Triticum monococcum ssp. monococcum has useful traits for bread wheat improvement. The synthesis of Triticum turgidum–T. monococcum amphiploids is an essential step for transferring genes from T. monococcum into bread wheat. In this study, 264 wide hybridization combinations were done by crossing 60 T. turgidum lines belonging to five subspecies with 83 T. monococcum accessions. Without embryo rescue and hormone treatment, from the 10,810 florets pollinated, 1983 seeds were obtained, with a mean crossability of 18.34% (range 0–89.29%). Many hybrid seeds (90.73%, 923/1017) could germinate and produce plants. A total of 56 new amphiploids (AABBAmAm) were produced by colchicine treatment of T. turgidum × T. monococcum F1 hybrids. The chromosome constitution of amphiploids was characterized by fluorescence in situ hybridization using oligonucleotides probes with different chromosome and sub-chromosome specificities. Sodium dodecyl sulphate polyacrylamide gel electrophoresis analysis indicated that the Glu-A1m-b, Glu-A1m-c, Glu-A1m-d and Glu-A1m-h proteins of T. monococcum were expressed in some amphiploids. Despite resistance reduction in several cases, 45 out of 56 amphiploids exhibited resistance to the current predominant Chinese stripe rust races at both the seedling and adult plant stage. These novel amphiploids provide new germplasm for the potential improvement of bread wheat quality and stripe rust resistance.

Woad (Isatis tinctoria L.) was the most important source of natural blue indigo, a pigment used mainly for dyestuff until the beginning of the 20th century, when the increased use of synthetic dyes by the industry lead to a decrease in the interest of natural dyes and to the abandonment of dye crops. The aim of this study was to characterize, according to morphological and agronomical traits, 11 woad accessions from different countries and to quantify the indigo precursor content by high-performance liquid chromatograph-diode array detection (HPLC-DAD). Qualitative traits revealed a low variability and a great variation was observed in quantitative traits. Principal component analysis (PCA) diagrams divided the accessions into four groups, primarily according to their geographic origin. The first three components of the PCA accounted for 76.8% of the total variation. Reciprocal interactions between indigo precursors and fresh leaf weight were compared through genetic diversity, with significant differences in isatan B (0.5–5.1 g/kg), indoxyl (0.3–2.0 g/kg) and residual indigo (0.3–0.5 g/kg). This information, together with genetic data, can be used to assist local farmers to re-introduce Isatis species in the European agricultural system, not only indicating the higher indigo yielding genotypes, but also the most suitable harvest time.

The starchy corms of taro (Colocasia esculenta) are consumed throughout the tropics and are essential for food security in many developing countries. Taro corms are increasingly processed into fries, chips, flours or flakes in urban areas, and varieties with attractive corm flesh colours are now needed. The identification of flavonoids in taro corms would add value to this crop. The present study developed a high-performance thin layer chromatography (HPTLC) protocol for the high-throughput screening of flavonoids in taro germplasm. Overall, 350 different accessions were analysed including 259 varieties from Vanuatu, one from Vietnam, eight from Thailand, eight from the Philippines, six from Malaysia, two from Japan and 18 from Indonesia. Forty-eight breeding lines (hybrids) including 21 from Vanuatu, 21 from Samoa, four from Hawaii and two from Papua New Guinea were also analysed. Ten flavones, namely luteolin-6-C-hexoside-8-C-pentoside, schaftoside, luteolin-3′,7-di-O-glucoside, homoorientin, isovitexin, orientin, luteolin-4′-O-glucoside, luteolin-7-O-glucoside, vitexin and apigenin-7-O-glucoside, were successfully detected in the corm and are responsible for the attractive yellow colour of the flesh and fibres. Quantitatively, luteolin-6-C-hexoside-8-C-pentoside and schaftoside were the most important of all the detected flavonoids. However, only 18% of the varieties analysed presented these two compounds and 80% presented poor flavonoid composition. No geographical structure of the variation was detected and the most flavone-rich varieties originated from Vanuatu, Thailand, the Philippines, Malaysia and Indonesia. The compounds detected in the present study were significantly and positively correlated, suggesting that there is potential for fast improvement through controlled crosses, subsequent evaluation of full-sib progenies and cloning of elite individuals.

