Habitat selection of Culicoides spp. (Diptera: Ceratopogonidae) is influenced by the physicochemical factors such as temperature, pH, salinity, moisture, conductivity, organic and inorganic compounds of substrates. These factors determine the life history traits of the vectors. We studied the influence of substrate salinity (0–40 parts per thousand, ppt) and pH (pH 1–13) on oviposition, egg hatching, larval survivability, and adult emergence of Culicoides peregrinus Kieffer under laboratory conditions. Most eggs (80.74%) were laid in 0 ppt and 95% in pH 7 but lowered with increased salinity and pH levels. It was observed that the females did not lay eggs in 30 ppt to 40 ppt salinity; pH 1 and pH 13 but interestingly up to 95% of the eggs were retained within the abdomen. Little effect of salinity and pH on egg hatching was observed up to 5 ppt and 10 ppt except at the extreme values of 40 ppt and pH 1, pH 13. Pupation did not occur in rearing plates with high salinities, 30 ppt and 40 ppt, although the few eggs hatched when exposed to such salinity. In low salinity (0 to 2 ppt), occurrence of adult emergence was more and then decreased with increasing salinity. Maximum emergence was seen when the rearing media was alkaline. This study deals with the suitability of breeding substrate of C. peregrinus when exposed to salinity and pH ranges. Our study suggests the ambient salinity and pH ranges to be maintained during laboratory rearing of this vector species.

Culicoides biting midges (Diptera: Ceratopogonidae) are the main vectors of livestock diseases such as bluetongue (BT) which mainly affect sheep and cattle. In Spain, bluetongue virus (BTV) is transmitted by several Culicoides taxa, including Culicoides imicola, Obsoletus complex, Culicoides newsteadi and Culicoides pulicaris that vary in seasonality and distribution, affecting the distribution and dynamics of BT outbreaks. Path analysis is useful for separating direct and indirect, biotic and abiotic determinants of species' population performance and is ideal for understanding the sensitivity of adult Culicoides dynamics to multiple environmental drivers. Start, end of season and length of overwintering of adult Culicoides were analysed across 329 sites in Spain sampled from 2005 to 2010 during the National Entomosurveillance Program for BTV with path analysis, to determine the direct and indirect effects of land use, climate and host factor variables. Culicoides taxa had species-specific responses to environmental variables. While the seasonality of adult C. imicola was strongly affected by topography, temperature, cover of agro-forestry and sclerophyllous vegetation, rainfall, livestock density, photoperiod in autumn and the abundance of Culicoides females, Obsoletus complex species seasonality was affected by land-use variables such as cover of natural grassland and broad-leaved forest. Culicoides female abundance was the most explanatory variable for the seasonality of C. newsteadi, while C. pulicaris showed that temperature during winter and the photoperiod in November had a strong effect on the start of the season and the length of overwinter period of this species. These results indicate that the seasonal vector-free period (SVFP) in Spain will vary between competent vector taxa and geographic locations, dependent on the different responses of each taxa to environmental conditions.

Vector-borne diseases are exceptionally sensitive to climate change. Predicting vector occurrence in specific regions is a challenge that disease control programs must meet in order to plan and execute control interventions and climate change adaptation measures. Recently, an increasing number of scientific articles have applied ecological niche modelling (ENM) to study medically important insects and ticks. With a myriad of available methods, it is challenging to interpret their results. Here we review the future projections of disease vectors produced by ENM, and assess their trends and limitations. Tropical regions are currently occupied by many vector species; but future projections indicate poleward expansions of suitable climates for their occurrence and, therefore, entomological surveillance must be continuously done in areas projected to become suitable. The most commonly applied methods were the maximum entropy algorithm, generalized linear models, the genetic algorithm for rule set prediction, and discriminant analysis. Lack of consideration of the full-known current distribution of the target species on models with future projections has led to questionable predictions. We conclude that there is no ideal ‘gold standard’ method to model vector distributions; researchers are encouraged to test different methods for the same data. Such practice is becoming common in the field of ENM, but still lags behind in studies of disease vectors.

We examined the effects of Haemoproteus infection on the survival and pathology caused in the biting midges. Forty-six females of Culicoides impunctatus were exposed experimentally by allowing them to feed on a naturally infected red-backed shrike infected with Haemoproteus lanii (lineage hRB1, gametocytaemia 5·2%). Seventeen females were fed on an uninfected bird (controls). Dead insects were collected, counted and used for dissection, histological examination and polymerase chain reaction-based testing. Parasites were present in all experimentally infected biting midges, but absent from control insects. Survivorship differed significantly between the control and infected groups. Twelve hours post-exposure (PE), 45 (98%) experimentally infected midges were dead, but all control midges remained alive, and many of them survived until 7 day PE. The migrating ookinetes of H. lanii overfilled midgut, markedly damaged the midgut wall, entered the haemocoel and overfilled the abdomen and thorax of exposed biting midges. Massive infection by migrating ookinetes led to damage of abdomen and thorax of biting midges. The parasites often present in large clumps in the haemocoel in abdomen and thorax, leading to the interruption of the haemolymph circulation. These are the main reasons for rapid death of biting midges after feeding on high-intensity infections of Haemoproteus parasites.

