The nutritional effects of giving raw (RSF) or heated (HSF) soya-bean flour to young guinea-pigs were investigated in trials 1 and 2, in which the levels of dietary protein were 120 and 190 g/kg diet respectively. The growth rate of animals fed on RSF was lower than that of those fed on HSF. Growth retardation of guinea-pigs fed on RSF was accompanied by a lower apparent digestibility of the protein (0.49–0.53) compared with HSF (0.67–0.76) and lower food conversion efficiency. In RSF-fed animals, increasing dietary protein affected growth and food conversion efficiency negatively. The pancreas of animals fed on RSF and HSF was similar in weight but secreted less trypsin, chymotrypsin and amylase, in RFS-fed animals. It was concluded that the mechanism by which raw soya-bean negatively affects the growth rate of guinea-pigs by reducing the activity of intestinal enzymes, differs from that suggested for rats and chicks, but is similar to that of pigs and calves.

A relationship between bile acid conjugation and hepatic taurine concentration was investigated in rats fed on citrus pectin. When rats were fed on the diets containing varying amounts of pectin (10, 30, 60 and 100 g/kg dietary levels), biliary excretion of bile acids increased as the dietary levels of pectin increased. The increase was entirely due to the glycine-conjugated bile acids. The biliary excretion of taurine-conjugated bile acid was somewhat decreased as the dietary level of the fibre increased. Consequently, most of the bile acids were conjugated with glycine in rats fed on the diet containing 100 g pectin/kg. On the other hand, dietary cellulose (60 and 100 g/kg) did not affect the biliary bile acid excretions. The major proportion of bile acids in rats receiving a fibre-free diet and the diets containing cellulose were conjugated with taurine. Hepatic taurine concentrations decreased as the dietary levels of pectin, but not of cellulose, increased. Although dietary pectin (100 g/kg) also slightly decreased the taurine concentration in the kidney, those concentrations in other non-hepatic tissues examined (heart, brain and serum) were unaffected by the dietary fibre. Supplementation of the diet containing 100 g pectin/kg with methionine (10 g/kg) and taurine (10 and 50 g/kg) strikingly increased hepatic taurine concentrations. In this situation, the conjugation of bile acid with glycine was almost abolished and taurine conjugates became abundant in the bile of these animals. It is suggested that dietary pectin mediated an increase in the biliary bile acid excretion which may have depleted the hepatic pool of taurine available for bile acid conjugation and, thus, increased glycine conjugation of bile acids.

The hypothesis that the energy cost of human pregnancy can be minimized by energy-sparing metabolic adaptations was tested using serial 24 h whole-body calorimetry. Eight healthy, well-nourished women were studied prepregnant and at 6, 12, 18, 24, 30 and 36 weeks gestation. Basal metabolic rate (BMR) showed highly characteristic changes within each subject and large inter-individual differences (F 3.5, P < 0.01). Some subjects showed a highly significant depression of metabolism up to 24 weeks gestation in support of the initial hypothesis. At 36 weeks BMR ranged from +8.6 to +35.4% relative to the prepregnant baseline. This wide variability was not explained by differences in the amount of lean tissue gained. Women displaying the energy-sparing suppression of BMR tended to be thin, suggesting that changes in metabolism may be responsive to initial energy status (ΔBMR ν. prepregnant body fat: r 0.84, P < 0.005). Changes in 24 h energy expenditure closely paralleled changes in BMR (r 0.98, P < 0.001), since the energy cost of minor voluntary activity and thermogenesis remained very constant within each individual. Pregnancy decreased the net cost of weight-dependent and weight-independent standard exercises when expressed per kg body-weight: stepping – 10 (sd 2)%, P < 0.001 at 18–36 weeks, cycling - 26 (sd 7)%, P < 0.01 at 12–36 weeks. The average integrated maintenance costs of pregnancy matched previous group estimates from well-nourished women, but individual estimates ranged from - 16 to + 276 MJ (coefficient of variation 93%). This high level of variability has important implications for the prescription of incremental energy intakes during pregnancy. It may also have had evolutionary significance.