Belgian agriculture could decrease its heavy dependency on imported protein crops by a local production of soybean. Unfortunately, soybean production is hampered by Belgium's short and cold growing season. We evaluated 409 varieties, breeding lines and genebank accessions planted at two planting dates in a row-plot experiment to explore the genotypes that are suitable for growing in Belgium. The current MG000 varieties may require additional crossings with very early-maturing genotypes to guarantee an optimal and safe harvest. Within such crossings, care must be taken to maintain the indeterminate or semi-determinate growth habit. Vegetative development was negatively correlated with flowering date and maturity date, but positively correlated with cold tolerance. Seed quality was mainly affected by mould infection (associated with strong lodging and late maturity) and mottling caused by soybean mosaic virus. Planting 3 weeks earlier resulted in 8 d earlier flowering and 7 d earlier maturing, without significant losses in seed yield per plant. The results of this row-plot experiment hold promise to select for genotypes adapted to the Belgian conditions.

Aegilops tauschii, the diploid progenitor of the wheat D-genome, is a valuable genetic resource for wheat breeders. In this study, we compared the efficiency of inter-simple sequence repeat (ISSR) (as an arbitrary technique) and start codon targeted (SCoT) (as a gene-targeting technique) markers in determining the genetic diversity and population structure of 90 accessions of Ae. tauschii. SCoT markers indicated the highest values for polymorphism information content, marker index and effective multiplex ratio compared to ISSR markers. The total genetic diversity (Ht) and genetic diversity within populations (Hs) parameters were comparably modest for the two marker systems. The results of the analysis of molecular variance showed that the genetic variation within populations was significantly higher than among them (ISSR: 92 versus 8%; SCoT: 88 versus 12%). Furthermore, SCoT markers discovered a high level of genetic differentiation among populations than ISSRs (0.19 versus 0.05), while the amount of gene flow detected by ISSR was higher than SCoT (2.13 versus 8.62). Cluster analysis and population structure of SCoT and ISSR data divided all investigated accessions into two and four main clusters, respectively. Our results revealed that SCoT and ISSR fingerprinting could be used to further molecular analysis in Ae. tauschii and other wild species. The high-genetic variability found in this study also indicates the valuable genetic potential present in the investigated Ae. tauschii germplasm, which could be utilized for future genetic analysis and linkage mapping in breeding programmes.

Bacterial blight (BB) of rice caused by Xanthomonas oryzae pv. oryzae (Xoo) is a major biotic constraint of rice production in all the major irrigated/lowland rice growing regions of Asia, including Punjab and its adjoining states in India. None of the individual BB resistant Xa/xa genes is effective against Punjab pathotypes. In the present study, we have screened 1176 accessions, comprising 1007 accessions of A genome species Oryza glaberrima, O. barthii, O. nivara, O. rufipogon, O. longistaminata, O. meridionalis, O. glumaepatula and 169 accessions from ten other wild species having CC, FF, EE, BBCC and CCDD genomes against two most recently evolved Xoo pathotypes viz. PbXo-10 and PbXo-8 in Punjab state of India, for two constitutive years 2014 and 2015. Based on 2 years of data, four accessions of O. glaberrima (IRGC102206, IRGC1022445, IRGC102512 and IRGC102520) and two of the O. longistaminata accessions (IRGC92624 and IRGC101754) were identified with immune reaction against PbXo-8. For PbXo-10, O. longistaminata showed large number of accessions with complete to partial resistance followed by O. rufipogon (8), O. nivara (2), O. punctata (2) and O. officinalis (1). Two of the O. longistaminata accessions IRGC92624 and IRGC92644 from Mali were found to have resistance against both the Xoo pathotypes indicating presence of BB resistance gene other than Xa21. These can be transferred to elite cultivars of O. sativa for better management of BB.

