Parasitic ‘turbellarians’ are known from various animals such as echinoderms, crustaceans, annelids, bivalve and gastropod molluscs. So far, however, no ‘turbellarians’ have been reported from cephalopods. In this paper we report a parasitic ‘turbellarian’ from the giant Antarctic octopus, Megaleledone setebos. We dissected two specimens of M. setebos caught in the Ross Sea (Antarctica) and found numerous worms in their intestine and liver. The worms were spherical or oblong and had two morphologically different poles. The frontal pole bears a small conical protrusion containing large elongated pear-shaped frontal glands and large polygonal cells. The ducts of the frontal glands open terminally to form the frontal organ. The caudal pole has an opening shaped as a folded tube connected by the genital pore with a common genital atrium, which continues into a canal with a muscular sheath. The worms were identified as ‘turbellarians’ from the family Notenteridae (Fecampiida). This family contains only one species, Notentera ivanovi, reported from the gut of a polychaete at the White Sea. The worms that we found in the gastrointestinal tract of the octopuses were morphologically similar to N. ivanovi but differed from it in several important respects. Phylogenetic analysis based on 28S rDNA gene showed that the newly found worm clustered together with other fecampiids in a highly supported clade and was closely related to N. ivanovi. On the basis of these morphological and molecular data, we described a new species, Octopoxenus antarcticus gen. nov., sp. nov. (Fecampiida: Notenteridae), establishing a new genus to accommodate it and provided an updated diagnosis of the family Notenteridae. This is the first report of a parasitic ‘turbellarian’ from a cephalopod mollusc.

The earlier work of this Institute on phenothiazine in lambs (Peters et al, 1941; J. Leiper et al, 1941) using egg-counts, worm-counts, and lamb-weights as criteria of anthelmintic efficacy, has revealed conflicting results as between one criterion and another. Thus, the lowest dosage rate used (0·15 gm. per Kg.) led to a reduction in the egg-count, whilst even the highest rate (1 gm. per Kg.) led to no reduction in the worm-count: by an irony of chance the worm-counts were lowest in the untreated controls in both experiments. In the first experiment weights were not affected by the drug, and in the second the only significant effect was a set-back in those lambs given tablets. Numerous other workers have reported increased weights due to the drug, in long-term experiments; ours were too short in duration to show such increases, yet the danger of reinfestation precluded any extension of the time, the lambs being kept outside on infective pastures. The most obvious explanation of the egg- and worm-count results appears to be that the drug has a temporary inhibiting effect on egg-laying, which would presumably return to normal in a few weeks; but here again a prolonged experiment increases the liability to reinfestation.

These being the relevant circumstances, the present experiment was designed to take account of them in the following ways. In the first place the lambs were kept inside throughout the experiment, their stalls being cleaned out thoroughly twice a week to obviate reinfestation.

In the autumn of 19G6 the writer received two samples of nematodes, containing over a hundred specimens, which were found in the pancreatic ducts of the mustelid Arctonyx collaris. The two host animals arrived from Bangkok, one already dead; the other died in the Zoological Garden of Emmen a week after arrival.

The present work forms a continuation of a survey undertaken three years ago, with a view to elucidating the nemic parasites commonly found in fishes of Canadian waters. These species have proved to be largely undescribed, and old species have always offered some new points of interest. The greatest difficulty has been encountered with the literature, much of which was difficult of access. Only those papers which have a definite bearing on the forms studied, and convey some pertinent information, are included in the appended bibliography.

It has been known for many years that the stem eelworm, Anguillulina dipsaci, can be carried in or upon the seeds of a number of different plants. Kühn (1858) originally found the parasite in the diseased seed heads and seed of the teasel or fuller's thistle. Bos (1888) discovered the worms in the flowers of the onion, Allium Cepa, and found in one case 3 per cent. of the seeds were infected. Godfrey (1924) found infestations of the parasite in the flower heads of cat's-ear, Hypochoeris radicata, and dandelion, Taraxacum officinale, and discovered in both hosts that living worms occurred within the seed coats. He also obtained diseased seedlings by sowing infected seed of both hosts. Hodson (1926) reported the occurrence of the parasite in quiescent form beneath the pales of oats harvested from fields badly affected with “tulip root” and obtained small numbers of infected seedlings by sowing such seed. The number of such cases was, however, so small that he considered their occurrence might be accidental. Robertson (1928) investigating “tulip root” in oats, found the parasite in the panicles of affected plants giving rise to abortive flowers. In a number of oat grains from distorted panicles he found small numbers of the parasite between the glumes and pales and between the pales and the kernel and was able to show that after storage for a few months the worms could be revived by soaking the grains in water. He did not pursue his studies to the extent of obtaining diseased seedlings from such infected seed. Cobb (1929) found samples of seed of red clover and lucerne, as cleaned by commercial processes, yielded the parasite in a living condition when soaked in water.

