We have previously described a rat mAb directed against a peptide derived from the vif protein of HIV-1 that recognized two Schistosoma mansoni (Sm) antigens with a major band at 65 kDa. Epitope mapping of this mAb using overlapping hexapeptides derived from the vif peptide revealed that the motif recognized was PLPSVT. The screening of a Sm cDNA library led to the identification of two clones, Sm70 and Sm65. The two deduced protein sequences did not share any common structural features apart from the epitope recognized by the mAb (see below), and did not show significant identity to sequences present in the data bases. However, the N terminus of the deduced sequence of the Sm70 protein exhibits a consensus sequence known to be an ATP/GTP binding site. Furthermore, the C terminus of the deduced Sm65 protein sequence was found to contain a conserved hexapeptide with a consensus sequence LPETGE reported to be an important motif of the surface proteins of gram-positive cocci. Both proteins exhibit a peptide sequence (PLRSVT for Sm70 and PVGSVT for Sm65) similar to the epitope recognized by the mAb anti-vif. Western blotting experiments showed that the mAb anti-vif reacted with both proteins. However, only Sm65 was recognized by sera from HIV-1- seropositive individuals, whereas both proteins were recognized by S. mansoni-infected patients.

Stage-specific expression of the mRNA encoding the cercarial-specific 8 kDa CaBP has been described previously. To gain information on possible function(s) of this protein we raised antibodies to the CaBP in rabbits immunized with a CaBP-TrpE fusion protein synthesized in bacteria. Western blots showed high levels of CaBP in cercariae and 3 h schistosomula, trace amounts in 24 h schistosomula, and none in miracidia sporocyst and adult worm, as found for the mRNA. The CaBP molecule has a short half-life (≤4 h) similar to that of the mRNA. Other experiments demonstrate that the CaBP may interact with a putative target molecule in a calcium-dependent manner to form a complex of 45 kDa. Immunogold electron microscopy showed CaBP in selected regions of cercariae and 3 h schistosomula: tegument, head gland, subtegumental cells, flame cells, intestinal wall and the body-tail junction. Other investigators have shown that the head gland and subtegumental cells synthesize and translocate granules to the tegument during transformation from cercariae (living freely in water) to schistosomula (residing in vertebrate host). These observations and the time-course of CaBP detection suggest that the CaBP synthesized in the head gland and subtegumental cells is translocated to the tegument where it plays a role in tegument modifications required for adaptation to parasite life in the host. CaBP was not found in muscles and mitochondria, suggesting that it is not involved in the rapid motility and aerobic metabolism characteristic of cercariae.

The surface lipophosphoglycans (LPG) of Leishmania promastigotes express stage- and species-specific polymorphisms that are defined by variations in the type and number of phosphorylated oligosaccharide repeats. We have studied how these polymorphic structures control the development of transmissible infections in the sandfly vector as well as the species-specificity of vectorial competence. Procyclic promastigotes displayed an inherent capacity to bind to midgut epithelial cells of a competent vector. This capacity was lost during their transformation to metacyclic promastigotes, permitting the selective release and anterior migration of infective-stage parasites for subsequent transmission by bite. Midgut attachment and release were found to be controlled by developmental modifications in terminally exposed saccharides on LPG, which, depending on the species of Leishmania, involved either substitution or capping of terminal side-chain sugars, loss of terminal side-chain sugars, substitution or loss of neutral capping sugars. The stage-specific terminal sugars involved in midgut adhesion are, in some cases, also species-specific, and the extent to which these differences affect midgut attachment, forcefully predicted vectorial competence.

