Thirty-seven lines from a population derived from the hull-less barley cultivar, Penthouse, were grown in a replicated trial over three seasons and assessed for grain yield. Following harvest, a rapid test to measure grain dimensions was applied to all samples, to look for novel variation in grain size and shape, as a possible way of detecting mutations. A range of grain and malt quality traits was also measured in two of the seasons, to detect genotype × season interactions and determine relationships between the measured traits. There were significant differences between years for all traits and between genotypes for most. Genotype × season interaction was significant for grain dimensions and some malting traits, but a correlation between malting quality and grain dimensions was only observed in one season. Line 30 showed very high yield potential in a comparatively wet season and gave a higher alcohol yield per unit area than a hulled control variety, while lines 21 and 33 contained putative additional mutations. Line 21, previously observed to have higher enzyme activity, appeared to contain an additional dwarfing gene and was characterized by smaller grain, later ear emergence and lower yield. Line 33, with malting potential, showed considerably altered grain length to width ratio and will be further investigated as a possible globosum type.

Bagasse and rice husk are two important agro-industrial by-products that are used as fuel in the sugar and rice mill industries, thus producing large quantities of bagasse ash (BA; 0·05 of bagasse) and rice husk ash (0·20 of rice husk) as waste material. Applying BA and rice husk ash (RHA) to agricultural land improves yield, nutrient uptake and chemical fertility of soil, particularly with special reference to available phosphorus (P) and potassium (K). The present field experiment was conducted for 3 years to evaluate the P fertilizer value of these agro-industrial waste materials in a wheat–rice system (WRS). The experiment was laid out in a split-plot design with RHA and BA applied at 10 t/ha and including a no-amendment control as the main plot treatments and three levels of fertilizer P (0, 13 and 26 kg P/ha; designated P0, P13 and P26, respectively) as sub-plot treatments to wheat in WRS. Application of fertilizer P increased the wheat grain yield up to P26 in the un-amended control treatment. However, a significant response of wheat to fertilizer P was also observed up to P13 in the presence of BA and RHA, thereby saving 50% of fertilizer P. Both RHA and BA increased wheat productivity by 12 and 16%, respectively, over the un-amended control. The subsequent rice crop also produced 14% higher paddy yield when the two ashes were applied along with P13 to the previous wheat crop. The increases in grain yield were accompanied by significant increases in the uptake of P and K, and P content (Olsen P) in the soil. The application of recommended P (P26) in un-amended plots resulted in a negative P balance of 9·3 kg P/ha/year. On the other hand, the application of BA alone and RHA along with P13 resulted in neutral/slightly positive P balance. A strong linear relationship (R2 = 0·98) was observed between P balance and Olsen-P build up in the soil. It may be concluded that application of BA and RHA has the potential to increase system productivity and reduce the cost of inputs in terms of reduced application of fertilizer P to wheat and rice.

Assessment of the value for cultivation and use (VCU) of a new cultivar, essential for its official registration, is done through a series of trials carried out over a 2–3-year period and across many locations. In a set of multi-environment VCU trials, evaluation of new genotypes can be a laborious task due to the presence of genotype by environment interactions, which can hide their true genetic value. In an attempt to reveal the true genetic value of new cultivars, a good starting point is investigation of the importance of various genetic and environmental sources of variation, which can be done by estimating relative magnitude of corresponding variance components within the mixed model framework.

Genotype × location × year (G × L × Y) data set for seven crops taken from the 10-year period 2001–10 was used in the present study to estimate the variance components for main effects and their interactions in Croatian VCU trials. Depending on the crop, the most important and least important components were Y or LY, and L or GL, respectively. Genotypic effect was relatively small, ranging from 2·1 to 13·4% of the total variation. The current results are comparable with the relative sizes of the variance components obtained in studies from four- to sixfold larger countries, indicating that the environments within Croatia, if sufficiently widely sampled, can provide as extreme cultivar responses as a geographically more dispersed set of VCU trials. The gap range in different crops is much wider (30–60%) than in Western Europe (up to 30%), but it remained constant over the 10-year period.

