Scedosporium and Lomentospora species are ubiquitous saprophytic filamentous fungi that emerged as human pathogens with impressive multidrug-resistance profile. The ability to form biofilm over several biotic and abiotic surfaces is one of the characteristics that contributes to their resistance patterns against almost all currently available antifungals. Herein, we have demonstrated that Scedosporium apiospermum, Scedosporium minutisporum, Scedosporium aurantiacum and Lomentospora prolificans were able to form biofilm, in similar amounts, when conidial cells were incubated in a polystyrene substrate containing Sabouraud medium supplemented or not with different concentrations (2%, 5% and 10%) of glucose, fructose, sucrose and lactose. Likewise, the glucose supplementation of culture media primarily composed of amino acids (SCFM, synthetic cystic fibrosis medium) and salts (YNB, yeast nitrogen base) did not modulate the biofilm formation of Scedosporium/Lomentospora species. Collectively, the present data reinforce the ability of these opportunistic fungi to colonize and to build biofilm structures under different environmental conditions.

Quantifying and comparing stocks of oysters (Crassostrea virginica) within and among estuaries across the Gulf of Mexico is difficult because the sampling equipment used is either inconsistent among studies, or inefficient. In Texas, USA, stock assessments of oyster populations are made using an oyster dredge, which is an inefficient sampling tool. We compared sampling densities estimated by oyster dredges with more accurate estimates taken by diver-quadrat samples to determine a dredge efficiency rate. Our calculated efficiency rate (0.125) was negatively affected by the number of dead oysters, and the number and volume of total oysters in an area, but not affected by sediment grain size, water quality, and other oyster metrics. The dredge efficiency rate calculated in this study can be applied to past and future dredge-collected oyster quantity data to provide more realistic estimates of oyster densities and allow more accurate stock assessments and comparisons among studies and regions.

The 10-item Autism-Spectrum Quotient (AQ10) is a self-report questionnaire used in clinical and research settings as a diagnostic screening tool for autism in adults. The AQ10 is also increasingly being used to quantify trait autism along a unitary dimension and correlated against performance on other psychological/medical tasks. However, its psychometric properties have yet to be examined when used in this way. By analysing AQ10 data from a large non-clinical sample of adults (n = 6,595), we found that the AQ10 does not have a unifactorial factor structure, and instead appears to have several factors. The AQ10 also had poor internal reliability. Taken together, whilst the AQ10 has important clinical utility in screening for diagnosable autism, it may not be a psychometrically robust measure when administered in non-clinical samples from the general population. Therefore, we caution against its use as a measure of trait autism in future research.

The glial fibrillary acidic protein (GFAP) is an intermediate filament widely used to identify and label astroglial cells, a very abundant and relevant glial cell type in the central nervous system. A major hurdle in studying its behavior and function arises from the fact that GFAP does not tolerate well the addition of protein tags to its termini. Here, we tagged human GFAP (hGFAP) with an enhanced green fluorescent protein (EGFP) for the first time, and substituted a previously reported EGFP tag on mouse GFAP (mGFAP) by a more versatile Halo Tag. Both versions of tagged GFAP were able to incorporate into the normal GFAP filamentous network in glioma cells, and Alexander disease-related mutations or pharmacological disruption of microtubules and actin filaments interfered with GFAP dynamics. These new tools could provide new fruitful venues for the study of GFAP oligomerization, aggregation and dynamics in living cells.

Acquired MDM2 inhibitor resistance is commonly caused by loss-of-function TP53 mutations. In addition to the selection of TP53-mutant cells by MDM2 inhibitors, MDM2 inhibitor-induced DNA damage may promote the formation of TP53 mutations. Here, we cultivated 12 sublines of the intrinsically MDM2 inhibitor-resistant TP53 wild-type acute myeloid leukaemia cell line PL21 for 52 passages in the presence of ineffective concentrations of the MDM2 inhibitor nutlin-3 but did not observe loss-of-function TP53 mutations. This suggests that MDM2 inhibitors select TP53-mutant cells after mutations have occurred, but do not directly promote TP53 mutations. Unexpectedly, many sublines displayed increased sensitivity to the anti-cancer drugs cytarabine, doxorubicin, or gemcitabine. Consequently, therapies can affect the outcome of next-line treatments, even in the absence of a therapy response. This finding is conceptually novel. A better understanding of such processes will inform the design of improved therapy protocols in the future.

Information on safe water, sanitation and hand washing obtained in large scale surveys are used to validate its responsiveness to childhood ailments. Definition of these variables are uniform to enable comparison within and across countries and devoid of the context and circumstance. Associating these variables with prevalence of diarrhoea overlooking the context seem to distort the relationship and lead to spurious results. An empirical verification of such an association in an Indian context based on the most recently conducted NFHS-4 data set brings to the fore apparent contradictions that cautions on the use of these variables as they are obtained. It calls for a redefinition of these variables prior to verifying their responsiveness to childhood diarrhoea as illustrated here.