In December, 1922, Sir John Russell called the writer's attention to some diseased potatoes which had been sent to the Rothamsted Experimental Station. Dr. W. B. Brierley, Head of the Mycology Laboratory, had examined them and had failed to find evidence of fungal attack though the diseased areas bore a superficial resemblance to the appearances presented by “blight” affected potatoes due to Phytophthora infestans. He had, however, found numerous eelworms in the diseased tissues and because of this the writer was asked to examine the material.

In February, 1924, Dr. Walton recorded Anoplocephala perfoliata, Ascaris equi, Oxyuris equi (curvula), and Cylicostomum tetracanthum from horses in North Wales; and in January 1925, Mr. Bissett, M.R.C.V.S., found, in a horse from Duffryn Mountains (South Wales), a large number of parasitic worms, including Anoplocephala mammillana, Cylicostomum tetracanthum, Strongylus edentatus, S. vulgaris, S. equinus, and one specimen of Oxyuris. These, it seems, are the only recent records concerning helminthic parasites of horses in Wales. It was therefore thought desirable to examine a few horses very carefully.

The antigenic properties of adult male and female of Dipetalonema viteae were studied by immunocytochemistry. Using antisera of the rodents Meriones unguiculatus and Mastomys natalensis infected with D. viteae, the binding of antibodies to sections of filariae embedded in Epon was assayed by the peroxidase-antiperoxidase (PAP) technique and by electron microscopy. The optimal staining intensity was obtained with an antiserum dilution of 1:5000. Control sera were obtained from sex and age matched uninfected animals.

Female D. viteae showed maximal antigen-antibody reactions within the uterus: in the inner uterus wall, in the “nutrient channels” between the maturing eggs and the differentiating microfilariae, on the eggshells, in the cuticula of microfilariae and in the spermatheca on the cell membrane of the mature spermatozoa. Male filariae showed binding of antibodies in the vesicula seminalis: in the nucleus and the nuclear membrane of primary spermatocytes and on maturing spermatids. Less pronounced antigen-antibody reactions in the cuticula, muscle and intestine were observed in both sexes.

The PAP-technique offers significant improvements in comparison with other techniques, e.g., immunofluorescence, used to detect antigens on filariae: the PAP-technique has an increased sensitivity with a concomitant reduction in nonspecific background and can be used for both light and electron microscopy; moreover, PAP-treated tissues can be stored indefinitely at room temperature.

1. Salting of pasture was explored as a means of controlling stomach worm infestation of grazing sheep.

2. Infestation with Haemonchus contortus was significantly decreased, but that with Ostertagia spp. and Trichostrongylus axei was not significantly affected.

The in vitro effects of ethanol and aqueous extracts of the medicinal plant Cardiospermum halicacabum on adult worms and microfilariae of Brugia pahangi were investigated. With or without the plant extracts in culture medium, the motility of adult worms, microfilariae and microfilarial release from female worms were monitored daily. After 7 days of culture, viability or tissue damage of adult worms was assessed using the MTT assay. At > 500 μg ml-1, the aqueous extract significantly reduced motility of adult females after 24 h of exposure and adult males after 3 days. The aqueous extract, at > 500 μg ml-1, also significantly reduced microfilarial release from female worms, starting on day 2. The reduction in the motility of adult worms and the pattern of microfilarial release from female worms were concentration and time dependent. The MTT assay results revealed that adult worms cultured in the presence of aqueous extracts at > 500 μg ml-1 were damaged. However, the aqueous extract did not affect the motility of microfilariae with the exception of those in higher concentration extracts. Higher concentrations of ethanol extracts (2 mg ml-1) inhibited both the motility of adult worms and the release of microfilariae from females. Little effect of ethanol extracts was detected by the MTT assay, as only slight damage was caused to worms exposed only to the highest concentration (2 mg ml-1). However, ethanol extract at 500 μg ml-1 rapidly reduced the motility of microfilariae on day 2. The present study revealed that an aqueous extract of C. halicacabum has mild but definite direct macrofilaricidal action on B. pahangi.

