Enzyme electrophoresis has been used to examine genetic diversity in a population of Proteocephalus exiguus La Rue, 1911 (Cestoda: Proteocephalidae), parasitizing salmonid and coregonid fishes. Among 16 loci tested, three polymorphic loci (Ada, Got, Pgm-2) were found. Six different genotypes at the Got locus distributed in Hardy-Weinberg equilibrium suggest remarkable genetic flexibility of P. exiguus. Balancing selection is proposed as the mechanism maintaining genetic variation within the species. Data of genetic variability parameters (Ho = 0.064; He = 0.07; P = 0.19) and outcrossing rate (t = 0.842) of P. exiguus population have been provided. Possible factors affecting these data are discussed.

A new species of polystomatid monogenean, Eupolystoma anterorchis sp. n. is described from the urinary bladder and kidneys of Bufo pardalis from the Cape Peninsula, Republic of South Africa. It is distinguished from other Eupolystoma species, and from all other species except one within the Polystomatidae, by the anterior pre-ovarian extension of a large proportion of the testis follicles. This and other morphological characteristics are incorporated into an amended generic diagnosis.

The morphology of E. alluaudi from Bufo regularis in West Africa is also described. The known range of this species is enlarged with new geographical records in the Cameroun and Ethiopia, and a new host record from Nectophrynoides malcolmi.

Material from each area shows that the eggs of Eupolystoma spp. complete development in utero and that all developmental stages of the parasite occur side by side within the same host individual. This represents a significant evolutionary departure from the pattern of transmission typical of Polystoma in most of the other anuran hosts.

Some parasitic nematodes can inhabit different definitive hosts, which raises the question of the intraspecific variability of the nematode genotype affecting their preferences to choose particular species as hosts. Additionally, the issue of a possible intraspecific DNA microheterogeneity in specimens from different parts of the world seems to be interesting, especially from the evolutionary point of view. The problem was analysed in three related species – Toxocara canis, Toxocara cati and Toxascaris leonina – specimens originating from Central Europe (Poland). Using specific primers for species identification, internal transcribed spacer (ITS)-1 and ITS-2 regions were amplified and then sequenced. The sequences obtained were compared with sequences previously described for specimens originating from other geographical locations. No differences in nucleotide sequences were established in T. canis isolated from two different hosts (dogs and foxes). A comparison of ITS sequences of T. canis from Poland with sequences deposited in GenBank showed that the scope of intraspecific variability of the species did not exceed 0.4%, while in T. cati the differences did not exceed 2%. Significant differences were found in T. leonina, where ITS-1 differed by 3% and ITS-2 by as much as 7.4% in specimens collected from foxes in Poland and dogs in Australia. Such scope of differences in the nucleotide sequence seems to exceed the intraspecific variation of the species.

The possible environmental effects of the massive use of Duddingtonia flagrans for controlling sheep nematodes were evaluated in two regions. Non-supplemented faeces and faeces from sheep supplemented with D. flagrans were deposited three times on pasture plots and samples were collected 7 and 14 days post-deposition. Samples were cultured in agar-water (2%) with Panagrellus spp. to recover D. flagrans and other nematophagous fungi, and soil nematodes were extracted using Baermann funnels and counted. No significant differences in the populations of soil nematodes and fungi colonizing sheep faeces (P > 0.05) were observed between supplemented and non-supplemented groups, except in one sample. The topsoil in contact with the faeces was sampled 1–4 months post-deposition, revealing that, with one exception, D. flagrans did not persist in soil beyond 2 months post-deposition. Duddingtonia flagrans does not affect faecal colonization by other fungi and soil nematodes and, once deployed on pasture, does not survive for long periods in the environment.

The bivalve Perumytilus purpuratus is a common species that is widely distributed throughout rocky intertidal zones in Chile. This bivalve is the first intermediate host for three trematode species: one bucephalid (an undetermined species) and two fellodistomids (Proctoeces lintoni and one undetermined species). A few studies based on morphological comparisons, experimental infection and molecular analyses have been performed to ascertain the taxon (at least at the family level) to which these trematodes belong; yet, there remains no clarification about the specific identity of these trematodes. Therefore, in this study, we compared the V4 region nucleotide sequences of the 18S rRNA of these three sporocyst species, classified as morphotypes, found in P. purpuratus and nine adult trematode species from intertidal fishes that are likely definitive hosts for these parasites. The sequences from two of the sporocyst morphotypes matched with adult trematodes from the intertidal fish: type 1 sporocyst was similar to Prosorhynchoides carvajali (Bucephalidae), with a mean genetic divergence of 0.78%, and type 2 sporocyst was similar to Proctoeces sp. (but not P. lintoni), with 0% genetic divergence. The third species (type 3 sporocyst) was classified to the family Fellodistomidae; however, the sequence from this species differed greatly from the three other fellodistomid species documented in the marine fish of Chile and from other fellodistomids in public databases. Moreover, this morphotype has a particular cercarial morphology that greatly differs from other fellodistomid species described thus far. Therefore, this intriguing trematode remains a mystery.