APETALA2/ethylene-responsive factor (AP2/ERF) family transcription factors play a vital role in plant growth and in response to hormones and abiotic stresses. In the current research, it is reported that OsDERF2, one of the drought-responsive ERF, is a member of the DREB sub-family. OsDERF2 is a nuclear-localized protein and has transcriptional activity in yeast. Expression of OsDERF2 was induced by drought and inhibited by abscisic acid (ABA). However, OsDERF2 RNA interference (RNAi) knock-down transgenic lines enhanced tolerance to drought stress at seedling stage and were much more sensitive to ABA treatment, which may result from the increased ABA level in vivo. The basic leucine zipper (bZIP) transcription factor family plays an important role in the ABA signalling pathway of abiotic stress. Quantitative real-time polymerase chain reaction analysis revealed that the bZIP family gene OsbZIP20 and ABA-response gene OsABA45 were up-regulated 25 times and 120 times, respectively, in OsDERF2 RNAi knock-down lines under drought stress, which were up-regulated five and seven times in wild type under drought stress. The current data reveal that OsDERF2 negatively modulates drought stress response in an ABA-mediated pathway through regulating gene expression of other ABA-response transcription factors.

Greenbug infestations to sorghum can cause severe and above economic threshold damage in the Great Plains of the United States. The current study was conducted to identify quantitative trait loci (QTLs) and potential candidate genes residing within the QTL region responsible for greenbug resistance in an advanced mapping population. Inter-crossed populations are useful in detecting QTLs tightly linked to genetic markers with high resolution. In the current study, QTLs responsible for greenbug resistance in sorghum were mapped using an inter-cross population derived from two parents, BTx623 (greenbug-susceptible line) and PI 607900 (greenbug-resistant line). Molecular markers for 115 loci were used to construct a linkage map which eventually facilitated tagging portions of the sorghum genome regions responsible for greenbug resistance. The molecular genetic map covered all the chromosomes of sorghum with a total genome length of 963·0 cM. The advanced mapping population revealed and confirmed the location of greenbug resistance loci, which explained a high phenotypic variation from 72·9 to 80·9% of greenbug resistance. The loci for greenbug resistance were mapped to the region flanked by markers Starssbnm 93 and Starssbnm 102 on chromosome 9 with an increased allelic effect from the resistant parent. The locations of these loci were compared with a previous study on QTL analysis using an F2 mapping population. The results from the present study were in agreement with the findings in the F2 QTL analysis and identified QTLs had a better confidence interval. The markers/QTLs identified from the current study can be effectively utilized in marker-assisted selection and map-based cloning experiments.

Identification of new effective strategies for improving crop yields under environmental stresses such as drought represent key priorities for researchers around the globe. In the present study, the effects of different methods of exogenous selenium (Se) supply viz. Se seed priming, Se fertigation and Se foliar spray on yield of spring wheat under normal and water deficit conditions were investigated. Two field experiments were conducted using one indigenous drought-tolerant genotype (Kohistan-97) and a sensitive genotype (Pasban-90) to understand the role of Se in improving wheat yield. The experiments were laid out in a split-split plot design with three replications during consecutive years (2011/12 and 2012/13) and the plants were exposed to water stress by withholding irrigation at two different wheat growth stages, viz. tillering and anthesis. It was noted that drought stress significantly affected the yield attributes of wheat; however, exogenous Se supply was observed to be helpful in improving the drought tolerance potential and yield of water-stressed wheat plants through maintenance of plant water status. A significant increase in wheat yield by Se supply was also noted under normal conditions. The normal plants fertigated with Se maintained the highest values for number of productive tillers, spike length, number of grains per spike, thousand-grain weight, biological and grain yield with no significant difference from Se foliar spray at the tillering stage, which was found to be the most effective method of exogenous Se supply for improving wheat yield under water deficit conditions. Moreover, Se fertigation and foliar spray resulted in the maximum accumulation of Se in shoots and gave the highest net return and cost-benefit ratio under drought stress conditions. The present study is one of the few reports on the role of Se in alleviating water stress for obtaining maximum profit in field grown spring wheat.

