Contemporary phytoplankton and palaeolimnological studies were made of the algal response to acidification and liming in three lakes in Hälsingland, central Sweden (Njupfatet, Sjösjön, Djuptjärn). Surveys and experimental studies of the phytoplankton response to liming were undertaken at Njupfatet, together with an experiment designed to determine the possible role of lake sediments as an inoculum for any new species arriving in the water column. Liming had little quantitative or qualitative effect on the phytoplankton diversity at Njupfatet, but did result in the loss of the dominant contributor to algal biomass, Merismopedia tenuissima. None of the four new species recorded in the lake following liming was hatched from the sediment inoculum experiment. A freeze core from Njupfatet was dated by 210Pb and carbonaceous fly-ash particle profiles were also determined for Njupfatet and Sjösjön, which provided an approximate chronology for the latter lake. Diatom analyses were made of the three lakes and pH and dissolved organic carbon inferred using weighted averaging methodology. Njupfatet had no planktonic diatoms over the last 200–300 years, and was dominated by benthic diatoms, but the diatom-inferred pH suggests that the lake was acidified to pH < 5 prior to liming in 1989. Both Djuptjärn and Sjösjön were dominated by planktonic diatoms, and were acidifying from (c. 1970) prior to liming, Sjösjön from pH 6 to ∼ 5 and Djuptjärn from 6·5 to ∼ 5·5. These pH inferences suggest that at least some lakes in this region are susceptible to atmospheric acidification. The response of the diatoms to liming is discussed, in particular the rapid expansion at Njupfatet, of Cyclotella glomerata a diatom which was also present in Sjösjön for a period prior to acidification, and at other lakes in south-west Sweden. Possible reasons for the expansion of this small centric diatom are discussed.

A Crypthecodinium cohnii-like dinoflagellate was found to prey on the unicellular red alga Porphyridium sp. The cytoplasm of the prey is ingested by myzocytosis within 10–30 s, and the contents of up to 20 Porphyridium cells can be taken up by one dinoflagellate. The feeding tube is retracted after each uptake process. Cytochalasin D disturbs the suction and the retraction of the feeding tube. Feeding behaviour depends on the light–dark regime: the dinoflagellates prey preferentially in the dark period and finish the trophic phase at the beginning of the light period. They then assemble and encyst, and digestion takes place in the light period. Cell division is restricted to the encysted stage. At the end of the light period excystment takes place, combined with defecation. Isogamy as well as anisogamy and nuclear cyclosis were observed. The trophonts have very thin thecal plates and a microtubular basket with two kinds of elongate vesicles. The microtubules of the tubular basket are also found in young cysts, formed after the end of the feeding period. The fine structure of the digestion vacuoles and of the faecal bodies is described.

The cyanobacterium Microcystis PCC7806, grown under continuous light, fermented endogenously stored glycogen to equimolar amounts of acetate and ethanol when incubated anaerobically in the dark. In addition, H2, CO2 and some L-lactate were produced. This fermentation pattern differed from that displayed by cells which had been grown under an alternating light/dark (16/8 h) cycle. Such cells produced much more ethanol than acetate, while no lactate was formed. These differences could not be related to the levels of key enzymes of fermentation, which were identical in the two cultures. The cultures grown under continuous light contained twice as much glycogen as the light/dark-grown cells and the former metabolized it at a rate approximately 3 times as fast as the latter culture. Fermentation in the culture grown under continuous light showed low carbon recovery (59–80%) and high oxidation/reduction balance (approximately 1·5). On the basis of calculations of ATP yield it was concluded that this culture was capable of growth driven by fermentation. The increase in structural cell material would account for the missing carbon and reduction equivalents.

The stress-70 protein family has previously been shown to be a useful tool for molecular phylogeny at the kingdom to family levels. Although sequences of many members of the stress-70 family are available, few genes from the Protoctista have been sequenced to date. Phylogenetic analyses of algae based on various molecules have not, as yet, provided clear results concerning relationships between major divisions. We cloned and sequenced several algal stress-70 genes in order to provide additional data and to further analyse phylogenetic relationships among algal divisions. New nuclear sequences were obtained from Guillardia theta (Cryptophyta), Ascophyllum nodosum (Heterokontophyta) and Cyanophora paradoxa (Glaucocystophyta). Phylogenetic trees of the stress-70 protein family calculated using different methods are presented. In our trees, the heterokont alga Ascophyllum nodosum is closely related to the slime mould Dictyostelium discoideum, while the nucleomorph (eukaryotic endosymbiont) of the cryptophyte Rhodomonas salina seems to be related to the chlorobiont lineage. The glaucocystophyte Cyanophora paradoxa and the nuclear sequence (host) of the cryptomonad alga Guillardia theta also seem to be closely related. The Cryptophyta and the heterokont algae have evolved from different secondary endosymbiotic events involving different hosts and probably different endosymbionts. However, until more stress-70 sequences of algal divisions become available no definitive conclusions can be drawn concerning branching of the major divisions.

