The current study investigated the coupling of groundwater and surface water nitrogen (N) dynamics over 3 years, and considered intensive agricultural land-management influences over this period where the risk of N loss to water was considered high. Groundwater N (as nitrate) was monitored monthly in different strata and zones in four hillslopes, two in each of two agricultural catchments of c. 10 km2, and stream water N flux was monitored sub-hourly in the catchment outlets. Field nutrient sources were connected to surface water via groundwater; the groundwater along hillslopes was seen to be influenced spatially and temporally by management, geology and weather as observed in the concentration variability of nitrate in groundwater. Based on spatio-temporal averages of nitrate-N concentration, groundwater status was considered good (at least below a maximum acceptable concentration (MAC) of 11·3 mg/l). However, zones coincident with land-use change (ploughing and reseeding, typical of a management event in intensive landscapes), showed high spatio-temporal variability in nitrate-N concentration, exceeding the MAC temporarily, before recovering. This spatio-temporal variability highlighted the need for insight into these differences when interpreting groundwater quality data from a limited number of basin-scale sampling points and occasions. In both catchments the 3-year mean nitrate-N concentration in stream water was similar to the spatio-temporal mean concentration in groundwater. The magnitude and variability of loads, however, were more related to changes in annual runoff rather than changes in annual groundwater nitrate-N status. In one wet year, nitrate-N loads exceeded 48 kg/ha from an Arable catchment and 45 kg/ha from a grassland catchment (close to double the loss in a dry year).

The current study was designed to determine the effect of roughage to concentrate ratio (R : C) on rumen pH, fermentation and bacterial population in dairy steers. Four rumen fistulated dairy steers (170±20 kg) were randomly assigned according to a 4×4 Latin square design, in which the steers were fed with four dietary treatments with different R : C ratios of 0·8 : 0·2, 0·6 : 0·4, 0·4 : 0·6 and 0·2 : 0·8, respectively. All animals were kept in individual pens and received feed according to the respective R : C ratios at 0·025 body weight (BW)/d; urea-treated rice straw (prepared using 3·5 kg urea+100 kg water sprayed onto 100 kg of rice straw) was used as a roughage source. The experiment was conducted for four periods of 21 days each. During the first 14 days, feed intake was measured and the animals were then moved to metabolism crates for total urine and faecal collection for 7 days. Total dry matter intake (DMI) was similar among treatments. Energy intake increased as the proportion of concentrate in the diet increased. Apparent digestibilities of dry matter (DM), organic matter (OM) and crude protein (CP) were improved, while neutral detergent fibre (NDF) and acid detergent fibre (ADF) were reduced when the levels of concentrate increased. A decreasing ratio of R : C reduced rumen pH linearly, from 6·4 to 5·9 at 0·2 : 0·8. High levels of concentrate impacted on volatile fatty acids (VFA) molar proportions and decreased acetate (C2) linearly, while propionate (C3) was increased, leading to decreased C2 : C3 ratio. Numbers of protozoa, fungi and proteolytic bacteria were not affected by R : C ratio. Cellulolytic bacteria decreased linearly while amylolytic bacteria increased linearly with 0·60 and 0·80 concentrates. Quantitative polymerase chain reaction (qPCR) based on 16S RNA revealed that Fibrobacter succinogenes numbers were increased when steers were fed with R : C ratio of 0·8 : 0·2. Conjugated linoleic acid (CLA)-producing bacteria, especially those of Butyrivibrio fibrisolvens, increased linearly with R : C ratios of 0·8 : 0·2 and 0·6 : 0·4, while Megasphaera elsdenii, a lactate-utilizing bacterium and reported producer of trans-10, cis-12 CLA increased linearly with R : C ratio of 0·8 : 0·2. In addition, microbial CP synthesis increased quadratically when steers were fed high levels of concentrate. However, the efficiency microbial N synthesis (EMNS) based on OM, truly digested in the rumen, was not affected by different R : C ratios. From the current study, it can be concluded that roughage to concentrate ratio of 0·4 : 0·6 had positive effects for the creation of healthy rumen (rumen pH and ecology), and improved energy intake and rumen fermentation, particularly propionic acid and microbial protein synthesis, in dairy steers fed urea-treated rice straw as a roughage source.