Culicoides vectors can transmit a diverse array of parasites and are globally distributed. We studied feeding preferences and seasonal variation of Culicoides (Diptera: Ceratopogonidae) vectors in an urban forest of Germany to determine whether humans living nearby are readily exposed to vector-borne parasites from wild animals. We used a fragment of the mtDNA COI gene to identify hosts from blood meals. We amplified a fragment of the mtDNA cyt b to detect haemosporidian infections in Culicoides abdomens and thoraxes. We detected a total of 22 Culicoides species. Fifty-eight blood meals (84%) were from humans, 10 from birds, and one from livestock. We found Culicoides kibunensis (considered ornithophilic) with 29 human blood meals. Host generalist Culicoides festivipennis and Culicoides obsoletus had 14 human blood meals. Culicoides clastrieri and Culicoides semimaculatus fed on birds; previously humans were their only known host. Six thoraxes and three abdomens were infected with either Haemoproteus pallidulus or Haemoproteus parabelopolskyi. There were changes in Culicoides community structure across months. Culicoides pictipennis was the dominant species during spring, C. kibunensis and C. clastrieri were dominant during summer, and C. obsoletus was dominant by early autumn. All dominant species were generalists feeding on birds, livestock and humans. Our results indicate that humans can serve as a blood source for dominant Culicoides species instead of the normal wild animal hosts in urban areas.

Biting midges of the genus Culicoides (Diptera: Ceratopogonidae) are vectors of a number of viral diseases worldwide. Following the unforeseen outbreak of bluetongue in northern Europe (2006–2009) there was a need to clarify on-farm breeding substrates utilized by temperate Culicoides spp. Six substrates (cow dung, cow slurry, horse dung, sheep dung, maize silage and soil) were investigated for Culicoides spp. emergence over a 31-week period. Overall, most Obsoletus group Culicoides emerged from the cow dung and the most Pulicaris group Culicoides emerged from the sheep dung. Furthermore, Culicoides of the Obsoletus group were found to be abundant in cow slurry and sheep dung. Temperature played a significant role in the emergence times of adult Culicoides. The Obsoletus group appear to have undergone 3 generations during the experimental period. The sex ratio of emergent Obsoletus group Culicoides was affected by substrate type, with a greater proportion of males emerging from cow dung and slurry compared with the other substrates.

Matrix-assisted laser desorption/ionization time of flight mass spectrometry (MALDI-TOF MS) was evaluated for the rapid identification of ceratopogonid larvae. Optimal sample preparation as evaluated with laboratory-reared biting midges Culicoides nubeculosus was the homogenization of gut-less larvae in 10% formic acid, and analysis of 0·2 mg/ml crude protein homogenate mixed with SA matrix at a ratio of 1:1·5. Using 5 larvae each of 4 ceratopogonid species (C. nubeculosus, C. obsoletus, C. decor, and Dasyhelea sp.) and of 2 culicid species (Aedes aegypti, Ae. japonicus), biomarker mass sets between 27 and 33 masses were determined. In a validation study, 67 larvae belonging to the target species were correctly identified by automated database-based identification (91%) or manual full comparison (9%). Four specimens of non-target species did not yield identification. As anticipated for holometabolous insects, the biomarker mass sets of adults cannot be used for the identification of larvae, and vice versa, because they share only very few similar masses as shown for C. nubeculosus, C. obsoletus, and Ae. japonicus. Thus, protein profiling by MALDI-TOF as a quick, inexpensive and accurate alternative tool is applicable to identify insect larvae of vector species collected in the field.

Blood meal identification can provide information about the natural host-feeding patterns or preferences of Culicoides species. Such information could indirectly provide data indicating which reservoirs are significant in associated vector-borne diseases.

We positively identified the host species through DNA sequencing of the cytochrome b gene in 144 of the 170 (84.7%) blood meal specimens tested. In the remaining samples, identification of the blood-meal source was unsuccessful, possibly due to the post-ingestion time prior to sampling or the availability of the species-specific cytochrome b gene sequences in the database. The majority of identified blood meals were derived from mammalian blood (95.8%), and only six contained chicken blood. We identified five species as mammalian hosts for Culicoides spp.: sheep (87.7%), human (6.5%), cattle (3.7%) and Savi's Pine Vole (Micrototus savii) (2.1%). The results suggested that large mammals, specifically ruminants, were most frequently fed upon by biting midges (Culicoides spp.), but evidence of opportunistic feeding behaviour was also found. Host feeding behaviour of Culicoides species may also be influenced by the relative abundance of a particular host species in the area being studied. In this sense, Savi's Pine Vole, a wild species, was found to be a locally relevant host and a putative reservoir for viruses transmitted by species of biting midges belonging to the Culicoides genus.

Finally, feeding on multiple potential host species was observed. One midge acquired blood meals from human and chicken hosts, while four other midges fed on two different sheep.