Occurrence of drought under rainfed conditions is the foremost factor responsible for yield reduction in soybean. Developing soybean cultivars with an inherent ability to withstand drought would immensely benefit the soybean production in rainfed areas. In the present study, F2 derived mapping populations were developed by crossing drought tolerant (PK 1180, SL 46) and susceptible (UPSL 298, PK 1169) genotypes to investigate the inheritance of seedling survival drought mechanisms and to identify simple-sequence repeat (SSR) markers associated with them, using bulked segregant analysis. Parents as well as a F2 derived mapping population were screened for drought tolerance based on seedling survivability under controlled conditions. Segregation analysis of F2 population derived from a cross between PK 1180 × UPSL 298 was previously shown to have a 3:1 tolerant to susceptible ratio and a probability of 0.61 at a χ2(3:1) value of 0.258. This was confirmed in another F2 population derived from a cross between PK 1169 × SL 46 with a χ2(3:1) value of 0.145 obtained at a probability of 0.70. One SSR marker Satt277 showed polymorphism between contracting bulks (tolerant and susceptible) out of 50 polymorphic markers identified during parental polymorphism. Single marker analysis suggested that the marker, Satt277 is linked to seedling survival drought tolerance and is located on chromosome linkage group C2 (chr 6) with a map distance of 3.40 cM. The tolerant genotypes identified could be used as a donor in soybean improvement programs. The marker identified can be used in marker-assisted selection while screening large collection of germplasm.

The genetic diversity of 39 Iranian black cumin (Nigella sativa L.) landraces was analysed using 14 polymorphic Start Codon Targeted (SCoT) markers. A total of 106 bands ranging from 3 (for SCoT70) to 13 (for SCoT23) were observed. Of them, 33 (31%) bands were polymorphic with a mean of 1.65 bands per primer. Polymorphism information content (PIC) per primer ranged from 0.035 (for SCoT12) to 0.133 (for SCoT70), with an average of 0.078. Besides PIC, Simpson's diversity (D) index was also calculated for each SCoT marker as an indication of discrimination power across population. The D index was used to adjust the PIC of the SCoT markers. As the adjusted PIC (PICD= PIC × D) was calculated based on both the PIC and the rate of band dispersion, this reflected the informativeness of a dominant marker more precisely than PIC. Genetic relationships were estimated using Jaccard's similarity coefficient-generated values between different pairs of genotypes that varied from 80 to 97% with an average of 88%. These coefficients were applied to construct a dendrogram using the UPGMA algorithm. A high genetic similarity was observed that might be due to the fact that N. sativa is a self-pollinated plant not originated from Iran and might have been imported from the Mediterranean regions.

Out-crossing between genetically modified herbicide-tolerant (GMHT) and non-GM rape cultivars was studied using GMHT source field plots of approximately 0.8 ha. Levels of cross-pollination between adjacent fully fertile rape varieties declined rapidly with increasing distance from the interface between plots. A varietal association with low levels of male sterility showed higher levels of out-crossing than other varieties. Out-crossing data were used to compare negative exponential and inverse power-law models for their fit to describe the observed relationship between cross-pollination and distance from source. Results showed that the inverse power-law model provided a better fit of the data.

Substantial variation in phenotypic traits and ISSR fingerprinting was observed among twenty populations of Achillea fragrantissima (Forssk.) Sch. Bip. in Egypt. Such variation was reflected in the clustering of the examined populations into two major groups: one representing populations in the mountainous area of South Sinai and the other comprising populations growing at low elevations in the middle of Sinai and the desert west of the Suez Canal from Suez in the east to Cairo in the west. Five populations in the eastern part of Sinai near Nuwieba and Taba on the Gulf of Aqaba were loosely assigned to the first group. The populations growing at high elevations in South Sinai, under lower temperature and higher humidity, were characterized by a higher number of total and polymorphic ISSR markers compared with other populations. Unique ISSR markers were more often observed in the fingerprinting of seven populations including five populations growing in the high mountains of Saint Catherine in South Sinai and two populations growing at low elevations but at Wadi Hof south-east of Cairo. Interestingly, unique bands were found in the populations that possessed traits associated with larger plant size and seed yield as well as better vigour. These are important criteria for the selection of A. fragrantissima populations for conservation and sustainable commercial use.

