The influence of temperature and water availability on seed germination can vary across the geographic range of species with a large distribution. Betula pendula is a widely spread European tree that has differentiated into two narrowly distributed taxa, endemic to Mediterranean mountains: Betula aetnensis in Sicily and B. fontqueri in Spain and Morocco. We tested the hypothesis that the regeneration niche, expressed by temperature and water potential thresholds, varies across these species and is influenced by the local climate. Seeds were collected from six populations of B. pendula, one of B. fontqueri and two of B. aetnensis. Germination tests were conducted between 5 and 30°C. The thermal thresholds were calculated before and after cold stratification. The osmotic potential tested ranged from 0 to −1.5 MPa. Time to reach 30 and 50% of germination was calculated by fitting non-linear models. Germination was promoted by high temperatures, but the response to stratification was heterogeneous. Tb and Ψb differed between and within species. Tb ranged between 2.22 and 8.94°C for unstratified seeds. Mediterranean species had higher drought tolerance, while B. pendula showed contrasting responses to low water potential. Ψb reached a minimum value of −1.15 MPa in B. fontqueri. High temperatures influenced the Tb of unstratified seeds negatively, while, after stratification, the Tb increased with precipitation in the driest month. The heterogeneity observed could reflect higher genetic variability in marginal populations of silver birch. Knowledge of their germination ecology may be useful to mitigate future impacts of climate change on core populations of B. pendula.

The plumules of newly-excised wampee embryos, which are more sensitive to dehydration than the roots, became more resistant to water loss when axes were allowed to sprout on woody plant medium [WPM; McCown and Lloyd (1981) Hortscience16, 453] before being dried. Pre-treatment of sprouting axes (seedlings) with sucrose incorporated in the WPM enhanced survival. Although the roots withered following further dehydration of seedlings cultured on WPM containing 60% sucrose, excised plumules were capable of generating adventitious roots when a combination of 10 mM α-napthaleneacetic acid and 10 mM indole-3-butyric acid was used during subsequent in vitro incubation.

Temperature is a key environmental signal regulating germination. A thorough understanding of how seed populations respond to various temperatures can inform end-users regarding effective establishment strategies and forms the basis for questions related to a taxon's thermo-biology. Although abundant information exists regarding germination responses of economically important crops to several temperature scenarios, much less is known concerning the seed biology of wild germplasm. To address this, we examined the germination response of non-dormant Rudbeckia mollis seeds to various doses of constant or simulated seasonal diel temperatures. Germination response was sigmoidal. Seeds of R. mollis were capable of germinating within a few days to high percentages (>95%) at relatively cool constant (15–25°C) or 12-hour alternating (22/11–33/24°C) temperatures, with optimum temperatures for germination occurring at 25°C or 29/19°C. Germination was inhibited as temperatures increased to 30°C or 33/24°C with early and late germinating phenotypes displaying differential responses at these temperatures. No germination occurred at 35°C. Results are discussed in terms of seedling establishment of R. mollis outside its natural range and implications of climate change on germination.

The pattern of seed dispersal in time and space can affect plant fitness and the soil seed bank, and thus information is needed on this aspect of the seed biology of a species before it is selected for use in habitat restoration projects. Zygophyllum xanthoxylon is a super-xerophilous shrub that is a potential pioneer species for use in revegetating highly disturbed areas of the cold deserts of northwest China. We studied fruit release and soil seed banks of Z. xanthoxylon for 3 years in two cold desert habitats characterized by different degrees of drought and wind velocity. In our study, fruit (a three-winged capsule) release began in summer (June 2010, August 2011, July 2012) and extended for 9–10 months, but plants can be found in the population with previous- and current-year fruits attached to them. More than 50% of the fruits were released in the first 3–4 months after maturity, while the others were released gradually over a 7–8 month period. The temporal pattern of fruit dispersal varied with habitat but not with amount of precipitation during summer. The pattern of fruit deposition on the soil surface was affected by neighbouring plants, wind velocity, wind direction and topography. In both habitats, >90% of the fruits were deposited beside large and small clusters of plants, mainly Ephedra przewalskii. To facilitate plant community development, we suggest that E. przewalskii should be planted (as a wind break) together with Z. xanthoxylon when native pioneer species are used for restoration of cold desert shrublands.

