The consequences of a low litter average birth weight phenotype for postnatal growth performance and carcass quality of all progeny, and testicular development in male offspring, were investigated. Using data from 25 sows with one, and 223 sows with two consecutive farrowing events, individual birth weight (BW) was measured and each litter between 9 and 16 total pigs born was classified as low (LBW), medium (MBW) or high (HBW) birth weight: low and high BW being defined as >1 standard deviation below or above, respectively, the population mean for each litter size. Litter average BW was repeatable within sows. At castration, testicular tissue was collected from 40 male pigs in LBW and HBW litters with individual BW close to their litter average BW and used for histomorphometric analysis. LBW piglets had a lower absolute number of germ cells, Sertoli cells and Leydig cells in their testes and a higher brain : testis weight ratio than HBW piglets. Overall, LBW litters had lower placental weight and higher brain : liver, brain : intestine and brain : Semitendinosus muscle weight ratios than MBW and HBW litters. In the nursery and grow–finish (GF) phase, pigs were kept in pens by BW classification (9 HBW, 17 MBW and 10 LBW pens) with 13 males and 13 females per pen. Average daily gain tended to be lower in LBW than HBW litters in lactation (P = 0.06) and throughout the nursery and GF phases (P < 0.01), resulting in an increasing difference in body weight between LBW, MBW and HBW litters (P < 0.05). Average daily feed intake was lower (P < 0.001) in LBW than HBW litters in the nursery and GF phases. Feed utilization efficiency (feed/gain) was similar for LBW and HBW litters in the nursery, but was lower (P < 0.001) in HBW than LBW litters in the GF phase. By design, slaughter weight was similar between BW classifications; however, LBW litters needed 9 more days to reach the same slaughter weight than HBW litters (P < 0.001). BW classification did not affect carcass composition traits. In conclusion, LBW litters showed benchmarks of intrauterine growth retardation, LBW had a negative impact on testicular development and germ and somatic cell populations, and was associated with decreased postnatal growth during all phases of production; however, no measurable effect on carcass composition traits was established.

The reliability of reverse transcription quantitative real-time PCR (RT-qPCR) depends on normalising the mRNA abundance using carefully selected, stable reference genes. Our aim was to propose sets of reference genes for normalisation in bovine or caprine adipose tissue (AT), mammary gland, liver and muscle. All of these tissues contribute to nutrient partitioning and metabolism and, thus, to the profitability of ruminant productions (i.e. carcasses, meat and milk). In this study, eight commonly used reference genes that belong to different functional classes (CLN3, EIF3K, MRPL39, PPIA, RPLP0, TBP, TOP2B and UXT) were analysed using the geNorm procedure to determine the most stable reference genes in bovine and/or caprine tissues. Abundances and rankings of reference genes varied between tissues, species and the combination of tissues and/or species. Therefore, we proposed 29 sets of reference genes that differed depending on the tissue and/or species. As examples of the 29 sets, EIF3K, TOP2B and UXT were proposed as the most stable reference genes in bovine AT; UXT, EIF3K and RPLP0 were the most stable reference genes in bovine and caprine AT. The optimal number of reference genes for data normalisation was 3 for 27 of the proposed 29 sets. In two of the 29 sets, four to five reference genes were necessary for data normalisation when the number of studied tissues was increased. For example, UXT, EIF3K, TBP, TOP2B and CLN3 were required for data normalisation in bovine mammary gland, AT, muscle and liver. We have evaluated some of our proposed sets of reference genes for the normalisation of CD36 gene expression. Normalisation using the three most stable reference genes has revealed downregulation of CD36 gene expression in bovine mammary gland by a concentrate-based diet that is supplemented with sunflower oil and upregulation of CD36 gene expression in caprine liver by including a rapidly degradable starch in the diet. The dietary regulation of the gene expression of CD36 has been erased by normalisation with the least stable reference genes, which may result in misinterpretation of CD36 gene regulation. To conclude, our results provide valuable reference gene sets for other studies that aim to measure tissue and/or species-specific mRNA abundance in ruminants.