By means of egg counts, faecal cultures and post-mortem examinations of sheep the course of an infestation of Ostertagia circumcincta was followed from March until the following January in a flock of 100 twelve-months-old wethers at Wellington, N.S.W. The highest adult infestations and egg counts occurred in March and January, while during the winter months there was a continual accumulation of inhibited fourth stage larvae in the abomasal mucosa. The data suggest that the adult population observed in January arose from the inhibited population observed during the previous winter and that one of the factors controlling the development of the inhibited population was larval intake.

Atlantic salmon, Salmo salar, is normally rather refractive to infection with the ectoparasitic monogenean Gyrodactylus derjavini but dexamethasone treatment of the host increases the susceptibility. The causative mechanisms were elucidated in this work. Groups of Atlantic salmon were treated by intra-peritoneal dexamethasone injections and subsequently infected with G. derjavini. It was shown that both the infection level and the mucous cell density of caudal and pelvic fins were affected by the treatment. Significantly higher mucous cell densities were found on infected and treated fish whereas non-infected and treated fish showed no significant elevation of cell density. This suggests that mucous cell discharge elicited by infection is inhibited by the drug. The association with elevated parasite counts in these fish can be explained either by decreased anti-parasitic mucus action or by parasite predilection for intact mucous cells.

Seasonal density fluctuations of Bulinus nasutus populations with accompanying Schistosoma haematobium infection rates in relation to rainfall and habitat water volumes were studied at Ukiriguru, Mwanza, Tanzania. Alongside the snail investigations, 50 school children initially negative for urinary schistosomiasis were examined regularly to determine seasonal incidence rates for the infection. Of the 17 646 B. nasutus collected in 2 years, 156 (0.88%) were found shedding cercariae. Snail populations fluctuated seasonally as influenced by rainfall through its effects on habitat water volume. Monthly snail infection rates ranged from 0.09% to 3.19% and were highest in February and March, at the time of the short dry period. Monthly incidence of S. haematobium in school children ranged between 2.6% and 12.5%, being highest in April and May. There was a significant linear association between monthly snail infection rates and S. haematobium incidence rates in school children ( in 1983/84) suggesting that the maximum transmission period for urinary schistosomiasis in the area occurs during the short dry period, sometime in February/March so that most of the infections in the community would be detected in April/May.

Twenty-nine Japanese wild boars (Sus scrofa leucomystax), collected during the hunting seasons of 2005 and 2006 in the western part of the mainland of Japan (Honshu), were examined for their visceral helminths. Eighteen helminth parasites were prevalent in them, including 17 Nematoda species (Metastrongylus elongatus, Metastrongylus salmi, Metastrongylus asymmetricus, Metastrongylus pudendotechus, Stephanurus dentatus, Gnathostoma doloresi, Physocephalus sexulata, Ascarops strongylina, Capillaria suis, Ascaris suum, Globocephalus samoensis, Globocephalus longimucronatus, Strongyloides ransomi, Trichuris suis, Bourgelatia diducta, Oesophagostomum dentatum, and Oesophagostomum watanabei), and one Cestoda species (Pseudanoplocephala nipponensis). Muscle digestion of the diaphragm (using an artificial gastric juice) of 24 wild boars detected a single diplostomulum of probably Pharyngostomum cordatum. In addition, four female and six male adults of Morgascaridia kugii sp. n. (Nematoda: Schneidernematidae) were recovered from the large intestine of a single wild boar. Examination of an additional 20 samples of the stomach and 27 samples of the large intestine could not find this species. To date, recorded species of the genus Morgascaridia are limited to M. sellsi collected from wild boars in Uganda and Congo several decades ago, and thence, no records of the recovery are available. Morgascaridia kugii sp. n. differed from M. sellsi by smaller body dimensions, shorter distance between the precloacal sucker and the cloaca, smaller sizes of the copulatory spicules and the gubernaculum, and smaller sizes of uterine eggs.

East African Fasciola gigantica apparently does not reach sexual maturity in mice, rats, hamsters, guinea pigs and rabbits. The order of recovery of flukes from the highest to the least was as follows : hamsters, guinea pigs, mice and rabbits. Rats were completely refractory to infection. The more highly susceptible animals also had a higher recovery rate of flukes. Mortality in animals appeared to be related to infection.

