Recently, two attentional modes have been associated with specific eye movement patterns during scene processing. Ambient mode, characterized by short fixations and long saccades during early scene inspection, is associated with localization of objects. Focal mode, characterized by longer fixations, is associated with more detailed object feature processing during later inspection phase. The aim of the present study was to investigate the development of these attentional modes. More specifically, we examined whether indications of ambient and focal attention modes are similar in infants and adults. Therefore, we measured eye movements in 3- to 12-months-old infants while exploring visual scenes. Our results show that both adults and 12-month-olds had shorter fixation durations within the first 1.5 s of scene viewing compared with later time phases (>2.5 s); indicating that there was a transition from ambient to focal processing during image inspection. In younger infants, fixation durations between two viewing phases did not differ. Our results suggest that at the end of the first year of life, infants have developed an adult-like scene viewing behavior. The evidence for the existence of distinct attentional processing mechanisms during early infancy furthermore underlines the importance of the concept of the two modes.

The mammalian retina contains approximately 30 different morphological types of amacrine cells, receiving glutamatergic input from bipolar cells. In this study, we combined electrophysiological and pharmacological techniques in order to study the glutamate receptors expressed by different types of amacrine cells. Whole-cell currents were recorded from amacrine cells in vertical slices of the mouse retina. During the recordings the cells were filled with Lucifer Yellow/Neurobiotin allowing classification as wide-field or narrow-field amacrine cells. Amacrine cell recordings were also carried out in a transgenic mouse line whose glycinergic amacrine cells express enhanced green fluorescent protein (EGFP). Agonist-induced currents were elicited by exogenous application of NMDA, AMPA, and kainate (KA) while holding cells at −75 mV. Using a variety of specific agonists and antagonists (NBQX, AP5, cyclothiazide, GYKI 52466, GYKI 53655, SYM 2081) responses mediated by AMPA, KA, and NMDA receptors could be dissected. All cells (n = 300) showed prominent responses to non-NMDA agonists. Some cells expressed AMPA receptors exclusively and some cells expressed KA receptors exclusively. In the majority of cells both receptor types could be identified. NMDA receptors were observed in about 75% of the wide-field amacrine cells and in less than half of the narrow-field amacrine cells. Our results confirm that different amacrine cell types express distinct sets of ionotropic glutamate receptors, which may be critical in conferring their unique temporal responses to this diverse neuronal class.

Previous studies indicate that cortical areas 17 and 18 play a prominent role in generating the direction selectivities of neurons in the superior colliculus of the cat. This hypothesis was tested by quantifying the activities of neurons in the superficial collicular layers in intact cats and cats which incurred ablation of areas 17 and 18 and part of area 19. In addition, since behavioral and anatomical studies suggest a functional adjustment in the superior colliculus following removal of inputs from areas 17, 18, and 19 in the neonatal cat, we included a group of neonatally lesioned cats. Computation of an index of directionality indicated that the majority of neurons in intact cats preferred movement in one direction, thus confirming reports of others. Following ablation of areas 17 and 18 and part of area 19 in both groups of lesioned cats, only modest changes in the population indices were detected when poorly responsive neurons were eliminated from the analyses. Based upon levels of visually evoked neuronal activity, our data suggest a physiological compensation by neurons in stratum griseum superficiale following removal of areas 17, 18, and 19 inputs. In the intact and neonatally operated groups, activity in stratum griseum superficiale is high, whereas in the adult lesioned group activity is low. In stratum opticum, neuronal activity was similar in all three groups of cats. These results show that neurons in stratum griseum superficiale undergo a physiological compensation following removal of immature areas 17 and 18.

