An aggregative dispersive dichotomy (ADD) model is presented to describe the distribution of parasites in host populations. The ADD model is a mathematical construct which provides two complementary measures extracted from a reformulated negative binomial (NBD) and an inequality model, which combine to capture observed patterns of a parasitic infection. The dispersion element is modelled using the NBD with the threshold set at a parasite level above zero. By applying binomial dichotomy, the host community is divided into two sub-populations, one including hosts harbouring parasites up to the threshold and the other with parasites above the threshold level. The k parameter, derived from the NBD, provides a cumulative probability. However, k is relatively insensitive to variations in the degree of aggregation, a known feature of the NBD model. The aggregation of parasites above the threshold in the host sub-population is evaluated by using an inequality model which is indexed by a scale-free parameter δ(δ ≥ 1) and provides an accurate measure of parasite aggregation. Applications of this model are made from field and simulated data in wood mouse populations infected with the trichostrongylid nematode Heligmosomoides polygyrus from a woodland site in Surrey.

The presence of helminths and protozoans in public squares and lawns of the city of Maringá, southern Brazil, during winter and summer was assessed in order to evaluate their seasonal fluctuations in relation to edaphic and climatic factors. Samples were collected from January 2003 through June 2004 in 90% (13) of all public squares covered by sand, and in 30% (4) of all lawns used as leisure areas. The samples were analysed quantitatively by modified centrifugal-flotation and sedimentation in water techniques, and qualitatively by a method based on positive larval thermo-hydrotropism. Meteorological data were recorded, and physical, chemical and structural characteristics of the soil were analysed. One hundred and thirty samples of sand from squares, 65 in summer and 65 in winter, and 40 samples of grass from lawns, 20 in each season, were collected. All samples from lawns, 62 (95.38%) from squares in winter and 45 (69.23%) in summer, contained protozoans and/or helminths. Eggs of Toxocara spp. were the most frequently observed parasites in both winter and summer in squares (P < 0.0001) and in lawns (P = 0.6142), being equally distributed among the different locations (P = 0.2038). Species diversity was lower in winter; fewer parasites were found in summer. This region, with a tropical climate and a mild winter dry season, has favourable edaphic and climatic conditions for soil contaminants to persist year-round. In addition, the high frequency of animals such as dogs and cats and the poor sanitary measures in force made it possible for zoonoses to be transmitted in the public spaces.

An epidemiological survey was undertaken of anisakids in 139 specimens (length: 13.2–24.5 cm) of pouting or bib (Trisopterus luscus) captured off the coast of northern Spain in the Cantabrian Sea. Third-stage larvae of two species of nematodes, Anisakis larvae type I and Hysterothylacium aduncum, were isolated. One adult female H. aduncum was also detected in the intestine of one pouting. Total prevalence of anisakids was 88.5%. Hysterothylacium aduncum and Anisakis showed, respectively, prevalence of 87.8% and 22.3%, mean intensity of 19.7 and 3.5, and mean abundance of 17.3 and 0.8. Analysis of infection parameters as a function of host length revealed a much higher prevalence in pouting specimens with length < 20 cm (94.4% for H. aduncum; 28.0% for Anisakis) than in those with length ≥ 20 cm (65.6% for H. aduncum; 3.1% for Anisakis). The high mean intensity of Anisakis in muscle of parasitized pouting (5.9) may pose human health risks, although these are minimized by eating only thoroughly cooked pouting, as is the custom in Spain.

The possible role of the cockroach Periplaneta americana in the transmission of Toxocara canis eggs and larvae via faeces and tissue migration was studied. Cockroaches fed with 3 × 105 and 5 × 105 embryonated eggs were found to harbour viable eggs and larvae from days 1 to 5 post-infection (DPI). At necropsy on 5 DPI, eggs and larvae were also recovered from the rectal contents but not from the tissues of cockroaches. In addition patent infections were established in pups fed on infected faeces of cockroaches, with eggs first appearing in the faeces of pups at 38 DPI. Adult worms of T. canis were also recovered at necropsy. Therefore the importance of cockroaches as good mechanical disseminators of ascarid eggs, especially T. canis, is discussed.

