The anabolic effects of androgen on skeletal muscles are thought to be mediated by androgen receptor (AR). Although multiple studies concerning the effects of AR in males have been performed, the molecular mechanisms of AR in skeletal muscles remain unclear. Here we first confirmed that satellite cells from mouse hindlimb muscles express AR. We then generated satellite cell-specific AR knockout mice using Pax7CreERT2 and ARL2/Y mice to test whether AR in satellite cells is necessary for muscle regeneration. Surprisingly, we found that muscle regeneration was compromised in both Pax7CreERT2(Fan)/+ control mice and Pax7CreERT2(Fan)/+;ARL2/Y mice compared to ARL2/Y mice. However, Pax7CreERT2(Gaka)/+;ARL2/Y;R26tdTomato/+ mice showed no significant differences between control and mutant muscle regeneration. These findings indicate that AR in satellite cells is not essential for muscle regeneration. We propose that Pax7CreERT2(Fan)/+ control mice should be included in all experiments, because these mice negatively affect the muscle regeneration and show the mild regeneration phenotype.

Inhibitory control can be divided into motor and cognitive inhibition. The current research is the first study exploring the impact of brief mindfulness training on motor inhibition, measured by a stop signal task in participants without any meditation experience. Motor inhibition performance was compared before and immediately after three different conditions; a brief mindfulness induction, a resting state and an active control session in which participants listened to their favorite music. Post-test learning effect on go-reaction times was seen for the resting and mindfulness conditions, but was absent in the music session, possibly due to emotional arousal might have led slower responses. Brief mindfulness training did not significantly alter inhibitory control, although marginal improvement in stop signal reaction time following the mindfulness induction was observed. Motor inhibition appears unresponsive to either short-term or long-term mindfulness practice. Future mindfulness studies should explore a broad spectrum of cognitive functions and populations.

An increase in oyster aquaculture as a sustainable method of shellfish production is one response to overharvest and degradation of natural oyster reefs over the past century. Successful aquaculture production requires determining the environmental conditions optimal for oyster growth. In this study, the salinity, temperature, chlorophyll a concentration and the growth of Crassostrea virginica were monitored at four locations within the Mission-Aransas Estuary, Texas (USA), a shallow subtropical estuary influenced by relatively low freshwater inflow. Mean growth of the oyster shell (0.205 mm d–1 and 0.203 g d–1) and soft tissues (3.447 mg d–1) was highest when salinity was low (mean = 15.5) and chlorophyll a concentration was high (8.4 μg l–1). Oyster growth also varied temporally with periods of spawning. In low-inflow estuaries such as the Mission-Aransas Estuary, oyster farms should be sited close to river mouths so that oysters can benefit from freshwater inflows and lower salinities.

During summer 2020, observations of the mesosphere using a 53.5 MHz radar on Svalbard, at 78.2°N 15.1°E, revealed the well-known Polar Mesospheric Summer Echoes (PMSE). At the same time, a co-located meteor detection radar, operating at 31 MHz detected corresponding echoes very distinct from those associated with meteor trails. Comparing as many days as possible during 2020, incontestable evidence arose to demonstrate that the meteor detection radar was capable of observing PMSE, although not in the optimised fashion of the 53.5 MHz system. We present examples of results from both systems, supplementing the earlier findings of Swarnalingam et al. (2009), and simultaneously show very first results from this particular geographical location.

Dynamic nuclear polarization (DNP) is a technique in magic-angle spinning (MAS) nuclear magnetic resonance (NMR) which leads to sensitivity enhancement and helps to overcome the issue of low polarization in detected nuclei. Recent research showed, that methyl groups, which show active reorientation dynamics and cause heteronuclear cross relaxation at typical DNP temperatures around 100 K, may be used as a pinpoint source of polarization for selective and site-specific probing. In this study, we investigated the cross-relaxation behavior of methyl groups in nicotine and caffeine under DNP. These effects could be useful for investigating receptor/ligand binding.

In this study, we define the cardinal temperatures and thermal time for germination and emergence of pigeonpea genotypes. Seeds of six genotypes were subjected to constant temperatures ranging between 5 and 50°C in petri dishes with filter paper (germination) and with media (emergence) were placed in a thermal gradient plate. A nonlinear bent-stick model fitted to the rate of development to germination and emergence resulted in parameters predicting cardinal temperatures including base (Tb), optimum (To), maximum (Tm), and thermal time. Estimated Tb for 50% germination and emergence were 8.4 and 10.8°C, respectively, with no significant differences between genotypes. Optimum temperatures were 33.8 and 37.9°C for germination and emergence, respectively, with genotypes differing significantly. Thermal time for 50% germination and emergence varied significantly among genotypes. The results suggest that genotypic responses to the temperature are typical for their tropical origin and hence their suitability for cropping in summer dominant rainfall regions insubtropical Australia.

