Shelled acanthors (=eggs) of two isolates of Moniliformis moniliformis (Acanthocephala) were tested for their capacity to respond to a hatching stimulus in vitro and for their retention of infectivity to a natural intermediate host (Periplaneta americana). The shelled acanthors were stored for more than 120 weeks (Australian isolate) and for 104 weeks (Texan isolate) together with rat faeces in an incubator maintained at 22·2±0·1°C. In both cases, infectivity to P. americana was shown to have been retained. In vitro tests of the hatching response were carried out on many occasions during this period of faecal storage. Shelled acanthors continued to respond and no differences were detected either between isolates or within an isolate through time.

In a paper recently published in the Philosophical Transactions of the Royal Society of London, the writer describes the discovery of certain small Tylenchoid nematodes in the swollen stems of the seedling oats suffering from frit-fly attack at Winches Farm, St. Albans. Adults of both sexes and larvæ undergoing the last two moults were found in the destroyed plant tissues surrounding the fly larvæ. The spermatized female worms were next discovered within the body-cavity of the frit-fly larvæ. In due course, they were obtained from pupæ of the fly, having increased in size, and finally were found within adult flies of both sexes lying coiled within the abdomen as comparatively large sausage-shaped worms. As a result of the presence of the worm, the flies are sterilised, being unable to develop their reproductive organs. The worms become viviparous and shed large numbers of larvæ into the body-cavity of the fly and these, after undergoing a certain amount of growth, make their way into the gut of the host and pass to the exterior via the anus. In the present communication a brief account is given of the principal observations on the new parasite, its life-history and the effects on the host, described in detail in the original paper.

The following collection of Cestodes has been brought together by Prof. A. S. Pearse, of the Duke University, Durham, N.C., during a sojourn in Nigeria for the purpose of studying the parasites of rodents and insectivores.

Fifteen female ICR mice, 6–8 weeks old, were each exposed to 25 metacercarial cysts of Echinostoma caproni. To determine the effects of withholding food on survival, size and distribution of E. caproni in ICR mice, two groups of five each were maintained without food for 1 day (group A) or 2 days (group B) prior to necropsy. Group C consisted of five mice maintained with food for the duration of the experiment. All mice were necropsied at 12 days postinfection. There was no significant difference in worm survival, body area or distribution between group A and C or group B and C mice. The results of this experiment show that short-duration removal of food has no effect on worm survival, size or distribution of E. caproni adults in the small intestine of the ICR mouse.

Three age groups of Bulinus natalensis, immature, mature but not yet egg-laying and mature egg-laying, were infected with miracidia of Schistosoma margrebowiei. The growth of infected, exposed but showing no signs of infection, and uninfected control groups, were examined at weekly intervals for ten weeks post exposure. Snails exposed to infection when immature, even in the group where no patent development ensued, showed a statistically significant reduction in growth and this was evident as early as 2–5 weeks post exposure. When infection occurred at the stage prior to egg-laying a significant reduction in growth was seen but only in the group which developed a patent infection. This was also the case for the egg-laying group although the reduction in growth was only significant in the final three weeks of the experiment. Thus in all three age groups, growth rate was reduced and the infected snails were significantly smaller at the end of ten weeks compared with the controls. The reasons for these effects are discussed.

Serine dehydratase was found to be five times more active than threonine dehydratase in the liver-fluke Fasciola indica. The latter was not affected by L-isoleucine. Optimum pH for both enzymes was found to be 9.0 in tris-universal buffer. Seven metallic ions tested did not accelerate the enzyme activity, but produced different effects on two enzymes. Two different enzymes probably exist for the two aminoacids, L-threonine and L-serine.

