Eosinophil infiltrations were observed in the intestine and the muscle of both Trichinella spiralis-infected (WBxC57BL/6)F1-W/Wv mice and their littermates, WBB6F1−+/+, +/W, +/Wv, almost to the same extent. W/Wv mice did not show infiltration of subepithelial mast cells and globule leucocytes in response to T. spiralis infection. Increased numbers of these cells were observed in their littermates. Worms in W/Wv mice were retained for longer periods than those in littermates. Also, no difference was noted in the production of specific serum antibodies between W/Wv mice and their littermates, as determined by passive cutaneous anaphylaxis (PCA) for specific IgE and by indirect haemagglutination (IHA). These results suggest a possible participation of SMC, GL and eosinophils in the expulsion of adult T. spiralis.

The study of populations of animals has received the attention of a large number of workers. Of these, however, but a few have been interested in the problem in its application to parasitic nematodes and in this field there is consequently a limited literature. Particularly is this so when the parasitic nematodes of sheep are considered, the incentive for study of which, in most cases, has been the pressing need for their destruction. This desire has been fulfilled to a greater or lesser degree, but at the expense of researches which would have contributed to a more fundamental knowledge of the ecological relationships of parasitic worms in general.

In recent decades, parasite community ecology has produced hundreds of studies on an ever-growing number of host species, and developed into an active sub-discipline of parasitology. However, this growth has been characterized by a lack of standards in the practices used by researchers, with many common approaches being flawed, unjustified or misleading. Here, in the hope of promoting advances in the study of parasite community ecology, I identify some of the most common errors or weaknesses in past studies, and propose ten simple rules for best practice in the field. They cover issues including, among others, taxonomic resolution, proper and justifiable analytical methods, higher-level replication, controlling for sampling effort or species richness, accounting for spatial distances, using experimental approaches, and placing raw data in the public domain. While knowledge of parasite communities has expanded in breadth, with more and more host species being studied, true progress has been very limited with respect to our understanding of fundamental general processes shaping these communities. It is hoped that the guidelines presented here can direct researchers away from the entrenched use of certain approaches flawed in design, analysis or interpretation, by offering a more rigorous and standardized set of practices, and, hopefully, a way forward.

Outbred LACA mice and inbred NIH mice were administered low (100 ova), medium (1000 ova), high (3000 ova) and trickle (4×250 ova) doses of Toxocara canis ova and the effect of infection on activity was examined with respect to: (i) the dose of ova administered and (ii) the number of larvae recovered from the brain. Larval recovery from the brain was significantly reduced in NIH mice compared to LACA mice for the 1000, 3000 and trickle doses. Mice from each strain were divided into larval intensity groupings based upon the number of larvae recovered from their brain. Activity for each mouse was measured pre- and post-infection by observing its behaviour in the home cage. Activity was assessed by monitoring six different independent categories of murine behaviour – ambulation, grooming, rearing, digging, climbing and immobility. Within each behavioural category, the duration of time spent at each behaviour per mouse within one thousandth of a second, the number of short bouts performed and the number of long bouts of behaviour performed were recorded over a 20 min period. Activity of LACA and NIH mice differed prior to infection. LACA mice spent more time immobile compared to NIH mice, which ambulated and climbed more. Variations in activity were also observed between groups of mice prior to infection. The effect of infection differed by strain, by dose and by larval intensity. Post-infection LACA mice became more immobile and ambulated less. NIH mice showed reduced immobility, but while ambulation decreased digging and climbing increased post-infection. Short bouts of activity remained unchanged among LACA mice post-infection but showed an increase for some behaviours in NIH mice.

To estimate prevalence of amphistomiasis and its association with bovine Fasciola gigantica infections in Zambia, 709 faeces of cattle presented for slaughter from three major cattle rearing areas of Central, Southern and Western provinces were analysed. The prevalence rate of amphistomes was 51.6%. Egg counts per gram (EPG) of faeces ranged from 0 to 385 with a mean (± SEM) of 11.96±1.07. The origin of the cattle had a significant influence (P<0.001) on the prevalence rate. Fasciola gigantica infections accounted for 46.7% of the cattle examined. The mean EPG count (± SEM) was 6.3±0.66 with a range of 0 to 223. A total of 34.6% were mixed infections while single Fasciola and amphistome infections represented 12.1% and 17.1%, respectively. Significantly more cattle (63.8%) were infected with either single or both trematode infections (P<0.0001) than not. Mixed trematode infections were highest in the Southern province (80.0%) while the lowest were recorded in Central province (16.3%). A positive correlation (r2=0.0428) was present in mixed infections. In order to minimize losses, epidemiology and cross-resistance of amphistomiasis and fascioliasis should be studied in different ecological regions of Zambia to formulate efficient control programmes.