Optimal nitrogen (N) management for maize in the film-mulched production systems that are widely used in dryland agriculture is difficult because top-dressing N is impractical. The current research determined how matching N supply and demand was achieved before and after silking stages, when single applications of controlled release urea (CRU) were combined with conventional urea in film-mulched maize production. The CRU: urea mixture was applied in a 1 : 2 or 2 : 1 ratio and all three fertilizer regimes (urea alone and CRU: urea at 1 : 2 or 2 : 1) were applied at N rates of 180 and 240 kg/ha over 2 years. The 1 : 2 CRU: urea mixture, applied once at 180 kg N/ha, was found to synchronize N supply with demand, thereby reducing N losses. The highest grain yields (11·8–12·0 t/ha), N uptake (232–239 kg/ha), N recovery (65·8–67·7%) and high net economic return were achieved with this regime. These results indicate that a single application of a mixture of CRU and urea can synchronize N supply with demand and provide higher yields and profits than conventional N fertilization in film-mulched maize systems.

Twenty Small Tailed Han (STH) and 20 Ujumqin (UJU) ewes naturally infected with gastrointestinal nematodes were randomly assigned to one of four treatments arranged in a 2 × 2 factorial design, receiving anthelmintic treatment (AT) or non-anthelmintic treatment (NonAT) prior to lambing. After lambing, the effects of AT on feed intake, digestion and milk yield in ewes, and the growth rates of lambs fed their mother's milk were assessed for 28 days. Faecal samples were collected to determine faecal egg counts (FECs), milk was collected to measure milk yield and ewes and lambs were weighed to quantify daily body weight change. The results showed that AT significantly increased ewe dry matter intake (2411 g/d for AT and 2209 g/d for NonAT) and decreased FECs (50 eggs/g for AT and 2655 eggs/g for NonAT). All ewes lost weight after lambing, but body weight loss in the AT (43 g/d) was significantly less than in NonAT (84 g/d), and STH ewes (70 g/d) lost more weight than UJU ewes (58 g/d). Anthelmintic-treated ewes produced more milk for their lambs to consume. However, the extent of these positive effects of AT differed between STH and UJU ewes. The average daily body weight gain of lambs in AT was higher than those in NonAT. In conclusion, effective AT in ewes before lambing benefits subsequent lactation in ewes and growth rate in lambs.

Maize (Zea mays L.)–common bean (Phaseolus vulgaris L.) intercropping is a recent practice in north-western Ethiopia and there is limited information on its productivity. A field experiment was conducted at South Achefer and Mecha in north-western Ethiopia during the 2012 and 2013 crop growing seasons to determine combinations of intercrop planting arrangement (IPA) with nitrogen (N) and phosphorus (P) rates for optimizing maize–common bean intercrop productivity and profitability. Treatments consisted of factorial combinations of two IPA (single row of common bean between maize rows and paired rows of common bean between paired rows of maize), two N rates (92 and 128 kg N/ha) and two P rates (20 and 40 kg P/ha). A sole crop maize with recommended fertilizer rate of 128/40 kg N/P/ha was used as a control treatment. The treatments were laid out in a randomized complete block design with three replications. Results indicated that land equivalent ratio was more than unity, and the intercrop system was 20% more productive relative to the sole crop. Maize equivalent yields were highest for most of the intercrop treatments relative to mono-crop maize with yield advantage of 14% from single row IPA with 128/20 kg N/P/ha. Single row IPA with 128/20 kg N/P/ha and paired row IPA with 92/20 kg N/P/ha increased financial returns by 16 and 8% relative to sole crop maize, respectively. Smallholder maize-based cropping of north-western Ethiopia could be nutritionally, agronomically and financially improved through maize–common bean intercropping of single row IPA with appropriate nutrient management.

