Quercetin, a vital flavonoid found in many medicinal plants, has shown anti-inflammatory, anti-cancerous, anti-aging, anti-tumour, anti-viral, anti-fungal, anti-bacterial, anti-obesity, anti-diabetic, and anti-protozoal activity. However, very little is known of its anthelmintic activity; there is no literature against tapeworm infection so far. The present study was performed to expose its cestocidal role by using the zoonotic tapeworm Hymenolepis diminuta as a parasite model. The parasite was exposed to different concentrations of 0.125, 0.25, 0.5, 1, 2.5, 5, 10, 20, and 40 mg/mL Quercetin prepared in RPMI 1640, with 1% Tween 20. Another set of parasites was treated with a standard dose of Praziquantel (0.001 mg/ml), and another set of parasites was kept as control. All experiments were maintained at 37°C ± 1°C in the incubator. Quercetin activity was assessed through viability test, and time of motility was observed through paralysis. After the experiment, worms were processed for light and electron microscopic analysis to observe the post-treatment effect on their tegument. Dose-dependent efficacy was observed in all the treatments. Time of paralysis and time of mortality for 20 mg/mL Quercetin dose was 1.40±0.03h and 2.35±0.03h, respectively, which is at par with the drug Praziquantel. Histological study showed constrictions in the tegument, while extensive damage in suckers and neck region with deformed and shrunken proglottids, sloughed-off microtriches and undistinguished nucleus with loss of envelope architecture were observed in treated parasites under electron microscopic studies, which indicates the negative activity of Quercetin on the parasite thus suggesting its cestocidal activity.

A new species of bothriocephalid cestode in the genus Anantrum is described from the intestine of the shorthead lizardfish Synodus scituliceps from the north Pacific coast of Costa Rica. The new species is described based on an integrative taxonomic approach that includes the use of light and scanning electron microscopy, 28S rDNA sequencing, and phylogenetic analysis. Anantrum gallopintoi sp. nov. is the third known member of this genus and can be distinguished from A. tortum (Linton, 1905) and A. histocephalum Jensen & Heckmann, 1977 by a combination of morphological and ecological traits and, in particular, by having a vaginal sphincter, different number of testes, and different type host and type locality. The molecular analysis and the phylogenetic reconstructions supported its status as a new taxon placing it within a well-supported separate branch of Anantrum spp. subclade. According to the present finding, S. scituliceps represents a new host record and Costa Rica a new geographical record for Anantrum species, and, in general, for a bothriocephalid cestode.

The endemic chub Squalius tenellus (Heckel, 1843) was introduced more than 100 years ago to Lake Blidinje (Bosnia-Herzegovina). Only 1 species of enteric helminth was found in a sample of 35 chubs, the tapeworm Caryophyllaeus brachycollis (Janiszewska, 1953). The paper includes histopathological investigation with identification of innate immune cells involved in host reaction and molecular data allowed correct designation of the cestode species. Of 35 specimens of chub examined, 21 (60%) harboured individuals of C. brachycollis and a total of 1619 tapeworms were counted, the intensity of infection ranged from 1 to 390 worms per fish (46.2 ± 15.3, mean ± s.e.). Histopathological and ultrastructural investigations showed strict contact between the worm's body and the epithelia and increase in the number of mucous cells, rodlet cells among the epithelial cells. Within the tunica propria-submucosa, beneath the site of scolex attachment, numerous neutrophils and mast cells were noticed. This is the first study of the occurrence of C. brachycollis in chub from Lake Blidinje and on the response of the innate immune cells of S. tenellus to this tapeworm. Interestingly, in 3 very heavily infected chubs, perforation of the intestinal wall was documented; this is uncommon among cestodes which use fish as a definitive host.

In this study, we report the first complete mitochondrial genome of the tapeworm Nippotaenia mogurndae in the order Nippotaeniidea Yamaguti, 1939. This mitogenome, which is 14,307 base pairs (bp) long with an A + T content of 72.2%, consists of 12 protein-coding genes, 22 transfer RNA (tRNA) genes, two rRNA genes, and two non-coding regions. Most tRNAs have a conventional cloverleaf structure, but trnS1 and trnR lack dihydrouridine arms of tRNA. The two largest non-coding regions, NCR1 (220 bp) and NCR2 (817 bp), are located between trnY and trnS2 and between nad5 and trnG, respectively. Phylogenetic analyses of mitogenomic data indicate that N. mogurndae is closely related to tapeworms in the order Cyclophyllidea.