Primitive epithelium and outer tegumental layer formation during early cercarial development was studied in Prosorhynchoides borealis using electron microscopy. It demonstrated that germinal cells freely floating in the sporocyst body cavity divide to give rise to naked cell aggregates. These early embryos are highly irregular in outline and are composed of blastomeres differing in size and structure. In embryos consisting of about 12–14 cells a few (possibly only two) superficial macromeres become concave and produce thin extensions which envelop the embryonic mass before fusing to form a syncytial primitive epithelium. This primitive epithelium forms syncytial connections with underlying embryonic cells. Primordial tegumental cells become apparent in late germinal balls below the primitive epithelium. These cells expand and fuse to give rise to an embryonic nucleated tegument. The embryonic tegument is connected to peripheral embryonic cells by thin cytoplasmic bridges until the basement lamina is formed. Subsequently, the primitive epithelium is shed by the embryos and the nuclei in the embryonic tegument undergo pyknotic degeneration. These results are analysed and compared with data from studies on other trematode species and it is concluded that the primitive epithelium is derived from the embryo in at least the majority of digeneans.

The abundances of different species in a parasite community are never similar: there is typically one or a few numerically dominant species and many species with low abundance. Here, we determine whether basic features of parasite communities are associated with strong dominance by one or a few species, among 39 component communities of gastrointestinal helminths in marine fishes from Brazil. First, we tested whether the shape of the species abundance distribution in these communities fits that predicted by several theoretical models, using a goodness-of-fit procedure. Only the canonical lognormal model could be rejected for 5 out of 39 communities; all other comparisons of observed and predicted abundance distributions showed no significant differences, although this may be due to limited statistical power. Second, we used the ratio between the abundance of the most abundant species and either the second or third most abundant species, as indices of dominance; these show, for instance, that the dominant species in a community is typically twice, but sometimes over ten times, as abundant as the next most abundant species. We found that these ratios were not influenced by either the community's species richness, the mean number of individual parasites per host, or the taxonomic identity of the dominant species. However, the abundance ratio between the first and third most abundant species in a community was significantly correlated with an independent index of species interactivity, based on the likelihood that the different parasite species in a component community co-occur in the same host individuals: the difference in abundance between the dominant and third most abundant species was greater in communities characterized by weak interactions. These findings suggest that strong interactions may lead to greater evenness in the abundance of species, and that numerical dominance is more likely to result from interspecific differences in recruitment rates.

The factors that determine parasite assemblages among the clariid fishes of Lake Victoria, Tanzania were studied between August 2003 and February 2005. Six hundred and fifty-six fish belonging to seven species were necropsied and examined for parasites, from which 31 species of metazoan parasites were recorded. The community was dominated by the nematodes both in species and numbers. Most species were generalists with only two trematodes, Diplostomum mashonense and Tylodelphys species, being specialists of Clarias gariepinus. Ten species were considered core and predictable. Parasite species richness, number of individuals per host and Shannon–Wiener diversity indices were generally high. At the compound community level, a mean number of 7.8 parasites were shared among different species of fish and the maximum number of parasites species per fish at the infracommunity level was seven. Levels of similarity in parasite species richness at the component community level ranged from 29.6 to 61.5%. The study concludes that parasite communities in clariid fishes of Lake Victoria are structured by ecological factors. At the infracommunity level, host size, diet and vagility promoted a richer parasite community. At the compound level, two factors were crucial, namely the intermixing of the waters in the lake and the predominant and mobile C. gariepinus.

Eighty-seven red foxes were investigated for the presence of Toxocara eggs on a sample of their hair from the peri-anal region. The worm burden of Toxocara in each fox intestine was also assessed and the relationship between eggs on the hair and worms in the intestine explored. Twenty-eight per cent of the foxes were found to have Toxocara eggs on their hair, with an average of 1.31 ± 3.21 eggs per gram of hair ( ± SD). Sixty-one per cent of foxes harboured Toxocara worms within their intestines, with a mean worm burden ( ± SD) of 4 ± 8. Host age and sex did not significantly influence the observed eggs on the hair or the worm burden. No significant correlation was found between the numbers of eggs on the hair and the worm burden within the intestine. These data collected from foxes are explored in the context of data from stray dogs and the possible epidemiological differences are discussed.

