Studies on the phenology of harbour seal moult have been carried out in the Atlantic and Pacific, however there has been no research into this process in the Republic of Ireland, at the southern edge of the species range in the north-east Atlantic. Population estimates of harbour seals are derived by counts primarily during the moulting seasons. In the absence of information on the moult phenology planning the optimal timing of such surveys is impossible. Furthermore, changes in moult phenology may reflect changes in resource availability or competition, or demographic changes. The phenology of the harbour seal moult was investigated in south-west Ireland in this study. Timing of the moult differed among all cohorts, yearlings began moulting first followed by adult females and finally adult males. The number of seals hauled out was generally positively related to the proportion of seals in active moult. The timing of the moult period was different to other parts of the species' range and should be considered in determining optimal timing of future surveys for assessing populations abundance and trends in Ireland.

This is the first study to examine the tolerance of Antarctic springtails (Collembola) to ultraviolet radiation (UV). Survival of extended attenuated exposure to sunlight was examined for both individuals and aggregations of the species Gomphiocephalus hodgsoni Carpenter over a 10 day period. Both individuals and aggregations demonstrated significantly higher survival and moult rates from control treatments kept in the dark to those exposed to UV. A photo-inhibitive element to moulting is indicated that may function to protect post-ecdysial springtails when their emergent cuticles are more sensitive to the external environment. DNA damage was measured in springtails directly exposed to sunlight for 5 h on a clear sunny day. Significant differences were found between treated animals and controls kept in the dark. There was some reduction of damage 12 and 24 h after exposure, when springtails had been placed in the dark to recover. This indicates the up-regulation of DNA repair mechanisms, with the 12 h treatment in particular showing no significant difference with controls. In addition to providing a first look at UV tolerance in these soil arthropods, these findings recommend employing strict protocols for collections of sample material for subsequent biological analysis in order to minimize the interactive effects of photo-damage.

Moulting is a complex process, induced by diet manipulation, which results in renewal of reproductive system and subsequent better production. Conventionally, moulting has been practised by utilising feed withdrawal in many developing countries, however, this has been associated with many drawbacks. Alternative types of moulting practices have been studied, however, these are not practised widely due to a range of undesirable effects. As a consequence of the beneficial effects on laying hens, better post-moult performance and a less criticised method according to poultry welfare advocates, Zn-induced moulting has been considered superior to forced-moulting. Different aspects of Zn-induced moulting related to physiology and production, with special reference to immunohistochemistry of pituitary gland as affected by Zn-induce moulting are included in this review.

Moulting is a natural phenomenon in birds. Temperature variation and photoperiodic changes induce circadian and circannual rhythms, manifested by changes in the neural and endocrine systems, ultimately causing physiological processes resulting in a moult. The concept of induced moulting by feed deprivation arose during the early part of the 20th century as a solution to economic challenges of decreased egg production in commercial layer flocks toward the end of the first laying cycle as they approached the moult phase. Efficiency of feed removal programs in enhancing post moult productivity and economical feasibility insured a gain in popularity by the 1930's. In 2003, more than 75% of all flocks were moulted in the U.S. However, moulting with feed withdrawal caused risk to the health of birds by impairing the immune system and increasing susceptibility to various pathogens, especially Salmonella enteritidis (SE). During the 1980s, increased incidence of gastroenteritis due to SE was linked to forced moulting. The importance of challenges for conventional forced moulting was realized and animal activists, welfare organizations, researchers, consumers, and producers realized that alternative moulting methods were necessary. Ultimately, the United Egg Producers Scientific Advisory Committee, having dealt with the issue of feed withdrawal for moulting since the early 1990's, amended the Animal Husbandry Guidelines for U.S. Egg Laying Flocks to specify the use of non feed withdrawal moult methods after January 1, 2006. Furthermore, moulting hens would have to be fed nutritionally adequate and palatable food suitable for non-egg producing hens. Though metals or hormones are efficient moult inducers, results of studies showed that they had disadvantages like residual effects and physiological alterations, respectively. Diets supplemented with low nutritious feed ingredients are safe and demonstrate promising outcomes. Unconventional feedstuffs such as coconut meal and cumin seed meal are successfully used in layer and non feed removal moults in Asian and Middle Eastern countries. In the western U.S. (especially in California), local ingredients like safflower meal and tomato pomace are promising alternatives.

