A schistosome infection is initiated when the parasite penetrates the skin of a susceptible host. Relatively large quantities of protein are released by transforming cercariae compared to later larval stages. This represents the first parasite material to which the host's immune system is exposed, yet little is known about the proteins which are released during the first few hours post-transformation. We have shown that antiserum raised against such molecules was capable of imparting protection against a schistosome challenge infection upon passive transfer to naïve mice. By screening a cercarial cDNA library with this serum, 38 positive clones were identified. Sequence analysis showed these to represent 8 different molecules which included Schistosoma mansoni 21·7 kDa antigen, calcium-binding-protein and the vaccine candidate glutathione S-transferase (Sm28GST). In addition, 5 clones were isolated, 1 of which had significant homology to many cytochrome C proteins, another with leukocyte elastase inhibitors and 3 which represented novel molecules. Four clones were expressed in a prokaryotic high-level expression vector, sera produced against each purified recombinant protein and used subsequently to probe Western blots and parasite sections. The leukocyte elastase inhibitor homologue and 2 unknowns induced significant proliferation by lymph node cells recovered from mice vaccinated with irradiated cercariae. More strikingly, the 2 novel proteins stimulated very high levels of interferon γ (IFNγ) secretion both by lymph node cells and those recovered by broncho-alveolar lavage from the lungs of vaccinated mice. Such results will be discussed in the context of vaccine development.

The in situ polymerase chain reaction (PCR) results revealed that mouse type A and type C retroviral sequences were transmitted horizontally from the host to schistosomes. The signals to these retroviral sequences were observed in the nuclei of the mesenchymal and reproductive cells of 8-week Schistosoma japonicum. These signals were also detected in the nuclei of the mesenchymal and reproductive cells and in the cytoplasm of the tegumental tubercles of 24-week S. mansoni. Furthermore, mouse type A retroviral sequence was detected in the DNA extracted from the cercariae of both species. However, mouse type C retroviral sequence and mouse type 2 Alu sequence (B2) were difficult to detect in the cercarial DNA of either species. These findings may indicate that some host sequences are propagated in the schistosome progeny, that is to say, not only horizontal but also vertical transfer of the host gene may occur in schistosomes.

Parasites should be better at infecting hosts from sympatric populations than allopatric populations most of the time (parasite local adaptation). In a previous study of a population of snail parasites (Microphallus sp.) from Lake Alexandrina, New Zealand, we found that Microphallus was more infective to snails (Potamopyrgus antipodarum) in shallow water but not in deep water. Here, we repeated the original study and also monitored the development of the parasite. We found that parasites from shallow water were more infective to hosts from shallow water and developed more rapidly in these hosts. In contrast, parasites from deep water were not more infective to hosts from deep water and did not develop more rapidly in them. These results suggest clinal variation in the susceptibility of these snails, with shallow-water snails more susceptible than deep-water snails. We offer 2 possible explanations for these results. First, gene flow in the Microphallus population is primarily from shallow to deep water, leading to an asymmetric pattern of local adaptation. Alternatively, snails from shallow water may be more susceptible for reasons independent of gene flow, perhaps due to differences in host condition between habitats.

Reproductive success of Gyrodactylus salaris from River Lierelva was compared experimentally on 3 stocks of salmon (12·5°C±0·2). Isolated fish from 2 susceptible Norwegian stocks (Rivers Lier and Alta) and 1 resistant Baltic stock of salmon (River Neva) were infected with a single gravid worm in order to record the temporal sequence of births and age at death of individual parasites. Establishment success (proportion of worms surviving to give birth) was generally low and mortality high, but significantly fewer worms survived on Neva (45% with mean survival of 3·5 days) compared to Alta and Lier fish (60%, mean survival 7·9 and 5·2 days, respectively). There was a dramatic difference in parasite fecundity between the host stocks: only 2 births occurred on Neva fish compared to third and fourth births on both Alta and Lier hosts. The timing of the first birth was more variable on Neva hosts and was significantly extended (mean 2·3 days) relative to that on Alta and Neva fish (1·8 days). However, timing of the second birth did not vary on any of the 3 salmon stocks. Age-specific mortality and fecundity data are consistent with exponential population growth of G. salaris on Alta and Lier fish but eventual extinction on Neva hosts. This is the first demonstration that gyrodactylids maintained on different host stocks exhibit variations in fecundity, development and mortality, which may in turn account for the variable virulence noted when G. salaris infects different salmonid hosts.

