The protein A encoding gene spa of four Staphylococcus aureus strains isolated from bovine clinical mastitis was amplified by PCR and sequenced. The four strains were selected after an initial screening of spa gene of 41 strains isolated from mastitic cows and were subjected to detailed investigations. According to the sequencing results the spa gene of three strains (M1, M2, M3) appeared with gene segments encoding five (E, D, A, B and C) and four (E, A, B and C) IgG binding domains for two (M1, M3) and one (M2) strain, respectively and with gene segments encoding four, two and two repeats of the octapeptide Xr-repeats for the strains M1, M2 and M3, respectively. For the remaining Staph. aureus strain (M4) gene segments encoding IgG binding domains E, D and A and a new domain BC with a size of 219 bp could be observed. The BC domain appears, with a deletion of a 123 bp segment from the border region between both domains, as fused domain of both previously characterized domains. The Xr-region of this strain had 11 octapeptide repeats.

The pH-dependent behaviour of soluble protein aggregates produced by the pre-heating of reconstituted skim milk at 90 °C for 10 min was studied, in order to understand the role of these aggregates in acid gelation of heated milk. The following milk samples were prepared: (1) control (unheated reconstituted milk, pH 6·5); (2) milk heat-treated at pH 6·5 (mHtd6·5) and (3) milk heat-treated at pH 7·2 (mHtd7·2). They were centrifuged and the supernatants (SPNT 1) pH-adjusted to yield a series of pH values ranging from 6·5 or 7·2 to 4·6 using HCl at 20 °C or GDL at 20 and 38 °C. pH-Adjusted SPNTs 1 were re-centrifuged. The resulting supernatants (SPNTs 2) were analysed by OD (at 600 and 280 nm) and SDS-PAGE in order to characterise proteins still soluble as a function of pH. Particle size in SPNTs 1 was analysed by Steric Exclusion Chromatography. The OD600 nm revealed that during acidification soluble casein in both control and heat-treated samples exhibits variations in its optical properties or size as previously shown with micellar casein. In heat-treated samples, soluble casein and heat-induced covalent soluble aggregates precipitate at the same pH value. A progressive acidification of the soluble phase did not separate them. Increasing the temperature of acidification from 20 to 38 °C resulted in an increase in the precipitation pH of the proteins. However choice of acidifier did not have a significant effect on OD profiles. The soluble covalent aggregates from mHtd7·2 were smaller, more numerous, and had a higher content of κ-casein than mHtd6·5. Both types of aggregates began to precipitate at the same pH value but precipitation occurred over a narrower pH-range for soluble aggregates prepared from mHtd7·2. This may explain the higher gelation pH of mHtd7·2 compared with mHtd6·5.

We investigated the effects of omitting one or two milkings weekly on milk yield and milk composition in dairy sheep. Ninety Spanish Assaf ewes were allocated to three experimental treatments: T0 (no milking omission; 14 milkings/week), T1 (omission of one evening milking per week; 13 milkings/week) and T2 (omission of two evening milkings per week; 12 milkings/week). The experiment was extended for 11 weeks, from week 7 after lambing to week 17 of lactation. Once a week, daily milk production was recorded just before and after the omission. Milk quality and composition was analysed in lactation weeks 9, 13 and 16. Omission of one or two evening milkings each week resulted in an increase in milk production recorded the day immediately after the omission (27% for T1 and 16% for T2), which can be attributed to accumulation of milk in the udder cisterns. This increase did not compensate completely for the loss of yield in the omitted milking. The lack of significant differences among the three milking strategies in milk production (on average 1903, 2062 and 1833 ml/d for T0, T1 and T2, respectively) recorded before omission throughout the trial, would indicate the absence of residual effects of the omission on production in subsequent weeks. Treatment T2 resulted in a loss of milk production of approximately 39% during those days when milkings were omitted, representing a decrease of approximately 10% of the estimated weekly milk production. In relation to milk composition, milking omission led to significant increases in the fat and protein concentrations of the milk collected after the omission, whereas differences were not significant for the milk obtained before the omission. Despite the slight increase observed in treatment T2 after the milking omission, the somatic cell count was always far below that considered as indicative of possible pathologies. In conclusion, at least one evening milking could be omitted each week in high-producing dairy sheep without adversely affecting milk yield and milk composition.