A study was conducted to determine the influence of environmental change on an endoparasite, Echinorhynchus gadi (Acanthocephala) of Atlantic cod (Gadus morhua) over a 30-year period off the coast of Labrador in the north-western Atlantic, North Atlantic Fisheries Organization subareas 2J–3K. Cod, once an abundant fish species that had been commercially exploited for many decades, declined precipitously during the mid-1980s onwards. This decline was attributed to climatic changes that affected the entire food chain from zooplankton to fish, sea birds and marine mammals. A monitoring programme was introduced, sampling cod by otter trawling using research vessels. The fish, after capture, were frozen at − 20°C, subsequently thawed and the digestive tract removed and examined for the parasite in 2006. Data from samples taken in 1976, 1980–81, 1986, 1990, 2000 and 2003 were compared statistically with those collected in 2006. The results indicate a decline in the prevalence and mean abundance of E. gadi in 1986 with a minimum in 2000 but increasing gradually in 2003 and 2006. These changes were coincident initially with a decline of oceanic temperature and the entire food web, including capelin (Mallotus villosus), a preferred prey of cod and primary source of E. gadi. The increase in prevalence and mean abundance of the parasite in 2006 were associated with an increase of oceanic temperature and the return of small schools of capelin to offshore areas. Cod older than 4 years harboured a greater abundance of E. gadi than younger fish, while no difference was observed between the sexes. The results suggest that the abundance of E. gadi can be useful as a bioindicator of environmental changes in the north-western Atlantic.

The influence of subclinical nematodosis on the kinetic disposition of albendazole was evaluated in goats following oral and intraruminal administration. The disposition curves of its metabolites indicated increased uptake of the drug in parasitized goats following intraruminal compared to oral dosing (P < 0.05). The midpoint for the pharmacologically active metabolite, albendazole sulphoxide, in the circulatory compartment was around 0.6 μg ml− 1 both in parasitized and naïve goats. The period of exposure to this concentration was around 14 h (oral route), 18 h (intraruminal route) and 16 h (oral route), 17 h (intraruminal route) in parasitized and naïve goats, respectively. As the duration of exposure of parasites to the toxic concentration of the anthelmintically active metabolite was prolonged, it could be assumed that intraruminal delivery of the drug would improve the efficacy of albendazole in parasitized goats.

According to the Brazilian Agricultural Ministry, the prevalence of cystic echinococcosis in the state of Rio Grande do Sul, Brazil has decreased between 1990 and 1998, possibly due to the common use of albendazole to control parasites in sheep. Therefore, albendazole (ABZ) action was tested on hydatid cysts of sheep experimentally infected with eggs of Echinococcus granulosus in Uruguaiana County (RS) with the aim of evaluating the observed changes in the parasite's prevalence. The sheep were divided into three groups of 15 animals each and treated as follows. Group 01 was a control (without treatment), groups 02 and 03 received 7.5 mg/kg and 15 mg/kg, respectively, of ABZ orally for 12 months, at intervals between 36 and 45 days. At the end of the studied period the animals were necropsied and the viability of the hydatid cysts was evaluated. Lung and hepatic cysts of sheep treated with ABZ were reduced in size. Degeneration and evident calcification were seen in 87.3% (group 02) and 75.7% (group 03), which were significantly different (P < 0.01) from the 17.3% demonstrated in the control group. It could be concluded that ABZ used early and over long periods, even in small doses, is capable of promoting lesions that devitalize the cysts. This leads to a decrease in fertility of E. granulosus in sheep and could be the reason why the prevalence of cystic echinococcosis has decreased in recent years.

The aim of this study was to examine the effect of different temperatures and humidities on the infectivity of Echinococcus granulosus protoscolices. Eighteen dogs (6 groups, n = 3 each) were fed with offal mince harbouring approximately 20,000 protoscolices of E. granulosus of different viabilities. Dogs were infected with E. granulosus protoscolices of: (1) 5% viability at − 10°C and 50% relative humidity (RH); (2) 30% viability at 0°C and 60% RH; (3) 20% viability at +10°C and 65% RH; (4) 15% viability at +30°C and 75% RH; (5) 11% viability at +40°C and 80% RH; (6) 68% viability (control group). Dogs in each group were necropsied at 29–49 days post-infection. Mean intensities of E. granulosus recovered from dogs were 256.7 ± 60.3 in the second group; 32.7 ± 7.1 in the third group; 40.3 ± 15.5 in the fourth group and 1533 ± 513 in the control group. However, no parasites were recovered from the first and fifth groups. Results obtained in the present study show that larval stages could be infective for 1 to 4 weeks during spring, autumn or winter months when maximal temperatures are approximately 0–10°C. In conclusion, cold-storage depots in slaughterhouses and abattoirs where sheep carcasses might be discarded should be kept at − 20°C for 2–3 days, dogs should be properly controlled and adequate control programmes must be established in areas where the disease is endemic.

