Synthetic geomicrobiology is a potentially new branch of synthetic biology that seeks to achieve improvements in microbe–mineral interactions for practical applications. In this paper, laboratory and field data are provided on three geomicrobiology challenges in space: (1) soil formation from extraterrestrial regolith by biological rock weathering and/or the use of regolith as life support system feedstock, (2) biological extraction of economically important elements from rocks (biomining) and (3) biological solidification of surfaces and dust control on other planetary surfaces. The use of synthetic or engineered organisms in these three applications is discussed. These three examples are used to extract general common principles that might be applied to the design of organisms used in synthetic geomicrobiology.

Infrared spectroscopic observations of planetary nebulae and proto-planetary nebulae have shown that complex organic compounds are synthesized in these objects over periods as short as a thousand years. These compounds are ejected into the interstellar medium and spread throughout the Galaxy. Evidence from meteorites has shown that these stellar grains have reached the Solar System, and may have showered the Earth during the heavy bombardment stage of the Early Earth. In this paper, we discuss the chemical structure of stellar organic grains and compare them to the organic matter found in meteorites, comets, asteroids, planetary satellites, and interplanetary particles. The possibility that the early Solar System was chemically enriched by organic compounds ejected from distant stars is presented.

The detection and atmospheric characterization of super-Earths is one of the major frontiers of exoplanetary science. Currently, extensive efforts are underway to detect molecules, particularly H2O, in super-Earth atmospheres. In the present work, we develop a systematic set of strategies to identify and observe potentially H2O-rich super-Earths that provide the best prospects for characterizing their atmospheres using existing instruments. First, we provide analytic prescriptions and discuss factors that need to be taken into account while planning and interpreting observations of super-Earth radii and spectra. We discuss how observations in different spectral bandpasses constrain different atmospheric properties of a super-Earth, including radius and temperature of the planetary surface as well as the mean molecular mass, the chemical composition and thermal profile of the atmosphere. In particular, we caution that radii measured in certain bandpasses can induce biases in the interpretation of the interior compositions. Second, we investigate the detectability of H2O-rich super-Earth atmospheres using the Hubble Space Telescope Wide Field Camera 3 spectrograph as a function of the planetary properties and stellar brightness. We find that highly irradiated super-Earths orbiting bright stars, such as 55 Cancri e, present better candidates for atmospheric characterization compared to cooler planets such as GJ 1214b even if the latter orbit lower-mass stars. Besides being better candidates for both transmission and emission spectroscopy, hotter planets offer higher likelihood of cloud-free atmospheres which aid tremendously in the observation and interpretation of spectra. Finally, we present case studies of two super-Earths, GJ 1214b and 55 Cancri e, using available data and models of their interiors and atmospheres.

A ‘habitable zone’ of a star is defined as a range of orbits within which a rocky planet can support liquid water on its surface. The most intriguing question driving the search for habitable planets is whether they host life. But is the age of the planet important for its habitability? If we define habitability as the ability of a planet to beget life, then probably it is not. After all, life on Earth has developed within only ~800 Myr after its formation – the carbon isotope change detected in the oldest rocks indicates the existence of already active life at least 3.8 Gyr ago. If, however, we define habitability as our ability to detect life on the surface of exoplanets, then age becomes a crucial parameter. Only after life had evolved sufficiently complex to change its environment on a planetary scale, can we detect it remotely through its imprint on the atmosphere – the so-called biosignatures, out of which the photosynthetic oxygen is the most prominent indicator of developed (complex) life as we know it. Thus, photosynthesis is a powerful biogenic engine that is known to have changed our planet's global atmospheric properties. The importance of planetary age for the detectability of life as we know it follows from the fact that this primary process, photosynthesis, is endothermic with an activation energy higher than temperatures in habitable zones, and is sensitive to the particular thermal conditions of the planet. Therefore, the onset of photosynthesis on planets in habitable zones may take much longer time than the planetary age. The knowledge of the age of a planet is necessary for developing a strategy to search for exoplanets carrying complex (developed) life – many confirmed potentially habitable planets are too young (orbiting Population I stars) and may not have had enough time to develop and/or sustain detectable life. In the last decade, many planets orbiting old (9–13 Gyr) metal-poor Population II stars have been discovered. Such planets had had enough time to develop necessary chains of chemical reactions and may carry detectable life if located in a habitable zone. These old planets should be primary targets in search for the extraterrestrial life.

