Temnocephalids are ectosymbionts of various freshwater animals. A species tentatively identified as Temnosewellia aff. vietnamensis (Platyhelminthes: Rhabdocoela: Temnocephalidae) is reported based on materials collected from the body surface of the freshwater crabs Eriocheir japonica (Brachyura: Varunidae) and Geothelphusa exigua (Potamidae) in Kagoshima, southern Japan. The temnocephalid is characterized as follows: the cirrus composed of a cone-shaped shaft and a cylindrical introvert 42–77 μm long; the introvert covered with approximately 30 vertical rows of fine sharp spines; the four seminal receptacles; and a long, curved oviduct with vaginal gland; a pair of gland cells (Haswell's cells) present anterior to the excretory ampullae. Bayesian inference trees using partial nuclear 28S rDNA (28S) and mitochondrial cytochrome c oxidase subunit I (COI) genes supported that the specimens collected from both crab species are conspecific but these also showed the geographical variations among them on both 28S and COI. The previous records of the genus Temnosewellia in East to South Asian countries are assembled and shown on the map (fig.7, this paper).

Temnocephala axenos Monticelli, 1898 was described based on specimens from an unidentified host collected in Blumenau, Santa Catarina, Brazil. Information about type locality was imprecise and the host was later identified as Aegla laevis (Latreille, 1818). However, it is known that A. laevis is not present on the eastern side of the Andes. Also, only histological preparations from one specimen studied by Monticelli are currently available in the Museum für Naturkunde Berlin, but it showed none of the taxonomic characters needed for the characterization of the species. Although the updated description of the species based on Uruguayan specimens, neither the author nor the several previous studies about the species showed a search for the type material, a resolution for the misidentification of the type host or the imprecise type locality due to the subsequent geographical division of the municipality cited in the description. The Uruguayan specimens were not even geographically close to the type locality and a neotype was not designed to validate the species’ taxonomic status again. Specimens from Santa Catarina and Paraná States, Brazil, were studied, as well as restudied Argentinean specimens. The new data were compared with the update description of the species. The historical background and the discussion about geographical origins and hosts of the species, as well as a designation of a neotype, allow comparative material of the type locality and type host to exist, eliminating doubts about the identification of T. axenos.

TPase and SDII activities were studied in various tissues of Ceylonocotyle scoliocoelium maintained in vitro in medium A for 15 days, using standard histochemical techniques. Results were compared with the normal activities of these enzymes in the tissues of worms freshly collected. Low ATPase and SDH enzyme activities in the tissues of cultured flukes indicated their low metabolic rate. Intensity of enzymatic reactions was also found to be tissue specific. The suitability of culture medium ‘A’ for the in vitro cultivation of amphistomes is discussed.

Nilocotyle duplicisphinctris sp. n. was recovered from the stomach of Hippopotamus amphibius in Ethiopia. The average body size (L × W) is 3.6 × 1.2 mm; pharynx 0.5 × 0.3, Dicranocoelium type; other diameter of the acetabulum 0.85, Nilocotyle type; anterior testis 0.34 × 0.49; posterior testis 0.32 × 0.27; ovary 0.27 × 0.19; genital opening 0.30 × 0.34, Duplicis phinctris type and eggs 0.15 × 0.07. The most characteristic specific feature is the presence of a double sphincter along the genital atrium.

The different stages of the life cycle of parasites are important components of ecosystems. Changes in environmental conditions may affect free-living stages, host–parasite interactions and ecosystem functioning. The larvae of Chordodes nobilii, which belongs to the parasitic phylum Nematomorpha, are susceptible to extreme temperatures and different pollutants, but the effects of pH and moderate temperature variations have not been evaluated yet. Our objective was to assess the effect of temperature, pH and their interaction on the infectivity of C. nobilii larvae to Aedes aegypti larvae over time. Larvae were treated with factorial combinations of temperature (18, 23 and 28°C), pH (7, 8 and 9) and time periods (24 and 48 h). Results show a highly significant interaction among all variables. The highest infectivity was recorded at 18°C and pH 7 at 24 and 48 h, and the lowest one at 28°C and pH 8 at 24 and 48 h. Infectivity differed significantly among the three temperatures only at pH 8 and 48 h, decreasing with increasing temperature. Our study is the first report of the effect of pH on a Nematomorpha species and suggests that the infectivity of C. nobilii larvae may be affected negatively by an increase in temperature and its interaction with pH and time. Since parasites must be considered for a better understanding of the effects of stressors on freshwater ecosystems, our results may help in the design and analysis of studies of anthropogenic impact.