Lameness in sows has an economic impact on pig production and is a major welfare concern. The aim of the present project was to develop methods to evaluate and quantify lameness in breeding sows. Five methods to study lameness were compared between themselves and with visual gait scoring used as a reference: footprint analysis, kinematics, accelerometers, lying-to-standing transition and foot lesion observation. Fifty sows of various parities and stages of gestation were selected using visual gait scoring and distributed into three groups: lame (L), mildly lame (ML) and non-lame (NL). They were then tested using each method. Kinematics showed that L sows had a lower walking speed than NL sows (L: 0.83 ± 0.04, NL: 0.96 ± 0.03 m/s; P < 0.05), a shorter stride length than ML sows (L: 93.0 ± 2.6, ML: 101.2 ± 1.5 cm; P < 0.05) and a longer stance time than ML and NL sows (L: 0.83 ± 0.03, ML: 0.70 ± 0.03, NL: 0.69 ± 0.02 s; P < 0.01). Accelerometer measurements revealed that L sows spent less time standing over a 24-h period (L: 6.3 ± 1.3, ML: 13.7 ± 2.4, NL: 14.5 ± 2.4%; P < 0.01), lay down earlier after feeding (L: 33.4 ± 4.6, ML: 41.7 ± 3.1, NL: 48.6 ± 2.9 min; P < 0.05) and tended to step more often during the hour following feeding (L: 10.1 ± 2.0, ML: 6.1 ± 0.5, NL: 5.4 ± 0.4 step/min standing; P = 0.06) than NL sows, with the ML sows having intermediate values. Visual observation of back posture showed that 64% of L sows had an arched back, compared with only 14% in NL sows (P = 0.02). Finally, footprint analysis and observation of lying-to-standing transition and foot lesions were not successful in detecting significant differences between L, ML and NL sows. In conclusion, several quantitative variables obtained from kinematics and accelerometers proved to be successful in identifying reliable indicators of lameness in sows. Further work is needed to relate these indicators with causes of lameness and to develop methods that can be implemented on the farm.

To reduce mortality among suckling piglets, lactating sows are traditionally housed in farrowing crates. Alternatively, lactating sows can be housed in farrowing pens where the sow is loose to ensure more behavioural freedom and consequently a better welfare for the sow, although under commercial conditions, farrowing pens have been associated with increased piglet mortality. Most suckling piglets that die do so within the first week of life, so potentially lactating sows do not have to be restrained during the entire lactation period. Therefore, the aim of the current study was to investigate whether confinement of the sow for a limited number of days after farrowing would affect piglet mortality. A total of 210 sows (Danish Landrace × Danish Yorkshire) were farrowed in specially designed swing-aside combination farrowing pens measuring 2.6 m × 1.8 m (combi-pen), where the sows could be kept loose or in a crate. The sows were either: (a) loose during the entire experimental period, (b) crated from days 0 to 4 postpartum, (c) crated from days 0 to 7 postpartum or (d) crated from introduction to the farrowing pen to day 7 postpartum. The sows and their subsequent litters were studied from introduction to the combi-pen ∼1 week before expected farrowing and until 10 days postpartum. Confinement period of the sow failed to affect the number of stillborn piglets; however, sows that were crated after farrowing had fewer live-born mortality deaths (P < 0.001) compared with the sows that were loose during the experimental period. The increased piglet mortality among the loose sows was because of higher mortality in the first 4 days after farrowing. In conclusion, the current study demonstrated that crating the sow for 4 days postpartum was sufficient to reduce piglet mortality.

In Chile, cow carcasses present the highest bruise prevalence compared with other cattle categories; however, the causes of the bruises are frequently unknown. In this study, 52 cull cows were transported to the slaughterhouse in three batches under identical transport conditions. A combination of direct observation and video analyses was used to determine moment, pre-slaughter stage and cause of potential bruising events during the period from loading on the farm until stunning at the slaughterhouse. After slaughter, number of bruises, location on the carcass and characteristics of the bruises were assessed. Seventy-eight bruises were observed on 37 carcasses. Fifty-two bruises were linked back to their causal event. Results showed that 46% of these bruises were a result of interactions between animal and facility, and most of them were inflicted in the stunning box, 27% of the bruises originated from animal–animal interactions and were mostly inflicted during lairage, another 27% was a result of human–animal interactions and were mostly inflicted during loading and unloading of animals. The percentages of potential bruising events resulting in a bruise were 43%, 9% and less than 1% for animal–facility, human–animal and animal–animal interactions, respectively. Most bruises on the back site were inflicted when the animal was in the stunning box (91.2%), whereas bruises on the pin site were mostly (75%) inflicted during loading at the farm. One may conclude that in relative short journeys (⩽4 h) directly from farm to the slaughterhouse and long lairage times (>12 h), most bruises are the result of circumstances at the slaughterhouse. A substantial amount of these bruises could be avoided by proper animal handling and adequate stunning facilities.