The brain metabolism of glutamate and γ-aminobutyrate (GABA) was investigated in thiamin-deficient and pair-fed control rats, in order to determine whether the GABA shunt may provide an important alternative to 2-oxo-glutarate dehydrogenase (EC 1.2.4.2) in energy-yielding metabolism in thiamin deficiency. Brains from thiamin-deficient animals contained less glutamate, 2-oxo-glutarate and GABA than those from control animals. The brain content of ATP was unaffected by thiamin deficiency. After intracerebroventricular injection of [14C]glutamate, the specific radioactivity of GABA in the brains from deficient animals was 45–50% higher than that in controls, suggesting a considerable increase in the metabolic flux through the GABA shunt in thiamin deficiency. Brain GABA showed a marked circumannual variation, with a peak in mid-summer and a minimum value in mid-winter.

The objective of the present study was to measure changes in splanchnic blood flow and oxygen consumption in sheep fed on a high-concentrate diet ad lib. (ADLIB) or an amount sufficient to maintain body-weight (MAINT) for 21 d. Eleven ram lambs were surgically implanted with chronic indwelling catheters in the portal, hepatic and mesenteric veins and mesenteric artery to measure blood flow and net O2 flux through the liver and portal-drained viscera (PDV). During the 21 d period, PDV (P < 0.05) and liver (P < 0.01) blood flow increased in ADLIB and decreased in MAINT lambs (treatment x day, linear). After 21 d, O2 consumptions in PDV and liver of MAINT lambs were 37 and 63% lower than in ADLIB lambs. In the control period, total splanchnic tissues represented an average of 52% of whole body O2 consumption. After 21 d, the relative contributions of PDV and liver to whole-body O2 consumption were 28 and 41% in ADLIB and 19 and 22% in MAINT lambs respectively. Allometric regression variables indicate that liver O2 consumption responds more rapidly to changes in metabolizable energy intake than portal O2 consumption. These results indicate that blood flow and O2 consumption in both PDV and liver are related to level of nutrition. Furthermore, splanchnic tissues represent a significant component of whole-body O2 consumption that is subject to manipulation by level of nutrition.

The quantity of re-ingested faeces was calculated by comparing faecal dry matter of unrestricted rats and coprophagy-restricted rats after correcting for differences in food intake. Due to high day-to-day variations of produced and re-ingested faeces it was not possible to calculate precisely the extent of coprophagy of an individual rat at a particular day with this difference method. Reliable quantitative estimates require at least two rats and a collection period of 7 d. When fed on a nutritionally complete diet, rats re-ingested 0-11 % of their faeces. When fed on low-protein diets (66 g egg albumin/kg) or diets diluted with 200 g cellulose/kg, coprophagy was not significantly increased. A high re-ingestion rate (6–25 %) was observed with thiamin and pantothenic acid deficiencies. After re-ingestion of faeces had been prevented for 1 week, the amount of faeces re-ingested during the subsequent week without tail-cups was increased twofold. It is concluded that rats are able to regulate the amount of faeces eaten precisely according to their requirements.

Experiments were made to estimate separately the amino acid requirements of growing pigs for maintenance and for protein accretion. The relationship between nitrogen retention and amino acid intake was estimated for each essential amino acid (except histidine) by giving, at rates of N intake of 0.25 and 2.0 g/kg body-weight (W)0.75 per d, diets in which one amino acid was made specifically deficient. From the regression coefficients it was calculated that, for the accretion of 1 g body protein, the dietary amino acid requirements were (mg) threonine 47, valine 53, methionine+cystine 36, methionine 19, isoleucine 43, leucine 78, phenylalanine+tyrosine 84, phenylalanine 41, lysine 68 and tryptophan 12. The daily amino acid requirements for N equilibrium were also estimated. From the relationship between N retention and amino acid intake the daily amino acid requirements for N equilibrium were estimated to be (mg/kg W0.75 per d) threonine 53, valine 20, methionine+cystine 49, methionine 9, isoleucine 16, leucine 23, phenylalanine+tyrosine 37, phenylalanine 18, lysine 36 and tryptophan 11. It was estimated that both for maintenance and for protein accretion tyrosine could provide close to half the total phenylalanine+tyrosine needs. Cystine could supply close to half the total sulphur amino acid needs for protein accretion but 0.8 of the needs for maintenance.