Populations of Pseudodiplorchis americanus infecting the desert toad, Scaphiopus couchii, have previously been shown to be remarkably stable from year to year, despite wide variation in conditions promoting invasion. The present study aimed to document the survival of both first-year and pre-existing adults under controlled laboratory temperatures. First-year worm survival in experimentally infected toads was shown to be very high for the first 5 months after migration to the definitive site, and there was no difference in survival at 25 °C or 15–20 °C. There was also no density-dependent survival during the same period: 20% of worms were recovered from the host urinary bladder, irrespective of initial intensities. After the first 5 months, there was a progressive loss of worms in toads maintained at 25 °C but not at 15–20 °C. Pre-existing adult populations were shown to be virtually identical to those under natural conditions for the first 4 months after toad capture. Following this, at 25 °C, populations declined and no pre-existing adults were recovered after 11–14 months. There was no such loss of pre-existing adults at 15–20 °C for up to 14 months. A diurnal temperature cycle of 20–34 °C (simulating temperatures during the desert summer) did not lead to a significant loss of worms. These observations suggest that during one annual cycle in the desert, when temperatures remain above 20 °C for less than 6 months, most adult worms established in the host urinary bladder will survive to the next transmission opportunity. However, the similar longevity of first-year and pre-existing adults in laboratory maintained toads shows that loss of worms cannot be due solely to parasite ageing. Temperature-dependent survival of P. americanus is suggestive of a host immune response. Low temperatures, which inhibit parasite growth and development, are essential for the survival of P. americanus.

A murine model using Heligmosomoides polygyrus and Trypanosoma congolense has been developed for studying the effects of concurrent chronic gastrointestinal nematode and trypanosome infections. Female outbred mice were infected either with 500 infective larvae (L3) of H. polygyrus or with 104 bloodstream forms of T congolense or both. In concurrent infections, animals were dosed with both parasites simultaneously or the trypanosomes were injected 5 or 10 days after the mice were infected with the nematode. The course of infection was monitored by routine parasitological and immunological techniques for 30 days after the H. polygyrus infection. Concurrently infected mice were severely compromised, except when T. congolense was superimposed on a 10-day-old (adult) H. polygyrus infection. In H. polygyrus-infected mice, simultaneous or subsequent infection with trypanosomes did not markedly influence worm establishment or fecundity, but the female worms were slightly stunted. Surviving mice displayed a markedly reduced antibody response to H. polygyrus antigens and a slightly reduced antibody response to T. congolense antigens.

Muscle fibres isolated from adult Schistosoma mansoni contracted in a dose-dependent manner when exposed to elevated K+ with a maximum response obtained with 25 mM K+. These contractions were dependent on extracellular Ca2+ since Co2+ (5 mM) or nicardipine (1 μM) blocked the high K+ contractions. Serotonin (300 nM or higher) was required for maintenance of high K+ contractions. With concentrations of serotonin less than 300 nM the response was dose dependent. 5-Methoxytryptamine or α-methylserotonin at 1 μM as well as 10 μM tryptamine were able to substitute for serotonin, but 1 μM 5-carboxyamidotryptamine was ineffective. The order of potency for antagonists (10 μM) was: methiothepin > metergoline > Ly-278,584 = ketanserin. This pattern of responsiveness does not fit well with any known mammalian serotonin receptor subtype. Since forskolin, an adenylate cyclase activator, is able to mimic the action of serotonin and H89, a protein kinase inhibitor, is able to block the effect of serotonin, the effect of serotonin on contractility of the muscle may be via a cAMP-dependent pathway.

Immunocytochemical techniques applied to sections and whole-mount preparations of cercariae from two species of trematodes, Trichobilharzia ocellata and Schistosoma mansoni, revealed the occurrence of immunoreactivity (IR) to several neurosubstances in the nervous system (NS). Immunostaining was localized in cerebral ganglia, in the main commissure, in anterior and posterior nerve trunks, as well as in a pair of nerve fibres running along the tail. In T. Ocellata, immunoreactivity (IR) was observed with antisera raised against: glutamate, FMRFamide, catch-relaxing peptide (CARP), small cardiac peptide B (SCPB), arg-vasotocin (AVT), arg-vasopressin (AVP), and substance P. In S. mansoni antisera raised against glutamate, FMRFamide, CARP, SCPB, α-caudodorsal cell peptides (α-CDCP), and cholecystokinin (CCK) showed neuronal IR. With the other 51 antisera tested no IR was observed. With anti-APGWamide, IR was observed outside the NS in cells of the wall of the daughter sporocyst and in flame cells of cercariae of T. ocellata. IR to FMRFamide was present in the escape glands of the intrasporocystic cercariae of T. ocellata and S. mansoni. IR to somatostatin was observed in subtegumental parenchymal cells of cercariae of S. mansoni. IR to met-enkephalin was present in cells of the cercarial embryos and in undifferentiated cells in developing cercariae. Trematodes are, together with cestodes, phylogenetically the oldest classes in which glutamate-like material and immunopositivity to a number of neuropeptides isolated from invertebrates has been demonstrated. The results are discussed in relation to immunocytochemical data obtained for other platyhelminths, to endogenous functions of the immunopositive materials, and to their possible role in parasite–host interactions.