The effect of fatty acids was studied on the developmental direction of Strongyloides ratti first-stage larvae (L1). The proportion of third-stage infective larvae increased markedly when L1 were cultured in faeces with added fatty acids such as palmitic (C16), stearic (C18), oleic (C18:1) and linoleic (C18:2) acids. Unsaturated fatty acids were more effective than saturated ones. Moreover, the proportion of infective larvae increased with quantity of linoleic acid but the triacylglycerols of any fatty acid had no effect. These results suggest that these free fatty acids cause physiological changes that determine the developmental course of L1 of S. ratti in nature.

2. The first intermediate host of the parasite in this area is shown to be the freshwater snail Lymnaea peregra.

3. Adult flukes were obtained experimentally in mice, ducklings and herring-gull chicks.

4. Significant differences are shown to occur between adult flukes recovered from different host species.

5. The implications of these findings in relation to the host specificity and the classification of the Strigeida are briefly discussed.

The host specificity and distribution of Eubothrium crassum (Bloch, 1779) and Eubothrium salvelini (Schrank, 1790), morphologically fairly similar pseudophyllidean tapeworms parasitizing salmonid fish, were critically assessed on the basis of morphological and genetic evaluation of extensive material collected from different definitive hosts and geographical regions in Europe. Eubothrium crassum occurs in fish of the genera Salmo, i.e. salmon (S. salar – both freshwater and marine), sea trout (S. trutta trutta), brown trout (S. trutta fario), and lake trout (S. trutta lacustris), and also in Danubian salmon (Hucho hucho) and vendace (Coregonus albula). Eubothrium salvelini parasitizes Arctic char (Salvelinus alpinus) and brook trout (Salvelinus fontinalis) in Europe, and also whitefish (Coregonus wartmanni). Rainbow trout (Oncorhynchus mykiss), which is not a native European fish species, was found to be a suitable definitive host for both Eubothrium species, which may occur simultaneously in the same fish. Previous records of E. crassum in Arctic char and brook trout, and those of E. salvelini in fish of the genus Salmo were most probably misidentifications. Most studies of Eubothrium have involved salmonids from the northern part of Europe, with few records from southern and south-eastern Europe. This study also confirmed the reliability of the morphology of the apical disc for the discrimination of E. crassum and E. salvelini.

Three new cestode species are described from the crocodile shark (Pseudocarcharias kamoharai) in Ecuador. All three were examined with light and scanning electron microscopy. The unique combination of morphological features in one of the new species prompted formal investigation of the non-monophyly of Paraorygmatobothrium relative to the morphologically similar genera Doliobothrium, Guidus, Marsupiobothrium, Nandocestus, Orectolobicestus, Ruhnkecestus and Scyphophyllidium. Sequence data generated for part of the 28S rDNA gene were subjected to maximum likelihood (ML) analysis. The resulting tree led to the synonymization of six of these seven genera with Scyphophyllidium, and transfer of their species to the latter genus. With the new species, the number of described members of Scyphophyllidium is now 45. The diagnosis of Scyphophyllidium is revised to accommodate these species. In addition, to expedite future descriptions, eight categories of Scyphophyllidium species are circumscribed, based largely on bothridial features. Scyphophyllidium timvickiorum n. sp. is a category 1 species. Beyond being the smallest category 1 species, it bears, rather than lacks, apical suckers and lacks, rather than bears, strobilar scutes. The two other new species are members of Clistobothrium. Clistobothrium amyae n. sp. differs from its congeners in bothridial shape, elongate cephalic peduncle and tiny size. Clistobothrium gabywalterorum n. sp. differs from the two of its congeners that also possess foliose bothridia in overall size and testis number. Despite their substantial morphological differences, the ML tree indicates they are sister taxa. Both are unique among their congeners in possessing cephalic peduncle spinitriches. The diagnosis of Clistobothrium is revised accordingly.

Despite the fact that tapeworms comprise the bulk of parasite communities of sharks in marine ecosystems, little is known about their life cycles and, more specifically, about the potential intermediate hosts they utilize as transmission routes. In the absence of morphological features required for specific identification of larval tapeworms from potential intermediate hosts, recent molecular advances have contributed to linking larval and adult parasites and, in some instances, uncovering unknown trophic links. Host–parasite checklists are often the first source of information consulted to assess the diversity and host specificity of parasites, and provide insights into parasite identification. However, these host–parasite checklists are only useful if they encompass the full spectrum of associations between hosts and parasites. A checklist of New Zealand fishes and their parasites has been published, but recent parasitological examinations of commercial fish species reveal that the checklist appears to be far from complete. We focused our current study on a comprehensive survey of macroparasites of a commercial species, the New Zealand sole (Peltorhamphus novaezeelandiae) off the coast of Otago, New Zealand. Specifically, we were expecting to recover marine tapeworms using sharks as their definitive hosts that are generally underreported in parasite surveys. The parasites recovered included tapeworms, flukes, round worms and thorny-headed worms. Surprisingly, a large proportion of the non-tapeworm parasites we recovered were not previously reported from this fish species. A discussion on the potential ecological roles played by this fish species in the transmission of parasites is included.

