The effectiveness of ATPase in germinated seed may play an important role in the vigour of germination. The activities of tonoplast and plasma membrane ATPases in two maize (Zea mays L.) lines with different vigour of germination were determined. ATP hydrolysis was measured in microsomal fractions from coleoptiles along with the responses to specific inhibitors for the plasma membrane, tonoplast and mitochondrial ATPases as well as for acid phosphatase. Nitrate-sensitive ATPase activity was 1.5–3.0 times lower in the low-vigour line than in the high-vigour line. Kinetic analysis of ATP hydrolysis at different substrate concentrations revealed the existence of two enzymes in the microsomal fractions of the two lines. The Vmax of enzyme 1 in the low-vigour line was a third of that in the high-vigour line. This enzyme was identified as the nitrate-sensitive or tonoplast ATPase on the basis of measurements of ATP hydrolysis in the presence of specific inhibitors at high (8.12mm) and low (0.77mm) ATP concentrations.

Seed germination of watermelon (Citrullus lanatus cv. Sugar Baby) was fully suppressed by intermittent far-red (FR) irradiation (1 min every 30 min). When the intervening dark period was increased, a linearly increasing final germination percentage was obtained. However, a 4-day intermittent FR treatment induced phytochrome-controlled dormancy and the longer the dark interval the deeper was the dormancy of the non-germinated seeds. When seeds were soaked in a fully inhibitory osmotic solution, no dormancy was imposed. However, a single FR pulse at the time of transfer induced partial, secondary dormancy; the kinetics of the imposition of dormancy followed a negative exponential curve (half-life 1.5 days; 3 days for the cultivar Crimson Sweet). Seeds osmotreated for 10 days in darkness and subsequently dehydrated (with and without a final FR pulse) acquired germination characteristics similar to those in light-requiring and dark-germinating achenes, respectively, of the lettuce cultivar Grand Rapids. In the light-requiring osmomanipulated seed population, the induction of germination was brought about by the low-energy reaction of phytochrome, chilling, dry storage and decoating. The transformation through osmomanipulation of the dark-germinating watermelon seeds (the inhibition of which required prolonged exposure to light) to positively or negatively photosensitive seeds (that responded to brief light pulses), might be attributed to the slow relaxation of existing meta-Fa and meta-Rb phytochrome intermediates to Pfr upon hydration.

Seeds of the Brussels sprouts cultivar Asmer Aries and the cauliflower cultivar Hipop were subjected to ageing at 20% moisture content and 45°C for 24 or 30 h, respectively; all seeds retained high germination after ageing. Aerated hydration of unaged and aged seeds of both cultivars for 4–8 h at a range of temperatures (10–30°C), followed by drying, resulted in improved performance, except that germination percentage and rate of cauliflower were lower at 10°C. Thus, all treated seeds showed greater germination rate and seedling root length than the control, which may have resulted from the advancement of the process of germination. The deleterious effect of aerated hydration at 10°C on cauliflower could be explained by damage due to rapid imbibition; seeds that had imbibed slowly to close to full imbibition (41% moisture content) before aerated hydration showed no decrease in germination. The improvement of aged seeds after aerated hydration was also revealed by higher germination after the controlled-deterioration test, which indicated less deterioration in treated seeds. Furthermore, the optimum improvements for all seeds were observed at 25°C and were greater when the water was aerated than non-aerated. These observations indicate the activation of metabolic repair processes during aerated hydration, leading to a reversal of the deterioration sustained during ageing.

The sorption isotherm for excised embryonic axes of recalcitrant (i.e. desiccation-sensitive) Quercus robur L. acorns was determined to find the relation between moisture content and water potential. Subsequently, physiological studies on the effect of desiccating the axes to a range of water potentials were undertaken. The respiratory capacity declined steeply after short exposure to water potentials from −5 to −30 MPa. The leachate conductivity increased significantly after exposure to −5 MPa and rose steeply after exposure to between −12 and −40 MPa. Axes were equilibrated at different relative humidities and the proline content showed a 15-fold increase with a peak value at −10 MPa. It was concluded that the critical water potential for initiation of damage was −5 MPa, and that axes accumulated proline as a response to desiccation stress. The embryonic axes from Q. robur behave more like typical vegetative tissue of angiosperms than like orthodox seeds.

On the premise that seed ageing may be largely a result of free-radical lipid autoxidation, a study was made of the relationship between lipid stability and longevity in seeds of soybean (Glycine max), lentil (Lens culinaris), mungbean (Vigna radiata), chickpea (Cicer arietinum), broadbean (Vicia faba), pea (Pisum sativum), lettuce (Lactuca sativa), and tomato (Lycopersicon esculentum). Seed lipids were examined for α-, γ- and δ-tocopherols, and levels of unsaturated and saturated fatty acids. Using this information, analysis was attempted using linear, multiplicative or exponential models to correlate aspects of lipid stability with seed longevity values from the published literature. No statistically significant correlations could be found between longevity and total lipid unsaturation, tocopherol levels or two protection formulae obtained from the oil chemistry literature. When values for tomato were excluded, a good correlation (r = 0.89, P = 0.007) was obtained using a multiplicative regression model for levels of linolenic acid per unit of total tocopherols in relation to longevity. Possible factors contributing to a lessening of the relationship between lipid stability and seed longevity are discussed.