Theileria-free waterbuck (Kobus defassa) born in captivity were successfully infected with Theileria parva sporozoites derived from ticks infected by feeding on African buffalo (Syncerus caffer). All waterbuck underwent mild infections with the development of sporadic schizont and piroplasm parasitosis when inoculated with sporozoite doses lethal to cattle. A carrier state of T. parva was demonstrated by feeding clean R. appendiculatus nymphs on two of these infected waterbuck. Tick batches from these waterbuck on 2 of 5 occasions transmitted lethal Theileria infections to cattle. In a separate experiment, waterbuck cells were infected and transformed in vitro by T. parva sporozoites derived from buffalo but not by cattle-derived T. parva (Muguga) sporozoites. Waterbuck cells infected in vitro with T. parva isolated from buffalo were inoculated into autologous waterbuck but no infections developed. Theileria parva isolates generated in this study from various sources were characterized using anti-T. parva schizont monoclonal antibodies (MAbs), and it was found that buffalo-derived and waterbuck-passaged isolates had different profiles. Species-specific synthetic oligonucleotide probes, restriction fragment length polymorphism (RFLP) analysis with cloned T. parva DNA probes, and DNA sequence analysis of the p67 sporozoite antigen gene confirmed that the waterbuck-passaged parasite was T. parva. The Tpr repetitive probe hybridization patterns from the waterbuck-passaged parasites were different from the other samples tested. The ribosomal genotype of the waterbuck-passaged T. parva was similar to that of cattle-derived T. parva Muguga. Analyses with both probes and MAbs suggested that a minor parasite population present within the T. parva 7014 buffalo- derived stock had been selected during waterbuck passage. A variable region of the p67 sporozoite antigen gene of the waterbuck-passaged T. parva was similar to that of cattle-derived T. parva stocks and different from that of buffalo- derived parasites. Based on these results, methods were suggested to confirm and quantitate the involvement of waterbuck in the epidemiology of cattle theileriosis.

This article presents an overview on phosphoglycan-containing components secreted by the insect and mammalian stages of several species of Leishmania, the causative agents of leishmaniasis in the Old and New World. Firstly, promastigotes of all three species considered, L. mexicana, L. donovani and L. major, shed lipophosphoglycan (LPG) into the culture medium possibly by release of micelles from the cell surface. Like the cell-associated LPG, culture supernatant LPG is arhphiphilic and composed of a lysoalkylphosphatidylinositol-phosphosaccharide core connected to species-specific phosphosaccharide repeats and oligosaccharide caps. Secondly, all three species release hydrophilic phosphoglycan. Thirdly, all three species appear to secrete proteins covalently modified by phosphosaccharide repeats and oligosaccharide caps. In the case of promastigotes of L. mexicana, these components are organized as two filamentous polymers released from the flagellar pocket: the secreted acid phosphatase (sAP) composed of a 100 kDa phosphoglycoprotein and a protein- containing high-molecular-weight-phosphoglycan (proteo-HMWPG) and fibrous networks likewise composed of phosphoglycan possibly linked to protein. Structural analyses and gene cloning suggest that the parasites can covalently modify protein regions rich in serine and threonine residues by the attachment of phosphosaccharide repeats capped by oligosaccharides. We propose that the networks formed in vitro correspond to fibrous material previously demonstrated in the digestive tract of infected sandflies. In the case of L. donovani, the sAP is also modified by phosphoglycans but contains neither proteo-HMWPG nor does it aggregate to filaments. Finally, L. mexicana amastigotes release proteo-HMWPG via the flagellar pocket into the parasitophorous vacuole of infected macrophages. This material appears to be released into the tissue of the infected mammal upon rupture of infected macrophages during lesion development. This secretory product may contribute to the pathology of lesion development.

