The use of an alkane mixture that remains liquid at 77 K to freeze specimens has advantages over the use of a pure alkane that is solid at 77 K. It was found that a mixture of methane and ethane did not give a cooling rate adequate to produce vitreous ice, but a mixture of propane and ethane did result in vitreous ice. Furthermore, the latter mixture produced less damage to specimens mounted on a very thin, fragile holey carbon substrate.

Roughness increases significantly after finishing procedures. The aim of this study was to assess by the atomic force microscope (AFM) the effect of finishing instruments on the surface roughness of composite resins. A nanofiller composite resin (Filtek Supreme, 3M–F) and a microhybrid composite resin (Point 4, Kerr–P) were selected. The finishing procedures were done with a 30-blade carbide bur (C) and a 30-μm finishing diamond bur (D). Standardized specimens were produced and divided into six experimental groups (n = 4) according to (1) composite resin, (2) absence of finishing (Mylar matrix–M), and (3) finishing instrument (FM, PM, FC, FD, PC, PD). The mean surface roughness was evaluated by AFM in the contact mode. FM and PM groups were assessed statistically by the Student's T test, and FC, FD, PC, PD groups were submitted to variance analysis (ANOVA), both at 5% significance. The mean surface roughness values, in nanometers, were FM, 23.63 (b); FC, 283.88 (c); FD, 510.55 (d); PM, 12.52 (a); PC, 343.98 (c); PD, 531.64 (d). Microhybrid composite displayed less roughness than nanofiller composite in the absence of finishing procedures. The 30-blade carbide bur produced less roughness compared to the extra fine diamond bur.

Intervessel pits play a key role in trees' water transport, lying at the base of drought-induced embolism, and in the regulation of hydraulic conductivity via hydrogels bordering pit canals. Recently, their microstructure has been the focus of numerous studies, but the considerable variation, even within species and the histochemistry of pit membranes, remains largely unexplained. In the present study, intervessel pits of the outermost wood were examined for Avicennia marina, of dry and rainy season wood separately for Rhizophora mucronata. The thickness of the pit membranes was measured on transmission electron micrographs while their topochemical nature was also analyzed via cellular UV microspectrophotometry. Pit membranes of R. mucronata were slightly thicker in dry season wood than in rainy season wood, but their spectra showed for both seasons a lignin and a yet unidentified higher wavelength absorbing component. It was suggested to be a derivative of the deposits, regularly filling pit canals. The vestures of A. marina chemically resembled pit membranes rather than cell walls.

Guiera senegalensis J. F. Gmel. (Combretaceae) is one of the most important West African medicinal plants, often used to treat a variety of microbial infections. The most frequently used plant part is the leaf, its medicinal use being corroborated by several in vitro antimicrobial activity studies. However, quality criteria for pharmaceutical use, including botanical identification, are not yet determined. Aiming the establishment of such criteria, the present work deepens today's knowledge on G. senegalensis leaf morphology and anatomy, the anatomical characters of both leaf transversal section and powdered leaves being hereby presented for the first time. The most useful characters for identification purposes are leaf isobilateral organization, with similar upper and lower surfaces and a palisade parenchyma on both surfaces; trichome insertion points on both epidermis, surrounded by polar arrangements of cells; tomentose indumentum on the upper epidermis consisting of compartmented and uniseriated trichomes with long, curved, or straight terminal cells; brown scales, isolated or inserted into the epidermis, formed by radially arranged cells surrounding a central cell, with a more or less circular form and a conspicuous base; calcium oxalate cluster crystals, isolated or inserted into the palisade parenchyma and absent in lateral veins.

A series of Polynesian pearls has been investigated with particular attention to the structural and compositional patterns of the early developmental stages of the pearl layer. These initial steps in pearl formation bear witness of the metabolic changes that have occurred during the pearl-sac formation. The resulting structurally and biochemically complex structures have been investigated using a variety of techniques that provide us with information concerning both mineral phases and the organic components. Results are discussed with respect to our understanding of the biomineralization mechanisms, as well as for the grafting process.

We previously reported that translocation of mitochondria from the oocyte cortex to the perinuclear area indicates positive developmental potential that was reduced in porcine somatic cell nuclear transfer (SCNT) embryos compared to in vitro–fertilized (IVF) embryos (Katayama, M., Zhong, Z.-S., Lai, L., Sutovsky, P., Prather, R.S. & Schatten, H. (2006). Dev Biol299, 206–220.). The present study is focused on distribution of donor cell mitochondria in intraspecies (pig oocytes; pig fetal fibroblast cells) and interspecies (pig oocytes; mouse fibroblast cells) reconstructed embryos by using either pig fibroblasts with mitochondria-stained MitoTracker CMXRos or YFP-mitochondria 3T3 cells (pPhi-Yellow-mito) as donor cells. Transmission electron microscopy was employed for ultrastructural analysis of pig oocyte and donor cell mitochondria. Our results revealed donor cell mitochondrial clusters around the donor nucleus that gradually dispersed into the ooplasm at 3 h after SCNT. Donor-derived mitochondria distributed into daughter blastomeres equally (82.8%) or unequally (17.2%) at first cleavage. Mitochondrial morphology was clearly different between donor cells and oocytes in which various complex shapes and configurations were seen. These data indicate that (1) unequal donor cell mitochondria distribution is observed in 17.2% of embryos, which may negatively influence development; and (2) complex mitochondrial morphologies are observed in IVF and SCNT embryos, which may influence mitochondrial translocation and affect development.