Seeds from an assortment of 28 grass species (including two cereals), chiefly representing different tribes of the sub-family Pooideae were tested for the presence of BASI-like proteins using Western blotting techniques and antisera raised against the barley amylase/subtilisin inhibitor. A single protein species of the same molecular weight as the BASI protein was detected in each member of the tribes Triticeae and Bromeae tested. Members of the Triticeae and Bromeae were the only species to produce high pi α-amylase isoenzymes following germination of the grain in addition to low pl forms which were present in all species tested. Anti-BASI antibody binding patterns for the other grasses examined were variable with the strongest staining being observed for Lolium species. In these species, either a double or single protein band fractionally larger than the BASI protein was recognized. In most other cases antibody binding was barely or not detectable. Inhibitor preparations from Hordeum vulgare and Lolium perenne were effective at inhibiting wheat α-amylase but neither had any effect against enzyme produced by germinating L. perenne.

A survey of proteinaceous inhibitors of the serine proteinases, bovine trypsin and chymotrypsin, that are extractable from dry seeds of several cultivars of pasture grasses has been undertaken. Using crude extracts, most cultivars screened contained inhibitors of chymotrypsin, whereas trypsin inhibition was not detectable. Seeds from four cultivars, Lolium perenne L. cv. Grasslands Ruanui, Lolium × boucheanum cv. Grasslands Greenstone, Festuca arundinacea Schreb. cultivars Grasslands Roa and Grasslands Garland, that contained more potent chymotrypsin inhibition were purified further. After gel filtration chromatography, both trypsin and chymotrypsin inhibition could be observed in all four cultivars, and each separated into two discrete native molecular weights; one of ca. 20–22 kDa and one of ca. 8–10 kDa. However, activity staining, after polyacrylamide gel electrophoresis, revealed an array of iso-inhibitors with molecular weights that ranged from ca. 3 kDa to 20 kDa. One of these, a dual trypsin/chymotrypsin inhibitor of ca. 12 kDa that is present in all four cultivars examined, was purified to homogeneity from F. arundinacea cv. Grasslands Garland using anhydro-trypsin affinity chromatography and reverse-phase HPLC. The protein was found to comprise two closely related peptides and N-terminal amino acid sequencing revealed highest identity with a trypsin inhibitor identified in rye (Secale cereale) seeds.

Climate variability is expected to increase due to climate change, with projected increases in temperature and erratic rainfall patterns. These changes will alter the environmental cues sensed by seeds, and therefore will impact plant recruitment. This study investigated the effects of seed functional traits (germinability, germination time, synchrony and seed mass) on germination responses of several sub-tropical native Australian plant species under different environmental factors (water stress, salinity and pH). The effect of a hot water pre-treatment was also tested on Fabaceae seeds with known physical dormancy. Seed traits, environmental factors and seed pre-treatments had significant effects on final germination percentage and germination time. Seed mass and time to 50% germination (t50) were also positively correlated. In contrast, pH did not affect germination and there was no interaction between pH and any of the measured seed functional traits. Some species showed a high thermal tolerance to germination and germination was indifferent to light conditions for all species. Results showed that certain seed functional traits interact with environmental factors to influence germination percentage and time. These findings highlight the importance of considering seed functional traits when determining a species germination response under a changing climate. In addition, the findings provide important knowledge to better guide seed-based land restoration programmes.

Scientists are becoming increasingly aware that disparities in opportunities for conducting and publishing research among scientists living under different socio-economic contexts have created pervasive biases and long-lasting impacts on our views of the natural world. These disparities are challenging the establishment of a global research agenda for a variety of disciplines, including seed ecology. Seed ecology has progressed enormously recently, but multiple barriers have hindered progress in the Global South where biodiversity and environmental complexity are highest. Here, we identify ten major challenges that seed ecologists from developing countries face in relation to planning, designing, conducting and publishing their research. We also propose several measures to overcome these challenges: (1) closing biodiversity knowledge shortfalls, (2) enhancing and creating long-term seed ecological networks, (3) supporting better infrastructure, (4) making fieldwork easier and safer, (5) unlocking funding opportunities, (6) promoting inclusive scientific meetings, (7) alleviating language barriers, (8) improving education, (9) shifting the notion of novelty and relevance and (10) supporting native seed markets. The authors recommend that the proposed solutions can be implemented by seed ecologists and the broader scientific community including funding agencies, research directors, journal editors and the academic publishing industry. Solutions can help mitigate multiple challenges simultaneously, thus offering a relatively inexpensive, fast and productive pathway for the development of seed ecology into a truly global research discipline that benefits scientists irrespective of their geographic location and background.