Nutritional status has a major impact on the immune system. Probiotic effects ascribed to fermented dairy products arise not only from whole microorganisms but also from metabolites (peptides, exopolysaccharides) produced during the fermentation. We recently demonstrated the immunomodulating capacity of kefir in a murine model. We now aimed at studying the immunomodulating capacity in vivo of the products derived from milk fermentation by kefir microflora (PMFKM) on the gut. BALB/c mice received the PMFKM for 2, 5 or 7 consecutive days. IgA+ and IgG+ cells were determined on histological slices of the small and large intestine. IL-4, IL-6, IL-10, IL-12, IFNγ and TNFα were determined in the gut, intestinal fluid and blood serum. IL-6 was also determined in the supernatant of a primary culture of small intestine epithelial cells challenged with PMFKM. PMFKM up-regulated IL-6 secretion, necessary for B-cell terminal differentiation to IgA secreting cells in the gut lamina propria. There was an increase in the number of IgA+ cells in the small and large intestine. The increase in the number of IgA+ cells was accompanied by an increase in the number of IL-4+, IL-10+ and IL-6+ cells in the small intestine. Effects of PMFKM in the large intestine were less widely apparent than the ones observed at the small intestine lamina propria. All cytokines that increased in the small intestine lamina propria, also did so in blood serum, reflecting here the immunostimulation achieved in the gut mucosa. We observed that the PMFKM induced a mucosal response and it was able to up and down regulate it for protective immunity, maintaining the intestinal homeostasis, enhancing the IgA production at both the small and large intestine level. The opportunity exists then to manipulate the constituents of the lumen of the intestine through dietary means, thereby enhancing the health status of the host.

Proteolysis is one of the major biochemical events which takes place during cheese ripening and its degradation products, amino acids and peptides, have a considerable influence on the sensory characteristics of cheese (Urbach, 1993). Primary proteolysis leads to the formation of large water-insoluble peptides and smaller water-soluble peptides. Several peptides from bovine milk cheeses have been isolated and identified, particularly from Cheddar cheese (e.g., McSweeney et al. 1994; Singh et al. 1994, 1995, 1997; Gouldsworthy et al. 1996; Fernández et al. 1998). However, there are few data available on the identification of peptides from ewes’ milk cheeses, although Michaelidou et al. (1998) identified some major peptides in the water-soluble fraction of Feta cheese (ewe's milk cheese).

The principal aim of this work was to compare Caciotta cheeses obtained from cow milk previously subjected to high pressure homogenisation (HPH) at 100 MPa with those produced from raw (R) or heat-treated (P) cow milk. HPH had both direct and indirect effects on cheese characteristics and their evolution during ripening. In particular, HPH treatment of milk induced a significant increase of the cheese yield; moreover, it affected the microbial ecology of both curd and cheese. Compared with the thermal treatment, the HPH treatment resulted in a decrease of about one log cfu/g of yeast and lactobacilli cell loads of the curd. The initial milk treatment also affected the evolution over time and the levels attained at the end of ripening of all the microbial groups studied. In fact, lactobacilli, microstaphylococci and yeast cell loads remained at lower levels in the cheeses obtained from HPH milk with respect to the other cheese types over the whole ripening period. Moreover, HPH of milk induced marked and extensive lipolysis. Cheeses from HPH milk showed the presence of high amounts of free fatty acids immediately after brining. The electrophoretic patterns of the different cheese types showed that Caciotta made from HPH-treated milk was characterized by a more extensive and faster proteolysis as well as a significant modification of its volatile molecule profile. The results obtained and the sensory analysis indicated that HPH treatment of milk was able to differentiate Caciotta cheese or to modify its ripening patterns.

