out to investigate the prevalence and long-term trend of hydatid disease in slaughtered herbivores in the large complex abattoir of Ahwaz (the capital of Khuzestan province, south-western Iran). A total of 3,583,417 animals including 2,815,982 sheep, 427,790 goats and 339,645 cattle were inspected macroscopically for hydatid cysts in the 10-year period, and overall 155,555 (4.24%) livers and 228,172 (6.37%) lungs were condemned. Cystic echinococcosis (CE) was responsible for 36.08% and 48.04% of total liver and lung condemnations, respectively. The prevalence of pulmonary hydatid disease in sheep, goats and cattle was 2.22, 5.43 and 6.99%, respectively; on the other hand, the prevalence of hepatic hydatid disease for those animals was 1.26, 2.57 and 2.80%, respectively. Data showed an overall downward long-term trend for CE in all livestock slaughtered during the study period (P < 0.01). Lung condemnation due to CE was significantly more common than liver condemnation for each animal separately (P < 0.001). The prevalence of liver and lung hydatidosis in sheep was significantly lower than that in other livestock (P < 0.001). The prevalence of hydatid disease recovered from the sheep, cattle and goats varied in different seasons, but there was no statistical difference between various seasons. The odds ratio of lung and liver condemnations due to hydatidosis showed a slightly different pattern in some years; however, the overall declining trend was still observed. The total annual economic loss incurred due to hydatidosis in all ruminants slaughtered at Ahwaz municipal abattoir was estimated to be US$459,659.6, based on the market prices in the year 2008. This number corresponds to a loss of US$300,620.4 for cattle, US$123,490.0 for sheep and US$35,549.2 for goats. The current results provide baseline data for the future monitoring of this potentially important disease in the region, and also suggest that a thorough investigation leading to a disease control strategy is required to reduce the economic and public health consequences of CE.

Members of the family Gastrothylacidae (Trematoda: Digenea: Paramphistomata) are parasitic in ruminants throughout Africa and Asia. In north-east India, five species of pouched amphistomes, namely Fischoederius cobboldi, F. elongatus, Gastrothylax crumenifer, Carmyerius spatiosus and Velasquezotrema tripurensis, belonging to this family have been reported so far. In the present study, the molecular phylogeny of these five gastrothylacid species is derived using the second internal transcribed spacer (ITS2) sequence and secondary structure analyses. ITS2 sequence analysis was carried out to see the occurrence of interspecific variations among the species. Phylogenetic analyses were performed for primary sequence data alone as well as the combined sequence-structure information using neighbour-joining and Bayesian approaches. The sequence analysis revealed that there exist considerable interspecific variations among the various gastrothylacid fluke species. In contrast, the inferred secondary structures for the five species using minimum free energy modelling showed structural identities, in conformity with the core four-helix domain structure that has been recently identified as common to almost all eukaryotic taxa. The phylogenetic tree reconstructed using combined sequence–structure data showed a better resolution, as compared to the one using sequence data alone, with the gastrothylacid species forming a monophyletic group that is well separated from members of the other family, Paramphistomidae, of the amphistomid flukes group. The study provides the molecular characterization based on primary sequence data of the rDNA ITS2 region of the gastrothylacid amphistome flukes. Results also demonstrate the phylogenetic utility of the ITS2 sequence–secondary structure data for inferences at higher taxonomic levels.

Identification of members of the family Schistosomatidae presents problems not only because of the imperfect state of their taxonomy, but also because no attempt has been made to collect and organise the available data. There are only two general keys on the schistosomes (Price, 1929 ; Yamaguti, 1958) both of which are incomplete. The result of this is that many authors, too numerous to mention, list genera which are no longer valid and erect new species without comparing them with species to which they are obviously closely related.

Fifteen lots of fifty individual cysts from three separate batches were exposed to the action of potato root diffusate, the hatching allowed to continue to completion and the total larvae liberated from each cyst were counted. The data obtained were analysed both as they stood and after three different transformations for variability, as well as for independence between mean and variance. Very high variability corresponding to a coefficient of variation of 1.00 was detected, and there was, moreover, a very significant correlation between mean and variance. The most suitable transformations appeared to be either logarithmic or angular—the latter being probably the more useful. The latter, moreover, tended to increase the sensitivity of analysis. The importance of residual counts when hatching is complete is emphasized even in the absence of an angular transformation of the data.

Species-specific identification of ascaridoid nematodes at any developmental stage is a prerequisite for detailed investigation of the life cycles, systematics and epidemiology of this important group, and is also crucial for the diagnosis of associated infections. The morphological identification of some species and/ or their larval stages can, however, present considerable difficulty. Recently, PCR-based methods, using genetic markers in the internal transcribed spacers (ITS) of ribosomal DNA, have been shown to provide reliable alternatives to more traditional methods for the specific identification of nematodes. This article provides an account of recent research on the development of PCR-based methods (utilizing ITS sequences) for the specific identification of ascaridoid nematodes of zoonotic potential, for the diagnosis of infections, and for the analysis of genetic variation within and among individual nematodes and their populations. Prospects for using these diagnostic and analytical tools to investigate epidemiological and population genetic questions relating to ascaridoid parasites are also discussed.

Laboratory experiments were performed to investigate the ability of isolates from the predacious fungus species Arthrobotrys conoides (one isolate) and A. robusta (four isolates) to trap and kill infective Haemonchus placei larvae. Three groups for each isolate were formed: group 1, fungi and infective larvae; group 2, fungi; group 3, infective larvae. There were statistically significant differences (P<0.05) between the antagonistic effects of three isolates of A. robusta, but there was no such difference between the isolate of A. conoides and isolates of A. robusta. This result indicates that there can be a variation in the predatory capacity of different isolates within a single fungus species. After 20 days of assay, there was greater recuperation of infective H. placei larvae in group 3 compared to group 1.