The Regenerative Enclosed Life Support Module Simulator (REMS) was designed to simulate the conditions aboard the International Space Station (ISS). This unique terrestrial, encapsulated environment for humans and their associated organisms allowed investigations into the microbial communities within an enclosed habitat system, primarily with respect to diversity, phylogeny and the possible impact on human health. To assess time- and/or condition-dependent changes in microbial diversity within REMS, a total of 27 air samples were collected during three consecutive months. The microbial burden and diversity were elucidated using culture-dependent and culture-independent molecular methods. The results indicate that during controlled conditions the total microbial burden detected by culture-dependent techniques (below a detectable level to 102 cells m−3 of air) and intracellular ATP assay was significantly low (102–103 cells m−3 of air), but increased during the uncontrolled post-operation phase (∼104 cells m−3 of air). Only Gram-positive and α-proteobacteria grew under tested culture conditions, with a predominant occurrence of Methylobacterium radiotolerans, and Sphingomonas yanoikuyae. Direct DNA extraction and 16S rDNA sequencing methodology revealed a broader diversity of microbes present in the REMS air (51 species). Unlike culture-dependent analysis, both Gram-positive and proteobacteria were equally represented, while members of a few proteobaterial groups dominated (Rhodopseudomonas, Sphingomonas, Acidovorax, Ralstonia, Acinetobacter, Pseudomonas, and Psychrobacter). Although the presence of several opportunistic pathogens warrants further investigation, the results demonstrated that routine maintenance such as controlling the humidity, crew’s daily cleaning, and air filtration were effective in reducing the microbial burden in the REMS.

Astrobiology is an extremely wide-ranging field and thus is in special need of conceptual and theoretical frameworks that can integrate its various arenas of study. In this paper I review recent work associated with a conceptual framework known as ‘ecological stoichiometry’ and even more recent extensions in the development of ‘biological stoichiometry’. Ecological stoichiometry is the study of the balance of energy and multiple chemical elements in ecological interactions and has developed rapidly in the study of nutrient cycling and energy flow in aquatic food webs. It identifies the elemental composition of interacting biota as central in understanding the nature of their interactions and dynamics, including key feedbacks via nutrient recycling. Biological stoichiometry extends this mode of thinking to all types of biological systems. It especially seeks to better understand, at the biochemical and genetic levels, the factors influencing the elemental composition of living things and the evolutionary forces that drive and constrain that elemental composition. By connecting key concepts of ecosystem ecology, evolutionary biology and biochemistry, stoichiometric theory integrates biological information into a more coherent whole that holds considerable promise for application in astrobiology. Several examples of potential astrobiological applications of stoichiometric analysis are offered, including ones related to pre-biotic evolution, the Cambrian explosion, biosignatures and biological feedbacks on planetary carbon cycling.

We propose that nanophase iron-oxide-bearing materials provided important niches for ancient photosynthetic microbes on the Earth that ultimately led to the oxygenation of the Earth's atmosphere and the formation of iron-oxide deposits. Atmospheric oxygen and ozone attenuate ultraviolet radiation on the Earth today providing substantial protection for photosynthetic organisms. With ultraviolet radiation fluxes likely to have been even higher on the early Earth than today, accessing solar radiation was particularly risky for early organisms. Yet, we know that photosynthesis arose early and played a critical role in subsequent evolution. Of primary importance was protection below 290 nm, where peak nucleic acid (~260 nm) and protein (~280 nm) absorptions occur. Nanophase ferric oxide/oxyhydroxide minerals absorb, and thus block, the lethal ultraviolet radiation, while transmitting light through much of the visible and near-infrared regions of interest to photosynthesis (400 to 1100 nm). Furthermore, they were available in early environments, and are synthesized by many organisms. Based on experiments using nanophase ferric oxide/oxyhydroxide minerals as a sunscreen for photosynthetic microbes, we suggest that iron, an abundant element widely used in biological mechanisms, may have provided the protection that early organisms needed in order to be able to use photosynthetically active radiation while being protected from ultraviolet-induced damage. The results of this study are broadly applicable to astrobiology because of the abundance of iron in other potentially habitable bodies and the evolutionary pressure to utilize solar radiation when available as an energy source. This model could apply to a potential life form on Mars or other bodies where liquid water and ultraviolet radiation could have been present at significant levels. Based on ferric oxide/oxyhydroxide spectral properties, likely geologic processes, and the results of experiments with the photosynthetic organisms, Euglena sp. and Chlamydomonas reinhardtii, we propose a scenario where photosynthesis, and ultimately the oxygenation of the atmosphere, depended on the protection of early microbes by nanophase ferric oxides/oxyhydroxides.