Four multiparous Holstein cows, each equipped with a duodenal cannula and an ultrasonic mammary blood flow probe, were assigned to a 4×4 Latin square to measure the effects of duodenal infusions of Leu (0, 40, 80 and 120 g/d) on lactational responses and mammary metabolism of nutrients. Cows were fed a diet of 67·0% corn silage, 5·3% grass hay, 14·2% peas, 7·8% maize starch, 1·0% each of molasses, urea, and sodium bicarbonate and 2·4% minerals and vitamins. Requirements of the remaining 9 essential amino acids were met by infusing into the duodenum 323 g/d of a mixture of amino acids including, Ile, Val, Met, Lys, Trp, Phe, His, Thr, Arg, Tyr and Glu. Milk protein content and yield were highest with 40 g/d of Leu and then progressively declined. Milk fat content and yield were significantly decreased over the entire range of Leu infusions. Arterial concentrations of Leu increased linearly and corresponding mammary arterio-venous differences increased to a plateau with 80 g/d of Leu. As Leu infusions increased, extraction rates of Leu decreased linearly, whereas those of Ile, Val, Lys, Arg, Thr and Tyr were significantly increased. Leu was taken up by the mammary gland according to milk output with 0 and 40 g/d of Leu infused and then largely in excess. Based on responses of protein yield and mammary uptake to milk output ratio, Leu concentration in total amino acids absorbed in the small intestine needs to be close to 8·9% for optimal milk protein synthesis.

Nineteen calves were reared in individual pens (IP), and twenty calves were grouped in four pens of five calves each (GP) to study the effects of grouping on behaviour and performance. All calves were on an enhanced-growth feeding programme and had unlimited access to starter. Total dry matter intake (DMI) body weight (BW), serum urea, non-esterified fatty acid (NEFA), cortisol, insulin, and glucose concentrations, and immune response to vaccination were measured. Behaviour patterns categorized as: non-nutritive oral behaviour, cross-sucking, inter-sucking, and self-grooming, were monitored 20 min following the morning and afternoon milk replacer consumption, and at the same times after weaning. There were no differences in final BW and starter DMI between treatments. The decrease of average daily gain (ADG) during the week following weaning was more pronounced in GP than in IP calves. Serum NEFA concentration increased at the beginning of the study in IP calves but serum urea concentrations were greater in GP than in IP calves the week after weaning. Self-grooming increased in both treatments throughout the study but GP calves increased non-nutritive oral behaviour and decreased cross-sucking and inter-sucking behaviours throughout the study. Rearing calves in groups following an enhanced-growth feeding programme did not increase starter intake. Furthermore, cross-sucking and inter-sucking behaviours in calves reared in groups did not seem to cause increased metabolic or immunological distress.

Organophosphorus pesticides (OPPs), widely used in agriculture, can cause toxic effects to humans and animals. The main purpose of the present work was to determine the contamination in raw milk by the main organophosphorus pesticides used in Italy and to evaluate the opportunity to start specific procedures of risk management along the milk production chain. The samples, collected in 4 Italian dairy plants directly from the tank trucks during the delivering, were representative of 920 tonnes of raw milk. The isolation of the OPPs (acephate, chlorpyriphos, chlorpyriphos-methyl, diazinon, methamidophos, methidathion, phorate, pirimiphos-methyl) was performed by liquid partition followed by clean-up with solid phase extraction. The analyses were carried out by dual column gas chromatography using two nitrogen-phosphorus detectors. Among the 135 samples analysed, 37 were positive in traces and 10 showed an OPP contamination ranging from 5 to 18 μg/kg. The higher results were recorded in the samples collected during the autumn-winter period. The main pollutants detected were acephate and chlorpyriphos. In every positive sample found, the OPP contamination was lower than the maximum residue level (MRL) fixed by the European Commission.