The success of Phasmarhabditis hermaphrodita (Schneider) Andrássy (Rhabditida: Rhabditidae) as a biological control agent of molluscs has led to a worldwide interest in phasmarhabditids. However, scant information is available on the lifecycle development of species within the genus. In the current study, the development of P. hermaphrodita, Phasmarhabditis papillosa, Phasmarhabditis bohemica and Phasmarhabditis kenyaensis were studied using ex vivo cultures, in order to improve our understanding of their biology. Infective juveniles (IJs) of each species were added to 1 g of defrosted homogenized slug cadavers of Deroceras invadens and the development monitored after inoculated IJ recovery, over a period of eight–ten days. The results demonstrated that P. bohemica had the shortest development cycle and that it was able to produce first-generation IJs after eight days, while P. hermaphrodita, P. papillosa and P. kenyaensis took ten days to form a new cohort of IJs. However, from the perspective of mass rearing, P. hermaphrodita has an advantage over the other species in that it is capable of forming self-fertilizing hermaphrodites, whereas both males and females are required for the reproduction of P. papillosa, P. bohemica and P. kenyaensis. The results of the study contribute to the knowledge of the biology of the genus and will help to establish the in vitro liquid cultures of different species of the genus.

Phyllodistomum pepirense n. sp. is described from the urinary bladder of Hoplias malabaricus (Bloch, 1794), sampled in the Jacaré-Pepira River in São Paulo state, Brazil. The isolates of the new species were recovered as a monophyletic group in the phylogenetic analysis of the 28S rRNA gene, which showed the new species as the sister taxa of Phyllodistomum virmantasi Pinacho-Pinacho, Sereno-Uribe, Hernández-Orts, García-Varela & Pérez-Ponce de León, 2021, a species sampled from an eleotrid fish in Southeastern Mexico. The new species differs morphologically from P. virmantasi by having a larger body size, slightly lobed testes and ovary, a mostly intercaecal uterus, slightly diverticulated caeca, and vitelline masses irregularly shaped. The new species is also readily distinguished from other species of Phyllodistomum Braun, 1899 reported from freshwater fishes in Brazil – namely, P. rhamdiae Amato & Amato, 1993 and P. spatula Odhner, 1902. The new species is herein described based on morphological characteristics, molecular data from D1–D3 domains of the 28S rRNA gene, host association, and geographical distribution.

In this study, a new dactylogyrid species is described from the gills of two ariid fish species of the Brazilian coast, Genidens barbus and Genidens genidens, by combining morphological characters with partial 18S rDNA sequences. The new species can be distinguished from its congeners by the following morphological characteristics: a male copulatory organ (MCO), a coiled tubular shaft of around one counterclockwise ring, base with a sclerotized cap; MCO with tapered distal region; a T-shaped accessory piece; a vagina, a cup-shaped vaginal vestibule, slightly sclerotized; and a pharynx comprising a muscular, glandular bulb. Supplementary taxonomic data for Chauhanellus velum from Sciades couma (type-host) are also presented and new 18S rDNA sequences of Chauhanellus spp. from other ariid fish from South America are provided. Phylogenetic analyses based on partial 18S rDNA gene sequences placed Chauhanellus riograndinensis n. sp. and Chauhanellus velum as two early divergent lineages within Chauhanellus from South America. Finally, a way to test the monophyly of Chauhanellus and Hamatopeduncularia is also discussed, which may be useful for future studies.