Elderly care is becoming a relevant issue with the increase of population ageing. Fall injuries, with their impact on social and healthcare cost, represent one of the biggest concerns over the years. Researchers are focusing their attention on several fall-detection algorithms. In this paper, we present a deep-learning solution for automatic fall detection from RGB videos. The proposed approach achieved a mean recall of 0.916, prompting the possibility of translating this approach in the actual monitoring practice. Moreover to enable the scientific community making research on the topic the dataset used for our experiments will be released. This could enhance elderly people safety and quality of life, attenuating risks during elderly activities of daily living with reduced healthcare costs as a final result.

Restless legs syndrome (RLS) is a neurological disorder characterized by an urge to move and uncomfortable sensations. Genetic studies have identified polymorphisms in up to 19 risk loci, including MEIS1 and BTBD9. Rodents deficient in either homolog show RLS-like phenotypes. However, whether MEIS1 and BTBD9 interact in vivo is unclear. Here, with C. elegans, we observed that the hyperactive egg-laying behavior caused by loss of BTBD9 homolog was counteracted by knockdown of MEIS1 homolog. This was further investigated in mutant mice with Btbd9, Meis1, or both knocked out. The double knockout mice showed an earlier onset of the motor deficit in a wheel running test but did not have increased sensitivity to heat stimuli as observed in single knock outs. Meis1 protein level was not influenced by Btbd9 deficiency, and Btbd9 transcription was not affected by Meis1 haploinsufficiency. Our results demonstrate that MEIS1 and BTBD9 do not regulate each other.

Previous work on aesthetic experience suggests that aesthetic judgments are self-referential. The self-reference effect (SRE) is the tendency for individuals to show improved memory for items that are judged in relation to themselves. The current study sought to understand if the SRE exists for aesthetic judgments of music. Participants heard musical excerpts (classical, jazz, and electronic) and rated either a) how much they liked the music (Self condition), b) how much a close relative or friend would like the music (Other condition), or c) the genre of the music (Genre condition). After a retention interval, participants completed a recognition memory task for the musical excerpts. Participants did not show improved memory for musical excerpts encoded in the Self condition. These results extend the concept of the SRE into the domain of aesthetic judgments, but do not provide support for a memory advantage when making aesthetic judgments in relation to the self.

Dynamic nuclear polarization (DNP) is a technique in magic-angle spinning (MAS) nuclear magnetic resonance (NMR) which leads to sensitivity enhancement and helps to overcome the issue of low polarization in detected nuclei. Recent research showed, that methyl groups, which show active reorientation dynamics and cause heteronuclear cross relaxation at typical DNP temperatures around 100 K, may be used as a pinpoint source of polarization for selective and site-specific probing. In this study, we investigated the cross-relaxation behavior of methyl groups in nicotine and caffeine under DNP. These effects could be useful for investigating receptor/ligand binding.

Telomeres are repeating DNA sequences found on the ends of chromosomes, which shorten with age and are implicated in senescence. Cross-species analyses of telomere shortening rates (TSR) and telomere lengths are important for understanding mechanisms underlying senescence, lifespan and life-history strategies of different species. Whittemore et al. (2019) generated a new dataset on variation in TSR, lifespan and body mass. In phylogenetically uncorrected analyses they found that TSR negatively correlates with lifespan. We re-ran analyses of their dataset using appropriate phylogenetic corrections. We found a strong phylogenetic signal in the association between TSR and body mass. We were able to corroborate Whittemore et al.’s major findings, including while correcting for body mass in a multivariate analysis. Since laboratory mice have different telomere lengths and potentially different telomere dynamics than wild mice, we removed mice from the analysis, which attenuates most associations.

One of the commonly used analytical approaches for measuring oxygen isotope ratios δ18O of solids (organic and inorganic) is to pyrolyze the samples to gaseous phases and then send the gas into an isotope ratio mass spectrometer system. Solid samples for δ18O measurements are usually stored in silver cups because of its low reactivity towards oxygen and other oxidants. Samples in silver cups can be dropped directly into the carbon column of the pyrolysis furnace. However, the silver cups can tarnish and then be oxidized over a prolonged storage period. We find that while a small amount of silver oxides does not affect measurements with appreciable sample sizes, it can skew isotope results of small samples. We thus recommend careful storage of samples in silver cups to minimize oxidation, such as under an air-isolated condition, and avoiding prolonged storage for accurate δ18O measurements.

In a recent study, mid-latitude ionospheric parameters were compared with solar activity; it was suggested that the relationship between these, earlier assumed stable, might be changing with time (Lastovicka, 2019). Here, the information is extended to higher latitude (69.6°N, 19.2E) and further back in time. For the ionospheric F-region (viz. the critical frequency, FoF2) the same behaviour is seen with a change-point around 1996. For the ionospheric E-region (viz. the critical frequency, foE), change-points are less obvious than in the mid-latitude study, presumably owing to the observation site lying under the auroral oval.

