Connections of the rabbit suprageniculate pretectal nucleus (SP) with the superior colliculus were explored by means of retrograde transport of horseradish peroxidase or Fluorogold. Large injections centered in the superficial and intermediate tectal layers resulted in bilateral retrograde transport to the medium-size multipolar neurons of the suprageniculate pretectal nucleus. Horseradish peroxidase was also transported anterogradely into the ipsilateral and contralateral neuropiles of the suprageniculate pretectal nucleus. The labeled cells in SP were dispersed throughout the nucleus, including its dorsal, wedge-shaped, internal portion. Labeling was mainly ipsilateral, and less abundant on the contralateral side.

To determine whether central communication of retinal signals is necessary for the development of an experimentally induced myopia, tree shrews were exposed to monocular deprivation (MD) while the action potentials of retinal cells in the deprived eye were blocked with intravitreally injected tetrodotoxin (TTX-MD animals). TTX injections (0.6 μ 3 μL) and MD began about 15 days after eye opening, at the start of the susceptible period for the development of lid-suture myopia. Six injections were given, one every second day to produce 12 days of MD and TTX-blockade. Control TTX animals (TTX-open) received TTX in one eye, but not MD, on the same injection schedule and were always found to be behaviorally unresponsive to visual stimuli through the injected eye indicating that TTX blocked central communication of action potentials. Other control animals received intravitreally injected saline in either an open eye (saline-open), or an MD eye (saline-MD). A sham-injected group (sham-inj-MD) received MD and all anesthetic and surgical manipulations except for penetration of the sclera. In all groups, one eye in each animal was an untreated control.

Two effects were found. All MD groups, including the TTX-MD animals, developed a significant vitreous chamber elongation in the deprived eye, indicating that an experimental myopia developed despite ganglion cell blockade. Thus, retinal mechanisms in tree shrew can detect the presence of a degraded visual image and produce an experimental myopia that does not depend on the receipt of visual messages by central neural structures. In addition, eyes in which the sclera was punctured had smaller vitreous chamber depths than comparable uninjected eyes, indicating that puncturing the sclera reduced the normal elongation. These data suggest that forces within the eye normally contribute to its expansion and may be resisted by the choroid and/or the sclera.

Extrastriate areas TEO and V4 have been associated with form and color vision. Area V4 has also been suggested to participate in processes concerned with attention, stimulus salience, and perceptual learning. In a continuing effort to elucidate the connectional interactions and microcircuitry of these areas, we describe in this report the pattern of feedback connections from TEO and V4. Connections were demonstrated by injections of the high-resolution anterograde tracers PHA-L or biocytin and further analyzed by reconstruction of 25 individual axons through serial sections. This analysis yielded several new results: (1) Both areas TEO and V4 have widespread feedback connections (defined by their preferential termination in layer 1 and avoidance of layer 4). From TEO, there are dense projections to area V4 and moderate ones to V2 and V1. From V4, there are dense projections to V2 and moderate ones to V3 and V1. (2) Terminal fields span large territories in area V1, up to 6.0 mm in the case of axons originating from TEO; up to 5.0 mm in the case of axons originating from V4. In V2, fields tend to be smaller, between 3.0–5.0 mm. (3) Many axons from TEO and some from V4 have terminations in both areas V1 and V2. (4) Because individual terminal clusters and segments are often larger than cytochrome oxidase compartments, especially in V1, we suggest they may not be correlated with this compartmental organization. These results are consistent with the hypothesis that feedback connections may contribute to processes other than perceptual discrimination.