The prevalence of toxocariasis was evaluated for 445 dogs, 105 cats and 92 foxes from the Poznan region during 1997–1998. Forty one cats were infected (39%), 140 dogs (32%) and 15 red foxes (16%). Toxocara canis was found most frequently in puppies up to 3 months old (58%) and T. cati in kittens 4–6 months old (64%). Toxocariasis was much more prevalent amongst adult foxes (14%) than adult dogs (3%). In contrast to cats, female dogs and foxes were less infected than males. The present study suggests that cats may constitute an underestimated risk of transmission of Toxocara spp. to humans and the progressive synatropization of red foxes may also increase the sources of environmental contamination with Toxocara eggs.

The paper summarises existing information on the role of animals in the transmission of filarial infections to man.

Wuclicreria bancrofti has not been found in animals and the parasite has never been transmitted to animals in the laboratory. Although bancrofti-typc microfilariae have been described from a potto (Perodicticus potto) in the Congo, there is no evidence that bancroftian filariasis is a zoonosis.

Wuchereria bancrofti var. vauceli occurs in man in Madagascar; the adults have not yet been found; the life cycle is unknown. For the time being it is best to regard this as a species inquirenda it is probably not a Wuchereria; it may occur in lemurs.

The sub-periodic form of Brugia malayi has been found in man and a variety of animals in Malaya. In the swamp forest areas there is probably a free interchange of parasites between man and monkeys; if this is confirmed then this is a definite zoonosis.

The purpose of this paper is to bring together all the information available on the helminth parasites known to occur in domesticated animals in Kenya. Several reports have been made by various workers over the past fifty years but these are scattered throughout the literature. Of the principal authors, Daubney (1926, 1933) has recorded many of the Nematodes, and Hudson (1934) published a list of the Cestodes known to occur in mammals, birds and reptiles in East Africa. Dinnik (1951, 1952, 1954, 1959, 1961a, 1961b, 1962) and Dinnik and Dinnik (1954–55, 1954, 1960) have recorded the Trematode species found in cattle, sheep and goats in East Africa.

Big variations have been reported in the extent of occurrence of the so-called “histotropic or tissue phase” (“dormant larvae”) especially in intestinal nematodes. In several species these variations were found by some authors, not by others, and also not always by the same author. From this fact, and from the well known retardation of growth of larvae during their stay in the tissue, it was concluded that it does not represent a phase in the sense of a behavioural pattern innate to the parasite, but rather a phenomenon dependent upon the degree of resistance in the host.

Three Jersey bull calves were immunized by subcutaneous injections of sonicated Onchocerca lienalis microfilariae suspended in phosphate buffered saline, and three control animals were injected with medium only. All calves received booster injections 27 days later, and were challenged with live microfilariae 44 days after the booster. Following challenge, only the immunized animals developed an elevated level of circulating eosinophils. When necropsied ten days after challenge there was a 97% reduction in recoveries of microfilariae from immunized animals compared to challenge controls. In human onchocerciasis it is the microfilariae which are the principal cause of pathology, and we believe that studies on O. lienalis in both the natural bovine host and in inbred rodents provide a promising model to investigate immunity to Onchocerca microfilariae.