To determine whether rabbits may serve as reservoir hosts for Lyme disease spirochaetes in Europe, we compared their competence as hosts for Borrelia afzelii, one of the most prevalent European spirochaetal variants, with that of the Mongolian jird. To infect rabbits or jirds, at least 3 nymphal or adult Ixodes ricinus ticks infected with spirochaetes fed to repletion on each animal. Whereas jirds readily acquired tick-borne Lyme disease spirochaetes and subsequently infected vector ticks, rabbits exposed to tick-borne spirochaetes rarely became infectious to ticks. Only the rabbit that was infectious to ticks developed an antibody response. To the extent that I. ricinus ticks feed on European rabbits, these mammals may be zooprophylactic by diverting vector ticks from more suitable reservoir competent hosts.

Using the filamentous actin marker, FITC-conjugated phalloidin, the major muscle systems of adult male and female schistosomes have been examined. The body wall musculature comprises an outer sheath of circular fibres, within which there is a compact layer of short, spindle-shaped longitudinal fibres and a lattice-like arrangement of inner diagonal fibres. Within the oral sucker and acetabulum 3 fibre types, circular, radial and longitudinal can be distinguished. The wall of the oesophagus is lined by a grid-like array of circular and longitudinal fibres, whereas the walls of the intestinal caeca contain only comparably broad circular fibres. Within the female reproductive system, only circular fibres are present in the oviduct, vitelline duct and uterus. In contrast, the wall of the ootype displays closely arranged circular and longitudinal muscle fibres. Antisera to previously identified myoactive compounds (serotonin [5-hydroxytryptamine, 5-HT], neuropeptide F [Moniezia expansa] and GYIRFamide [Bdelloura candida, Dugesia tigrina]) were used as neuronal markers in a preliminary study of the spatial inter-relationships of specific nerve fibres and various muscle systems. Serotoninergic fibres innervate both suckers and also constitute a subtegumental nerve net. In males they provide innervation to the dorso-ventral muscle fibres of the gynaecophoric canal, and in females they innervate the circular and longitudinal muscle fibres of the ootype. Neuropeptide F and the FMRFamide-related peptide, GYIRFamide are both localized within nerve plexuses associated with the dorso-ventral fibres of the gynaecophoric canal, and are evident in the innervation of the ventral and oral sucker.

The hypothesis that intestinal helminth communities in freshwater brown trout are dissimilar in composition and structure to those in the European eel was tested by an analysis of component communities from 72 localities and of infracommunities from 34 localities in the British Isles and Norway. Derived indices were then compared with published data from eels. Composition of helminth communities differed considerably between the two hosts as a group of 4 species occurred commonly in trout and so gave greater predictability to the community composition. These 4 species were trout specialists and in 97% of the localities a trout specialist dominated the community rather than a generalist acanthocephalan as is typical for eels. By contrast all measures of community structure and indices of richness and diversity indicated that helminth communities in trout were isolationist in character, species poor and exhibited low diversity at both component and infracommunity levels. All values of indices for trout helminth communities were strikingly similar to those obtained from eels. Evidence of interspecific interactions within the trout helminth communities and a limit of 4 to infracommunity species richness further enhanced the similarities and suggested a common determinant of community structure. The hypothesis was thus supported in respect of species composition but refuted in respect of community structure.