A recent model for two-dimensional motion processing in MT has demonstrated that perceived direction can be accurately predicted by combining Fourier and non-Fourier component motion signals using a vector sum computation. The vector sum direction is computed by a neural network that weights Fourier and non-Fourier components by the cosine of the component direction relative to that of each pattern unit, after which competitive inhibition extracts the signals of the most active units. It is shown here that a minor modification of the connectivity in this network suffices to predict transitions from motion coherence to transparency under a wide range of circumstances. It is only necessary that the cosine weighting function and competitive inhibition be limited to directions within ± 120 deg of each pattern unit's preferred direction. This network responds by signaling one pattern direction for coherent motion but two distinct directions for transparent motion. Based on this, neural networks with properties of MT and MST neurons can automatically signal motion coherence or transparency. In addition, the model accurately predicts motion repulsion under transparency conditions.

We have developed a method that utilizes repeated sequences of pseudorandomly modulated stimuli for calculation of the SNR either in the time or frequency domains. The method has the advantage that the distribution of SNR over relevant frequencies is readily observed. In addition, a SNR value, calculated as the ratio of the corresponding variances, is an estimate of the true SNR because it has been weighted by the cell's frequency response. The procedure offers significant advantages when studying signal transmission in nonspiking cells like photoreceptors.

In the retina of the African clawed frog (Xenopus laevis), endogenous dopamine release increases in light and decreases in darkness. Exogenous melatonin and several chemical analogs of melatonin suppressed light-evoked dopamine release from frog retina in a concentration-dependent manner. The rank order of potency for inhibition of light-evoked dopamine release was melatonin » 5-methoxytryptamine ≥ N-acetylserotonin > 5-methoxytryptophol ⋙ serotonin. Melatonin did not suppress dopamine release below levels seen in darkness. The putative melatonin receptor antagonist luzindole inhibited the effect of melatonin. Luzindole enhanced dopamine release in darkness but had little effect in light. These data suggest a role for endogenous melatonin in dark-induced suppression of retinal dopamine.

Picrotoxin and bicuculline, GABA-A receptor antagonists, blocked melatonin-induced suppression of dopamine release. In the presence of melatonin, bicuculline was significantly less potent in stimulating dopamine release. These results suggest that melatonin enhances GABAergic inhibition of light-evoked dopamine release. This mechanism may underlie the light/dark difference in dopamine release in vertebrate retina.

The present study was performed as part of a systematic examination of the transmitter specificity of neuronal populations in the larval tiger salamander retina. Backfill-labeling of ganglion cells from the optic tectum was combined with double-label immunofluorescence histochemistry to determine if substance P and GABA are localized to ganglion cell populations in the tiger salamander retina. The triple-label analysis revealed the presence of substance P- and GABA-ganglion cells in both central and peripheral regions of the retina. Substance P-immunoreactive ganglion cells comprised 2% of the total population of backfill-labeled ganglion cells, while less than 1% of backfill-labeled ganglion cells expressed GABA immunoreactivity. Ganglion cells were not found to co-label for both substance P and GABA. Backfill-labeled displaced ganglion cells, which comprised 1.4% of the ganglion cell population, were not observed to be immunoreactive for either substance P or GABA. Forty-six point nine percent of substance P-cells in the ganglion cell layer were backfill-labeled and were identified as ganglion cells. GABA ganglion cells comprised less than 1% of GABA-immunoreactive cells in the ganglion cell layer.

Postembedding immunocytochemistry was used to determine the retinal distribution of the amino acid glutamine, and characterize amino acid signatures in the avian retinal ganglion cell layer. Glutamine is a potential precursor of glutamate and some glutamatergic neurons may use this amino acid to sustain production of glutamate for neurotransmission. Ganglion cells, cells in the inner nuclear layer, and some photoreceptors exhibited glutamine immunoreactivity of varying intensity. Ganglion cells demonstrated the highest level of immunoreactivity which indicates either slow glutamine turnover or active maintenance of a large standing glutamine pool relative to other glutamatergic neurons. Müller's cells in the avian retina are involved in glutamate uptake and carbon recycling by the rapid conversion of glutamate to glutamine, thus explaining the low glutamate and high glutamine immunoreactivity found throughout Müller's cells. Most chicken retinal ganglion cells are glutamate (E) and glutamine (Q) immunoreactive but display diverse signatures with presumed functional subsets of cells displaying admixtures of E and Q with GABA (7) and/or glycine (G). The four major ganglion cell signatures are (1) EQ; (2) EQγ; (3) EQG; and (4) EQγG.