Capture by water surfaces is a frequent but stochastic phenomenon for Antarctic collembolans (springtails), with implications for both the survival and dispersal of populations. This paper provides the first field description of the characteristics of rafting aggregations of the springtail Gomphiocephalus hodgsoni and provides the first observation of the inclusion of the mite, Stereotydeus mollis, within these aggregations. Field observations of aggregations in meltwater pools are used to develop a picture of the ecological implications of rafting for Antarctic springtails. The potential for marine capture was also demonstrated by additional observations of two springtails aerially deposited on to sea ice as well as springtails floating in the tidewater cracks between the sea ice and land.

Nematodes represent one of the most abundant and species-rich groups of animals on the planet, with parasitic species causing chronic, debilitating infections in both livestock and humans worldwide. The prevalence and success of the nematodes is a direct consequence of the exceptionally protective properties of their cuticle. The synthesis of this cuticle is a complex multi-step process, which is repeated 4 times from hatchling to adult and has been investigated in detail in the free-living nematode, Caenorhabditis elegans. This process is known as moulting and involves numerous enzymes in the synthesis and degradation of the collagenous matrix. The nas-36 and nas-37 genes in C. elegans encode functionally conserved enzymes of the astacin metalloprotease family which, when mutated, result in a phenotype associated with the late-stage moulting defects, namely the inability to remove the preceding cuticle. Extensive genome searches in the gastrointestinal nematode of sheep, Haemonchus contortus, and in the filarial nematode of humans, Brugia malayi, identified NAS-36 but not NAS-37 homologues†. Significantly, the nas-36 gene from B. malayi could successfully complement the moult defects associated with C. elegans nas-36, nas-37 and nas-36/nas-37 double mutants, suggesting a conserved function for NAS-36 between these diverse nematode species. This conservation between species was further indicated when the recombinant enzymes demonstrated a similar range of inhibitable metalloprotease activities.

Various catecholamines and catecholamine antagonists have been examined for their effects on the third larval moult of the parasitic nematode, Dirofilaria immitis, cultured in vitro. The non-selective α and β agonist, noradrenaline, and the β agonist, isoprenaline, had no effect on the timing of the third stage moult when used at a concentration of 10−5M. The α-adrenergic antagonist. phentolamine, resulted in worm mortality at 10−5M. At 10−7M, both phentolamine and the β-antagonist, propranolol caused a significant reduction in the numbers of larvae capable of completing the third stage moult. Idazoxan, an a2-antagonist, at 10−5M did not affect worm mortality but did completely prevent ecdysis. The potential of these compounds as possible filaricides is discussed.

The effects of the insect hormones, ecdysone and 20-hydroxyecdysone, certain non-steroidal ecdysteroid agonists (RH compounds) and the inhibitor, azadirachtin, on the timing of the 3rd-stage moult of Dirofilaria immitis were investigated. 20-Hydroxyecdysone and RH 5849 when used at a concentration of 10−5 M, resulted in a premature timing of this moult. Azadiracthin, at a similar concentration, prevented moulting of most of the larvae to the 4th stage. The results are discussed in relation to the possibility of a hormonal role for ecdysteroids and neuropeptide-like compounds in the control of ecdysis in filarial nematodes, that maybe somewhat comparable to the system which is found in insects.

Phase-interference microscopic examination of the infective, post-infective 3rd-stage and 4th-stage larvae of Dirofilaria immitis has identified a single cell body in each of the paired lateral amphidial nerves which undergoes characteristic morphological change during the development from 3rd- to 4th-stage larvae. Acetaldehyde—fuchsin staining of worm sections revealed fuchsinophilic material in the precise location of the amphidial nerve-cell bodies observed by phaseinterference microscopy. This material was found in 70% of infective larvae recovered from mosquitoes and in 100% of larvae recovered from micropore chambers 24 h after implantation into BALB/C mice. In pre-moult larvae recovered at 42 h (Experiment 1) and at 48 h (Experiment 2) fuchsinophilic material was demonstrable, but no staining was observed in those larvae in which separation of the 3rd- and 4th-stage cuticles had occurred. No such material was observed in 4thstage larvae recovered from chambers after 74 h, and in these larvae the amphidial nerve-cell bodies were not discernible. These cytological observations are consistent with a cycle of elaboration and release of neurosecretion associated with moulting in D. immitis.

