Articular cartilage consists of hyaline cartilage, is a major constituent of the human musculoskeletal system and has critical functions in frictionless joint movement and articular homoeostasis. Osteoarthritis (OA) is an inflammatory disease of articular cartilage, which promotes joint degeneration. Although it affects millions of people, there are no satisfying therapies that address this disease at the molecular level. Therefore, tissue regeneration approaches aim at modifying chondrocyte biology to mitigate the consequences of OA. This requires appropriate biochemical and biophysical stimulation of cells. Regarding the latter, mechanotransduction of chondrocytes and their precursor cells has become increasingly important over the last few decades. Mechanotransduction is the transformation of external biophysical stimuli into intracellular biochemical signals, involving sensor molecules at the cell surface and intracellular signalling molecules, so-called mechano-sensors and -transducers. These signalling events determine cell behaviour. Mechanotransducing ion channels and gap junctions additionally govern chondrocyte physiology. It is of great scientific and medical interest to induce a specific cell behaviour by controlling these mechanotransduction pathways and to translate this knowledge into regenerative clinical therapies. This review therefore focuses on the mechanotransduction properties of integrins, cadherins and ion channels in cartilaginous tissues to provide perspectives for cartilage regeneration.

Porphyrinic compounds are of increasing interest to the materials science community, yet little attention has been paid to crystallographically controlled voids and channels in these materials. We have conducted an initial survey of the voids and channels in a random subset of 1000 porphyrinic compounds with known crystal structures. From calculations using a rolling-probe subroutine, we have found that about 5% of these compounds have line-of-sight channels, which differ in their topology depending on the crystallography. A small but significant number of porphyrinic compounds have calculated void contents of >25 volume %. We discuss in detail the void and channel characteristics, including pore-size distribution, of four representative compounds, with technological implications.

Glycosomes are specialized peroxisomes found in all kinetoplastid organisms. The organelles are unique in harbouring most enzymes of the glycolytic pathway. Matrix proteins, synthesized in the cytosol, cofactors and metabolites have to be transported across the membrane. Recent research on Trypanosoma brucei has provided insight into how these translocations across the membrane occur, although many details remain to be elucidated. Proteins are imported by a cascade of reactions performed by specialized proteins, called peroxins, in which a cytosolic receptor with bound matrix protein inserts itself in the membrane to deliver its cargo into the organelle and is subsequently retrieved from the glycosome to perform further rounds of import. Bulky solutes, such as cofactors and acyl-CoAs, seem to be translocated by specific transporter molecules, whereas smaller solutes such as glycolytic intermediates probably cross the membrane through pore-forming channels. The presence of such channels is in apparent contradiction with previous results that suggested a low permeability of the glycosomal membrane. We propose 3 possible, not mutually exclusive, solutions for this paradox. Glycosomal glycolytic enzymes have been validated as drug targets against trypanosomatid-borne diseases. We discuss the possible implications of the new data for the design of drugs to be delivered into glycosomes.

Do tidal channels have a characteristic length? Given the sediment characteristics, the inlet conditions and the degree of channel convergence, can we predict it? And how is this length affected by the presence of tidal flats adjacent to the channel? We answer the above questions on the basis of a fully analytical treatment, appropriate for the short channels typically observed in coastal wetlands. The equilibrium length of non-convergent tidal channels is found to be proportional to the critical flow speed for channel erosion. Channel convergence causes concavity of the bed profile. Tidal flats shorten equilibrium channels significantly. Laboratory and field observations substantiate our findings.

Variations in velocity detection thresholds for full-field visual rotation about various axes are compatible with a simple channel-based system for coding the axis and velocity of the rotation (Harris & Lott, 1995). The present paper looks at the frame of reference for this system. The head-centered, craniotopic reference system and the retinal-based, retinotopic reference systems were separated by using eccentric eye positions. We measured the threshold for detecting full-field visual rotation about a selection of axes in the sagittal plane with the eyes held either 22½ degs up, straight ahead or 22½ degs down in the head. The characteristic features of the variation in detection thresholds did not stay stable in craniotopic coordinates but moved with the eyes and were constant in retinotopic coordinates. This suggests that the coding of head rotation by the visual system is in retinotopic coordinates.

Traditionally, it has been thought that the processing of binocular disparity for the perception of stereoscopic depth is accomplished via three types of disparity-selective channels – “near,” “far,” and “tuned.” More recent evidence challenges this notion. We have derived disparity-tuning functions psychophysically using a subthreshold summation (i.e. low-level masking) technique. We measured correlation-detection thresholds for dynamic random-element stereograms containing either one or two surfaces in depth. The resulting disparity-tuning functions show an opponent-type profile, indicating the presence of inhibition between disparity-tuned units in the visual system. Moreover, there is clear inhibition between disparities of the same sign, obviating a strict adherence to near-far opponency. These results compare favorably with tuning functions derived psychophysically using an adaptation technique, and with the tuning profiles from published single-unit recordings. Our results suggests a continuum of overlapping disparity-tuned channels, which is consistent with recent physiological evidence as well as models based on other psychophysical data.

An aftereffects paradigm was used to behaviorally measure contrast sensitivity of cats to gratings of three different test spatial frequencies after adaptation to gratings of various spatial frequencies, contrasts, and durations. Post-adaptation reductions in sensitivity occurred even after short periods of adaptation (<7 s) and could be as large as 1.0 log unit under some conditions. The magnitude of the adaptation effect varied monotonically with (1) adaptation grating contrast, (2) duration, and (3) the contrast sensitivity for the test grating. Average half-width (at half-height) of the spatial-frequency tuning curves constructed from the data was 1.4 octaves, and was not dependent upon the level of adaptation or the spatial frequency of the test grating. Post-adaptation psychometric functions of the cats showed reduced slopes and maxima suggesting that, unlike humans, in cats apparent contrast grows more slowly with increases in physical contrast after contrast adaptation. All of the characteristics observed are in excellent agreement with electrophysiologically measured properties of neurons in striate cortex of cats. In addition, there was a remarkable similarity of the cat tuning functions, both in shape and bandpass, to those measured in man with a similar paradigm suggesting that (1) the two visual systems are sufficiently similar to make the cat a useful spatial vision model and (2) there is a common functional plan to all mammalian visual systems despite significant anatomical differences between species.

Movement detection thresholds for full-field visual motion about various axes were measured in three subjects using a two-alternative forced-choice staircase method. Thresholds for 1-s exposures to rotation about different rotation axes varied significantly over the range 0.139 ± 0.05 deg/s to 0.463 ± 0.166 deg/s. The highest thresholds were found in response to rotation about axes closely aligned to the line of sight. Variations among the thresholds for different axes could not be explained by different movement patterns in the fovea or variations in motion sensitivity with eccentricity. The variations can be well simulated by a three-channel model for coding the axis and velocity of full-field visual motion. A three-channel visual coding system would be well suited for extracting information about self-rotation from a complex pattern of retinal image motion containing components due to both rotation and translation. A three-channel visual motion system would also be readily compatible with vestibular information concerning self-rotation arising from the semicircular canals.