Many studies of visual neurons make use of stimuli that are sinusoidally modulated in time, and take as the response the fundamental Fourier component of the firing. This is a study of the variability of the fundamental sinusoidal components.

A theoretical analysis shows that the variance of sinusoidal components should be nearly independent of their amplitudes; this is expected despite the observation that variance of firing rate increases with increasing firing rate. However, this result applies only to the variance of the complex amplitude, defined as the complex Fourier amplitude in response to each stimulus cycle. This variance is called the complex variance. The variance of the scalar amplitude, which is simply the amplitude in response to each stimulus cycle disregarding phase (scalar variance) is expected to shrink by a factor of up to 2⅓ as the response magnitude approaches zero.

If the relationship between variance of rate and rate is linear, complex variance should be independent of amplitude. If the relationship between variance of rate and rate is characterized by a compressive nonlinearity (as has been observed), the complex variance should very slightly decrease with increased amplitude, despite the main trend of increased variance of rate with increased rate.

Data from cat ganglion cells stimulated with sinusoidally modulated lights of various contrasts agree with the theory, although some individual cases show trends that may be indicative of nonlinearity in the relationship between variance of rate and rate.

We studied the effect of cooling the striate cortex on parvocellular lateral geniculate nucleus (PLGN) neurons in awake monkeys. Cooling the striate cortex produced both facilitation and inhibition of the responses of all neurons, depending on the stimulus presented. Cooling the striate cortex also altered the temporal distribution of spikes in the responses of PLGN neurons. Shannon's information measure revealed that cooling the striate cortex reduced the average stimulus-related information transmitted by all PLGN neurons. The reduction in transmitted information was associated with both facilitation and inhibition of the response. Cooling the striate cortex reduced the amount of information transmitted about all of the stimulus parameters tested: pattern, luminance, spatial contrast, and sequential contrast. The effect of cooling was nearly the same for codes based on the number of spikes in the response as for codes based on their temporal distribution. The reduction in transmitted information occurred because the differences among the responses to different stimuli (signal separation) were reduced, not because the variability of the responses to individual stimuli (noise) was increased. We conclude that one function of corticogeniculate feedback is to improve the ability of PLGN neurons to discriminate among stimuli by enhancing the differences among their responses.

In the δAsn20 Drosophila stock, the N-linked glycosylation site of opsin in Rl-6 receptors (Rhl) is absent. We used electroretinography (ERG), microspectrophotometry (MSP), and electron microscopy (EM) to quantify visual cell defects. Positive controls, w9, had wild type Rhl. MSP revealed minimal photopigment in δAsn20 for 6 days posteclosion; w9 had near normal visual pigment. ERG sensitivity and prolonged depolarizing afterpotential (PDA) were compared for δAsn20 and w9. δAsn20's Rl-6 function is decreased 100–fold at eclosion and diminishes until only R7/8 functions at 11 days. What little rhodopsin is routed to the rhabdomere functions. Morphometry showed smaller Rl-6 rhabdomeres in δAsn20 for 8 days posteclosion. Rhabdomeres in w9 were normal. A negative control, ninaE0117, a deletion of the Rhl gene, also has small rhabdomeres. δAsn20 and ninaE0117 lack the extreme rhabdomere elimination of ora (outer rhabdomeres absent), a nonsense mutant interrupting Rhl's coding sequence. δAsn20 and ora have surplus membrane while ninaE0117 does not. Freeze fracture reveals that δAsn20's rhabdomeric P-face particle count is as low as for vitamin A deprivation, consistent with an opsin defect. High particle density, organized into rows, is present in adjacent plasmalemma where surplus membrane accumulates. In summary, δAsn20 interferes with either synthesis, deployment, or maintenance of opsin.