Excretory/secretory (ES) products obtained by in vitro culture of infective-stage larvae of Trichinella spiralis and T. pseudospiralis were injected intramuscularly at various intervals into mice. Mini-osmotic pumps containing T. spiralis ES products were also implanted subcutaneously and intraperitoneally into rats. The introduction of ES materials into muscles elicited extensive lesions which included dissolution of myofibres, mobilization of mononuclear and polymorphonuclear leucocytes, angiogenesis, hypertrophy of myonuclei, myotube formation, mitosis, muscle bundles becoming rounded and separated from each other, disappearance of Z, I and A bands of sarcomeres, increase in endoplasmic reticulum and Golgi complexes, decrease in glycogen and relocation of mitochondria. These are considered as degenerative/regenerative changes of muscles to injury. Immunodominant epitopes of specific 45–53 kDa glycoproteins in ES antigens of T. spiralis could not be detected in hypertrophic nuclei of injected muscles by using polyclonal and monoclonal antibodies and immunocytochemical methods. ES products of T. spiralis failed to stimulate unsensitized lymphocytes in the lymphocyte transformation test. Infective-stage larvae of T. spiralis released from muscles were found capable of forming nurse cells after injection subcutaneously into rats. It is postulated that the invasion of muscles by trichinellids elicits two independent events, i.e. a general degenerative/regenerative response of muscles and a specific change in genomic expression of myonuclei. The two events are probably mediated by different effector molecules.

Relatively simple protocols employing non-radioactive DNA probes have been used for the detection of African trypanosomes in the blood of mammalian hosts and the saliva of live tsetse flies. In combination with the polymerase chain reaction (PCR), the protocols revealed trypanosomes in buffy-coat samples from antigenaemic but aparasitaemic cattle and in the saliva of live, infected tsetse flies. Furthermore, the protocols were used to demonstrate concurrent natural infections of single tsetse flies with different species of African trypanosomes.

The effects of the novel benzimidazole, triclabendazole (TCBZ) (‘Fasinex’, Ciba-Geigy), in its active sulphoxide metabolite form (TCBZ-SX), on the tegumental ultrastructure of Fasciola hepatica were determined in vitro by transmission electron microscopy (TEM), using both intact flukes and tissue-slice material. At a concentration of 15 µ/ml, the tegument of the whole adult fluke showed ultrastructural changes only after prolonged time-periods, with vacuolation at the base of the syncytium and accumulation of T2 secretory bodies in the tegumental cells. At a concentration of 50 µ/ml, with both whole flukes and tissue-slices, the tegument appeared extremely abnormal with accumulation of secretory bodies towards the base of the syncytium. With longer incubation times, the tegument was completely sloughed away and the tegumental cells became synthetically inactive. The tegument of the 3-week-old juvenile became progressively convoluted at the apex, while in the basal regions there was severe vacuolation. In the tegumental cells, there were accumulations of T1 secretory bodies. These results confirm TCBZ as a potent fasciolicide, being very effective in disrupting the fluke tegument. They may go some way to explain the mode of action of this important fasciolicide.

Parasitological and immunological interactions between Eimeria vermiformis or E. pragensis and Trichinella spiralis were investigated during concurrent infections in NIH, BALB/c and B10.G inbred mice. The establishment of T. spiralis was unaffected by the presence of either coccidium, but expulsion of adult worms was delayed significantly in mice infected with E. vermiformis; E. pragensis did not have this effect. Replication of E. vermiformis was enhanced in concurrent infections with T. spiralis, but that of E. pragensis was reduced. Specific immune responses to each parasite were unaffected in mice infected with T. spiralis and E. pragensis, but levels of some responses were reduced when T. spiralis and E. vermiformis were combined. Thus both in vitro antigen-induced proliferation of mesenteric lymph node cells (MLNC) and intestinal mastocytosis were lower than in singly infected mice. Mitogen (Con A) responsiveness of MLNC was not affected in mice infected with T. spiralis and E. vermiformis, and cells from these mice were capable of transferring protective immunity to the nematode in naive recipients. Injection of monoclonal antibody to interferon gamma, a major component of the cytokine response to E. vermiformis, did not prevent delay of worm expulsion in concurrent infections. The results are discussed in terms of possible interactions between the T helper cell subsets or the inflammatory components of the responses induced by each parasite.