We use the critical step model to study the major transitions in evolution on Earth. We find that a total of five steps represents the most plausible estimate, in agreement with previous studies, and use the fossil record to identify the potential candidates. We apply the model to Earth-analogs around stars of different masses by incorporating the constraints on habitability set by stellar physics including the habitable zone lifetime, availability of ultraviolet radiation for prebiotic chemistry, and atmospheric escape. The critical step model suggests that the habitability of Earth-analogs around M-dwarfs is significantly suppressed. The total number of stars with planets containing detectable biosignatures of microbial life is expected to be highest for K-dwarfs. In contrast, we find that the corresponding value for intelligent life (technosignatures) should be highest for solar-mass stars. Thus, our work may assist in the identification of suitable targets in the search for biosignatures and technosignatures.

SHARK is a proposal aimed at investigating the technical feasibility and the scientific capabilities of high-contrast cameras to be implemented at the Large Binocular Telescope (LBT). SHARK foresees two separated channels: near-infrared (NIR) channel and visible, both providing imaging and coronagraphic modes. We describe here the SHARK instrument concept, with particular emphasis on the NIR channel at the level of a conceptual study, performed in the framework of the call for proposals for new LBT instruments. The search for giant extra-Solar planets is the main science case, as we will outline in the paper.

The subsurface exploration of other planetary bodies can be used to unravel their geological history and assess their habitability. On Mars in particular, present-day habitable conditions may be restricted to the subsurface. Using a deep subsurface mine, we carried out a program of extraterrestrial analog research – MINe Analog Research (MINAR). MINAR aims to carry out the scientific study of the deep subsurface and test instrumentation designed for planetary surface exploration by investigating deep subsurface geology, whilst establishing the potential this technology has to be transferred into the mining industry. An integrated multi-instrument suite was used to investigate samples of representative evaporite minerals from a subsurface Permian evaporite sequence, in particular to assess mineral and elemental variations which provide small-scale regions of enhanced habitability. The instruments used were the Panoramic Camera emulator, Close-Up Imager, Raman spectrometer, Small Planetary Linear Impulse Tool, Ultrasonic drill and handheld X-ray diffraction (XRD). We present science results from the analog research and show that these instruments can be used to investigate in situ the geological context and mineralogical variations of a deep subsurface environment, and thus habitability, from millimetre to metre scales. We also show that these instruments are complementary. For example, the identification of primary evaporite minerals such as NaCl and KCl, which are difficult to detect by portable Raman spectrometers, can be accomplished with XRD. By contrast, Raman is highly effective at locating and detecting mineral inclusions in primary evaporite minerals. MINAR demonstrates the effective use of a deep subsurface environment for planetary instrument development, understanding the habitability of extreme deep subsurface environments on Earth and other planetary bodies, and advancing the use of space technology in economic mining.

The mean air temperature of the Icelandic interior is below 10 °C. However, we have previously observed 16S rDNA sequences associated with thermophilic lineages in Icelandic basalts. Measurements of the temperatures of igneous rocks in Iceland showed that solar insolation of these low albedo substrates achieved a peak surface temperature of 44.5 °C. We isolated seven thermophilic Geobacillus species from basalt with optimal growth temperatures of ~65 °C. The minimum growth temperature of these organisms was ~36 °C, suggesting that they could be active in the rock environment. Basalt dissolution rates at 40 °C were increased in the presence of one of the isolates compared to abiotic controls, showing its potential to be involved in active biogeochemistry at environmental temperatures. These data raise the possibility of transient active thermophilic growth in macroclimatically cold rocky environments, implying that the biogeographical distribution of active thermophiles might be greater than previously understood. These data show that temperatures measured or predicted over large scales on a planet are not in themselves adequate to assess niches available to extremophiles at micron scales.