Hordeum chilense Roem. et Schultz. is a diploid wild barley native to Chile and Argentina. The high crossability of this species with other members of the Triticeae tribe promoted the development of the new species × Tritordeum Ascherson et Graebner. Hexaploid tritordeum was developed from the hybrid derived from the cross between H. chilense (used as female parent) and durum wheat. The interest of H. chilense is based on the presence of traits potentially useful for wheat breeding, including high endosperm carotenoid content, septoria tritici blotch resistance and abiotic stress tolerance. Besides, the variability at cytoplasm level is also important in this species. The development of common wheat–H. chilense alloplasmic lines (nucleus from wheat and cytoplasm from H. chilense) results in fertile or male sterile genotypes, depending on the accession donating the cytoplasm. Furthermore, these alloplasmic lines constitute an ideal system for deepening our knowledge on nuclear–cytoplasm interactions. In conclusion, H. chilense is an interesting source of variability for wheat breeding.

The Drake Passage arises as a likely route for gene flow into Antarctica, as it is the shortest path between this continent and the rest of the world. Despite this, long-distance dispersion into Antarctica could be particularly complex for terrestrial biota. To compare the levels of genetic diversity between Antarctic and South American populations of the Antarctic pearlwort, Colobanthus quitensis, we conducted the first estimation of genetic diversity in this species using amplified fragment length polymorphism. Four populations across the Drake Passage were selected and their genetic composition was characterized. Differences among the levels of genetic diversity were found between the populations analysed as well as between their allelic identities. However, interestingly, their spatial distribution across the Drake Passage suggests a north-to-south gradient of increasing genetic diversity.

The narrow genetic base of existing commercial oil palm cultivars has prompted oil palm breeders to give increased importance to augmenting these genetic resources because the sustainable development of the crop depends largely on the availability of genetic diversity and its use. Therefore, the purpose of this study was to conduct a molecular characterization of an ex situ collection of oil palm Elaeis guineensis Jacq. populations from the Republic of Cameroon using microsatellite molecular markers. Overall, 31 simple sequence repeats were polymorphic, with a total of 223 alleles, 78.4% of which were found at low frequency. The total genetic diversity was relatively high (HT= 0.673). The genetic differentiation between geographical regions was low (GST= 0.023, P= 0.001), and between families it was high (GST= 0.166, P= 0.001), showing greater variation between families than among geographical regions. The molecular data indicate that genetic diversity among the genotypes evaluated is mainly distributed within regions, suggesting that there is no isolation by geographical distance and that all the sampled individuals form a single diverse population. Therefore, it was concluded that a relatively low number of accessions (120 in the analysed case) that includes at least one representative of each family would allow us to efficiently collect almost the entire genetic diversity of Cameroon within the collection studied. This will allow for the efficient use of genetic resources and a reduction in morpho-agronomic characterization costs.

Drought stress ‘particularly at seedling stage’ causes morpho-physiological differences in wheat which are crucial for its survival and adaptability. In the present study, 209 recombinant inbred lines (RILs) from synthetic wheat (W7984)× ‘Opata’ (also known as SynOpRIL) population were investigated under well-watered and water-limited conditions to identify quantitative trait loci (QTL) for morphological traits at seedling stage. Analysis of variance revealed significant differences (P < 0.01) among RILs, and water treatments for all traits with moderate to high broad sense heritability. Pearson's coefficient of correlation revealed positive correlation among all traits except dry root weight that showed poor correlation with fresh shoot weight (FSW) under water-limited conditions. A high-density linkage map was constructed with 2639 genotyping-by-sequencing markers and covering 5047 cM with an average marker density of 2 markers/cM. Composite interval mapping identified 16 QTL distributed over nine chromosomes, of which six were identified under well-watered and 10 in water-limited conditions. These QTL explained from 4 to 59% of the phenotypic variance. Six QTL were identified on chromosome 7B; three for shoot length under water-limited conditions (QSL.nust-7B) at 64, 104 and 221 cM, two for fresh root weight (QFRW.nust-7B) at 124 and 128 cM, and one for root length (QRL.nust-7B) at 122 cM positions. QFSW.nust-7B appeared to be the most significant QTL explaining 59% of the phenotypic variance and also associated with FSW at well-watered conditions. These QTL could serve as target regions for candidate gene discovery and marker-assisted selection in wheat breeding.