Growing in a saline environment causes changes in important physiological processes that are directly related to plant growth and development. In this study we evaluated the effect of salinity on transpiration of sorghum plants in semi-arid conditions and found that the highest rates of transpiration were observed in the hottest hours of the day, between 10 a.m. and 3 p.m., with plants subjected to the saline environment having their transpiration reduced by up to 70% when compared to the non-saline environment. This behavior can be reflected in reductions in plant growth and development due to reduced water absorption by the roots, consequently causing an imbalance of nutrients in the plant due to low absorption rate and competition between nutrients and salts in the preferred routes of absorption in the roots.

Motor adaptation is a process by which the brain gradually reduces error induced by a predictable change in the environment, e.g., pointing while wearing prism glasses. It is thought to occur via largely implicit processes, though explicit strategies are also thought to contribute. Research suggests a role of the cerebellum in the implicit aspects of motor adaptation. Using non-invasive brain stimulation, we sought to investigate the involvement of the cerebellum in implicit motor adaptation in healthy participants. Inhibition of the cerebellum was attained through repetitive transcranial magnetic stimulation (rTMS), after which participants performed a visuomotor-rotation task while using an explicit strategy. Adaptation and aftereffects of the TMS group showed no difference in behaviour compared to a Sham stimulation group, therefore this study did not provide any further evidence of a specific role of the cerebellum in implicit motor adaptation. However, our behavioral findings replicate those in the seminal study by Mazzoni and Krakauer (2006).

Evaluation of neural activity during natural behaviours is essential for understanding how the brain works. Here we show that neuron-specific self-evoked firing patterns are modulated by an object’s presence, at the electrosensory lobe neurons of tethered-moving Gymnotus omarorum. This novel preparation shows that electrosensory signals in these pulse-type weakly electric fish are not only encoded in the number of spikes per electric organ discharge (EOD), as is the case in wave-type electric fish, but also in the spike timing pattern after each EOD, as found in pulse-type Mormyroidea. Present data suggest that pulsant electrogenesis and spike timing coding of electrosensory signals developed concomitantly in the same species, and evolved convergently in African and American electric fish.

BPIFA2 (PSP, SPLUNC2, C20orf70) is a major salivary protein of uncertain physiological function. BPIFA2 is downregulated in salivary glands of spontaneously hypertensive rats, pointing to a role in blood pressure regulation. This study used a novel Bpifa2 knockout mouse model to test the role of BPIFA2 in sodium preference and blood pressure. Blood pressure did not differ between wild-type male and female mice but was significantly lower in male knockout mice compared to male wild-type mice. In contrast, blood pressure was increased in female knockout mice compared to female wild-type mice. Female wild-type mice showed a significant preference for 0.9% saline compared to male mice. This difference was reduced in the knockout mice. BPIFA2 is an LPS-binding protein but it remains to be determined if the reported effects are mediated by the LPS-binding activity of BPIFA2.

In recent years the availability of geolocation data has increased considerably and can be found in various portable devices such as smartphones. These devices are intended for navigation in general, but can be used to carry out topographical surveys that do not require high accuracy of the surveyed data. To verify the applicability and accuracy of these devices we conducted the topographic survey in an area of approximately 5 ha using a GPS with RTK technology as reference, a Commercial Navigation Receiver (RNC) and a popular brand smartphone with the mobile applications C7 GPS Data and GPS Essentials previously installed. The GPS RNC showed the best planimetric results and the Smartphone with C7 GPS Data obtained the best result altimetric. None of the receivers analyzed showed high accuracy in results obtained. However, they can be used for tasks where high precision is not required.

Type I/II interferons (IFNα,β/IFNɣ) are cytokines that activate signal-transducer-and-activator-of-transcription-1 (STAT1). The STAT1 N-terminal domain (NTD) mediates dimerization and cooperative DNA-binding. The STAT1 DNA-binding domain (DBD) confers sequence-specific DNA-recognition. STAT1 has been connected to growth inhibition, replication stress and DNA-damage. We investigated how STAT1 and NTD/DBD mutants thereof affect fibrosarcoma cells. STAT1 and indicated mutants do not affect proliferation of resting and IFNα-treated cells as well as checkpoint kinase signaling, and phosphorylation of the tumor-suppressive transcription factor p53 ensuing ɣ-irradiation. Of the STAT1 reconstituted U3A cells those with STAT1 NTD mutants accumulate the highest levels of the replication stress/DNA-damage marker S139-phosphorylated histone H2AX (ɣH2AX). This is similarly seen with a STAT1 NTD/DBD double mutant, indicating transcription-independent effects. Furthermore, U3A cells with STAT1 NTD mutants are most susceptible to apoptotic DNA fragmentation and cleavage of the DNA repair protein PARP1. These data provide novel insights into the relevance of the STAT1 NTD.