The influence of diet type and pre-treatment fasting on the kinetic disposition of albendazole was evaluated in Sahiwal heifers following oral and intra-ruminal administration of the drug. The anthelmintically active moiety albendazole sulphoxide appeared early and was eliminated early in cattle offered green fodder, with decreased maximum concentration (C max) and area under concentration–time curve (AUC) when the drug was administered both through oral and intra-ruminal routes. Further, the elimination half-life (t ½β) revealed significantly increased values for albendazole sulphoxide in cattle administered albendazole through the intra-ruminal route. An increased AUC and t ½β is reflective of increased bioavailability of albendazole in animals offered dry fodder. Increased values (P <  0.05) of C max, time to C max (T max), AUC and t ½β for albendazole sulphoxide occurred in cattle with a pre-treatment 24-h fast, resulting in its increased bioavailability. Extrapolation of data of the active metabolite albendazole sulphoxide levels in terms of drug–parasite contact revealed increased exposure of parasites to the drug in cattle administered albendazole through the intra-ruminal route and with 24-h pre-treatment fasting.

Papaya latex has been demonstrated to be an efficacious anthelmintic against murine, porcine, ovine and canine nematode parasites, and even those infecting poultry, and it has some efficacy against rodent cestodes. The active ingredients of papaya latex are known to be cysteine proteinases (CPs). The experiments described in this paper indicate that CPs in papaya latex, and also those in pineapples, are highly efficacious against the equine cestode Anoplocephala perfoliatain vitro, by causing a significant reduction in motility leading to death of the worms. The susceptibility of A. perfoliata to damage by CPs was considerably greater than that of the rodent cestodes Hymenolepis diminuta and H. microstoma. Our results are the first to report anthelmintic efficacy of CPs against an economically important equine helminth. Moreover, they provide further evidence that the spectrum of activity of CPs is not restricted to nematodes and support the idea that these plant-derived enzymes can be developed into useful broad-spectrum anthelmintics.

A new species of the genus Diomedenema, a spiruromorph nematode, collected from the lung of Spheniscus magellanicus (Sphenisciformes) found on the southern coast of the state of Rio Grande do Sul, Brazil, is described. The new species is differentiated from the only previously described species of the genus, D. diomedeae Johston & Mawson, 1952, by males possessing a set of caudal papillae with three pairs of precloacal, two pairs of adcloacal and one pair of postcloacal papillae; precloacal papillae with the papillae of the first two pairs being closer to each other than those of the third pair; a longer and pointed tail in males; and females with the vulva at mid-body. This is the first report of a nematode infecting the lung of a sphenisciforme host.

Acetylcholine is the major endogenous classical neurotransmitter in the central and peripheral nervous system of trematodes and mammals. This study investigates the effects of cholinergic drugs on muscle activity in the amphistome, Gastrothylax crumenifer. In the present investigation, acetylcholine (10− 7–10− 3 m) did not produce any marked effect, whereas carbachol (10− 7–10− 3 m) elicited a concentration-dependent decrease in amplitude, baseline tension and frequency of contractions as compared to the control. Nicotine (10− 7–10− 3 m) produced a significant decrease in the amplitude and frequency of spontaneous muscular activity in a concentration-dependent manner, as compared to control amplitude (0.5 ± 0.01 g) and frequency (58.5 ± 3.45 per 5 min). However, the baseline tension was also reduced significantly by 10− 3 m nicotine. Atropine (10− 7–10− 3 m) elicited a concentration-dependent increase in amplitude and baseline tension, whereas there was no significant effect on the frequency of the spontaneous contractions of rumen flukes. These observations indicate that G. crumenifer has an inhibitory cholinergic system and that the inhibitory activity of nicotine is more pronounced than that of carbachol or acetylcholine.

Extracts of Anisakis simplex third (L3) and fourth (L4) larval stages were assayed for protein content and activity and properties of α-amylase, glucoamylase and glycogen phosphorylase. Protein content in L4 was twice that in L3. SDS–PAGE applied to both larval stages revealed 22 protein fractions in each, including five stage-specific fractions in each larval stage. The L3 extracts contained three amylase isoenzymes: α1, α2 and α3; their molecular weights were 64, 29 and 21 kDa, respectively. Only one amylase isoenzyme (64 kDa) was found in the L4 extracts. Glycogen in L3 was found to be broken down mostly by hydrolysis because of low glycogen phosphorylase activity. The α-amylase activity in L4 was higher than that in L3 by half and the glycogen phosphorylase activity was ten times higher. In addition, the same enzymes isolated from L3 and L4 were found to differ in their properties. These differences could be manifestations of metabolic adaptations of A. simplex larvae to host switch from fish (L3) to mammals (L4), i.e. adaptations to a new habitat.