An estimated 1049 million persons harbour T. trichiura, including 114 million preschool-age children and 233 million school-age children. The prevalence of T. trichiura is high and may reach 95% in children in many parts of the world where protein energy malnutrition and anaemias are also prevalent and access to medical care and educational opportunities is often limited. The Trichuris dysentery syndrome (TDS) associated with heavy T. trichiura, which includes chronic dysentery, rectal prolapse, anaemia, poor growth, and clubbing of the fingers constitutes an important public health problem, as do lighter but still heavy infections, even if not strictly TDS, especially in children. The profound growth stunting in TDS can be reversed by repeated treatment for the infection and, initially, oral iron. However findings from Jamaica strongly suggest that the significant developmental and cognitive deficits seen are unlikely to disappear without increasing the positive psychological stimulation in the child's environment. The severe stunting in TDS now appears likely to be a reaction at least in part to a chronic inflammatory response and concomitant decreases in plasma insulin-like growth factor-1 (IGF-1), increases in tumor necrosis factor-α (TNF-α) in the lamina propria of the colonic mucosa and peripheral blood (which likely decrease appetite and intake of all nutrients) and a decrease in collagen synthesis. Improvements in cognitive performance have been found after treatment for relatively heavy infections (without chronic dysentery) in school-going children; it is unclear precisely how much T. trichiura interferes with children's ability to access educational opportunities, but treatment of infections whenever possible is obviously sensible. The blood loss that can occur in T. trichiura infection is likely to contribute to anaemia, particularly if the child also harbours hookworm, malaria and/or has a low intake of dietary iron. Community control is important, particularly for the individuals within a population who harbour heavy worm burdens; this means children, with special attention to girls who will experience increased iron requirements and blood loss due to menstruation, pregnancies, and lactation. Mebendazole and albendazole, both of which are on the WHO Essential Drugs List, are very effective against T. trichiura; multiple doses are needed to attain complete parasitological cure in all cases. However the goal of control programmes in endemic areas is morbidity reduction, which follows when intensity of infection is significantly reduced.

This paper describes, for the human onchocerciasis focus of southern Venezuela, the age profiles of Onchocerca volvulus microfilarial (mf) and nodule prevalence, mf intensity, and mf aggregation for the whole examined population (836 Yanomami people) living in 20 villages, and for these communities classified according to endemicity levels (hypoendemic: [les ] 20%; mesoendemic: 21–59%; hyperendemic: [ges ] 60% infected). Mf prevalence and intensity increased with age, particularly in the hyperendemic areas, and there were no marked differences between the sexes. The prevalence of nodules followed the same age pattern. Fifty percent mf prevalence was reached in the 15–19 year age-class when the population was taken as a whole; nearly in the 10 to 14-year-olds for the hyperendemic level, in those aged 20–29 years in mesoendemic areas, and not reached at all in hypoendemic villages. The degree of mf aggregation was measured by the k value of the negative binomial distribution and by the variance to mean ratio (VMR). The relationship between the standard deviation (S.D.) of mf counts and the mean mf density was also explored. These 3 indices (k, VMR, and S.D.) showed a tendency to increase with both mean mf load and host age. Since infection intensity and host age were themselves positively related, it was not possible to draw definite conclusions about age-specific changes of parasite aggregation. There was not a significant decrease of mf intensity after an earlier peak neither was there a shift towards younger ages of the maximum no. of mf/mg reached as the endemicity level increased. These results are discussed in relation to detection of density dependence in the human host, selection of an indicator age-group for rapid epidemiological assessment (REA) methods, and strategies of ivermectin distribution in the Amazonian focus. It is recommended that, for the Amazonian onchocerciasis focus, the indicator group for REA consists of all those aged 15 years and over.

Ecological factors may influence the number of parasites encountered and, thus, parasite species richness. These factors include diet, gregarity, conspecific and total host density, habitat, body size, vagility, and migration. One means of examining the influence of these factors on parasite species richness is through a comparative analysis of the parasites of different, but related, host species. In contrast to most comparative studies of parasite species richness of fish, which have been conducted by using data from the literature, the present study uses data obtained by the investigators. Coral reef fishes vary widely in the above ecological factors and are frequently parasitized by a diverse array of parasites. We, therefore, chose to investigate how the above ecological factors influence parasite species richness in coral reef fishes. We investigated the endoparasite species richness of 21 species of butterfly fishes (Chaetodontidae) of New Caledonia. We mapped the diet characters on the existing butterfly fish phylogeny and found that omnivory appears to be ancestral. We also mapped the estimated endoparasite species richness, coded from low to high parasite species richness, on the existing butterfly fish phylogeny and found that low parasite species richness appears to be associated with the ancestral state of omnivory. Different dietary and social strategies appear to have evolved more than once, with the exception of obligate coralivory, which appears to have evolved only once. Finally, after controlling for phylogenetic relationships, we found that only the percentage of plankton in the diet and conspecific host density were positively correlated with endoparasite species richness.