The epidemiological picture of Taenia saginata infections in Kenya is fragmented with limited available data. Although Sarcocystis species are significant meat-borne parasites, few studies have explored their occurrence in Kenya. This study aimed to estimate the occurrence of bovine cysticercosis and screen for the presence of Sarcocystis spp. A meat inspection-based survey was conducted in ten abattoirs in Narok County, Kenya, and inspection for T. saginata cysticerci was limited to the Triceps brachii muscle. The apparent occurrence of the parasite was 5.4% (95% CI, 3.8, 7.6, n=573). Molecular confirmation of T. saginata was done via nested polymerase chain reaction targeting the mitochondrial 12S ribosomal RNA gene and restricted fragment length polymorphism. Sarcocystis species were identified using a multiplex polymerase chain reaction method targeting the 18S ribosomal RNA gene sequences and the mitochondrial cytochrome c oxidase subunit I gene. Of the 31 cystic lesions tested, 26/31 (83.9%) were confirmed to be T. saginata. Sarcocystis cruzi and S. hominis were detected in 8/31 (25.8%) and 1/31 (3.2%) of the cystic lesions, respectively. Co-infections of S. cruzi and T. saginata were found in 6/31 lesions (19.4%). The confirmation of bovine cysticercosis and S. hominis is suggestive of the presence of risky culinary and sanitation practices that facilitate transmission. This is the first report and molecular confirmation of Sarcocystis spp. in cattle in the country. The presence of both zoonotic S. hominis and pathogenic S. cruzi highlights an underexplored concern of veterinary and human health significance, warranting further epidemiological investigation.

Many important studies on resistance reversion, anthelmintic efficacy and, especially, new molecules with antiparasitic effects are performed in laboratories using gerbils (Meriones unguiculatus) as the experimental model. This study aimed to evaluate the use of corticosteroids (dexamethasone and methylprednisolone acetate) in gerbils experimentally infected with different doses of infective larvae (sheathed or exsheathed) of Haemonchus contortus. In the first experiment, 28 gerbils were divided into seven groups infected by 2–6 × 103 larvae, with or without immunosuppression using corticosteroids. In the second experiment, eight gerbils were divided into two groups infected by 2 × 103 sheathed or exsheathed larvae. For the third assay, seven immunosuppressed gerbils were infected with 2 × 103 sheathed larvae and were killed 15 days post infection (PI). The highest number of parasites was recovered from methylprednisolone-immunosuppressed animals. We observed red and white blood cell alterations and biochemical parameters in infected animals that had undergone immunosuppression with methylprednisolone. We highlight that in the first and second experiments a satisfactory number of worms was recovered using sheathed larvae and immunocompetent animals. When exsheathed larvae were used, the number of worms recovered was unsatisfactory. A considerable larval burden was recovered from immunosuppressed gerbils 15 days PI, and body weight did not influence establishment of larvae.

Host–parasite coevolution may result in life-history changes in hosts that can limit the detrimental effects of parasitism. Fecundity compensation is one such life-history response, occurring when hosts increase their current reproductive output to make up for expected losses in future reproduction due to parasitic infection. However, the potential trade-offs between this increase in quantity and the quality of offspring have been relatively unexplored. This study uses the trematode, Schistosoma mansoni, and its snail intermediate host, Biomphalaria glabrata, to better understand how this host life-history response, fecundity compensation, impacts host reproduction. Measures of host reproductive output as well as offspring hatching success and survival were collected to assess the reproductive consequences of infection. Infected snails exhibited fecundity compensation by increasing the number of eggs laid and the overall probability of laying eggs compared to uninfected snails. Parental infection status did not play a significant role in hatching or offspring survival to maturity. Offspring from a later reproductive bout demonstrated a higher hatching success rate. Overall, the lack of an apparent trade-off between quantity and quality of offspring suggests that infected parental snails invest more resources towards reproduction not only to increase reproductive output, but also to maintain the fitness of their offspring, possibly at the expense of their own longevity.