Deploying resistant cultivars can reduce the prohibitive cost associated with managing Sclerotinia sclerotiorum in rapeseed production worldwide. The present paper reports the results of an analysis of inheritance of leaf and stem resistance involving a single inter-specific cross between Brassica incana and Brassica oleracea var. alboglabra. Detached leaves and stems of parental lines, F1, F2 and the backcrosses were obtained from Southwest University, Chongqing Field Station, Chongqing China in the 2009/10 and 2010/11 field seasons and inoculated with S. sclerotiorum to determine resistance. Significant differences were detected across the two growing seasons between parents and some of the progeny for measures of both leaf and stem resistance. Continuous variation patterns among the segregating generations suggest the quantitative nature of resistance in both leaf and stem. Dominant and additive × additive epistatic interactions controlled the genetic effects for both traits. Broad- and narrow-sense heritability estimates were moderately high for leaf resistance, but were intermediate to high for stem resistance in the two field seasons. Low estimates of the minimum number of genes for leaf and stem resistance were recorded in the two field seasons. The results indicate that selection gains and the identification of quantitative trait loci can be maximized in marker-assisted-selection through differential selection (tissue-based selection) on a replicated plot basis.

Sainfoin is a temperate legume that contains condensed tannins (CT), i.e. polyphenols that are able to bind proteins and thus reduce protein degradation in the rumen. A reduction in protein degradation in the rumen can lead to a subsequent increase in amino acid flow to the small intestine. The effects of CT in the rumen and the intestine differ according to the amount and structure of CT and the nature of the protein molecular structure. The objective of the present study was to investigate the degradability in the rumen of three CT-containing sainfoin varieties and CT-free lucerne in relation to CT content and structure (mean degree of polymerization, proportion of prodelphinidins and cis-flavanol units) and protein structure (amide I and II bands, ratio of amide I-to-amide II, α-helix, β-sheet, ratio of α-helix-to-β-sheet). Protein molecular structures were identified using Fourier transform/infrared-attenuated total reflectance (FT/IR-ATR) spectroscopy. The in situ degradability of three sainfoin varieties (Ambra, Esparcette and Villahoz) was studied in 2008, during the first growth cycle at two harvest dates (P1 and P2, i.e. 5 May and 2 June, respectively) and at one date (P3) during the second growth cycle (2 June) and these were compared with a tannin-free legume, lucerne (Aubigny). Loss of dry matter (DMDeg) and nitrogen (NDeg) in polyester bags suspended in the rumen was measured using rumen-fistulated cows. The NDeg of lucerne compared with sainfoin was 0·80 v. 0·77 at P1, 0·78 v. 0·65 at P2 and 0·79 v. 0·70 at P3, respectively. NDeg was related to the rapidly disappearing fraction (‘a’) fraction (r=0·76), the rate of degradation (‘c’) (r=0·92), to the content (r=−0·81) and structure of CT. However, the relationship between NDeg and the slowly disappearing fraction (‘b’) was weak. There was a significant effect of date and species×date, for NDeg and ‘a’ fraction. The secondary protein structure varied with harvest date (species×date) and was correlated with the fraction ‘b’. Both tannin and protein structures influenced the NDeg degradation. CT content and structure were correlated to the ‘a’ fraction and to the ‘c’. Features of the protein molecular secondary structure were correlated to the ‘b’ fraction.

The effects of melamine on gas production (GP) kinetics, methane (CH4) production and fermentation of diets differing in forage content (low-forage (LF) diet: 200 g/kg and high-forage (HF) diet: 800 g/kg) by rumen micro-organisms in vitro were studied using batch cultures. Rumen contents were collected from three Simmental×Luxi crossbred beef cattle. Melamine was added to the incubation bottles to achieve final concentration of 0 (control), 2, 6, 18, 54, 162 and 484 mg/kg of each diet. Cumulative GP was continuously measured in an automated gas recording instrument during 72 h of incubation, while fermentation gas end-products were collected to determine molar proportions of carbon dioxide (CO2), CH4 and hydrogen gas (H2) in manually operated batch cultures. Differences in GP kinetics and fermentation gases were observed in response to the nature of the diets incubated. Although melamine addition did not affect GP kinetics and fermentation gas pattern compared to the control, the increase of melamine addition stimulated the yield of CH4 by decreasing CO2, especially during the fermentation of the HF diet. The concentrations of ammonia nitrogen (N), amino acid N and microbial N in culture fluids were greater in the fermentation of LF- than HF diets, and these concentrations were increased by the increase of melamine addition after 72-h fermentation. The concentrations of total volatile fatty acids (VFA) were greater in HF than LF diets. The addition of melamine decreased total VFA concentrations and this response was greater in HF than LF diet fermentations. Melamine addition did not affect molar proportions of acetate, butyrate, propionate and valerate compared with the control; however, branched-chain VFA production, which was lower in the HF than the LF diet, was increased by the melamine addition, especially in the HF diet fermentation. The ratio of non-glucogenic to glucogenic acids was lower in the HF than the LF diet, but it was not affected by melamine addition. In brief, the greater reduction in the rate and extent of rumen fermentation found for the HF diet in comparison with the LF diet suggested that rumen fermentation rate and extent in vitro depended mainly on the nature of the incubated substrate, and that they could be further inhibited by the increase of melamine addition.