The glycinergic system in goldfish retina was studied by immunocytochemical localization of glycine antiserum at the light-microscopical level. Numerous amacrine cells, a type of interplexiform cell, interstitial cell, and displaced amacrine cell were glycine-immunoreactive (IR). Amacrine cells, accounting for 97% of the glycine-IR neurons, were of four types based solely on their level of dendritic stratification: stratified amacrine cells of the first, third, and fifth sublayers and bistratified amacrine cells of the first and fifth sublayers. Double-labeling experiments were carried out to determine possible co-localization of glycine-IR with GABA-IR, serotonin-IR, substance P-IR and somatostatin-IR. No evidence for co-localization of glycine-IR with these other transmitter substances was found, despite reports of co-localization of these substances in retinas of other species. Glycinergic neurons in goldfish retina appear to consist of a heterogeneous population of at least seven morphologically distinct subtypes that are also neurochemically distinct in regard to GABA, serotonin, substance P, and somatostatin. Since dendritic stratification in the inner plexiform layer is correlated with ON-, OFF-response types, we suggest that the subtypes of glycine-IR amacrine cells play different roles in the encoding of visual information.

Choroidal blood flow (ChBF) in birds is regulated by a neural circuit whose components are the retina, the suprachiasmatic nucleus, the medial division of the Edinger-Westphal nucleus (EWM), the ciliary ganglion, and the choriod. We have previously shown that lesions of EWM appear to result in pathological alterations in the retina. To determine whether EWM lesions also lead to altered visual functions, we have examined the effects of EWM lesions on visual acuity in pigeons. Bilateral lesions of EWM were made electrolytically, and visual acuity for high-contrast, square-wave gratings was determined behaviorally about 1 year later and compared to that of a group of pigeons that had received sham lesions of EW about 1 year prior to acuity testing. Because lesions targeting EWM invariably resulted in damage to the adjoining lateral part of the Edinger-Westphal nucleus (EWL), which controls pupillary constriction and accommodation, two additional control groups were studied. In one such control group, bilateral lesions in the area pretectalis (AP), which innervates the pupillary control part of EWL and thereby controls pupillary constriction, were made and the effects on visual acuity determined about 1 year later. In the second such control group, the effects of acute accommodative and pupillary dysfunction on acuity were studied in pigeons made cycloplegic. The accuracy of all lesions was later confirmed histologically. The mean acuities of birds with AP lesions (9.1 ± 1.4 cycles/deg) and sham lesions (7.1 ± 1.5 cycles/deg) were not significantly different from normal, based on published normative data on pigeons. In contrast, pigeons with lesions that completely destroyed EW bilaterally showed visual acuity (2.7 ± 0.1 cycles/deg) that was well below the acuity of the sham and AP-lesion control groups. The acuity of the cycloplegic pigeons (4.8 ± 0.3 cycles/deg) and one pigeon with a nearly complete bilateral EWL but a unilateral EWM lesion (6.4 cycles/deg) indicated that only about half of the loss with a bilateral EW lesion could be attributed to accommodative dysfunction. Thus, bilateral destruction of EWM appears to have led to a loss in visual acuity. This conclusion suggests that disruption of adaptive neural regulation of ChBF may impair visual function. Destruction of EWM was, however, associated with damage to the somatic components of the oculomotor and trochlear nuclei. The possibility cannot be excluded that such damage also contributed to the acuity loss.

During visual fixation, small eye movements keep the retinal image continuously in motion. It is known that neurons in the visual system are sensitive to the spatiotemporal modulations of luminance resulting from this motion. In this study, we examined the influence of fixational eye movements on the statistics of neural activity in the macaque's retina during the brief intersaccadic periods of natural visual fixation. The responses of parvocellular (P) and magnocellular (M) ganglion cells in different regions of the visual field were modeled while their receptive fields scanned natural images following recorded traces of eye movements. Immediately after the onset of fixation, wide ensembles of coactive ganglion cells extended over several degrees of visual angle, both in the central and peripheral regions of the visual field. Following this initial pattern of activity, the covariance between the responses of pairs of P and M cells and the correlation between the responses of pairs of M cells dropped drastically during the course of fixation. Cell responses were completely uncorrelated by the end of a typical 300-ms fixation. This dynamic spatial decorrelation of retinal activity is a robust phenomenon independent of the specifics of the model. We show that it originates from the interaction of three factors: the statistics of natural scenes, the small amplitudes of fixational eye movements, and the temporal sensitivities of ganglion cells. These results support the hypothesis that fixational eye movements, by shaping the statistics of retinal activity, are an integral component of early visual representations.