Three new species of ‘tetraphyllideans’ in the family Serendipeidae are described from cownose rays off Senegal. Duplicibothrium jeannettae n. sp. and Duplicibothrium collosum n. sp. parasitize both Rhinoptera marginata and Rhinoptera peli. Duplicibothrium jillae n. sp. parasitizes R. marginata. Duplicibothrium jeannettae n. sp. and D. jillae n. sp. differ from one another and their described congeners in size, number of proglottids, and locular configuration. The generic assignment of D. collosum n. sp. was initially unclear because its scolex more closely resembles that of Serendip than Duplicibothrium. To help inform generic placement, sequence data were generated for the 28S rDNA gene (D1–D3 region) for the three new species as well as for Duplicibothrium minutum and Duplicibothrium n. sp. 2 from Rhinoptera bonasus off North Carolina, Duplicibothrium n. sp. 3 from Rhinoptera brasiliensis off Belize, Duplicibothrium n. sp. 4 and Duplicibothrium n. sp. 5 from Rhinoptera jayakari off Mozambique, and Duplicibothrium n. sp. 6 from Rhinoptera neglecta off Australia. Comparable data were obtained from GenBank for adults and larvae of Duplicibothrium collected from the Gulf of Mexico. The tree resulting from a maximum likelihood analysis (MLA) placed D. collosum n. sp. robustly within Duplicibothrium; the generic diagnosis is emended accordingly. This raises a question regarding the independence, and thus also validity, of the three genera of the Serendipeidae – a question that must await a molecular analysis that includes Serendip and Glyphobothrium. These results extend the hosts of Duplicibothrium to include R. brasiliensis, R. jayakari, R. marginata, R. neglecta and R. peli, and the distribution to include the western Pacific Ocean, eastern Atlantic Ocean, and western Indian Ocean (Zoobank Registration: 97BB5020-BFFF-4FEA-AE07-B4711D1110FC).

Manipulative parasites with complex life cycles are known to induce behavioural and physiological changes in their intermediate hosts. Cyathocephalus truncatus is a manipulative parasite which infects Gammarus pulex as intermediate host. G. pulex males display pre-copulatory mate guarding as a response to male-male competition for access to receptive females. In this paper, we tested the influence that C. truncatus-infection might have on male G. pulex sperm number and pairing success. We considered 3 classes of G. pulex males in our experiments: (i) uninfected males found paired in the field, (ii) uninfected males found unpaired in the field, or (iii) infected males found unpaired in the field. Both infected males and uninfected unpaired males paired less with a new female than uninfected paired males did. Furthermore, infected males appear to be at a strong disadvantage when directly competing for females with a healthy rival male, and had fewer sperm in their testes. We discuss the potential effect of male and female mating strategies on such male host mating alteration.

Reproductive parameters of Ligula intestinalis-infected roach (Rutilus rutilus) which were held under long-tem laboratory conditions with unlimited food supply were investigated. Although uninfected and infected roach showed no difference in condition factor and both groups deposited perivisceral fat, the gonadosomatic-index was significantly lower in infected female and male roach. Quantitative histological analysis revealed that gonad development was retarded upon parasitization in both genders. In contrast to the phenotype described in the field, infected females were able to recruit follicles into secondary growth, but a high percentage of secondary growth follicles underwent atresia. In both genders, the histological data corresponded well with reduced expression of pituitary gonadotropins and lowered plasma concentrations of sex steroids, as revealed by real-time RT-PCR and ELISA, respectively. Furthermore, a reduction of vitellogenin mRNA and modulated expression of sex steroid receptors in the liver was demonstrated. Like in the field, there was a significant adverse impact of L. intestinalis on host reproductive physiology which could not be related to parasite burden. Our results show, for the first time, that maintenance under laboratory conditions can not abolish the deleterious effect of L. intestinalis on gametogenesis in roach, and indicate a specific inhibition of host reproduction by endocrine disruption.