The main problem regarding the chemotherapy of filariasis is that no safe and effective drug is available yet to combat the adult human filarial worms. Setaria cervi, the causal organism of setariasis and lumbar paralysis in cattle, is routinely employed as a model organism for conducting biochemical and enzymatic studies on filarial parasites. In view of the practical difficulties in procuring human strains of Wuchereria bancrofti and Brugia malayi for drug screening, the bovine filarial parasite S. cervi, resembling the human species in having microfilarial periodicity and chemotherapeutic response to known antifilarial agents, is widely used as a model in such studies. For a rational approach to antifilarial chemotherapy, knowledge of the biochemical composition and metabolic pathways of this helminth parasite may be of paramount importance, so that more potent antifilarial agents based on specific drug targets can be identified in drug discovery programmes. The present review provides an update on the biochemistry of the important metabolic pathways functioning within this potentially important bovine parasite, that have so far been studied, and on those that need to be investigated further so as to identify novel drug targets that can be exploited for designing new antifilarial drugs.

1. Examination of 289 fowl showed that 69·2% were infected with intestinal helminths. The following 6 species were identified: Ascaridia galli (33·6%), Capillaria anatis (10·4%), C. caudinflata (15·9%), C. obsignata (45·3%), Davainea proglottina (0·7%) and Heterakis gallinarum (60·9%).

2. No parasites were found in the upper intestines of 151 fowl.

3. The results are compared with those of previous surveys and are discussed in relation to the age and management of the birds.

When the beef measle bladder worm is ingested by man it is believed that the head attaches itself to the mucosa of the jejunum, the caudal bladder disintegrates and segments develop in the neck region immediately behind the head at the rate of about 6–12 per day. Thus, segments immediately behind the head one day are pushed away from the proximity of the head by the formation of new segments each day. While a segment is being pushed further from the head it increases in size and alters its shape; its sex organs, both male and female, develop and perform their functions. About 80 days are required for a segment to become ripe or for a bladder worm to develop into a complete tapeworm. The ripe posterior segments are detached almost daily. These are replaced by the formation of a similar number immediately behind the head. In this way a worm once fully grown maintains approximately the same length.

Groups of ferrets inoculated with Dracunculus insignis were treated with various anthelminthic compounds to evaluate the potential use of drugs in controlling the human parasite D. medinensis. The three primary compounds tested were diethylcarbamazine (DEC), albendazole (ALBZ), and ivermectin (IVER); they were administered in dosages of 60 mg/kg, 10 mg/kg, and 1 mg/kg, respectively. Groups of animals received treatment either once at 60 days after inoculation, once at 60 and 90 days, or for 3 days at either 60 or 90 days postinoculation. The most marked decrease occurred in the animals that received treatment once at 60 and at 90 days after inoculation. This was observed for all three drugs tested. Increased dosages, i.e., 3 days of treatment at 60 or 90 days did not result in decreased worm burdens. In no group was there a statistically significant reduction in worm burden when compared with controls. Two other compounds, metrifonate and CGP 6140, were tested in a more limited manner, but again the worm recovery rates were comparable with those in control groups. It would appear that existing drugs commonly used to treat helminthic infections are poor candidates for use in the campaign to eradicate guinea worm disease.

A total of 358 cattle was examined for schistosome infection in Zambian slaughterhouses. A total of 542 worms collected from 104 infected individuals was examined for glucose-6-phosphate dehydrogenase and phosphoglucomutase using isoelectric focusing. The overall prevalence of infection was 51%. Ninety three percent of the infected animals had less than 100 worm pairs in the mesenteric veins. Schistosoma mattheei was the predominant species (75%); S. leiperi (12%) and S. margrebowiei (2%) were also identified. The remaining 11% of the worms showed one of two distinct heterozygote patterns. Pattern A is identical to that of a laboratory-produced Fl S. mattheei × S. haematobium hybrid, but could also represent a S. mattheei × S. leiperi hybrid. Further studies are required to elucidate the origins of pattern B.
