Digenetic trematodes of the genus Clinostomum are cosmopolitan parasites infecting fishes, amphibians, reptiles, and snails as intermediate hosts. Despite the broad geographical distribution of this genus, debate about the number of species and how they vary in host use has persisted. To better understand patterns of infection among host species and across life stages, we used large-scale field surveys and molecular tools to examine five species of amphibians and seven species of fishes from 125 California ponds. Among the 12,360 examined hosts, infection was rare, with an overall prevalence of 1.7% in amphibians and 9.2% in fishes. Molecular evidence indicated that both groups were infected with Clinostomum marginatum. Using generalized linear mixed effects models, host species identity and host life stage had a strong influence on infection status, such that Lepomis cyanellus (green sunfish) (49.3%) and Taricha granulosa (rough skinned newt) (9.2%) supported the highest overall prevalence values, whereas adult amphibians tended to have a higher prevalence of infection relative to juveniles (13.3% and 2.5%, respectively). Experimentally, we tested the susceptibility of two amphibian hosts (Pseudacris regilla [Pacific chorus frog] and Anaxyrus boreas [western toad]) to varying levels of cercariae exposure and measured metacercariae growth over time. Pseudacris regilla was 1.3× more susceptible to infection, while infection success increased with cercariae exposure dose for both species. On average, metacarcariae size increased by 650% over 20 days. Our study highlights the importance of integrating field surveys, genetic tools, and experimental approaches to better understand the ecology of host–parasite interactions.

Much interest has centred recently on the role of adaptive trade-offs between the immune system and other components of life history in determining resistance and parasite intensities among hosts. Steroid hormones, particularly glucocorticoids and sex steroids, provide a plausible mechanism for mediating such trade-offs. A basic assumption behind the hypothesis, however, is that steroid activity will generally correlate with reduced resistance and thus greater parasite intensities. Here, we present some findings from a field study of bank voles (Clethrionomys glareolus) in which we have looked at associations between parasite intensities, anatomical and morphometric measures relating to endocrine function and life history variation in three local populations inhabiting similar but mutually isolated woodland habitats. In general, sites with greater parasite intensities were those in which male C. glareolus had significantly larger adrenal glands, testes and seminal vesicles for their age and body size. Females also showed a site difference in adrenal gland weight. Some aspects of site-related parasite intensity were associated with asymmetry in adrenal gland weight and hind foot length, which may have reflected developmental effects on glucocorticoid activity.

Porcine cysticercosis is an emerging zoonosis with public health and economic importance. A cross-sectional study was undertaken to investigate the disease in free-range pigs on 182 smallholder farms in Busia District, Kenya. The survey households were selected using a snowballing technique. Serum samples were obtained from 284 pigs of all ages at farm level and 37 pigs from slaughter slabs in the study area. The samples were analysed for the presence of cysticercus antigen using an antigen enzyme-linked immunosorbent assay (ELISA). A structured questionnaire was administered to determine the risk factors for porcine cysticercosis on the study farms. At pig level, the total number of pigs testing positive were 11, resulting in a seroprevalence of 4% (95% confidence interval (CI): 1.9–6.2%), while the farms with a positive pig were 9% (95% CI: 3.9–14.1%). All pigs examined in the slaughter slab survey were seronegative. The distribution of possible risk factors for porcine cysticercosis that were observed at farm level was as follows: free-range pig keeping (100%), history of human taeniosis infection in a family (51%), slaughtering of pigs at home (20%), lack of meat inspection (15%) and absence of latrines (15%). The only significant (χ2 = 4.4, P = 0.034, odds ratio (OR) = 3.8) risk factor associated with the occurrence of cysticercosis was lack of latrines at household level. The study shows that porcine cysticercosis is prevalent in free-range pigs in Busia District, Kenya and thus control measures need to be instituted.

Using an autopsy method of examination, an incidence of infection with Toxocara canis in 250 dogs was 6·4% and with Toxascaris leonina it was 2%. Of 100 cats examined, 8% harboured Toxocara cati and 3% Toxascaris leonina. Other British records concerning the incidence of the dog parasites are discussed and attention is drawn to certain reservations that must be placed on their acceptance.