The objective of the current study is to determine the beef production traits of purebred Nordic Red (NR) and NR×beef breed crossbred bulls. The data collected from slaughterhouses included observations of 164 812 purebred NR, 2329 NR×Aberdeen Angus, 1466 NR×Blonde d'Aquitaine, 1044 NR×Charolais, 782 NR×Hereford, 5293 NR×Limousin and 1270 NR×Simmental bulls. For estimating valuable cuttings, a separate dataset including a total of 16 827 observations was collected. Crossbreeding NR cows with late-maturing breeds (Blonde d'Aquitaine, Charolais, Limousin or Simmental) had favourable effects on carcass gain, conformation and proportion of high-value joints of the progeny when compared with purebred NR bulls. No advantages in proportion of valuable cuttings seemed to be obtained by crossbreeding with Aberdeen Angus or Hereford breeds, while the improvements in gain and conformation were intermediate compared with the late-maturing crossbreds. A feeding experiment comprised 36 bulls: nine purebred NR, nine NR×Aberdeen Angus (NR×Ab), nine NR×Limousin (NR×Li) and nine NR×Blonde d'Aquitaine (NR×Ba). The animals were offered total mixed ration (TMR) ad libitum. The dry matter (DM) of the TMR consisted of grass silage (500 g/kg DM), rolled barley (425 g/kg DM) and rapeseed meal (75 g/kg DM). There were no differences in DM, energy or nutrient intakes between NR and crossbred bulls during the feeding experiment. Instead, the carcass gain of the NR×Ba bulls was 13% higher than that of the pure NR bulls. In addition, the carcass gain of the NR×Li bulls tended to be 8% higher than that of the NR bulls. The feed (kg DM/kg carcass gain) and energy (MJ/kg carcass gain) conversion rates of the NR×Ba bulls tended to be better compared with purebred NR bulls. There were no differences in feed or energy conversion between NR, NR×Ab and NR×Li bulls. In conclusion, crossbreeding, especially with late-maturing bulls, largely improved carcass production compared with purebred NR bulls. The feeding experiment indicates that there is no difference in DM intake between pure NR and crossbred bulls when animals are fed with high-energy rations. Therefore, differences in growth and carcass traits describe well the economic superiority of crossbreds compared with pure dairy bulls from beef producers’ point of view.

Many studies have shown the efficacy of the nitrification inhibitor dicyandiamide (DCD) in reducing nitrous oxide (N2O) emissions and nitrate (NO3−) leaching. However, there is no information on the effect of DCD on gross soil N transformations under field conditions, which is key information if it is to be used as a mitigation strategy to reduce N losses. The current field study was conducted to determine the effect of DCD on ten gross nitrogen (N) transformations in soil following cattle slurry (CS) application to grassland in Northern Ireland on three occasions (June 2010, October 2010 and March 2011).

Ammonium (NH4+) oxidation (ONH4) was the dominant process in total NO3− production (ONH4+ONrec (oxidation of recalcitrant organic N to NO3−)) following CS application, accounting for 0·894–0·949. Dicyandiamide inhibited total NO3− production from CS by 0·781, 0·696 and 0·807 in June 2010, October 2010 and March 2011, respectively. The lower inhibition level in October 2010 was thought to be due to the higher rainfall and soil moisture content in that month compared to the other application times. As DCD strongly inhibited NH4+ oxidation following CS application, it also decreased the rate of total NO3− consumption, since less NO3− was formed. The rates of mineralization from recalcitrant organic-N (MNrec) were higher than from labile organic-N (MNlab) on all occasions. The DCD significantly increased total mineralization (MNrec+MNlab) following CS application in June 2010 and March 2011, but had no significant effect in October 2010. In contrast, the rate of immobilization of labile organic-N (INH4_Nlab) was higher than from recalcitrant organic-N (INH4_Nrec) on all occasions, accounting for 0·878–0·976 of total NH4+ immobilization from CS. The DCD significantly increased total immobilization (INH4_Nrec+INH4_Nlab) when CS was applied in June 2010, but had no significant effect at other times of the year.

Dicyandiamide was shown to be a highly effective inhibitor of ammonium oxidation at this grassland site. Although there was evidence that it increased both NH4+ mineralization and immobilization following CS application, its effect on these processes was inconsistent. Further work is required to understand the reason for these inconsistent effects: future improvements in 15N tracer models may help.