Photocurrents from isolated rods of adults and sub-adults of three species of amphibians, Rana pipiens, Ambystoma tigrinum, and Xenopus laevis, were measured with suction pipette electrodes. The intensity for a half-maximal response was 0.91 ± 0.48 photons μm−2 flash−1 (mean ± s.d., 10-ms flashes) for Rana, 0.92 ± 0.44 for Ambystoma, and 6.14 ± 1.33 for Xenopus. The mean number of photoisomerizations at half-saturation was 22 ± 12 for Rana, 50 ± 24 for Ambystoma, and 221 ± 48 for Xenopus. The photocurrent per photoisomerization is several times smaller in Xenopus rods than in the other two species. Spectral sensitivity was measured from 277–737 nm with light polarized both parallel and perpendicular to the planes of the membrane disks. Dichroism fell in the near UV and was absent in the region of absorption by tryptophan and tyrosine. Maximum sensitivity of Rana was at 503.9 ± 2.6 nm (n = 86), and of Ambystoma, 505.8 ± 1.8 nm (n = 24). Animals from these same batches that were sampled by HPLC had no 3-dehydroretinal (retinal2). Xenopus containing about 94% retinal2 and 6% retinal1 had λmax at 519.3 ± 2.7 nm (n = 11). Spectral position of the β-band, estimated by the method of Stavenga et al. (1993), appears to be at longer wavelengths in amphibian photoreceptors than in other vertebrates. Fits of log sensitivity to a normalized-frequency template that tracks the long-wavelength tail of the α-band (Lamb, 1995) show that the rod pigments of Rana and Ambystoma are slightly narrower than those found in the photoreceptors of fish and mammals.

Nitric oxide (NO) is known to be synthesized in several tissues and to increase the formation of cyclic GMP through the activation of soluble guanylate cyclases. Since cyclic GMP plays an important role in visual transduction, we investigated the presence of nitric oxide synthesizing activity in retinal rod outer segments. Bovine rod outer segments were isolated intact and separated into membrane and cytosolic fractions. Nitric oxide synthase activity was assayed by measuring the conversion of L-arginine to L-citrulline. Both membrane and cytosolic fractions were active in the presence of calcium and calmodulin. The activity in both fractions was stimulated by the nitric oxide synthase cofactors FAD, FMN, and tetrahydrobiopterin and inhibited by the L-arginine analog, L-monomethyl arginine. The Km for L-arginine was similar, about 5 μM for the enzyme in both fractions. However, the two fractions differed in their calcium/calmodulin dependence: the membrane fraction exhibited basal activity even in the absence of added calcium and calmodulin while the cytosolic fraction was inactive. But the activity increased in both fractions when supplemented with calcium/calmodulin: in membranes from about 40 to 110 fmol/min/mg of protein and in the cytosol from near zero to about 350 fmol/min/mg of protein in assays carried out at 0.3 μM L-arginine. The two enzymes also responded differently to detergent: the activity of the membrane enzyme was doubled by Triton X-100 while that of the cytosolic enzyme was unaffected. These results show that NO is produced by cytosolic and membrane-associated enzymes with distinguishable properties. Investigations on the purity of isolated ROS showed that about 50% of the NOS activity is endogenous to the outer segments, and that the rest is due to membrane vesicles rich in Na, K-ATPase activity. If and how NO influences the rod outer segment physiology remains to be investigated.