The effects of potassium deficiency on growth, K content and protein synthesis have been compared in 4–13-week-old rats. When maintained on K-deficient fodder (1 mmol/kg) rats ceased to grow within a few days, and the incorporation of [3H]leucine into skeletal muscle protein in vivo was reduced by 28–38%. Pair-feeding experiments showed that this inhibition was not due to reduced energy intake. Following 14 d on K-deficient fodder, there was a further reduction (39–56 %) in the incorporation of [3H]leucine into skeletal muscle protein, whereas the incorporation into plasma, heart and liver proteins was not affected. The accumulation of the non-metabolized amino acid α-aminoisobutyric acid in the heart and skeletal muscles was not reduced. The inhibitory effect of K deficiency on 3H-labelling of muscle protein was seen following intraperitoneal (10–240 min) as well as intravenous (10 min) injection of [3H]leucine. In addition, the incorporation of [3H]phenylalanine into skeletal muscle protein was reduced in K-depleted animals. Following acute K repletion in vivo leading to complete normalization of muscle K content, the incorporation of [3H]leucine into muscle protein showed no increase within 2 h, but reached 76 and 104% of the control level within 24 and 72 h respectively. This was associated with a rapid initial weight gain, but normal body-weight was not reached until after 7 weeks of K repletion. Following 7 d on K-deficient fodder the inhibition of growth and protein synthesis was closely correlated with the K content of the fodder (1–40 mmol/kg) and significant already at modest reductions in muscle K content. In vitro experiments with soleus muscle showed a linear relationship between the incorporation of [3H]leucine into muscle protein and K content, but the sensitivity to cellular K deficiency induced in vitro was much less pronounced than that induced in vivo. Thus, in soleus and extensor digitorum longus (EDL) muscles prepared from K-deficient rats, the incorporation of [3H]leucine was reduced by 30 and 47 % respectively. This defect was completely restored by 24 h K repletion in vivo. It is concluded that in the intact organism protein synthesis and growth are very sensitive to dietary K deficiency and that this can only partly be accounted for by the reduction in cellular K content per se. The observations emphasize the need for adequate K supplies to ensure optimum utilization of food elements for protein synthesis and growth.

Characterization of the carbohydrates of pea (Pisum sativum) hulls, carrot and cabbage using both colorimetric and gas–liquid chromatographic techniques permitted a detailed investigation into the extent of digestion of differing types of fibre. These digestion studies were greatly aided by the development of a rapid bioassay employing starved adult cockerels. Total collection of undigested residues, uncontaminated by food spillage, could be made from trays placed under the cockerels. Chemical analysis showed that pea hulls consisted mainly of fibre with very little available carbohydrate present, whereas more than half of freeze-dried carrot and cabbage consisted of available carbohydrate (sucrose, glucose, fructose, starch) and consequently considerably less fibre was present. The fibre of carrot and cabbage was similarly composed of nearly equal amounts of neutral and acidic polysaccharides, whereas pea-hull fibre had four times as much neutral as acidic polysaccharides. The digestibility of total neutral polysaccharides from all three foodstuffs was extremely low. However, there appeared to be preferential digestion of polysaccharides composed of rhamnose, arabinose and galactose residues, all associated with pectic material, in contrast to the indigestibility of polysaccharides composed of fucose, xylose and glucose. Acidic polysaccharides were digested to a greater extent than neutral ones, and those of carrot and cabbage more so than pea hulls. The polysaccharides which were the most soluble were also the most digestible, but due to the arbitrariness of polysaccharide solubility, quantification of their total digestibility per se was considered not possible.