There is increasing concern about balancing agronomic and environmental gains from nitrogen (N) usage on dairy farms. Data from a 3-year (2009–2011) survey were used to assess farm-gate N balances and N use efficiency (NUE) on 21 intensive grass-based dairy farms operating under the good agricultural practice (GAP) regulations in Ireland. Mean stocking rate (SR) was 2·06 livestock units (LU)/ha, mean N surplus was 175 kg/ha, or 0·28 kg N/kg milk solids (MS), and mean NUE was 0·23. Nitrogen inputs were dominated by inorganic fertilizer (186 kg N/ha) and concentrates (26·6 kg N/ha), whereas outputs were dominated by milk (40·2 kg N/ha) and livestock (12·8 kg N/ha). Comparison with similar studies carried out before the introduction of the GAP regulations in 2006 would suggest that N surplus, both per ha and per kg MS, have significantly decreased (by 40 and 32%, respectively) and NUE increased (by 27%), mostly due to decreased inorganic fertilizer N input and improvements in N management, with a notable shift towards spring application of organic manures, indicating improved awareness of the fertilizer value of organic manures and good compliance with the GAP regulations regarding fertilizer application timing. These results would suggest a positive impact of the GAP regulations on dairy farm N surplus and NUE, indicating an improvement in both environmental and economic sustainability of dairy production through improved resource-use efficiencies. Such improvements will be necessary to achieve national targets of improved water quality and increased efficiency/sustainability of the dairy industry. The weak impact of SR on N surplus found in the present study would suggest that, with good management, increased SR and milk output per ha may be achievable, while decreasing N surplus per ha. Mean N surplus was lower than the overall mean surplus (224 kg N/ha) from six studies of northern and continental European dairy farms, while mean NUE was similar, largely due to the low input/output system that is more typical in Ireland, with seasonal milk production (compact spring calving), low use of concentrates, imported feed and forages, high use of grazed grass and lower milk yields per ha.

The grassland steppe of Inner Mongolia is traditionally used for sheep grazing. However, overgrazing reduced vegetation cover in winter, thereby increasing soil erosion and consequently, degradation of the steppe vegetation. Grazing intensity (GI) is still the most important factor in pasture management. Hence, the aim of the current study was to evaluate the effect of GI on grassland and sheep performance. A grazing experiment was conducted from July until September in 2005, 2006 and 2007 in which six different GI ranging from very light (GI 1), light (GI 2), light-moderate (GI 3), moderate (GI 4) and heavy (GI 5) to very heavy (GI 6) were tested. Each GI treatment comprised two adjacent plots that were alternately used for grazing or hay-making each year. Variables measured included herbage mass (HM) and chemical composition, digestibility of ingested organic matter (dOM), organic matter intake (OMI) and live weight gain (LWG) of sheep. The HM decreased significantly with increasing GI from 1·01 t (GI 1) to 0·45 t dry matter (DM)/ha (GI 6). There were only minor effects of GI on chemical composition and digestibility of standing herbage. Moreover, dOM, OMI and hence, digestible OMI did not differ between GI. Across all study years, LWG of sheep was not influenced by GI so that LWG per hectare increased with increasing GI, reaching a maximum of 730 g/d at GI 6 compared with 181 g/d at GI 1. However, a strong decrease in LWG per sheep with increasing stocking rate was found in 2005 when annual rainfall was less than half of the long-term average, resulting in a similar LWG per hectare across the range of tested stocking rates. The results therefore show that intensive grazing does not reduce growth of individual animals in most years, but increases LWG per unit of land area and thus, income of farmers. The alternating use of pastures for grazing or hay-making might have mitigated the negative effects of heavy grazing on herbage and animal performance. Nevertheless, high GI may negatively affect grassland productivity in the long term and the lack of HM on offer on heavy grazed pastures in dry years will require supplement feeding at the end of the vegetation period or the untimely sale of animals.