The metacestode stage of Echinococcus granulosus is of zoonotic importance. In general, the 14-3-3 protein is involved in multiplication and survival of eukaryotes. Therefore, this communication presents succinct information on relative expression of the 14-3-3 protein in six different morphotypes of cysts of E. granulosus. All isolated E. granulosus belonged to the common sheep strain (G1). Relative expression of the 14-3-3 protein was higher in fertile cysts when compared to sterile cysts. The predicted amino acid sequence of the 14-3-3 protein was closely clustered with zeta-type isoforms 1 and 2 of the 14-3-3 protein. In addition, the present study demonstrates the presence of the 14-3-3 protein which until now had not been detected in the germinal layer. Our findings indicate that the expression of this biomolecule in the germinal layer of sterile organisms may contribute to the development and survival of the parasite in the host. The uniform expression of actin II conclusively proves the survivability of the harvested organisms.

Polycephalic larvae of Taenia endothoracicus were found from naturally infected gerbils Meriones lybicus in a rural area of Arak, central Iran. A large cyst containing 19 protoscolices was located in the peritoneum, attached to the large omentum. The characteristics of the protoscolices and rostellar hooks confirmed the identification of these larvae as T. endothoracicus.

A 30 kDa antigen was characterized as a hydrophobic polypeptide containing 16 amino acids and evaluated as a potential candidate vaccine against infection by Schistosoma mansoni. CD1 albino mice immunized at 0, 14, and 21 days with 25 or 50 μg of the 30 kDa antigen per mouse with and without alum developed high levels of IgG antibodies (predominantly IgG2a and IgG2b isotypes). When immunized mice were infected with 200 S. mansoni cercariae, the highest protection levels (61% and 65% reduction in worm burden in two separate experiments) were obtained using the 50-μg antigen without alum adjuvant. The granuloma size decreased to 10%, a non-significant level in mice immunized using alum adjuvant. The results demonstrate the ability of the 30 kDa antigen with and without alum adjuvant to protect mice against S. mansoni infection.

Climate change is expected to affect parasitic nematodes and hence possibly parasite–host dynamics and may have far-reaching consequences for animal health, livestock production, and ecosystem functioning. However, there has been no recent overview of current knowledge to identify how studies could contribute to a better understanding of terrestrial parasitic nematodes under changing climates. Here we screened almost 1,400 papers to review 57 experimental studies on the effects of temperature and moisture on hatching, development, survival, and behaviour of the free-living stages of terrestrial parasitic nematodes with a direct life cycle in birds and terrestrial mammals. Two major knowledge gaps are apparent. First, research should study the temperature dependency curves for hatching, development, and survival under various moisture treatments to test the interactive effect of temperature and moisture. Second, we specifically advocate for more studies that investigate how temperature, and its interaction with moisture, affect both vertical and horizontal movement of parasitic nematodes to understand infection risks. Overall, we advocate for more field experiments that test environmental effects on life-history traits and behaviour of parasitic nematodes in their free-living stages under natural and realistic circumstances. We also encourage studies to expand the range of used hosts and parasitic nematodes because 66% of results described in the available studies use sheep and cattle as hosts and 32% involve just three nematode species. This new comprehension brings attention to understudied abiotic impacts on terrestrial parasitic nematodes and will have broader implications for livestock management, wildlife conservation, and ecosystem functioning in a rapidly warming climate.

Schistosoma haematobium and S. mansoni are two medically important schistosomes, commonly occurring sympatrically in Africa and so potentially able to infect the same human host. Experiments were designed to study the mating behaviour of these two species in mixed infections in hamsters. Analysis of the data obtained showed that both heterospecific and homospecific pairs readily form. No significant difference was seen between the two species in their ability in forming pairs, however, S. mansoni showed a greater homospecific mate preference. Analysis of the data using the Mantel-Haenszel test suggests that mating competition does occur between S. haematobium and S. mansoni, the former being the more dominant species. Both species appeared to be able to change mate, with S. haematobium showing a greater ability in taking S. mansoni females away from S. mansoni males when introduced into a pre-established S. mansoni infection highlighting the competitiveness of S. haematobium. The significance of the results is discussed in relation to the epidemiological consequences occurring in Senegal, and other areas where both species are sympatric.