Generally, orchids produce dust-like seeds in which endosperm reduction and embryo undifferentiation represent a derived state shared with species in about 11 other plant families. Orchid seeds are proposed to have a special kind of morphological or morphophysiological dormancy. We test this proposition, overcoming several design limitations of earlier studies, specifically that the in vitro germination method for orchid seeds uses pro-oxidants for disinfection and incorporates nitrate in the medium; both ‘treatments’ might contribute to dormancy breaking, potentially confounding judgement on the depth and nature of the dormant state. Seeds of the tropical orchid Dendrobium cruentum Rchb. f., were sown both in vitro, on a nutrient medium, and ex vitro, on plain agar omitting prior disinfection with sodium hypochlorite. Seeds previously stored and fresh seeds were incubated under combinations of vitro conditions, light treatments, constant or alternating temperatures and nitrate concentration. Seeds of D. cruentum are very small but have a large embryo that occupies most of the seed. Over a range of constant temperature seeds germinated to the spherical protocorm stage just as well ex vitro as in vitro. Neither light nor nitrate were prerequisites for ex vitro germination. The ability of D. cruentum seed to germinate in the absence of environmental or chemical stimuli suggests that mature seed can be non-dormant. Our results support the proposition that neither all DUST seed fit a dormancy class nor all orchids produce morphological or morphophysiological seeds. Finally, embryo/seed volume determinations in orchids may prove as valuable in studies on the evolution and ecology of germination and dormancy as embryo:seed ratios in other angiosperm species.

We investigated the thermal thresholds to seed germination and the variations in abscisic acid (ABA) levels and oxidative metabolism during seed dormancy-breaking and germination in two palm species with differences in desiccation tolerance. We used Mauritia flexuosa (buriti palm, desiccation-sensitive seeds) from swampy habitats (Veredas) and Attalea speciosa (babassu, desiccation-tolerant seeds) from the transition zone between the forest and semi-arid region (drained soils). Germination was evaluated at 15–40°C after dormancy-breaking (operculum removal). At optimal temperature for both species (30°C), embryos were sampled in distinct germination phases – dry, imbibed, after operculum removal and at early germination – and used for quantifying ABA and hydrogen peroxide (H2O2) content, antioxidant enzyme activities and for histolocalization of superoxide anion (O2−). Seeds of M. flexuosa germinated only in a narrow temperature range (25–35°C), while A. speciosa seeds germinated between 15 and 40°C. After operculum removal, reduced ABA levels in embryos of M. flexuosa were accompanied by constant H2O2 levels, while in A. speciosa, similar levels of ABA and H2O2 were maintained throughout all germination phases. The presence of O2− was restricted to the haustorium, and an increase in O2− accumulation was observed in both species after operculum removal. Similarities were noted between both species regarding enzyme activities; however, the activities were higher in embryos from M. flexuosa. The presence of O2− only in the haustorium indicates that this region of the embryo is an active structure following imbibition and is involved in germination itself, not just functioning in reserve mobilization.

The variation for protein content has been assessed in a series of pea (Pisum sativum L.) lines differing in mean seed size. The range of seed size within each genotype was manipulated by growing the plants in different environments and by altering the structure of the plant. The response of the lines in terms of seed weight to the environments and treatments was inconsistent, but 90% of the total variation was accounted for by differences between genotypes. In contrast, for percentage protein, the genetic component was much weaker with nearly half the variation being accounted for by within (residual) variation. The correlations between seed weight and percentage protein were highly genotype specific. For the largerseeded genotype, percentage protein increased with increasing seed size (r= +0.9, P < 0.01), while for the small-seeded genotype there was mid-range negative value (r = −0.5, P <0.01). It is suggested that any assessment of percentage protein in pea genotypes must take account of seed size.