In view of environmental concerns with regard to phosphorus (P) pollution and the expected global P scarcity, there is increasing interest in improving P utilization in dairy cattle. In high-producing dairy cows, P requirements for milk production comprise a significant fraction of total dietary P requirements. Although variation in P content of milk can affect the efficiency of P utilization for milk production (i.e. the fraction of ingested P that is incorporated in milk), this variation is poorly understood. It was hypothesized that the P content of milk is related to both milk protein and milk lactose content, but not necessarily to milk fat content. Three existing experiments comprising individual animal data on milk yield and fat, protein, lactose and P content of milk (in total 278 observations from 121 cows) were analysed to evaluate this hypothesis using a mixed model analysis. The models including the effects of both protein and lactose content of milk yielded better prediction of milk P content in terms of root-mean-square prediction error (RMSPE) and concordance correlation coefficient (CCC) statistics than models with only protein included as prediction variable; however, estimates of effect sizes varied between studies. The inclusion of milk fat content in equations already including protein and lactose did not further improve prediction of milk P content. Equations developed to describe the relationship between milk protein and lactose contents (g/kg) and milk P content (g/kg) were: (Expt 1) P in milk=−0·44(±0·179)+0·0253(±0·00300)×milk protein+0·0133(±0·00382)×milk lactose (RMSPE: 5·2%; CCC: 0·71); (Expt 2) P in milk=−0·26 (±0·347)+0·0174(±0·00328)×milk protein+0·0143 (±0·00611)×milk lactose (RMSPE: 6·3%; CCC: 0·40); and (Expt 3) P in milk=−0·36(±0·255)+0·0131(±0·00230)×milk protein+0·0193(±0·00490)×milk lactose (RMSPE: 6·5%; CCC: 0·55). Analysis of the three experiments combined, treating study as a random effect, resulted in the following equation to describe the same relationship as in the individual study equations: P in milk=−0·64(±0·168)+0·0223(±0·00236)×milk protein+0·0191(±0·00316)×milk lactose (RMSPE: 6·2%; CCC: 0·61). Although significant relationships between milk protein, milk lactose and milk P were found, a considerable portion of the observed variation remained unexplained, implying that factors other than milk composition may affect the P content of milk. The equations developed may be used to replace current fixed milk P contents assumed in P requirement systems for cattle.

Effects of the correction of microbial contamination (using 15N techniques) and of considering the comminution rate (kc) of particles in the rumen on effective estimates of the ruminally undegraded (RU) fraction and its intestinal effective digestibility (IED) were examined in a sample of dehydrated sugar beet pulp (DBP) generating composite samples (from rumen-incubated residues) representative of the chemical composition of RU. Tested fractions were dry matter (DM), organic matter (OM, tested only for RU), crude protein (CP) and amino acids (AA). The study was performed on three rumen and duodenum cannulated wethers fed with a 2 : 1 (fresh weight basis) chopped oat hay-to-concentrate diet supplied at 40 g DM/kg BW0·75 in six equal meals per day. The DBP showed sigmoid degradation kinetics: the fractional degradation rate increased by 5·8 times as time (h) increased from 0 to ∞. The kc rate (measured in the diet concentrate) represented 5·74% of the total rumen retention time of particles. As a result, the RU of DM was over-evaluated by 6·53% when kc was not considered. Microbial contamination of RU was high as in DM as in CP. Therefore, the overestimation of RU of DM was increased to 12·2% when this contamination was not corrected. The lack of this correction also led to large over-evaluations of RU and IED of CP and AA. As a result, the overestimation of the intestinal digested fraction was 40·9% for CP and 45·0% for total analysed AA. This overestimation varied largely among AA (from 18·9 to 88·7%). Corrected proportions of RU and IED were also variable among AA. Hypotheses on the causes of this variability are given. Resultant changes in the AA profile of the intestinal digested protein had some negative impact on the supply of essential AA and cysteine without affecting lysine. This problem is limited because the microbial protein synthesized from DBP fermentation in the rumen is largely predominant in the AA supply to the host.

Nitrification inhibitors are used in agriculture for the purpose of decreasing nitrogen (N) losses, by limiting the microbially mediated oxidation of ammonium (NH4+) to nitrate (NO3−). Successful inhibition of nitrification has been shown in numerous studies, but the extent to which inhibitors affect other N transformations in soil is largely unknown. In the present study, cattle slurry was applied to microcosms of three different grassland soils, with or without the nitrification inhibitor dicyandiamide (DCD). A solution containing NH4+ and NO3−, labelled with 15N either on the NH4+ or the NO3− part, was mixed with the slurry before application. Gross N transformation rates were estimated using a 15N tracing model. In all three soils, DCD significantly inhibited gross autotrophic nitrification, by 79–90%. Gross mineralization of recalcitrant organic N increased significantly with DCD addition in two soils, whereas gross heterotrophic nitrification from the same pool decreased with DCD addition in two soils. Fungal to bacterial ratios were not significantly affected by DCD addition. Total gross mineralization and immobilization increased significantly across the three soils when DCD was used, which suggests that DCD can cause non-target effects on soil N mineralization–immobilization turnover.