Effects of extracellular calcium on signal transmission between photoreceptors and horizontal cells (HCs) are studied in superfused isolated retina of the larval tiger salamander. Horizontal cell light response is optimal when extracellular Ca2+ is maintained between 1–2 mM. Ca2+ levels beyond this range in either direction significantly reduce the HC light response amplitude. When extracellular Ca2+ is lowered from 2 mM to 0.5 mM, the rod input to HCs is reduced whereas the cone input is not affected. In comparison, the peak voltage responses of rods are not changed whereas the cone voltage responses are enhanced in 0.5 mM Ca2+. The selective suppression of rod input to HCs is probably due to the interplay of three factors: (1) the photocurrents, (2) voltage- and time-dependent membrane currents in photoreceptors, and (3) the Ca2-dependent synaptic gain between photoreceptors and HCs.

White noise stimuli were used to estimate second-order kernels for complex cells in cortical area VI of the macaque monkey, and drifting grating stimuli were presented to the same sample of neurons to obtain orientation and spatial-frequency tuning curves. Using these data, we quantified how well second-order kernels predict the normalized tuning of the average response of complex cells to drifting gratings.

The estimated second-order kernel of each complex cell was transformed into an interaction function defined over all spatial and temporal lags without regard to absolute position or delay. The Fourier transform of each interaction function was then computed to obtain an interaction spectrum. For a cell that is well modeled by a second-order system, the cell’s interaction spectrum is proportional to the tuning of its average spike rate to drifting gratings. This result was used to obtain spatial-frequency and orientation tuning predictions for each cell based on its second-order kernel. From the spatial-frequency and orientation tuning curves, we computed peaks and bandwidths, and an index for directional selectivity.

We found that the predictions derived from second-order kernels provide an accurate description of the change in the average spike rate of complex cells to single drifting sine–wave gratings. These findings are consistent with a model for complex cells that has a quadratic spectral energy operator at its core but are inconsistent with a spectral amplitude model.

Biological visual systems can detect positional changes that are finer than these systems' acuity to sine-wave gratings, a property known as hyperacuity. Some systems can even detect changes finer that the photoreceptor spacing. We report here that rabbit's directionally selective ganglion cells not only detect positional changes in the hyperacuity range, but also discriminate the direction of their motion. Our experiments show that directional selectivity occurs for edges of light moving as little as 1.1 μm (26” of visual angle) across the retina. This distance corresponds to a hyperacuity, since the acuity to sine-wave gratings of rabbit's On-Off DS ganglion cells is about 125 μm (50′). In addition, this distance is smaller than the minimal spacing between rabbit photoreceptors (1.9 μm or 46”), as estimated from cell-density studies (Young & Vaney, 1991). Such a hyperacuity suggests low-noise high-gain signal transmission from photoreceptors to ganglion cells and that directional selectivity can arise in small portions of retinal dendritic processes.

Rhodopsin gene expression has been used as a model system to study the mechanisms regulating photoreceptor gene expression. Previous transgenic experiments using rhodopsin promoter/lacZ fusion constructs identified some of the cis-acting DNA elements responsible for photoreceptor cell-specific expression. However, the issue of rod specificity vs. photoreceptor (rod and cone) specificity of the elements was not resolved. To address this issue, the specificity of reporter gene expression in the retinas of transgenic mice carrying bovine rhodopsin promoter/lacZ (ß-galactosidase) fusion genes was assessed using X-gal staining and electron microscopy. Two independent transgenic lines, one carrying a rhodopsin promoter fragment extending from −2174 to +70 base pairs (bp) relative to the messenger RNA start site and another line carrying a fragment from −222 to +70 bp, both showed reporter gene expression in cones as well as rods, although the level of staining appeared to be less in the cones than in the rods. These results demonstrate that the −2174 to +70 bp and −222 to +70 bp bovine rhodopsin promoter fragments are not rod-specific in transgenic mice and indicate that the existence of rod promoter mediated-expression in cones must be considered when interpreting results from transgenic experiments utilizing the rhodopsin promoter.