In 1991, the Committee on Medical Aspects of Food Policy produced a report on the dietary reference values for food energy and nutrients for groups of people in the United Kingdom. The resulting recommendations, which included specific limits for intakes of total, saturated, trans- and cis-polyunsaturated fatty acids (PUFA) have remained a cornerstone of public health policy ever since, and similar recommendations have been adopted by the World Health Organization. These recommendations were made largely on the basis of specific effects of these fatty acids on the risk of developing atherosclerotic cardiovascular disease (CVD). The intervening years have seen a plethora of human epidemiological and intervention trials to further elucidate the specific relationship between dietary fatty acid intake, plasma lipids and lipoproteins and cardiovascular morbidity and mortality. A number of recent meta-analyses and systematic reviews have revisited the role of specific dietary fatty acid classes and CVD risk. In general, these continue to support a link between saturated fatty acids (SFA) and CVD morbidity/mortality. They also highlight the potent adverse effects of trans fatty acids derived from partially hydrogenated vegetable oil. The most recent data suggest that replacing SFA with cis-PUFA (primarily linoleic acid) has the greatest impact on reducing CVD risk. Evidence of specific beneficial effects of n-3 PUFA is generally stronger for secondary, rather than primary, CVD risk, and it is restricted to very long chain fatty acids of marine origin as opposed to alpha-linolenic acid. Taken together, these data suggest that recent focus on dietary n-6-to-n-3 PUFA ratios may have been misguided, and that future strategies should focus on replacing dietary SFA with total PUFA, rather than concentrating on n-6 : n-3 PUFA ratio.

The growth dispersion of farmed fish is a subject of increasing interest and one of the most important factors in stocking density. On a duration of 60 days, the effect of stocking density on the growth, coefficient of variation and inter-individual variation of feed intake (CVFI) of juvenile Nile tilapia Oreochromis niloticus L. (14.9 ± 1.2 g) were studied in an experimental tank-based flow-through system. Groups of fish were stocked at four stocking densities: 200, 400, 600 and 800 fish/m3, corresponding to a density of ∼3, 6, 9 and 12 kg/m3 and referred to as D1, D2, D3 and D4, respectively. Each treatment was applied to triplicate groups in a completely randomized design. No treatment-related mortality was observed. The fish densities increased throughout the experiment from 3 to 23.5, 6 to 43.6, 9 to 56.6 and 12 to 69 kg/m3. Results show that mass gain and specific growth rate (SGR, %M/day) were negatively correlated with increased stocking density. Groups of the D1 treatment reached a mean final body mass (FBM) of 119.3 g v. 88.9 g for the D4 groups. Feed conversion ratios (FCRs) were 1.38, 1.54, 1.62 and 1.91 at D1, D2, D3 and D4 treatments, respectively. Growth heterogeneity, expressed by the inter-individual variations of fish mass (CVM), was significantly affected by time (P < 0.001), stocking density (P < 0.001) and their interaction (P < 0.05). The difference in CVM was particularly conspicuous towards the end of the experiment and was positively correlated with stocking density. Similarly, radiographic study shows that CVFI was also found to be significantly greater for groups reared at high stocking densities (D3 and D4) than the other treatments (D1 and D2). These differences in both CVM and CVFI related to the stocking density need to be taken into account by husbandry practices to assure the production of more homogeneous fish size. A simple economic analysis indicates a parabolic relationship between profit and density with optimal final density at the peak of the curve. Given reasonable assumptions about production costs, the optimal final density (Dopt) is 73.7 kg/m3. A sensitivity analysis shows that changes in the fixed cost have no effects on the optimal final density. However, small change in variable costs, such as feed and juvenile costs, may have substantial effect on the optimal density.