Protein composition of sunflower seeds (Helianthus annuus L.) was analysed to identify a possible marker for germination capacity and to evaluate seed viability precociously after harvest and after natural ageing during storage. Electrophoresis on 2-D gels was used to compare total soluble and in vivo synthesized proteins in two seed lots showing different germination capacities. Differences were observed to a greater extent in in vivo protein synthesis profiles than in total soluble proteins. Inhibition of [35S]methionine uptake and incorporation in seeds with low germinability was also observed. The precise relationship between protein composition and prediction of germinability could not be determined. Other aspects such as membrane damage, capacity for protein synthesis and seed lot homogeneity in relation to germination are discussed.

Norway maple (Acer platanoides L.) seeds were harvested at different stages of seed development and maturation in 1989–91. As maturation drying progressed, the seed populations showed increasing desiccation tolerance: at 67–69% moisture content, no seeds survived desiccation below 10% moisture content; maturation drying to 55–57% moisture content (values corresponding with the end of the seed-filling phase) improved desiccation tolerance, but nevertheless most seeds were unable to withstand desiccation to 5–7% moisture content; further maturation drying to 27–28% moisture content enabled the seeds to survive considerable desiccation, no loss in viability occurring in seeds dried to 3% moisture content. This considerable improvement in desiccation tolerance after the end of the seed-filling phase was correlated (P<0.05) with the progress of maturation drying and may be associated with the increase in the potential longevity of seeds of other species that occurs during seed development subsequent to seed filling.

Six pasture legumes, common in Syrian grasslands, were investigated. Medicago orbicularis and M. rotata had the largest number of hard seeds, with >90% still hard 5 months after shedding; Trifolium stellatum had the least with <30% of hard seeds; T. campestre, M. rigidula and T. tomentosum were intermediate. Hard-seededness in M. orbicularis and M. rotata varied between years. Scarification of hard seeds resulted in almost complete germination in all species. Alternating temperatures (10/20°C) reduced the rate of germination of scarified T. campestre seeds, but had no effect on germination of scarified or unscarified seeds of T. stellatum. The breakdown of hard-seededness in the field in Syria did not begin until 3 months after shedding and greatly increased when seeds overwintered. Breakdown of hard-seededness was slower when seeds were buried at 5 and 10 cm than when seeds remained on the soil surface. Seedling emergence after the rains started in November was highly correlated with the number of soft seeds observed in October. Similarly, a high correlation was found between the percentage of hard seeds in October and carry-over in the seed bank. In small-seeded species (Trifolium spp.), 39% of the initial seed bank was lost in 2 years by causes other than emergence as seedlings. Such losses were only 7% in larger-seeded species (Medicago spp.). The implications of hard-seededness on the utilization and survival of legumes in grasslands are discussed.

The seeds of three medics (Medicago orbicularis, M. rigidula and M. rotata) and three clovers (Trifolium stellatum, T. campestre and T. tomentosum) were collected from a pasture in north-west Syria on three occasions: immediately after seed set, at the end of the dry season, i.e. 4 months after seed set, and 16 months after seed set. Complete and sectioned seeds were observed with a scanning electron microscope. Morphological changes and differences in thickness of the seed coat were related to the hard-seededness of the samples collected in the seed bank before the onset of the rainy season. The morphology and structure of the seed coat of all six species were typical of papilionoid legumes. The species with the softest seeds (T. stellatum) showed a very thin and discontinuous cuticular layer at seed maturation (0.16 μm), while a thick and continuous cuticle (4.24 μm) was characteristic of the hardest-seeded species (M. orbicularis). The lens region of seed of all species except T. campestre was weakened after a few months in the field. This weakening was also found in seeds which were certainly hard, but the cracks in the lens region were apparently not deep enough to allow imbibition. Even when large areas of the palisade layer were removed from the lens region of seeds of M. rotata, this was not sufficient to permit water uptake. Differences in the seed coat thickness of the six species accounted for differences in seed dormancy only when seed mass was taken into account.

In order to examine the occurrence of a glassy state in the cytoplasm of maize embryos as a function of water content, isolated embryos were examined using spin-probe electron spin resonance. The glass transition temperature was determined at various degrees of hydration (h) in the range 0.05–0.25 g H2O g−1 dry sample weight. The obtained phase diagram indicates that, at standard storage temperature (−5°C), the cytoplasm of embryos drier than 0.15 g H2O g−1 is in a glassy state.
