The description of the molecular processes which underlie visual excitation is the fundamental problem in understanding vision at the level of a single photoreceptor. Thus far only a general outline of photoreceptor function has emerged with little known about actual biochemical and biophysical mechanisms.

Glucose is one of the substrates that is controlled with the most efficient hormonal mechanisms in higher organisms. The presence of tissues such as the central nervous system which, under normal conditions, depend solely on glucose as substrate, and the sporadic type of food intake with periods of fasting of various lengths in the mammalians necessitate that the distribution of energy-rich substrates among various tissues be continuously adjusted by changes in the secretion of a number of hormones. The efficiency of this system is evidenced by the stability of the blood glucose level in man, in whom after a carbohydrate-rich meal more than 70% of the glucose that has been ingested will be retained in the liver during a single passage of portal blood, resulting in only small changes of the glucose concentration in peripheral blood. Likewise, periods of fasting up to24–36 h are followed by modest to minimal reductions of the blood glucose level, the liver now supplying the circulation with the hexose.

The basic principles underlying fluctuation phenomena in thermodynamics have long been understood (for reviews see Kubo, 1957; Kubo, Matsuo & Kazuhiro 1973 Lax, 1960). Classical examples of how fluctuation analysis can provide an insight into the corpuscular nature of matter are the determination of Avogadro's number according to Einstein's theory of Brownian motion (see, e.g. Uhlenbeck & Ornstein, 1930; Kac, 1947) and the evaluation of the electronic charge from the shot noise in vacuum tubes (see Van der Ziel, 1970).

It is our purpose to review recent experiments on haemoglobin in order to discuss them in terms of the two state model of cooperativity. Excellent previous reviews are available of the chemistry of haemoglobin (Antonini & Brunori, 1971; Gibson, 1959b) which are referred to when possible. The study of differentiation is concerned with the analysis of the processes responsible for ‘the cellular changes in macromolecular synthesis and composition, patterned in time and space and resulting in specialized functions, forms and organization’ (Moscona, 1973).

To the Chemist, it has long been second nature to work with separate, purified substances. Biochemists and molecular biologists have now largely achieved the same working position, with their powerful separation techniques of ultracentrifugation, electrophoresis, chromatography and the like. For the cell biologists, it is a much more recent phenomenon that widespread attention is being directed towards quantitative measurement of properties in isolated, homogeneous cell types. The richness and diversity of cell systems beg for cleverness on the part of the experimenter.

The concept that the liquid crystalline or mesomorphic condition was of importance to biological systems is a relatively old idea. Thus Bernal (1933) when discussing the different types of arrangements of molecules in liquid crystals commented ‘Such structures belong to the liquid crystal as a unit and not to its molecules which may be replaced by others without destroying them and they persist in spite of the complete fluidity of the substance. These are just the properties to be required for a degree of organization between that of the continuous substance, liquid or crystalline solid and even the simplest living cell.’ Stewart (1961) some thirty years later also stated that ‘It is this property – the combination of flow and lability with a preferred and relatively stable molecular orientation – that makes the mesomorphic (i.e. liquid crystal) phase uniquely appropriate to the structure of protoplasm and living tissue.’

Several functions of the animal body can take place in cell or tissue cultures with almost unreduced efficiency and precision. Functions, where only one cell type is involved, often do so, but also some differentiation steps where interactions between two or more cell types are clearly needed can take place in tissue culture (Saxén et al. 1968).

Most immune responses require collaboration between two or more cell types (Claman, Chaperon & Triplett, 1966; Miller & Mitchell, 1968; Feldmann & Nossal 1972c). Some of them can be easily induced in vitro but others cannot. Even when antibody responses can be induced in vitro their intensity varies a great deal. With some antigens and under some circumstances a response in vitro can be nearly as strong as one in vivo. A crude comparison can be derived from responses in vitro and in vivo to the same antigen, conjugate of hapten NIP and pneumococcal polysaccharide type III (NIP-SIll, Nakamura, Ray & Mäkelä, 1973).

Most theories of determination or differentiation assume that embryonic cells differ from mature cells. Embryonic cells are thought to have metastable control mechanisms. These labile controls are believed to become progressively more stabilized as the cells differentiate. Zygote, blastula, neural plate, limb bud, somite, or ‘stem’ cells are conceived of as undifferentiated, totipotent, or multipotential cells. As such, these cells supposedly have available for activation a larger repertoire of phenotypic programmes than their progeny. A necessary corollary to this view is that the activation of one particular phenotypic programme out of the many available is a function of instructive exogenous inducing molecules.

Fluorescence has been used in biochemical studies for many years but only recently has the information content and the practical applicability of the fluorescence method been fully realized.

Following the early studies of Newton (1954) and Weber (11954) and after the initial utilization of fluorescent probes by Chance and coworkers (Azzi et al. 1969) and Tasaki et al. (1968), in the study of membranes, the use of fluorescence to provide structural information at microscopic or molecular levels in biological membranes has become widespread. widespread. The application of the fluorescence technique to biological systems has progressed parallel to the development of a theoretical basis for fluorescence data interpretation and the synthesis of a large number of fluorescent probes, organic molecules having fluorescence characteristics that are dependent on their environment.