South American tomato moth, Tuta absoluta, a serious pest of tomato (Solanum lycopersicum) in tropics and subtropics, is rapidly spreading world over. Twenty one wild/cultivated/advanced breeding lines of tomato were screened for resistance to T. absoluta under greenhouse conditions (choice bioassay) and promising genotypes were evaluated further for their antibiosis activity through no choice bioassay under in-vitro conditions. From 21 genotypes screened, six wild accessions viz., S. pennellii (LA 1940); S. chilense (LA 1963); S. arcanum (LA 2157); S. lycopersicum (LA1257) and S. corneliomulleri (LA 1292, LA1274) were relatively resistant to T. absoluta based on mean percent damage and were further studied under in-vitro conditions. These genotypes recorded relatively more larval mortality, prolonged larval and pupal duration with reduced adult emergence of T. absoluta. Among these six genotypes, S. pennellii (LA-1940) showed resistance both under choice and no choice bioassays with higher number of type IV trichomes, highest total flavonoids and phenols. In general, glandular trichomes (GTs) (type I, IV, VII) showed negative correlation in different genotypes of tomato with reference to larval number/plant, percent damage and adult activity, whereas type V (non-GTs) showed negative correlation with number of larvae/plant.

Crop wild relatives (CWR) are valuable gene pools for crop improvement and offer unique potential and opportunity for enhancing food security and adaptation to climate change. However, current actions towards conservation of plant genetic resources in Zambia do not adequately cover CWR occurring in the country. The article describes the process leading to the development of a national strategic action plan (NSAP) for the conservation and sustainable use of priority CWR in Zambia. Based on 59 prioritized crops, a partial checklist of 459 CWR taxa was generated from the national flora checklist of 6305 taxa. The generated CWR taxa were prioritized based on the socio-economic value of the related crop, their utilization potential in crop improvement, relative distribution and threat status to produce 30 prioritized CWR taxa. Occurrence data were compiled for all CWR inventory taxa and used in spatial analyses to establish species distribution, species richness, gaps in in situ conservation and genebank collections, and to identify priority sites for in situ conservation and ex situ collecting. Consistent with the national developmental agenda, along with the contribution of national stakeholders, spatial analyses of occurrence data of priority CWR taxa are valuable input for the development of the NSAP for the conservation and sustainable use of the priority CWR.

Red bean, also known as azuki bean [Vigna angularis (Willd.) Ohwi & Ohashi], belongs to a group of legumes (family Fabaceae). The name azuki is a transliteration of the native Japanese name from the Chinese word Shōzu, which means small bean. In Korea, it is known as pat. In total, 178 red bean accessions were taken to analyse the genetic diversity, population structure and gene flow using 39 polymorphic simple sequence repeat markers. A total of 431 alleles were detected, with an average of 11 alleles per locus, among the 178 tested red bean accessions. Forty-six specific alleles were identified with 20 loci. Locus CEDG090 had the highest number (n = 22) of alleles, whereas only two alleles were observed at loci CEDG144 and CEDC018. The proportion of different alleles for microsatellite loci was analysed using a microsatellite toolkit. In locus CEDG029, one allele was shared in all the three groups of varieties and species, and three alleles were shared between the wild ancestors and cultivated varieties, while in locus CEDG090, one allele was shared in all the three groups and 12 alleles were shared between the wild ancestors and cultivated varieties. Our findings describe the genetic relationships and population structure of the red bean in Korea and will be useful for designing effective breeding programmes and broadening the genetic base of commercial varieties. Moreover, the results demonstrate substantial gene flow from the red bean to nearby wild relatives in a given region.