Culicoides biting midges are of great importance as vectors of pathogens and elicitors of allergy. As an alternative for the identification of these tiny insects, matrix-assisted laser desorption/ionization time of flight mass spectrometry (MALDI-TOF MS) was evaluated. Protein mass fingerprints were determined for 4–5 field-caught reference (genetically confirmed) individuals of 12 Culicoides species from Switzerland, C. imicola from France, laboratory-reared C. nubeculosus and a non-biting midge. Reproducibility and accuracy of the database was tested in a validation study by analysing 108 mostly field-caught target Culicoides midges and 3 specimens from a non-target species. A reference database of biomarker mass sets containing between 24 and 38 masses for the different species could be established. Automated database-based identification was achieved for 101 of the 108 specimens. The remaining 7 midges required manual full comparison with the reference spectra yielding correct identification for 6 specimens and an ambiguous result for the seventh individual. Specimens of the non-target species did not yield identification. Protein profiling by MALDI-TOF, which is compatible with morphological and genetic identification of specimens, can be used as an alternative, quick and inexpensive tool to accurately identify Culicoides biting midges collected in the field.

The transmission of bovine Onchocerca spp. in Britain was investigated by studying the Ceratopogonidae and Simuliidae which attacked bait cattle between April and October at two sites in North Wales, where either Onchocerca lienalis and O. gutturosa (Cynwyd), or O. lienalis alone (Pentrefoelas) occurred. Based on the relative abundance of each species, their seasonal variation in abundance and preferential feeding sites on cattle, the presence of Onchocerca sp. infective larvae and the development of patent infections in bait cattle, vectors were identified as Simulium ornatum s.1. at Pentrefoelas and S. reptans at Cynwyd (0·5% and 1·5% of parous flies infective, respectively). There was no significant difference between the lengths of the infective larvae from vectors at either site (Pentrefoelas: mean = 543·5 μm, SD = 29·45; Cynwyd: mean = 550·86 μm, SD = 35·26; p > 0·1), which were consistent with descriptions of O. lienalis. The identity of a vector for O. gutturosa was not determined and the role of certain candidates is discussed. In this respect, the absence of Culicoides nubeculosus was notable. Transmission of Onchocerca sp. was limited to a short period of about 8 weeks, by a combination of vector abundance and parasite uptake from dermal skin layers, and a mean daily temperature requirement of 15°C or more for complete parasite development in the vector. The effect of this temperature restriction in relation to the geographical distribution of the two species is discussed.

A total of 2083 Culicoides spp. were collected in an ultraviolet light trap hung from the rafters of a laying chicken house at a location in Sebele, giving a mean daily catch of 208.3 biting midges. C. imicola was the most abundant and constituted 57.5% of the total catch. C. leucosticus and C. engubandei constituted 20.5% and 14.770, respectively, of the catch. A comprehensive questionnaire conducted at three sites in the village indicated that many chickens in the homesteads in this location had died as a result of fowl pox. A high incidence of Haemoproteus columbae gametocytes was also found in the cytoplasm of erythrocytes of domestic pigeons (Colurnbiae livia domestica).

Species of Culicoides feeding on horses were investigated in Ibadan and Ilorin in the southwestern part of Nigeria using light, sticky and emergence traps. A totalof 2949 Culicoides consisting of 22 species were caught over a period of 22 months. Culicoides quinquelineatus was the most numerous, followed by C. imicola. The greatest proportion of Culicoides was caught with the sticky traps followed by the emergence traps during the period of moderate to heavy rains. Males were scarceand the proportion of fed females was generally low.

A significantly greater number of Culicoides was caught during the rains than during the dry season. The results are discussed within the context of previous investigations on Culicoides in Nigeria. The possibility of some species, especially C. imicola and C. quinquelineatus being vectors of African horse sickness is also discussed.

In a bluetongue (BT) disease project, light trappings of Culicoides were conducted in 19 localities and 39,000 biting midges were examined. BT virus type 16 was isolated in 1968 at Devora from a pool consisting mainly of Culicoides schultzei gp and C. imicola; BT virus type 4 was isolated in 1969 at Yesodot from a pool of 183 C. imicola; BT virus type 2 was isolated in 1979 at Talmé El'azar from pools consisting of 2462 C. imicola; and BT virus type 6 was isolated 3 times in 1980 at Bet Dagan from pools of 204, 258 and from 143 C. imicola, respectively. In the Akabane project (1976 and 1977) 4632 Culicoides and 1867 mosquitoes from four localities were trapped and examined. An arbovirus which was not Akabane, BT or turkey meningo-encephalitis (TME) was isolated from a pool of 44 engorged Culex pipiens complex trapped in Gan Shemuel. In the Rift Valley fever project, which was conducted in 1978 and 1979 due to the presence of an epidemic in Egypt, 351 mosquitoes of four species and 7098 Culicoides of 14 species were tested and none were found to harbour an arbovirus. Within the TME project more than 12,000 Culicoides and 9000 mosquitoes from 14 localities were tested. Positive isolations of TME were made from an unsorted pool of mosquitoes from Gesher haziw, an unsorted pool of 13 Culicoides from Nahalal, a pool of 223 unidentified Culicoides from Zetan and a pool of 12 engorged Cx pipiens complex from the same locality. Other pathogens which were not identified to species level were also isolated.