Previous studies have suggested an immunomodulatory and even protective role for Enterobius vermicularis, the least pathogenic human intestinal helminth. Here, in a study using haematological and serological parameters, we tested a total of 215 children from central Greece, with a mean age of 8.39, of whom 105 (48.84%) were infected with E. vermicularis and 110 (51.16%) were matched healthy controls. In particular, we analysed eosinophil counts (EO), serum eosinophil cationic protein (ECP), total and specific serum immunoglobulin E (IgE) and the ECP/EO ratio. The atopic status and the potential occurrence of clinically expressed allergic diseases were both taken into account. Eosinophils, ECP and IgE were found to be higher in infected than in uninfected children, indicating a type-2 immune response activation during infection. Atopic infected children exhibited higher IgE levels compared to non-atopic ones. EO and ECP were found to be lower in atopic children who had a history of allergic disease than in those with no such history. The type-2 oriented immune response elicited against E. vermicularis could contribute to a balanced activation of the immune system in the examined children. Interestingly, although the atopic children showed a stronger activation, they did not exhibit any symptoms and, moreover, there seemed to be some indication of immunosuppression in those children with a positive history of allergic disease.

The present study was based on assessments of the molluscicidal and antiparasitic activities of Solanum nigrum villosum. This plant has been collected in Tozeur's traditional oases (south-western Tunisia). Molluscicidal activities of leaves, ripe fruit and unripe fruit extracts of S. n. villosum have been assessed in experimental and semi-field conditions on uninfected Galba truncatula. Antiparasitic activities have also been assessed on larval stages (rediae, intraredial germinal masses and cercariae) of the parasite Fasciola hepatica infecting G. truncatula naturally. The unripe fruit extract (LC50 = 41.2 mg l− 1) was more toxic than the leaf (LC50 = 132.5 mg l− 1) and the ripe fruit (LC50 = 172.8 mg l− 1) extracts on the snail after 48 h of treatment. One mg l− 1 of cupric chloride produced the death of 97% of G. truncatula after 25 days of treatment. Solanum n. villosum extracts used at the lethal laboratory concentration in semi-field conditions were also toxic to G. truncatula. The unripe fruit extract was more toxic than the leaf and the ripe fruit extracts on larval stages of F. hepatica. Maximum deterioration rates were obtained with the unripe fruit extract: 65.6% for rediae, 77.8% for cercariae and 27.6% for intraredial germinal masses after 48 h of treating the snails. The possibility of the unripe fruit extract application for the control of G. truncatula and F. hepatica larval stages should be considered.

1. A new species of Wuchereria is described, from the lymphatic systems of dogs, cats and Genet Cats (Genetta tigrina) on Pate Island off the Kenya coast. It is named W. patei after the island on which it was first discovered.

2. The new species belongs to the “W. malayi” group which are characterized in the microfilarial stage by the presence of one terminal and one sub-terminal nucleus in the tail; and in the adults by the presence of only 5 pairs of adanal papillae in the male and by the well-developed centre section in the left spicule.

Immunoreactive components of Opisthorchis viverrini and Clonorchis sinensis were analysed by enzyme-linked immunosorbent assay (ELISA), radioimmunoprecipitation and immunoblotting. Somatic extracts from these two liver flukes as well as from other related parasites, together with the metabolic products, were tested for their reactivities with sera from patients with opisthorchiasis and clonorchiasis. A significant cross-reactivity in the ELISA was noted between Opisthorchis and Clonorchis. Immunoblotting and radioimmunoprecipitation analyses showed that the 89-kD protein which was previously shown to be a predominant metabolic product of O. viverrini reacted with sera from both groups of patients. However, an antigen with a molecular weight of 16 kD, apparently a predominant somatic component, appeared to be specific for O. viverrini.

Hesperidin has been reported to exert a wide range of pharmacological effects, including antifungal, antiviral, antioxidant, anti-inflammatory and anticarcinogenic activities. Herein, the schistosomicidal activity of this compound was evaluated in vitro and in vivo. Using an in vitro assay, a concentration of 200 μg/ml of hesperidin resulted in the mortality of 100% adult worms of Schistosoma (S.) mansoni within 72 h and a partial tegumental alteration in 10% of worms. However, after 144 h incubation, 50 and 100 μg/ml concentrations showed 0% and 10% mortality in adult worms, respectively, without any changes to the tegument. Sublethal doses did not influence egg output nor the development of eggs deposited by pairs of adult worms. In an in vivo study, mice infected with S. mansoni and treated with 600 mg hesperidin/kg body weight showed a respective reduction of 50, 45.2, 50 and 47.5% of males, females, worm pairs and total worm burden. In addition, a respective reduction, based on the number of eggs/g tissue, of 41.5, 63.7 and 58.6% was observed in the liver, intestine and liver/intestinal tissue combined. Furthermore, S. mansoni-specific IgG level significantly increased with hesperidin treatment, whereas IgA and IgE levels were not significantly changed. IgM levels decreased in response to cercarial antigen preparation but were not altered in response to soluble worm or soluble egg antigen. As in hesperidin-treated mice, praziquantel-treated mice showed a similar pattern of specific antibody response to S. mansoni antigens. The present study represents the first report on the effects of the schistosomicidal activity of hesperidin.

