Since the exogenous compound tris(hydroxymethyl)aminomethane (Tris) showed a potent chemoattractant activity for Brugia pahangi infective third-stage larvae (L3), it was assumed that, in natural infection to a host, filarial L3 can be expected to recognize an endogenous Tris-related compound. In addition, a few amino acids have been identified as water-soluble attractants for second-stage juveniles of Meloidogyne incognita, a plant parasitic nematode. Therefore, the present study assesses the in vitro chemotactic responses of B. pahangi L3 to Tris-related compounds and amino acids using an agar-plate assay. Among Tris-related compounds, 2-amino-1,3-propanediol (APD) and 2-amino-2-methyl-1,3-propanediol (AMPD) exhibited a potent chemoattractant activity for filarial L3 at a level similar to Tris. Furthermore, arginine (Arg) was identified as a potent attractant for filarial L3 among amino acids. In addition, filarial L3 were attracted to Arg, APD and AMPD in mild alkaline conditions rather than acidic conditions. The chemoattractant activity of the three compounds for filarial L3 was observed in concentrations between 6.3 and 200 mm. This is the first report to demonstrate that Arg, APD and AMPD are potent chemoattractants for B. pahangi L3. Endogenous Arg and APD, in particular, may be involved in the regulation of the chemotactic behaviour of filarial L3 in the infection to a host. The present results will help to elucidate the mechanism of filarial skin-penetrating invasion of a host.

We describe the first case of angiostrongyliasis in a water rat, Hydromys chrysogaster, a large rodent adapted to aquatic life, which is endemic to Australia, New Guinea and adjacent islands.

The prevalence of helminths in pigs was investigated in five rural communities situated on the embankment of Dongting Lake in Zhiyang County, Hunan Province, People's Republic of China, in an area known to be endemic for Schistosoma japonicum. The helminth prevalences identified on the basis of faecal egg count analysis were: Oesophagostomum spp. (86.7%), Ascaris suum (36.7%), Metastrongylus spp. (25.8%), Strongyloides spp. (25.8%), Trichuris suis (15.8%), Globocephalus spp. (6.7%), Gnathostoma spp. (4.2%), Schistosoma japonicum (5.0%) and Fasciola spp. (1.3%). Post mortem examinations of a small number of pigs depositing eggs of different helminth species revealed the presence of Oesophagostomum dentatum, O. quadrispinulatum, A. suum, Metastrongylus apri, M. pudendotectus, T. suis, G. hispidum and Ascarops dentata. Prevalences of all helminths, with the exception of Oesophagostomum spp., were higher in young pigs (< 8 months old) compared with adult pigs. Prevalences of trematodes were very low, especially for S. japonicum which had decreased dramatically compared with previous reports from this area of P.R. China, whereas prevalences of nematodes were generally in agreement with those reported from other Yangtze River Provinces. Results from helminth prevalence studies in pigs, conducted in other provinces of P.R. China between 1987 and 1997, are presented and discussed. It was concluded that a government helminth control programme, implemented in 1995 to control S. japonicum infection in pigs in Hunan Province, may have resulted in a greatly reduced prevalence of S. japonicum in pigs in this region.

Acanthocephalans constitute a small taxonomic group related to rotifers and specialized in a parasitic lifestyle. Anurans act as paratenic and definitive hosts and infections always occur trophically. Our objective is to describe and summarize the richness of acanthocephalans in Neotropical anurans. We conducted a literature review in the main research databases, compiling data published until August 2021. We identified 66 articles with records of acanthocephalan-anuran association, 53.03% were carried out in Brazil. We detected 108 species of anurans from 11 families parasitized by acanthocephalans. With the exception of Bufonidae, Hylidae and Leptodactylidae, which are relatively well-studied families, interaction with acanthocephalans remains largely unexplored for most anuran species. We found six families of acanthocephalans: Centrorhynchidae, Echinorhynchidae, Oligacanthorhynchidae, Cavisomidae, Neoechinorhynchidae and Plagiorhynchidae. Centrorhynchidae and Echinorhynchidae presented the largest number of taxa associated with anurans. The largest number of records corresponded to acanthocephalans in the larval stage (cystacanths), for which anurans act as paratenic hosts. We observed a lack of specific taxonomic resolution in the identifications of most reports, because a large part of the records in the larval stage make morphological identification difficult. Brazil, Mexico, Paraguay, Argentina, Ecuador and Peru are the countries with the most records, while Costa Rica, Venezuela, Colombia, Chile and Uruguay exhibited the lowest publication numbers, resulting in gaps in the distribution of acanthocephalans. We expanded the known number of anuran species parasitized by acanthocephalans, compared to the last published review. Overall, we aim to contribute to the understanding of diversity within this intriguing but understudied group.