Honeybee foraging can transfer exogenous genes from genetically modified (GM) oilseed rape (Brassica napus L.) to closely related plants, which not only induces potential ecological risks but also contaminates non-GM seeds or honey products with GM ingredients. These events may lead to international trade disputes. Chinese honeybees (Apis cerana cerana Fabricius) and a herbicide (glufosinate)-resistant GM strain of B. napus (Z7B10) were studied to examine the effects of honeybee short-range foraging on oilseed rape gene flow and honey ingredients. Results showed variable frequencies of gene flow between GM and non-GM oilseed rape cultivars, with the highest frequency under nylon net isolation with artificially stocked honeybees, the lowest frequency under nylon net isolation alone, and an intermediate frequency under natural pollination, suggesting the important role of honeybee foraging in gene flow frequency. Additionally, GM pollen grains were found in honey collected from honeybees foraging on both GM and non-GM oilseed rape cultivars. The phosphinothricin acetyltransferase protein was also detected in both unbroken pollen-containing and pollen-free honey by protein testing strips, suggesting that honeybee foraging on GM oilseed rape could lead to contamination with GM ingredients. Overall, the results provide a direct scientific basis for the ecological risk assessment and safety management of GM oilseed rape.

Physiological traits can be used to improve the efficiency of selecting suitable genotypes to grow under nitrogen (N) limitation. The objective of the current study was to investigate the relationship between physiological characteristics and storage root yield of three cassava genotypes under three rates of N fertilizer. The experiments were conducted from 2014 to 2016 at farm fields in Thailand. A split-plot randomized complete block design with four replications was used. Three different rates of N fertilizer, i.e., 46·9, 90·0 and 133·2 kg N/ha and three cassava genotypes, Rayong 9, Rayong 11 and Kasetsart 50, were used. Kasetsart 50 had the highest mean performance for most crop traits. Growth rate of stem (SGR), storage root (SRGR) and crop (CGR) during 180–210 days after planting (DAP) and leaf area index (LAI) at 120 DAP were related to storage root dry weight for all three rates of N fertilizer. Storage root growth during 90–120 DAP, CGR during 180–210 DAP and specific leaf area (SLA) at 210 DAP contributed most to storage root dry weight of the three genotypes grown at 46·9 kg N/ha, while the combination of SRGR during 90–120 DAP, SRGR during 180–210 DAP, LAI at 210 DAP and SLA at 210 DAP was best for N fertilizer at 90·0 kg N/ha and the combination of leaf growth rate (LGR) during 180–210 DAP and LAI at 210 DAP was best for N fertilizer at 133·2 kg N/ha.

Diminishing irrigation water availability and increased crop evapotranspiration (ET) have already been shown to pose threats to agricultural productivity. The aim of the current study was to estimate the values and trends of both ET and net irrigation requirements (NIR) of those crops consuming most water due to the size of their cultivated area (citrus and potatoes) in a water-scarce region with typical Mediterranean climate, such as Cyprus, analyse possible spatial and temporal variations of these parameters, and discuss implications of this analysis on agricultural water conservation. A linear regression analysis of ET and NIR values of potatoes and citrus during recent decades in their four main cultivated areas uncovered both increasing and decreasing trends for these parameters. Overall, however, the results did not show any change in irrigation water demands for these crops during recent years. In accordance with this outcome, average ET and NIR values of the majority of cultivated crops on the island showed no statistically significant differences between the periods 1976–2000 and 1990–2014. Conversely, this was not the case when data were analysed within each weather station across crops. It is suggested, therefore, that agricultural water resource management schemes should not be focused merely on a regional analysis of changes but that emphasis should be given to site-specific values and trends of ET and NIR estimations, ultimately serving the conservation of agricultural water.

Crops often experience combined soil stresses. Root responses to soil penetration resistance (PR) and drought stress can be an important basis for crop management. In 2013/14, a 2-year experiment was conducted to evaluate the effect of tillage treatment and drought stress (no irrigation for 4, 12 and 20 days during the V10–V16 growth stage) on the root length, diameter, cortex and cortical cell, and grain yield of maize (Zea mays L.) in a clayey red soil in southern China. Total root length and average root diameter were significantly correlated with soil PR and moisture. The cortical cell file number increased with soil PR and drought stress, while cortical cell size increased only with soil PR. Soil PR and moisture played different roles in maize root morphology modifications, but were both affected by tillage practices. Deep ploughing and conventional tillage increased soil moisture under severe drought stress conditions, whereas soil compaction and no-till significantly increased soil PR. The results indicate that high PR in clayey red soil was responsible for a decrease in maize root size and grain yield under drought conditions.

