Summary
Introduction
People often wonder what discourse analysis is and what the field of study includes. As a newer field, discourse analysis is still determining its own boundaries. It is different from traditional linguistics in several ways – it uses natural data for analysis, and it deals with utterances, as opposed to sentences. If you spend time observing everyday talk, you will notice that these utterances rarely look like those grammatical sentences seen in the grammar books and often described in linguistic research about syntax and grammar. In everyday interaction, people do not always use complete “sentences,” they leave out information, they add meaning with their voice, or their faces, and they leave their comments unfinished. Yet, we still understand each other. An utterance, then, is the real-life expression of people's thoughts, ideas, and feelings.
Simply put, discourse analysis is the study of language in use. In discourse analysis, we study how people interact by expressing meaning using language within a context. This includes much more than just studying the words or signs that make up an utterance. It includes all the aspects of those utterances: pacing, facial expression, body shifting, gestures, and any other features that add meaning to an utterance. It also includes building understanding one utterance at a time, from beginning to end, in real time.
Neuropsychological Profile of Parkin Mutation Carriers with and without Parkinson Disease: The CORE-PD Study

The cognitive profile of early onset Parkinson’s disease (EOPD) has not been clearly defined. Mutations in the parkin gene are the most common genetic risk factor for EOPD and may offer information about the neuropsychological pattern of performance in both symptomatic and asymptomatic mutation carriers. EOPD probands and their first-degree relatives who did not have Parkinson’s disease (PD) were genotyped for mutations in the parkin gene and administered a comprehensive neuropsychological battery. Performance was compared between EOPD probands with (N = 43) and without (N = 52) parkin mutations. The same neuropsychological battery was administered to 217 first-degree relatives to assess neuropsychological function in individuals who carry parkin mutations but do not have PD. No significant differences in neuropsychological test performance were found between parkin carrier and noncarrier probands. Performance also did not differ between EOPD noncarriers and carrier subgroups (i.e., heterozygotes, compound heterozygotes/homozygotes). Similarly, no differences were found among unaffected family members across genotypes. Mean neuropsychological test performance was within normal range in all probands and relatives. Carriers of parkin mutations, whether or not they have PD, do not perform differently on neuropsychological measures as compared to noncarriers. The cognitive functioning of parkin carriers over time warrants further study. (JINS, 2011, 17, 1–10)

1.Two experiments of Latin square design were made, each with four Friesian bull calves fitted with re-entrant duodenal and ileal cannulas at 4–10 d of age. The calves were used to studythe effect of giving milk-substitutes containing 0, 300, 500 and 700 g bacterial protein (Pruteen)/kg total protein on apparent digestibility of nitrogen fractions and amino acids and true digestibility of 3H-labelled milk protein and 35S-labelled bacterial protein in the small intestine. A third experiment of Latin square design with four intact Friesian calves was used to measure apparent digestibility of nutrients throughout the alimentary tract and retention of N, calcium and phosphorus.
2.At the duodenum, volume of outflow, its pH, and outflow of total-N (TN), protein-N (PN) and non-protein-N (NPN) decreased with time after feeding. At the ileum, volume of outflow and TN outflow were unaffected by time after feeding but PN outflow decreased; NPN outflow at the ileum increased to a maximum 6 h after feeding and then declined.
3.Increased inclusion of Pruteen did not affect the volume of outflow at the duodenum or ileum, but duodenal PN outflow increased. At the ileum, pH values were lower and TN, PN and NPN outflows were higher with increasing concentration of Pruteen in the diet. Apparent digestibility in the small intestine tended to decrease with greater amounts of Pruteen, but was only significant for NPN. Apparent digestibility from mouth to ileum significantly decreased for TN and PN as Pruteen inclusion increased.
4.Amino acid concentration in duodenal outflow, with the exception of that of arginine, reflected intake. The total amount of each amino acid in ileal outflow increased and the apparent digestibility of most amino acids decreased with greater amounts of Pruteen in the diet. Apparent digestibility of nucleic acid-N from Pruteen was very high.
5.True digestibility in the small intestine and between mouth and ileum of 3H-labelled milk protein was high and did not differ between dietary treatments. True digestibility of 36S-labelled Pruteen was low for the milk-protein diet and tended to increase linearly as more dietary Pruteen was included.
6.Dry matter concentration in faeces and a high apparent digestibility throughout the whole alimentary tract of carbohydrates did not differ between treatments. Apparent digestibility of dry matter, organic matter, crude protein and fat, apparent absorption of Ca, P and ash throughout the tract, retention of N, Ca and P and biological value of the protein decreased with inclusion rates greater than 300 g Pruteen/kg total dietary protein. The amount of N apparently absorbed in the large intestine was estimated as 0.9 g/d.
7.Comparison of intake of apparently absorbed essential amino acids with requirement suggests that histidine is likely to be the limiting amino acid, assuming that arginine is synthesized in the body.
8.Efficiencies of utilization of protein for tissue synthesis and to cover obligatory loss are estimated as 0.80, 0.75, 0.66 and 0.47 for diets containing 0, 300, 500 and 700 g Pruteen/kg total protein in the diet. Efficiencies of utilization of individual essential amino acids were also estimated.

1. Two experiments of Latin square design, with four Friesian bull calves fitted with re-entrant duodenal cannulas at 4–10 d of age, were made to study the effect of giving varying levels of single-cell protein on the abomasal outflow and composition of digesta.
2. In Expt 1, diets in which 0,220,440, and 660 g/kg milk protein were replaced by the bacterial protein Pruteen were compared from 14 d of age. In Expt 2, which began at about 61 d of age, a comparison was made of diets in which 0, 220, 440 and 660 g/kg milk protein were replaced by the yeast protein Toprina.
3. Collection of abomasal outflow was made for 8 h after feeding for 2 d within each 6 d period of the Latin square design experiment. The amount of diet offered daily was 50 g dry matter/kg body-weight0.75. Polyethylene glycol (PEG), which was used as an indigestible marker, total nitrogen (TN), protein-N (PN), fat, and potassium, sodium and chloride ion outflows were measured together with pepsin (EC 3.4.23.1) and chymosin (EC 3.4.23.4) activities, pH and titratable acidity.
4. In Expt 1 there was little difference in the outflow of liquid digesta between diets and about 0.9 of the dietary PEG was recovered within the 8 h collection period. With increasing amounts of Pruteen in the diet, outflows of TN, PN, fat and Na+ increased, and the pH of digesta increased. However, the volume of 'apparent secretion' into the abomasum (outflow - intake), pepsin activity, chymosin activity, titratable acidity, (outflow of C1--outflow of Na+) as a measure of outflow of HC1, and outflows of K+ and of C1- were reduced. All outflows decreased with the time interval after feeding, except (Cl--Na+) outflow.
5. In Expt 2, the same trends as in Expt 1 were apparent, but since one calf had to be slaughtered and the experiment had to be analysed as a randomized block, only PN and K+ outflows and pH were significantly affected by dietary treatment, with K+ outflow increasing, rather than decreasing, with increasing concentration of single- cell protein in the diet.
6. Reduced proteolysis in the abomasum, associated with a faster and greater outflow of protein as a result of poor or no coagulation of protein in the abomasum, and a reduction in secretion of enzymes and in acidity may partly explain the poor protein digestibility and growth rate obtained in other experiments when diets containing more than 100 g single-cell protein/kg diet (about 200 g protein/kg total protein) were given to young calves.