Habitats characterized by geographic isolation such as islands have been studied using different organisms as models for understanding the dynamic and insular patterns of biodiversity. Determinants of parasite richness in insular host populations have been conducted mainly with mammals and birds, showing that parasite richness decreases in insular areas. In the present study, we predicted that the type of environment (insular or continental) can influence the richness, diversity and abundance of parasites associated with the endemic frog Haddadus binotatus (Spix, 1824). We sampled frogs in two insular and two mainland fragments to survey their helminth parasites. The total richness was composed of 15 taxa of Nematoda and two of Acanthocephala, and the community composition of the two islands had more similarities between them than the two mainland localities. The insular effect was positive for richness and abundance of helminths, and no significant effect was observed on helminth diversity – even the mean diversity presented high numbers for the islands. We presumed that insular hosts could have lost some parasites in the colonization process when these continental islands were separated from the mainland, approximately 11,000 years ago. However, the high richness and abundance on islands can be explained by an epidemiological argument, which considers high population density due to insularity and other features of the host as factors that increase parasite transmission success among individuals.

The metacercaria of Indodidymozoon pearsoni (Digenea: Didymozoidae) is identified by DNA sequencing of second internal transcribed spacer (ITS2) ribosomal DNA and described. Its development into the adult is documented. Morphological features that may assist the matching of didymozoid metacercariae to adults are discussed. Those that may be of greatest use are the size of the oral and ventral suckers and the presence or absence of a chamber at the posterior extremity of the oesophagus.

Ligula (Cestoda: Pseudophyllidea) infections in gudgeon (Gobio gobio) and roach (Rutilus rutilus) differ markedly in the pathology that is observed in the host, particularly with respect to a tissue response and the extent of inhibition of gonadal development. The entire internal transcribed spacer (ITS) region (ITS-1, 5.8S and ITS-2) and the large subunit domains D1–D3 were sequenced and compared in parasites from these fish from Lough Neagh, Northern Ireland, together with a single specimen from minnow (Phoxinus phoxinus) from Wales. Sufficient differences were observed between parasites from R. rutilus and G. gobio to support the suggestion that they may represent different strains/species. In contrast, Ligula from P. phoxinus closely resembled those from R. rutilus. Ligula infections in G. gobio were recorded prior to the introduction of R. rutilus. The co-existence of separate strains or species of Ligula in Lough Neagh probably resulted from the introduction of R. rutilus to these waters, correlated with an increase in the number of great crested grebes (Podiceps cristatus).

The northern section of the Caspian Sea and lower reaches of the Zhaiyk (Ural) River is an important fishery for Kazakhstan. In the present study, a total of 1597 individuals of ten fish species were analysed. The fish were caught over three years, from 2018 to 2020. For each species studied – Abramis brama, Alosa saposchnikowii, Atherina boyeri caspia, Carassius gibelio, Chelon auratus, Cyprinus carpio, Leuciscus aspius, Rutilus caspius, Sander marinus and Sander volgensis – between 100 and 200 individuals were examined. A series of generalized linear models (GLMs) were used to examine the association between individual parasite intensity of infection and the Fulton index, age, year the fish was captured, where the fish was captured (northern Caspian or Zhaiyk River) and sex. For each GLM, the best-fitting probability distribution was used –either Poisson, zero-inflated Poisson, negative binomial or zero-inflated negative binomial. For some fish/parasite species, an increased Fulton index was associated with higher intensities of parasite infection, whilst, for others, the Fulton index decreased with the intensity of parasite infection. This was also true of age-related intensity of infection, with some parasites having an increased intensity of infection with age whilst others had a decreased intensity of infection with age. There was also some evidence of variation in intensity of parasite infection between different years when the fish were caught. For some species of fish that are endemic to both the fresh waters of the Zhaiyk River and the low-saline waters of the northern Caspian, there were variations in intensity of parasite infection between the two environments. The best-fitting probability distribution also gave some information about the dynamics of infection. No fish species had a Poisson distribution of parasites, which is consistent with an entirely random infection process, with all fish being potentially exposed. For some parasites, the distribution was a zero-inflated Poisson, which is consistent with either the fish being exposed to parasite infection or not; and, if exposed, infection was a random process. Other parasites had a negative binomial distribution, consistent with the entire fish population being exposed, but the infection process was clumped or there were variations in the susceptibility of infection between fish. Finally, some of the parasites had a zero-inflated negative binomial distribution, which can be interpreted as part of the fish population not being exposed and the remainder of the population being exposed to a clumped or aggregated infection process and/or a variation in individual susceptibility to infection.