Mesocestoides vogae tetrathyridia infection in mice causes hepatocyte injury, hepatic granulomatous inflammmation, liver fibrosis and chronic peritonitis manifested with portal hypertension. To reduce the detrimental effect of parasites on the host liver, the effect of the anthelmintic drug praziquantel (PZQ) in combination with natural products silymarin (an antioxidant) and β-glucan (an immunomodulator) was investigated. The therapeutic effect of drugs was assessed by means of aminotransferase (alanine aminotransferase (ALT) and aspartate aminotransferase (AST)) activities, content of albumin, total proteins and hyaluronic acid (HA) in sera of ICR mice infected with M. vogae larvae. Animals were treated with PZQ suspended in oil emulsion (Group 1), PZQ combined with silymarin incorporated into lipid microspheres (LMS) (Group 2), PZQ combined with β-glucan incorporated into liposomes (LG) (Group 3), PZQ co-administered with LMS and LG (Group 4). Untreated animals (Group 5) served as the control. Treatment of animals started at the early chronic phase of infection (day 14 p.i.) and lasted 10 days; serum samples were collected on days 0, 7, 14, 25, 28, 31, 35 and 45 p.i. ALT and AST activities were significantly (P < 0.05) decreased in Groups 2, 3 and 4. HA content was significantly (P < 0.05 and 0.01) lower in Groups 2 and 4. Albumin levels were decreased in Groups 2 and 4, total protein concentration decreased in Groups 1 and 3 (P < 0.05 and 0.01). These results showed that combined treatment of PZQ with silymarin and/or β-glucan was able to ameliorate or suppress fibrogenesis in the liver, protect liver cells from oxidative damage and, possibly, stimulate regeneration of the parenchyma.

Twenty extracts from plants from Sierra de Huautla Biosphere Reserve, Morelos, Mexico were evaluated against Haemonchus contortus infective larvae in an in vitro assay. The plant species evaluated were Bursera copallifera, B. grandifolia, Lippia graveolens, Passiflora mexicana, Prosopis laevigata, Randia echinocarpa and Urtica dioica. The plants were separated into their parts and macerated with different solvents (n-hexane, acetone, ethanol and methanol). An in vitro assay was used to evaluate the anthelmintic activity against unsheathed third stage H. contortus infective larvae. The experiment was carried out in 24-well cell culture plates at room temperature with three replicates per treatment and using a concentration of 20 mg ml− 1. Ten 5 μl aliquots were taken from the corresponding wells and deposited on a slide for microscopical observation at 24, 48, 72 and 96 h post-exposure. The evaluation criteria were based on the average numbers of live and/or dead larvae in the different treatments. Alive and dead larval numbers were statistically analysed through the ANOVA test (P>0.01). The Tukey test was used as a complementary tool to determine which treatment was different from the other treatments (P>0.05). The highest mortality was observed with P. laevigata hexanic extract from stem and leaves combined, which produced 51%, 81% and 86% larval mortality at 24, 48 and 72 h post-exposure, respectively. On the other hand, B. copallifera stem acetonic extract exhibited 18%, 59% and 66% nematicidal activity after 24, 48 and 72 h of exposure, respectively.

Environmental and socio-cultural variables influencing the distribution of helminth eggs in 608 soil samples were studied in 14 playgrounds that differ in socio-economic status in Kaduna metropolis, Nigeria, using a modified sieving method and a sucrose flotation medium of specific gravity 1.27. Helminth eggs were found in 62% of the soil samples and the distribution was as follows: Toxocara spp. 50.4%, Taenia spp./Echinococcus spp. 36.9%, Dipylidium caninum 26.3%, Ancylostoma spp. 9.0%, Ascaris spp. 7.2%, Trichuris spp. 3.7% and Ascaridia spp. 1.9%. A higher prevalence (68.1%) was recorded during the dry harmattan period while in the rainy period the rate was 58.1%. Mean egg densities ranged from 1.11 ± 0.32 to 3.92 ± 2.47 in areas moderately rated. Samples from site 14, which was highly rated, were more contaminated (78.1%) than those collected from other sites, while the intensity of contamination (14.0%) was more in moderately rated site 4 than in the rest of the sites. There were significant associations between the prevalence of helminth eggs and rainy period of the study (odds ratio (OR) = 0.38; 95% confidence interval (CI) on OR: 0.20 < OR < 0.70), presence of dogs (OR = 0.56; 95% CI on OR: 0.37 < OR < 0.85) and grass (vegetation) (OR = 1.44; 95% CI on OR: 1.03 < OR < 2.04) in the sites. On the other hand, there was no association between the prevalence of helminth eggs and the dry period of the study, presence of refuse in the playgrounds, topography of playgrounds, depth of sample collection and socio-economic status of people in playgrounds (P>0.05). This study shows that the period of study, the presence of dogs and vegetation influence the prevalence of helminth eggs in soil in Kaduna metropolis.