Enteromorpha intestinalis grows along the Swedish west coast in rockpools which are isolated from the open seawater for long time periods and where, therefore, the inorganic carbon content is low and the pH is high during the day. In order to investigate how E. intestinalis could grow under such apparently CO2-constraining conditions, we measured its photosynthetic responses to inorganic carbon in the presence of an inhibitor of external/surface-bound carbonic anhydrase (acetazolamide) as well as an inhibitor of HCO−3 transport via anion exchange (4,4′-diisothiocyanatostilbene-2,2′-disulfonate). The results show that both HCO−3 dehydration via surface-bound carbonic anhydrase and HCO−3 transport via a 4,4′-diisothiocyanatostilbene-2,2′-disulfonate-sensitive mechanism were present in E. intestinalis, but only HCO−3 uptake via the putative transporter was operative in rockpool water during most of the photic period (pH > 9·4, inorganic carbon < 0·45 mol m−3 and CO2 < 0·05 mmol m−3). The advantage of such a mechanism, rather than extracellular HCO−3 dehydration, is discussed with regard to photosynthesis of marine macroalgae under in situ conditions conducive to high pH values adjacent to the thalli.

Caulerpa (Chlorophyta, Ulvophyceae) is a common marine tropical-subtropical genus of about 70 species, inhabiting the eulittoral zone on rocks and corals as well as mangroves. The genus is particularly diverse and abundant along southern Australian coasts, where it is thought to have originated. Here, we compare sequences from the nuclear ribosomal cistron among five species of Caulerpa, including nine populations of Caulerpa filiformis from two biogeographic regions: five from Australia and four from South Africa. Species relationships were well resolved by internal transcribed spacer (ITS) sequences and supported by high bootstrap values as follows: (C. geminata (C. simpliciuscula (C. trifaria (C. scalpelliformis (C. filiformis – Australia, C. filiformis – South Africa))))). Nucleotide divergence within C. filiformis was low with four and five nucleotide differences present in ITS1 and ITS2 respectively. Unexpectedly, the 5.8S rRNA gene showed eight nucleotide differences between the Australian and South African populations and may indicate cryptic species. The Australian C. filiformis is probably not a South African introduction. These sequences provide an independent means for assessment of species relationships and appear, in so far as limited taxon sampling permits, to follow chloroplast ultrastructural groupings proposed 20 years ago.

DNA of Chara vulgaris thalli contains target sequences for the methylation-sensitive restriction enzymes, Sau3A, HpaII and MspI. These enzymes were used with the in situ nick-translation radioactive technique to evaluate by grain counting in autoradiograms changes in cytosine methylation during antheridial filament development and spermiogenesis. In successive generations of dividing antheridial filaments the level of DNA methylation gradually increases, then decreases slightly in the early stages of spermiogenesis. Spermatozoids display the highest level of DNA methylation.

Polypeptides of 21, 36 and 37 kDa are induced in the unicellular green alga Chlamydomonas reinhardtii when cells are transferred from high (5%) to low (0·03%) CO2 concentrations. The synthesis of these polypeptides is correlated with the induction of the CO2-concentrating mechanism. Interaction between the induction of low-CO2-inducible polypeptides, the CO2-concentrating mechanism and photorespiration has been studied in wild-type C. reinhardtii with the aim of clarifying whether the glycolate pathway is involved in algal acclimation to limiting CO2 conditions. Our results showed that the induction of the 37 kDa periplasmic carbonic anhydrase and 21 kDa polypeptide under low-CO2 conditions was not observed in the presence of aminooxyacetate, an inhibitor which completely blocks glycolate metabolism. However, the induction of the 36 kDa polypeptide was not affected by this inhibitor. The presence of aminooxyacetate during the acclimation to low CO2 conditions also inhibited the increase in the photosynthetic affinity for inorganic carbon shown by low-CO2-acclimated Chlamydomonas cells without the inhibitor. Our results suggest that the induction of the CO2-concentrating mechanism may require the function of the glycolate pathway. In addition, our results also indicate that there is differential regulation of the induction of these three low-CO2-inducible polypeptides in Chlamydomonas reinhardtii.