The aim of the present study was to determine how the long-term metabolic adaptations to winter shearing of the pregnant ewe result in significant changes in the rates of lipid mobilization and utilization of non-esterified fatty acids (NEFA) in comparison with unshorn controls. Continuous infusions of [1-14C]palmitic acid, [2-3H]glycerol and NaH14CO3 were used to measure whole-body lipid metabolism in fed (estimated metabolizable energy (ME) intake 9.54 MJ/d) and under-fed (estimated ME intake of 3 MJ/d), shorn and unshorn sheep over the final 4 weeks of pregnancy. Whole-body carbon dioxide, estimated heat production, total NEFA entry and oxidation rates were all significantly higher in fed shorn ewes compared with unshorn controls, even though there was no difference in the arterial plasma NEFA concentration. These differences may be mediated via an increase in the plasma concentrations of thyroid hormones in shorn animals. As a result of under-feeding any significant differences in lipid metabolism between shorn and unshorn groups were removed. In all sheep the mean total NEFA entry rate as measured using [1-14C]palmitic acid was 3.4 times the value obtained using [2-3H]glycerol. It is concluded that when sheep are fed on a diet from which no more than half the required ME for late pregnancy is obtained, then lipolysis of body fat depots occurs via the incomplete breakdown of adipose tissue triglycerides. This effect is significantly greater in the fed shorn pregnant ewe which exhibits higher entry and oxidation rates of NEFA.

The concentrations of holotrichs, reducing sugars and fermentation end-products, and the fluid dilution rates, in the cranial (CR), ventral (V) and caudal (CA) regions of the reticulo-rumen of cows were compared. Additionally, holotrich numbers in fluid and solid digesta samples taken from the dorsal (D) region were determined at different time-intervals. Holotrich numbers expressed per ml fluid at site D were almost twofold those expressed per g solid digesta, and the fluctuation of their numbers in fluid preceded that in solid digesta. Holotrich concentrations at site CR were higher before feeding than after feeding, while those at sites V and CA were lower before feeding than for a few hours after feeding. At sites V and CA, holotrich concentrations fluctuated in good agreement with reducing sugar concentrations, but the increase in the former always preceded that in the latter. The concentration of volatile fatty acids (VFA) tended to decrease from site CA to site CR, while the dilution rate was highest in site CR. The results suggest that migration of holotrichs from the reticulum to the rumen and vice versa are the cause of fluctuation in their numbers, and also the mechanism by which they are retained in the rumen of cattle.

During early lactation ruminants can mobilize considerable amounts of body fat to maintain milk production. The aim of the present study was to evaluate the efficacy of tritiated water (TOH) and non-esterified fatty acid (NEFA) kinetics as means of monitoring adipose tissue fat mobilization in lactating goats. Body fat, as estimated by a two-pool model of TOH kinetics, and NEFA entry rate were measured in four primiparous goats at days 11, 37 and 72 post partum. Estimated body fat decreased by an average of 64 g/d between days 11 and 37 of lactation, tending to increase between days 37 and 72. Plasma NEFA concentrations and NEFA entry rate decreased as lactation advanced, being significantly lower at day 72 than at day 11 of lactation. Both plasma concentrations of NEFA and NEFA entry rate were negatively correlated with calculated energy balance. Plasma NEFA concentrations and NEFA entry rate at days 11 and 37 of lactation were positively related to average body fat losses over the subsequent stage of lactation. These results demonstrate that NEFA kinetics reflect fat mobilization in primiparous lactating goats, particularly during negative energy balance.