The tegument of schistosomes is a source of many potential anti-Schistosoma vaccine molecules. This work aimed to assess the protective effects of the adult Schistosoma mansoni tegument treated (TT) with sub-curative praziquantel (PZQ), whether in vivo (in vivo TT) or in vitro (in vitro TT), in murine schistosomiasis. In vitro TT and in vivo TT showed great similarity, and they differed from untreated tegument antigen (Teg) in terms of quantity and quality of protein bands on SDS–PAGE. Two immunization trials were performed, each with 50 mice, divided randomly into five groups of 10 mice each: (1) uninfected control mice (UC), (2) infected mice given phosphate buffer saline + adjuvant (PBS + adjuvant), (3) infected, Teg vaccinated, (4) infected, in vivo TT vaccinated, and (5) infected, in vitro TT vaccinated. All the immunizations with antigens induced mixed Th1/Th2 immune responses, as indicated by significantly high (P < 0.001) specific IgG2a and IgG1 levels, with Th1 predominating, as shown by a diminished IgG1/IgG2a ratio, as well as a high serum concentration of IFN-γ, an absence of IL-4 and increased IL-10. In vitro TT gave the most pronounced response. With respect to reduction of total worm burden, relative to PBS + adjuvant mice, in vitro TT achieved the highest significant (P < 0.001) results, followed by in vivo TT and Teg (51.8–57.04%, 44.6–50.2% and 35.2–39.3%, respectively). In scanning electron microscopy studies, all the tested antigens caused tegumental changes in adult worms, with the worst occurring with in vitro TT, such as retracted ventral sucker, an effect on the gynaecophoric canal, and changes to tubercles. In conclusion, in vitro TT, which is cheap to prepare, could be a potential vaccine against S. mansoni.

The Didymozoidae (Trematoda: Hemiuroidea) is among the most speciose trematode families, known from a wide range of marine teleost fishes. Despite their richness, however, didymozoid life cycles are unusually poorly known; only two first intermediate hosts are known, a marine bivalve (Anadara trapezia) and a pelagic gastropod (Firoloida desmarestia). This study uses multi-locus molecular sequence data to identify a novel first intermediate host for the family, a sessile gastropod of the genus Thylacodes Guettard (Vermetidae). The didymozoid infection is not identified to species but, based on molecular phylogenetic analyses, it is close to Saccularina magnacetabula Louvard et al., 2022, which uses a bivalve as a first intermediate host. The distribution of known first intermediate hosts of didymozoids (a bivalve, a holoplanktonic gastropod and a sessile gastropod that feeds with the use of mucus nets) suggests that first intermediate host use within the Didymozoidae has been opportunistically driven by the trophic ecology of potential mollusc hosts and has involved significant host-switching events.

Elaphostrongylus cervi produces a subclinical cerebrospinal disease in many wild and domestic ruminants from Europe, North America and New Zealand and has recently been described in Spain. To determine some aspects of its epidemiology, 121 red deer (Cervus elaphus) from central Spain were sampled during 2000. The prevalence (7%) and mean worm burden (3.8 worms per brain) were similar to the values previously recorded in other European areas. The infection was only detected in young deer during the winter. The estimation of larval production in the faeces was not a reliable method of diagnosing E. cervi infection.

Potato tubers infected with Heterodera marioni having been received at this Institute, it was decided to attempt to infect a small field plot with this eelworm for experimental purposes. The tubers were planted in the plot but the infection never became established. This was thought to be due to the heavy nature of the soil, since H. marioni has been recorded as a field parasite in Britain (Triffitt, 1931), and it is therefore unlikely that climatic conditions were the cause of its failure to become established in this instance. In order to verify this, and to find out how long this parasite can survive in soil in the absence of a host, the following experiment was carried out.

Histochemical tests indicate the presence of tanning precursors namely protein, phenols and phenolase in the vitelline glands of the temnocephalid Paracaridinicola platei. This suggests the occurrence of a tanned egg shell in this organism, a feature it shares with many other parasitic and free living platyhelminths. The large eggs are laid singly and are attached to the gill lamellae of the host.

A Sudanese isolate of Schistosoma bovis from experimentally infected sheep was found to be highly pathogenic to albino mice. Eggs were first found in the livers 42 days after exposure. The distribution of eggs in the liver and small and large intestines changed little during the course of infection. Results arc compared with others using an Iranian isolate of S. bovis, which causes only a mild infection, and S. mattheci.

Buckley and Fairley (1950) working in England found segments of a species of Bertiella passed by an eight year old girl who had lived in Kenya for 14 months during 1946–47. Since only the terminal segments were available for study, a specific identification could not be made, though the authors thought that it was probably Bertiella studeri (Blanchard, 1891). Nelson (1963) has found that baboons in Kenya are commonly infected with species of Bertiella.