Feed intake behavior was studied between 9 weeks of age and slaughter in a total of 36 gilts, 32 immunocastrates, 33 surgically castrated barrows and 33 boars from 36 litters. Consequences for the concentration of substances contributing to off odor of pork (skatole, indole) were evaluated. Animals were kept in groups of 12 pigs of the same sex and treatment and fed ad libitum (13.4 MJ ME, 17% CP, 1.1% lysine). Individual feed intake behavior was recorded continuously by an electronic feeder. Immunocastration was carried out with two injections of Improvac with at least 4 weeks between both injections (1st: 12 to 17 weeks of age, 2nd: 19 to 21 weeks of age). Feed intake/day increased from an average of 0.91 ± 0.02 kg/day up to 3.15 ± 0.04 kg/day before slaughter. This increase was associated with a 50% reduction in the number of meals/day (from 15.8 ± 0.44 to 7.2 ± 0.29 meals/day). The larger meal sizes resulted from an increase in both, the duration of feed intake/meal and the feed intake rate (g/min). In addition, sex and treatment differences were observed: Feed intake in boars was lower than in all other groups due to a reduction in the number of meals/day and in the time spent feeding/day. In females, time spent feeding/day was quite similar to boars, but resulted from a higher number of meals of shorter duration. Barrows had a significantly higher feed intake because of a higher number of meals/day resulting in more time spent feeding/day. The feed intake rate was similar in boars, gilts and barrows and showed an increasing trend during the study, starting from about 15 g/min up to four times the amount. Immunocastration affected feed intake behavior severely, especially the meal size increased dramatically because of higher feed intake rate, which exceeded that of all other groups by 25% at the end of the study. The number of meals/day was not influenced by immunocastration and was almost identical to that of boars. Highest skatole concentrations were measured in fat of boars, whereas indole concentrations were higher in immunocastrates than in all other groups. In gilts and barrows, skatole concentrations were related to growth rate. Additionally, the feeding rate was an important factor explaining the variability in skatole/indole concentrations in adipose tissue. The physiological mechanisms however need further clarification.

Mounted police horses have to cope with challenging, unpredictable situations when on duty and it is essential to gain insight into how these horses handle stress to warrant their welfare. The aim of the study was to evaluate physiological and behavioral responses of 12 (six experienced and six inexperienced) police horses during police training. Horses were evaluated during four test settings at three time points over a 7-week period: outdoor track test, street track test, indoor arena test and smoke machine test. Heart rate (HR; beats/min), HR variability (HRV; root means square of successive differences; ms), behavior score (BS; scores 0 to 5) and standard police performance score (PPS; scores 1 to 0) were obtained per test. All data were statistically evaluated using a linear mixed model (Akaike's Information criterium; t > 2.00) or logistic regression (P < 0.05). HR of horses was increased at indoor arena test (98 ± 26) and smoke machine test (107 ± 25) compared with outdoor track (80 ± 12, t = 2.83 and t = 3.91, respectively) and street track tests (81 ± 14, t = 2.48 and t = 3.52, respectively). HRV of horses at the indoor arena test (42.4 ± 50.2) was significantly lower compared with street track test (85.7 ± 94.3 and t = 2.78). BS did not show significant differences between tests and HR of horses was not always correlated with the observed moderate behavioral responses. HR, HRV, PPS and BS did not differ between repetition of tests and there were no significant differences in any of the four tests between experienced and inexperienced horses. No habituation occurred during the test weeks, and experience as a police horse does not seem to be a key factor in how these horses handle stress. All horses showed only modest behavioral responses, and HR may provide complimentary information for individual evaluation and welfare assessment of these horses. Overall, little evidence of stress was observed during these police training tests. As three of these tests (excluding the indoor arena test) reflect normal police work, it is suggested that this kind of police work is not significantly stressful for horses and will have no negative impact on the horse's welfare.