Molecular techniques are an alternative for the diagnosis of strongyloidiasis, produced by Strongyloides stercoralis. However, it is necessary to determine the best amplification target for the populations of this parasite present in a geographical area and standardize a polymerase chain reaction (PCR) protocol for its detection. The objectives of this work were the comparison of different PCR targets for molecular detection of S. stercoralis and the standardization of a PCR protocol for the selected target with the best diagnostic results. DNA extraction was performed from parasite larvae by saline precipitation. Three amplification targets of the genes encoding ribosomal RNA 18S (18S rDNA) and 5.8S (5.8S rDNA) and cytochrome oxidase 1 (COX1) of S. stercoralis were compared, and the PCR reaction conditions for the best target were standardized (concentration of reagents and template DNA, hybridization temperature, and number of cycles). The analytical sensitivity and specificity of the technique were determined. DNA extraction by saline precipitation made it possible to obtain DNA of high purity and integrity. The ideal target was the 5.8S rDNA, since the 18S rDNA yielded non-reproducible results and COX1 never amplified under any condition tested. The optimal conditions for the 5.8S rDNA-PCR were: 1.5 mM MgCl2, 100 μM dNTPs, 0.4 μM primers, and 0.75 U DNA polymerase, using 35 cycles and a hybridization temperature of 60 °C. The analytical sensitivity of the PCR was 1 attogram of DNA, and the specificity was 100%. Consequently, the 5.8S rDNA was shown to be highly sensitive and specific for the detection of S. stercoralis DNA.

The present paper describes the taxonomy of two new monogeneans, namely, Thaparocleidus longiphallus sp. n. and T. siloniansis sp. n., based on morphological, morphometric and molecular biological analysis, collected from the fish Wallago attu (Bloch & Schn.) and Silonia silondia (Ham.), respectively, at Meerut, UP, India. Genetic characterization of the two new species is based on sequence analyses of the rDNA 28S gene using neighbour-joining and maximum-parsimony techniques. These methods are congruent in depicting T. longiphallus sp. n. and T. siloniansis sp. n. as closely related species, but distinct from each other and forming a subclade with other species of the genus Thaparocleidus Jain, 1952. Secondary-structure models of the large subunit rDNA of the two species were also predicted using a combined comparative and thermodynamic approach. Molecular morphometric and phylogenetic relationships of the isolates of the Thaparocleidus species are discussed in detail.

The mallard Anas platyrhynchos is the most abundant water bird species in Austria, but there is no record of its helminth community. Therefore, this work aimed to close that gap by recording and analysing the parasite community of a large number of birds from Austria for the first time. A total of 60 specimens shot by hunters in autumn were examined for intestinal parasites. The following taxa were recovered (prevalence given in parentheses): Cestoda: Diorchis sp. (31.7%) and Fimbriarioides intermedia (1.7%); Acanthocephala: Filicollis anatis (5%), Polymorphus minutus (30%) and one cystacanth unidentified (1.7%); Trematoda: Apatemon gracilis (3.3%), Echinostoma grandis (6.7%), Echinostoma revolutum (6.7%) and Notocotylus attenuatus (23.3%); Nematoda: Porrocaecum crassum (1.7%) and one not identified (1.7%). The frequency distribution of parasites showed a typical pattern in which 39 birds (65%) were either not parasitized or were harbouring up to five worms, whereas more intense infestations occurred in a lesser number of hosts. Compared to other studies from central and eastern Europe, an extremely depauperate helminth community, particularly of the cestodes and nematodes, was found. Polymorphus minutus was observed as having highly variable morphology and, therefore, molecular genetic characterization by DNA barcoding was carried out. Species identification was confirmed by comparing data with the reference cytochrome c oxidase subunit 1 gene sequence from P. minutus available in GenBank.

The diagnosis of cystic echinococcosis (CE) is based on imaging. Detection of a focal lesion with morphological characteristics of Echinococcus granulosus sensu lato metacestode is the starting point for the diagnostic workup. In organs explorable with ultrasound (US), this is the method of choice for both aetiological diagnosis of CE and staging of the CE cyst. Staging in terms of lesion morphology is also needed when serology is added to the diagnostic workflow when imaging alone is inconclusive. Finally, staging guides the clinical management of uncomplicated CE, especially in the liver. This commentary provides an overview of the most up-to-date evidence backing the above-mentioned role of US in the diagnosis and clinical management of CE. Finally, we outline future perspectives for the improvement of CE diagnosis.