Lignin is a polyphenolic compound found in plant tissues, especially wood and bark. The lignin content determines the quality of wood biochar in agroecological uses, and is used in the production of synthetic resins and adhesives. Despite its importance in plant physiology and its agricultural and industrial utility, there exists a wide gap of knowledge of lignin contents of tropical hardwood trees of South Asia, except for a few species. We present here the first estimation of lignin content in wood and bark of 48 species from tropical deciduous forests of India. We show that some species are characterized by greater wood lignin (WL) compared with bark lignin (BL) content, contrary to the generalization held for hardwood trees, and suggest a plausible correspondence between the WL to BL ratio and timber quality.

Motor adaptation is a process by which the brain gradually reduces error induced by a predictable change in the environment, e.g., pointing while wearing prism glasses. It is thought to occur via largely implicit processes, though explicit strategies are also thought to contribute. Research suggests a role of the cerebellum in the implicit aspects of motor adaptation. Using non-invasive brain stimulation, we sought to investigate the involvement of the cerebellum in implicit motor adaptation in healthy participants. Inhibition of the cerebellum was attained through repetitive transcranial magnetic stimulation (rTMS), after which participants performed a visuomotor-rotation task while using an explicit strategy. Adaptation and aftereffects of the TMS group showed no difference in behaviour compared to a Sham stimulation group, therefore this study did not provide any further evidence of a specific role of the cerebellum in implicit motor adaptation. However, our behavioral findings replicate those in the seminal study by Mazzoni and Krakauer (2006).

Expansion of cultivated lands and field management impacts greenhouse gas (GHG) emissions from agriculture soils. Soils naturally cycle GHGs and can be sources or sinks depending on physical and chemical properties affected by cultivation and management status. We looked at how cultivation history influences GHG emissions from subtropical soils. We measured CO2, N2O, and CH4 fluxes, and soil properties from newly converted and continuously cultivated lands during the summer rainy season in calcareous soils from south Florida. Newly converted soils had more soil organic matter (OM), more moisture, higher porosity, and lower bulk density, leading to more GHG emissions compared to historically cultivated soils. Although more nutrients make newly converted lands more desirable for cultivation, conversion of new areas for agriculture was shown to release more GHGs than cultivated lands. Our data suggest that GHG emissions from agricultural soils may decrease over time with continued cultivation.

The use of natural pozzolans in concrete applications is gaining more attention because of the associated environmental, economic, and technical benefits. In this study, reference cemented mine backfill samples were prepared using Portland cement, and experimental samples were prepared by partially replacing Portland cement with 10 or 20 wt.% fly ash as a byproduct (artificial) pozzolan or pumice as a natural pozzolan. Samples were cured for 7, 14, and 28 days to investigate uniaxial compressive strength development. Backfill samples containing 10 wt.% pumice had almost a similar compressive strength as reference samples. There is strong potential for pumice to be used in cemented backfill to minimize costs, improve backfill properties, and promote the sustainability of the mining industry.

Flood and drought events cause significant freshwater inflow fluctuations in estuaries, potentially leading to physiological stress and altered abundances of pathogens such as Vibrio vulnificus and Perkinsus marinus in oysters. To assess the effects of freshwater pulses to oyster reefs in subtropical estuaries in Texas, this study accomplished two goals: 1) reconstructed a reef-specific history of freshwater pulses through shell stable isotope analysis, 2) quantified the abundance of V. vulnificus and P. marinus through culture-dependent and culture-independent microbiology analyses. Oysters from a low-relief and high-relief reef experienced similar fluctuations in shell isotopes, indicating similar ranges of past environmental conditions. V. vulnificus and P. marinus were detected throughout the study but the abundance of these microorganisms was not correlated with environmental parameters or one another. Importantly, the P. marinus infection intensity was always lower at the high-relief reef, which suggests that high-relief reefs may experience lower infection frequencies.

Quantifying and comparing stocks of oysters (Crassostrea virginica) within and among estuaries across the Gulf of Mexico is difficult because the sampling equipment used is either inconsistent among studies, or inefficient. In Texas, USA, stock assessments of oyster populations are made using an oyster dredge, which is an inefficient sampling tool. We compared sampling densities estimated by oyster dredges with more accurate estimates taken by diver-quadrat samples to determine a dredge efficiency rate. Our calculated efficiency rate (0.125) was negatively affected by the number of dead oysters, and the number and volume of total oysters in an area, but not affected by sediment grain size, water quality, and other oyster metrics. The dredge efficiency rate calculated in this study can be applied to past and future dredge-collected oyster quantity data to provide more realistic estimates of oyster densities and allow more accurate stock assessments and comparisons among studies and regions.

Green–Griffiths–Kerr introduced Hodge representations to classify the Hodge groups of polarized Hodge structures, and the corresponding Mumford–Tate subdomains. We summarize how, given a fixed period domain $ \mathcal{D} $ , to enumerate the Hodge representations and corresponding Mumford–Tate subdomains $ D \subset \mathcal{D} $ . The procedure is illustrated in two examples: (i) weight two Hodge structures with $ {p}_g={h}^{2,0}=2 $ ; and (ii) weight three CY-type Hodge structures.