Indian gooseberry, popularly known as amla, is harvested from two species of Phyllanthus, namely Phyllanthus emblica and Phyllanthus indofischeri. Although these two species are characterized on vegetative and reproductive features, development of reliable and authentic identification based on molecular approaches is necessary for precise identification. Most of the commercial orchards of Indian gooseberry are established with mixture of the two species. Introduction of unauthentic clones and seedlings in the establishment of orchards and differences in the growth of these two species under different elevations are the major constrains in identification. The present work aims to study the changes in vegetative and reproductive features under different elevations and to develop a simple molecular tool for precise identification of these two species based on single nucleotide polymorphisms in trnL (UAA) intron sequences of chloroplast DNA. Genomic DNA of leaf tissues of P. emblica and P. indofischeri, collected from the authentic source, was subjected to polymerase chain reaction amplification using trnL (UAA) intron gene-specific primers. The amplified product with the molecular weight of 540 bp was sequenced and used as reference for identification of these two species. The trnL (UAA) introns sequenced from 60 individual trees in three different orchards were compared with trnL (UAA) intron of authentic samples, and confirmed that these orchards have 76% population of P. indofischeri and remaining 24% is occupied by P. emblica. Thus, trnL (UAA) intron is a potential DNA marker for precise identification of these two species. The importance of the present study and its practical application in genetic improvement of Indian gooseberry are discussed.

Galantamine (GAL), a morphine-like alkaloid produced by some members of the Amaryllidaceae plant family, is a possible therapeutic agent in Alzheimer's disease because of its central cholinergic effects. GAL has been extracted from the plant sources or produced synthetically for pharmaceutical use. Limited supply of the natural source and high cost of synthetic production has led to a search for alternative sources of this valuable compound. In the present study, a total of six Galanthus nivalis populations (GNPs) and 11 Narcissus tazetta populations (NTPs) were collected across different regions of Iran and were then subjected to the high-performance liquid chromatography analysis for their GAL quantification. The GAL content ranged from 0.05 to 0.36 mg/g dry weight (DW) in the bulbs of GNPs, and from 0.03 to 0.33 mg/g DW in the bulbs of NTPs. Maximum content of GAL (0.36 and 0.33 mg/g DW) was measured in the Zirab population of G. nivalis and Ghaemshahr population of N. tazetta, respectively. Our results provided a suitable material for further agronomical and biotechnological strategies for enhanced production of valuable GAL compound on a large scale.

This study reviews the various conservation strategies applied to the four redwood species, namely coast redwood (Sequoia sempervirens), Sierra redwood or giant sequoia (Sequoiadendron giganteum), dawn redwood (Metasequoia glyptostroboides) and South American redwood or alerce (Fitzroya cupressoides), which are endemic in the USA, China and South America, respectively. All four redwood genera belong to the family Cupressaceae; they are monospecific, share a number of common phenotypic traits, including red wood, and are threatened in their native ranges due to human activity and a changing climate. Therefore, the management objective should be to conserve representative populations of the native species with as much genetic diversity as possible for their future survival. Those representative populations exhibiting relatively high levels of genetic diversity should be selected for germplasm preservation and monitored during the conservation phase by using molecular markers. In situ and ex situ strategies for the preservation of germplasm of the redwoods are discussed in this study. A holistic in situ gene conservation strategy calls for the regeneration of a large number of diverse redwood genotypes that exhibit adequate levels of neutral and adaptive genetic variability, by generative and vegetative methods for their preservation and maintenance in their endemic locations. At the same time, it would be desirable to conserve the redwoods in new ex situ reserves, away from their endemic locations with similar as well as different environmental conditions for testing their growth and survival capacities. In addition, other ex situ strategies involving biotechnological approaches for preservation of seeds, tissues, pollen and DNA in genebanks should also be fully exploited in the face of global climate change.