Dehydration can be extremely damaging to the performance and welfare of indigenous chickens. The effect of water restriction on haematological and biochemical parameters was compared in Naked Neck (NNK) and Ovambo (OVB) chickens. A total of 54 8-week-old pullets each of NNK and OVB chickens with an initial average weight of 641 ± 10 g/bird were randomly assigned to three water intake treatments with three replications, each having six birds. The water restriction treatments were ad libitum, 70% and 40% of ad libitum intake. Nine experimental pens with a floor space of 3.3 m2 per strain were used. Feed was provided ad libitum. Packed cell volume (PCV), erythrocyte count (RBC), mean corpuscular volume (MCV) and total leucocyte count (WBC), and biochemical parameters (uric acid (UA)), creatinine (CREAT), total protein (TP), albumin (ALB), globulin (GLOB), triglyceride (TGA), total cholesterol (TC), high- (HDLC) and low- (LDLC) density lipoprotein cholesterol and activity of alanine transaminase (ALT), alkaline phosphatase (ALP) and aspartate transaminase (AST) were determined from blood collected after 60 days of water restriction. PCV was higher (P < 0.05) in NNK than OVB chickens offered water ad libitum, but similar in birds offered 70% and 40% of ad libitum. There were no differences in RBC and MCV values between strains, but MCV was higher in birds on 40% than 70% of ad libitum water intake, irrespective of strain. Naked neck chickens had higher (P < 0.05) WBC values than OVB at 40% restriction level, but lower WBC than OVB at 70% water restriction level. UA, CREAT, TGA, TC, LDLC, TP and GLOB increased (P < 0.05) with each increment in water restriction, but the increase in CREAT and TC was more pronounced in OVB than NNK chickens. The opposite was observed for UA. ALT activity indicated that liver function was not affected by water restriction. It was concluded that the two strains can withstand up to 40% of ad libitum water restriction, but NNK tolerated water stress better than OVB chickens.

Data from 113 Dutch organic farms were analysed to determine the effect of cross-breeding on production and functional traits. In total, data on 33 788 lactations between January 2003 and February 2009 from 15 015 cows were available. Holstein–Friesian pure-bred cows produced most kg of milk in 305 days, but with the lowest percentages of fat and protein of all pure-bred cows in the data set. Cross-breeding Holstein dairy cows with other breeds (Brown Swiss, Dutch Friesian, Groningen White Headed, Jersey, Meuse Rhine Yssel, Montbéliarde or Fleckvieh) decreased milk production, but improved fertility and udder health in most cross-bred animals. In most breeds, heterosis had a significant effect (P < 0.05) on milk (kg in 305 days), fat and protein-corrected milk production (kg in 305 days) and calving interval (CI) in the favourable direction (i.e. more milk, shorter CI), but unfavourably for somatic cell count (higher cell count). Recombination was unfavourable for the milk production traits, but favourable for the functional traits (fertility and udder health). Farm characteristics, like soil type or housing system, affected the regression coefficients on breed components significantly. The effect of the Holstein breed on milk yield was twice as large in cubicle housing as in other housing systems. Jerseys had a negative effect on fertility only on farms on sandy soils. Hence, breed effects differ across farming systems in the organic farming and farmers can use such information to dovetail their farming system with the type of cow they use.

The present study was designed to measure the use of various, simultaneously available resources in a complex housing environment in juvenile blue foxes. Twelve blue fox sibling (male–female) pairs were housed in two-section experimental cages from the age of 8 weeks until the age of 7 months (from June to December). Each experimental cage was furnished with two platforms, a nest box, a sand box and a wooden block. This housing set-up provided the foxes with social contact, and an opportunity for oral manipulation, scratching and nesting, as well as the choice of staying on a solid floor material or on an elevated location. The foxes’ behaviour was recorded at three time points during autumn (September, November and December). The foxes used all available resources. The most utilised resource was the nest box, possibly because it could be utilised in several ways (as a shelter, an elevated location, an object for scratching and for oral manipulation). The foxes also stayed more in the cage section containing the nest box than in the cage section containing a sand box. The foxes rested much on the cage floor, but they also used the interior of the nest box and elevated locations for resting. Social contact often occurred during resting. Thus, the nest box and elevated location, in conjunction with social contact seem to be valuable while resting. While active, the foxes utilised the cage floor and roof of the nest box instead of the platforms. Scratching, digging and an interaction with the wooden block were seldom observed. Activity occurred mainly on the ‘empty’ cage area. In conclusion, all studied resources provided blue foxes with a distinct value, as they all were used in the complex housing environment. The nest box is used most and for most variable behaviours.