In continuing studies on the taxonomy of non-geniculate, coralline algae from South Africa, stipes of the kelp, Ecklonia maxima, in the subtidal zone of Western Cape Province, were frequently observed to be covered with rosy-pink encrusting coralline algae. The reproductive morphology and anatomy of these plants show that they belong to the subfamily Melobesioideae and the genus Synarthrophyton. The specimens agree closely with the holotype of Lithothamnion capense f. eckloniae described by Foslie on the basis of South African material. Synarthrophyton eckloniae is described in detail; the species appears to be endemic to Western Cape Province. Another crustose coralline alga from the subtidal zone of South Africa was found to be conspecific with a syntype of Lithothamnion magellanicum and the lectotype and isolectotype of Lithothamnion muelleri f. neglecta. These are now shown to conform to the generic concept of Synarthrophyton on the basis of their concordance with the characters of that genus. South African material of S. magellanicum is compared with the type specimen of Lithophyllum schmitzii with which it had been synonymized and it is concluded that the two species are distinct. It can be distinguished from type specimens of Lithothamnion lamellatum and L. chatamense, with which previous authors had suggested it might be conspecific, on the basis of anatomical characters, despite similarity in habit. The last two species have not been recorded in South Africa.

Monoecious, bisexual and unisexual clones of Achnanthes longipes were isolated from the Black Sea and studied in laboratory culture. Clones differed in their growth characteristics: in monoecious clones the cells formed tufted aggregations while in other clones they were more dispersed. Bisexual and unisexual clones exhibited intraclonal (monoecious) reproduction, but only at a very low frequency and usually within a more restricted size range than in monoecious clones. Interclonal crosses were made in all possible pairwise combinations. Abundant auxosporulation took place in all crosses, except where unisexual clones of the same sex were incubated together. Auxosporulation was more vigorous and occurred over a wider size range in interclonal crosses than during monoecious reproduction. Sexual reproduction is isogamous. In the commonest pattern of auxosporulation, two paired gametangia each produce two gametes, which fuse to give two auxospores. More rarely (9% of pairs), the gametangia produce only one gamete apiece, and hence only one auxospore. In addition, very small cells can enlarge vegetatively, although genetic or cytological damage sometimes compromises their long-term viability.

The marine red alga known as Laurencia obtusa var. snackeyi (Weber–van Bosse) Yamada (Rhodomelaceae, Ceramiales) is characterized as follows: (1) production of 4 periaxial cells from each axial cell; (2) a few, percurrent, terete, thick main axes arising from a single discoid holdfast without stolon-like branches; (3) alternate-spiral branching; (4) presence of longitudinally oriented secondary pit-connections between contiguous superficial cortical cells; (5) absence of a palisade-like layer; (6) absence of projecting superficial cortical cells; (7) absence of lenticular thickenings in the walls of medullary cells; (8) parallel arrangement of tetrasporangia; and (9) presence of one corps en cerise per superficial cortical cell and trichoblast cell. It differs from L. obtusa (Hudson) Lamouroux in the absence of stolon-like branches with secondary attachment discs and the presence of thicker axes. The following binomial is proposed: Laurencia snackeyi (Weber–van Bosse) Masuda, stat. nov. Halogenated secondary metabolites of this species were investigated for samples from Vietnam and Malaysia. The major metabolites of the Vietnamese sample are palisadin A and aplysistatin, both sesquiterpenoids, whereas the Malaysian samples contain these compounds along with another monocyclofarnesol-derived sesquiterpenoid, 5-acetoxypalisadin B. L. snackeyi is also characterized by this set of secondary metabolites.

The combination of nitrogen deprivation and increased cultivation light intensity resulted in the synthesis of secondary carotenoids in flagellates of Haematococcus lacustris. The pigment pattern was characterized by high-pressure liquid chromatography analysis during the accumulation period and in response to inhibitors of carotenoid biosynthesis (diphenylamine, norflurazon and tetcyclacis). Diphenylamine treatment resulted in (i) a decrease in ketocarotenoids and (ii) an accumulation of β-carotene and zeaxanthin due to inhibition of the β-carotene oxygenase. Our results indicate that astaxanthin synthesis in H. lacustris follows the biosynthetic pathway elucidated in the marine bacterium Agrobacterium aurantiacum.

Phytoplankton studies in the North Atlantic have revealed coccoliths of Neosphaera coccolithomorpha and Ceratolithus cristatus occurring on the same cell. This may indicate that each of these species represents a stage of one coccolithophorid life cycle, in which each phase possesses a different heterococcolith type. However, definitive evidence for this is lacking.