The deep subsurface of other planetary bodies is of special interest for robotic and human exploration. The subsurface provides access to planetary interior processes, thus yielding insights into planetary formation and evolution. On Mars, the subsurface might harbour the most habitable conditions. In the context of human exploration, the subsurface can provide refugia for habitation from extreme surface conditions. We describe the fifth Mine Analogue Research (MINAR 5) programme at 1 km depth in the Boulby Mine, UK in collaboration with Spaceward Bound NASA and the Kalam Centre, India, to test instruments and methods for the robotic and human exploration of deep environments on the Moon and Mars. The geological context in Permian evaporites provides an analogue to evaporitic materials on other planetary bodies such as Mars. A wide range of sample acquisition instruments (NASA drills, Small Planetary Impulse Tool (SPLIT) robotic hammer, universal sampling bags), analytical instruments (Raman spectroscopy, Close-Up Imager, Minion DNA sequencing technology, methane stable isotope analysis, biomolecule and metabolic life detection instruments) and environmental monitoring equipment (passive air particle sampler, particle detectors and environmental monitoring equipment) was deployed in an integrated campaign. Investigations included studying the geochemical signatures of chloride and sulphate evaporitic minerals, testing methods for life detection and planetary protection around human-tended operations, and investigations on the radiation environment of the deep subsurface. The MINAR analogue activity occurs in an active mine, showing how the development of space exploration technology can be used to contribute to addressing immediate Earth-based challenges. During the campaign, in collaboration with European Space Agency (ESA), MINAR was used for astronaut familiarization with future exploration tools and techniques. The campaign was used to develop primary and secondary school and primary to secondary transition curriculum materials on-site during the campaign which was focused on a classroom extra vehicular activity simulation.

The interest of exo-moons for Astrobiology is that they widen significantly the potential niches of exolife. In this paper, after a review of standard detection methods, we investigate new ways to characterize exo-moons and to use them to characterize their home planetary system.

The discovery of thousands of exoplanets in the last two decades has uncovered a wide diversity of planets that are very different from those in our own Solar System. Ideas for how to detect signs of life in the variety of planetary possibilities, by way of biosignature gases, are expanding, although they largely remain grounded in study of familiar gases produced by life on Earth and how they appear in Earth's spectrum as viewed as an exoplanet. What are the chances we will be able to observe and identify biosignature gases on exoplanets in the coming two decades? I review the status of the search for habitable planets and biosignature gases framed by a ‘Biosignature Drake Equation’.

There is increasing evidence that Mars may have once been a habitable environment. Gypsum is targeted in the search for Martian biosignatures because it can host extensive cryptoendolithic communities in extreme terrestrial environments and is widespread on Mars. In this study the viability of using different spectroscopy-based techniques to identify the presence of gypsum endolithic communities was investigated by analysing various cryptoendoliths collected from the Lake St. Martin impact crater (LSM), a Mars analogue site found in Manitoba, Canada. Concurrently, the cryptoendolithic microbial community structure present was also analysed to aid in assigning spectroscopic features to microbial community members. Two main morphologies of endolithic communities were collected from gypsum deposits at LSM: true cryptoendolithic communities and annular deposits on partially buried boulders and cobbles <1 cm below the soil surface. Endolithic communities were found to be visibly present only in gypsum with a high degree of translucency and could occur as deep as 3 cm below the exterior surface. The bacterial community was dominated by a phylum (Chloroflexi) that has not been previously observed in gypsum endoliths. The exterior surfaces of gypsum boulders and cobbles are devoid of spectroscopic features attributable to organic molecules and detectable by reflectance, Raman, or ultraviolet-induced fluorescence spectroscopies. However, exposed interior surfaces show unique endolithic signatures detectable by each spectroscopic technique. This indicates that cryptoendolithic communities can be detected via spectroscopy-based techniques, provided they are either partially or fully exposed and enough photon–target interactions occur to enable detection.

Impact-generated hydrothermal systems are commonly proposed as good candidates for hosting primitive life on early Earth and Mars. However, evidence of fossil microbial colonization in impact-generated hydrothermal systems is rarely reported in the literature. Here we present the occurrence of putative fossil microorganisms in a hydrothermal system of the 89 Ma Dellen impact structure, Sweden. We found the putative fossilized microorganisms hosted in a fine-grained matrix of hydrothermal alteration minerals set in interlinked fractures of an impact breccia. The putative fossils appear as semi-straight to twirled filaments, with a thickness of 1–2 μm, and a length between 10 and 100 μm. They have an internal structure with segmentation, and branching of filaments occurs frequently. Their composition varies between an outer and an inner layer of a filament, where the inner layer is more iron rich. Our results indicate that hydrothermal systems in impact craters could potentially be capable of supporting microbial life. This could have played an important role for the evolution of life on early Earth and Mars.