We have used two monoclonal antibodies, 128C3/3 and 504B1, to immunolocalize their carbohydrate epitopes in different developmental stages of Schistosoma mansoni. Both epitopes contain fucose: mAb 128C3/3, as we have shown previously, recognizes fucose in a novel, possibly internal linkage (Levery et al. 1992) while mAb 504B1, as we show here, bound to the Lex epitope, which contains fucose α1 → 3 linked to N-acetyl-glucosamine. The tissue expression of these epitopes was strikingly different and both elicit an immune response in infected hosts. The mAb 128C3/3-defined epitope was exposed on the surface of all larval stages but not on adult worms; however, it was found in the excretory system of adult worms of both sexes. In contrast, surface expression of the Lex epitope was initiated after the transformation of cercariae to schistosomula and was maintained throughout the adult life in both sexes.

Gyrdicotylus gallieni is a viviparous gyrodactylid monogenean that infects the buccal cavity and pharynx oi Xenopus laevis laevis. Offspring directly recolonize the same host as their parent and transmission is carried out exclusively by adult worms which are capable of leech-like stepping locomotion. The infrapopulation dynamics (at 20 °C) of this species were studied by experimental exposure to single worms of previously uninfected hosts (which were dissected at various time intervals post-infection). Infrapopulations increased exponentially until 50 days post-infection (p.i.), during which time the intrinsic rate of increase was estimated by regression as 0·065/parasite/day. After this, infrapopulation sizes showed greater variability, with established infections becoming extinct 2–5 months p.i.: elimination was probably due to someform of host reaction, as parasite lineages were maintained for up to 10 months by transfers to successive uninfected hosts.The development of infrapopulations is similar to that reported for other viviparous gyrodactylids parasitic on the bodysurface of teleosts. However, the intrinsic rate of increase was comparatively slow in G. gallieni and the duration of infection in isolated hosts prolonged. Slow-growing infrapopulations may elicit a host response more slowly and extend the period during which dispersal to a new host or host population can occur. This might be important in G. gallieni for which the internal site of infection could limit the transmission rate, as worm migration from the oral cavity or accidental detachment and expulsion is necessary for host-host transfer to occur. Exposure of wild caught X. l. laevis of unknown infectionhistory to 1 (n = 33) or 10 (n = 10) worms produced only 2 established infrapopulations (in both cases hosts initially infected by single worms), compared with establishment of over 60% in naive hosts infected with single worms: this suggests that host resistance may be an important factor in the population dynamics of G. gallieni.

Foraging strategies of eight species of entomopathogenic nematodes were predicted from their response to host volatile cues and dispersal behaviour on 2-dimensional substrates. Positive directional response to chemical cues and similar distances travelled on smooth (agar) or nictation substrates (agar overlaid with sand grains) by Heterorhabditis bacterio-phora, Heterorhabditis megidis, Steinernema anomali, and Steinernema glaseri suggest their cruising approach to finding hosts. The absence of directional response and less distance travelled on nictation substrate, than on smooth agar by Steinernema carpocapsae and Steinernema scapterisci suggest their ambushing mode of foraging. Steinernema feltiae and Steinernema sp. responded directionally to host volatiles, but travelled less distance on the nictation substrate than on smooth agar; the two species also did not nictate. The cruiser species located hosts more effectively in the sand columns, whereas the ambushers were more effective at finding hosts on filter paper. Steinernema feltiae and Steinernema sp. performed equally on filter paper and in the sand column. We conclude that H. bacteriophora, H. megidis, S. anomali and S. glaseri cruise to find hosts, whereas S. carpocapsae and S. scapterisci ambush hosts. Steinernema feltiae and Steinernema sp. are intermediary in the search continuum sharing some characteristics of both ambush and cruise foragers.