The current study considers the distribution of a small sample of 138 Bulinus snails, across 28 localities within eight Nigerian states. Snails were identified using a combination of molecular methods involving both DNA sequencing of a partial cytochrome oxidase subunit 1 (cox1) fragment and restriction profiles obtained from ribosomal internal transcribed spacer (its) amplicons. The results showed that the majority of Bulinus samples tested belonged to the species Bulinus truncatus while only two were Bulinus globosus. The use of RsaI restriction endonuclease to cleave the ribosomal its of Bulinus, as a method of species identification, was adopted for the majority of samples, this being a quicker and cheaper method better suited to small laboratory environments. Polymerase chain reaction (PCR) amplification of the schistosome Dra1 repeat within each of the collected Bulinus samples was employed to determine the extent and distribution of infected snails within the sample areas. Successful amplification of the Dra1 repeat demonstrated that 29.7% of snails were infected with schistosomes. Sequencing of the partial schistosome its from a small subset of snail samples suggested that some snails were either penetrated by both Schistosoma haematobium and Schistosoma bovis miracidia or hybrid miracidia formed from the two species.

The chronology of the life cycle of Trichostrongylus retortaeformis (Zeder, 1800) (Nematoda, Trichostrongyloidea) is studied in its natural host Oryctolagus cuniculus. The free living period lasted 5 days at 24°C. Worm-free rabbits were each infected per os with T. retortaeformis larvae. Rabbits were killed at 12 h post-infection (p.i.) and every day from one day to 13 days p.i. By 12 h p.i., all the larvae were exsheathed and in the small intestine. The third moult occurred between 3 and 5 days p.i. and the last moult between 4 and 7 days p.i. The prepatent period lasted 12 to 13 days. The patent period lasted five and a half months. The four known life cycles of species of Trichostrongylus in ruminants were compared with that of T. retortaeformis. No significant difference was found except in the duration of the prepatent period. These similarities in the life cycles confirm the previously formulated hypotheses on the relationship between the parasites of the two host groups ().

We report on the intestinal helminth fauna of 56 South American sea lions, Otaria flavescens, and 5 South American fur seals, Arctocephalus australis, from northern Patagonia, Argentina. A total of 97,325 helminth specimens were collected from sea lions. Gravid individuals were represented by 6 species of parasites: 1 digenean (Ascocotyle (Ascocotyle) patagoniensis), 1 cestode (Diphyllobothrium spp.), 3 nematodes (Uncinaria hamiltoni, Contracaecum ogmorhini s.s., Pseudoterranova cattani) and 1 acanthocephalan (Corynosoma australe). In addition, third-stage larvae of 2 nematodes (Contracaecum sp. and Anisakis sp. type I) and 3 juvenile acanthocephalans (Andracantha sp., Profilicollis chasmagnathi and Corynosoma cetaceum) were also collected. Andracantha sp., C. ogmorhini s.s. and P.chasmagnathi represent new host records. A total of 1516 helminth specimens were collected from fur seals. Gravid individuals were represented by three species of parasites, namely, Diphyllobothrium spp., C. ogmorhini s.s. and C. australe. In addition, larvae of Contracaecum sp. and P. cattani, juveniles of C. cetaceum and immature cestodes (Tetrabothriidae gen. sp.) were also collected. Corynosoma australe was the most prevalent and abundant parasite in both hosts, accounting for >90% of all specimens. Sea lions and furs seals from northern Patagonia harbour the intestinal helminth communities that could be predicted for otariids, i.e. the combination of species of the genera Corynosoma, Diphyllobothrium, Pseudoterranova, Contracaecum and, in pups, Uncinaria. Additionally, both species of otariid are apparently unsuitable hosts (i.e. non-hosts) for as many as five parasite taxa. The inclusion or exclusion of these species affects estimation of species richness at both component community (11 versus 6 species in sea lions; 7 versus 3 species in fur seals) and infracommunity (mean: 3.1 versus 2.6 in sea lions; 2.2 versus 1.7 species) levels. Information about the reproductive status of helminth species is often lacking in parasitological surveys on otariids and other marine vertebrates, but it is of significance to improve precision in parascript studies or ecological meta-analyses.