Schistosomiasis remains a major neglected tropical disease that afflicts over 200 million people globally. Schistosomes, the aetiological agent of schistosomiasis, are parasitic flatworms that propagate between molluscan and mammalian hosts. Inside the mammalian host, schistosomes rapidly grow over 100-fold in size and develop into a sexually mature male or female that thrives in the bloodstream for several decades. Recent work has identified schistosome stem cells as the source that drives parasite transmission, reproduction and longevity. Moreover, studies have begun to uncover molecular programmes deployed by stem cells that are essential for tissue development and maintenance, parasite survival and immune evasion. Such programmes are reminiscent of neoblast-driven development and regeneration of planarians, the free-living flatworm relative of schistosomes. Over the last few decades, research in planarians has employed modern functional genomic tools that significantly enhanced our understanding of stem cell-driven animal development and regeneration. In this review, we take a broad stroke overview of major flatworm organ systems at the cellular and molecular levels. We summarize recent advances on genetic regulators that play critical roles in differentiation and maintenance of flatworm cell types. Finally, we provide perspectives on how investigation of basic parasite biology is critical to discovering new approaches to battle schistosomiasis.

We first sequenced and characterised the complete mitochondrial genome of Toxocara apodeme, then studied the evolutionary relationship of the species within Toxocaridae. The complete mitochondrial genome was amplified using PCR with 14 specific primers. The mitogenome length was 14303 bp in size, including 12 PCGs (encoding 3,423 amino acids), 22 tRNAs, 2 rRNAs, and 2 NCRs, with 68.38% A+T contents. The mt genomes of T. apodemi had relatively compact structures with 11 intergenic spacers and 5 overlaps. Comparative analyses of the nucleotide sequences of complete mt genomes showed that T. apodemi had higher identities with T. canis than other congeners. A sliding window analysis of 12 PCGs among 5 Toxocara species indicated that nad4 had the highest sequence divergence, and cox1 was the least variable gene. Relative synonymous codon usage showed that UUG, ACU, CCU, CGU, and UCU most frequently occurred in the complete genomes of T. apodemi. The Ka/Ks ratio showed that all Toxocara mt genes were subject to purification selection. The largest genetic distance between T. apodemi and the other 4 congeneric species was found in nad2, and the smallest was found in cox2. Phylogenetic analyses based on the concatenated amino acid sequences of 12 PCGs demonstrated that T. apodemi formed a distinct branch and was always a sister taxon to other congeneric species. The present study determined the complete mt genome sequences of T. apodemi, which provide novel genetic markers for further studies of the taxonomy, population genetics, and systematics of the Toxocaridae nematodes.

Parasites can have strong effects on invertebrate host behaviour, fecundity and survival in marine ecosystems. However, parasites are often poorly documented and still rarely integrated into marine ecological modelling; comprehensive surveys of infection in marine invertebrates are sporadic at best. For example, rock crabs are an important part of Californian coastal ecosystems, both as regulators of mussel populations and non-native species, and as prey items for predators like sea otters, but their parasite communities and potential effects on crab population dynamics are seldom studied or understood. Here, we present the first report of infection by the trematode Helicometrina nimia in the economically and ecologically important red rock crab (Cancer productus) and Pacific rock crab (Romaleon antennarium). As intermediate hosts, they are a missing link for infection by H. nimia in Californian fish that was unreported until now. Based on these findings, we advocate for further research into parasite diversity and their potential effects on ecologically and commercially important species.

In his “Monograph on the Anguillulidae, or Free Nematoids, Marine, Land and Freshwater; with Descriptions of 100 New Species,” published in 1865, Bastian described a nematode which he found in the yellow lichen Xanthoria (then known as Parmelia) parietina. He named it Aphelenchus parietinus, giving a short description and two drawings. His drawings are reproduced in Fig. 1 and his description is as follows:

This study shows that some cercariae of S. haematobium and S. mansoni die during penetration of mouse or hamster skin.

Approximately 30–38% of cercariae of both species die in mouse skin and 14–16% die in hamster skin. The greater number of cercariae which die in the skin of mice seems to account for the higher yield of adult worms recovered in hamsters. Adult worm recoveries from animals infected with S. haematobium were, however, only about half the worm recoveries from hosts infected with S. mansoni.