The present study examines the role of host genetics in predisposition to Ascaris lumbricoides and Trichuris trichiura infection, by comparing the associations between age-standardized infection intensities of parents and their children (genetically related), with age-standardized infection intensities of parents alone (not related), within families previously demonstrated to exhibit familial predisposition. The lack of a consistent trend in infection intensity associations within families, in particular the lack of a stronger association between parents and their children than between unrelated parents, suggests that host genetic factors are not a major determinant of infection status. If there is a genetic basis for predisposition, then the data suggest that the effects of this genetic basis are overwhelmed by other, environmental or behavioural features of the family household.

Macroparasites almost invariably exhibit overdispersed distributions of parasites/host, yet the specific causes of such aggregations remain poorly understood. The present study focused on the distribution of the parasitic nematode Howardula aoronymphium among its hosts, several species of mycophagous Drosophila. The distribution of parasites/host is close to random for cohorts of flies of a given host species emerging from single mushrooms. At the level of Howardula populations, overdispersion of parasites among hosts results primarily from variation among subgroups of hosts in their exposure to infective-stage nematodes. The sources of variation identified in this study include Drosophila host species, the site where flies bred, mushroom species within sites, and, most importantly, individual mushrooms within mushroom species at a site. For the mean intensity of parasitism observed in this study, the degree of aggregation is typical of macroparasites in general. Combinations of random distributions with different means, resulting from variation among groups in exposure to parasites, may be a common cause of the overdispersion of macroparasites among hosts.

The ability of Trypanosoma cruzi to induce erythrocyte membrane destabilization in vitro was studied. Epimastigote forms adhered to human erythrocytes and caused fusion or lysis of the red cells, depending on the conditions of the interaction. Red cells were fused in the presence of calcium, while haemolysis was induced in the absence of the cation. Dextran 60 C facilitated fusion but delayed lysis. Optimum pH and temperature for fusion were 7·4 and 37 °C, respectively. Lipid alterations were produced in the plasma membrane of the red cell during the interaction with the parasite. A Ca2+-independent increase of lysophospholipids and free fatty acids was common to both the lysis and fusion processes. A relative increase of 1, 2-diacylglycerides was unique to the fusion process and these changes were dependent on Ca2+. The transfer of free fatty acids and lysophospholipids from T. cruzi to erythrocyte membranes was demonstrated using parasites pre-labelled with radioactive phospholipids. Pre-treatment of parasites with exogenous phospholipase A2 abolished the fusogenicity, while lysis was increased. Neither fusion nor haemolysis occurred when the parasites were pre-treated with fatty acid free albumin, phospholipase A2 inhibitors or when these compounds were present in the medium during the parasite-erythrocyte interaction. Our results suggest that T. cruzi induces erythrocyte membrane destabilization in vitro by transfer of lipid material in a calcium independent manner and that this ion is necessary for other membrane alterations that lead to erythrocyte fusion.

Glucose is utilized as a significant source of metabolic energy by Leishmania parasites. This sugar is accumulated by the parasite via a specific carrier-mediated transport system located in the parasite membrane. Parasites may also contain another transporter that shuttles glucose between the cytoplasm and the glycosome, a membrane-bound organelle where the early steps of glycolysis occur. The transport systems of both the insect stage promastigotes and the intracellular amastigotes have been characterized and shown to have kinetic properties that are consistent with the different physio-logical environments of the insect gut and the macrophage phagolysosome. Several genes have been cloned from Leishmania species which encode proteins with substantial sequence similarity to glucose transporters from mammals and lower eukaryotes. Two of these genes are expressed preferentially in the promastigote stage of the life cycle, where glucose is more readily available and more rapidly transported and metabolized than in the intracellular amastigotes. One of these two developmentally-regulated genes has been functionally expressed in Xenopus oocytes and shown to encode a glucose transporter. A third gene encodes a protein that is also a member of the glucose transporter family on the basis of sequence similarity and proposed secondary structure. However, the significant differences between this protein and the other two suggest that it is likely to transport a different substrate. Functional expression will be required to define the specific biochemical role of each gene within the parasite.