Twelve enzymes (encoded by 14 loci) in liver flukes of Fasciola species originating from Japan (parthenogenetic triploids), Korea (parthenogenetic diploids), the United States of America (USA) and Australia (all sexual diploids) were analysed using starch gel electrophoresis. Variation in electrophoretic patterns between samples was detected at five enzyme loci (Ak, Got, Gpi, 6-Pgd and Pgm-2). Japanese worms (31, of which six were established as uniparental laboratory strains), which reproduce by parthenogenesis, exhibited three different isozyme patterns. This indicates that triploidy has arisen more than once in Japanese flukes. Japanese Fasciola sp. can be separated into three types on morphological grounds. For the six laboratory strains of Japanese worms, the parental morphological type was known. Each of the three isozyme patterns observed was restricted to one morphological type. Most alleles detected in the Japanese triploids were also found in diploid worms from the other countries: the only alleles not represented elsewhere were four at the Got locus and two at the Pgm locus. Flukes from a laboratory strain derived from a single Korean diploid worm resembled the Japanese worms in genotype more closely than did American (seven uniparental laboratory strains) or Australian (30 worms) specimens. Worms from the last two countries were closely related.

Three groups of golden Syrian hamsters received intraperitoneally either, (1) 1 × 107 spleen cells, (2) 0.5 ml. serum or, (3) 1 × 107 spleen cells plus 0.5 ml. serum, from donors infected 6 weeks previously with 25 Opisthorchis viverrini metacercariae, one day before and at the same time as challenge with 25 metacercariae. Three groups of control animals received the same quantities of spleen cells, serum or an admixture of both from normal non-infected donors in the same manner followed by challenge with 25 metacercariae.

Animals were killed six weeks after challenge, livers and extrahepatic biliary systems carefully removed, and liver worm burdens estimated. Additionally, parasitic egg counts were performed on pooled faeces samples collected one hour prior to death.

Animals receiving spleen cells, serum, or an admixture of both, from normal non-infected donors had mean worm burdens of 11.44, 12.00 and 12.66, respectively. Animals receiving spleen cells, serum, or both, from infected donors had mean worm burdens of 9.88, 7.77 and 12.00, respectively. There were no significant differences in mean worm burdens between control and experimental groups. However, a substanial reduction in parasitic faecal egg counts and subsequently mean egg production per worm was observed in all 3 groups of animals receiving spleen cells, serum, or both, from infected donors when compared to their respective control group.

These findings are discussed in relation to adoptive transfer studies performed with other helminths and their possible relevance to naturally acquired immunity is commented on.

The pathology of the infection in the trout of Poulaphouca Reservoir is described.

An annual mortality was found to occur in the trout during the summer months. This was caused by peritonitis which is attributed to plerocercoids becoming active as a result of the increase in temperature.

It is demonstrated that the infection in trout is probably of a secondary nature resulting from their feeding on sticklebacks primarily infected from the Copepod hosts.

Among the Brazilian marsupials, the species of the genus Didelphis are the most parasitized by helminths. This study aimed to describe the species composition and to analyse the helminth communities of the Atlantic Forest common opossum Didelphis aurita at infracommunity and component community levels using the Elements of Metacommunity Structure Analysis, considering peri-urban, sylvatic and rural environments in the state of Rio de Janeiro, Brazil. Seventy-three specimens of D. aurita were captured during the study. Fourteen species of helminths were collected: nine of the phylum Nematoda (Trichuris minuta, Trichuris didelphis, Globocephalus marsupialis, Heterostrongylus heterostrongylus, Travassostrongylus orloffi, Viannaia hamata, Aspidodera raillieti, Cruzia tentaculata and Turgida turgida); four of the phylum Platyhelminthes (three species of Trematoda, Duboisiella proloba, Brachylaima advena and Rhopalias coronatus, and one species of the class Cestoda); and one species of the phylum Acanthocephala (Oligacanthorhynchus microcephalus). More than 95% of the animals were infected with parasites. The most abundant parasite species were V. hamata, T. turgida, C. tentaculata and T. orloffi. The nematodes T. turgida, C. tentaculata and A. raillieti were the most dominant species in the three environments. The analysis of the metacommunity structure indicated a structured pattern as a function of the environmental gradient, but only when all localities were considered together. At the infracommunity and component community levels, quasi-nested and nested structures with stochastic species loss were observed, respectively. The results indicate that the settlement of helminth species in the opossums can be attributed to the heterogeneity among individual hosts in relation to their exposure to parasites and to extrinsic factors, which vary among habitats, considering a regional scale. On a local scale, the characteristics of this host species may promote homogenization among individual hosts in terms of their exposure to the helminths. This study constitutes a new host record for the species G. marsupialis and T. didelphis and represents a pioneering investigation of the helminth metacommunity of a Neotropical marsupial.