Digenean larvae of hermaphroditic generation – cercariae – are known to be polymorphic at genetic and behavioural levels. Cercariae arise as a result of parthenogenetic reproduction of intramolluscan stages, and represent a clone if a snail was infected with a single miracidium. Here we investigated cercarial clones of Himasthla elongata – namely, the infectivity of cercariae with normal (negative) and deviant (positive) photoreaction. In our study, most H. elongata clones showed intraclonal variance in their response to light. The proportion of photopositive cercariae ranged between 0.2% and 60% in different H. elongata clones. Photopositive larvae demonstrated significantly reduced rates of encystment in Mytilus edulis haemolymph in vitro and in young mussels. We discuss the possible mechanisms behind intraclonal variations, such as non-specific genomic rearrangements.

The Asian fish tapeworm (Schyzocotyle acheilognathi) is an important fish parasite with a wide host range that infects over 300 species of fish worldwide. Schyzocotyle acheilognathi has been reported from eastern coastal areas of Australia, but has not been previously reported in Western Australia (WA). During a control program for invasive freshwater fishes in south-western WA, a region with a unique and highly endangered freshwater fish fauna, tapeworms identified as S. acheilognathi from their distinctive scolex morphology were found at a prevalence of 3.3% in goldfish (Carassius auratus), 37.0% in koi carp (Cyprinus carpio haematopterus) and 65.0% in eastern gambusia (Gambusia holbrooki) in a small suburban lake to the north of Perth. For molecular confirmation, the 18S ribosomal RNA gene was targeted at hypervariable V4 region. Koi carp isolates were 100% identical to S. acheilognathi isolated from varying hosts, including the red shiner (Cyprinella lutrensis) and a human sample. Sequences obtained from two eastern gambusia were identified as S. acheilognathi, but formed a discrete cluster and may represent a novel genotype. Isolates from two other eastern gambusia and two goldfish formed a distinct clade with only 91.9% similarity to previously sequenced isolates of S. acheilognathi. This emphasizes the importance of molecular identification methods in addition to morphological identification. The presence and potential for transmission of these parasites in south-western WA may threaten the health of native fishes, which are immunologically naïve to this introduced parasite. Immediate control or containment measures should be implemented to halt the spread of these parasites.

Parasites may affect host demographic characteristics because they can directly or indirectly cause the death of their hosts and/or influence their reproduction. Parasitism is therefore recognized as a factor that influences the composition and structure of populations and communities. One of these behaviours is the compensatory response: the host can compensate for the parasite losses effect, modifying the reproductive effort to enhance fitness. Ovigerus female Palaemonetes argentinus was collected and sorted into two groups according to the degree of development of their embryos: newly spawned embryos and embryos ready to hatch. The number of embryos and their dry weight for each female were determined. All parts of the female body were checked for parasites. The females of P. argentinus were parasitized by Microphalus szidati. We found that parasitized females produce more embryos but had more egg loss during development and the percentage of embryonic loss was higher in the parasitized females than in non-parasitized. Parasitized females produced lighter eggs than those from uninfected females. This supports the compensatory reproduction hypothesis suggested for this species. Parasitism can change life history traits in a way that fecundity can be compensated; this co-evolution between host and parasites will be population or context dependent. Parasites are a functional part of any ecosystem and as our results show, deleting parasites in life history traits and reproduction studies in free living organisms could lead to an incomplete picture of the true processes that happen in nature.

Fully developed, sexually mature small male and female acanthocephalans, Microsentis wardae Martin & Multani, 1966 (Neoechinorhynchidae) reaching only 2.25 mm and 2.42 mm, respectively, were collected from the rectum of longjaw mudsuckers, Gillichthys mirabilis Cooper, in the salty marches of Anaheim Bay and San Diego Bay, California. Our specimens were half the size of those reported in the original description from the same host in Scammon's Lagoon over 700 km to the south. The ratio of proboscis and receptacle size to trunk size was markedly higher in our specimens compared to the larger specimens measured in the original description. The anatomy of all structures in our specimens from Anaheim Bay was comparable to that of the larger Scammon's Lagoon specimens that have apparently realized more growth in the Scammon's Lagoon. We have observed more structures that are not reported in the original description, especially evident from our scanning electron microscopy images, which are not possible to observe in the original line drawings. In our specimens, the micropores were unusually widely spaced and the energy dispersive X-ray analysis showed longitudinal hook sections with high levels of sulphur and phosphorus and moderate levels of calcium, but the whole hooks showed highest levels of sodium and magnesium – the biochemical hook signature of this species. Sequences of the small subunit (18S) of the nuclear ribosomal DNA were generated and compared with acanthocephalan sequences available from GenBank. As M. wardae comprises a monotypic genus, therefore, phylogenetic analyses inferred from the 18S gene showed its relationship with other species of closely related genera of Eoacanthocephala. This is the first report of molecular data of M. wardae.