This study aimed to determine the prevalence of gastrointestinal parasites in alpacas on selected farms in Poland. In July and August 2019 and August 2021, 223 samples from six commercial farms were examined using coproscopic techniques. The total percentage of alpacas infected with intestinal parasites was 57.7%. Eggs of Nematodirus sp. were found in 28.9%, Trichostrongylus sp. in 15.5%, Strongyloides sp. in 13.4%, Camelostrongylus sp. in 11.3%, other strongyle-type in 12.4%, Trichuris sp. in 3.1%, Capillaria spp. in 2.1%, Oesophagostomum sp. in 1.0% and eggs of Moniezia sp. in 1.0% of individuals. Oocysts of Eimeria macusaniensis were found in 8.2%, Eimeria sp. in 4.1%, and Cryptosporidium sp. in 3.1% of animals. Redundancy analysis showed that parasites and their number in faeces were related to the individual’s country of origin, sex and age. Females had significantly more eggs of parasites than males. More significant parasite infection was recorded in younger individuals. Moreover, the most infected were individuals from Germany. Some of the described parasites in tested alpacas have zoonotic potential. Due to the possibility of introducing parasites native to alpacas and acquiring species parasitising wild and farmed animals in Europe, permanent veterinary monitoring of animals imported from other regions is necessary.

The ingestion of soil in parks and public places containing eggs of Toxocara may constitute a significant health risk, particularly to children. To determine the most efficient method for extracting eggs from experimentally contaminated soil, two consecutive studies were undertaken. Four techniques, including washing, sieving, vacuum, and the one recommended by the World Health Organization, were evaluated. Recovery rates of over 85% were recorded with both washing and sieving methods. Using the washing technique, all combinations of the four pre-treatment solutions, distilled water, acetoacetic solution pH 5, 0.1 N sodium hydroxide and 1% Tween 20, and seven flotation fluids with different specific gravities (S.G.) ranging from 1.20 to 1.35 were assayed. The association of distilled water and saccharose solution with an S.G. of 1.27 showed the best results, with a recovery rate of 99.91%.

A larvoscopic examination of faeces collected from localities inhabited by chamois in the Tatra National Park (TANAP) in 1997 demonstrated the presence of the lung nematodes Muellerius spp. (likely to be M. tenuispiculatus and M. capillaris) and Neostrongylus linearis. The overall prevalence of lung nematodes in chamois herds in TANAP was 48.4% with prevalences of 45.6% and 11.9% for Muellerius spp. and N. linearis, respectively. No significant differences in lung nematode prevalences were observed in the biotopes of TANAP with prevalence values of 44.9% being recorded in the High Tatras and 58.5% in the Belianske Tatras. Individual species were in equal proportion in both biotopes, although N. linearis was significantly less prevalent (11.2–13.8%). The prevalence of lung nematodes in the High Tatras varied from 25.0 to 84.2% within individual localities, while in the Belianske Tatras it was more proportionate (50.0–85.7%). In the High Tatras, the prevalence of lung nematodes in the chamois herds peaked during August, declining to its lowest in October. A similar prevalence was also recorded for Muellerius species, while the minimum prevalence of N. linearis was found in July. In the Belianske Tatras, the prevalence of lung nematodes including both species of Muellerius peaked in July and gradually decreased until October. On the other hand, N. linearis was most prevalent in October. The mean L1 count per gram faeces was low (7.6 ± 13.2 larvae g-1).

