Mongolian gerbils (Meriones unguiculatus) are a widely used model species in parasitology and epilepsy research. Under standard laboratory housing conditions gerbils develop stereotypic digging, a behavioural abnormality, which is caused by the lack of a burrow in the laboratory cage. Here we show that gerbils reared with access to an opaque artificial burrow (nest-box with access tube) develop significantly less stereotypic digging than those reared with transparent artificial burrows. Subadult gerbils also preferred opaque artificial burrows to transparent ones. Based on these findings we developed an artificial burrow system that could be integrated into laboratory Makrolon Type IV cages to prevent the development of stereotypic digging in gerbils by addressing their behavioural needs. Faecal cortisol levels were measured as a non-invasive method of comparing stress reactions in gerbils given access to the new integrated artificial burrow system or to an equivalent transparent burrow. Behavioural differences were observed between gerbils in the two housing conditions, but faecal cortisol levels were unaffected. We conclude that simple refinement of housing is effective in improving gerbil welfare in the laboratory.

The early development of bar-gnawing was studied in young gerbils kept in standard laboratory cages. Bar-gnawing was performed on the bars of the cage lid and developed shortly after eye-opening at an age of 18 days. It increased in bout length and duration with age and there was considerable variability in behaviour between individuals. It is discussed as to whether bar-gnawing is a stereotypy and in which behavioural context it develops.

The presence of the prostate in female mammals has long been known. However, pieces of information related to its development are still lacking. The aim of this study was to explore the budding dynamic during the initial prostate development in female gerbils. Pregnant females were timed, the fetuses were euthanized, and the urogenital sinus was dissected out between the embryonic days 20 and 24 (E20–E24 groups). Newborn pups (1-day-old; P1 group) underwent the same procedures. The female prostate development was based on epithelial buds which arose far from the paraurethral mesenchyme (PAM). The epithelial buds reached the PAM at prenatal day 24, crossing a small gap in the smooth muscle layer between the periurethral mesenchyme (PEM) and the PAM. Steroid nuclear receptors such as the androgen receptor and estrogen receptor alpha were localized in the PEM through the urethral wall, although some epithelial labeling was also present in the urogenital sinus epithelium (UGE). P63-positive cells were found only in the UGE, becoming restricted to the basal compartment after the 23rd prenatal day. The results showed that the gerbil female prostate exhibits a distinct budding pattern as compared to the male prostate development.

Transplanted infections of Dipetalonema viteae and Brugia pahangi have been evaluated as tools for experimental chemotherapy. Attempts were made to establish these filariae in similar pharmacokinetic sites within the same host, so that direct comparisons of in vivo drug susceptibilities could be made. Unfortunately, it was not possible to establish B. pahangi in the subcutaneous tissues, the preferred site of D. viteae. Therefore, intraperitoneal B. pahangi and subcutaneously implanted D. viteae in gerbils were used for the study. D. viteae infections were significantly enhanced by concomitant infections with B. pahangi, while B. pahangi infection rates were unaffected by the presence of D. viteae. Experiments with amoscanate, CGP6140 and Mel W demonstrated the importance of employing both B. pahangi and D. viteae for antifilarial discovery work and the fundamental effect of parasite location on drug efficacy. D. viteae rapidly migrate from the peritoneal cavity of gerbils following implantation; twenty one hours after infection 73% of transplanted worms were found in the subcutaneous tissues. It was shown that the migration response could be used as a stringent parameter for demonstrating antifilarial activity. D. viteae were exposed to antifilarial drugs for 24 hours in vitro, washed and implanted into the peritoneal cavity of gerbils. At autopsy, 5 days later, 10−8M ivermectin and milbemycin D had prevented migration; CGP6140, amoscanate, suramin, flubendazole and furapyrimidone were also detected at <10−6M using this parameter. In all cases the migration response was more sensitive to drugs than parasite kill. Ivermectin's ability to inhibit worm migration through the tissues is discussed, with respect to the role of itinerant males in the reproductive cycle of Onchocerca volvulus.

Gerbils with well developed peritoneal cysts of Echinococcus granulosus were randomized to albendazole 50 mg/kg/day or untreated control. Treated animals had less disease at post mortem after 3 months of treatment. Cysts were then taken from both albendazole-treated and control animals and cultured in vitro either with or without albendazole sulphoxide (Alb Sx) 500 μg/L for 14 days. Viability of cysts was then established by implantation of whole cysts into gerbils. Whilst naive cysts were affected by Alb Sx (only 2 cysts developed/gerbil) cysts from animals treated with albendazole were not sensitive to further therapy (6.4 cysts/gerbil).