This paper presents a first attempt at modelling the possible cost and effectiveness of reaching non-enrolled children through school-based programmes using empirical data from Egypt. A sex/school-attendance/age-structured population dynamic model was used to predict trends in infection and early disease. Four treatment delivery strategies were compared: school-based (coverage of 85%) and school-aged targeted (coverage of 25, 50 and 85%). The school-aged targeted strategies also included the school-based programme. For each alternative strategy, the maximum unit cost was calculated to obtain a cost-effectiveness ratio equal or smaller to the one obtained with the school-based programme (unit cost of US$ 0·60). The analysis showed that, for S. mansoni in Lower Egypt, a programme where only 85% of children attending school were treated would still prevent 77% of the early disease cases prevented with a programme where 85% of all school-age children were treated. However, using the school-aged targeted strategy, from US$ 0·06 to US$ 1·03 extra unit costs could be spent to reach non-enrolled children and still be more cost-effective. Treating non-enrolled children is an important consideration in maximizing the effectiveness of treatment programmes while maintaining a cost-effectiveness comparable to school-based delivery.

In the previous paper it was concluded that those aged [ges ] 15 years of both sexes could comprise the indicator group for rapid epidemiological assessment (REA) of onchocerciasis in the Amazonian focus. This paper explores relationships between community microfilarial (mf) prevalence, intensity, and nodule prevalence in 20 Yanomami communities, that would allow identification of REA methods in the region. The mean nodule ratio (prevalence of nodules/prevalence of mf) was 0·54 when onchocercomata in the indicator group were considered. The Spearman correlation coefficient between mf and nodule prevalence was 0·686 (P = 0·001). Palpation of nodules had 92% specificity and 32% sensitivity when compared to skin-snipping for the diagnosis of onchocerciasis. The predictive value positive increased from 75% to 81% when the indicator group was used. A microfilarial prevalence > 75% in this group would be indicative of hyperendemic status in the village, between 30 and 75% of mesoendemicity, and < 30% of hypoendemicity. For the assessment of infection intensity, biopsies may be taken from the iliac crest for all endemicity levels. Five of the hyperendemic villages surveyed in this work had a community microfilarial load (CMFL) greater than 10 mf/skin snip; the remaining 5 had a CMFL between 5 and 9. These levels of infection merit high priority ivermectin treatment. In Latin America, communities at both moderate and severe risk are included in mass chemotherapy programmes (i.e. when mf prevalence is over 20%). Roughly, a nodule prevalence in the indicator group > 10% would suggest a community mf prevalence > 20% with a sensitivity of 85% and a specificity of 71%. A multiple linear regression model of the arc-sine transformed mf prevalence in the village (all ages) on nodule prevalence in those aged [ges ] 15 years and altitude of the village explained 72% of the variance. The model combining nodule and altitudinal information had a sensitivity of 92% and a specificity of 71% in comparison to an estimated mf prevalence of 21% or more. It is suggested that the usefulness of the REA methods proposed be assessed in other areas of the Amazonian onchocerciasis focus.

Resistance to the murine intestinal nematode Trichuris muris requires the development of a strong Th2 response. In a reconstituted SCID mouse model, CD4+ Th2 cells can mediate resistance to infection in the absence of antibody (Else & Grencis, 1996). The data presented here address the issue of how CD4+ T cells mediate this protective immunity within the SCID host. These studies demonstrate that timing and cell dose are critical if transfer is to result in resistance, with a minimum of 5 × 106 immune donor cells required to confer immunity. Furthermore, this CD4-mediated protective immunity only operates against the larval stages of the parasite. When the molecules necessary for activated CD4+ T cell migration to the GALT are inhibited with a cocktail of anti-integrin/addressin antibodies (anti-β7, anti-MAdCAM-1 and anti-αE), the resistance conferred by immune donor cells is completely abrogated. This implies that the effector mechanism acts locally at the level of the gut. CD4+ mediated cytotoxicity, directed against the epithelial cells inhabited by the parasite, could represent a novel, locally acting effector mechanism. However, Fas and Fas ligand-deficient mice, which are unable to mount CD4-mediated cytotoxic responses, readily expel T. muris indicating that the mechanism by which CD4+ T cells mediate protective immunity is unlikely to involve killing of infected gut epithelial cells.