A total of 140 trout were examined from four lakes in north west Ireland, Loughs Craghy, Waskel, Meela and Owennamarve. Eleven species of metazoan parasite were recorded, ten of which were of freshwater origin and Anisakissp. was the only marine species. No acanthocephalan species were found. Diphyllobothrium ditremum, D. dendriticum and Anisakissp. were the only allogenic species. Lough Owennamarve had the largest component community with nine species recorded. Lough Meela, the only brackish water lake had the smallest component community with six species recorded. Lough Waskel and Craghy had intermediate component community values. Lough Meela showed the lowest levels of similarity to any of the other lakes. Values for the Shannon-Wiener diversity index were highest for Lough Owennamarve and lowest for Lough Meela and intermediate for the other two lakes. Lough Meela had the highest values for the Simpson's dominance index and the Berger-Parker index. Lough Owennamarve had the highest Brillouin's index and Lough Meela had the lowest. The use of multiple regression techniques to control for the potentially confounding effect of geographical distance as a predictor of community similarity was discussed. No parasite species found could be considered as regionally and locally abundant and therefore described as a core species. The helminth community appears to have some degree of predictability, but chance colonization events are also important.

Seventy female lambs (6–7 months old) which were exposed to natural infections of Haemonchus contortus were designated as responders or non-responders on the basis of 10 weekly cumulative faecal egg counts. Selected responder and non-responder lambs were treated with ivermectin, housed separately and 6 weeks post-housing, seven lambs from each group were given a trickle infection of Haemonchus contortus at 1000 L3 daily for 5 days per week up to 2 weeks and examined weekly for 10 weeks after first infection. Analysis of data revealed significantly lower mean faecal egg counts and non-significantly less weight loss in responder than non-responder lambs. Mean values of haemoglobin, packed cell volume, total serum protein and peripheral eosinophil counts were significantly higher in responders than non-responders. In contrast, serum pepsinogen concentration was significantly less in responders than in non-responders. At 10 weeks post-infection, there were fewer pathological lesions and significantly lower worm burdens in responders than in non-responders. These results demonstrate a distinct resistance in responders to Haemonchus contortus infection.

Acanthocephalans are a group of obligate endoparasites that alternate between vertebrates and invertebrates to complete their life cycles. Occasionally, the same individual host acts as a definitive or paratenic host for different acanthocephalan species. In this study, acanthocephalans were sampled in marine fish in three localities of the Yucatán Peninsula; adults and cystacanths were recovered from the intestine and body cavity, respectively, of Haemulon plumierii from off the coast of Sisal, Yucatán. Ribosomal DNA sequences (small and large subunits) were used to test the phylogenetic position of the species of the genus Dollfusentis, whereas the mtDNA gene cox 1 was used for assessing species delimitation. The cox 1 analysis revealed an independent genetic lineage, which is recognized herein as a new species, Dollfusentis mayae n. sp. The new species is morphologically distinguished from the other six congeners by having a cylindrical proboscis armed with 22–25 longitudinal rows bearing 12 hooks each. The cystacanths were morphologically identified as Gorgorhynchus medius by having a cylindrical trunk covered with tiny irregular spines on the anterior region, and a cylindrical proboscis armed with 17–18 longitudinal rows of 21 hooks each; small and large subunit phylogenetic analyses yielded G. medius within the family Isthomosacanthidae, suggesting that Gorgorhynchus should be transferred to this family from Rhadinorhynchidae where it is currently allocated.

Six species of freshwater turtles dominate the Chaco-Pampa Plain in southern South America and their parasites have been relatively understudied, with most records concentrated in Brazil. Particularly in Argentina, there are only scattered records of parasites for most of the turtles that inhabit the region, leaving a large knowledge gap. The purpose of the present contribution is to increase the knowledge of the internal parasites of six species of freshwater turtles from Argentina, after 15 years of fieldwork, by providing new hosts and additional geographic records for many host-parasite relationships. Some molecular sequences of the studied parasites were provided as a tool for better species identification. We processed 433 stomach and fecal samples from live individuals and visceral and soft tissue samples from 54 dissected turtles collected from a wide range and different ecoregions. We found 6230 helminths belonging to 18 taxa (one cestode, 11 digeneans and six nematodes). Fourteen new parasite-host associations are reported here, and for the first time parasites are recorded for Phrynops williamsi. This work contributes significantly to the knowledge of the parasitofauna in freshwater turtles in Argentina, providing a detailed list of parasites present in each turtle species and reporting molecular characters for future studies.