A total of 549 birds from four orders were examined for nematodes in their respiratory system from 1995 to 2000. Twelve individuals of Falconiformes (n = 503), one of Gruiformes (n = 22) and one of Ciconiformes (n = 1), but no bird of the order Strigiformes (n = 23) were infected with syngamids. The syngamid species included Hovorkonema variegatum, Syngamus trachea and Cyathostoma trifurcatum from the trachea, bronchi and air sacs, with H. variegatum being the most prevalent. Cyathostoma trifurcatum from a black stork Ciconia nigra is a new record for Germany. The marsh harrier Circus aeruginosus and the white-tailed sea eagle Haliaeetus albicilla are new hosts for H. variegatum. Morphological characters such as the dorsal rays of the bursa copulatrix, length of the spicules and the mouth capsule are used to differentiate species of the family Syngamidae. Egg size is different between S. trachea and H. variegatum. In addition to morphological characters, the nucleotide sequence of the SSU ribosomal gene was determined for H. variegatum. Pairwise comparisons with the SSU sequence of S. trachea (AF036606) revealed sequence difference of 2.6%. The nucleotide sequence of the second internal transcribed spacer of ribosomal DNA for different populations of H. variegatum was also determined. Pairwise comparisons revealed two separate strains with a sequence difference of 14.0% to 14.5% suggesting the existence of a cryptic species. Pathological findings associated with H. variegatum were found in 7 of 12 cases and consisted of thickened air sac walls and lesions or granuloma at the site of attachment of the worm, which occasionally involved the underlaying tissues. Lymphoplasmocytic air sacculitis was the most prominent histological lesion found.

Schistosoma japonicum and S. mansoni were tested for reactivity with an anti-inducible nitric oxide (iNOS) antibody and the distribution of iNOS was studied by immunofluorescent tests in different stages of the parasites. Reactivity was associated with the tegument in both larval schistosomes (sporocysts and cercariae) and eggs. With adult worms, the majority of the immunofluorescence was predominantly subtegumental in S. japonicum and parenchymal in S. mansoni. Fluorescence was also observed in host tissues (snails and mouse liver). In Western blots, the enzyme of S. japonicum had an apparent molecular weight of about 210 kDa. The possible role of worm and host iNOS in the parasite–host interrelation remains to be clarified.

Inequalities in body size among adult helminths can result in inequalities in reproductive output, with consequences for population dynamics and genetics. These inequalities can result from growth differences among larval worms inside intermediate hosts that persist into the adult stage. Here, we investigate the effects of both host body size and intensity of infection on the sizes of metacercariae of the trematode Maritrema novaezealandensis (Microphallidae) inside their second intermediate host, the isopod Paridotea ungulata (Idoteidae). Among the more than 1500 metacercariae recovered and individually measured, there was no relationship between the mean diameter of metacercarial cysts per isopod and isopod body length. However, intensity of infection correlated negatively with the mean diameter of cysts within an isopod, i.e. metacercariae in crowded infections attained smaller sizes on average. In contrast, the variability in cyst sizes per isopod, measured as the coefficient of variation, was independent of both isopod body length and infection intensity. Our results show that a disproportionate number of relatively small metacercariae come from the relatively few hosts in which a large fraction of all metacercariae are aggregated. The combination of aggregation and intensity-dependent growth generates inequalities in sizes among metacercariae that will be passed on to adult worm populations in definitive hosts.

A substantial level of resistance to a lictcrologous challenge with Fasciola hepatica was demonstrated insheep harbouring primary non-patent (two- to three-week-old) and newly patent (seven- to cight-wcck-old)Schistosoma hovis infections, the liver-fluke burdens being reduced by 70% and 93%, respectively, ( p< 0·01) inthese groups compared with that of the challenge control group.