Analyses of laboratory-based helminth-rodent model systems have been immensely useful in delineating the workings of the mammalian immune system. Investigations in the 1970s–1980s on the fate of the rat tapeworm, Hymenolepis diminuta, in rats and mice and the systemic and local responses evoked following infection have contributed directly to our knowledge of how permissive and non-permissive hosts respond to the challenge of infection with a helminth parasite. This convenient laboratory model system has, in the authors' opinion, regrettably received considerably less attention in recent years. With the goal of highlighting the utility of this model system, data is presented on: (1) the immune and enteric responses of rats and mice to infection with H. diminuta; (2) the ability of excretory or secretory products derived from H. diminuta to significantly reduce T cell and macrophage activation in vitro; and (3) how assessment of H. diminuta-rodent models can be used to identify immune effector or regulatory mechanisms that can be translated into novel treatments for inflammatory and autoimmune disorders.

Temporal changes in glucose transport capacity in excysted scoleces of Hymenolepis diminuta were examined. Assays involved incubation for 1 min in [3H]glucose after pre-incubation for 1 min to 8 h in saline. There were two abrupt increases in uptake velocity, a relatively small one between 15 and 75 min, and a large one between 5 and 6 h, during which the Vmax increased from 0·36 to 2·49 nmol/25 larvae/h. The second increase was unaffected when the pre-incubation saline contained 5 mM glucose, but it was completely blocked when the excysted larvae were pre-incubated in Ca2+-free saline. Abrupt glucose transport changes did not occur in intact cysticercoids or in scoleces when the substrate was [3H]leucine or [3H]uracil. Arrhenius plots (log V versus 1/temperature, 10–42 °C) were linear for intact cysticercoids, but were biphasic for both scoleces and adults with discontinuities at 20 ± 1 °C. Thus, ‘activation’ of the excysted scolex seemed to involve a specific, Ca2+ -dependent increase in number of glucose transporters functioning in the worm surface. The Arrhenius plots indicated that development in the final host does not involve a major change in lipid composition of the parasite's membranes.

Hymenolepis diminuta migrates up the small intestine in response to feeding the host 1 g of glucose. Locomotion during migration may result from fixed patterns of retrograde peristaltic-like waves in the strobila of the tapeworm which propel the organism against the normal expulsive forces in the small intestine. The peristaltic-like locomotory waves occur in a gradient along the strobila with a frequency of 24·9±0·9 cycles/min in the anterior segments of the worm, decreasing linearly to 6·6±1·4 cycles/min in the posterior segments of the worm. Chemical signals, isolated from the small intestine of fed hosts, which stimulate migration behaviour in vivo do not alter the behaviour of the scolex or strobila in vitro. Removal of the scolex containing the cerebral ganglia does not alter the frequency or pattern of locomotory activity in the strobila. After the worm is cut into pieces, each region generates the pattern of locomotory activity that is appropriate for that region. These data suggest that the peripheral nervous system, and not the central nervous system, is responsible for the coordination of the fixed patterns of locomotory activity in these tapeworms.

Many significant diseases of human civilization are thought to have arisen concurrently with the advent of agriculture in human society. It has been hypothesised that the food produced by farming increased population sizes to allow the maintenance of virulent pathogens, i.e. civilization pathogens, while domestic animals provided sources of disease to humans. To determine the relationship between pathogens in humans and domestic animals, I examined phylogenetic data for several human pathogens that are commonly evolutionarily linked to domestic animals: measles, pertussis, smallpox, tuberculosis, taenid worms, and falciparal malaria. The majority are civilization pathogens, although I have included others whose evolutionary origins have traditionally been ascribed to domestic animals. The strongest evidence for a domestic-animal origin exists for measles and pertussis, although the data do not exclude a non-domestic origin. As for the other pathogens, the evidence currently available makes it difficult to determine if the domestic-origin hypothesis is supported or refuted; in fact, intriguing data for tuberculosis and taenid worms suggests that transmission may occur as easily from humans to domestic animals. These findings do not abrogate the importance of agriculture in disease transmission; rather, if anything, they suggest an alternative, more complex series of effects than previously elucidated. Rather than domestication, the broader force for human pathogen evolution could be ecological change, namely anthropogenic modification of the environment. This is supported by evidence that many current emerging infectious diseases are associated with human modification of the environment. Agriculture may have changed the transmission ecology of pre-existing human pathogens, increased the success of pre-existing pathogen vectors, resulted in novel interactions between humans and wildlife, and, through the domestication of animals, provided a stable conduit for human infection by wildlife diseases.