Peritoneal Echinococcus granulosus in gerbils was treated with albendazole. Both early and late infections were studied; response to albendazole therapy and the ability of the parasite to recover after treatment was found to depend on dose and length of therapy. Even after treatment at 50 mg/kg for 2 months late infections retained the ability to recover over 3 months.

The present report compares the macrophage function in rodent hosts susceptible and resistant to the human lymphatic filariid Brugia malayi. Macrophages from both mastomys (resistant) and gerbil (susceptible) infected intraperitoneally (i.p.) with the infective larvae (L3) of B. malayi were isolated from peritoneal lavage at different time-intervals and formation rate of NO, H2O2, O2−, TNF-α, glutathione peroxidase and reductase was assayed. NO release was found to be significantly increased in resistant mastomys as compared to gerbils and the release was markedly suppressed by i.p. administration of the NOS inhibitor aminoguanidine (AG). The AG-treated mastomys also demonstrated significantly greater establishment of larvae which correlated well with suppressed formation of NO. Nitric oxide synergizes with superoxide to form peroxynitrite radical (potent oxidant), which is known to be more toxic per se than NO. Results indicate the possible involvement of peroxynitrite in the rapid killing of larvae in the peritoneal cavity of mastomys. In contrast, the production of H2O2 was found to be enhanced in both species indicating that B. malayi L3 could withstand the toxic effects of H2O2. The higher level of glutathione peroxidase and reductase, as observed in mastomys compared with the gerbil after larval introduction, possibly protects the cell against the injurious effect of H2O2. The TNF-α level remained virtually unchanged in both the hosts, suggesting an insignificant role for this cytokine in parasite establishment.

The Mongolian gerbil has been used as laboratory animal since 1935. Breeding gerbils as an isolated laboratory population for decades may have led to a domestication process whose effects include changes in brain size. Quantitative changes in testicular activity could be assumed. Comparative intraspecific measurements were performed in 34 adult males of the laboratory strain (LAB) and in males raised as offspring of wild Mongolian gerbils (WILD) caught in central Mongolia (F1, n = 16; F2, n = 17). LAB and WILD were examined in January. Testicular spermatozoa were counted, proportions of different cell types were analysed using DNA flow cytometry, and mitotic and meiotic activity was calculated from DNA histograms. Intratesticular testosterone concentrations were measured with an enzyme immunoassay. In the WILD, testicular activity was lower and varied more. The overall weight, the efficiency of spermatogenesis (sperm/g testis) and resulting total sperm per testis were significantly less in offspring of wild gerbils. This corresponded with lower levels of haploid cells, total germ cell transformation of diploid cells to spermatids and meiotic transformation of spermatocytes to spermatids. The most profound difference was found in testicular testosterone concentration: the mean level was 405.7 ± 41.2 ng/g testis in LAB vs 6.4 ± 2.0 ng/g in WILD F1. All parameters changed in WILD F2 generation compared with F1 and diminished the differences with LAB. Differences between F1 and F2 were significant for testis mass, testis/body weight ratio, percentages of haploid cells and cells in G2/M phase, both germ cell transformations and testosterone concentration. The results suggest rapid, adaptive changes of male reproductive physiology in the early offspring generations from wild populations under laboratory breeding conditions. The breeding of Mongolian gerbils in the laboratory has influenced the testicular function resulting in increased spermatogenic activity and highly stimulated testosterone production.

The effects of oat saponins (a mixture of avenacosides A and B) on plasma and liver lipids in gerbils (Meriones unguiculatus) and rats were investigated. Cholesterol-containing diets high in total and saturated fat and with different avenacoside contents (zero (ethanol-extracted oats), normal (oats) and twice normal (ethanol-extracted oats plus added avenacosides)) were used. Compared with a cellulose control group the oat diets in both species gave a significantly higher cholesterol content in the HDL fraction and a significantly lower liver cholesterol content. No significant differences in total plasma cholesterol, HDL-cholesterol and plasma triacylglycerols were found, however, between the groups fed on oats with different avenacoside content. The liver weight, total liver cholesterol and free liver cholesterol were also similar, whereas the liver lipid content was significantly lower in rats given the highest amount of avenacosides compared with zero or normal amounts. The tendency was the same in gerbils. Thus, the oat saponins had only minor effects on lipid metabolism in gerbils and rats.