The aim of this study was to compare the effects of supplementing dairy cows with 1000 IU/day of all-rac-α-tocopheryl acetate (SynAc), RRR-α-tocopheryl acetate (NatAc), or RRR-α-tocopherol (NatAlc), from approximately 3 weeks before estimated calving until 2 weeks after calving, on the concentration of α-tocopherol and its stereoisomers (RRR-, RSS-, RRS-, RSR- and the four 2S-forms of α-tocopherol) in blood and milk. An unsupplemented group was included as control. Blood samples were collected at 3, 2 and 1 weeks before estimated calving, at calving, and 3, 7 and 14 days after calving, while milk samples were taken twice within 24 h after calving and at 7 and 14 days in milk. Overall, time and treatment had significant effects on plasma α-tocopherol with higher concentrations in NatAc than in the other groups. In addition, SynAc had higher concentrations than Control, and NatAlc tended to be higher than Control. The lowest plasma concentrations were observed at calving and 3 days after calving. Independent of treatment, the concentration was higher in colostrum than in milk day 7 and 14 after calving. Analyses of the stereoisomer distribution in plasma and milk showed that, irrespective of dietary treatment, RRR-α-tocopherol was the most predominant form, constituting more than 86%, whereas the remaining part of α-tocopherol was made up by the three synthetic 2R isomers, while the 2S isomers only contributed less than 1% of the total α-tocopherol. In control cows and cows supplemented with natural vitamin E, the proportion of RRR-α-tocopherol in plasma and milk constituted more than 98% of the total α-tocopherol. In conclusion, the results indicate that daily oral supplementation of dairy cows with RRR-α-tocopheryl acetate gives the highest blood concentrations of α-tocopherol in the periparturient period. Analyses of the distribution of the individual stereoisomers of α-tocopherol further indicate that the bioavailability of RRR-α-tocopherol relative to synthetic stereoisomers in cattle is considerably higher than officially accepted until now.

The objective was to quantify the strength of the relationship between body condition score (BCS) and live weight (LW) in pasture-based Holstein-Friesian dairy cattle, and to determine the kg LW per unit BCS. A total of 26021 test-day records with information on both BCS (1–10 scale, where 1 is emaciated and 10 is obese) and LW across 1110 lactations from one research farm were used in the analysis. Correlation and regression analyses were used to determine the degree of association between BCS and LW in different parities, stages of the inter-calving interval and years. Correlations between BCS and LW were relatively consistent, with the mean correlation between BCS and LW across all data of 0·55 implying that differences in BCS explain approximately 30% of the variation in LW. Significantly different regressions of LW on BCS were present within stage of inter-calving interval by parity subclasses. Excluding calving, LW per unit BCS varied from 17 kg (early to mid lactation in parity 1) to 36 kg (early lactation in parity 4 and 5). However, LW per unit BCS was greatest at calving varying from 44 kg in first parity animals to 62 kg in second parity animals. On average, 1 BCS unit equated to 31 kg LW across all data.

The effects of high pressure treatments (100–300 MPa; 15 min; 9 °C or 20 °C) on the distribution of minerals and proteins of raw skim milk (RSM) and of a dispersion of industrial phosphocaseinate (PC) were studied after separation of the micellar and soluble phases by ultracentrifugation (UCF). Whatever the temperature of high pressure treatments, the pressure-induced dissociation of the casein micelles was accompanied by calcium (Ca), phosphorus (P) and casein release from the micelles. The released Ca and P were or became bound to soluble proteins since progressive increases in Ca and P concentrations were observed in the UCF supernatants of RSM and of the PC dispersion but not in the ultrafiltrates from these UCF supernatants (free of soluble proteins). Simultaneously, αS1-, αS2-, β- and κ-caseins were progressively released from the micelles, as seen by electrophoretic analysis. The pressure-induced solubilisation of αS1- and αS2-caseins, essentially located in the core of the micelles, suggests that high pressure destabilised micelles including their internal structure.