Crossed-beam experiments on the reactions of cyano CN(X2Σ+) and ethinyl C2H(X2Σ+) radicals with the unsaturated hydrocarbons acetylene, ethylene, methylacetylene allene and benzene have been carried out under single-collision conditions to investigate synthetic routes to form nitriles, polyynes and substituted allenes in hydrocarbon-rich atmospheres of planets and their moons. All reactions were found to proceed without an entrance barrier, to have exit barriers well below the energy of the reactant molecules and to be strongly exothermic. The predominant identification of the radical versus atomic hydrogen exchange channel makes these reactions compelling candidates for the formation of complex organic chemicals – precursors to biologically important amino acids – in Solar system environments and in the interstellar medium.

Cloud formation modelling has entered astrophysics as a new field of research for planetary and brown dwarf atmospheres. Clouds are a chemically and physically very active component of an atmosphere since they determine the remaining gas phase and change the object's albedo depending on their material composition. The grains can also provide a surface where the molecular constituents for life can be physisorbed for possible pre-biotic evolution. This paper summarizes our model for the kinetic formation of dirty dust grains which make up the atmospheric clouds of extraterrestrial giant gas planets. We include seed formation, surface growth and evaporation, the gravitational settling that influences the dust formation, element depletion that determines the remaining gas phase abundances, and convective overshooting that is needed for a dust model to be applicable to hydrostatic atmosphere simulations. We demonstrate the evolution of the material composition of the cloud itself and the distribution of the grain sizes in the cloud layer, exemplary for a giant gas planet parameter combinations (Teff, log g). In general, substellar clouds are composed of small, dirty grains with a high silicate content at the cloud deck. They grow in size and gradually purify to iron/corundum grains when they move into denser and hotter atmospheric regions. Comparing these results with experimental data from condensation experiments leads to the conclusion that cloud grains that gravitationally settle in the atmosphere of a giant planet can easily change their lattice structure from the disordered amorphous state they are forming in, into the ordered lattice of a crystal.

In the frame of the EXPOSE-R mission outside the Russian Zvezda Module of the International Space Station (ISS) passive thermoluminescence dosimeters were applied to measure the radiation exposure of biological samples. The detectors were located beneath the sample carriers to determine the dose levels for maximum shielding. The dose measured beneath the sample carriers varied between 317 ± 10 and 230 ± 2 mGy, which amount to an average dose rate of 381 ± 12 and 276 ± 2 μGy d−1. These values are close to those assessed for the interior of the ISS and reflect the high shielding of the biological experiments within the EXPOSE-R facility. As a consequence of the high shielding (several g cm−2), the biological samples were predominantly exposed to galactic cosmic heavy ions and trapped protons in the Earth's radiation belts, whereas the trapped electrons did not reach the samples.

The polar desert is one of the most extreme environments on Earth. Endolithic organisms can escape or mitigate the hazards of the polar desert by using the resources available in the interior of rocks. We examined endolithic communities within crystalline rocks that have undergone shock metamorphism as a result of an asteroid or comet impact. Specifically, we present a characterization of the heterotrophic endolithic community and its environment in the interior of impact-shocked gneisses and their host polymict breccia from the Haughton impact structure on Devon Island, Nunavut, Canadian High Arctic. Microbiological colonization of impact-shocked rocks is facilitated by impact-induced fissures and cavities, which occur throughout the samples, the walls of which are lined with high abundances of biologically important elements owing to the partial volatilization of minerals within the rock during the impact. 27 heterotrophic bacteria were isolated from these shocked rocks and were identified by 16S rDNA sequencing. The isolates from the shocked gneiss and the host breccia are similar to each other, and to other heterotrophic communities isolated from polar environments, suggesting that the interiors of the rocks are colonized by microorganisms from the surrounding country rocks and soils. Inductively coupled plasma–atomic emission spectroscopy (ICP-AES), scanning electron microscopy (SEM) and energy dispersive X-ray (EDX) analysis were used to identify the chemical composition of the shocked materials and to document the in situ growth of microbes in their interiors. The identification of these heterotrophic communities within impact-shocked crystalline rocks extends our knowledge of the habitable biosphere on Earth. The colonization of the interiors of these samples has astrobiological applications both for considering terrestrial, microbiological contamination of meteorites from the Antarctic ice sheet and for investigating possible habitats for microbial organisms on the early Earth, and more speculatively, on Mars and other planetary bodies.