Isozyme analysis (12 enzymes: 14 loci) was conducted on 99 isolates of Trypanosoma cruzi: 77 from Guatemala, 5 from Mexico and 17 from South American countries. Analyses of 4 population-genetic indices were undertaken to assess the possibility of genetic exchange occurring among Guatemalan isolates. The results provide evidence for a degree of genetic exchange occurring among isolates from this relatively small geographical area. Previous studies of population genetics on T. cruzi might have failed to detect this phenomenon because they tended to use isolates originating far from one another, rendering gene exchange unlikely for geographical reasons. Phylogenetic data, presented here, show considerable differences in genetic structure between Central and South American isolates, suggesting that different biological and clinical properties might be expected. For example, there are differences in clinical syndromes between Central and South America, a situation discussed further here.

In this paper we discuss several established and potential animal models for human parasitic infection, with a focus on rodent, pig and primate models and the nematodes Ascaris, Trichuris and Toxocara spp. Firstly, we discuss the relevance of choosing a suitable animal host to fit the particular study hypothesis, and the interaction between mathematical modelling and animal models. Secondly, we review the use of animal models for the study of nutrition-parasite interaction, evaluation of treatment and control strategies, and bacteria-parasite interactions. We show that rodent, pig and primate models are all very useful in parasitological research, and that each model has its limitations. However, based on recent experience with the pig-Ascaris and pig-Trichuris models, a more extensive use of the pig-parasite model is advocated, especially for the study of the interaction between human malnutrition and helminth infection, and congenital helminth infection.

Eggs of Neoheterocotyle rhinobatidis, Troglocephalus rhinobatidis and Merizocotyle icopae (Monogenea: Monocotylidae) from the shovelnose ray Rhinobatos typus (Rhinobatidae) have a distinct hatching pattern linked to light periodicity. Larvae of these 3 monogenean species emerge only during daylight when exposed to natural illumination or when incubated in alternating 12 h periods of light and dark (light on 06.00 h, light off 18.00 h). N. rhinobatidis larvae emerge with a distinct peak during the first 2 h of light; this peak is not as pronounced in T. rhinobatidis or M. icopae. Eggs of N. rhinobatidis incubated in a reverse light/dark cycle (light on 18.00 h, light off 06.00 h) hatched only during periods of illumination, again with a peak during the first 2 h of light. Evidence suggests that the hatching patterns observed in all 3 species represent true circadian rhythms because eggs incubated in 24 h light or 24 h dark conditions continued to hatch with a rhythm. Shadows, disturbance and host tissue did not promote hatching in N. rhinobatidis or T. rhinobatidis but there were indications that host tissue may promote hatching in M. icopae. The hatching patterns observed are discussed with respect to their adaptive responses to host behaviour and predation pressure.

This comprehensive review briefly describes the history and pharmacology of albendazole as an anthelminthic drug and presents detailed summaries of the efficacy and safety of albendazole's use as an anthelminthic in humans. Cure rates and % egg reduction rates are presented from studies published through March 1998 both for the recommended single dose of 400mg for hookworm (separately for Necator americanus and Ancylostoma duodenale when possible), Ascaris lumbricoides, Trichuris trichiura, and Enterobius vermicularis and, in separate tables, for doses other than a single dose of 400mg. Overall cure rates are also presented separately for studies involving only children 2–15 years. Similar tables are also provided for the recommended dose of 400mg per day for 3 days in Strongyloides stercoralis, Taenia spp. and Hymenolepis nana infections and separately for other dose regimens. The remarkable safety record involving more than several hundred million patient exposures over a 20 year period is also documented, both with data on adverse experiences occurring in clinical trials and with those in the published literature and/or spontaneously reported to the company. The incidence of side effects reported in the published literature is very low, with only gastrointestinal side effects occurring with an overall frequency of just >1%. Albendazole's unique broad-spectrum activity is exemplified in the overall cure rates calculated from studies employing the recommended doses for hookworm (78% in 68 studies: 92% for A. duodenale in 23 studies and 75% for N. americanus in 30 studies), A. lumbricoides (95% in 64 studies), T. trichiura (48% in 57 studies), E. vermicularis (98% in 27 studies), S. stercoralis (62% in 19 studies), H. nana (68% in 11 studies), and Taenia spp. (85% in 7 studies). The facts that albendazole is safe and easy to administer, both in treatment of individuals and in treatment of whole communities where it has been given by paramedical and nonmedical personnel, have enabled its use to improve general community health, including the improved nutrition and development of children.