The aim was to investigate the interdependence of udder quarters within cow towards the incidence of intramammary infections during the dry period in herds under selective dry-cow antibiotic therapy. A total of 368 cows among 28 herds were included in a survey. Quarter milk samples collected at the last milking before drying-off and on day 3 after calving were submitted to microbiological procedures. An expected distribution of cows according to their number of newly infected quarters was calculated based on a binomial probability distribution from the overall quarter incidence (or from the quarter incidence in each herd) and compared with the observed distribution. Incidence of newly infected quarters ranged from 0·0 to 39·3%, depending on the herd (median: 17·7%). Interdependence of quarters towards new infection during the dry period was observed whatever the pathogen type, for both treated and untreated cows. Calculation of an expected distribution of cows according to their number of newly infected quarters using the quarter incidence in each herd (instead of the overall quarter incidence) reduced the distance to the observed distribution, but interdependence was still observed. Our results support the application of selective antibiotic therapy at the cow level rather than at the quarter level.

Milk powder taken to Antarctica on Shackelton's British Antarctic Expedition in 1907 was produced in New Zealand by a roller drying process in the first factory in the world dedicated to this process. Thermophilic bacilli are the dominant contaminants of modern spray-dried milk powders and the 1907 milk powder allows a comparison to be made of contaminating strains in roller-dried and spray-dried powders. Samples of milk powder obtained from Shackelton's Hut at Cape Royds had low levels of thermophilic contamination (<500 cfu ml−1) but the two dominant strains (Bacillus licheniformis strain F and Bacillus subtilis) were typical of those found in spray-dried powders. Soil samples from the floor of the hut also contained these strains, whereas soils distant from the hut did not. Differences in the RAPD profiles of isolates from the milk powder and the soils suggest that contamination of the milk from the soil was unlikely. It is significant that the most commonly encountered contaminant strain in modern spray-dried milk (Anoxybacillus flavithermus strain C) was not detected in the 1907 sample.

A data file on 36396 lactations was used to determine the strength of the phenotypic relationship between productive, reproductive and lifespan traits for 7935 Spanish Churra ewes. These ewes first lambed between 1989 and 1997 and belonged to 23 flocks. The study took into account four lifespan traits (lifetime, productive life, useful life and lifetime score), three productive traits (total milk yield produced during lifetime, lambs weaned during ewe lifetime and total revenues from sold milk and weaned lambs during lifetime) and two reproductive traits (age at first lambing and mean interval between successive lambings). Moreover, milk yield and revenues from sold milk and weaned lambs were calculated per day of lifetime, productive life and useful life. Partial lifespan traits were considered for the first three parities. The model included flock, birth year within flock and season of birth of the ewe as fixed effects. The first two effects contributed significantly to variation in all traits, while season significantly affected lifespan traits, productive traits and age at first lambing. Milk production level was added to the model for lifespan traits only. It significantly contributed to explaining the variation in all lifespan traits with high percentage of variance explained averaging 14·91%. Lifetime averaged 2324 d. Productive life accounted for 57% of lifetime while useful life represented 50% of productive life. Age at first lambing averaged 622 d and average days dry during lifetime was 560. During lifetime, ewes gave an average of 4·6 parities, 6·5 weaned lambs and 636 l of milk. Average revenue from milk and lambs during lifetime was €673. Milk/day of lifetime, productive life and useful life averaged 0·26, 0·51 and 0·93 l, respectively. The corresponding per-day revenues from sold milk and weaned lambs were, €0.27, €0.55 and €1.01, respectively. Lifespan and productive traits had strong relationships (rp among these traits ranged between 0·75 and 0·95). Two-parity and three-parity per-day milk yield had moderately high correlation (0·70–0·83) with total lifetime per-day milk yield traits. Therefore, good use of these traits would be helpful in determining best individuals early in life.