Crystalline properties of synthetic nanostructured hydroxyapatite (n-HA) were studied using high-resolution transmission electron microscopy. The focal-series-restoration technique, obtaining exit-plane wavefunction and spherical aberration-corrected images, was successfully applied for the first time in this electron-beam-susceptible material. Multislice simulations and energy dispersive X-ray spectroscopy were also employed to determine unequivocally that n-HA particles of different size preserve stoichiometric HA-like crystal structure. n-HA particles with sizes of twice the HA lattice parameter were found. These results can be used to optimize n-HA sinterization parameters to improve bioactivity.

Experimental low-loss electron (LLE) yields were measured as a function of loss energy for a range of elemental standards using a high-vacuum scanning electron microscope operating at 5 keV primary beam energy with losses from 0 to 1 keV. The resulting LLE yield curves were compared with Monte Carlo simulations of the LLE yield in the particular beam/sample/detector geometry employed in the experiment to investigate the possibility of modeling the LLE yield for a series of elements. Monte Carlo simulations were performed using both the Joy and Luo [Joy, D.C. & Luo, S., Scanning11(4), 176–180 (1989)] expression for the electron stopping power and recent tabulated values of Tanuma et al. [Tanuma, S. et al., Surf Interf Anal37(11), 978–988 (2005)] to assess the influence of the more recent stopping power data on the simulation results. Further simulations have been conducted to explore the influence of sample/detector geometry on the LLE signal in the case of layered samples consisting of a thin C overlayer on an elemental substrate. Experimental LLE data were collected from a range of elemental samples coated with a thin C overlayer, and comparisons with Monte Carlo simulations were used to establish the overlayer thickness.

High-resolution transmission electron microscopy and electron energy loss spectroscopy (EELS) were performed on electrochemically anodized niobium and niobium oxide. Sintered anodes of Nb and NbO powders were anodized in 0.1 wt% H3PO4 at 10, 20, and 65 V to form surface Nb2O5 layers with an average anodization constant of 3.6 ± 0.2 nm/V. The anode/dielectric interfaces were continuous and the dielectric layers were amorphous except for occurrences of plate-like, orthorhombic pentoxide crystallites in both anodes formed at 65 V. Using EELS stoichiometry quantification and relative chemical shifts of the Nb M4,5 ionization edge, a suboxide transition layer at the amorphous pentoxide interface on the order of 5 nm was detected in the Nb anodes, whereas no interfacial suboxide layers were detected in the NbO anodes.

The molecular distribution in nanocolloids of poly(dimethyl siloxane) (PDMS) and an organic copolymer (methyl acrylate co-methyl methacrylate co-vinyl acetate) preserved in a frozen aqueous solution was investigated using cryovalence electron energy-loss spectroscopy (EELS) coupled with a scanning transmission electron microscope. Low energy-loss spectra depend upon valence electron structure, and we show that they are substantially different for the PDMS, the copolymer, and the vitrified water studied here. Combining a high efficiency detection system and the use of high-signal low-loss spectra in EELS, we achieved a spatial resolution of 8 nm without serious beam-induced specimen damage in this radiation-sensitive soft-materials system. To obtain quantitative phase maps of silicone and copolymer composition within individual nanoparticles, spectrum datasets were processed via multiple least squares fitting. Quantitative line profiles from the resulting compositional maps indicate that the PDMS lobe of biphasic nanoparticles contained a significant amount of the copolymer and a diffuse interface was formed. Since the nanoparticle synthesis involves polymerization of acrylate monomer dissolved in PDMS nanoparticle precursors, these results suggest that the evolution of the nanocolloid morphology during synthesis is kinetically frozen as the acrylate copolymer achieves some critical molecular weight.

The ability of electron microscopes to analyze all the atoms in individual nanostructures is limited by lens aberrations. However, recent advances in aberration-correcting electron optics have led to greatly enhanced instrument performance and new techniques of electron microscopy. The development of an ultrastable electron microscope with aberration-correcting optics and a monochromated high-brightness source has significantly improved instrument resolution and contrast. In the present work, we report information transfer beyond 50 pm and show images of single gold atoms with a signal-to-noise ratio as large as 10. The instrument's new capabilities were exploited to detect a buried Σ3 {112} grain boundary and observe the dynamic arrangements of single atoms and atom pairs with sub-angstrom resolution. These results mark an important step toward meeting the challenge of determining the three-dimensional atomic-scale structure of nanomaterials.