The liver fluke, Opisthorchis viverrini, and the minute intestinal fluke, Haplorchis taichui, are prevalent in many Asian countries. This study analysed the patterns of infections of O. viverrini and H. taichui in Lahanam and Thakhamlien villages (Savannakhet Province, Lao PDR), in two cross-sectional investigations. Out of a total of 207 human participants, post-anthelmintic treatment positivity rates for expelled worms were 170 (82.1%) for H. taichui and 65 (31.4%) for O. viverrini. Both these species co-exist in the study villages. When each parasite was analysed separately, H. taichui infections reached a plateau among people aged >20 years. Opisthorchis viverrini infection rates were highest in the age group 21–30 years, with decreasing infection rates after the age of 30. Our findings indicated that fish-borne trematode infections were more prevalent among adults. Fish, common intermediate hosts, were acquired in the study area for analysis. The examination of 35 species of fish as intermediate hosts found O. viverrini metacercariae in only six species, and these were found mostly during the month of November. Many farmers who live on the rice fields obtain their food from their immediate environment, including these intermediate-host fish, potentially putting them at greater risk of O. viverrini infection. By contrast, H. taichui metacercariae were found in three species of fish obtained from the market, meaning that anyone could consume them and become infected. If people who work in rice fields limit the species of fish they consume, or avoid consuming raw fish during the month of November, they may reduce their risk of O. viverrini infection.

In the past two decades, considerable evidence has accumulated in the literature about the differences in the susceptibility of various intermediate hosts of Schistosoma mansoni to different strains of the parasite. Comprehensive studies on this aspect of host-parasite relationship have been published by Files & Cram (1949), Abdel-Malek (1950) and Files (1951). The results of more recent studies have been reported by Wright (1962) and Saoud (1964).

In the present paper, the writer has studied the susceptibility of four intermediate hosts of S. mansoni from Brazil, Puerto Rico, Egypt and Tanganyika to some strains of the parasite.

The model of Onchocerca lienalis microfilariae (mf) injected into inbred CBA/Ca mice was studied for its usefulness as an additional primary/secondary drug screen for onchocerciasis. Invermectin, DEC, suramin, flubendazole, mebendazole, levamisole, Mel W, furapyrimidone, metrifonate, amoscanate and the new Ciba-Geigy compounds CGP 6140, CGP 20'376 and CGI 17658 all significantly reduced levels of mf at a dose of 5x100 mg/kg or less. An early dosing protocol, on days 3–7 after infection, was found to be generally more effective than dosing on days 11–15, followed by necropsy on day 18. In some cases there were important differences in levels of drug activity depending on whether the drug was administered by the subcutaneous or oral route, indicating that new compounds should be tested via both routes. Ivermectin was by far the most active compound examined, virtually clearing mf from the skin at a dose of 5x0.0063 mg/kg and producing a significant mf reduction (63.5%) at 5x0.008 mg/kg following subcutaneous administration. In comparison, DEC was much less active, producing a 32.4% mf reduction at 5x25 mg/kg ranging up to a maximum of 72% reduction at 5x100 mg/kg. CGI 17658 was the most active compound examined next to ivermectin, almost 100% effective against skin mf at a dose of 5x6.25 mg/kg via the oral route while being less effective via subcutaneous administration (65% reduction). The lowest effective dose examined was 5x3.13 mg/kg (per os) which reduced mf levels by 64%. CGP 20'376 was also very active, resulting in a 46% (subcutaneous) and 62% (per os) reduction at a dose of 5x6.25 mg/kg. This mouse model has clearly identified all the known microfilaricides examined and also, to a lesser extent, those compounds considered to be principally macrofilaricides. We believe it has value as an additional drug screen for onchocerciasis, which will enable the evaluation of novel compounds against skin-dwelling Onchocerca mf at the primary/secondary level, providing complementary information to new in vitro screens using adult Onchocerca.

1. The lower intestinal tracts (duodenum to rectum) of 267 adult domestic fowls were examined for helminth parasites and 91% were found to be infested. The average numbers of worms and species present in infested birds were 240.6 and 2.42 respectively. Twelve species of parasites were recovered but only 5 occurred in more than 20% of the birds.