Extrinsic factors such as maternal microbiota, bacterial load of the environment, diet and medication modulate the intestinal microbiota. Maturation and function of the immune system is influenced by established gut microbiota. In this work we describe the immunological effects of the dietary deprivation of fermented foods of healthy volunteers. Significant decreases in faecal lactobacillus and total aerobes counts and concentration of short chain fatty acids were observed following deprivation of fermented food of the normal diet. Moreover, a decrease in phagocytic activity in leukocytes was observed after two weeks of restricted diet. Therefore, the dietary deprivation of fermented foods could induce a decrease in innate immune response that might affect the capacity to respond against infections. The ingestion of a probiotic product containing the strains Lactobacillus gasseri CECT5714 and Lactobacillus coryniformis CECT5711 or a standard yogurt containing a conventional starter Lactobacillus delbrueckii sp. bulgaricus counteracted the fall in the immune response, although the probiotic product was more effective than the standard yogurt.

Simplified designs of milk-composition recording, based on information from a single monthly milking, adjusted or not for interval between milkings and for milk yield, were simulated and evaluated for 2553 ewe-test-day records belonging to 155 lactations of Churra dairy ewes. Accuracy of simplified methods was evaluated by comparing estimated trait values (fat, protein, casein and total solid yields) with those observed both in a reference plan, where the two daily milkings were recorded at weekly intervals (A1), and in the official A4 milk recording (monthly records of the two daily milkings). Trait yields per lactation were estimated and adjusted to the only milking period (days in milk 30–120). Estimates of milk component traits were less precise when monthly designs, including the A4 design, were compared with a weekly sampling of both a.m. and p.m. milkings, with fat yield being the most difficult trait to estimate. All options with one daily milking every month were more accurate when the corresponding plan was based on, or began with, the a.m. milking. Adjustment for the preceding interval between milkings or milk yield did not improve sampling accuracy. The design alternating a.m. and p.m. milkings every month, without adjustment, is suggested for recoding milk component traits in dairy ewes.

A study was carried out to determine genetic variants of β-lactoglobulin gene and to explore associations between these and milk composition traits in riverine buffalo. Single strand conformation polymorphism was employed to detect the genetic variants of the gene. Two fragments of this gene i.e. 119 bp of exon I and 400 bp spanning exon IV and intron IV were included in the study. For 119 bp fragment, three alleles namely, A, B and C were observed in all the buffalo breeds whereas four alleles (A, B, C and D) were detected for 400 bp fragment. The frequency distribution of alleles was different in different breeds of buffaloes for both the fragments. For exon I fragment, the milk composition traits such as total SNF, protein, solid, fat and whey protein yield were found to be significantly (P<0·05) associated with genotypes in Murrah and Bhadawari buffalo whereas in Mehsana breed genotypes were significantly (P<0·05) co-related with total SNF, solid and fat yield. Genotypes of 400 bp fragment, only total fat yield in Mehsana buffalo was found to be significantly (P<0·05) associated with genotypes.

In studies of bovine mastitis, it has been repeatedly confirmed that milking is associated with entrance of microorganisms into the mammary gland (International Dairy Federation, 1987; Bramley, 1992). As different husbandry practices apply between cattle and dairy sheep (cows are machine milked, ewes are usually hand milked), results from cows may not always apply to ewes. The objective of this investigation was to determine whether there were differences in bacterial flora populations in the teat duct and mammary secretion of ewes before and after milking.

The goal of this study was to measure the economic impacts of reduced milk production associated with papillomatous digital dermatitis (PDD) in dairy cows in the USA, and of specific risk factors for PDD, in 1996. The method applied was an economic-welfare analysis of producer and consumer surplus, with the GUM Workbench used to analyse uncertainties in the measurements. Reduced milk production associated with PDD was found to reduce consumer surplus by $750 million±$580 million, and to increase the economic surplus of producers by $560 million±$470 million, with a net economic loss of $190 million±$130 million. An examination of the economic effects of specific epidemiologic risk factors for PDD showed that having dairy cows that were not born on the operation had important economic consequences associated with the disease, as did the type of land to which dairy cows had access during the winter months and the type of flooring on which cows walked. Washing hoof-trimming equipment between cows was an important biosecurity measure that was associated with reduced PDD. The epidemiologic model used also implicated hoof trimmers who trimmed cattle hooves on other operations as having an important economic impact associated with this disease, although this finding may have been erroneous.