Hazard identification is the first step in assessing the risk of a genetically modified (GM) crop. It employs the concept of substantial equivalence to evaluate crop safety. The current process relies on subjective opinions to integrate various comparisons among the GM crop, the non-GM counterpart and an assortment of non-GM references over an array of key endpoints measured in field trials. The pre-eminent need to control the consumer's risk in hazard identification has been left unaddressed. The current paper develops statistical strategies to resolve this issue. Hypotheses of individual tests are explicitly defined to reflect the study objectives. They are then grouped into families and connected by logical operators according to decision rules commonly used in crop safety evaluation. This pre-specification of hypotheses arranged in an organized layout leads to a simple, transparent decision-making process where the consumer's risk can be managed directly. A two-stage multiplicity adjustment procedure is created by applying fundamental principles for multiple testing to the newly assembled families of hypotheses. The practical utility of the proposed procedure is shown in a real-world example. Besides being easy to implement and convey, the proposed statistical strategies accommodate the addition of supportive evidence for safety and allow the nature of the genetic modification to be taken into account.

Assimilated carbon during vegetative and early reproductive growth in wheat is temporarily stored in stem internodes and leaf sheaths (LSs), and can later be remobilized and transported to developing grain. The present study was undertaken to determine the effect of several peduncle (the uppermost internode) morpho-anatomical and biochemical traits on grain weight, and to assess the contribution of the peduncle water-soluble carbohydrate (WSC) reserves shortly after anthesis to its variation. In 2-year field trials, 61 wheat genotypes were used (27 F4:5 families, 17 parents used for the crosses and the 17 current best standards), comparing intact control plants (CP) with plants that were defoliated (DP) by cutting off all leaf blades 10 days after anthesis. Estimated contributions of peduncle (culm (C) and flag LS) assimilate reserves to grain weight/spike were from 0·06 to 0·31 and from 0·11 to 0·45 in CP and DP plants, respectively. In both CP and DP plants, a higher contribution was from the LS than from the C. High peduncle reserve mobilization efficiency, a longer exposed part of the peduncle and larger C storage capacity (through higher parenchyma and/or lower lignified area) were of specific benefit for maintaining grain weight in defoliated plants. F4:5 families had higher transport capacity in the peduncle, but without any improvement in WSC-related traits compared with the best standards.

Soluble starch synthase II (SSII) plays an important role in the biosynthesis of starch and in rice it consists of three isoforms encoded by SSII-1, SSII-2 and SSII-3. However, the genetic effects of various SSII alleles on grain quality have not been systematically characterized. In the present study, the japonica alleles on SSII-1, SSII-2 and SSII-3 (SSIIa) loci from a japonica cultivar, Suyunuo, were respectively introgressed by molecular marker-assisted selection into a typical indica cultivar, Guichao2, through successive backcrossing, generating three sets of near-isogenic lines (NILs). Grain quality and starch property analysis showed that NIL-SSII-3j exhibited significant decreases in the following parameters: amylose content, average granule size, and setback viscosity and consistency; but increases in peak viscosity, hot paste viscosity, gelatinization temperature and relative crystallinity. Moreover, the proportion of short amylopectin chains and branching degree also increased when compared with those of NIL-SSII-3i (Guochao2). Similar effects were observed in NIL-SSII-1j , and certain alterations in the fine structure of starch (granule size) were revealed. However, NIL-SSII-2j did not exert significant effect on grain quality and starch properties. In brief, among the SSII gene family, the functional diversity occurred on SSII-1 and SSII-3, and not on SSII-2. Therefore, it appears that more attention should be directed to SSII-1 and SSII-3 loci for improving the eating and cooking quality of rice.