The eel population in Neusiedler See has been maintained by regular massive stocking since 1958. After the establishment of the National Park Neusiedler See-Seewinkel in 1993, eel stocking was prohibited and the population, together with the specific parasites of eels, was predicted to decline to extinction within 10 years. This investigation was undertaken to document the decline and extinction of the Anguillicola crassus population in eels. From 1994 to 2001, 720 eels were collected from two sites in the lake. Prevalence and abundance of A. crassus were lower in spring than in summer and autumn and larger eels harboured more parasites than smaller ones. Neither year of study nor sampling site were correlated with parasite infection levels. No significant trend in the population parameters of A. crassus was detected over the 8 years of the survey. This suggested that there had been no significant decline in the eel population. This suggestion was confirmed by investigations of the fishery, which also found evidence of regular illegal stocking. The stability of the A. crassus population over the past decade seems to reflect the lack of change in eel population density. No mass mortalities of eels occurred over the period despite the many similarities between Neusiedler See and Lake Balaton in Hungary. Differences in eel size, eel diet and the lack of large-scale insecticide use are discussed as possible explanations for the absence of eel mass mortalities in Neusiedler See.

In this paper we report an investigation of the utility of coprological analysis as an alternative technique to study parasite specificity whenever host sampling is problematic; acanthocephalans from marine mammals were used as a model. A total of 252 scats from the South American sea lion, Otaria flavescens, and rectal faeces from 43 franciscanas, Pontoporia blainvillei, from Buenos Aires Province, were examined for acanthocephalans. Specimens of two species, i.e. Corynosoma australe and C. cetaceum, were collected from both host species. In sea lions, 78 out of 145 (37.9%) females of C. australe were gravid and the sex ratio was strongly female-biased. However, none of the 168 females of C. cetaceum collected was gravid and the sex ratio was not female-biased. Conversely, in franciscanas, 14 out of 17 (82.4%) females of C. cetaceum were gravid, but none of 139 females of C. australe was, and the sex ratio of C. cetaceum, but not that of C. australe, was female-biased. In putative non-hosts, the size of worms was similar to that from specimens collected from prey. Results suggest that both acanthocephalans contact sea lions and franciscanas regularly. However, C. australe and C. cetaceum cannot apparently reproduce, nor even grow, in franciscanas and sea lions, respectively. Coprological analysis may represent a useful supplementary method to investigate parasite specificity, particularly when host carcasses are difficult to obtain.

Trichuris trichiura and Trichuris suis parasitize (at the adult stage) the caeca of humans and pigs, respectively, causing trichuriasis. Despite these parasites being of human and animal health significance, causing considerable socio-economic losses globally, little is known of the molecular characteristics of T. trichiura and T. suis from China. In the present study, the entire first and second internal transcribed spacer (ITS-1 and ITS-2) regions of nuclear ribosomal DNA (rDNA) of T. trichiura and T. suis from China were amplified by polymerase chain reaction (PCR), the representative amplicons were cloned and sequenced, and sequence variation in the ITS rDNA was examined. The ITS rDNA sequences for the T. trichiura and T. suis samples were 1222–1267 bp and 1339–1353 bp in length, respectively. Sequence analysis revealed that the ITS-1, 5.8S and ITS-2 rDNAs of both whipworms were 600–627 bp and 655–661 bp, 154 bp, and 468–486 bp and 530–538 bp in size, respectively. Sequence variation in ITS rDNA within and among T. trichiura and T. suis was examined. Excluding nucleotide variations in the simple sequence repeats, the intra-species sequence variation in the ITS-1 was 0.2–1.7% within T. trichiura, and 0–1.5% within T. suis. For ITS-2 rDNA, the intra-species sequence variation was 0–1.3% within T. trichiura and 0.2–1.7% within T. suis. The inter-species sequence differences between the two whipworms were 60.7–65.3% for ITS-1 and 59.3–61.5% for ITS-2. These results demonstrated that the ITS rDNA sequences provide additional genetic markers for the characterization and differentiation of the two whipworms. These data should be useful for studying the epidemiology and population genetics of T. trichiura and T. suis, as well as for the diagnosis of trichuriasis in humans and pigs.