Unilateral partial ablation of the superior colliculus in the hamster results in a compression of the retinotopic map onto the remaining tectal fragment. In a previous electrophysiological study (Pallas & Finlay, 1989a), we demonstrated that receptive-field properties of single tectal units (including receptive-field size) remain unchanged, despite the increased afferent/target convergence ratios in the compressed tecta. The present study was done to investigate the mechanism that produces increased convergence from retina to tectum at the population level while maintaining apparent stability of convergence at the single neuron level. We injected comparable quantities of horseradish peroxidase into the tecta of normal adult hamsters and adult hamsters that had received neonatal partial tectal ablations of varying magnitude. We then compared the area of retina backfilled from the injection and the number and density of labeled retinal ganglion cells within it to the size of the remaining tectal fragment.

As expected from earlier anatomical (Jhaveri & Schneider, 1974) and physiological (Finlay et al., 1979a; Pallas & Finlay, 1989a) studies demonstrating compression of the retinotectal projection, we found that the area of retina labeled from a single tectal injection site increases linearly with decreasing tectal fragment size. However, for fragment sizes down to 30% of normal, total number of retinal ganglion cells projecting to the injection site remains in or above the normal range. For large lesions (less than 30% of tectum remaining), total number of labeled retinal ganglion cells declines from normal, despite the fact that a larger absolute area of retina is represented on each unit of tectum under these conditions. Comparison of retinal ganglion cell density with tectal fragment size shows an initial decline with decreasing fragment size, which becomes sharper with very large lesions (small tectal fragments).

The maintenance of the normal number of retinal ganglion cells innervating each patch of tectum could be accomplished by an elimination of the tectal collaterals of some retinal ganglion cells. Our results suggest that, in addition to collateral elimination, reduction in the size of ganglion cell arbors is occurring, since the peak density of backfilled ganglion cells declines less rapidly than backfilled retinal area increases, especially for small lesions. However, arbor reduction and collateral elimination must occur in such a way that individual tectal cells represent the same amount of visual space as normal.

Thus, collateral elimination and arbor reduction are two mechanisms that operate to maintain afferent/target convergence ratios (and thus receptive-field properties) over large variations in afferent availability. This compensation may occur through an activity-dependent stabilization mechanism that does not change its selectivity even when excess afferents are available. For very large lesion sizes, receptive-field size and innervating ganglion cell number and density are not preserved, thus demonstrating a limit to the afferent/target matching mechanism. The same ontogenetic mechanisms might provide a buffer for normal variations in afferent populations, and could help to align topographic maps with differing numbers of afferents.

We previously reported that two types of visual thalamic interneurons (class III and class V) could be distinguished by the presence or absence of the enzyme brain nitric oxide synthase (bNOS; Bickford et al., 1999). In the present study, we found that further immunocytochemical characterization can be used to isolate class V and class III neurons: class V neurons express GABA and bNOS but not the calcium binding protein calbindin, whereas class III neurons express GABA and calbindin, but not bNOS. By pairing staining for bNOS or calbindin with staining for GABA, we were able to distinguish class III and class V neurons, thereby allowing us to characterize the synaptic inputs to these two interneuron populations within the cat pulvinar nucleus. We found class III and class V interneurons participate in distinctly different circuitries. Class III cells receive the majority of their input from large terminals with round vesicles (RL profiles, that presumably originate from cortical layer V) while class V cells receive all of their input from small terminals with round vesicles (RS profiles, that presumably originate from cortical layer VI). These two types of interneurons also target different regions of the dendritic arbors of thalamocortical cells; class III cells contact large caliber (presumably proximal) thalamocortical dendrites within glomeruli, while class V cells contact the small caliber (presumably distal) thalamocortical cell dendrites within the extraglomerular neuropil. Thus, a dual inhibitory system exists within the visual thalamus that integrates different types of corticothalamic input and targets distinct regions of thalamocortical cell dendritic arbors.