Chrysochromulina and Prymnesium are important bloom-forming organisms in marine and brackish waters, respectively. Both genera include toxic species, which are primarily implicated in fish kills. Previous analyses of small subunit (SSU) rDNA sequences from Chrysochromulina and Prymnesium spp. indicate that Chrysochromulina is paraphyletic. C. polylepis, which produced a spectacular, harmful bloom in 1988, is more closely related to toxic Prymnesium species than to most other Chrysochromulina species based on rDNA sequence comparisons. Signatures were identified in the SSU rRNA gene specific for a clade that comprised primarily toxic taxa (C. polylepis, P. parvum, P. patelliferum and P. calathiferum) and that recognized C. polylepis alone. Oligonucleotide probes complementary to these regions were designed, and their specificity tested using dot-blot hybridization on PCR products of the SSU rRNA gene from 28 strains of Chrysochromulina and Prymnesium. Whole-cell hybridizations were performed with FLUOS- as well as Cy3-labelled probes on cultured species from both genera, and were detected with both epifluorescence microscopy and flow cytometry. The probes afforded easy identification of clonal isolates of C. polylepis and a cluster of closely related species including C. polylepis and Prymnesium spp. The feasibility of using these probes for species identification and studies of population dynamics in the field is discussed.

Single coccoliths of Emiliania huxleyi grown in mesocosm enclosures (60°16′N, 05°14′E, May–June 1991) under different N[ratio ]P regimes were analysed in a scanning electron microscope. The results indicate that only E. huxleyi with Type A coccoliths was present in the enclosures. Approximately 80–90% of the total coccolith assemblages had developed normally, whilst the remainder were malformed, incompletely grown or dissolved. Severely under-calcified specimens were rare and dissolution and breakage less than 5%. The coccoliths were of larger size than normal, as has been found previously in fjords of southwestern Norway, supporting the conclusion that a local population of E. huxleyi has developed, specific to these waters. Both phosphorus and nitrogen stress caused significant changes in coccolith size and evidence of malformation was clear, particularly in the low-phosphate enclosure. Although the observations presented here concern only Type A coccoliths and it is not known how nutrient stress may affect the coccoliths of the other types of E. huxleyi, they do serve to stress the fact that environmental conditions may possibly obscure genetically determined features. Following our observations on coccolith morphology in relation to nutrient status in enclosures, it will be of interest to test whether a similar correlation can be detected in the natural environment.

Examination of type material, comparative studies on vegetative characters and data from published accounts of sporangia, gametangia and spermatozoids are used here to further develop and strengthen generic and species concepts in Zonaria and Homoeostrichus. Zonaria is characterized by octosporangia which lack a stalk cell and occur among whitish paraphyses, by the presence of cortical cells in indusiate sporangial and oogonial sori, and by slightly elongate sterile cells bordering the antheridial sori. Other characters (mucilage in sporangial and oogonial sori, biflagellate sperm with a row of many tall spines along the anterior flagellum) may also be diagnostic of Zonaria but have not been demonstrated in the generitype. Eight species are retained in Zonaria. Generic placement of Zonaria stipitata, described on vegetative characters, requires confirmation. Homoeostrichus, as typified by Homoeostrichus sinclairii, is defined by the presence of brown paraphyses in nonindusiate sporangial and oogonial sori and bordering antheridial sori, a multicellular sporangial stalk, oogonia without a stalk cell, and uniflagellate spermatozoids. Zonaria harveyana and Homoeostrichus olsenii are similar in sporangial structure, but they cannot be assigned to either Zonaria or Homoeostrichus as now defined and consequently are transferred to the new genus, Exallosorus, as Exallosorus harveyanus (Pappe ex Kützing) comb. nov. and Exallosorus olsenii (Womersley) comb. nov. Exallosorus is distinguished from Zonaria and Homoeostrichus using sporangial characters: tetrasporangia have a stalk cell and are closely packed into indusiate sori which lack paraphyses and mucilage. Problems in obtaining reliable species identification for the five southern Australian species of Zonaria are partially resolved. This study demonstrates again the essential contribution of reproductive characters to the definition of taxa and supports the position that decisions to erect, subsume or transfer members of the Dictyotales should be based on more than vegetative characters.