Raffinose, stachyose and phytin are undesirable compounds for soybean food and animal feed products. In seeds, raffinose and stachyose are believed to contribute to desiccation and cold stress tolerance. Thus, removal of these compounds from soybean by genetic mutation has resulted in a more commercially desirable composition, but potentially less physiologically viable seeds. In an effort to develop a method to improve viability and seed storability in soybean, stem–leaf–pod explants of three low raffinose, low stachyose lines, two of which were also low in phytin, and a check line were fed solutions containing d-chiro-inositol, myo-inositol or d-pinitol, free cyclitols which unload through the seed coat to the developing embryo where they accumulate as fagopyritols, galactinol and galactopinitols, respectively, during seed maturation. Increased galactopinitol and fagopyritol accumulation may substitute for the roles of raffinose and stachyose in low raffinose, stachyose and phytin seeds. Explants of all lines unloaded d-chiro-inositol, myo-inositol and d-pinitol. Fed d-chiro-inositol accumulated in leaf tissues demonstrating uptake into explants. Fed d-chiro-inositol and myo-inositol accumulated in pod wall and seed coat tissues of one or more lines. The results indicate that d-chiro-inositol was unloaded from the seed coat to the embryo in increased amounts after feeding. The potential use of increased maternal d-chiro-inositol for synthesis of fagopyritols in embryos to improve seed performance in low-stachyose and low-phytin soybean seeds is supported. The seed coat cup unloading of fed free cyclitols may provide a model system to test effective unloading of upregulated maternally synthesized cyclitols.

Only a few studies have been performed on seed germination of perennial ephemeral species native to the cold deserts of central Asia. We hypothesized that seeds of the cold desert perennial ephemeral Leontice incerta exhibit versatility in the timing of germination, that is, having the capacity to germinate at any time in summer, autumn and next spring. At dispersal in late May, only about 30% of the seeds could germinate; thus, a high percentage of the seeds was dormant. Seeds had a fully developed embryo, and dry storage, cold stratification, warm stratification and gibberellin promoted germination; we concluded that they have non-deep physiological dormancy. Seeds buried under natural conditions during summer germinated to 57–86% in autumn (late October) when exhumed and incubated at 5/2–25/15°C. However, seeds were sown in soil exposed to natural temperature and (low) precipitation did not germinate until next spring when the soil was moist. Thus, like various cold desert annuals, seeds of the perennial L. incerta can germinate in summer, autumn and next spring, depending on the availability of soil moisture (rainfall). Rainfall in cold deserts can play an important role in shaping seed germination traits of desert plants.

In small seeds, light often promotes germination and longer-term exposure to darkness reduces light sensitivity. In cacti inhabiting harsh environments, a rapid response to light exposure is potentially advantageous for seedling establishment. We exposed dark-imbibed seeds of the cactus Cereus repandus to doses of red (RED) light and far-red (FR) light. The seeds exhibited positive photoblastism to RED light. Although the initial levels of germination varied between seed lots, the sensitivity to increasing the RED dose did not. As little as 5 min per day for 4 d was sufficient to saturate the light requirement for germination. The effects of RED light were reversed by FR exposure as long as the interval between RED and FR did not extend to 2 d, by which time the seeds had ‘committed’ to germinate. Dark incubation for 1–2 weeks prior to RED exposure reduced light sensitivity in two seed lots, such that RED only promoted around 20% germination. Phytochrome is assumed to mediate the reversibility of the RED:FR response. High sensitivity to light spectral quality suggests that seeds of C. repandus are able to germinate quickly in high-quality microsites, but seed burial or shading may commit the seeds to form a soil seed bank. The light characteristics of the germination trait in this species are typical of many small seeded species of the drylands.

Viburnum is a temperate-zone genus that also occurs in mountains of South America and Malesia, and seeds of many species have morphophysiological dormancy (MPD). Information on the level of MPD in seeds of species in various clades of Viburnum potentially would increase our understanding of the evolutionary relationships between the nine levels of MPD. Our aim was to determine the level of MPD in seeds of Viburnum plicatum var. formosanum that is endemic to mountains (1800–3000 m a.s.l.) in Taiwan and a member of the Lutescentia clade. The temperature requirements for embryo growth and root and shoot emergence and response of seeds to gibberellic acid (GA) were determined. No fresh seeds germinated during 16 weeks of incubation at 15/5, 20/10, 25/15, 30/20 or 25°C. Embryo growth and root emergence occurred during moist cold stratification at 5°C or at a temperature sequence of 15/5 to 5°C. During cold stratification, embryos length increased from 0.76 ± 0.06 to 3.40 ± 0.26 mm and the embryo length:seed length ratio from 0.20 ± 0.02 to 0.68 ± 0.07. In a temperature sequence simulating field conditions, embryos grew inside seeds at 5°C, roots emerged at 15/5°C and shoots emerged at 20/10°C. The optimum temperature for embryo growth was 5°C. Neither GA3 nor GA4 was effective in promoting root emergence. We conclude that seeds of V. plicatum var. formosanum have deep complex MPD, which is a first report for Viburnum. Dormancy release during the cool season at high elevations helps to ensure that seeds germinate at the beginning of the warm season.