Moulting is a natural phenomenon in birds during which they replace old plumage with new feathers, reduce feed intake, lose body weight and suspend reproduction. Moulting in laying hens can be induced by using photoperiods, feed deprivation or restriction or diets containing minerals or variable amounts of other ingredients. Induced moulting can result in higher egg production and improved quality. It reduces mortality, production costs and investments in new farms and hatcheries. While feed withdrawal has been a most effective way to induce moult in poultry birds, it is illegal on welfare grounds in the UK and Europe. This may have implications for the global poultry industry. Therefore, efforts to find a non feed removal method as a desirable alternative to induce moulting in poultry birds are underway in different countries. This paper reviews the history of induced moulting and its future implications by examining different methods that have been tested in the past and their potential to become a more acceptable alternative to feed removal method of inducing moulting in laying hens.

Sodium fluoride (NaF) is an anion that has been previously shown to block the moulting process of Ascaris suum larvae. This study describes moulting and development-specific protein expression profiles of A. suum lung-stage L3 (AsLL3) following NaF exposure. AsLL3s cultured in the presence or absence of NaF were prepared for protein analysis using two-dimensional (2D) electrophoresis. NaF exposure inhibited at least 22 proteins in AsLL3 compared with moulted larvae (i.e. AsLL4). A further comparison of AsLL4 with those of pre-cultured AsLL3 and NaF-exposed AsLL3 revealed 8 stage-specifically and 4 over-expressed proteins. Immunoblot analysis revealed an inhibition by NaF of 19 immunoreactive proteins. Enzyme assay and immunochemical data showed an inhibition of the moulting-specific inorganic pyrophosphatase activity by 41% and a decreased expression in NaF-treated larvae, indicating its significance in the moulting process. A protein spot associated with NaF inhibition was isolated and identified by peptide mass spectrometry and bioinformatics approaches to be a member of 3–hydroxyacyl–CoA dehydrogenase/short-chain dehydrogenase enzyme families. These results have implications for the identification of proteins specific to the moulting process as potential chemotherapeutic targets.

The ovulatory cycle and moulting are associated with physiological and endocrinological mechanisms involving a complex interplay of light stimulus, hypothalamus, pituitary, gonads, thyroid and adrenal glands. During ovulation, the LH surge triggers the release of a mature follicle. With advance in the age of a laying hen, egg production decreases coinciding with decrease in ovarian steroids and gonadotropins. Moulting occurs when oestrogen, progesterone and luteinizing hormone are low while thyroid hormones and corticosterone are high. The physiological processes observed during induced moulting are common to those found in natural moulting. Moulting results in the rejuvenation of reproductive tissues of old laying hens, preparing them for another laying cycle. Vitellogenin provides the two proteins (lipovitellin and phosvitin) to the developing yolk, at the same time serving as their transport protein through the blood stream. Low levels of vitellogenin in the plasma of moulting hens are attributable to decrease in oestrogen during moulting since production of vitellogenin from the liver is stimulated by oestrogen.

This is the first demonstration of the in vitro development of the 3rd-stage larvae (L3) of Hysterothylacium aduncum to the adult. This was achieved in a semi-defined medium that is easy to prepare and to reproduce. The L3, collected from the peritoneal cavity of horse mackerel (Trachurus trachurus), were individually inoculated into RPMI-1640 medium +20% heat-inactivated fetal bovine serum (IFBS). It has been demonstrated that the optimum temperature for development is around 13 °C and is stimulated by the presence of 5% CO2 in the growth atmosphere, increasing the percentage moulting to the 4th larval stage (L4) by 1.9-fold (from 44 to 82%) and the average survival of the nematodes by 1.6 times (from 60 to 96 days). When the larvae were grown at different pHs, optimum development occurred at pH 4.0. Under these conditions, all the larvae moulted to the L4 and more than two-thirds transformed to the adult stage – in which 25–30% of the females laid eggs – and reached an average survival of over 4 months. When this medium was supplemented with 1% (w/v) of commercial pepsin, all the larvae reached the adult stage, at least 45% of the females oviposited, laying around 12-fold more eggs per female than in the medium without pepsin. The mean size of the eggs (non-fertilized) obtained was 56.8×47.6 μm. The mean length of the adult males obtained was between 3.2 and 5.2 cm and the females were between 3.0 and 6.5 cm. The adult specimens were morphologically identified as Hysterothylacium aduncum aduncum. This culture medium (RPMI-1640+20% (v/v) IFBS+1% commercial pepsin, at pH 4.0, 13 °C and 5% CO2 in air) could facilitate the identification of at least some of the larvae of the genus Hysterothylacium – and perhaps other anisakids – for which the specific identification and the biological study of these parasites is often difficult.