Autotrophic picoplankton (APP) abundance, primary production and vertical distribution were studied in the oligotrophic Lake Stechlin (northeastern Germany) in 1994 and 1995. Within the euphotic zone APP contributed 34% of phytoplankton biomass and 35% of primary production. Annual average APP cell number was 209×103 cells ml−1, of which 95% were unicellular cyanobacteria, 2% were colonial cyanobacteria and 3% were eukaryotes. Three ecologically and/or morphologically different groups of APP were recognized: (i) unicellular cyanobacteria belonging to the genus Cyanobium, (ii) eukaryotic species growing in early spring under isothermal conditions and (iii) cyanobacteria, partly colonial species, growing in the stratified period in the euphotic zone. Three species of eukaryotic green algae were identified: Choricystis minor, Neocystis diplococca and Pseudodictyosphaerium jurisii, the latter two being colonial. This is the first record of the occurrence of colonial eukaryotes potentially of APP size in fresh waters. In summer picocyanobacteria were highly productive so the low net increase rates indicate that losses must be high. The dominant, Cyanobium population started growing in February with maximum abundance in late April, contributing significantly to the spring peak in phytoplankton biomass. During this growth period, the population was evenly distributed in the 60 m water column. By the time the maximum biomass occurred, inorganic nutrients had decreased below analytically detectable levels. Parallel to the onset of stratification a part of the population was grazed, most probably in the microbial loop and primarily in the upper 10–15 m. The rest of the Cyanobium population accumulated in a narrow layer in the upper hypoliminon. The APP remaining from the spring was persistent for much of the summer in this cold, high-nutrient (especially nitrate)/low-light environment. Short phosphorus-turnover times suggest that APP is probably phosphate-limited. The stability of the thermocline and the pattern of thermocline development in May affected the accumulation of the APP cells in the upper hypolimnion. Thus, this process is sensitive to the physical stability of the water column.

In Porphyra umbilicalis, circadian rhythms of nuclear division and growth activity persisted for at least 7 cycles in continuous white fluorescent light, with a period of 21 h for the free-running growth rhythm at 15 μmol m−2 s−1 and 10°C. Growth rhythmicity was lost at irradiances above 20 μmol m−2 s−1. The growth and mitotic rhythms seem to be driven in parallel by circadian rhythmicity, and the details of growth kinetics must be due mainly to the growth behaviour of non-dividing cells. This was inferred from the finding that individual cells continued to grow from one cell division to the next, with generation times of 2–6 days. Transfer from continuous light to 12[ratio ]12 h light[ratio ]dark synchronized the free-running growth rhythm, with a high growth peak appearing every 24 h at the start of the light phase and an increase in growth rate during the dark phase. The ascending portion of the free-running growth curve was thus shifted into the night phase of the diurnal regime and the descending portion into the light phase. This behaviour of decreasing growth activity after the morning hours, previously found in a brown and a green marine macroalga, ensures that cell activity is available primarily for photosynthesis during the day.

Growth rates in terms of area expansion per 30 min were measured in the red alga Porphyra leucosticta under different light/dark (LD) cycles, irradiance levels and light qualities. Twelve hours per day under white light (35 μmol m−2 s−1) promoted a dramatic and rapid increase in thallus expansion during the first part of the light period followed by low values during the remaining light and dark phase. Light/dark regimes of 8[ratio ]8 h, 6[ratio ]6 h, 3[ratio ]3 h and 1·5[ratio ]1·5 h resulted in a discontinuous growth pattern where thallus expansion was restricted to the light phase while no growth occurred in the dark phase, and there was even a significant thallus contraction just after light-off. Growth rates in terms of percentage area increased with irradiance at LD 12[ratio ]12. This was due to: (1) the phase of high growth rate following the initial ‘morning peak’ extending from the first 6 h of the light phase to cover almost the entire light phase, and (2) an increase in growth rate during the second half of the night phase. Saturation was reached at an irradiance of 125 μmol m−2 s−1. An irradiance of 500 μmol m−2 s−1 decreased thallus expansion rate during the light and dark culture phases relative to the other irradiances used. Light quality also caused changes in growth rate: the maximum value was reached under yellow light followed by red; both of these were higher than the growth rate under white light. Blue light (35 μmol m−2 s−1) caused a decrease of more than 50% in thallus expansion compared with white or red light due to a very dramatic decrease in the ‘morning peak’ of thallus expansion and very low values in the rest of the light and dark phases. Increasing the percentage of blue light in different white light sources also decreased thallus expansion. It is suggested that light influences the growth of Porphyra in three ways: as an external signal matching thallus expansion with light and dark cycles; as an energy source promoting thallus expansion with increasing irradiance; and, at a morphogenetic level, inhibiting growth in blue light.