Temperature may regulate seed dormancy and germination and determine the geographical distribution of species. The present study investigated the thermal limits for seed germination of Polygonum ferrugineum (Polygonaceae), an aquatic emergent herb distributed throughout tropical and subtropical America. Seed germination responses to light and temperature were evaluated both before (control) and after stratification at 10, 15 and 20°C for 7, 14 and 28 d. Germination of control seeds was ~50% at 10 and 15°C, and they did not germinate from 20 to 30°C. The best stratification treatment was 7 d at 10°C, where seed germination was >76% in the dark for all temperatures, except at 30°C, and < 60% in light conditions. A thermal time approach was applied to the seed germination results. Base temperature (Tb) was 6.3°C for non-dormant seeds and optimal temperature (To) was 20.6°C, ceiling temperature (Tc (<50)) was 32.8°C, and thermal time requirement for 50% germination was 44.4°Cd. We concluded that a fraction of P. ferrugineum seeds is dormant, has a narrow thermal niche to germinate (10 and 15°C) and that cold stratification (10°C) alleviated dormancy and amplified the thermal range permissive for germination of the species. Consequently, P. ferrugineum is expected to occur in colder environments, for example, at high altitudes. Higher temperatures decrease the probabilities of alleviate dormancy and the ability of their seeds to germinate.

Size-class structure was used to monitor the growth of underdeveloped embryos in two seed cohorts of Aristolochia macrophylla, A. tomentosa and A. californica and the results are related briefly to differences in seed dormancy among and within the three species. The size-class method of presenting embryo growth data allows more insight to be gained into the embryo-level cause(s) of seed dormancy than does the cumulative growth curve. This appears to be the first report on use of size-class structure to monitor growth of underdeveloped embryos.

Imbibed embryos of after-ripened and secondarily thermo- and FR-dormant Agrostemma githago seeds were investigated as to their ability to synthesize DNA, RNA and proteins with the aim of finding characteristic differences connected with the induction and maintenance of developmental arrest. A gradual decrease in DNA synthesis was observed during the induction of thermodormancy. However, DNA synthesis was stimulated up to that of embryos of 30–h-imbibed after-ripened seeds within 24 h approximately after transferring the thermodormant seeds into temperatures which normally allow germination. DNA synthesis of embryos of FR-dormant seeds remained constant at a relatively low level during 7 d FR and another 7 d dark incubation. RNA synthesis decreased to different extents during induction of thermo- and FR-dormancy when it was arrested at a relatively low level in seeds transferred to temperatures which normally allow germination. Processes leading to an increase in RNA synthesis such as in embryos of after-ripened seeds appeared to be quantitatively and/or qualitatively repressed. Interestingly, protein synthesis was extremely depressed during induction of thermodormancy whereas it was slightly stimulated during induction of FR-dormancy. Nevertheless two-dimensional protein PAGE revealed several polypeptides which were new, increased, decreased or not synthesized predominantly in axes of thermo- and FR-dormant seeds in comparison to germinating after-ripened seeds. It is suggested that a connection exists between these polypeptides and the repression of germination. After transferring seconarily dormant seeds to temperatures which normally allow germination, a temporary stimulation of protein synthesis could be observed in both cases.

The capacity for protein synthesis in embryonic axes of Phaseolus vulgaris L. cv. Top Crop seeds was reduced markedly by soaking in airsaturated and CO2-saturated water. The former treatment induces soaking injury while the latter treatment prevents injury. After soaking, the capacity for protein synthesis in axes of seeds soaked in air-saturated water declined and the pattern of synthesis remained almost similar to that at the time of termination of soaking. However, the capacity and pattern of protein synthesis in axes of seeds soaked in CO2-saturated water recovered subsequent to soaking and approached that of non-soaked seeds.