Spodoptera litura (F.) (Lepidoptera: Noctuidae) is a serious pest of several crops in India and other countries. Prothoracicotropic hormone (PTTH) release from the brain of penultimate fifth instars of the noctuid was followed in the laboratory and a spiracle index developed to assess PTTH action. PTTH release from the brain occurred during the scotophase at 4.30 h of day 1 (AZT). Ecdysone secreted in the prothoracic gland was released from the 7th hour of day 1 (AZT), reaching a maximum in the haemolymph on the 9th hour of day 1 (AZT). The prothoracic gland showed three distinct phases, namely growth, synthesis and release. Injected PTTH of 5th instar brain extract induced larval moulting in head-ligated larvae. Partially purified PTTH from 6th-instar S. litura brain induced moulting in 6th instars. Abdomen-ligated penultimate instars administered β-ecdysone showed variable moulting response. The phenomenon of hormonal interaction with development of the penultimate instar of S. litura is discussed.

This paper describes the in vitro cultivation of the 3rd-larval stage (L3) of Anisakis simplex to adulthood in a much simpler and easier to prepare medium than those described to date. The adult males obtained are between 3.8 and 6.5 cm long and the females between 4.5 and 8.0 cm. Some individually cultivated females laid eggs which had an average size of 44.4×50.5 μm. The culture conditions were as follows: medium RPMI-1640 supplemented with 20% heat-inactivated fetal bovine serum and 1% commercial pepsin, at pH 4.0 and a temperature of 37 °C, and in air atmosphere with 5% CO2. The pepsin was found to be the key to the success of the culture. The average survival of the worms in the culture increased from 50 to 88 days, due to the fact that the survival of the adults practically doubled (increasing by 1.9 times). Furthermore, the number of worms that completed the 4th moulting (M4) increased by 4.2 times, from 22.9 to 95.6%. This culture medium may facilitate, due to its simplicity, the study of anisakids, or at least of A. simplex, constituting another step towards achieving a complete in vitro life-cycle for these parasites.

Moulting and copulation by the barnacle Tetraclita japonica attached to an intertidal concrete pier were observed continuously with a video camera housed in a plastic case in summer 1996 and 1997. In a total of 779 individual-day observations, 10 individuals became sexually receptive as functional females and 14 moulted. Copulation occurred not only immediately after, but also before moulting. Duration of receptivity varied from 2 min to nearly 12 h, during which functional females accepted between two to 582 penis insertions by between one to 11 surrounding males. Each insertion time was short, ranging from 0.6 to 42 s, with a median of 2.4 s. Sex role changes were observed between two animals located close to each other, the shortest interval for which was 3.3 h. No significant size differences were detected between functional males and females. A temporal pattern of copulation, with a high frequency of penis insertions of short durations, is discussed in relation to natural habitat conditions.

There is abundant evidence of injury, teratology and pathological conditions in trilobites although it is commonly difficult to distinguish between their morphological results. Many injuries were probably sustained during moulting, with spines, bilamellar fringes and narrow gaps between dorsal exoskeleton and doublure being most vulnerable. Some injuries were the result of predaceous attack and provide important clues to the trophic structure of Palaeozoic communities. Many injuries show evidence of repair. Some supposedly teratological conditions, most notably the number of paradoxidid thoracic segments, are so common that they must be considered part of normal variation. Others represent genetic mutation or ontogenetic malfunction, the latter possibly externally induced. Undoubted pathological conditions are restricted to swellings and borings presumably caused by parasitic infestation.

Abnormalities affecting lateral areas are more common than those involving the rachis and glabella and this probably reflects both greater mortality when the axial region is involved, and greater vulnerability of peripheral regions to predaceous and moulting damage. The first explanation also accounts for the rarity of cephalic abnormalities compared to those of the pygidium.

Batches of engorged Rhipicephalus appendiculatus ticks (females, nymphs and larvae) were placed in gauze-covered vials in the field at monthly intervals and left to complete development. The ticks were placed in open areas and tree shaded areas; the latter being consistently cooler.

The duration of development varied seasonally and was inversely related to environmental temperature. The mean time to oviposition ranged from 5 days during the hot dry season to 25 days during the cool dry season. There was a high larval mortality during the larval to nymphal moult. However, the available data shows that the mean time to larval–nymphal moult ranged from less than 2 weeks during the hot dry season to 7 weeks in the cool season. The mean time to nymphal–adult moult ranged from less than 3 weeks in the hot season to over 13 weeks in the cool dry season. Egg hatching ranged between 3 weeks in the hot dry season and 15 weeks in the cool dry season.