The effects of feeding total mixed ration (TMR) or pasture forage from a perennial sward under a management intensive grazing (MIG) regimen on grain intake and enteric methane (EM) emission were measured using chambers. Chamber measurement of EM was compared with that of SF6 employed both within chamber and when cows grazed in the field. The impacts of the diet on farm gate greenhouse gas (GHG) emission were also postulated using the results of existing life cycle assessments. Emission of EM was measured in gas collection chambers in Spring and Fall. In Spring, pasture forage fiber quality was higher than that of the silage used in the TMR (47.5% v. 56.3% NDF; 24.3% v. 37.9% ADF). Higher forage quality from MIG subsequently resulted in 25% less grain use relative to TMR (0.24 v. 0.32 kg dry matter/kg milk) for MIG compared with TMR. The Fall forage fiber quality was still better, but the higher quality of MIG pasture was not as pronounced as that in Spring. Neither yield of fat-corrected milk (FCM) which averaged 28.3 kg/day, nor EM emission which averaged 18.9 g/kg dry matter intake (DMI) were significantly affected by diet in Spring. However, in the Fall, FCM from MIG (21.3 kg/day) was significantly lower than that from TMR (23.4 kg/day). Despite the differences in FCM yield, in terms of EM emission that averaged 21.9 g/kg DMI was not significantly different between the diets. In this study, grain requirement, but not EM, was a distinguishing feature of pasture and confinement systems. Considering the increased predicted GHG emissions arising from the production and use of grain needed to boost milk yield in confinement systems, EM intensity alone is a poor predictor of the potential impact of a dairy system on climate forcing.

The high pre-weaning mortality in farm animal species and poor welfare conditions of reproductive females question modern industrial farming acceptability. A growing body of literature has been produced recently, investigating the impact of maternal stress during gestation on maternal and offspring physiology and behavior in farm animals. Until now, the possible impact of prenatal stress on neonatal health, growth and survival could not be consistently demonstrated, probably because experimental studies use small numbers of animals and thus do not allow accurate estimations. However, the data from literature synthesized in the present review show that in ungulates, maternal stress can sometimes alter important maternal parameters of neonatal survival such as colostrum production (ruminants) and maternal care to the newborn (pigs). Furthermore, maternal stress during gestation can affect maternal immune system and impair her health, which can have an impact on the transfer of pathogens from the mother to her fetus or neonate. Finally, prenatal stress can decrease the ability of the neonate to absorb colostral immunoglobulins, and alter its inflammatory response and lymphocyte functions during the first few weeks of life. Cortisol and reproductive hormones in the case of colostrogenesis are pointed out as possible hormonal mediators. Field data and epidemiological studies are needed to quantify the role of maternal welfare problems in neonatal health and survival.

The aim of this study was to assess unconsciousness in pigs during and after the exposure to gas mixtures of 70% nitrogen (N2) and 30% carbon dioxide (CO2) (70N30C), 80% N2 and 20% CO2 (80N20C) and 85% N2 and 15% CO2 (85N15C) compared with 90% CO2 in air (90C) by means of the Index of Consciousness®(IoC), their behaviour and the absence of brain stem reflexes. The experiment included three trials of 24 pigs divided into four groups according to the number of treatments. Half of the group was exposed for a short time and the other half for a long time (3 and 5 min for the N2/CO2 mixtures exposure and 2 and 3 min in 90C exposure, respectively). During exposure, the IoC and the electroencephalography suppression rate (ESR) were assessed, as well as the time to onset and percentage of gasping, loss of balance, vocalizations, muscular excitation and gagging. At the end of the exposure, the corneal reflex, rhythmic breathing and sensitivity to pain were each assessed at 10 s intervals for 5 min. Brain activity decreased significantly (P < 0.05) 37.60 s after the start of the exposure to 90% CO2, which was significantly earlier than in 70N30C, 80N20C and 85N15C exposure, (45.18 s, 46.92 s and 43.27 s, respectively). Before brain activity decreased, all pigs experienced gasping and loss of balance and a 98% muscular excitation. The duration of the muscular excitation was longer in animals exposed to 70N30C, 80N20C and 85N15C than 90C (P < 0.01). After a long exposure time, all animals exposed to 90C died, whereas the 30.4% of animals exposed to N2/CO2 gas mixtures survived. Pigs exposed to 85N15C recovered corneal reflex and sensitivity to pain significantly earlier than when exposed to 90C. Exposure to 90C causes a higher aversive reaction but a quicker loss of consciousness than N2/CO2 gas mixtures. Exposure to N2/CO2 gas mixtures causes a lower percentage of deaths and an earlier recovery of the brain stem activity than 90C, whereas the time to recover the cortical activity is similar. In conclusion, the inhalation of N2/CO2 gas mixtures reduces the aversion compared with high concentrations of CO2; however, the period of exposure for inducing unconsciousness may be longer in N2/CO2 gas mixtures, and the signs of recovery appear earlier, compared to CO2.