The prophylactic and curative effects of praziquantel and oxamniquine on a Saudi Arabian strain of Schistosoma mansoni in MF-1 mice were assessed. The drugs were administered orally. At 240 mg/kg praziquantel, there was a reduction of 89·1% in adult worm recovery and a marked reduction in tissue deposited eggs. The reduction in adult worm recovery after dosing with 50 mg/kg oxamniquine was 89·2%. At low doses (40 mg/kg praziquantel and 30 mg/kg oxamniquine) administered at 11 days, 5 days and 3 h before and 5, 21 and 49 days after infection, the reduction in adult worm recovery was 0·0%, 65·1%, 58·8%, 33·6%, 0·0% and 76·0% for praziquantel and 0·0%, 66·0%, 60·0%, 41·3%, 10·8% and 79·0% for oxamniquine. Numbers of lung schistosomula and the size of hepatic granulomata were also reduced.

The existence of adult Schistosoma haematobium worm proteins (Sh31/32) homologous to the diagnostic Sm31/32 of S. mansoni is shown. Sm31/32 and Sh31/32, adsorbed on nitrocellulose and kept dry on the bench before immunoblot analysis, were antigenically stable for at least 4 years including storage for 17 months in tropical climates. The antigens react with patient sera in the absence of defined buffers under rather simple conditions (“humid chamber blot”). The results add to the use of these antigens for serodiagnosis of schistosomiasis in endemic areas.

The consequences of previous and concurrent infection with two related species of cestodes, Hymenolepis nana and H. diminuta, were studied in CD1 mice. A H. diminuta infection strongly affected the establishment and the survival of a secondary H. nana egg or cyst infection administered 30 days later. An infection of 20 H. nana eggs strongly protected against a 5-cyst H. diminuta challenge, whereas an infection of 10 H. nana cysts was ineffective; 20 H. nana eggs also protected against a challenge with 5 cysts of H. diminuta administered 5 days later. No effects were observed in either parasite during a concurrent infection established by administration of cysts. An H. nana egg-infection was unable to affect the establishment of a secondary H. nana cyst-infection given 1 month later; however a significant decrease in growth was found. Similar results were found when a primary H. nana egg-infection was followed 5 days later by the homologous cyst-infection. But an infection with 5 H. nana cysts was unable to protect against a homologous challenge of 5 cysts or 20 eggs. The reciprocal cross immunity between the heterologous parasites and the failure of protection of homologous challenges are discussed in relation to light infections.

Sheep reared worm free were used as indicators of the availability of sheep nematode infective larvae during the summer on pasture grazed by ordinary flock ewes and their lambs.

Data were collected in terms of worms found in the test sheep at post mortem examination after periods of three weeks' exposure in the field.

Incidence of mating pairs in 40 worm infections in 5 week old female CBA mice increased up to day 30 post infection. By this time, and thereafter until the experiment ended on day 70, approximately 60% of the worm population occurred as mating pairs. Mating began during the ontogenetic migration of the worms. There was no difference between the percentage of worms mating in the more highly populated anterior sections of the small intestine as compared to the less densely populated more posterior sections. Significantly more female than male worms were recovered between days 8 and 20 post infection (p<0.02 to 0.001) and there was a significant decline in overall recoveries during the experimental period (r = −0.77, p<0.01). Low intensity infections, with 4 to 16 worms per mouse, demonstrated that a larger percentage of worms in the infection were mating at an earlier time post infection than in the 40 worm infections. Ontogenetic migration was reduced in low intensity infections because the initial distribution of worms was more restricted.

Two new species of the genus Oochoristica Lühe, 1898 from Australian dasyuried marsupials are described and their affinities discussed. The new species are O. antechini sp. nov. from Antechinus macdonnellensis and O. eremophlia sp. nov. from Antechinus rosamondae.