In addition to a modulatory function, somatostatin (SS) is likely to exert a morphogenetic and/or trophic role in the developing nervous system. In this study, a mouse monoclonal antibody directed to SS was used to investigate the posthatching development of SS-immunoreactivity (SS-ir) in the pigeon retina to provide a basis for a better understanding of the role of this peptide in retinal maturation. In the adult, SS-ir was observed in amacrine cells located in the inner nuclear layer (INL) of the entire retina. Two cell types were recognized according to their morphology. They showed a differential density distribution. Cell type indicated as “adult 1” (AD1) was characterized by pear-shaped cell bodies with single primary processes directed to the inner plexiform layer (IPL) and was mostly present in the red field. In contrast, cell type indicated as “adult 2” (AD2) was characterized by round-shaped somata with 1–3 primary processes and was highly represented in the fovea and the dorsal periphery. Posthatching maturation of the pigeon retina was characterized by drastic changes in the pattern of SS-ir. Over the first days posthatching, SS-ir was observed in sparsely distributed somata mostly located in the ganglion cell layer (GCL). This cell type indicated as “hatch” (H) was characterized by dense granular staining and became extremely rare at 7 days. Over the same period, growing SS-positive axons displaying enlarged growth cones were found in the optic tract (TrO). These observations suggest the possibility that ganglion cells transiently expressing SS are present at early stages of posthatching development. Of the two types of SS-containing cells observed in the adult, the first to be recognized morphologically was cell type AD1 which appeared at 2 days after hatching in the INL. These cells were virtually adult-like in morphology by 7 days. In contrast, cell type AD2 was not apparent until 7 days posthatching. The density (defined as number of cells/mm2 of retinal tissue) and the total number of SS-containing cells changed during posthatching maturation. In particular, the adult number of cell type AD1 was reached at about 10 days, while the number of cell type AD2 was reached at about 3 weeks posthatching. At this stage, both cell types also displayed their mature density distribution. The present findings suggest a temporal relationship between the maturation of SS-ir and developmental events which include the onset of light-driven activity and the maturation of retinal acuity. Our results also demonstrate significant differences in the pattern of SS-ir between the avian and the mammalian retina and suggest that SS-containing neurons represent important intraretinal association neurons in the retina of birds.

Ciliary ganglia from the pigeon, cat, and monkey were investigated for the presence of NADPH-diaphorase reactivity by use of a standard histochemical method. In the pigeon, where the ganglion is known to control lens and pupil function, and the choroidal vasculature, about one-third of the ganglion cells were densely stained and most other somata were lightly stained. In some cases, preganglionic terminals with a cap-like morphology were also darkly stained. The pattern of NADPH-diaphorase staining in mammals was very different from that seen in pigeons. In both mammalian species, where the ganglion is known to control lens and pupil function, a small number (less than 2%) of the ganglion cells were shown to be densely NADPH-diaphorase positive, revealing their neuronal processes. The presence of NADPH-diaphorase positive cells in pigeon, cat, and monkey ciliary ganglia suggests that nitric oxide may be used for intercellular communication in this ganglion, or in light of the known importance of nitric oxide in vascular control, some of these positive neurons may participate in the control of choroidal vasodilation.

Previous research suggests binocular rivalry disrupts extrastriate, but not striate processes, although the locus along the visual pathway at which such disruption first occurs is uncertain. It has been argued that subjective contours arise via a two-stage process in which end-stopped cells feed into orientation-sensitive neurones in V2, and that orientation aftereffects induced with subjective contours are the product of mechanisms similar to those giving rise to real contour aftereffects. If binocular rivalry disrupts the acquisition of subjective contour aftereffects, then it follows from this model that rivalry disrupts processing in V2. Experiments reported here confirm this and provide evidence which suggests binocular rivalry arises through interactions between binocular neurones, rather than via some type of specialized binocular rivalry mechanism.