Lameness in sows is an emerging disease condition with major effects on animal welfare and economics. Yet the direct impact on reproduction results remains unclear. The present field study investigated the impact of lameness and claw lesions throughout the reproductive cycle on (re)production results of sows. In five farms, a total of 491 group-housed sows were followed up for a period of one reproductive cycle. Sows were assessed for lameness every time they were moved to another area in the farm. Claw lesions were scored at the beginning and at the end of the cycle. Reproduction results included the number of live-born piglets, stillborn piglets, mummified fetuses and crushed piglets, weaning-to-oestrus interval and the presence of sows not showing oestrus post weaning, returning to service and aborting. Sows that left the group were recorded and the reason was noted. A mean prevalence of lameness of 5.9% was found, although it depended on the time in the productive cycle. The highest percentage of lame sows (8.1%) was found when sows were moved from the post-weaning to the gestation stable. No significant associations were found between lameness and reproduction parameters with the exception of the effect on mummified foetuses. Wall cracks, white line lesions, heel lesions and skin lesions did have an effect on farrowing performance. Of all sows, 22% left the group throughout the study, and almost half of these sows were removed from the farm. Lameness was the second most important reason for culling. Sows culled because of lameness were significantly younger compared with sows culled for other reasons (parity: 2.6 ± 1.3v. 4.0 ± 1.8). In conclusion, the present results indicate that lameness mainly affects farm productivity indirectly through its effect on sow longevity, whereas claw lesions directly affect some reproductive parameters. The high percentage of lame sows in the insemination stable indicate that risk factor studies should not only focus on the gestation stable, but also on housing conditions in the insemination stable.

This study was designed to determine whether methyl-β-cyclodextrin (MCD) can substitute for albumin in incubation medium for neonatal swine adipose tissue explants, or whether MCD affects metabolism and cytokine expression. Subcutaneous adipose tissue explants (100 ± 10 mg) were prepared from 21-day-old pigs. Explants were incubated in medium 199 supplemented with 25 mM HEPES, 1.0 nM insulin at 37°C. The medium also contained bovine serum albumin (BSA) or MCD at 0%, 0.05%, 0.1%, 0.2% or 0.3%. Tissue explants were treated with these media for 1 h and then switched to the same basal incubation medium containing 0.05% BSA. Explants were removed from basal medium at 2 or 8 h of incubation, and real-time PCR was performed to assess expression of tumor necrosis α (TNF) and interleukin 6 (IL6), acetyl CoA carboxylase (ACAC) and fatty acid synthase (FASN). Alternatively, rates of 14C-glucose oxidation and lipogenesis were monitored ± insulin (100 nM), following MCD treatment. Incubation with BSA had minimal effects on gene expression or adipose tissue metabolism, only producing a doubling in TNF mRNA abundance (P < 0.01). Treatment with MCD increased TNF mRNA abundance by eightfold (P < 0.009), whereas IL6 gene expression increased a 100-fold (P < 0.001) with a suppression in ACAC and FASN expression (P < 0.01). This was paralleled by MCD inhibition of insulin-stimulated glucose oxidation and lipogenesis (P < 0.001). Addition of a TNF antibody to the incubation medium alleviated this inhibition of insulin-stimulated glucose metabolism by ∼30% (P < 0.05).

The objective of this study was to compare some husbandry procedures on the base of physiological stress parameters and evaluate the welfare status in sheep. Forty ewes were used as the study material. Measurements were taken during several routine husbandry procedures such as milking, shearing, weighing, loading and hoof care. Data regarding time spent for each application, as well as heart and respiratory rates were recorded during the applications. Blood samples were taken 15 min before and after each application and malondialdehyde (MDA), glutathione-peroxidase (GSH-Px), cortisol T3 and T4 parameters were measured. In addition, changes in the same parameters between pre- and post-application periods were evaluated. According to the results, machine milking caused less stress than hand milking. No significant difference was seen between shearing methods for hand shearer or clipper; however, both applications caused stress in animals. The results for weighing methods of animals demonstrated significant differences in cortisol, T3 and T4 values in favor of traditional method. Cortisol, T3 and T4 levels were significantly higher in manual loading compared with loading by ramp. Regarding hoof care, all the examined parameters differed in favor of modern method. On the other hand, significant differences were determined between the stress parameters regarding pre- and post-applications. All values differed for hand milking while no significant difference was observed in MDA and T3 values in machine milking group. Parameters in weighing groups changed significantly. For loading process, GSH, cortisol, T3 and T4 values differed in both treatment groups. With regard to hoof care, parameters except T4 in laying group differed significantly. An increase occurred in minute-based measurements of heart and respiratory rates parallel to physiological data. The number of the respiratory rates during the applications differed except for the shearing process. All the parameters displayed significant differences between groups in terms of heart rates. Time spent for each application also differed between groups. Time saved for milking, shearing, weighing, loading and hoof care was 3.23 min, 4.37 min, 1.71 min, 7.85 s and 1.55 min, respectively. These results appear to provide a tangible advantage of using new husbandry methods to the breeders. It was concluded that using new methods in sheep husbandry procedures provided advantages in terms of saving time and reducing labor, as well as improved conditions for welfare of animals. In addition, it facilitated the routine works and flock husbandry.