2. Twenty-six casualty and diseased chickens between 2 and 12 weeks old were examined. Fourteen were infested with helminth parasites and the average infestation was 499.9 worms of 2.79 species.

3. The total numbers of worms of all species present in individual fowls were shown to bear a relationship to the number of species comprising the infestations. Linear and quadratic equations which demonstrate the relationship have been derived from the observed figures.

1. The adult stage of Polystoma integerrimum feeds on blood which is digested by a combination of intraluminar and intracellular processes, haemolysis and partial digestion in the gut lumen being followed by absorption and completion of digestion by the cells of the gastrodermis.

2. The end products of intracellular digestion are the pigment haematin and a crystalline substance identified as haematoidin, which is chemically identical with vertebrate bilirubin, and these are passed back into the lumen to be eventually voided to the exterior.

3. It is probable that the expulsion of haematin from the gut cells results in a simultaneous release of enzymes, traces of which persist in the lumen to effect the early extracellular stages in the digestion of the next meal, so that intraluminar digestion in Polystoma appears to be an incidental extension of the primitive intracellular process arising from the blood-feeding habit and the consequent need to eliminate residues of haemoglobin breakdown from the gastrodermis.

4. Since haematin formation has been described in certain other trematodes it is suggested that their digestive processes probably follow a course similar to that described for Polystoma but an alternative method must exist within the class since other blood feeders such as Haplometra and Gorgodera do not form this pigment.

The nematode communities in spruce forests were compared with the short-term effects of forest damage, caused by windstorm, wildfire and management practices of forest soils. Soil samples were collected in June and October from 2006 to 2008 in four different sites: (1) forest unaffected by the wind (REF); (2) storm-felled forest with salvaged timber (EXT); (3) modified forest affected by timber salvage (wood removal) and forest fire (FIR); and (4) storm-felled forest where timber had been left unsalvaged (NEX). Nematode analysis showed that the dominant species in all four investigated sites were Acrobeloides nanus and Eudorylaimus silvaticus. An increase of A. nanus (35% of the total nematode abundance) in the first year in the FIR site led to the highest total abundance of nematodes compared with other sites, where nematode abundance reached the same level in the third year. In the FIR site bacterial feeders appeared to be the most representative trophic group, although in the second and third year, after disturbance, the abundance of this trophic group gradually decreased. In the NEX site, the number of nematode species, population densities and Maturity Index were similar to that recorded for the FIR site. In EXT and NEX sites, the other dominant species was the plant parasitic nematode Paratylenchus microdorus. Analyses of nematodes extracted from different forest soil samples showed that the highest number of species and diversity index for species (H'spp) were in the REF site. Differences between the nematode fauna in REF and other localities were clearly depicted by cluster analysis. The greatest Structure Index and Enrichment Index values were also in REF. In the EXT site, the number of nematode species, their abundance, H'spp and Maturity Index were not significantly different from those recorded in the reference site.

Entomopathogenic nematodes (EPNs) of the families Steinernematidae and Heterorhabditidae have a symbiotic association with bacteria which makes them virulent against insects. EPNs have been mass produced using in vivo and in vitro methods, including both solid and liquid fermentation. This study assessed the effect of nematode inoculum age on the production of Steinernema feltiae in liquid, solid and biphasic processes. Several physical parameters were also assessed: the effect of medium viscosity, flask size and aeration speed on the recovery and yield of infective juveniles (IJs). Inoculum age treatments included inoculum liquid cultures that were 7, 14, 21 and 28 days old. Nematodes from the same inoculum were added to one liquid medium (liquid culture), one solid medium with bacteria previously grown in sponge (solid culture) and a variation of the solid medium (a biphasic culture), in which the bacteria were first grown in liquid and, then, soaked into the sponges, with the purpose of providing a more homogeneous bacterial culture before nematode inoculation. Experiments were conducted in Erlenmeyer flasks. Eight treatments were established involving combinations of three variables: two media (with and without 0.2% agar), two flask sizes (250 and 150 ml) and two agitation speeds (180 and 280 rpm). The study showed increases in nematode yield for liquid cultures, but not for solid or biphasic cultures, with the advance of the inoculum age up to 28 days of growth. Furthermore, the addition of 0.2% agar to the liquid medium and increasing the aeration rate by using larger flasks with higher agitation speed may increase nematode recovery and final yield. The experiments were conducted using shake flasks but the results may also be applicable for bioreactors.