A range of options was explored to test the hypothesis that diets for dairy cows could be formulated to reduce the carbon footprint (CFP) of feed, increase efficiency of conversion of potentially human-edible feed into milk, increase nitrogen use efficiency (NUE) and reduce methane (CH4) emissions per kg milk. Diets based on grazed grass, grass silage, maize silage or straw, supplemented with raw material feeds, were formulated to meet requirements for metabolizable energy and metabolizable protein for a range of daily milk yields. At similar levels of milk yield, NUE, predicted CH4 emissions and diet CFP were generally higher for diets based on maize silage than for those based on grazed grass, grass silage or straw. Predicted CH4 emissions and human-edible proportion decreased, while NUE increased with the increasing level of milk yield. It is concluded that there is potential to reduce the environmental impact of milk production by altering diet formulation, but the extent to which this might occur is likely to depend on availability of raw material feeds with low CFPs.

The current experiment was conducted to evaluate the effects of different dietary protein levels and rumen-protected folic acid (RPFA) supplementation on ruminal fermentation, microbial enzyme activity, bacterial populations and urinary excretion of purine derivatives (PD) in growing beef steers. Low-protein (LP) or high-protein (HP) diets were fed to eight ruminally cannulated first-generation cross-bred (Blonde d'Aquitaine × Simmental) beef steers with or without RPFA supplementation. Steers were fed a total mixed ration, and dietary concentrate to maize silage ratio was 50 : 50 (dry matter (DM) basis). No interaction between dietary crude protein (CP) levels and RPFA supplementation was observed during the experiment. Ruminal pH was unaffected by RPFA supplementation, but decreased with increasing dietary CP levels. Ruminal total volatile fatty acid concentration increased with increasing dietary CP levels or RPFA supplementation. Molar proportion of acetate increased with RPFA supplementation, but tended to decrease with increasing dietary CP levels. The proportion of propionate decreased with RPFA supplementation, but was unaffected by dietary CP levels. As a result, the acetate to propionate ratio increased with RPFA supplementation, but tended to be lower for the HP diets than the LP diets. Ammonia-nitrogen content decreased with RPFA supplementation, but increased with increasing dietary CP levels. In situ ruminal degradability of maize straw and concentrate increased with increasing dietary CP levels or RPFA supplementation. Microbial enzyme (carboxymethyl-cellulase, cellobiase, xylanase, pectinase, α-amylase and protease) activity, bacterial populations (Ruminococcus albus, Ruminococcus flavefaciens, Butyrivibrio fibrisolvens, Prevotella ruminicola, Fibrobacter succinogenes and Ruminobacter amylophilus) and urinary PD excretion increased with increasing dietary CP levels or RPFA supplementation. The current study showed that increasing dietary CP levels from 109·1 to 130·7 g/kg DM or supplementing 75 mg RPFA improved ruminal fermentation and microbial protein synthesis by increasing bacterial population and microbial enzyme activity.

The current experiment was undertaken to investigate the effect of including white clover (Trifolium repens L.; WC) into perennial ryegrass (Lolium perenne L.; PRG) swards (PRG/WC) receiving 250 kg nitrogen (N) per hectare (ha) per year compared with PRG only swards receiving 250 kg N/ha/year, in an intensive grass-based spring calving dairy production scenario. Forty spring-calving cows were allocated to graze either a PRG/WC or PRG sward (n = 20) from 6 February to 31 October 2012. Fresh herbage was offered daily (17 kg dry matter (DM)/cow) supplemented with concentrate in times of herbage deficit (total supplementation 507 kg/cow). Pre-grazing herbage mass (HM), sward WC content and milk production were measured for the duration of the experiment. Herbage DM intake was estimated in May, July and September. Pre-grazing HM (±s.e.) was similar (1467 ± 173·1 kg DM/ha) for both treatments, as was cumulative herbage production (14 158 ± 769 kg DM/ha). Average WC content of the PRG/WC swards was 236 ± 30 g/kg DM. The PRG/WC cows had greater average daily milk yield and milk solids yield from June onwards. Cumulative milk yield and milk solids yield were greater for the PRG/WC cows compared with the PRG cows (5048 and 4789 ± 34·3 kg milk yield/cow, and 400 and 388 ± 1·87 kg milk solids/cow, respectively). Cows had similar DM intake in all measurements periods (15·1 ± 0·42 kg DM/cow/day). In conclusion, including WC in N-fertilized PRG swards increased milk production from cows grazing the PRG/WC swards compared with PRG, particularly in the second half of the lactation.