The telencephalic target of the thalamofugal visual pathway in birds, the visual wulst, is part of the hyperstriatum accessorium/dorsale in the bird's brain. In this study, we tried to determine the exact location of the visually responsive area in the zebra finch by recording visually evoked potentials (VEPs) from different sites throughout the hyperstriatum and calculating current source densities (CSDs). In addition, we examined the influence of ipsilateral and contralateral stimuli on stimulus processing within this area, and tried to get insight into the neuronal machinery of the thalamofugal pathway by application of drugs such as tetrodotoxin (TTX) and picrotoxin.

About two-thirds of the hyperstriatum is responsive to contralateral stimuli but only a small portion responds to ipsilateral stimuli. Contralateral visual information arrives in the hyperstriatum dorsale (HD) and is processed further to the hyperstriatum accessorium (HA).

The small influence of ipsilaterally evoked potentials is not due to inhibition by the activity of the contralateral eye, as could be demonstrated previously for the ectostriatum. Instead, our results show that ipsilaterally evoked potentials are inhibited at least in part by a projection from the contralateral visual wulst.

We found that L-glutamate (L-Glu) inhibits L-type Ca2+ currents (ICa) in rod photoreceptors. This inhibition was studied in isolated rods or rods in retinal slices from tiger salamander using perforated patch whole cell recordings and Cl−-imaging techniques. Application of L-Glu inhibited ICa by ∼20% at 0.1 mM and ∼35% at 1 mM. L-Glu also produced an inward current that reversed around ECl. The metabotropic glutamate receptor (mGluR) agonists t-ADA (Group I), DCG-IV (Group II), and L-AP4 (Group III) had no effect on ICa. However, the glutamate transport inhibitor, TBOA (0.1 mM), prevented L-Glu from inhibiting ICa. D-aspartate (D-Asp), a glutamate transporter substrate, also inhibited ICa with significantly more inhibition at 1 mM than 0.1 mM. Using Cl− imaging, L-Glu (0.1–1 mM) and D-Asp (0.1–1 mM) were found to stimulate a Cl− efflux from terminals of isolated rods whereas the ionotropic glutamate receptor agonists NMDA, AMPA, and kainate and the mGluR agonist, 1S,3R-ACPD, did not. Glutamate-evoked Cl− effluxes were blocked by the glutamate transport inhibitors TBOA and DHKA. Cl− efflux inhibits Ca2+ channel activity in rod terminals (Thoreson et al. (2000), Visual Neuroscience17, 197). Consistent with the possibility that glutamate-evoked Cl− efflux may play a role in the inhibition, reducing intraterminal Cl− prevented L-Glu from inhibiting ICa. In summary, the results indicate that activation of glutamate transporters inhibits ICa in rods possibly as a consequence of Cl− efflux. The neurotransmitter L-Glu released from rod terminals might thus provide a negative feedback signal to inhibit further L-Glu release.

The aim is to study chromatic visual evoked potentials (VEP) to isoluminant red-green (R-G) stimulus in schoolchildren. Sixty children (7–19 years) with normal color vision were examined, 30 binocularly and 30 monocularly. The isoluminant point was determined for each child subjectively by using heterochromatic flicker photometry, and objectively from recordings. The stimulus was a 7° circle composed of horizontal sinusoidal gratings, with spatial frequency 2 cycles/degrees and 90% contrast, presented in onset-offset mode. VEP were recorded from Oz (mid-occipital) position. Age-dependent waveform changes and changes of the positive and negative wave were studied to both binocular and monocular R-G stimulation. Age-dependent waveform changes were observed to binocular and monocular R-G stimulation. In younger children the positive wave was prominent, whereas in older children also the negative wave became more evident. The latency of the positive wave decreased linearly with age to R-G binocular stimulation. To monocular stimulation no significant changes of the latency were observed. The amplitude of the positive wave dropped exponentially with age to binocular and monocular stimulation. The latency of the negative wave increased linearly with age to binocular and monocular stimulation, whereas the amplitude did not show age-dependent changes. These findings suggest that the chromatic VEP response undergoes evident age-dependent changes during the school-age period.