Spiroxys ankarafantsika, sp. nov. is described from Madagascar in two species of freshwater turtles, Pelusios castanoides and Pelomedusa subrufa. Spiroxys ankarafantsika differs from other species of the genus in having pseudolabial teeth only on the median lobes and no other cuticular prominences, a smooth cuticular collar, deirides as cervical minute spine-like projections, and a gubernaculum without tubes. This is the first record of a nominal species of the genus Spiroxys from the Ethiopian region.

A modification of gDNA extraction was developed for the polymerase chain reaction (PCR) technique, intended for the detection and differentiation of Toxocara spp. eggs in soil or sediments. Sand samples from sandpits confirmed as being contaminated with Toxocara spp. eggs by the flotation technique were analysed by PCR. The use of proteinase K made it possible to obtain genomic DNA from the sample without needing to isolate eggs using flotation or to inactivate PCR inhibitors present in the sand. Specific primers in the PCR reaction allowed discrimination between T. canis and T. cati eggs. The modification simplified the procedure, thanks to eliminating the step of gDNA isolation from eggs, which is both laborious and difficult.

The use of parasite body size (i.e. body mass) is a promising proxy to improve the study of patterns in parasite infracommunities, which are usually analysed using only numerical descriptors. This study deals with the importance and effect of the presence of a large endoparasite species, the cestode Anonchocephalus chilensis in a marine fish species, Genypterus maculatus, on the structure of the parasite infracommunities. Numerical and volumetric measures of aggregated properties of parasite infracommunities were compared and their correlation examined. The highly dominant presence of A. chilensis by volume causes a dramatic change in the patterns observed, including a smaller total volume of the remaining species when this volumetrically dominant species is present. However, C-scores and V-ratios, both indices based on null models of species occurrence, do not support the idea of communities structured by interspecific competition. Analyses reveal that numerical and volumetric community descriptors are complementary ways to search for patterns and to reveal processes within these systems.

Although hookworms are known to stimulate inflammatory responses in the intestinal mucosa of their hosts, there is little quantitative data on this aspect of infection. Here we report the results of experiments conducted in hamsters infected with Ancylostoma ceylanicum. Infection resulted in a marked increase in goblet cells in the intestinal mucosa, which was dependent on the number of adult worms present and was sustained as long as worms persisted (over 63 days) but returned to baseline levels within 7 days of the removal of worms by treatment with ivermectin. Increased mast cell responses were also recorded. Levels were again dependent on the intensity of worm burdens and lasted as long as 63 days after infection. When worms were eliminated, mast cell numbers took over 2 weeks to return to normal. Paneth cell numbers fell soon after infection, the degree of reduction being dependent on the worm burden. After clearance of worms, Paneth cell numbers returned to normal within a week, but then rebounded and numbers rose to higher levels than those in control naïve animals. The time course of the response was similar whether animals experienced a chronic low-intensity infection without loss of worms or a higher intensity infection during the course of which worm burdens were gradually reduced. Clearly, A. ceylanicum was able to induce a marked inflammatory response in its host's intestine which was sustained for over 9 weeks after infection, and which hamsters appeared able to tolerate well. Our data draw attention to the resilience of hookworms which, unlike many other nematodes, are able to survive for many weeks in a highly inflamed intestinal tract.

Twenty-three adults (only one male) and two fourth-stage larvae of Anisakis, recovered from the stomach of a Blainville's beaked whale (Mesoplodon densirostris) stranded in Galicia (NW Spain), were studied morphologically and molecularly. The polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) and sequencing studies carried out on the ribosomal DNA spacers and on the cox2 mitochondrial gene confirm that these nematodes belong to the taxon Anisakis sp. A, which was previously detected as a larval stage in fishes from Madeiran waters. In addition, our molecular studies demonstrate that Anisakis sp. A and the taxon Anisakis sp., previously found in other Mesoplodon spp., are the same species. The adults of Anisakis sp. A are morphologically similar to A. ziphidarum but with a shorter body length and longer spicules (right spicule 2.42 mm, left spicule 2.30 mm). Since the poor condition of the male tail cuticle prevented a proper description of the caudal plates and the pattern of caudal papillae, we propose to retain the name Anisakis sp. A until new males are described correctly.
