It is relatively infrequent these days for a novel mineral to be discovered in the natural environment (in this context, this implies on the Earth). On the rare events of such a discovery, the new mineral is typically found in milligram quantities. In an article by Ronald Peterson, William Nelson, Bruce Madu, and Herbert Shervell, they describe the discovery of kilograms of a mineral that had only been synthesized previously, but never before detected in the natural environment. As if that were not impressive enough, they went looking for this mineral because they thought it had been observed on Mars!

The rapid technical development of FIM (Focused Ion Beam) technology has spawned an increase in spatial resolution capability in scanning ion microscopy (SIM) technology. Furthermore, FIM has been used for preparation of thin specimens in transmission electron microscopy and micro-fabrication of electronic devices in the semiconductor industry. Recently, a scanning ion microscope with a helium field ion source has been developed. Thus, the contrast formation of emission electron images in scanning ion microscopy has been the object of study for analyzing images of materials specimens, similar to the theory behind scanning electron microscope (SEM) contrast formation. Furthermore, whether the electron emission yield γ induced by ion impact is periodic or non-periodic as a function of Z2 (the atomic number of the target) has not been well studied in the low energy region from several keV to the several tens of keV values used in SIM.

High-content screening (HCS) and other quantitative cellular imaging assay methods that use fluorescence microscopy require effective fluorescent labeling and identification of the target cell(s). Fluorescent probes that stain cells as the primary objects are used to identify and count individual cells, as well as to define the region(s) of each cell to which target-specific image analysis is applied. For this purpose, the primary object can be a major component of the cell, such as the nucleus or a large organelle, or the whole cell itself. When the whole cell is the primary object, a high-quality cell stain is needed to delineate the intact cell from bordering cells without also interfering in target-specific detection and analysis. Thermo Scientific Cellomics ® Whole Cell Stains (Thermo Fisher Scientific Inc., Rockford, Ill.), available in blue, green, orange and red, provides image staining and the ability to quantify the whole cell volume in HCS and fluorescence microscopy assays.

Scanning electron microscopes have traditionally been used for observation and microanalysis of samples. Positioning and testing of samples has usually been performed out of the SEM chamber e.g. electrical test benches, sample preparation. However, due to miniaturization in semiconductor technology, optics, micro-mechanics, medicine, gene- and biotechnology, highly precise positioning techniques are becoming increasingly important. This may be performed using an optical microscope, or more commonly, within the SEM chamber itself.

In the earliest years of microscopy, it was well understood that visualization of the finest details was often limited by the ‘intrusion’ of light from out-of-focus planes. For many decades the only solution to this dilemma was to limit microscopic studies to very thin samples or to physically squash or slice thicker samples. More recently, biomedical research has driven the need for microscopes that can resolve very fine detail in 3 dimensions within intact, and often living, specimens. The use of fluorescence labeling further exacerbates the problem of out-of-focus light because signal is generated throughout the volume of the sample. The following article describes the most prevalent techniques for 3D imaging and important optical considerations for achieving highest image quality.

Automated scanning transmission electron microscopy (STEM) metrology provides critical dimension (CD) measurements an order of magnitude more precise than comparable scanning electron microscopy (SEM) measurements. New developments in automation now also provide throughput and response time sufficient to support high volume microelectronic manufacturing processes. The newly developed methodology includes automated, focused ion beam (FIB) based sample preparation; innovative, ex-situ sample extraction; and automated metrology. Although originally developed to control the production of thin film magnetic heads for data storage, the technique is fully applicable to any wafer-based manufacturing process.

Coupling between electrical and mechanical phenomena is extremely common in inorganic materials, and nearly ubiquitous in biological systems, underpinning phenomena and devices ranging from SONAR to cardiac activity and hearing. This paper briefly summarizes the Scanning Probe Microscopy (SPM) approach, referred to as Piezoresponse Force Microscopy (PFM), for probing electromechanical coupling on the nanometer scales, and delineates some existing and emerging applications to probe local structure and functionality in inorganic ferroelectrics, calcified and connective tissues, and complex biosystems based on electromechanical detection.

Automated Crystal Orientation Microscopy (ACOM) on a grain specific level has proved to be an invaluable new tool for characterizing polycrystalline materials. It is usually based on scanning facilities using electron diffraction , due to its high sensitivity and spatial resolution, but also attempts have been made which rely upon X-ray or hard synchrotron radiation diffraction. The grain orientations are commonly mapped in pseudo-colors on the scanning grid to construct Crystal Orientation Maps (COM), which represent “images” of the microstructure with the advantage of providing quantitative orientation contrast. In a similar way, misorientations across grain boundaries, Σ values of grain boundaries, or other microstructural characteristics are visualized by mapping the grains in the micrograph with specific colors. The principal objectives are the determination of quantitative, statistically meaningful data sets of crystal orientations, misorientations, the CSL character (Σ) of grain boundaries, local crystal texture (pole figures, ODF, MODF, OCF) and derived entities, phase discrimination and phase identification.

Preparation for TEM of resin-embedded monolayer cell cultures usually requires scraping cells from the culture substrate before or after primary fixation, which disturbs the monolayer and often results in mechanical trauma to the cells. After harvest, the cells are centrifuged and processed as a pellet in a microcentrifuge tube through a prolonged procedure of post-fixation, dehydration, infiltration, and finally embedding to ensure a “well done” sample block for subsequent sectioning, staining, and observing under a transmission electron microscope. Other disadvantages to this method include the loss of cellular orientation and information on cellular interaction, as well as difficulty in collecting a large quantity of cells to form a sizable pellet for processing, which is of special importance when samples are differentiated cells and neurons cultured in low density. In an alternative method, cells can be seeded on either glass or plastic cover slips or Petri dishes, and then processed and embedded directly as a monolayer.

The application of nanotechnology in disciplines as varied as medicine and electronics is advancing rapidly with carbon nanoparticles (CNPs) such as fullerenes (C60) and nanotubes at the forefront. However, a lack of understanding of the interaction of such small structures with cellular material has resulted in concerns over their impact on human health and since the individual structures have a diameter of ~1 nm they are potentially small enough to penetrate through ion channels or diffuse through pores in the nuclear membrane. Assessing their toxicity is imperative. In response to these concerns there has been an increase in the number of papers addressing the toxicity of carbon nanoparticles over the last few years but much of this data appears contradictory. It is therefore essential to understand how the human body interacts with CNPs and more specifically to elucidate pathways by which CNPs enter the cell and their distribution within.

2D-crystallization in combination with transmission electron microscopy (TEM) is one of the few methods for the structural analysis of membrane proteins in their native state. However, the parameters for the generation of large crystalline sheets are typically difficult to identify for a given protein. Many repetitive and time consuming screening steps by TEM are therefore necessary to find the best crystallization and preparation conditions. Although several software packages offer the possibility to control an electron microscope, none is completely adapted for a fully automated and completely integrated acquisition and analysis of 2D crystals. Here we report on the development of a fully automatic screening and on-line analysis software for the fast and automatic survey of large quantities of negatively stained EM samples for 2D crystallography.