Recently developed psychophysical techniques permit the biasing of the processing of the stimulus by early visual channels so that responses reflect characteristics of either magno- or parvocellular pathways (Pokorny & Smith, 1997). We used such techniques to test psychophysically whether the global magnocellular dysfunction reported in schizophrenia also affects early processes. Seven schizophrenic patients and 19 normal controls participated. The task was a four-alternative forced-choice luminance discrimination, using a 2 × 2 configuration of four 1-deg squares. Target luminance threshold was determined in three conditions: the stimulus, including the target, was pulsed for 17 ms (pulse paradigm); the target was presented on a steady background of four squares (steady paradigm), or the target was presented alone (no background paradigm). We replicated previous results demonstrating magnocellular and parvocellular signatures in control participants. No evidence for an early magnocellular deficit could be detected as the thresholds of all schizophrenic observers were higher both in the steady paradigm (presumed magnocellular mediation) and in the pulse paradigm (presumed parvocellular mediation). Magnocellular dysfunction, if present in schizophrenia, must concern more integrated processes, possibly at levels at which parvocellular and magnocellular paths interact.

The chicken has been extensively studied as an animal model for myopia because its eye growth is tightly controlled by visual experience. It has been found that the retina controls the axial eye growth rates depending on the amount and the sign of defocus imposed in the projected image. Glucagonergic amacrine cells were discovered that appear to encode for the sign of imposed defocus. It is not clear whether the downstream neurons, the retinal ganglion cells, still have access to this information—and whether it ultimately reaches the brain. We have analyzed the spike rates of chicken retinal ganglion cells in vitro using a microelectrode array. For this purpose, we initially defined spatial resolution and contrast sensitivity in vitro. Two classes of chicken retinal ganglions were found, depending on the linearity of their responses with increasing contrast. Responses generally declined with increasing defocus of the visual stimulus. These responses were well predicted by the modulation transfer function for a diffraction-limited defocused optical system, the first Bessel function. Thus, the studied retinal ganglion cells did not distinguish between a loss of contrast at a given spatial frequency due to reduced contrast of the stimulus pattern or because the pattern was presented out of focus. Furthermore, there was no indication that the retinal ganglion cells responded differently to defocus of either sign, at least for the cells that were recorded in this study.

The thalamocortical (TC) relay neuron of the dorsoLateral Geniculate Nucleus (dLGN) has borne its imprecise label for many decades in spite of strong evidence that its role in visual processing transcends the implied simplicity of the term “relay”. The retinogeniculate synapse is the site of communication between a retinal ganglion cell and a TC neuron of the dLGN. Activation of retinal fibers in the optic tract causes reliable, rapid, and robust postsynaptic potentials that drive postsynaptics spikes in a TC neuron. Cortical and subcortical modulatory systems have been known for decades to regulate retinogeniculate transmission. The dynamic properties that the retinogeniculate synapse itself exhibits during and after developmental refinement further enrich the role of the dLGN in the transmission of the retinal signal. Here we consider the structural and functional substrates for retinogeniculate synaptic transmission and plasticity, and reflect on how the complexity of the retinogeniculate synapse imparts a novel dynamic and influential capacity to subcortical processing of visual information.

We investigated the effect of auditory–visual sensory integration on visual tasks that were predominantly dependent on parvocellular processing. These tasks were (i) detecting metacontrast-masked targets and (ii) discriminating orientation differences between high spatial frequency Gabor patch stimuli. Sounds that contained no information relevant to either task were presented before, synchronized with, or after the visual targets, and the results were compared to conditions with no sound. Both tasks used a two-alternative forced choice technique. For detecting metacontrast-masked targets, one interval contained the visual target and both (or neither) intervals contained a sound. Sound–target synchrony within 50 ms lowered luminance thresholds for detecting the presence of a target compared to when no sound occurred or when sound onset preceded target onset. Threshold angles for discriminating the orientation of a Gabor patch consistently increased in the presence of a sound. These results are compatible with sound-induced activity in the parvocellular visual pathway increasing the visibility of flashed targets and hindering orientation discrimination.