The recently sequenced cattle (Bos taurus) genome unraveled the unique genomic features of the species and provided the molecular basis for applying a systemic approach to systematically link genomic information to metabolic traits. Comparative analysis has identified a variety of evolutionary adaptive features in the cattle genome, such as an expansion of the gene families related to the rumen function, large number of chromosomal rearrangements affecting regulation of genes for lactation, and chromosomal rearrangements that are associated with segmental duplications and copy number variations. Metabolic reconstruction of the cattle genome has revealed that core metabolic pathways are highly conserved among mammals although five metabolic genes are deleted or highly diverged and seven metabolic genes are present in duplicate in the cattle genome compared to their human counter parts. The evolutionary loss and gain of metabolic genes in the cattle genome may reflect metabolic adaptations of cattle. Metabolic reconstruction also provides a platform for better understanding of metabolic regulation in cattle and ruminants. A substantial body of transcriptomics data from dairy and beef cattle under different nutritional management and across different stages of growth and lactation are already available and will aid in linking the genome with metabolism and nutritional physiology of cattle. Application of cattle genomics has great potential for future development of nutritional strategies to improve efficiency and sustainability of beef and milk production. One of the biggest challenges is to integrate genomic and phenotypic data and interpret them in a biological and practical platform. Systems biology, a holistic and systemic approach, will be very useful in overcoming this challenge.

The nature of human–animal interactions is an important factor contributing to animal welfare and productivity. Reducing stress during routine husbandry procedures is likely to improve animal welfare. We examined how the type of early handling of calves affected responses to two common husbandry procedures, ear-tagging and disbudding. Forty Holstein–Friesian calves (n = 20/treatment) were exposed to one of two handling treatments daily from 1 to 5 weeks of age: (1) positive (n = 20), involving gentle handling (soft voices, slow movements, patting), and (2) negative (n = 20), involving rough handling (rough voices, rapid movements, pushing). Heart rate (HR), respiration rate (RR) and behaviour (activity, tail flicking) were measured before and after ear-tagging and disbudding (2 days apart). Cortisol was measured at −20 (baseline), 20 and 40 min relative to disbudding time. There were no significant treatment differences in HR, RR or behaviour in response to either procedure. However, the following changes occurred across both treatment groups. HR increased after disbudding (by 14.7 ± 4.0 and 18.6 ± 3.8 bpm, positive and negative, respectively; mean ± s.e.m.) and ear-tagging (by 8.7 ± 3.1 and 10.3 ± 3.0 bpm, positive and negative, respectively). After disbudding, there was an increase in RR (by 8.2 ± 3.4 and 9.3 ± 3.4 breaths/min, positive and negative, respectively), overall activity (by 9.4 ± 1.2 and 9.9 ± 1.3 frequency/min, positive and negative, respectively) and tail flicking (by 13.2 ± 2.8 and 11.2 ± 3.0 frequency/min, positive and negative, respectively), and cortisol increased from baseline at 20 min post procedure (by 10.3 ± 1.1 and 12.3 ± 1.1 nmol/l positive and negative, respectively). Although we recorded significant changes in calf responses during ear-tagging and disbudding, the type of prior handling had no effect on responses. The effects of handling may have been overridden by the degree of pain and/or stress associated with the procedures. Further research is warranted to understand the welfare impact and interaction between previous handling and responses to husbandry procedures.