Seven measurements of the effect of clenbuterol on basal nitrogen excretion (UNE), and protein turnover were made in six female sheep. The sheep were sustained by the intraruminal infusion of energy as volatile fatty acids to provide maintenance, but given no protein (N-free) for 12 d (6 d control, 6 d clenbuterol). Clenbuterol reduced UNE by 20 %, but only on day 2 of the 6 d subperiod. Protein flux (equivalent to degradation on N-free nutrition), measured on day 6 by the irreversible loss of leucine was significantly increased (12%) by clenbuterol. Amino-N oxidation measured by N excretion was unchanged and, therefore, protein synthesis was also increased. During the 12 d N-free period, the recovery of urinary total N (Kjeldahl) as the sum of urea, ammonia, creatinine and purine derivatives, declined from 87.7 to 74.2 %. The form of this missing N was not identified. The effect of clenbuterol of increasing both degradation and synthesis is unlike that reported in the literature for animals receiving protein when, in general, synthesis is unchanged and degradation reduced. This could be due to a different effect of clenbuterol in the N-free state, or to unchanged effects on protein pools other than muscle whose relative contribution to protein metabolism is different in the N-free state.

The effect of post-rumen supply of nutrients on the rumen digesta load and voluntary consumption of roughage by Border Leicester × Merino ram lambs was investigated. A chopped wheaten hay was offered to the ram lambs (n 24), whose nutrient intake was altered by infusing into the abomasum a liquid supplement, containing reconstituted cow's milk, sodium caseinate, minerals and vitamins, for a period of at least 30 d. Either two or three lambs were allocated by live weight to each of ten rates of nutrient supplementation. The lambs were slaughtered at a target live weight of 31 kg. Voluntary intake of hay was estimated over the last 7 d of feeding. The amounts of digesta and organic matter (OM) in the rumen were measured by emptying after slaughter. The particle size distribution of the digesta was measured by wet sieving and the fractional outflow rate of particulate matter by reference to the marker lignin. Growth rate of the lambs increased linearly as total energy infused increased. The linear increase in growth rate indicated that roughage was not substituted for infused nutrients on a direct energy basis. OM intake from hay declined linearly as the amount of nutrients infused increased. The decline in intake was associated with a decline in both total digesta and the amount of rumen OM. Both the fractional digestion rate of OM and the fractional outflow rate of particulate matter from the rumen were unaffected by the amount of nutrients infused. The results indicated that the rumen digesta load of young sheep fed on a single roughage is directly related to their energy deficit, that is the difference between the capacity of the animals to use energy and the energy available to them for metabolism.

Oxidation of dl-2-hydroxy 4-methylthiobutanoic acid (dl-HMB), dl-methionine (dl-MET) and l-methionine (l-MET) in chicken tissue homogenates was compared using 1-14C-labelled tracers. The pattern of oxidation of the substrates was similar at both low (0.7 mm) and high (20 mm) concentrations. The rate of conversion to 2-keto 4-methylthiobutanoic acid (KMB) was highest for dl-MET and lowest for l-MET in kidney, liver and intestinal mucosa. In breast muscle, rates for dl-MET and l-MET were similar at 0.7 mm, but dl-HMB showed the highest rate at 20 mm. Kidney contained the highest specific activity for oxidation of all three substrates. Raising the pH of liver and kidney homogenates from 7.5 to 8.6 increased the oxidation of dl-MET, exclusively. Experiments with inhibitors of D-2-hydroxy acid dehydrogenase (EC 1.1.99.6) and L-2-hydroxy acid oxidase (EC 1.1.3.15) suggested that d- and l-HMB were stereospecifically oxidized by the enzymes. KMB stimulated l-MET oxidation in kidney yet inhibited l-MET oxidation in liver homogenates. The effect of KMB on dl-MET and dl-HMB oxidation also varied between tissues. Amino-oxyacetate inhibited l-MET oxidation completely and dl-MET and dl-HMB oxidation almost completely in both kidney and liver. L-Cycloserine was less potent than amino-oxyacetate and decreased l-MET oxidation more in kidney than in liver. It can be calculated from the results that, at low substrate concentrations, the liver contributes principally to the whole body oxidation of both dl-HMB and dl-MET. At high (greater than physiological) concentrations, dl-HMB would be oxidized principally in skeletal muscle. At all concentrations, l-MET would be converted to KMB mainly in the muscle.