In order to provide a better understanding of the interaction between the liver fluke (Fasciola hepatica) and the immune system of its mammalian host immunoreactive λ bacteriophage clones containing F. hepatica cDNA have been isolated. Plasmids from these clones were sequenced and found to encode a family of proteins containing certain common elementsThe nucleotide sequence data reported in this paper are available in the GenBank database with the accession number AF153056.. All the clones contained a coding repeating sequence (RRRXCA) which is conserved at the nucleic acid level followed by a non-repeating element coding for the C terminal used by the proteins which shows conservation of amino acids at certain positions. Antisera raised against a β-galactosidase fusion protein with one of these sequences as a terminal extension was used to localize the immunoreactive antigens. Binding was predominantly in the tegument of the juvenile fluke but was reduced in the adult tegument. The wall of the uterus showed strong reactivity in the adult. Rats immunized with the β- galactosidase fusion protein showed enhanced resistance to challenge infections. The role of these antigens in the host response to infection by F. hepatica is discussed.

Host tissue penetration, feeding and immune evasion by helminth parasites may be mediated by both mechanical processes and histolytic products released by the parasite. The aim of this study was to investigate potential histolytic products released during in vitro maintenance of exsheathed third (L3) and 4th larval stage (L4) and adult Ostertagia ostertagi. Therefore, the pH optima, substrate specificity, molecular size and inhibitor sensitivity of in vitro released (IVR) proteinases were analysed by spectrophotometry and substrate gel electrophoresis. It was shown that L3, L4 and adult IVR proteinases degrade a variety of protein substrates in a pH-dependent and stage-specific manner. At alkaline pH, gelatin, casein and fibrinogen were degraded by metallo- and serine proteinases. In contrast, mucin, fibrinogen, albumin and haemoglobin were degraded at acidic pH by aspartyl protease- and cathepsin L-like activity. At pH 3, the heavy chain of bovine IgG was completely degraded by an aspartyl proteinase secreted by all 3 parasitic stages. The specificity of the L3, L4 and adult Ostertagia ostertagi proteinases against the different substrates indicates variable functional requirements.

The present study describes a novel heat-stable, water-stress-related protein with a molecular mass of 47 kDa (designated Desc47) in the entomopathogenic nematode Steinernema feltiae (IS-6). The protein was accumulated about 10-fold (from 7·84±1·85 to 74·09±4·35% relative content level [RCL]) in dehydrated clumps of infective juveniles (IJs), which had lost 34·4% of their initial water content (from 65·1±1·7% to 42·7±0·72%) in a desiccation-tolerance-inducing treatment (97% relative humidity [RH] for 3 days). The appearance of Desc47 was accompanied by trehalose accumulation (from 300 to 600 mg trehalose/g protein) during the process of inducing the IJs into a quiescent anhydrobiotic state. A second cycle of IJ dehydration did not alter the RCL of Desc47 (79·3% for the first cycle and 73·3% for the second cycle). Desc47 retained its high RCL (69·7%) in rehydrated active IJs for 3 days, reaching 51·2% of its initial RCL only after a week. No homology to other known proteins was found by mass-spectrometry electrospray-ion-trap analysis. However, of the 5 sequences obtained from the protein (ranging from 11 to 21 amino acids), the 21-amino-acid peptide NVASDAVETVGNAAGQAG(D/T)AV showed excellent homology (74% identity in 19 amino acids) to the cold-responsive protein COR14b (g6564861) from Triticum aestivum. In the Caenorhabditis elegans predicted proteome database search, the N21 yielded the first-best identity score (59% identity in 17 amino acids) to the CE-LEA homologue protein (g2353333). In plants, COR and LEA are related proteins, heat-stable, which are expressed in response to both dehydration and cold acclimation. The implication of the involvement of Desc47 and the osmoprotectant trehalose in the desiccation-tolerance mechanisms of S. feltiae is discussed.