Glycine is a major inhibitory neurotransmitter in the mammalian retina and has been shown to influence the responses of ganglion cells. Midget and parasol ganglion cells serve distinct physiological roles in the primate retina and show differences in their response characteristics to light stimuli. In the present study, we addressed the question of whether the expression of glycine receptors differs in midget and parasol ganglion cells. Ganglion cells in the retinae of marmoset and macaque monkeys were injected with Neurobiotin in a live in vitro retinal whole-mount preparation. Retinal pieces were then processed with an antibody against the α1 subunit of the glycine receptor. Strong punctate immunoreactivity indicative of synaptic localization is present in the ON and OFF sublamina of the inner plexiform layer. Many of the immunoreactive puncta coincide with the dendrites of both midget and parasol ganglion cells. Immunoreactive puncta are present on distal and proximal dendrites of ON and OFF cells. These results suggest that ON and OFF midget and parasol cells do not differ with respect to the distribution of the α1 subunit of the glycine receptor.

Direction selectivity and orientation selectivity were examined in the direction-selective (DS) and orientation- selective (OS) ganglion cells in the rabbit retina. Spike activities were recorded in vivo using tungsten-in-glass electrodes. Three types of visual stimuli (flashing slit, moving edges behind a slit, and whole-field drifting gratings) were used to distinguish these attributes. Fourier analysis was performed on data obtained using drifting gratings as proposed by Worgotter and Eysel (1987). Results from both angular and Fourier (polar angle frequency) domains were consistent. DS cells had strong directional components and varying strength of orientational components; they also had intact inhibitory surrounds. The phase of the biased orientation did not have a consistent relationship with the preferred direction. OS cells had predominant orientational components and very weak directional components. We conclude that the orientation bias does not contribute to the generation of direction selectivity, and that the mechanism of this bias of DS cells is rather different from the mechanism that generates orientation selectivity in rabbit OS cells. There are indications that it is similar to the mechanism shown to cause orientation bias in cat concentric cells.

Recent physiological experiments support behavioral and morphological evidence for a fourth type of cone in the turtle retina, maximally sensitive in the ultraviolet (UV). This cone type has not yet been included in the models proposed for connectivity between cones and horizontal cells. In this study, we examined the inputs of UV, S, M, and L cones to horizontal cells. We used the high-resolution Dynamic Constant Response Method to measure the spectral sensitivity of horizontal cells without background light and after adaptation to UV, blue (B), green (G), and red (R) light. We concluded the following: (1) Tetrachromatic input to a Y/B horizontal cell was identified. The spectral-sensitivity curves of the cell in three of the adaptation conditions were well represented by L-, M-, and S-cone functions. Adaptation to blue light revealed a peak at 372 nm, the same wavelength location as that determined behaviorally in the turtle. A porphyropsin template could be closely fitted to the sensitivity band in that region, strong evidence for input from a UV cone. (2) The spectral-sensitivity functions of R/G horizontal cells were well represented by the L- and M-cone functions. There was no indication of UV- or S-cone inputs into these cells. (3) The spectral sensitivities of the monophasic horizontal cells were dominated by the L cone. However, the shape of the spectral-sensitivity function depended on the background wavelength, indicating secondary M-cone input. Connectivity models of the outer retina that predict input from all cone types are supported by the finding of tetrachromatic input into Y/B horizontal cells. In contrast, we did not find tetrachromatic input to R/G and monophasic horizontal cells. Chromatic adaptation revealed the spectral-sensitivity function of the turtle UV cone peaking at 372 nm.