A variety of machines are currently being used for mechanical excavation in mining and civil industries. A series of research works have been conducted at McGill University in the past decade to study the effects of microwave (MW) irradiation on rock mechanical properties. The idea is to enhance the excavation performance by improving the rate of penetration and decreasing the wear rate on the cutting tools. These two effects would eventually translate into economic benefits for mine operators. The effectiveness of MW on weakening rocks is proven, however the most efficient method to employ MW in mines is still under investigation. This article presents some experimental results on the effects of cooling- rate on rock strength. Brazilian Tensile Strength (BTS) of microwave treated samples were compared in natural air-cooled and water rapid-cooled conditions.

Restless legs syndrome (RLS) is a neurological disorder characterized by an urge to move and uncomfortable sensations. Genetic studies have identified polymorphisms in up to 19 risk loci, including MEIS1 and BTBD9. Rodents deficient in either homolog show RLS-like phenotypes. However, whether MEIS1 and BTBD9 interact in vivo is unclear. Here, with C. elegans, we observed that the hyperactive egg-laying behavior caused by loss of BTBD9 homolog was counteracted by knockdown of MEIS1 homolog. This was further investigated in mutant mice with Btbd9, Meis1, or both knocked out. The double knockout mice showed an earlier onset of the motor deficit in a wheel running test but did not have increased sensitivity to heat stimuli as observed in single knock outs. Meis1 protein level was not influenced by Btbd9 deficiency, and Btbd9 transcription was not affected by Meis1 haploinsufficiency. Our results demonstrate that MEIS1 and BTBD9 do not regulate each other.

During military operations, soldiers are required to successfully complete numerous physical and cognitive tasks concurrently. Understanding the typical variance in research tools that may be used to provide insight into the interrelationship between physical and cognitive performance is therefore highly important. This study assessed the inter-day variability of two military-specific cognitive assessments: a Military-Specific Auditory N-Back Task (MSANT) and a Shoot-/Don’t-Shoot Task (SDST) in 28 participants. Limits of agreement ±95% confidence intervals, standard error of the mean, and smallest detectable change were calculated to quantify the typical variance in task performance. All parameters within the MSANT and SDST demonstrated no mean difference for trial visit in either the seated or walking condition, with equivalency demonstrated for the majority of comparisons. Collectively, these data provided an indication of the typical variance in MSANT and SDST performance, while demonstrating that both assessments can be used during seated and walking conditions.

Understanding the effects of predicted rising sea levels, combined with changes in precipitation and freshwater inflow on key estuarine ecosystem engineers such as the eastern oyster would provide critical information to inform restoration design and predictive models. Using oyster ladders with shell bags placed at three heights to capture a range of inundation levels, oyster growth of naturally recruited spat was monitored over the course of 6 months. Oyster numbers and shell heights were consistently highest in bottom and mid bags experiencing greater than 50% inundation (mid: 63 ± 7%; bottom: 95 ± 3%). Identifying thresholds for optimal oyster growth and survival to enhance restoration engineering would require finer scale evaluation of inundation levels.

Growing in a saline environment causes changes in important physiological processes that are directly related to plant growth and development. In this study we evaluated the effect of salinity on transpiration of sorghum plants in semi-arid conditions and found that the highest rates of transpiration were observed in the hottest hours of the day, between 10 a.m. and 3 p.m., with plants subjected to the saline environment having their transpiration reduced by up to 70% when compared to the non-saline environment. This behavior can be reflected in reductions in plant growth and development due to reduced water absorption by the roots, consequently causing an imbalance of nutrients in the plant due to low absorption rate and competition between nutrients and salts in the preferred routes of absorption in the roots.

Scedosporium and Lomentospora species are ubiquitous saprophytic filamentous fungi that emerged as human pathogens with impressive multidrug-resistance profile. The ability to form biofilm over several biotic and abiotic surfaces is one of the characteristics that contributes to their resistance patterns against almost all currently available antifungals. Herein, we have demonstrated that Scedosporium apiospermum, Scedosporium minutisporum, Scedosporium aurantiacum and Lomentospora prolificans were able to form biofilm, in similar amounts, when conidial cells were incubated in a polystyrene substrate containing Sabouraud medium supplemented or not with different concentrations (2%, 5% and 10%) of glucose, fructose, sucrose and lactose. Likewise, the glucose supplementation of culture media primarily composed of amino acids (SCFM, synthetic cystic fibrosis medium) and salts (YNB, yeast nitrogen base) did not modulate the biofilm formation of Scedosporium/Lomentospora species. Collectively, the present data reinforce the ability of these opportunistic fungi to colonize and to build biofilm structures under different environmental conditions.

One of the commonly used analytical approaches for measuring oxygen isotope ratios δ18O of solids (organic and inorganic) is to pyrolyze the samples to gaseous phases and then send the gas into an isotope ratio mass spectrometer system. Solid samples for δ18O measurements are usually stored in silver cups because of its low reactivity towards oxygen and other oxidants. Samples in silver cups can be dropped directly into the carbon column of the pyrolysis furnace. However, the silver cups can tarnish and then be oxidized over a prolonged storage period. We find that while a small amount of silver oxides does not affect measurements with appreciable sample sizes, it can skew isotope results of small samples. We thus recommend careful storage of samples in silver cups to minimize oxidation, such as under an air-isolated condition, and avoiding prolonged storage for accurate δ18O measurements.
